Three-dimensional solid modeling of an OBS anti-trawl sinking coupling frame was carried out using the ANSYS, and modal simulation analysis of the overall structure of the anti-trawl sinking coupling frame was made through the ANSYS Workbench. The natural frequencies and mode shape contour diagrams of the first six modes were extracted. The displacement response spectrum was analyzed, and the equivalent stress and directional displacement contour diagrams were obtained. Simulation results show that the natural frequency of the anti-trawl sinking coupling frame evades the working frequency band of the OBS, which verifies the effectiveness of the anti-trawl structure design.

Structural stabilization for a membrane structure under high-frequency vibration is still a recognized problem. In nature, honeybee wings with non-uniform material properties demonstrate excellent anti-interference ability. However, the correlation between the structural stabilization and mechanical properties of insect wings has not been completely verified. Here we demonstrate that the sclerotization diversity partially distinguishes the stiffness inhomogeneity of the wing structure. Furthermore, a wing cross-section model with diversity in elastic modulus is constructed to analyze the effect of stiffness distribution on stress optimization during flight. Our results demonstrate that the heterogeneous stiffness promotes the stress distribution and structural stabilization of the wing during flight, which may inspire more optimal designs for anisotropic high-strength membrane structures.

The ladybird beetle (Coccinella septempunctata) is known for swift deployment of its elytra, an action that requires considerable power. However, actuation by thoracic muscles alone may be insufficient to deploy elytra at high speed because the maximum mechanical power that elytral muscles can produce is only 70% of that required for initiation of deployment. Nevertheless, the elytra open rapidly, within 3 ms in the initial phase, at a maximum angular velocity of 66.49±21.29 rad s−1, rivaling the strike velocity of ant lion (Myrmeleon crudelis) mandibles (65±21 rad s−1). Here, we hypothesize that elytra coupling may function as an energy storage mechanism that facilitates rapid opening by releasing elastic strain energy upon deployment. To test this hypothesis and better understand the biomechanics of elytra deployment, we combined micro-computed tomography and scanning electron microscopy to examine the microstructure of the coupling of paired elytra. We found that two rows of setae on the internal edges of the elytra coupling structure undergo elastic deformation when the elytra are locked together. Kinematics observations and mathematical modeling suggest that the elastic potential energy stored in the compressed setae generates 40% of the power required for deployment of elytra. Our findings broaden insights into how ladybirds actuate elytra opening by a strategy of using both muscles and elastic microstructures, and demonstrate a distributed pattern of actuation that adapts to geometrical constraints in elytra locking.

When we flip a coin, we expect a 50 percent chance of it landing on heads. But what happens when an insect jumps? In this study, we report the self-righting strategy of the flea beetle, Altica cirsicola, which lives in a dense maze of leaves. How can the flea beetle coordinate landing on surfaces of such a range of orientation and position? In this experimental study, we film the take-off, flight, and landing of flea beetles on a configurable angled platform. Dead flea beetles tossed through the air have a 46% chance of landing on their legs, similar to a fair coin. However, live beetles land successfully 85% of the time, suggesting the importance of active self-righting strategies to accommodate the different orientations of the landing platform. We observe that beetles may extend their wings to slow down and reduce impact as well as perform numerous rotations. These behaviors may inspire jumping robots that can land softly.

The flea beetle, Altica cirsicola, escapes predators by jumping and landing in a dense maze of leaves. How do they land on such varied surfaces? In this experimental study, we filmed the take-off, flight, and landing of flea beetles on a configurable angled platform. We report three in-flight behaviors: winged, wingless, and an intermediate winged mode. These modes significantly affected take-off speed, acceleration, and the duration that wings were deployed. When wings were closed, flea beetles rolled or pitched up to five times in the air. This work may help to understand how insects can jump and right themselves onto variable surfaces.

According to the requirements of pressure resistance and sealing performance detection of deep-sea instruments and equipment under full sea pressure, a 120MPa deep-sea test chamber for ultra-high pressure environment simulation is designed in this paper. The structural dimensions of the test chamber were designed and calculated, including the cabin wall thickness, the cabin bottom head thickness, the sealing top cover, and the locking nut. The strength of the cylinder structure and the sealing cover of the test chamber were verified and analyzed through finite element analysis, which validates the reliability of the test chamber structure. The 120MPa deep-sea test chamber for ultra-high pressure environment simulation designed in this paper can provide important technical support for testing the pressure resistance and the sealing performance of deep-sea instruments and equipment.

