Monoclonal antibodies have been administered to kidney transplant recipients (KTRs) with a poor or non-responder status to SARS-CoV-2 vaccination. The cellular response to SARS-CoV-2 has been poorly studied in this context. We assessed the T cell response to SARS-CoV-2 in 97 patients on the day of the injection of tixagevimab/cilgavimab using an IFNγ enzyme-linked immunospot assay (ELISPOT). Among the 97 patients, 34 (35%) developed COVID-19 before the injection. Twenty-nine (85.3%) had an ELISPOT compatible with a SARS-CoV-2 infection. There was no difference between KTRs under belatacept or tacrolimus treatment. Sixty-three patients (64.9%) had no known COVID-19 prior to the ELISPOT, but nine (14.3%) had a positive ELISPOT. In 21 KTRs with a positive ELISPOT who received a booster dose of a bivalent mRNA vaccine, median antibody titers and spike-reactive T cells increased significantly in patients under tacrolimus but not belatacept. Our study emphasizes the potential usefulness of the exploration of immune cellular response to SARS-CoV-2 by ELISPOT. In KTRs with a positive ELISPOT and under CNI therapy, a booster dose of mRNA vaccine seems effective in inducing an immune response to SARS-CoV-2.

Background: Kidney transplant recipients (KTRs) are likely to develop severe COVID-19 and are less well-protected by vaccines than immunocompetent subjects. Thus, the use of neutralizing anti–SARS-CoV-2 monoclonal antibodies (mAbs) to confer a passive immunity appears attractive in KTRs. Methods: This retrospective monocentric cohort study was conducted between 1 January 2022 and 30 September 2022. All KTRs with a weak antibody response one month after three doses of mRNA vaccine (anti spike IgG < 264 (BAU/mL)) have received tixagevimab-cilgavimab in pre-exposure (group 1), post-exposure (group 2) or no specific treatment (group 3). We compared COVID-19 symptomatic hospitalizations, including intensive care unit hospitalizations, oxygen therapy, and death, between the three groups. Results: A total of 418 KTRs had SARS-CoV-2 infection in 2022. During the study period, we included 112 KTRs in group 1, 40 KTRs in group 2, and 27 KTRs in group 3. The occurrence of intensive care unit hospitalization, oxygen therapy, and COVID-19 death was significantly increased in group 3 compared to group 1 or 2. In group 3, 5 KTRs (18.5%) were admitted to the intensive care unit, 7 KTRs (25.9%) needed oxygen therapy, and 3 KTRs (11.1%) died. Patients who received tixagevimab-cilgavimab pre- or post-exposure had similar outcomes. Conclusions: This retrospective real-life study supports the relative effectiveness of tixagevimab-cilgavimab on COVID-19 infection caused by Omicron, used as a pre- or post-exposure therapy. The continued evolution of Omicron variants has made tixagevimab-cilgavimab ineffective and reinforces the need for new therapeutic monoclonal antibodies for COVID-19 active on new variants.

Control of CNS pathogens by CD8 T cells is key to avoid fatal neuroinflammation. Yet, the modalities of MHC I presentation in the brain are poorly understood. Here, we analyze the antigen presentation mechanisms underlying CD8 T cell-mediated control of the Toxoplasma gondii parasite in the CNS. We show that MHC I presentation of an efficiently processed model antigen (GRA6-OVA), even when not expressed in the bradyzoite stage, reduces cyst burden and dampens encephalitis in C57BL/6 mice. Antigen presentation assays with infected primary neurons reveal a correlation between lower MHC I presentation of tachyzoite antigens by neurons and poor parasite control in vivo. Using conditional MHC I-deficient mice, we find that neuronal MHC I presentation is required for robust restriction of T. gondii in the CNS during chronic phase, showing the importance of MHC I presentation by CNS neurons in the control of a prevalent brain pathogen.

Resumo: Para contribuir para o debate sobre a chegada dos jovens ao ensino médio, esta pesquisa analisou um grupo de estudantes de um distrito que, após a conclusão do ensino fundamental na escola local, passou a estudar na sede do município. Para compreender como percebiam seus processos de transição para o ensino médio, a pesquisa de campo foi realizada com base em entrevistas reflexivas com oito jovens e o material gerado foi analisado por meio de perfis de configuração com informações organizadas individualmente. Constatou-se que a transição para o ensino médio, especialmente para jovens do interior, é um momento no qual se amplia a rede de sociabilidade e surgem complexos desafios. Espera-se que os resultados da pesquisa contribuam para as discussões sobre os sentidos do ensino médio para jovens das camadas populares, sobretudo associados ao que planejam para o futuro.

