The aim of this study is to set up statistical quality control for monitoring of volumetric modulated arc therapy (VMAT) delivery error using machine log data. Eclipse and Clinac iX linac with the RapidArc system (Varian Medical Systems, Palo Alto, USA) is used for delivery of the VMAT plan. During the delivery of the RapidArc fields, the machine determines the delivered motor units (MUs) and gantry angle position accuracy and the standard deviations of MU (sigma_MU; dosimetric error) and gantry angle (sigma_GA; geometric error) are displayed on the console monitor after completion of the RapidArc delivery. In the present study, first, the log data was analyzed to confirm its validity and usability; then, statistical process control (SPC) was applied to monitor the sigma_MU and sigma_GA in a timely manner for all RapidArc fields: a total of 195 arc fields for 99 patients. The sigma_MU and sigma_GA were determined twice for all fields, that is, first during the patient-specific plan QA and then again during the first treatment. The sigma_MU and sigma_GA time series were quite stable irrespective of the treatment site; however, the sigma_GA was strongly dependent on the gantry rotation speed. The sigma_GA of the RapidArc delivery for stereotactic body radiation therapy (SBRT) was smaller than that for the typical VMAT. Therefore, SPC was applied for SBRT cases and general cases respectively. Moreover, the accuracy of the potential meter of gantry rotation is important, since the sigma_GA can be dramatically changed due to its condition. By applying SPC to the sigma_MU and sigma_GA, we could monitor the delivery error efficiently. 5. Absorption spectrum of Gafchromic EBT2 film with angular rotation

It is important to study absorption spectrum in film dosimetry because the spectral absorbance of the film relates to the film's total absorption dose. We investigated the absorption spectra of Gafchromic EBT2 film with various rotational angles in a visible wavelength band. The film was irradiated with 6 MV photon beams and a total dose of 300 cGy. Absorption spectra were taken under different rotational angles after 24 h after irradiation and we fitted the spectra using Lorentzian functions. There were two dominant absorption peaks at approximately 586 nm (green) and 634 nm (red). The measured spectrum was decomposed 542 nm, 558 nm, 578 nm, 586 nm, 626 nm, 634 nm, and 641 nm. The maximum total area of the red band absorption spectrum was at 45{\deg}(225{\deg}) and the minimum at 90{\deg}(270{\deg}). As the angle of rotation changed, the intensity and integrated area of the blue and green peaks also changed with 180{\deg} period, with minima at 90{\deg} and 270{\deg}, and maxima at 0{\deg} and 180{\deg}, although the overall absorbance is very low. The spectral peak wavelengths remained constant within 2.4 nm for all angles. There was no hysteresis of absorption spectrum of the film; spectra taken at 0{\deg} and 360{\deg}were substantially the same and showed similar behavior for all rotational angles. The change of absorbance with rotational angle of the film affected the dosimetric properties, resulting in rotationalvariations of film dosimetry in each red-green-blue channel., Comment: 9pages,9figures. Background/Aim: To evaluate the early effect of radiation dose on liver function in breast cancer patients undergoing free-breathing volumetric modulated arc therapy (FB-VMAT). Patients and Methods: Medical records of 125 patients with breast cancer who underwent curative surgery followed by postoperative radiotherapy using FB-VMAT during 2018-2021 were reviewed. Results of the liver function test (LFT), performed within 1-week before and 6-months after radiotherapy, were collected and compared. The LFTs analyzed albumin, total and direct bilirubin, aspartate transaminase, alanine transferase, and alkaline phosphatase levels. The mean dose and relative liver volume receiving at least 10 Gy, 20 Gy, or 30 Gy were calculated. Results: Median follow-up time was 21.4 months. One patient experienced locoregional and distant failures. The mean liver irradiation dose was 325.9 centigray (cGy) for all patients. Ostreobium is a genus of siphonous green algae that lives as an endolith in carbonate substrates under extremely limited light conditions and has recently been gaining attention due to its roles in reef carbonate budgets and its association with reef corals. Knowledge about this genus remains fairly limited due to the scarcity of strains available for physiological studies. Here, we report on 10 strains of Ostreobium isolated from coral skeletons from the Great Barrier Reef. Phenotypic diversity showed differences in the gross morphology and in few structures. Phylogenetic analyses of the tufA and rbcL put the strains in the context of the lineages identified previously through environmental sequencing. The chloroplast genomes of our strains are all around 80k bp in length and show that genome structure is highly conserved, with only a few insertions (some containing putative protein-coding genes) differing between the strains. The addition of these strains from the Great Barrier Reef to our toolkit will help develop Ostreobium as a model species for endolithic growth, low-light photosynthesis and coral-algal associations.