Nitrogen-fixing root nodules are formed by symbiotic association of legume hosts with rhizobia in nitrogen-deprived soils. Successful symbiosis is regulated by signals from both legume hosts and their rhizobial partners. HmuS is a heme degrading factor widely distributed in bacteria, but little is known about the role of rhizobial hmuS in symbiosis with legumes. Here, we found that inactivation of hmuS

A drogue is used to stabilise and straighten seismic arrays so that seismic waves can be well-received. To embed the effect of a cone-shaped drogue into the numerical modelling of the deep-towed seismic survey system, one surrogate model that maps the relationship between the hydrodynamic characteristics of the drogue and towing conditions was obtained based on data-driven simulations. The sample data were obtained by co-simulation of the commercial software RecurDyn and Particleworks, and the modelling parameters were verified by physical experiments. According to the Morison formula, the rotational angle, angular velocity, angular acceleration, towing speed, and towing acceleration of the drogue were selected as the design variables and drag forces and aligning torque were selected as the research objectives. The sample data of more than 8500 sets were obtained from virtual manoeuvres. Subsequently, both polynomial and neural network regression algorithms were used to study these data. Finally, analysis results show that the surrogate model obtained by machine learning has good performance in predicting research objectives. The results also reveal that the neural network regression algorithm is superior to the polynomial regression algorithm, its largest error of mean square is less than 0.8 (N2/N2 mm2), and its R-squared is close to 1.

NAD+ supplementation has significant benefits in compromised settings, acting largely through improved mitochondrial function and DNA repair. Elevating NAD+ to physiological levels has been shown to improve the function of some adult stem cells, with implications that these changes will lead to sustained improvement of the tissue or system. Here, we examined the effect of elevating NAD+ levels in models with reduced hematopoietic stem cell (HSC) potential, ATM-deficient and aged WT mice, and showed that supplementation of nicotinamide riboside (NR), a NAD+ precursor, improved lymphoid lineage potential during supplementation. In aged mice, this improved lymphoid potential was maintained in competitive transplants and was associated with transcriptional repression of myeloid gene signatures in stem and lineage-committed progenitor cells after NR treatment. However, the altered transcriptional priming of the stem cells toward lymphoid lineages was not sustained in the aged mice after NR removal. These data characterize significant alterations to the lineage potential of functionally compromised HSCs after short-term exposure to NR treatment.

We recently reported that some cancers induce accumulation of bone marrow (BM) B-cell precursors in the spleen to convert them into metastasis-promoting, immunosuppressive B cells. Here, using various murine tumor models and samples from humans with breast and ovarian cancers, we provide evidence that cancer cells also coopt differentiation of the extra nodal B-cell precursors to generate macrophages (termed B-MF). We link the trans-differentiation to a small subset of CSF1R+ Pax5Low cells within BM pre-B and immature B cells and cancer-secreted M-CSF that downregulates Pax5 via CSF1R signaling. Thus, cancer generates tumor-associated macrophages (TAM) from B-cell precursors in addition to their primary source, monocytes. Based on their differences from monocyte-derived TAM, such as a superior ability to induce FoxP3+ Tregs, suppress proliferation of T cells and more efficiently phagocytize apoptotic cells, we propose that cancer generates B-MF to mediate cancer escape.

A method of obtaining bottom backscattering strength by employing an omnidirectional projector and omnidirectional hydrophone is proposed. The backscattering strength is extracted from monostatic backscattering data. The method was adopted in an experiment conducted in the South Yellow Sea of China. The seafloor surface was relatively smooth and covered by a small quantity of shell fragments, as observed through a digital camera system. Sampling data showed that the main component of the sediment at this experimental site was fine sand. In this paper, we detail the calculation method. Preliminary results of backscattering strength as a function of grazing angle (20°–70°) in the frequency range of 6–24 kHz are presented. The measured backscattering strength increased with the grazing angle and changed more rapidly at large grazing angles (60°–70°). A comparison of the data at different frequencies reveals that the measured backscattering strength substantially rises with the increase of acoustic frequency. A fitting curve of Lambert’s law against the measured data shows that the backscattering strength deviates from Lambert’s law at large grazing angles.

