Abstract Optimal root system architecture (RSA) is critical for efficient resource capture in soils, hence being an interest in crop breeding. Seminal root angle (SRA) at the seedling stage in durum wheat has been suggested to be a good indicator of RSA. However, research on correlating such laboratory‐based seedling root phenotyping to RSA at later phases of plant growth is limited, resulting in the importance of root trait variation seen in seedlings often being overstated. To explore the role of SRA in modifying RSA at later phases of plant growth, we assessed 11 genotypes contrasting in SRA (wide and narrow), grown in a rhizobox designed for phenotyping root systems of plants during late‐tillering. Aboveground traits and root dry mass in different soil depths and across the entire soil volume were measured manually, while root architectural traits were extracted using image analysis and summarised by multiple factor analysis to describe RSA. When comparing the wide and narrow genotypes, no differences were detected for aboveground traits and total root dry mass. However, differences were observed in the allocation of root dry mass at different depths. The wide and narrow genotypes showed distinct RSAs, particularly in the upper soil (0–30 cm). The wide genotypes exhibited a ‘spread‐out’ root system with dense and thin roots, whereas the narrow genotypes had a compact root system with fewer but thicker roots. Our study demonstrated a clear difference in RSA between the wide and narrow genotypes, highlighting the association between SRA on the direction and distribution of root growth in plants at later growth stages.

Background: Sex-differential biology may contribute to the consistently male-biased prevalence of autism spectrum disorder (ASD). Gene expression differences between males and females in the brain can indicate possible molecular and cellular mechanisms involved, although transcriptomic sex differences during human prenatal cortical development have been incompletely characterized, primarily due to small sample sizes. Methods: We performed a meta-analysis of sex-differential expression and co-expression network analysis in 2 independent bulk RNA sequencing datasets generated from cortex of 273 prenatal donors without known neuropsychiatric disorders. To assess the intersection between neurotypical sex differences and neuropsychiatric disorder biology, we tested for enrichment of ASD–associated risk genes and expression changes, neuropsychiatric disorder risk genes, and cell type markers within identified sex-differentially expressed genes (sex-DEGs) and sex-differential co-expression modules. Results: We identified 101 significant sex-DEGs, including Y-chromosome genes, genes impacted by X-chromosome inactivation, and autosomal genes. Known ASD risk genes, implicated by either common or rare variants, did not preferentially overlap with sex-DEGs. We identified 1 male-specific co-expression module enriched for immune signaling that is unique to 1 input dataset. Conclusions: Sex-differential gene expression is limited in prenatal human cortex tissue, although meta-analysis of large datasets allows for the identification of sex-DEGs, including autosomal genes that encode proteins involved in neural development. Lack of sex-DEG overlap with ASD risk genes in the prenatal cortex suggests that sex-differential modulation of ASD symptoms may occur in other brain regions, at other developmental stages, or in specific cell types, or may involve mechanisms that act downstream from mutation-carrying genes.

Abstract As the human–primate interface expands, many nonhuman primate (NHP) populations exploit anthropogenic foods to survive, while some populations opportunistically target them. Though anthropogenic food consumption is sometimes associated with greater reproductive output and survival in these populations, there is a dearth of research on possible health effects. We explore how differential exposure to anthropogenic foods is linked to variation in isotopic compositions (δ13C and δ15N) and body weights in Barbary macaques (Macaca sylvanus) in the Upper Rock Nature Reserve, Gibraltar. We placed monkeys into three categories based on anthropogenic food exposure. We then analyzed individuals for isotopic signatures (N = 147) and body weight measurements (N = 80). Using the lowest exposure category as the comparison, we found body weights and δ15N values, but not δ13C values, significantly differed across key categories. Within categories, we found no significant associations between sex and δ13C or δ15N values, suggesting that individuals within categories consumed similar foods regardless of sex. We found a significant interaction effect between category and sex for predicting body weights. These results suggest that sex plays a role in how anthropogenic foods are accessed and consumed regardless of exposure, which may result in differential health profiles for female and male macaques.

Abstract Genomic selection in sugarcane faces challenges due to limited genomic tools and high genomic complexity, particularly because of its high and variable ploidy. The classification of genotypes for single nucleotide polymorphisms (SNPs) becomes difficult due to the wide range of possible allele dosages. Previous genomic studies in sugarcane used pseudo‐diploid genotyping, grouping all heterozygotes into a single class. In this study, we investigate the use of continuous genotypes as a proxy for allele‐dosage in genomic prediction models. The hypothesis is that continuous genotypes could better reflect allele dosage at SNPs linked to mutations affecting target traits, resulting in phenotypic variation. The dataset included genotypes of 1318 clones at 58K SNP markers, with about 26K markers filtered using standard quality controls. Predictions for tonnes of cane per hectare (TCH), commercial cane sugar (CCS), and fiber content (Fiber) were made using parametric, non‐parametric, and Bayesian methods. Continuous genotypes increased accuracy by 5%–7% for CCS and Fiber. The pseudo‐diploid parametrization performed better for TCH. Reproducing kernel Hilbert spaces model with Gaussian kernel and AK4 (arc‐cosine kernel with hidden layer 4) kernel outperformed other methods for TCH and CCS, suggesting that non‐additive effects might influence these traits. The prevalence of low‐dosage markers in the study may have limited the benefits of approximating allele‐dosage information with continuous genotypes in genomic prediction models. Continuous genotypes simplify genomic prediction in polyploid crops, allowing additional markers to be used without adhering to pseudo‐diploid inheritance. The approach can particularly benefit high ploidy species or emerging crops with unknown ploidy.

