7. Future Climate and Land Use Change Impacts on River Flows in the Tapajós Basin in the Brazilian Amazon

Abstract

Land conversion and changing climate are expected to significantly alter tropical forest hydrology. We used a land surface model integrated with a river routing scheme to analyze the hydrological alterations expected in the Tapajós River basin, a large portion of the Brazilian Amazon, caused by two environmental drivers: climate and land use. The model was forced with two future climate scenarios (years 2026–2045) from the Earth System Model HadGem2-ES with moderate (+4.5 W/m2 radiative forcing value in the year 2100 with respect to preindustrial levels) and severe (+8.5 W/m2) representative atmospheric carbon dioxide pathways (Representative Concentration Pathways). We tested the sensitivity of our results to the uncertainty in future climate projections by running simulations with IPSL-CM5 (wettest scenarios) and GISS-E2 (driest scenarios). Human land use effects on vegetation were evaluated using a limited and an extreme deforestation scenario. Our analysis indicates that climate change is predicted to reduce river flows across seasons (up to 20%) and bring a considerable shift in flow seasonality toward a later onset (nearly 1.5 months) and increase in interannual variability. While land use change partially counteracts the climate-driven diminishing trend in river flows, it is expected to contribute to a further increase in interannual and intraannual variability. From a water management perspective, the overall reduction of river flows and their increased variability, combined with the shift and the shortening of the wet season, could potentially affect the productivity of the large hydropower systems planned for the region and the growing demand for agricultural and transport expansion.

Abstract

Oxidized phospholipids (oxPAPC) induce endothelial dysfunction and atherosclerosis. Here we show that oxPAPC induce a gene network regulating serine-glycine metabolism with the mitochondrial methylenetetrahydrofolate dehydrogenase/cyclohydrolase (MTHFD2) as a causal regulator using integrative network modeling and Bayesian network analysis in human aortic endothelial cells. The cluster is activated in human plaque material and by atherogenic lipoproteins isolated from plasma of patients with coronary artery disease (CAD). Single nucleotide polymorphisms (SNPs) within the MTHFD2-controlled cluster associate with CAD. The MTHFD2-controlled cluster redirects metabolism to glycine synthesis to replenish purine nucleotides. Since endothelial cells secrete purines in response to oxPAPC, the MTHFD2-controlled response maintains endothelial ATP. Accordingly, MTHFD2-dependent glycine synthesis is a prerequisite for angiogenesis. Thus, we propose that endothelial cells undergo MTHFD2-mediated reprogramming toward serine-glycine and mitochondrial one-carbon metabolism to compensate for the loss of ATP in response to oxPAPC during atherosclerosis.

23. Investigation of swelling mechanisms in self-adherent microneedles.

Abstract

Swellable microneedles (MNs) expand to mechanically interlock with wet biological tissue, offering improved adhesion and enhanced drug delivery over non-swellable counterparts. This study numerically evaluates how the material and geometric parameters of swellable MN arrays influence shape change. Using finite element simulation, MNs were subjected to unconstrained swelling, approximated via a thermal-strain analogy. Optimal MN design must support mechanical interlocking to prevent dislodgement. We observed that wet in vivo environments induce unwanted swelling-mediated curvature, hindering contact and interlocking. We quantified this bending and calibrated gel material swellability using experimental data. To counteract curling, we introduced a design approach to shift the direction of the unwanted curling and improve MN array conformability. [ABSTRACT FROM AUTHOR]