Microstructure evolution and hardening effect of pure tungsten and W–1.5%ZrO2 alloy under carbon ion irradiation are investigated by using transmission electron microscopy and nano-indentation. Carbon ion irradiation is performed at 700 °C with irradiation damages ranging from 0.25 dpa to 2.0 dpa. The results show that the irradiation defect clusters are mainly in the form of dislocation loop. The size and density of dislocation loops increase with irradiation damages intensifying. The W–1.5%ZrO2 alloy has a smaller dislocation loop size than that of pure tungsten. It is proposed that the phase boundaries have the ability to absorb and annihilate defects and the addition of ZrO2 phase improves the sink strength for irradiation defects. It is confirmed that the W–1.5%ZrO2 alloy shows a smaller change in hardness than the pure tungsten after being irradiated. From the above results, we conclude that the addition of ZrO2 into tungsten can significantly reduce the accumulation of irradiated defects and improve the irradiation resistance behaviors of the tungsten materials.

A variety of models have been used in mating bioassays of insects to assess the contribution of chemical and visual signals to mate location and mate selection. Although the use of such ‘dummies’ has had varying degrees of success, some insect species refuse to accept simplistic models. In the present study, we developed a 3D-printed model to explore whether more realistic models will be more successful than simplistic models in mating assays of difficult to manipulate species such as the flea beetle Altica fragariae. We ran five experiments to test (1) whether males could discriminate between males and females solely based on differences in cuticular hydrocarbons (CHCs), (2) whether males use shape or (3) color to choose mates, and (4) whether males can discriminate between 3D-printed models and freshly killed beetles either with or (5) without legs and antennae. The results of these experiments confirmed that male A. fragariae preferred models coated with CHCs of females over that of male CHCs, providing strong support for the role of CHCs in mate choice in Altica. We also showed that males use both shape and color in mate selection, and that males are capable of discriminating between the models and real beetle specimens. Together, the results indicate that 3D-printed models can provide a feasible and cost-effective method for mating studies of insects.

The efficacy of susceptible variants derived from genome-wide association studies (GWAs) optimizing discriminatory accuracy of colorectal cancer (CRC) in Chinese remains unclear. In the present validation study, we assessed 75 recently identified variants from GWAs. A risk predictive model combining 19 variants using the least absolute shrinkage and selection operator (LASSO) statistics offered certain clinical advantages. This model demonstrated an area under the receiver operating characteristic (AUC) of 0.61 during training analysis and yielded robust AUCs from 0.59 to 0.61 during validation analysis in three independent centers. The individuals carrying the highest quartile of risk score revealed over 2-fold risks of CRC (ranging from 2.12 to 2.90) compared with those who presented the lowest quartile of risk score. This genetic model offered the possibility of partitioning risk within the average risk population, which might serve as a first step toward developing individualized CRC prevention strategies in China.

Acoustic bottom scattering is the main source of reverberation background in target detection and recognition near the seafloor. Accurate estimation of bottom scattering strength is necessary for prediction of sonar performance in the presence of reverberation in shallow-water environments. A measurement of bottom backscattering strength at midfrequency in a shallow-water environment of the South Yellow Sea of China was carried out in April 2016, where the water depth was 39 m. Finally, the fitted result of a scattering model to the measured data suggests that seafloor roughness scattering is larger than that from sediment volume heterogeneity in the measurement frequency band.

Cellular senescence is a well-orchestrated programmed process involved in age-related pathologies, tumor suppression and embryonic development. TGF-β/Smad is one of the predominant pathways that regulate damage-induced and developmentally programmed senescence. Here we show that canonical TGF-β signaling promotes senescence via miR-29-induced loss of H4K20me3. Mechanistically, oxidative stress triggers TGF-β signaling. Activated TGF-β signaling gives rise to acute accumulation of miR-29a and miR-29c, both of which directly suppress their novel target, Suv4-20h, thus reducing H4K20me3 abundance in a Smad-dependent manner, which compromises DNA damage repair and genome maintenance. Loss of H4K20me3 mediated by the senescent TGF-β/miR-29 pathway contributes to cardiac aging in vivo. Disruption of TGF-β signaling restores H4K20me3 and improves cardiac function in aged mice. Our study highlights the sequential mechanisms underlying the regulation of senescence, from senescence-inducing triggers to activation of responsive signaling followed by specific epigenetic alterations, shedding light on potential therapeutic interventions in cardiac aging., Cellular senescence is associated with epigenetic remodeling. Here, the authors report that TGF-β signaling promotes miR-29 mediated loss of H4K20me3 thus accelerating senescence.