Summary: Background: Bacille Calmette-Guérin (BCG) vaccination has off-target (non-specific) effects that are associated with protection against unrelated infections and decreased all-cause mortality in infants. We aimed to determine whether BCG vaccination prevents febrile and respiratory infections in adults. Methods: This randomised controlled phase 3 trial was done in 36 healthcare centres in Australia, Brazil, the Netherlands, Spain, and the United Kingdom. Healthcare workers were randomised to receive BCG-Denmark (single 0.1 ml intradermal injection) or no BCG in a 1:1 ratio using a web-based procedure, stratified by stage, site, age, and presence of co-morbidity. The difference in occurrence of febrile or respiratory illness were measured over 12 months (prespecified secondary outcome) using the intention-to-treat (ITT) population. This trial is registered with ClinicalTrials.gov, NCT04327206. Findings: Between March 30, 2020, and April 1, 2021, 6828 healthcare workers were randomised to BCG-Denmark (n = 3417) or control (n = 3411; no intervention or placebo) groups. The 12-month adjusted estimated risk of ≥1 episode of febrile or respiratory illness was 66.8% in the BCG group (95% CI 65.3%–68.2%), compared with 63.4% in the control group (95% CI 61.8%–65.0%), a difference of +3.4 percentage points (95% CI +1.3% to +5.5%; p 0.002). The adjusted estimated risk of a severe episode (defined as being incapacitated for ≥3 consecutive days or hospitalised) was 19.4% in the BCG group (95% CI 18.0%–20.7%), compared with 18.8% in the control group (95% CI 17.4%–20.2%) a difference of +0.6 percentage points (95% CI −1.3% to +2.5%; p 0.6). Both groups had a similar number of episodes of illness, pneumonia, and hospitalisation. There were three deaths, all in the control group. There were no safety concerns following BCG vaccination. Interpretation: In contrast to the beneficial off-target effects reported following neonatal BCG in infants, a small increased risk of symptomatic febrile or respiratory illness was observed in the 12 months following BCG vaccination in adults. There was no evidence of a difference in the risk of severe disease. Introduction: The first case of mpox in Louisiana was identified 2 months ahead of Southern Decadence Festival in New Orleans, the largest LGBTQ+ Pride festival in the South. With mpox case numbers reflecting racial disparities, the objective was to mount an equitable vaccination response. Methods: The Louisiana Department of Health rapidly pivoted its COVID-19 resources and strategies—specifically, using vaccine strike teams and mobile events, in-state vaccine redistribution through centralized warehousing and shipping support, and community partnerships—to now control mpox transmission. Here, the authors have evaluated state-based Immunization Information System data to examine whether the vaccination response was geographically and racially equitable. Geographic equity was measured by taking into account vaccine availability as well as uptake in areas with high Social Vulnerability Index. Results: A total of 113 providers were enrolled in the vaccination program, and 96 mobile vaccination events were held in locations frequented by at-risk populations. Racial disparities among vaccine recipients decreased over time, and vaccine availability and uptake were equitable in areas with high Social Vulnerability Indices. However, Black, female, and Hispanic/Latinx patients had significantly higher risk of not completing the 2-dose series than their counterparts. Conclusions: The mpox vaccination response in Louisiana was geographically equitable, though some demographic disparities remained.