Soil associated with sea freight (shipping containers, flat racks and used machinery) arriving at New Zealand seaports was sampled for bacteria, fungi, nematodes, macro-invertebrates and plant seeds. Pseudomonads were selectively isolated, as several significant plant pathogens fall within this bacterial group. The mean and median sample weight collected from sea freight was found to be 417.3 and 152.7 g, respectively, with most recovered soil (73%) collected from the underside of shipping containers and flat rack containers. Likewise, for used machinery, most recovered soil (75%) was found under the machinery. Flat rack containers had significantly higher soil contamination compared to shipping containers and used machinery, but generally the counts and incidence of taxa were significantly lower compared to these other freight types. Viable bacteria, fungi, nematodes, seeds and arthropods were associated with the soil, with both counts g-1 and prevalence in samples varying with taxa, freight type, and location on the freight. Various regulated biosecurity organisms were recovered from the samples, including Aphelenchoides besseyi (rice white tip nematode), and seeds from genera such as Brachiaria, Cortaderia, Digitaria, Eragrostis and Sonchus. There were also live arthropod taxa that were not recorded as being present in New Zealand. No known plant pathogenic pseudomonads were identified through sequencing of the 16S ribosomal RNA gene. Shipping containers were found to be an important introduction pathway for exotic species, and therefore require careful monitoring and management. Comparisons of the incidence and mean number of organisms associated with soil on sea freight compared to a previous study with soil on footwear, generally showed that incidence and counts of many taxa were lower on sea freight, indicating that biosecurity risk can vary with pathway. However, prioritising one soil pathway over another according to the risks they present, and differentially allocating resources is problematic because the relative risk is dynamic, dictated by factors such as new pests or diseases entering the respective pathways.

Abstract The function of dreams is a longstanding scientific research question. Simulation theories of dream function, which are based on the premise that dreams represent evolutionary past selective pressures and fitness improvement through modified states of consciousness, have yet to be tested in cross-cultural populations that include small-scale forager societies. Here, we analyze dream content with cross-cultural comparisons between the BaYaka (Rep. of Congo) and Hadza (Tanzania) foraging groups and Global North populations, to test the hypothesis that dreams in forager groups serve a more effective emotion regulation function due to their strong social norms and high interpersonal support. Using a linear mixed effects model we analyzed 896 dreams from 234 individuals across these populations, recorded using dream diaries. Dream texts were processed into four psychosocial constructs using the Linguistic Inquiry and Word Count (LIWC-22) dictionary. The BaYaka displayed greater community-oriented dream content. Both the BaYaka and Hadza exhibited heightened threat dream content, while, at the same time, the Hadza demonstrated low negative emotions in their dreams. The Global North Nightmare Disorder group had increased negative emotion content, and the Canadian student sample during the COVID-19 pandemic displayed the highest anxiety dream content. In conclusion, this study supports the notion that dreams in non-clinical populations can effectively regulate emotions by linking potential threats with non-fearful contexts, reducing anxiety and negative emotions through emotional release or catharsis. Overall, this work contributes to our understanding of the evolutionary significance of this altered state of consciousness.

Abstract Mungbean (Vigna radiata (L.) R. Wilczek var. radiata) is a significant food and cash crop grown in tropical and subtropical regions. Mungbean production and consumer demand have increased substantially over the last two decades, owing to its agronomic, nutritional and economic benefits. Despite increased breeding efforts and the expansion of mungbean production in various agro‐climatic regions, further production is hindered by low yield and variability, which is partly attributed to the impacts of abiotic stress. Abiotic stress impacts on the physiology, morphology and reproductive ability of mungbean which influences yield. Exposure to abiotic stresses at the reproductive stage is considered the most critical for yield production. In this review, we evaluate how abiotic stress impacts mungbean growth and productivity when occurring during the reproductive stage and traits that may confer adaptation. We present the limitations of current research including limited number of genotypes, lack of field experiments and detailed experimental information. We highlight the opportunities to exploit new tools and technologies, such as high‐throughput phenotyping platforms, gene editing, and genomic selection, to accelerate breeding efforts to develop more resilient mungbean cultivars for today and tomorrow.

Mate-allocation strategies in breeding programs can improve progeny performance by harnessing non-additive genetic effects. These approaches prioritise predicted progeny merit over parental breeding value, making them particularly appealing for clonally propagated crops such as sugarcane. We conducted a comparative analysis of mate-allocation strategies, exploring utilising non-additive and heterozygosity effects to maximise clonal performance with schemes that solely consider additive effects to optimise breeding value. Using phenotypic and genotypic data from a population of 2,909 clones evaluated in final assessment trials of Australian sugarcane breeding programs, we focused on three important traits: tonnes of cane per hectare (TCH), commercial cane sugar (CCS), and Fibre. By simulating families from all possible crosses (1,225) with 50 progenies each, we predicted the breeding and clonal values of progeny using two models: GBLUP (considering additive effects only) and extended-GBLUP (incorporating additive, non-additive, and heterozygosity effects). Integer linear programming was used to identify the optimal mate-allocation among selected parents. Compared to breeding value-based approaches, mate-allocation strategies based on clonal performance yielded substantial improvements, with predicted progeny values increasing by 57% for TCH, 12% for CCS, and 16% for fibre. Our simulation study highlights the effectiveness of mate-allocation approaches that exploit non-additive and heterozygosity effects, resulting in superior clonal performance. However, there was a notable decline in additive gain, particularly for TCH, likely due to significant epistatic effects. When selecting crosses based on clonal performance for TCH, the inbreeding coefficient of progeny was significantly lower compared to random mating, underscoring the advantages of leveraging non-additive and heterozygosity effects in mitigating inbreeding depression. Thus, mate-allocation strategies are recommended in clonally propagated crops to enhance clonal performance and reduce the negative impacts of inbreeding.

