Abstract

Polarizing beam splitters (PBSs) are fundamental components of optical systems and are crucial for sensing, communication, and imaging tasks. Traditional PBS devices, assembled using right-angle prisms with dielectric coatings, face challenges such as bulkiness and limited versatility in deflection directions. To address these limitations, we meticulously make metasurfaces for enhanced PBS performance. Metasurfaces, composed of subwavelength structures, manipulate wavefronts, polarization, and light intensity. Using metasurfaces in the design of PBS devices, we can precisely tailor the structure to manipulate the deflection angles of light beams, ensuring that they align with the desired specifications. Our experimental results closely align with simulation outcomes, showcasing deflection angles of a 1.5 mm diameter metasurface near ±15 degrees for s- and p-polarizations in a wavelength of 940-nm. Abstract

For an analog‐based design of novel Wee1 inhibitors, we profiled in vitro ADMET and in vivo PK properties of adavosertib. Based on the properties of adavosertib, we aimed to improve its metabolic stability by designing a novel target compound 1a with an aminosulfonyl group instead of the 2‐hydroxypropan‐2‐yl moiety in adavosertib. Derivatives of target compound 1a were synthesized and evaluated for Wee1 enzyme inhibition and liver microsomal phase I stability. We identified compound 1a as a sub‐nanomolar Wee1 inhibitor and 10 additional compounds with one‐digit nanomolar Wee1 inhibitory activity, among which seven compounds including 1a exhibited improved metabolic stabilities compared with adavosertib. However, MDA‐MB‐231 cell growth inhibitory activities of all synthesized compounds and Wee1 substrate phosphorylation inhibitory activities of selected compounds were inferior to adavosertib overall. It was renamed as telomeric zinc finger-associated protein (TZAP) binding to elongated telomeres. However, its expression level was not measured in cancers.Patients and Methods: We analyzed TZAP mRNA levels in 60 colorectal cancers (CRC) and its correlation with telomere length and TERT was studied.Results: TZAP mRNA in CRC was higher statistically than that in paired non-cancerous tissues (p = 0.033). Higher TZAP was found in carcinoembryonic antigen (CEA)-positive CRCs (> 5 ng/mL) (p = 0.012). Shorter telomere was found in CRCs with high TZAP expression than that with low TZAP expression (p = 0.010). According to quantitative correlation analysis, TZAP has a correlation with age (r = − 0.349, p = 0.007), TERT (r = 0.279, p = 0.041) and telomere length (r = − 0.305, p = 0.021). TZAP expression did not harbor prognostic value in CRC. Inhibition of TZAP expression by siRNA suppresses cell growth in HT29 cells; however, it resulted in increased cell viability in HCT116 cells. TZAP inhibition induces a decrease in mRNA levels of TERT in both HT29 and HCT116 cells. TCGA data analysis showed higher expression of TZAP showed poorer overall survival in colon cancer (p = 0.001); however, it did not have a significance in rectal cancer (p = 0.951).Conclusion: We suggested that TZAP may be a possible biomarker for CRC.Keywords: TZAP, telomere, colorectal cancer, ZBTB48