An inversion method using backscattering strength is used to obtain bottom parameters. A backscattering model based on the effective density fluid approximation was established as the forward model. The objective function used to scale the mismatch between the model predictions and the measured data (obtained by employing two monostatic transducers with narrow directivity) was established on the assumption that the data errors follow a Gaussian distribution. The maximum a posteriori estimations of inversion parameters were obtained by using a two-step hybrid optimization algorithm combining the differential evolution algorithm and the particle swarm optimization algorithm. The uncertainty and the correlation of inversion parameters were presented using marginal probability distributions and a covariance matrix through Bayesian inversion. Using this method, we can acquire not only geoacoustic parameters (including sound speed and attenuation) but also partial physical parameters of marine sediments and statistical character parameters of seafloor roughness and sediment heterogeneity. Finally, the efficiency of this inversion method was verified through comparison between inversion results and sampling data from sand sediment at the bottom of a water tank.

In this paper, a thermal insulation structure with silica aerogel felt as filler material was designed for the requirements of deep-sea fluid thermal insulation sampling technology for Jiaolong human occupied vehicle. Simulation analysis of thermal insulation performance was carried out and an experimental prototype was developed for the thermal insulation structure. Experimental study on thermal insulation performance was conducted with the variation characteristics of the operation environment for Jiaolong human occupied vehicle being taken into account. Results show that the silica aerogel felt with a thickness of 30 mm filled in the radial space between the inner and outer cylinders can achieve the expected thermal insulation effect during the diving-sampling-transferring process, with maximum temperature rise of 8.5 °C, and can meet the requirements of deep-sea fluid thermal insulation sampling technology.

The matricellular glycoprotein products of the SPP1 and SPARC genes play critical roles in many aggressive tumor phenotypes including gastric cancer. We sought to test whether the polymorphisms of these two genes, individually or jointly, influence gastric cancer susceptibility. Nine potentially functional, tagging single nucleotide polymorphisms (tagSNPs) of SPP1 and SPARC were selected and detected using the Kompetitive Allele Specific PCR method in 301 gastric cancer cases and 1441 healthy control subjects. We found that the genotype frequencies of SPP1 rs4754 in gastric cancer were significantly different from those in controls. The rs4754 TT genotype conferred an increased risk of gastric cancer, with unadjusted and adjusted ORs ranging from 1.75 to 1.95 (all P0.05). The assessment of the effect modifications of sex and age on the genetic effects also confirmed the statistically significant association of the rs4754 TT genotype with increased gastric cancer risk. Epistatic interactions were found between SPP1 rs4754 and SPARC rs1054204, rs3210714 and rs3549 (all P values for interaction0.05). During the assessment of the epistatic effects between pairs of interacting factors, increased gastric cancer risk was observed in the combined presence of the SPP1 rs4754 TT genotype and the common genotypes of interacting SPARC SNPs, with ORs ranging from 3.94 to 4.41. When the genetic influence of SPP1 rs4754 TT was excluded, the genetic effects of the SPARC rs1054204, rs3210714 and rs3549 common genotypes on gastric cancer susceptibility switched from being risky to beneficial. These data reveal an association between the SPP1 rs4754 polymorphism and altered risk of gastric cancer and highlight an important role of the epistatic effects of SPP rs4754 with SPARC polymorphisms in gastric carcinogenesis. Additional functional experiments and independent large-scale studies, especially in other ethnic populations, are needed to confirm our results.

Rationale: The antitumor activity of high-dose ascorbate has been re-evaluated recently, but the mechanism underlying cell-specific sensitivity to ascorbate has not yet been clarified. Methods: The effects of high-dose ascorbate on gastric cancer were assessed using cancer cell lines with high and low expression of GLUT1 via flow cytometry and colony formation assays in vitro and patient-derived xenografts in vivo. Results: In this study, we demonstrated that gastric cancer cells with high GLUT1 expression were more sensitive to ascorbate treatment than cells with low GLUT1 expression. GLUT1 knockdown significantly reversed the therapeutic effects of pharmacological ascorbate, while enforced expression of GLUT1 enhanced the sensitivity to ascorbate treatment. The efficacy of pharmacological ascorbate administration in mice bearing cell line-based and patient-derived xenografts was influenced by GLUT1 protein levels. Mechanistically, ascorbate depleted intracellular glutathione, generated oxidative stress and induced DNA damage. The combination of pharmacological ascorbate with genotoxic agents, including oxaliplatin and irinotecan, synergistically inhibited gastric tumor growth in mouse models. Conclusions: The current study showed that GLUT1 expression was inversely correlated with sensitivity of gastric cancer cells to pharmacological ascorbate and suggested that GLUT1 expression in gastric cancer may serve as a marker for sensitivity to pharmacological ascorbate.