In modern plant breeding, genomic selection is becoming the gold standard to select superior genotypes in large breeding populations that are only partially phenotyped. Many breeding programs commonly rely on single-nucleotide polymorphism (SNP) markers to capture genome-wide data for selection candidates. For this purpose, SNP arrays with moderate to high marker density represent a robust and cost-effective tool to generate reproducible, easy-to-handle, high-throughput genotype data from large-scale breeding populations. However, SNP arrays are prone to technical errors that lead to failed allele calls. To overcome this problem, failed calls are often imputed, based on the assumption that failed SNP calls are purely technical. However, this ignores the biological causes for failed calls—for example: deletions—and there is increasing evidence that gene presence–absence and other kinds of genome structural variants can play a role in phenotypic expression. Because deletions are frequently not in linkage disequilibrium with their flanking SNPs, permutation of missing SNP calls can potentially obscure valuable marker–trait associations. In this study, we analyze published datasets for canola and maize using four parametric and two machine learning models and demonstrate that failed allele calls in genomic prediction are highly predictive for important agronomic traits. We present two statistical pipelines, based on population structure and linkage disequilibrium, that enable the filtering of failed SNP calls that are likely caused by biological reasons. For the population and trait examined, prediction accuracy based on these filtered failed allele calls was competitive to standard SNP-based prediction, underlying the potential value of missing data in genomic prediction approaches. The combination of SNPs with all failed allele calls or the filtered allele calls did not outperform predictions with only SNP-based prediction due to redundancy in genomic relationship estimates.

Abstract Introduction Globally, soil‐borne plant pathogens are a significant contributor to plant yield loss in agricultural systems, including pasture production. The extent of soil biological constraints in high‐value dairy systems was assessed across three major dairying regions of New Zealand. Materials and Methods Regional measures of disease pressure were obtained by comparing white clover, perennial ryegrass and plantain growth in 30 farm soils with (non‐pasteurised) or without (microwave pasteurised) their normal complement of microbial and nematode pathogens. Results Pasteurising soils from the Waikato region led to significant average increases in clover (35%) and ryegrass (19%) shoot dry matter. Individual site yields increased up to 74% for clover and 38% for ryegrass. For Canterbury and Southland, there was no increase in plant yield with pasteurisation on a regional basis, but increases in either clover or ryegrass were significant at three individual farm sites. Across all regions, negative linear relationships between disease pressure (% growth change) and a proxy of soil organic matter quality (anaerobically mineralizable nitrogen:total nitrogen ratio were found for both clover and ryegrass, accounting for 34% of the variation in both clover and ryegrass growth. A positive linear relationship was found between disease pressure and Heterodera cyst nematode abundance, accounting for 33% and 17% of the variation in clover and ryegrass growth change, respectively. The most prevalent putative fungal and oomycete pathogens isolated from the roots and stem bases of clover and ryegrass seedlings were Fusarium oxysporum, Fusarium culmorum, and various Pythium species. Conclusion Our results suggest that root pathogens on New Zeland dairy farms are most prevalent and damaging in the Waikato region where economic modelling estimated clover and ryegrass root disease to cost 211 kg MS ha−1 year−1 in milk production and $909 ha−1 year−1 in farm profitability. Given the scale of these costs, targeted management of soil borne disease could present an economically viable approach to improving the resilience of these multi‐plant multi‐pathogen ecosystems.

Abstract Background The incorporation of root traits into elite germplasm is typically a slow process. Thus, innovative approaches are required to accelerate research and pre-breeding programs targeting root traits to improve yield stability in different environments and soil types. Marker-assisted selection (MAS) can help to speed up the process by selecting key genes or quantitative trait loci (QTL) associated with root traits. However, this approach is limited due to the complex genetic control of root traits and the limited number of well-characterised large effect QTL. Coupling MAS with phenotyping could increase the reliability of selection. Here we present a useful framework to rapidly modify root traits in elite germplasm. In this wheat exemplar, a single plant selection (SPS) approach combined three main elements: phenotypic selection (in this case for seminal root angle); MAS using KASP markers (targeting a root biomass QTL); and speed breeding to accelerate each cycle. Results To develop a SPS approach that integrates non-destructive screening for seminal root angle and root biomass, two initial experiments were conducted. Firstly, we demonstrated that transplanting wheat seedlings from clear pots (for seminal root angle assessment) into sand pots (for root biomass assessment) did not impact the ability to differentiate genotypes with high and low root biomass. Secondly, we demonstrated that visual scores for root biomass were correlated with root dry weight (r = 0.72), indicating that single plants could be evaluated for root biomass in a non-destructive manner. To highlight the potential of the approach, we applied SPS in a backcrossing program which integrated MAS and speed breeding for the purpose of rapidly modifying the root system of elite bread wheat line Borlaug100. Bi-directional selection for root angle in segregating generations successfully shifted the mean root angle by 30° in the subsequent generation (P ≤ 0.05). Within 18 months, BC2F4:F5 introgression lines were developed that displayed a full range of root configurations, while retaining similar above-ground traits to the recurrent parent. Notably, the seminal root angle displayed by introgression lines varied more than 30° compared to the recurrent parent, resulting in lines with both narrow and wide root angles, and high and low root biomass phenotypes. Conclusion The SPS approach enables researchers and plant breeders to rapidly manipulate root traits of future crop varieties, which could help improve productivity in the face of increasing environmental fluctuations. The newly developed elite wheat lines with modified root traits provide valuable materials to study the value of different root systems to support yield in different environments and soil types.