Embedded RF contacts are integrated within the ITER ICRH launcher to allow assembling, sliding and to lower the thermo-mechanical stress. They have to withstand a peak RF current up to 2.5 kA at 55 MHz in steady-state conditions, in the vacuum environment of themachine.The contacts have to sustain a temperature up to 250{\textdegree}Cduring several days in baking operations and have to be reliable during the whole life of the launcher without degradation. The RF contacts are critical components for the launcher performance and intensive R&D is therefore required, since no RF contactshave so far been qualified at these specifications. In order to test and validate the anticipated RF contacts in operational conditions, CEA has prepared a test platform consisting of a steady-state vacuum pumped RF resonator. In collaboration with ITER Organization and the CYCLE consortium (CYclotronCLuster for Europe), an R&D program has been conducted to develop RF contacts that meet the ITER ICRH launcher specifications. A design proposed by CYCLE consortium, using brazed lamellas supported by a spring to improve thermal exchange efficiency while guaranteeing high contact force, was tested successfully in the T-resonator up to 1.7 kA during 1200 s, but failed for larger current values due to a degradation of the contacts. Background. Kidney transplantation and dialysis are two major risk factors for severe forms of coronavirus disease 2019 (COVID-19). The dynamics of the immune response to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in this population remain largely unknown. Methods. We report here the analysis of anti–SARS-CoV-2 antibody– and T cell–mediated immune responses in 26 kidney transplant recipients (KTRs) and 11 dialyzed patients (DPs) who recovered from COVID-19. Results. After a mean time of 83 ± 26 d post–symptom onset for KTRs and 97 ± 31 d for DPs, 20 KTRs (76.9%) and 10 DPs (90.9%) displayed anti-S1 immunoglobulin G SARS-CoV-2 antibodies (P = 0.34), at similar titers in both groups. SARS-CoV-2–specific interferon-γ–producing T cells were evidenced in 26 KTRs (100%) and 10 DPs (90.9%). Total numbers of SARS-CoV-2–reactive T cells were high and not statistically different between the 2 groups. No correlation between the severity of the disease and the number of reactive T cells was found in KTRs. In 5 KTRs, also evaluated 10 mo after COVID-19, weak or absent antibody response was observed, whereas specific memory T-cell response was detected in all cases. Conclusion. T-cell response persisted up to 3 mo post–symptom onset, even in KTRs in whom full immunosuppressive regimen was reinstated at recovery, and seems to be present up to 10 mo after infection. Our findings have implications in the understanding of the natural course of the disease in transplant patients and DPs.

The objective of this work is to propose a standard classification of seismic signals generated by gravitational processes and detected at close distances ( km). We review the studies where seismic instruments have been installed on unstable slopes and discuss the choice of the seismic instruments and the network geometries. Seismic observations acquired at 13 unstable slopes are analyzed in order to construct the proposed typology. The selected slopes are affected by various landslide types (slide, fall, topple and flow) triggered in various material (from unconsolidated soils to consolidated rocks). We investigate high-frequency bands (>1 Hz) where most of the seismic energy is recorded at the 1 km sensor to source distances. Several signal properties (duration, spectral content and spectrogram shape) are used to describe the sources. We observe that similar gravitational processes generate similar signals at different slopes. Three main classes can be differentiated mainly from the length of the signals, the number of peaks and the duration of the autocorrelation. The classes are the “slopequake” class, which corresponds to sources potentially occurring within the landslide body; the “rockfall” class, which corresponds to signals generated by rock block impacts; and the “granular flow” class, which corresponds to signals generated by wet or dry debris/rock flows. Subclasses are further proposed to differentiate specific signal properties (frequency content, resonance, precursory signal). The signal properties of each class and subclass are described and several signals of the same class recorded at different slopes are presented. Their potential origins are discussed. The typology aims to serve as a standard for further comparisons of the endogenous microseismicity recorded on landslides.