We present MADLens a python package for producing non-Gaussian lensing convergence maps at arbitrary source redshifts with unprecedented precision. MADLens is designed to achieve high accuracy while keeping computational costs as low as possible. A MADLens simulation with only $256^3$ particles produces convergence maps whose power agree with theoretical lensing power spectra up to $L{=}10000$ within the accuracy limits of HaloFit. This is made possible by a combination of a highly parallelizable particle-mesh algorithm, a sub-evolution scheme in the lensing projection, and a machine-learning inspired sharpening step. Further, MADLens is fully differentiable with respect to the initial conditions of the underlying particle-mesh simulations and a number of cosmological parameters. These properties allow MADLens to be used as a forward model in Bayesian inference algorithms that require optimization or derivative-aided sampling. Another use case for MADLens is the production of large, high resolution simulation sets as they are required for training novel deep-learning-based lensing analysis tools. Summary: Background: The COVID-19 pandemic led to the rapid development and deployment of several highly effective vaccines against SARS-CoV-2. Recent studies suggest that these vaccines may also have off-target effects on the immune system. We sought to determine and compare the off-target effects of the adenovirus vector ChAdOx1-S (Oxford-AstraZeneca) and modified mRNA BNT162b2 (Pfizer-BioNTech) vaccines on immune responses to unrelated pathogens. Methods: Prospective sub-study within the BRACE trial. Blood samples were collected from 284 healthcare workers before and 28 days after ChAdOx1-S or BNT162b2 vaccination. SARS-CoV-2-specific antibodies were measured using ELISA, and whole blood cytokine responses to specific (SARS-CoV-2) and unrelated pathogen stimulation were measured by multiplex bead array. Findings: Both vaccines induced robust SARS-CoV-2 specific antibody and cytokine responses. ChAdOx1-S vaccination increased cytokine responses to heat-killed (HK) Candida albicans and HK Staphylococcus aureus and decreased cytokine responses to HK Escherichia coli and BCG. BNT162b2 vaccination decreased cytokine response to HK E. coli and had variable effects on cytokine responses to BCG and resiquimod (R848). After the second vaccine dose, BNT162b2 recipients had greater specific and off-target cytokine responses than ChAdOx1-S recipients. Interpretation: ChAdOx1-S and BNT162b2 vaccines alter cytokine responses to unrelated pathogens, indicative of potential off-target effects. The specific and off-target effects of these vaccines differ in their magnitude and breadth. The clinical relevance of these findings is uncertain and needs further study. In veterinary medicine and livestock production, ensuring good animal husbandry is vital for the physical and emotional wellbeing of animals under our care. Pain poses challenges for assessment and mitigation, especially in species unable to express pain overtly. This review examines current pain mitigation interventions in routine husbandry, focuses on the duration of pain after procedures and implications for animal welfare. Pain behaviours have been observed for days or weeks after regular husbandry procedures, and many studies have noted pain-related behaviour persisting until study finalisation, suggesting potential undocumented pain beyond study completion. Current products registered in Australia for pain mitigation in livestock primarily target immediate pain associated with procedures. The future of pain relief in livestock demands longer-acting solutions to address post-procedural pain adequately. Providing pain relief for at least 72 h post surgery is recommended, but current products require retreatment intervals to achieve this, posing practical challenges, especially in livestock. Methods of pain relief provision, such as voluntary consumption of medicated feed, transdermal medication delivery and long-acting formulations offer potential solutions for prolonged pain relief, with research ongoing in these areas. There is a need for further research and development of longer-acting pain relief to ensure optimal welfare of livestock.

Asprosin is a recently discovered fasting-induced hormone that promotes hepatic glucose production. Here we demonstrate that asprosin in the circulation crosses the blood-brain barrier and directly activates orexigenic AgRP+ neurons via a cAMP-dependent pathway. This signaling results in inhibition of downstream anorexigenic proopiomelanocortin (POMC)-positive neurons in a GABA-dependent manner, which then leads to appetite stimulation and a drive to accumulate adiposity and body weight. In humans, a genetic deficiency in asprosin causes a syndrome characterized by low appetite and extreme leanness; this is phenocopied by mice carrying similar mutations and can be fully rescued by asprosin. Furthermore, we found that obese humans and mice had pathologically elevated concentrations of circulating asprosin, and neutralization of asprosin in the blood with a monoclonal antibody reduced appetite and body weight in obese mice, in addition to improving their glycemic profile. Thus, in addition to performing a glucogenic function, asprosin is a centrally acting orexigenic hormone that is a potential therapeutic target in the treatment of both obesity and diabetes.