Abstract Background High-density SNP arrays are now available for a wide range of crop species. Despite the development of many tools for generating genetic maps, the genome position of many SNPs from these arrays is unknown. Here we propose a linkage disequilibrium (LD)-based algorithm to allocate unassigned SNPs to chromosome regions from sparse genetic maps. This algorithm was tested on sugarcane, wheat, and barley data sets. We calculated the algorithm’s efficiency by masking SNPs with known locations, then assigning their position to the map with the algorithm, and finally comparing the assigned and true positions. Results In the 20-fold cross-validation, the mean proportion of masked mapped SNPs that were placed by the algorithm to a chromosome was 89.53, 94.25, and 97.23% for sugarcane, wheat, and barley, respectively. Of the markers that were placed in the genome, 98.73, 96.45 and 98.53% of the SNPs were positioned on the correct chromosome. The mean correlations between known and new estimated SNP positions were 0.97, 0.98, and 0.97 for sugarcane, wheat, and barley. The LD-based algorithm was used to assign 5920 out of 21,251 unpositioned markers to the current Q208 sugarcane genetic map, representing the highest density genetic map for this species to date. Conclusions Our LD-based approach can be used to accurately assign unpositioned SNPs to existing genetic maps, improving genome-wide association studies and genomic prediction in crop species with fragmented and incomplete genome assemblies. This approach will facilitate genomic-assisted breeding for many orphan crops that lack genetic and genomic resources.

Category: Hindfoot; Trauma Introduction/Purpose: Calcaneus fractures are common injuries to the hindfoot. The safety of in-patient versus outpatient treatment in patients with calcaneus fractures remains unclear. The aim of the present study was to assess differences in wound complications and readmissions in operative calcaneus fractures treated with open reduction and internal fixation (ORIF) in an in- patient versus outpatient setting. Methods: Patients undergoing ORIF for calcaneus fractures from 2012 to 2020 were reviewed. Inclusion criteria were age greater than 18 years and an operative calcaneus fracture treated with the sinus tarsi approach (STA). Exclusion criteria consisted of a minimum of three months follow-up, open calcaneal fractures or fracture dislocations, inpatients with polytrauma, and patients without a preoperative computed tomography (CT) scan. A total of 113 patients met inclusion criteria with 24 (21%) managed inpatient and 89 (79%) managed as outpatient. The primary outcomes were deep infection defined as return to the operating room for debridement with positive cultures and readmissions. Secondary outcomes included implant related pain and unplanned return to the operating room. Results: Inpatients had a higher percentage of ASA classification 3&4 patients (58.3% vs 29.2%, p=0.008). Outpatients had a longer delay in days between injury and definitive fixation (mean 8 (8.9 SD, 0-31 range) vs. 14 (12.4 SD, 0-91 range) days, p=0.009). There were no statistically significant differences in the incidence of deep infections (8.3% vs. 4.5%, p=.606), implant related pain (8.3% vs. 15.7%, p=.516), return to the operating room (16.7% vs. 15.7%, p=1.0) or readmissions (4.2% vs. 3.4%, p=1.0) between inpatient and outpatient groups including in binary logistic regression models (p>.3 for all). In our retrospective study of patients undergoing operative repair of isolated calcaneus fractures with STA, there was no increased risk of wound complications or readmissions when calcaneus fractures were treated in an outpatient setting. Conclusion: In our retrospective study of patients undergoing operative repair of isolated calcaneus fractures with STA, there was no increased risk of wound complications or readmissions when calcaneus fractures were treated in an outpatient setting.

Abstract Sleep studies in small-scale subsistence societies have broadened our understanding of cross-cultural sleep patterns, revealing the flexibility of human sleep. We examined sleep biology among BaYaka foragers from the Republic of Congo who move between environmentally similar but socio-ecologically distinct locations to access seasonal resources. We analyzed the sleep–wake patterns of 51 individuals as they resided in a village location (n = 39) and a forest camp (n = 23) (362 nights total). Overall, BaYaka exhibited high sleep fragmentation (50.5) and short total sleep time (5.94 h), suggestive of segmented sleep patterns. Sleep duration did not differ between locations, although poorer sleep quality was exhibited in the village. Linear mixed effect models demonstrated that women’s sleep differed significantly from men’s in the forest, with longer total sleep time (β ± SE = − 0.22 ± 0.09, confidence interval (CI) = [− 0.4, − 0.03]), and higher sleep quality (efficiency; β ± SE = − 0.24 ± 0.09, CI = [− 0.42, − 0.05]). These findings may be due to gender-specific social and economic activities. Circadian rhythms were consistent between locations, with women exhibiting stronger circadian stability. We highlight the importance of considering intra-cultural variation in sleep–wake patterns when taking sleep research into the field.