Entering the high school means, for many students, an important moment for the establishment of their identity. This article is the result of a study that sought to understand how young people from a small district that started to travel daily to the municipal center to study at the High School realized this transition process. Eight young people were heard, and their statements were analyzed using configuration profiles. It was found that, for them, as well as for others in the same situation, entering the High School is a delicate moment of transition that, besides the change of school, requires changes in their routines and interfere in their relations with the space and their sociability ties.

Documenting landslide activity over long periods and monitoring standards (sensors, acquisition rates, quality-control) is critical for understanding the landslide forcing factors, develop process-based models, identify the effect of climate change on their behavior, and ultimately define warning thresholds.The French Landslide Observatory (Observatoire Multi-Disciplinaire des Instabilités de Versants) OMIV is the service of the French Research Institute (CNRS) in charge of deploying, acquiring, exploiting and disseminating multi-parametric sensor data over several large landslides in France. OMIV has developed, since more than 15 years, standards in terms of sensor types, using both high-grade and low-cost sensing in order to construct reference and spatially dense monitoring time series. The service provides open access to records of landslide kinematics, landslide micro-seismicity, landslide hydro-meteorology and landslide hydro-geophysics. Combined, these four categories of observations are unique worldwide for long-term landslide observations. OMIV is currently supervisizing the acquisition and dissemination of sensor data on 8 permanent unstable slopes (Avignonet/Harmallière, La Clapière, Séchilienne, Super-Sauze/La Valette, St-Eynard, Pégairolles, Vence, Villerville) and on unstable slopes currently experiencing gravitational crises (La Clape, Viella, Marie-sur-Tinée, Aiguilles). The service is organized around the dissemination of qualified data (in international reference file format) and products for 5 categories of observation (Geodesy, Seismology, Hydrology, Meteorology, Hydrogeophysics). For each categories of observation, specific FAIR data repository and access portals have been developed and automated processing methods have been proposed to meet the needs of the landslide research community. The products being generated are time series of GNSS and total station positions, catalogue of endogeneous landslide micro-seismicity, resistivity tomography datasets, and hydro-meterological parameters).OMIV provides consistent and harmonized landslide monitoring data in order to identify the physical processes that control the landslide dynamics, both for slopes affected by slow-moving slides and cliffs affected by rockfalls, use these datasets to develop and validate landslide deformation/propagation models, extract (from the long-term observations) the patterns that may characterize changes in the landslide dynamics (annual, seasonal, event) and propose possible forerunners. The OMIV observations aim at contributing at identifying the key controlling parameters of different landslide types (e.g. soft/hard rock, cohesion/friction, slip/fracture, localized/diffuse damage) and at monitoring their evolution in time and space (deceleration or acceleration according to the triggering factors, sliding- flowing transition).The objectives are to present the OMIV datasets, sensing standards and automated processing methods that has been developed, both for the science community and for operational partners in charge of landslide risk management (ONF-RTM, BRGM, CEREMA), for some of the monitored landslides. The objectives are also to present the future directions of the service with a focus on the modelling of the landslide processes using both process-based and machine learning approaches.

- Abstract
Envirosciences is developing an integrated multi-parameter low-cost monitoring station encompassing the geohazard and geophysical community needs. It consists in integrating co-located sensors ((meteorology, seismology, GNSS) on the same data acquisition card with modular configurations compatible with the EPOS - European Plate Observing System- specifications (sensor type, data sampling, noise level, data and metadata format). On-line data dissemination sand on-demand processing services are further being developed in order to propose advanced products such as GNSS position time series, advanced hydro-meteorological variables and seismic/micro-seismic catalogues.A dense network of 45 stations is currently being implemented in the Western and Central Pyrenees (South France). The network consists of a seismological, meteorological and geodetic (GNSS) antennas. The measurement network is semi-permanent with at least ten years of observation. It will allow to create catalogues of hydro-geomorphological and tectonic events, to document Pyrenean tectonic uplift, and to better constrain local micro-meteorology from the valley bottoms to the summit ridges (by combining co-localised measurements of classic meteorological parameters - wind, temperature, pressure, humidity, precipitation - and tomography of vertical water vapor profiles from GNSS delays).The objective of the presentation is to present the technological development of the station which combines several types of sensors (2 Hz seismometers, dual-frequency GNSS receivers and meteorological stations), a high-frequency geophysical digitization module, a communication module (WiFi or4G) and a power supply module (by solar energy or 220V). Softwares to control the station have been created, as well as software to supervise the database and codes to interpret the measurements.We will further present the processing worklows and the time series of data acquired since November 2022 on 8 measurement stations already deployed in the Pyrenees. By the end of 2023, the full network of 45 autonomous real-time stations will be deployed with inter-station distances of around 5 km.

