Abstract

Tuning the lower critical solution temperature (LCST) of temperature-responsive recombinant elastin-like polypeptides has usually been achieved by designing different protein sequences, in terms of amino acid composition and length, implying tedious molecular cloning steps. In the present work, we have explored the chemoselective alkylation of methionine as an easy means to modify elastin repeat side chains and easily modulate the LCST of the polypeptides. Such a versatile synthetic method shall practically be exploited to modulate any properties of recombinant polymers.

Abstract

Cyclic polymers display unique physicochemical and biological properties. However, their development is often limited by their challenging preparation. In this work, we present a simple route to cyclic poly(alpha-peptoids) from N-alkylated-N-carboxyanhydrides (NNCA) using LiHMDS promoted ring-expansion polymerization (REP) in DMF. This new method allows the unprecedented use of lysine-like monomers in REP to design bioactive macrocycles bearing pharmaceutical potential against Clostridioides difficile, a bacterium responsible for nosocomial infections.

Abstract

Trehalose diesters exhibiting $��$,$��$-unsaturation are glycolipids which can be easily polymerized by ADMET (acyclic diene metathesis) polymerization. In this paper, enzymatic esterification was performed to selectively esterify primary hydroxyl groups of trehalose (6 and 6'-positions) with vinyl undecenoate. The vinyl ester was beforehand obtained by palladium-catalyzed transesterification of undecenoic acid with vinyl acetate. The resulting trehalose diundecenoate was homopolymerized and copolymerized with undecenyl undecenoate in order to obtain random copolymers with different compositions. The synthesis of such copolymers was confirmed by 1 H NMR spectroscopy and size exclusion chromatography (SEC). Their solid-state phase separation were investigated by DSC and X-ray scattering as function of temperature and their solution self-assembly was investigated by dynamic light scattering (DLS) in water.

Abstract

The normal interaction and the behavior under shear of mica surfaces covered by two different triblock copolymers of polylysine-polydimethysiloxane-polylysine were studied by combining the capabilities of the surface forces apparatus and the atomic force microscopy. At low pH values these copolymers spontaneously adsorb on the negatively charged mica surfaces from aqueous solutions as a consequence of the positive charge of the polylysine moieties. The morphology of the adsorbed layer is determined by the molecular structure of the particular copolymer investigated. This morphology plays a fundamental role on the behavior of the adsorbed layers under shear and compression. While nonadhesive smooth layers oppose an extremely small resistance to sliding, the presence of asperities even at the nanometric scale originates a frictional resistance to the motion. The behavior of uniform nonadhesive nanorough surfaces under shear can be quantitatively understood in terms of a simple multistable thermally activated junction model. The electric charge of the adsorbed copolymer molecules and hence the adhesion energy between the coated surfaces can be modified by varying the pH of the surrounding media. In the presence of an adhesive interaction between the surfaces the behavior under shear is strongly modified. Abstract

International audience; Using "click chemistry" as an easy and versatile synthetic strategy to combine hyaluronan and polyglutamate blocks, we have prepared nanovesicles (polymersomes) that present a controlled size, excellent colloidal stability, and a high loading capacity for hydrophilic and hydrophobic drugs. The unique feature of our concept is the use of hyaluronan, a polysaccharide with known capacity for targeting cancer-related protein receptors, as the hydrophilic portion of a block copolymer system. The cytotoxicity and internalization mechanism of doxorubicin-loaded polymersomes have been evaluated in C6 glioma tumor cell lines. The dual purpose served by hyaluronan, as both a hydrophilic block critical to vesicle formation and as a binding agent for biological targets, breaks new ground in terms of multifunctional nanomaterial design for drug delivery.