24. Pharyngeal Structure and Dysphagia in Patients with Parkinson's Disease and Related Disorders.

Abstract

Pharyngeal muscle changes occur in patients with Parkinson's disease and related disorders (PRD); however, the association between the structural alterations in the pharynx and the symptoms of dysphagia remains unclear. We assessed structural changes and contractile forces by measuring pharyngeal wall thickness and width. We aimed to define the pharyngeal measurements and determine their value as diagnostic tools for dysphagia. The pharyngeal wall thickness (PWT), pharyngeal width at rest (PWR), and shortest pharyngeal width at swallowing (PWS) were measured using lateral neck roentgenograms and videofluoroscopic swallowing study. We compared the PWR and PWT between the PRD and control groups using an independent t-test. The Kendall correlation test was performed on the radiological data of the pharynx (PWT, PWR and PWS), dysphagia scales (Penetration–Aspiration scale [PAS] and Dysphagia Outcome and Severity Scale [DOSS]), and Hoehn and Yahr scale (HY scale). The PWT was smaller and the PWR greater in the PRD than in the control group (p < 0.05). The dysphagia scales (PAS and DOSS) were correlated with the radiological data (PWT and PWS) and the HY scale (p < 0.05). The HY scale score also correlated with the PWT (p < 0.05). The optimal cutoff points of the PWT and PWR for predicting aspiration were 4.05 and 16.05 mm in the PRD group, respectively. Using the PWT, PWR and PWS, muscle atrophy and contractile strength of the pharynx can be estimated. Abstract

Identifying accurate biomarkers of cognitive decline is essential for advancing early diagnosis and prevention therapies in Alzheimer's disease. The Alzheimer's disease DREAM Challenge was designed as a computational crowdsourced project to benchmark the current state-of-the-art in predicting cognitive outcomes in Alzheimer's disease based on high dimensional, publicly available genetic and structural imaging data. This meta-analysis failed to identify a meaningful predictor developed from either data modality, suggesting that alternate approaches should be considered for prediction of cognitive performance.

Abstract

The objective of this study was to develop an inclusive fashion design (IFD) process using digital tools for educational purposes. The study consisted of three stages: development of IFD process prototypes 1 and 2 with different phases for incorporating diverse users' information (before and after the ideation phase), conducting an experiment on ten undergraduate students using two types of IFD process prototypes, and analysing the IFD process prototypes through protocol analysis, statistical analysis, and experts' assessment. The study found that the timing of incorporating user information can have a significant impact on inclusive fashion design. The user-centered Type 1 prioritised social and symbolic expressions and functional aspects over other design elements. Type 2, which focused more on aesthetics, better reflected users' expressive needs and allowed for more adaptable designs based on diverse users. 41. Dynamical Forecasts of Tropical Terrestrial Carbon Fluxes with the NASA S2S Retrospective Forecast System

Abstract

Recent advances in the ability to predict climate anomalies at sub-seasonal to seasonal (S2S) timescales allow us to explore the possibility of forecasting carbon flux anomalies. Although carbon flux forecasting is a relatively new concept, it is potentially beneficial as it can help us better understand global and regional land-atmosphere carbon feedbacks associated with climate variations and can provide guidance for future field mission design. Here we evaluate the skill of forecasted terrestrial carbon anomalies generated from meteorological anomalies produced with the NASA Global Modeling and Assimilation Office (GMAO) S2S forecast system. We focus here on three representative time periods (the most recent 2015-2016 El Nino, 2011 La Nina, and 2014 as a neutral year), with each corresponding 9-month forecast comprising four ensemble members initialized in the preceding December. The meteorological variables produced by the GMAO forecast system were bias-corrected using a climatology derived from the Modern Era Retrospective analysis for Research and Applications, version 2 (MERRA-2) before being used to drive a suite of offline simulations with the NASA Catchment-CN terrestrial biosphere model, a model that computes water-energy-carbon dynamics. Forecasts are evaluated by comparing against satellite-driven estimates of gross primary production (GPP) and inverse model estimates of net carbon flux that incorporate satellite carbon dioxide measurements. We find that the restrospectively predicted carbon fluxes in the tropics reasonably reproduce the signs and magnitudes of the observed anomalies between the 2015-2016 El Nino and the 2011 La Nina for both net flux and GPP. For instance, for the El Nino period, the magnitude of the forecasted negative GPP anomaly in the South American tropics (which undergoes anomalously warm and dry conditions) agrees with the observed GPP anomaly at leads of up to three or four months. Overall, this study demonstrates potential skill in the forecast of biospheric carbon fluxes a few months in advance, a capability that could contribute to attribution studies focusing on carbon flux variations and support innovative observation strategies in the future.
