Abstract

Much environmental enforcement in the United States has historically relied on either self-reported data or physical, resource-intensive, infrequent inspections. Advances in remote sensing and computer vision, however, have the potential to augment compliance monitoring by detecting early warning signs of noncompliance. We demonstrate a process for rapid identification of significant structural expansion using Planet's 3m/pixel satellite imagery products and focusing on Concentrated Animal Feeding Operations (CAFOs) in the US as a test case. Unpermitted building expansion has been a particular challenge with CAFOs, which pose significant health and environmental risks. Using new hand-labeled dataset of 145,053 images of 1,513 CAFOs, we combine state-of-the-art building segmentation with a likelihood-based change-point detection model to provide a robust signal of building expansion (AUC = 0.86). A major advantage of this approach is that it can work with higher cadence (daily to weekly), but lower resolution (3m/pixel), satellite imagery than previously used in similar environmental settings. It is also highly generalizable and thus provides a near real-time monitoring tool to prioritize enforcement resources in other settings where unpermitted construction poses environmental risk, e.g. zoning, habitat modification, or wetland protection.

12. Public School Teachers' Perceptions of What Promotes or Hinders Their Use of Outdoor Learning Spaces

Abstract

The creation of outdoor learning spaces (OLS) such as school gardens is increasing in frequency in elementary and secondary schools in Canada. Previous research has indicated that students benefit from learning in OLS, but little is known about teachers' perceptions of their experiences using school gardens or other OLS. Given that teacher buy-in is essential for effective use of these learning environments, better understanding of the factors which affect teachers' adoption and continued use of OLS is needed. Thus, we interviewed elementary- and high-school teachers who used school gardens in Richmond, British Columbia, Canada, to explore their perceptions of their experiences of using school gardens and other OLS. We analysed interview transcripts, guided by Roger's Diffusion of Innovations theory, and identified key factors influencing teachers' adoption of school gardens or other OLS in their pedagogy: personal values, gardening experience, environmental education, and administrative support. Teachers' continued use of various OLS was supported by students' enthusiasm for learning in those spaces. Barriers to using OLS included lack of administrative support, principals' disapproval, and lack of professional development regarding their use. Insights from this study can be applied to support teachers in the adoption of school gardens and other OLS in their teaching.

Abstract

Manduca sexta, known as the tobacco hornworm or Carolina sphinx moth, is a lepidopteran insect that is used extensively as a model system for research in insect biochemistry, physiology, neurobiology, development, and immunity. One important benefit of this species as an experimental model is its extremely large size, reaching more than 10 g in the larval stage. M. sexta larvae feed on solanaceous plants and thus must tolerate a substantial challenge from plant allelochemicals, including nicotine. We report the sequence and annotation of the M. sexta genome, and a survey of gene expression in various tissues and developmental stages. The Msex_1.0 genome assembly resulted in a total genome size of 419.4 Mbp. Repetitive sequences accounted for 25.8% of the assembled genome. The official gene set is comprised of 15,451 protein-coding genes, of which 2498 were manually curated. Extensive RNA-seq data from many tissues and developmental stages were used to improve gene models and for insights into gene expression patterns. Genome wide synteny analysis indicated a high level of macrosynteny in the Lepidoptera. Annotation and analyses were carried out for gene families involved in a wide spectrum of biological processes, including apoptosis, vacuole sorting, growth and development, structures of exoskeleton, egg shells, and muscle, vision, chemosensation, ion channels, signal transduction, neuropeptide signaling, neurotransmitter synthesis and transport, nicotine tolerance, lipid metabolism, and immunity. This genome sequence, annotation, and analysis provide an important new resource from a well-studied model insect species and will facilitate further biochemical and mechanistic experimental studies of many biological systems in insects.

42. Vitamin E and egg production in heat stressed laying hens

Abstract

The objective of the work described in this thesis was, firstly, to confirm that vitamin E has a beneficial effect on egg production in heat stressed laying hens and, secondly, to investigate the molecular basis of its effect. During exposure to a chronic period of heat (4 weeks at 32°C), egg production was improved in birds fed vitamin E-enriched diets. Distribution of a diet containing 250 mg vitamin E/kg diet before (4 weeks), during and after (8 weeks) the period of stress increased egg production by 12% (P<0.02) during and by 13% (P<0.02) in the four weeks following the stress control birds (fed 10 mg vitamin E/kg diet). Egg formation involves the mutually dependent activities of the ovary (uptake of yolk precursors and synthesis of oestrogens) and of the liver (oestrogen induced-synthesis of yolk precursors). Neither heat stress nor vitamin E were found to have direct effects on the in vivo uptake of the main yolk precursor protein, vitellogenin, by oocytes or on the circulating concentration of 17β-oestradiol. Although the amount of vitellogenin messenger RNA in hepatocytes and vitellogenin protein in the circulation were significantly depressed during heat stress in non-supplemented birds, the concentration of vitellogenin protein in hepatocytes was dramatically increased in these birds (+63%, P<0.01) compared to pre-stress values. In vitamin E-supplemented birds, this accumulation of vitellogenin protein in hepatocytes did not occur and the circulating vitellogenin concentration was higher than in non-supplemented birds. It is therefore proposed that, during heat stress, vitamin E improved the export of vitellogenin from the liver into the circulation. This effect is probably related to the known antioxidant properties of vitamin E and to its consequent protective effect on the integrity of organelle and plasma membranes which is required for proper export of proteins through the secretory pathway.

Abstract

Parents of adolescents with cancer (AWC) provide critical support throughout the cancer journey and could offer key insights into support needs. This prospective study aimed to obtain parent perspectives on peer support needs of AWC. Ten individual parents (9 mothers and 1 father) completed a survey and a semistructured interview. Four themes were identified: cancer journey challenges; emotions, reactions, and coping; personal support preferences; and AWC's support needs. Parents recognized that AWC require various support, but lacked insight into their specific peer support desires. Next step interventions should focus on peer support for AWC, while also incorporating peer support for parents. [ABSTRACT FROM AUTHOR]
