Abstract

Cognitive impairments play a key role in both schizophrenic as well as highly schizotypal individuals. Additionally, there is evidence of alterations in the metacognitive processes that accompany cognition, perception and motor control in schizophrenia. Less is known about metacognition in schizotypy. The goal of this study is to gain a better understanding of the relationship between possible metacognitive impairments associated with the subclinical trait of schizotypy. To investigate whether highly schizotypal individuals indeed show impairments in metacognition, we will assess 1) self-reported metacognitive beliefs as well as 2) task-based metacognition via trial-by-trial confidence ratings in an episodic memory framework. Based on the existing research on this topic, which has been summarized in our prior systematic literature review (https://osf.io/f9k26/), we will examine the following primary questions: 1. Are higher levels of schizotypy associated with variations in self-reported metacognitive beliefs? 2. Are higher levels of schizotypy associated with variations in metacognitive bias? 3. Are higher levels of schizotypy associated with impaired metacognitive performance in an episodic memory task? Further exploratory analyses will also investigate the possible relationship between schizotypy and a global pre-task estimate of task performance (~ prospective metacognition), and between schizotypy and subscale scores of the Self-Reflection and Insight Scale (SRIS).

Abstract

To characterize modes of action of substances as well as their cytotoxic effects Bionas GmbH has developed a new screening system to allow the continuous recording of how an active substance can act (Bionas® 2500 analyzing system). In the pharmaceutical and chemical industry as well as in environmental science it is important to acquire as much information as possible about the metabolic effects of an active substance or there cytotoxicity. With the Bionas® 2500 analyzing system metabolically relevant data including oxygen consumption, acidification rate and the adhesion (cell impedance) of cells can be measured in parallel, on-line and label-free. Using e.g. ion-sensitive field effect-transistors (ISFET) and electrode structures it is possible to observe metabolic parameters non-invasively and continuously over longer periods of time. The system has already been established for several cell models, cell lines as well as primary cells. The strength of our system can be found in the continuous data collection during the whole application period. Dynamic, reversible and / or regenerative processes can be observed in one and the same culture. Adaptation effects through repeated addition of compounds can also be observed. A long application period (hours to days) allows more realistic compound concentrations as it is possible in short-term experiments. This advantages result in a higher information content compared to end-point-methods. It also offers the advantage that regenerative effects can be observed during the same test run. A better understanding of the multifunctional cellular processing of input- and output-signals in living cells is fundamental for basic research, development of drugs and for environmental monitoring e.g. the detection of biotoxic agents. For on-line monitoring of cellular reactions we develop(ed) several Cell Monitoring Systems (CMS®). Abstract

International audience; In the past, we have deeply investigated the development of a Bêta-alumina potentiometric sensor with two coplanar metallic electrodes. Recently, many papers have demonstrated an interest for similar non-Nernstian devices based on stabilised zirconia (YSZ) electrolyte. The common particularity of such sensors is related to the use a dissymmetry of electrodes on an electrolyte and to work in a single gas cell. Our main experimental results obtained with Bêta-alumina sensors are reported with the special attention to develop gas sensors for exhaust applications. The sensors are evaluated under CO, HC and NO, CO. The selectivity depends on the sensor temperatures. The performances appear sufticient to satisfy in the future automotive applications like the control of NOxTrap or DeNOx devices. The measure of NH3 concentrations seems also possible. The comparison of performances of sensors replacing bêta-alumina electrolyte by YSZ is also presented. Similar behaviours are obtained for both types of sensors. These comparisons reinforce our proposal of a capacitive model mainly based on the difference of reactivity of the electrodes. It is pointed out that mixed potentials cannot explain the behaviour of our sensors under oxygen. Very good agreements are obtained between this capacitive model and the experimental results, not only versus oxygen, but also with mixture of oxygen and CO.

Abstract

Prediction of liver toxicity and compound responses continues to be a major challenge for the pharmaceutical industry. In vitro studies on liver cells have been developed to reduce or replace animal experiments. However, most of the tests in use are based on cell lines which do not necessarily represent normal cell physiology. We compared the response of primary human hepatocytes from two donors with primary rat hepatocytes and the cell line HepG2 to the test compound acetaminophen (AAP) by measuring oxygen consumption, extracellular acidification and cell adhesion as dynamic parameters of cell metabolism. Primary human hepatocytes were cultured on collagen pre-coated sensor chips or in conventional two-dimensional cultures in chemically defined Human Hepatocyte Maintenance Medium. This medium allows cultivation of functionally differentiated hepatocytes for several weeks. Sensor chip based results were compared with conventional assays for hepatocytes like albumin release and urea release. The hepatocytes were exposed to AAP (50-2815 mg/l) for 24 h. Cell respiration was inhibited by AAP concentrations of 500 mg/l and more in all three cell types, whereas only the cellular acidification rates and cell adhesion of the rat hepatocytes and the HepG2 cells were affected by AAP. In conventional cultures of human hepatocytes, AAP had no effect on cellular viability. Whereas high doses of AAP (2815 mg/l) diminished albumin secretion by 70-80%.