Summary: Background: Recurrences of herpes simplex virus (HSV) in the orofacial region (herpes labialis or cold sores) impact quality-of-life. We aimed to study whether the bacille Calmette-Guérin (BCG) vaccine can attenuate cold sore recurrences through off-target immunomodulatory effects. Methods: In this nested randomised controlled trial within the multicentre, phase 3 BRACE trial, 6828 healthcare workers were randomised in 36 sites in Australia, the Netherlands, Spain, the United Kingdom and Brazil, to receive BCG-Denmark or no BCG (1:1 ratio using a web-based procedure) and followed for 12 months with 3-monthly questionnaires. Exclusion criteria included contraindication to BCG vaccine or previous vaccination with BCG within the past year, any other live-attenuated vaccine within the last month, or any COVID-specific vaccine. The intervention group received one intradermal dose of 0.1 mL of BCG-Denmark corresponding to 2−8 x 105 colony forming units of Mycobacterium bovis, Danish strain 1331. The primary outcome was the difference in restricted mean survival time (i.e., time to first cold-sore recurrence), in participants with frequent recurrent herpes labialis (≥4 recurrences/year), analysed by intention-to-treat. Secondary outcomes addressed additional questions, including analyses in other sub-populations. Adverse events were monitored closely during the first 3 months and were reported in all participants who received one dose of study drug according to intervention received. The BRACE trial is registered with ClinicalTrials.gov, NCT04327206. Findings: Between March 30, 2020 and February 18, 2021, 84 individuals with frequent recurrent cold sores were randomly assigned to BCG (n = 38) or control (n = 46). The average time to first cold-sore recurrence was 1.55 months longer in the BCG group (95% CI 0.27–2.82, p = 0.02) than the control group (hazard ratio 0.54, 95% CI 0.32–0.91; intention-to-treat). The beneficial effect of BCG was greater in the as-treated population (difference 1.91 months, 95% CI 0.69–3.12, p = 0.003; hazard ratio 0.45, 95% CI 0.26–0.76). In prespecified subgroup analyses, only sex modified the treatment effect (interaction p = 0.007), with benefit restricted to males. Over 12 months, a greater proportion of participants in the BCG group compared with the control group reported a decrease in duration (61% vs 21%), severity (74% vs 21%), frequency (55% vs 21%), and impact on quality of life (42% vs 15%) of cold sore recurrences. In participants who had ever had a cold sore, there was also a decrease in self-reported burden of recurrences in the BCG group. In participants who had never had a cold sore, there was an increased risk of a first episode in the BCG group (risk difference 1.4%; 95% CI 0.3–2.6%, p = 0.02). There were no safety concerns. Interpretation: BCG-Denmark vaccination had a beneficial effect on herpes labialis, particularly in males with frequent recurrences, but may increase the risk of a first cold sore. In this paper, we introduce Notice, Wonder, Feel, Act, and Reimagine (NWFAR) to promote social justice in data science (DS) education. NWFAR draws on intersectional feminist DS to scaffold critical perspectives towards systems of power and oppression and attend to students’ experiences in designs for learning. NWFAR adds three practices that are typically not emphasized in learning designs for DS: feel—engaging emotions and the physical body; act—challenging, inspiring, or informing others towards change; and reimagine—envisioning how data, data methods, and data technologies could pursue different problems, solutions, and perspectives. We illustrate NWFAR through two design-based research projects from prior empirical work. Through these two examples, we demonstrate what thinking with NWFAR could look like in practice and highlight future possibilities for learning. We conclude with a discussion that focuses on the reimagining dimension, in which we highlight social-justice oriented theories.

Abstract Effective teams are essential for optimally functioning societies. However, little is known regarding the neural basis of two or more individuals engaging cooperatively in real-world tasks, such as in operational training environments. In this exploratory study, we recruited forty individuals paired as twenty dyads and recorded dual-EEG at rest and during realistic training scenarios of increasing complexity using virtual simulation systems. We estimated markers of intrinsic brain activity (i.e., individual alpha frequency and aperiodic activity), as well as task-related theta and alpha oscillations. Using nonlinear modelling and a logistic regression machine learning model, we found that resting-state EEG predicts performance and can also reliably differentiate between members within a dyad. Task-related theta and alpha activity during easy training tasks predicted later performance on complex training to a greater extent than prior behaviour. These findings complement laboratory-based research on both oscillatory and aperiodic activity in higher-order cognition and provide evidence that theta and alpha activity play a critical role in complex task performance in team environments.

This study marks a significant stride in nanotechnology, particularly in catalyst development, focusing on magnesium oxide nanoparticles and plant extracts from Cajanus cajan (L.) and Melaleuca leucadendra (L.), noted for their antioxidant properties. We aimed to unravel how these substances, both as traditional extracts and as nanoparticles, perform in various applications due to their antioxidant potential. Utilizing DPPH and FRAP assays, the research revealed that Ascorbic acid consistently exhibited strong antioxidant capabilities, serving as a reliable benchmark. Interestingly, Cajanus cajan (L.) and M. leucadendra (L.) extracts varied in their antioxidant effectiveness. A key finding was the pronounced increase in antioxidant efficacy when these extracts, particularly from Cajanus cajan (L.), were transformed into nanoparticles, as reflected in elevated FRAP values. This observation underscores the potential of nanoparticles to significantly enhance the effectiveness of plant extracts. The implications of this advancement are far-reaching, opening new avenues in the pharmaceutical and nutraceutical industries for developing therapeutic agents and antioxidant-rich foods. This research contributes notably to pharmaceutical sciences, emphasizing the vital role of nanoparticle technology in enhancing the antioxidant qualities of plant-based substances. It lays a solid foundation for further exploration into the mechanisms underlying nanoparticle-mediated improvements, offering valuable insights into the application of nanotechnology in health and nutrition.