Abstract Ice cores and other paleotemperature proxies, together with general circulation models, have provided information on past surface temperatures and the atmosphere's composition in different climates. Little is known, however, about past temperatures at high altitudes, which play a crucial role in Earth's radiative energy budget. Paleoclimate records at high‐altitude sites are sparse, and the few that are available show poor agreement with climate model predictions. These disagreements could be due to insufficient spatial coverage, spatiotemporal biases, or model physics; new records that can mitigate or avoid these uncertainties are needed. Here, we constrain the change in upper‐tropospheric temperature at the global scale during the Last Glacial Maximum (LGM) using the clumped‐isotope composition of molecular oxygen trapped in polar ice cores. Aided by global three‐dimensional chemical transport modeling, we exploit the intrinsic temperature sensitivity of the clumped‐isotope composition of atmospheric oxygen to infer that the upper troposphere (effective mean altitude 10–11 km) was 6–9°C cooler during the LGM than during the late preindustrial Holocene. A complementary energy balance approach supports a minor or negligible steepening of atmospheric lapse rates during the LGM, which is consistent with a range of climate model simulations. Proxy‐model disagreements with other high‐altitude records may stem from inaccuracies in regional hydroclimate simulation, possibly related to land‐atmosphere feedbacks.

Abstract This paper presents the response to anthropogenic forcing in the GISS‐E2.1 climate models for the 21st century Shared Socioeconomic Pathways emission scenarios within the Coupled Model Intercomparison Project Phase 6 (CMIP6). The experiments were performed using an updated and improved version of the NASA Goddard Institute for Space Studies (GISS) coupled general circulation model that includes two different versions for atmospheric composition: A non‐interactive version (NINT) with prescribed composition and a tuned aerosol indirect effect and the One‐Moment Aerosol model (OMA) version with fully interactive aerosols which includes a parameterized first indirect aerosol effect on clouds. The effective climate sensitivities are 3.0°C and 2.9°C for the NINT and OMA models, respectively. Each atmospheric version is coupled to two different ocean general circulation models: The GISS ocean model (E2.1‐G) and HYCOM (E2.1‐H). We describe the global mean responses for all future scenarios and spatial patterns of change for surface air temperature and precipitation for four of the marker scenarios: SSP1‐2.6, SSP2‐4.5, SSP4‐6.0, and SSP5‐8.5. By 2100, global mean warming ranges from 1.5°C to 5.2°C relative to 1,850–1,880 mean temperature. Two high‐mitigation scenarios SSP1‐1.9 and SSP1‐2.6 limit the surface warming to below 2°C by the end of the 21st century, except for the NINT E2.1‐H model that simulates 2.2°C of surface warming. For the high emission scenario SSP5‐8.5, the range is 4.6–5.2°C at 2100. Due to about 15% larger effective climate sensitivity and stronger transient climate response in both NINT and OMA CMIP6 models compared to CMIP5 versions, there is a stronger warming by 2100 in the SSP emission scenarios than in the comparable Representative Concentration Pathway (RCP) scenarios in CMIP5. Changes in sea ice area are highly correlated to global mean surface air temperature anomalies and show steep declines in both hemispheres, with the largest sea ice area decreases occurring during September in the Northern Hemisphere in both E2.1‐G (−1.21 × 106 km2/°C) and E2.1‐H models (−0.94 × 106 km2/°C). Both coupled models project decreases in the Atlantic overturning stream function by 2100. The largest decrease of 56%–65% in the 21st century overturning stream function is produced in the warmest scenario SSP5‐8.5 in the E2.1‐G model, comparable to the reduction in the corresponding CMIP5 GISS‐E2 RCP8.5 simulation. Both low‐end scenarios SSP1‐1.9 and SSP1‐2.6 also simulate substantial reductions of the overturning (9%–37%) with slow recovery of about 10% by the end of the 21st century (relative to the maximum decrease at the middle of the 21st century).

This large, international dataset contains survey responses from N = 12,570 students from 100 universities in 35 countries, collected in 21 languages. We measured anxieties (statistics, mathematics, test, trait, social interaction, performance, creativity, intolerance of uncertainty, and fear of negative evaluation), self-efficacy, persistence, and the cognitive reflection test, and collected demographics, previous mathematics grades, self-reported and official statistics grades, and statistics module details. Data reuse potential is broad, including testing links between anxieties and statistics/mathematics education factors, and examining instruments’ psychometric properties across different languages and contexts. By tracking a group of individuals through time, cohort studies provide fundamental insights into the developmental time course and causes of health and disease. Evolutionary life history theory seeks to explain patterns of growth, development, reproduction and senescence, and inspires a range of hypotheses that are testable using the longitudinal data from cohort studies. Here we review two decades of life history theory-motivated work conducted in collaboration with the Cebu Longitudinal Health and Nutrition Survey (CLHNS), a birth cohort study that enrolled more than 3000 pregnant women in the Philippines in 1983 and has since followed these women, their offspring and grandoffspring. This work has provided evidence that reproduction carries “costs” to cellular maintenance functions, potentially speeding senescence, and revealed an unusual form of genetic plasticity in which the length of telomeres inherited across generations is influenced by reproductive timing in paternal ancestors. Men in Cebu experience hormonal and behavioural changes in conjunction with changes in relationship and fatherhood status that are consistent with predictions based upon other species that practice bi-parental care. The theoretical expectation that early life cues of mortality or environmental unpredictability will motivate a “fast” life history strategy are confirmed for behavioural components of reproductive decision making, but not for maturational tempo, while our work points to a broader capacity for early life developmental calibration of systems like immunity, reproductive biology and metabolism. Our CLHNS findings illustrate the power of life history theory as an integrative, lifecourse framework to guide longitudinal studies of human populations.