DNA methylation shows complex correlations with gene expression, and the role of promoter hypermethylation in repressing gene transcription has been well addressed. Emerging evidence indicates that gene body methylation promotes transcription; however, the underlying mechanisms remain to be further investigated. Here, using methylated DNA immunoprecipitation sequencing (MeDIP-seq), bisulfite genomic sequencing, and immunofluorescent labeling, we show that gene body methylation is indeed positively correlated with rRNA gene (rDNA) transcription. Mechanistically, gene body methylation is largely maintained by DNA methyltransferase 1 (DNMT1), deficiency or downregulation of which during myoblast differentiation or nutrient deprivation results in decreased gene body methylation levels, leading to increased gene body occupancy of plant homeodomain (PHD) finger protein 6 (PHF6). PHF6 binds to hypomethylated rDNA gene bodies where it recruits histone methyltransferase SUV4-20H2 to establish the repressive histone modification, H4K20me3, ultimately inhibiting rDNA transcription. These findings demonstrate that DNMT1-mediated gene body methylation safeguards rDNA transcription by preventing enrichment of repressive histone modifications, suggesting that gene body methylation serves to maintain gene expression in response to developmental and/or environmental stresses.

To evaluate the effectiveness of minimally invasive bridging for the lateral malleolar fractures in the open comminuted ankle fractures with dislocation. Methods The clinical data of 24 patients [19 males and 5 females aged 40 to 65 years,mean(47.5±8.6)years] with open comminuted ankle fractures with dislocation who were treated in the Second Hospital of Tangshan from September 2015 to June 2017 were retrospectively analyzed.All patients were treated with minimally invasive bridging for the lateral malleolar fractures.The ankle function after treatment was assessed with the Olerud Molander Ankle(OMA)score. Results All the 24 patients were followed up from 12-26 months [mean:(14.5±2.6)months].Good fracture union was achieved in all patients after 2 - 5 months(mean:3 months).No deep infection,skin necrosis,or bone nonunion occurred after 12 months of follow-up.Only one patient suffered from partial skin necrosis at lateral malleolus,which was cured after changing wound dressings.The OMA score was 93.5(range:85-100)after 12 months(excellent in 19 cases and good in 5 cases). Conclusions Minimally invasive bridging for the lateral malleolar fractures is effective in treating the open comminuted ankle fractures with dislocation.It can obtain good reduction and fixation,reduce complications,and achieve high union rate.

To enhance the wear resistance of high chromium cast iron (HCCI, Cr26), a new wear resistant alloy with high vanadium and chromium contents (HCCI-V, 15Cr5V4MoSiMn) was designed and prepared by sand mold casting. The microstructure and the phase composition were analyzed by SEM, EDS and XRD, and the abrasive wear property was tested compared with HCCI. Results show that the new wear resistant alloy is characterized by multi-scale and multi-type carbides distributed in a metal matrix composed of martensite and retained austensite. The carbides include VC, M7C3, M2C and M23C6 (M stands for (Cr, Fe)), with dimensions ranging from tens of nanometers to tens of micrometers. The hardness and impact toughness of HCCI-V are 65 ± 0.2 HRC and 10 ± 0.12 J cm−2, respectively, far higher than that of HCCI (57 ± 0.2 HRC, 8 ± 0.12 J cm−2). When the abrasive particle size and load are 6.5 μm and 1.41 MPa respectively, the wear weight loss of HCCI and HCCI-V are 5.6 ± 0.1 mg and 0.8 ± 0.1 mg respectively, and the relative wear resistance of HCCI-V is 7. The excellent wear resistance of HCCI-V is attributed to the multi-scale carbides. The micro-scale carbides resist scratch, and the nano-scale carbides strengthens matrix. The multi-scale carbides can work collaboratively to resist abrasive wear efficiently.