Social intelligence empowers individuals not only to comprehend and interpret written discourse but also to respond effectively and appropriately. In the context of job applications, the ability to demonstrate the presence of social intelligence through documents such as resumes can significantly persuade recruiters’ interest. However, many future graduates are not concerned about producing impactful resumes. The way a job advertisement is presented can affect how a resume is crafted. This paper explores how future graduates interpret job advertisements and shape their resumes. One postgraduate student from a technical university in Malaysia participated in this study. This study employed think-aloud protocols to examine the cognitive processes behind the resume composition. The data from the think-aloud and the language used in the advertisements and resumes were analyzed through thematic and discourse analysis, respectively. The results generated from the analysis were mapped against the social intelligence dimensions to determine the presence of social intelligence. The key finding highlights that social intelligence plays an important role in helping future graduates understand and interpret job advertisements to enable them to produce impactful resumes. The study raises the relevance of social intelligence for future professionals.

Abstract Background Land plants respond to drought and salinity by employing multitude of sophisticated mechanisms with physiological and developmental consequences. Abscisic acid-mediated signaling pathways have evolved as land plant ancestors explored their habitats toward terrestrial dry area, and now play major roles in hyperosmotic stress responses in flowering plants. Green algae living in fresh water habitat do not possess abscisic acid signaling pathways but need to cope with increasing salt concentrations or high osmolarity when challenged with adverse aquatic environment. Hyperosmotic stress responses in green algae are largely unexplored. Results In this study, we characterized hyperosmotic stress-induced cytoskeletal responses in Chlamydomonas reinhardtii, a fresh water green algae. The Chlamydomonas PROPYZAMIDE-HYPERSENSITEVE 1 (PHS1) tubulin kinase quickly and transiently phosphorylated a large proportion of cellular α-tubulin at Thr349 in G1 phase and during mitosis, which resulted in transient disassembly of microtubules, when challenged with > 0.2 M sorbitol or > 0.1 M NaCl. By using phs1 loss-of-function algal mutant cells, we demonstrated that transient microtubule destabilization by sorbitol did not affect cell growth in G1 phase but delayed mitotic cell cycle progression. Genome sequence analyses indicate that PHS1 genes evolved in ancestors of the Chlorophyta. Interestingly, PHS1 genes are present in all sequenced genomes of freshwater Chlorophyta green algae (including Chlamydomonas) but are absent in some marine algae of this phylum. Conclusion PHS1-mediated tubulin phosphorylation was found to be partly responsible for the efficient stress-responsive mitotic delay in Chlamydomonas cells. Ancient hyperosmotic stress-triggered cytoskeletal remodeling responses thus emerged when the PHS1 tubulin kinase gene evolved in freshwater green algae.

Two site-directed mutants of isocitrate dehydrogenase (IDH) of Escherichia coli have been studied by site-directed mutagenesis kinetic and structural studies. Substitution of phenylalanine for tyrosine at position 160 (Y160F) showed 0.4% of the kcat of wild-type with isocitrate as substrate, while the Km for isocitrate remained unchanged. When the postulated intermediate, oxalosuccinate, was enzymatically decarboxylated, Y160F showed a higher kcat and a similar Km to the wild type values. The rate of reduction of oxalosuccinate to isocitrate by the Y160F mutant was greatly decreased relative to the wild-type. Substitution of methionine for lysine at position 230 decreased kcat to 1.1% of that of the wild-type and Km increased by a factor of 500-600. The decarboxylation of oxalosuccinate was undetectable for the K230M mutant. The structure of the site-directed mutants of IDH with and without a bound complex of isocitrate and Mg2+ was solved at 2.5 A resolution and compared by difference mapping against previously determined enzyme structures. The structural studies show that (i) the overall protein-folding side chain conformations and active sites of both mutants are isomorphous with wild-type enzyme, (ii) isocitrate and magnesium bind to both enzyme mutants with the same relative conformation and binding interactions as wild-type enzyme, and (iii) the mutated side chains (Phe 160 and Met 230) are positioned for catalysis in a similar conformation as that observed for the wild-type enzyme. Hence, the alteration of the side chain functional groups is directly related to the loss of enzyme activity. Possible roles of the active site tyrosine and lysine are discussed.

The optical properties of colored wing scales of the Chrysiridia croesus moth were investigated experimentally and theoretically by reflection spectroscopy and ellipsometry. Transmission electron microscope micrographs show that the outer surfaces of these scales incorporate a fairly regular layered structure of alternating dense and less-dense material, which reflects light by the well-known thin-film interference process. A Monte Carlo–type simulation of the reflection process is discussed, which permits the determination of the complex index of refraction of the scale material.