Due to the climate change and an increased frequency of drought, it is of enormous importance to identify and to develop traits that result in adaptation and in improvement of crop yield stability in drought-prone regions with low rainfall. Early vigour, defined as the rapid development of leaf area in early developmental stages, is reported to contribute to stronger plant vitality, which, in turn, can enhance resilience to erratic drought periods. Furthermore, early vigour improves weed competitiveness and nutrient uptake. Here, two sets of a multi-reference nested association mapping (MR-NAM) population of bread wheat (Triticum aestivum ssp. aestivum L.) were used to investigate early vigour in a rain-fed field environment for 3 years, and additionally assessed under controlled conditions in a greenhouse experiment. The normalised difference vegetation index (NDVI) calculated from red/infrared light reflectance was used to quantify early vigour in the field, revealing a correlation (p < 0.05; r = 0.39) between the spectral measurement and the length of the second leaf. Under controlled environmental conditions, the measured projected leaf area, using a green-pixel counter, was also correlated to the leaf area of the second leaf (p < 0.05; r = 0.38), as well as to the recorded biomass (p < 0.01; r = 0.71). Subsequently, genetic determination of early vigour was tested by conducting a genome-wide association study (GWAS) for the proxy traits, revealing 42 markers associated with vegetation index and two markers associated with projected leaf area. There are several quantitative trait loci that are collocated with loci for plant developmental traits including plant height on chromosome 2D (log10 (P) = 3.19; PVE = 0.035), coleoptile length on chromosome 1B (–log10 (P) = 3.24; PVE = 0.112), as well as stay-green and vernalisation on chromosome 5A (–log10 (P) = 3.14; PVE = 0.115).

Effective surveillance for early detection of invasive alien species in natural ecosystems, or on valued plants found in modified areas, could prevent potentially devastating and costly impacts (whether environmental, economic or cultural) of new invasions on the invaded country. Surveillance technologies are often constrained by a range of factors. Determining which species present a significant risk before they reach the border is an effective strategy to minimize the possibility of invasion and/or the impact of invasion. Surveillance of sentinel plants provides an important tool to strengthen biosecurity programs assisting with i) detecting and identifying insect pests, nematodes and plant diseases that could potentially invade uncolonized countries, and ii) developing pest risk analysis profiles to eliminate or mitigate the risk of arrival. This review examines some of the challenges and opportunities provided by sentinel plant research and discusses the factors that could affect the success of their use for biosecurity risk assessment and surveillance in the New Zealand context.

Abstract Farmers around the world have recently experienced significant crop losses due to severe heat and drought. Such extreme weather events and the need to feed a rapidly growing population have raised concerns for global food security. While plant breeding has been very successful and has delivered today’s highly productive crop varieties, the rate of genetic improvement must double to meet the projected future demands. Here we discuss basic principles and features of crop breeding and how modern technologies could efficiently be explored to boost crop improvement in the face of increasingly challenging production conditions.

Landfills are a source of malodors, greenhouse gases, harmful pollutants, pests, noise, and litter. To reduce their impact on neighboring communities, landfill facilities and the policies they follow must reduce emissions of trace gases such as hydrogen sulfide (H2S) and methane (CH4). However, a comprehensive understanding of the spatial variability of both pollutants at landfills should first be established to obtain a clear picture of emissions at landfills. This study measured the mixing ratios of H2S and CH4 at two landfills in New York State (Fresh Kills Landfill and Seneca Meadows Landfill) in November 2021 using laser-based methods deployed in a mobile lab. H2S emission fluxes were estimated based on a mass balance calculation. The highest mixing ratios of both H2S and CH4 were measured at Fresh Kills Landfill, at up to 7 parts per billion (ppb) and ~140 parts per million (ppm), respectively, yet these values resulted in a low ΔH2S/ΔCH4 ratio, at approximately 5.2 ± 2.6 × 10−5 mol mol−1 and a H2S emission flux of 0.02 ± 0.01 mg m−2 day−1. The highest ΔH2S/ΔCH4 ratio was observed at the Seneca Meadows Landfill at 8.6 ± 4.3 × 10−4 mol mol−1 and yielded a H2S emission flux estimate of 17.7 ± 12.9 mg m−2-day−1. The variability in mixing ratios and ΔH2S/ΔCH4 ratios measured at the landfills can be attributed to various factors, including facility operations and design, landfill age, meteorology, types of waste, and pH levels, but further multiday measurements are needed at each landfill to improve emission estimates and determine a more accurate and resolute reasoning behind these variations.

Here the authors present the crystal structure of human MAJIN-TERB2 and combine biophysical approaches and structured illumination microscopy analysis of mouse meiotic chromosomes to characterize the molecular architecture of the wider MAJIN-TERB2-TERB1 complex and its interactions with TRF1.

