Abstract

Peptide fragmentation and activity mapping of human lysozyme was carried out in order to define the structural and sequence requirements for anti-HIV activity. The findings suggest that the effects of HL9 on signaling pathways involved in survival, stress, TGF(beta), p53, NF(kappa)B, protein kinase C and hedgehog signaling may reflect changes in host cells that affect susceptibility to infection.

Abstract

The structure of anti-tumor and anti-HIV-1 plant protein MAP30, a ribosome-inactivating protein, has been investigated and structural insights into multiple functions gained. MAP30 comes from Momordica charantia, bitter melon, the extracts of which have long been used as therapeutic agents. The protein is among the largest single-chain proteins the solution structures of which have been found without a prior X-ray crystal structure. It behaves as would a DNA glycosylase/apurinic lyase.

Abstract

Human chorionic gonadotropin (hCG) preparations contain activity against HIV type 1 (HIV-1). However, there has been controversy about whether some biological activities of hCG [Beta]-subunit (hCG[Beta]) preparations are caused by the [Beta]-subunit itself or other proteins present in the preparations. We report here the purification, characterization, and identification of three enzymes with anti-HIV activity present in the [Beta]-core fraction of hCG[Beta] prepared from the urine of pregnant women. The N-terminal amino acid sequence of one protein is identical to human urinary lysozyme C, and those of the other two are identical to human RNase A and urinary RNase U. We thus refer to these proteins as AVL (antiviral lysozyme) and AVR (antiviral RNases). In addition to HIV-1 inhibition, AVL is capable of lysing Micrococcus lysodeikticus. AVR digests a variety of RNA substrates, including RNA from HIV-1-infected cells. We also find that lysozyme from chicken egg white, human milk, and human neutrophils and RNase A from bovine pancreas possess activity against HIV-1. These findings may offer additional strategies for the treatment of HIV-1 infection.

Abstract

Experiments were performed to measure the dipolar couplings of 1H-15N, 1H-13C' and 15N-13C' in a perdeuterated protein under high magnetic field conditions. Analysis of the data revealed the strengthening of 15N line-narrowing through the use of relaxation interference between 15N-1H and 15N chemical shift anisotropy. Findings suggested that the measurement of small heteronuclear dipolar couplings can extend the limit of protein structure determination by nuclear magnetic resonance.
