Abstract

Unmanned aircraft systems (UAS) – also called unmanned aerial vehicles (UAVs) or drones – are an emerging tool that may provide a safer, more cost-effective, and quieter alternative to traditional research methods. We review examples where UAS have been used to document wildlife abundance, behavior, and habitat, and illustrate the strengths and weaknesses of this technology with two case studies. We summarize research on behavioral responses of wildlife to UAS, and discuss the need to understand how recreational and commercial applications of this technology could disturb certain species. Currently, the widespread implementation of UAS by scientists is limited by flight range, regulatory frameworks, and a lack of validation. UAS are most effective when used to examine smaller areas close to their launch sites, whereas manned aircraft are recommended for surveying greater distances. The growing demand for UAS in research and industry is driving rapid regulatory and technological progress, which in turn will make them more accessible and effective as analytical tools.

Abstract

In regulated rivers, managers must evaluate competing flow release scenarios that attempt to balance both human and natural needs. Meeting these natural flow needs is complex due to the myriad of interacting physical and hydrological factors that affect ecosystems. Tools that synthesize the voluminous scientific data and models on these factors will facilitate management of these systems. Here, we present the Riverine Environmental Flow Decision Support System (REFDSS), a tool that enables evaluation of competing flow scenarios and other variables on instream habitat. We developed a REFDSS for the Upper Delaware River, USA, a system that is regulated by three headwater reservoirs. This version of the REFDSS has the ability to integrate any set of spatially explicit data and synthesizes modeled discharge for three competing management scenarios, flow-specific 2-D hydrodynamic modeled estimates of local hydrologic conditions (e.g., depth, velocity, shear stress, etc.) at a fine pixel-scale (1 m2), and habitat suitability criteria (HSC) for a variety of taxa. It contains all individual model outputs, computationally integrates these data, and outputs the amount of potentially available habitat for a suite of species of interest under each flow release scenario. Users have the flexibility to change the time period of interest and vary the HSC. The REFDSS was developed to enable side-by-side evaluation of different flow management scenarios and their effects on potential habitat availability, allowing managers to make informed decisions on the best flow scenarios. An exercise comparing two alternative flow scenarios to a baseline scenario for several key species is presented. The Upper Delaware REFDSS was robust to minor changes in HSC (± 10 %). The general REFDSS platform was developed as a user-friendly Windows desktop application that was designed to include other potential parameters of interest (e.g., temperature) and for transferability to other riverine systems.

Abstract

Escherichia coli O157:H7 and other Shiga toxin (Stx)-producing E. coli (STEC) bacteria are not enteroinvasive but can cause hemorrhagic colitis. In some STEC-infected individuals, a life-threatening sequela of infection called the hemolytic uremic syndrome may develop that can lead to kidney failure. This syndrome is linked to the production of Stx by the infecting organism. For Stx to reach the kidney, the toxin must first penetrate the colonic epithelial barrier. However, the Stx receptor, globotriaosylceramide (Gb3), has been thought to be absent from human intestinal epithelial cells. Thus, the mechanisms by which the toxin associates with and traverses through the intestine en route to the kidneys have been puzzling aspects of STEC pathogenesis. In this study, we initially determined that both types of Stx made by STEC, Stx1 and Stx2, do in fact bind to colonic epithelia in fresh tissue sections and to a colonic epithelial cell line (HCT-8). We also discovered that globotetraosylceramide (Gb4), a lower-affinity toxin receptor derived from Gb3, is readily detectable on the surfaces of human colonic tissue sections and HCT-8 cells. Furthermore, we found that Gb3 is present on a fraction of HCT-8 cells, where it presumably functions to bind and internalize Stx1 and Stx2. In addition, we established by quantitative real-time PCR (qRT-PCR) that both fresh colonic epithelial sections and HCT-8 cells express Gb3 synthase mRNA. Taken together, our data suggest that Gb3 may be present in small quantities in human colonic epithelia, where it may compete for Stx binding with the more abundantly expressed glycosphingolipid Gb4.

Abstract

The yeast ABC (ATP-binding cassette protein) multidrug transporter Pdr5p transports a broad spectrum of xenobiotic compounds, including antifungal and antitumor agents. Previously, we demonstrated that substrate size is an important factor in substrate-transporter interaction and that Pdr5p has at least three substrate-binding sites. In this study, we use a combination of whole cell transport assays and photoaffinity labeling of Pdr5p with [(125)I]iodoarylazidoprazosin in purified plasma membrane vesicles to study the behavior of two series of novel substrates: trityl (triphenylmethyl) and carbazole derivatives. The results indicate that site 2, defined initially by tritylimidazole efflux, requires at least a single hydrogen bond acceptor group (electron pair donor). In contrast, complete inhibition of rhodamine 6G efflux and [(125)I]iodoarylazidoprazosin binding at site 1 requires substrates with three electronegative groups. Carbazole and trityl substrates with two groups show saturating, incomplete inhibition at this site. This type of inhibition is frequently observed in bacterial multidrug-binding proteins that use a pocket with multiple binding sites. The presence of multiple sites with different requirements for substrate-Pdr5p interaction may explain the broad specificity of xenobiotic compounds transported by this protein.