Abstract Simulations of the CMIP6 historical period 1850–2014, characterized by the emergence of anthropogenic climate drivers like greenhouse gases, are presented for different configurations of the NASA Goddard Institute for Space Studies (GISS) Earth System ModelE2.1. The GISS‐E2.1 ensembles are more sensitive to greenhouse gas forcing than their CMIP5 predecessors (GISS‐E2) but warm less during recent decades due to a forcing reduction that is attributed to greater longwave opacity in the GISS‐E2.1 pre‐industrial simulations. This results in an atmosphere less sensitive to increases in opacity from rising greenhouse gas concentrations, demonstrating the importance of the base climatology to forcing and forced climate trends. Most model versions match observed temperature trends since 1979 from the ocean to the stratosphere. The choice of ocean model is important to the transient climate response, as found previously in CMIP5 GISS‐E2: the model that more efficiently exports heat to the deep ocean shows a smaller rise in tropospheric temperature. Model sea level rise over the historical period is traced to excessive drawdown of aquifers to meet irrigation demand with a smaller contribution from thermal expansion. This shows how fully coupled models can provide indirect observational constraints upon forcing, in this case, constraining irrigation rates with observed sea level changes. The overall agreement of GISS‐E2.1 with observed trends is familiar from evaluation of its predecessors, as is the conclusion that these trends are almost entirely anthropogenic in origin.

Abstract This paper describes the GISS‐E2.1 contribution to the Coupled Model Intercomparison Project, Phase 6 (CMIP6). This model version differs from the predecessor model (GISS‐E2) chiefly due to parameterization improvements to the atmospheric and ocean model components, while keeping atmospheric resolution the same. Model skill when compared to modern era climatologies is significantly higher than in previous versions. Additionally, updates in forcings have a material impact on the results. In particular, there have been specific improvements in representations of modes of variability (such as the Madden‐Julian Oscillation and other modes in the Pacific) and significant improvements in the simulation of the climate of the Southern Oceans, including sea ice. The effective climate sensitivity to 2 × CO2 is slightly higher than previously at 2.7–3.1°C (depending on version) and is a result of lower CO2 radiative forcing and stronger positive feedbacks.

Abstract Introduction The impact of crisaborole ointment, a nonsteroidal phosphodiesterase 4 inhibitor for the treatment of mild to moderate atopic dermatitis (AD), on quality of life (QoL) was assessed in two identically designed phase 3 studies (AD-301: NCT02118766; AD-302: NCT02118792, both at http://www.clinicaltrials.gov). Methods In both studies, patients aged ≥ 2 years with mild to moderate AD per the Investigator’s Static Global Assessment were randomly assigned 2:1 to receive crisaborole or vehicle twice daily for 28 days. QoL was assessed using the Children’s Dermatology Life Quality Index (CDLQI) (2–15 years), the Dermatology Life Quality Index (DLQI) (≥ 16 years), and the Dermatitis Family Impact Questionnaire (DFI) (parents/caregivers/family of patients aged 2–17 years). Established QoL score severity bands provided clinical context. Results Greater mean improvement in QoL was observed in crisaborole-treated patients than in vehicle-treated patients at day 29 [mean change from baseline (∆BL), CDLQI: − 4.6 vs. − 3.0; P < 0.001; DLQI: − 5.2 vs. − 3.5; P = 0.015]. At baseline, more than half the patients had a “moderate effect” or higher of AD on QoL. At day 29, there was a trend toward more crisaborole- than vehicle-treated patients having “small effect” to “no effect”, The QoL of parents/caregivers/family improved more for crisaborole-treated than for vehicle-treated patients (∆BL, DFI: − 3.7 vs. − 2.7; P = 0.003). Conclusion Crisaborole treatment results in clinically meaningful improvement in QoL for patients and their parents/caregivers/families. Aim: To determine if field-typical storage and collection conditions are related to salivary testosterone (T), cortisol (cort) and secretory immunoglobulin A (sIgA) as measured using commercially available kits. Subjects and methods: This study assessed whether storage time (∼6 months to 1.5 years) at −35 °C impacted levels of the measured biomarkers (n = 10,247 samples). For a sub-set of T samples (n = 2954), we also evaluated the impact of collection conditions, such as time spent at room temperature in participants’ homes (0–39 hours) and time spent in coolers (0.3–10 hours) in transit. Results: T was unrelated to storage and collection variables and there was no evidence of reduced sample fidelity at longer storage times. Abstract Background Plant breeding requires numerous generations to be cycled and evaluated before an improved cultivar is released. This lengthy process is required to introduce and test multiple traits of interest. However, a technology for rapid generation advance named ‘speed breeding’ was successfully deployed in bread wheat (Triticum aestivum L.) to achieve six generations per year while imposing phenotypic selection for foliar disease resistance and grain dormancy. Here, for the first time the deployment of this methodology is presented in durum wheat (Triticum durum Desf.) by integrating selection for key traits, including above and below ground traits on the same set of plants. This involved phenotyping for seminal root angle (RA), seminal root number (RN), tolerance to crown rot (CR), resistance to leaf rust (LR) and plant height (PH). In durum wheat, these traits are desirable in environments where yield is limited by in-season rainfall with the occurrence of CR and epidemics of LR. To evaluate this multi-trait screening approach, we applied selection to a large segregating F2 population (n = 1000) derived from a bi-parental cross (Outrob4/Caparoi). A weighted selection index (SI) was developed and applied. The gain for each trait was determined by evaluating F3 progeny derived from 100 ‘selected’ and 100 ‘unselected’ F2 individuals. Results Transgressive segregation was observed for all assayed traits in the Outrob4/Caparoi F2 population. Application of the SI successfully shifted the population mean for four traits, as determined by a significant mean difference between ‘selected’ and ‘unselected’ F3 families for CR tolerance, LR resistance, RA and RN. No significant shift for PH was observed. Conclusions The novel multi-trait phenotyping method presents a useful tool for rapid selection of early filial generations or for the characterization of fixed lines out-of-season. Further, it offers efficient use of resources by assaying multiple traits on the same set of plants. Results suggest that when performed in parallel with speed breeding in early generations, selection will enrich recombinant inbred lines with desirable alleles and will reduce the length and number of years required to combine these traits in elite breeding populations and therefore cultivars.

