Abstract

In vivo insulin sensitivity can be assessed using “open loop” clamp or “closed loop” methods. Open loop clamp methods are static, and fix plasma glucose independently from plasma insulin. Closed loop methods are dynamic, and assess glucose disposal in response to a stable isotope labeled glucose tolerance test. Using PPARα−/− mice, open and closed loop assessments of insulin sensitivity/glucose disposal were compared. Indirect calorimetry done for the assessment of diurnal substrate utilization/metabolic flexibility showed that chow fed PPARα−/− mice had increased glucose utilization during the light (starved) cycle. Euglycemic clamps showed no differences in insulin stimulated glucose disposal, whether for chow or high fat diets, but did show differences in basal glucose clearance for chow fed PPARα−/− versus SV129J-wt mice. In contrast, the dynamic stable isotope labeled glucose tolerance tests reveal enhanced glucose disposal for PPARα−/− versus SV129J-wt, for chow and high fat diets. Abstract

De novo lipogenesis and dietary fat uptake are two major sources of fatty acid deposits in fat of obese animals. To determine the relative contribution of fatty acids from these two sources in obesity, we have determined the distribution of c16 and c18 fatty acids of triglycerides in plasma, liver, and epididymal fat pad of Zucker diabetic fatty (ZDF) rats and their lean littermates (ZL) under two isocaloric dietary fat conditions. Lipogenesis was also determined using the deuterated water method. Conversion of palmitate to stearate and stearate to oleate was calculated from the deuterium incorporation by use of the tracer dilution principle. In the ZL rat, lipogenesis was suppressed from 70 to 24%, conversion of palmitate to stearate from 86 to 78%, and conversion of stearate to oleate from 56 to 7% in response to an increase in the dietary fat-to-carbohydrate ratio. The results suggest that suppression of fatty acid synthase and stearoyl-CoA desaturase activities is a normal adaptive mechanism to a high-fat diet. In contrast, de novo lipogenesis, chain elongation, and desaturation were not suppressed by dietary fat in the ZDF rat. Abstract

Underestimation of gluconeogenesis by the [[U-.sup.13][C.sub.6]]glucose method: effect of lack of isotope equilibrium. Am J Physiol Endocrinol Metab 282: E376-E385, 2002. First published October 16, 2001; 10.1152/ajpendo.00210.2001.--Among the many tracer methods to indirectly estimate gluconeogenesis in humans, the [[U-.sup.13][C.sub.6]]glucose method as proposed by Tayek and Katz (Am J Physiol Endocrinol Metab 270: E709-E717, 1996; Am J Physiol Endocrinol Metab 272: E476-E484, 1997) has the advantage of being able to simultaneously estimate hepatic glucose output and fractional gluconeogenesis. However, Landau et al. (Landau BR, J Wahren, K Ekberg, SF Previs, D Yang, and H Brunengraber. Am J Physiol Endocrinol Metab 274: E954-E961, 1998) have shown that this method underestimates the rate of gluconeogenesis. The underestimation has been attributed to tracer dilution by other three-carbon substrates and the lack of isotopic steady state. Using a computer simulation of [[U-.sup.13][C.sub.6]]glucose infusion, we demonstrate that the lack of isotope equilibrium in both the lactate and glucose compartments contributes substantially to the underestimation of gluconeogenesis. [[U-sup.13][C.sub.6]]glucose experiments were performed with the addition of a primed constant infusion of [[U-.sup.13][C.sub.3]]lactate and the delay in M3 glucose equilibrium estimated from the isotopic steady-state value determined by modeling M3 glucose to a single-exponential fit. We found that, even with the addition of [[U-.sup.13][C.sub.3]]lactate infusion, the M3 glucose enrichment of the last timed sample was ~20% less than the isotopic steady-state value. Thus the lack of isotopic equilibrium of the glucose compartment potentially accounts for 20% of the underestimation of gluconeogenesis. The underestimation of gluconeogenesis using [[U-.sup.13][C.sub.6]] glucose without the additional infusion of [[U-.sup.13][C.sub.3]] lactate in previous publications is expected to be even greater because of the lack of isotope equilibrium in both the lactate and glucose compartments. Abstract

Two techniques of measuring rates of gluconeogenesis based on label redistribution after the introduction of (U-13C)glucose into the whole body were investigated. These techniques were contrasted with methods previously derived for carbon-14 tracers. Results showed that the three approaches (stoichiometric, dilution and combinatorial) are equivalent, provided the same set of assumptions are employed.

Abstract

Background. This study sought to compare the ultrasound tissue characteristics of the muscles (including muscle thickness, echo intensity, and stiffness) and tendons (including thickness, stiffness, and peak spatial frequency radius (PSFR)) of participants with or without diabetes mellitus. Moreover, the study sought to determine any relationships between the muscle stiffness and strength or tendon PSFR and the results of blood tests, including the glycation or lipid profiles, of the diabetics. Methods. Twenty-three participants with type 2 diabetes mellitus and nineteen controls without a history of diabetes were recruited. Results. The diabetic muscles exhibited less thickness (P=0.024), greater echo intensity (P=0.033 and 0.002), and lower muscle stiffness (P=0.015 and 0.009) than the control muscles. Furthermore, the diabetic tendons exhibited a lower PSFR (P ranged between 0.037 and <0.001). There were correlations between the resting stiffness of the gastrocnemius muscle and the height of heel lifting (r=0.450, P=0.031), between the PSFR in the patellar tendon and the hemoglobin A1c level (r= -0.539, P=0.017), and between the PSFR in the Achilles tendon and the high-density lipoprotein cholesterol level (r=0.545, P=0.019). Conclusions. The diabetic muscles and tendons exhibited morphomechanical changes associated with force capacity or markers of insulin resistance. Clinical applications of musculoskeletal ultrasound techniques to diabetics include using them to design exercise strategies and for microstructural screening. [ABSTRACT FROM AUTHOR]

