Abstract

During the COVID-19 pandemic, a considerable proportion of patients developed a severe condition that included respiratory failure, shock, or multiple organ dysfunction. Acute Kidney Injury (AKI) has been recognized as a possible cause of severe COVID-19 development. Given this, this study investigates the occurrence and consequences of AKI in Mexican patients to contribute to better knowledge and management of this problem. Methods: Using a retrospective observational cohort methodology, we investigated 313 cases from a cohort of 1019 patients diagnosed with COVID-19 at the IMSS Zacatecas General Hospital of Zone No. 1 in 2020. The prevalence of AKI was determined using the AKIN criteria based on serum creatinine levels and a detailed review of demographic characteristics, medical history, comorbidities, and clinical development. Results: The data showed a 25.30% prevalence of AKI among patients infected with severe COVID-19. Remarkably, these patients with AKI exhibited an advanced age (>65 years), arterial hypertension, a higher number of white blood cells during admission and the hospital stay, and elevated levels of C-reactive protein, serum creatinine, and blood urea nitrogen (BUN). Clinically, patients with AKI had signs of prostration, pneumonia, and the requirement for ventilatory assistance when compared to those without AKI. Finally, those diagnosed with AKI and COVID-19 had a 74% death rate. Relative risk analyses indicated that age (>65 years), arterial hypertension, high creatinine levels, endotracheal intubation, and pneumonia are associated with the development of AKI. On the other hand, among the protective factors against AKI, high hemoglobin levels and the consumption of statins during COVID-19 were found. Conclusions: The findings of this study underscore the significance of promptly identifying and effectively managing AKI to potentially alleviate the negative consequences of this complication within the Mexican population during COVID-19. Abstract

Once regarded as inert organelles with limited and ill-defined roles, lipid droplets (LDs) have emerged as dynamic entities with multifaceted functions within the cell. Recent research has illuminated their pivotal role as primary energy reservoirs in the form of lipids, capable of being metabolized to meet cellular energy demands. Their high dynamism is underscored by their ability to interact with numerous cellular organelles, notably the endoplasmic reticulum (the site of LD genesis) and mitochondria, which utilize small LDs for energy production. Beyond their contribution to cellular bioenergetics, LDs have been associated with viral infections. Evidence suggests that viruses can co-opt LDs to facilitate their infection cycle. Furthermore, recent discoveries highlight the role of LDs in modulating the host's immune response. Observations of altered LD levels during viral infections suggest their involvement in disease pathophysiology, potentially through production of proinflammatory mediators using LD lipids as precursors. Abstract

Human Adenoviruses are a diverse family of viruses that can infect a variety of tissues causing acute or persistent infection. Viruses induce numerous cellular alterations as they hijack cellular functions to promote viral progeny. Recent research has shed light on the functions of viral proteins in orchestrating viral production, revealing that many of these functions overlap with oncogenesis or metabolic disruption. Studies of the Adenovirus family (Adenoviridae) have identified oncogenic members, such as Adenovirus (Ad-)2, 5, 9, and 12, and also Ad-36, which is most extensively studied for its ability to induce metabolic alterations. Specifically, Adenoviruses encode a gene product known as early region 4 open reading frame 1 (E4orf1), which has emerged as an oncoprotein and regulator of metabolism depending on the lineage of the infected host cell. Abstract

Viruses depend on cell machinery to replicate, which strategically modulates the metabolism and physiology of host cells, changing cell architecture and functions. In this sense, it has been shown that alphavirus induces changes in the membrane, alters glycolysis rates, and even affects autophagy/apoptosis. The genus Alfavirus belongs to the family Togaviridae, including Venezuelan equine encephalitis (VEEV) and Chikungunya virus (CHIKV). The impact of CHIKV on mitochondria structure has not been determined, so this project aimed to evaluate the effects of CHIKV on mitochondria. For this purpose, the immunofluorescence technique evaluated changes in morphology, distribution, and number of mitochondria at 12, 14, 16, and 18 hpi. The data obtained suggest perinuclear clustering, a decrease in the number of mitochondria, and modification of mitochondrial morphology depending on the time of infection. Subsequently, the Macintosh MitoProt program was used to determine whether structural proteins or peptides (capsid, E1, E2, E3, and 6K) had a mitochondrial target, with the capsid protein having the highest score.