Pruritus occurs in all patients with atopic dermatitis and requires quick relief to reduce disease exacerbation and improve quality of life. Crisaborole ointment is a non-steroidal phosphodiesterase 4 inhibitor for the treatment of mild-to-moderate atopic dermatitis. This post hoc analysis explores crisaborole ointment for early relief of pruritus in patients with mild to moderate atopic dermatitis from 2 phase III studies. Patients received crisaborole or vehicle twice daily for 28 days. Pruritus was graded on a 4-point scale of none (0) to severe (3). Early improvement in pruritus required a score of none (0) or mild (1), with a ≥ 1-grade improvement from baseline on day 6. Plant parasitic nematodes (PPN) are known to survive periods of desiccation, an ability that increases the risk of them surviving unintentional transport between countries. To investigate nematode survival in soil subject to prolonged storage, soil collected from a native forest and an organic orchard was stored separately in cupboards at ambient temperature for 36 months. Subsamples were taken at 0, 3, 6, 12, 13, 24 and 36 months to determine the presence of plant parasitic and total nematodes using a standard misting technique. Pratylenchus was used as a model to determine if PPNs that had been under prolonged storage were able to infect plant hosts at 13, 24 and 36 months. Overall, the total number of nematodes recovered from stored soil declined over time, with differences in species diversity determined by molecular methods, related to soil origin. No PPN were recovered in soil stored beyond 13 months using the three-day misting technique. By comparison, Pratylenchus nematodes, using a baiting method, were found to successfully invade host plant roots (ryegrass and white clover) even after 36 months storage and were observed to produce offspring at 13 months. Baiting was not effective for Pratylenchus found in soil originally collected from the forest but was for orchard soil, a result attributed to the lack of suitable host plants for the Pratylenchus species found in forest soil. This study demonstrated, that in protected environments, nematodes could survive for at least 36 months and were observed to produce offspring at 13 months. Baiting with a host plant was more sensitive in detecting nematodes than using the misting extraction technique, although this approach only works where the host plant is known. Without a priori knowledge of the nematode-plant host association, plant baiting may also produce false negatives. In the context of plant biosecurity and providing an accurate risk assessment in soil contaminants, the development of a generic test for PPN that induces nematodes in a resting stage to emerge and respond to a cue would enhance the probability of detection. However, as assessments at the border are often time limited, a molecular based bioassay that can be used to indicate the presence of multiple species of live PPN species may be a more feasible option for risk assessments.

Abstract Introduction Testosterone and oxytocin are psychobiological mechanisms that interrelate with relationship quality between parents and the quantity and quality of parenting behaviors, thereby affecting child outcomes. Their joint production based on family dynamics has rarely been tested, particularly cross‐culturally. Methods We explored family function and salivary testosterone and oxytocin in mothers and fathers in a small‐scale, fishing‐farming society in Republic of the Congo. Fathers ranked one another in three domains of family life pertaining to the local cultural model of fatherhood. Results Fathers who were viewed as better providers had relatively lower oxytocin and higher testosterone than men seen as poorer providers, who had lower testosterone and higher oxytocin. Fathers also had higher testosterone and lower oxytocin in marriages with more conflict, while those who had less marital conflict had reduced testosterone and higher oxytocin. In contrast, mothers in conflicted marriages showed the opposite profiles of relatively lower testosterone and higher oxytocin. Mothers had higher oxytocin and lower testosterone if fathers were uninvolved as direct caregivers, while mothers showed an opposing pattern for the two hormones if fathers were seen as involved with direct care. Conclusions These results shed new light on parents' dual oxytocin and testosterone profiles in a small‐scale society setting and highlight the flexibility of human parental psychobiology when fathers' roles and functions within families differ across cultures.

The optimal root system architecture (RSA) of a crop is context dependent and critical for efficient resource capture in the soil. Narrow root growth angle promoting deeper root growth is often associated with improved access to water and nutrients in deep soils during terminal drought. RSA, therefore is a drought-adaptive trait that could minimize yield losses in regions with limited rainfall. Here, GWAS for seminal root angle (SRA) identified seven marker-trait associations clustered on chromosome 6A, representing a major quantitative trait locus (qSRA-6A) which also displayed high levels of pairwise LD (r2 = 0.67). Subsequent haplotype analysis revealed significant differences between major groups. Candidate gene analysis revealed loci related to gravitropism, polar growth and hormonal signaling. No differences were observed for root biomass between lines carrying hap1 and hap2 for qSRA-6A, highlighting the opportunity to perform marker-assisted selection for the qSRA-6A locus and directly select for wide or narrow RSA, without influencing root biomass. Our study revealed that the genetic predisposition for deep rooting was best expressed under water-limitation, yet the root system displayed plasticity producing root growth in response to water availability in upper soil layers. We discuss the potential to deploy root architectural traits in cultivars to enhance yield stability in environments that experience limited rainfall.