Introdução: Hérnia umbilical é uma protuberância anormal na região do umbigo devido a um defeito no fechamento da cicatriz umbilical ou por fatores adquiridos durante a vida. O tratamento cirúrgico da hérnia umbilical envolve uma vasta gama de opções existentes. Revisão da literatura: Os números relatados foram encontrados nos 30 artigos lidos nas plataformas PubMed, Medline e Lilacs dos últimos 6 anos. No total, 1785 pacientes foram submetidos ao reparo com malha, ocorrendo recidiva em 72 casos (4,03%). Já o reparo primário simples foi realizado em um total de 2739 pacientes analisados, sendo a recidiva presente em 196 casos (7,16%). O uso laparoscópico no reparo de hérnias umbilicais compreendeu um total de 337 cirurgias e 10 recorrências (2,97%). A prótese de duas camadas foi realizada em 1720 pacientes com 62 recorrências (3,6%). Discussão: A técnica mais indicada é o reparo com malha em controvérsia com o de sutura que apresentou maior número de recidivas, a posição da malha não apresentou diferença nesse quesito. Em cirróticos, a Button Hole se mostrou bastante eficaz. Conclusão: Portanto, o reparo com malha é a técnica mais recomendada e com baixo índice de recorrências.

International audience; ObjectivesLupus nephritis (LN) is a frequent complication of systemic lupus erythematosus (SLE). Severe (proliferative) forms of LN are treated with induction immunosuppressive therapy (IST), followed by maintenance IST, to target remission and avoid relapses. The optimal duration of maintenance IST is unknown. The WIN-Lupus trial tested whether IST discontinuation after 2‒3 years was non-inferior to IST continuation for two more years in proliferative LN.MethodsWIN-Lupus was an investigator-initiated multicentre randomised controlled trial. Patients receiving maintenance IST with azathioprine or mycophenolate mofetil for 2–3 years, and hydroxychloroquine, were randomised (1:1) into two groups: (1) IST continuation and (2) IST discontinuation. The primary endpoint was the relapse rate of proliferative LN at 24 months. Main secondary endpoints were the rate of severe SLE flares, survival without renal relapse or severe flare, adverse events.ResultsBetween 2011 and 2016, 96 patients (out of 200 planned) were randomised in WIN-Lupus: IST continuation group (n=48), IST discontinuation group (n=48). Relapse of proliferative LN occurred in 5/40 (12.5%) patients with IST continuation and in 12/44 (27.3%) patients with IST discontinuation (difference 14.8% (95% CI −1.9 to 31.5)). Non-inferiority was not demonstrated for relapse rate; time to relapse did not differ between the groups. Severe SLE flares (renal or extrarenal) were less frequent in patients with IST continuation (5/40 vs 14/44 patients; p=0.035). Adverse events did not differ between the groups.ConclusionsNon-inferiority of maintenance IST discontinuation after 2‒3 years was not demonstrated for renal relapse. IST discontinuation was associated with a higher risk of severe SLE flares.Trial registration numberNCT01284725.

The experience resulted in this article is derived from the creation of the network Professores transformadores (Transformative Teachers), a project based on content exchange, dialog and development among teachers, educators, managers, and undergraduate students and all of those interested in a more reflective and committed education. Created in 2015, the network aims to bring together teachers who are the authors of innovative and transformative actions, calling attention to the importance of their practices in the students’ development process as autonomous, critical and creative citizens. With reinforcement strategies to leadership and teaching appreciation, the network intends to contribute to reframe the professional practice of those involved by reflecting on possibilities of overcoming adversity and some of the Brazilian Education dilemmas. The proposals operate in two ways. Digitally, members have access to content collectively created and published on a website, on a page and among a group on a social network. Locally, in Ouro Preto and Mariana towns, in the state of Minas Gerais, Brazil, the network works by mobilizing teachers with activities such as lectures, film exhibitions and guided studies. The results so far indicate receptivity to the proposal, with teachers articulating, empowering themselves and building exchange relationships with colleagues across the country.