Abstract

The in-plane aligned MnAs nanowires have been grown by molecular-beam epitaxy on GaAs(001) substrates at high growth temperature (≥450 °C). A discontinuous growth with break intervals (50 s’ interval per 10 s’ growth) was employed. The obtained nanowires were identified to be mainly type-B hexagonal MnAs. The influences of growth temperature and As4/Mn flux ratio on the nanowires’ morphology were investigated. Both high growth temperature and high As4/Mn flux ratio are necessary for the growth of uniaxially aligned MnAs nanowires with high aspect ratio. The magnetic anisotropy of the nanowires and their multimodal size distributions contribute to the large coercivity and special shape of the M-H loops along the magnetic easy axis, which is [1102]MnAs∥[110]GaAs. However, the longer growth time would lead to the both azimuthal alignments of the MnAs wires and the weakening of the magnetic anisotropy. [ABSTRACT FROM AUTHOR]

Am J Physiol Endocrinol Metab 279: E425-E432, 2000.--We present here a study on the role of leptin in the regulation of lipogenesis by examining the effect of dietary macronutrient composition on lipogenesis in the leptin receptor-defective Zucker diabetic fatty rat (ZDF) and its lean litter mate (ZL). Animals were pair fed two isocaloric diets differing in their fat-to-carbohydrate ratio providing 10 and 30% energy as fat. Lipogenesis was measured in the rats using deuterated water and isotopomer analysis. From the deuterium incorporation into plasma palmitate, stearate, and oleate, we determined de novo synthesis of palmitate and synthesis of stearate by chain elongation and of oleate by desaturation. Because the macronutrient composition and the caloric density were controlled, changes in de novo lipogenesis under these dietary conditions represent adaptation to changes in the fat-to-carbohydrate ratio of the diet. De novo lipogenesis was normally suppressed in response to the high-fat diet in the ZL rat to maintain a relatively constant amount of lipids transported. The ZDF rat had a higher rate of lipogenesis, which was not suppressed by the high-fat diet. 30. Effect of substrate orientation on magnetic properties of (Ga, Mn)As.

Abstract

Structural and magnetic properties of Ga0.93Mn0.07As layers grown on (001) and (311)A GaAs substrates by molecular-bean epitaxy are investigated. The as-grown (001) and (311)A Ga0.93Mn0.07As layers exhibit the same Curie temperature (TC) of 80 K. However, upon annealing, the TC’s of the (001) and (311)A Ga0.93Mn0.07As layers are enhanced by 80 and 60 K, respectively. X-ray diffraction studies reveal that the AsGa defects cannot be removed by low-temperature annealing, and a higher concentration of AsGa defects exist in the (311)A layers than in the (100) reference layers. The less enhancement in TC by annealing for the (311)A Ga0.93Mn0.07As layer can be ascribed to the larger amount of AsGa defects in the material. [ABSTRACT FROM AUTHOR]

Abstract

Semantic-similarity measures quantify concept similarities in a given ontology. Potential applications for these measures include search, data mining, and knowledge discovery in database or decision-support systems that utilize ontologies. To date, there have not been comparisons of the different semantic-similarity approaches on a single ontology. Such a comparison can offer insight on the validity of different approaches. We compared 3 approaches to semantic similarity-metrics (which rely on expert opinion, ontologies only, and information content) with 4 metrics applied to SNOMED-CT. We found that there was poor agreement among those metrics based on information content with the ontology only metric. The metric based only on the ontology structure correlated most with expert opinion. Our results suggest that metrics based on the ontology only may be preferable to information-content-based metrics, and point to the need for more research on validating the different approaches.

Abstract

Hepatic steatosis is associated with increased insulin resistance and tricarboxylic acid (TCA) cycle flux, but decreased ketogenesis and pyruvate dehydrogenase complex (PDC) flux. This study examined whether hepatic PDC activation by inhibition of pyruvate dehydrogenase kinase 2 (PDK2) ameliorates these metabolic abnormalities. Wild-type mice fed a high-fat diet exhibited hepatic steatosis, insulin resistance, and increased levels of pyruvate, TCA cycle intermediates, and malonyl-CoA but reduced ketogenesis and PDC activity due to PDK2 induction. Hepatic PDC activation by PDK2 inhibition attenuated hepatic steatosis, improved hepatic insulin sensitivity, reduced hepatic glucose production, increased capacity for β-oxidation and ketogenesis, and decreased the capacity for lipogenesis. These results were attributed to altered enzymatic capacities and a reduction in TCA anaplerosis that limited the availability of oxaloacetate for the TCA cycle, which promoted ketogenesis. The current study reports that increasing hepatic PDC activity by inhibition of PDK2 ameliorates hepatic steatosis and insulin sensitivity by regulating TCA cycle anaplerosis and ketogenesis.
