Uropathogenic Escherichia coli (UPEC) can undergo extensive filamentation in the host during acute urinary tract infections (UTIs). It has been hypothesised that this morphological plasticity allows bacteria to avoid host immune responses such as macrophage engulfment. However, it is still unclear what properties of filaments are important in macrophage-bacteria interactions. The aim of this work was to investigate the contribution of bacterial biophysical parameters, such as cell size and shape, and physiological parameters, such as cell surface and the environment, to macrophage engulfment efficiency. Viable, reversible filaments of known lengths and volumes were produced in the UPEC strain UTI89 using a variety of methods, including exposure to cell-wall targeting antibiotics, genetic manipulation and isolation from an in vitro human bladder cell model. Quantification of the engulfment ability of macrophages using gentamicin-protection assays and fluorescence microscopy demonstrated that the ability of filaments to avoid macrophage engulfment is dependent on a combination of size (length and volume), shape, cell surface and external environmental factors. UTI89 filamentation and macrophage engulfment efficiency was also found to occur independently of the SOS-inducible filamentation genes, sulA and ymfM in both in vivo and in vitro models of infection. Compared to filaments formed via antibiotic inhibition of division, the infection-derived filaments were preferentially targeted by macrophages. With several strains of UPEC now resistant to current antibiotics, our work identifies the importance of bacterial physiological and morphological states during infection. Author summary: Urinary tract infections (UTIs) are one of the most common bacterial infections worldwide with 50% of women suffering from a UTI during their lifetime. Escherichia coli is the primary bacteria responsible for UTIs and is usually found in short rod forms. However, during UTIs E. coli can elongate into extremely long thin shapes called 'filaments'. Filaments are thought to be advantageous during infections because they are too long to be engulfed and killed by immune cells called macrophages. Due to increasing antibiotic resistance in bacteria there is a strong need for the discovery of new ways to treat infections and this is only possible once we thoroughly understand the mechanisms bacteria employ to overcome our immune response. Therefore, we investigated the effect of E. coli filamentation on macrophage engulfment along with other aspects of bacteria reported to influence engulfment. We found that the ability of filaments to avoid macrophage engulfment is dependent on a combination of size (length and volume), shape, surface and external environmental factors. Our research has highlighted the importance of bacterial shape changes during infections and provided a foundational understanding of macrophage engulfment of filaments. Catalytic ozonation is a new method used for removal of NH4OH solution. Therefore, high catalytic performance (activity and selectivity) should be achieved. In this work, we report the synthesis and catalytic performance of Fe2O3-Co3O4 modified dolomite in the catalytic ozonation of NH4OH solution. Dolomite was successfully activated and modified with Fe2O3 and Co3O4. Firstly, dolomite was activated by heating at 800°C for 3 h and followed by KOH treatment. Activated dolomite was modified with Fe2O3 by the atomic implantation method using FeCl3 as Fe source. Fe2O3 modified dolomite was further modified with Co3O4 by precipitation method. The obtained catalysts were characterized by X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), energy-dispersive X-ray spectroscopy (EDS), scanning electron microscopy (SEM), N2 adsorption–desorption (BET), and temperature-programmed reduction (H2-TPR). From SEM image, it was revealed that nano-Fe2O3 and Co3O4 particles with the size of 80–120 nm. Catalytic performance of activated dolomite, Fe2O3 modified dolomite, and Fe2O3-Co3O4 modified dolomite in catalytic ozonation of NH4+ solution was investigated and evaluated. Among 3 tested catalysts, Fe2O3-Co3O4 modified dolomite has the highest NH4+ conversion (96%) and N2 selectivity (77.82%). Selectivity toward N2 over the catalyst was explained on the basis of bond strength M-O in oxides through the standard enthalpy Δ H ° f of oxide. Catalyst with lower Δ H ° f value has higher N2 selectivity and the order is the following: Co3O4 (Δ H ° f of 60 kcal (mole O)) > Fe2O3 (Δ H ° f of 70 kcal (mole O)) > MgO (Δ H ° f of 170 kcal (mole O)). Moreover, high reduction ability of Fe2O3-Co3O4 modified dolomite could improve the N2 selectivity by the reduction of NO3- to N2 gas. [ABSTRACT FROM AUTHOR]

Ferns are among the most popular groups of plants in the tropics and subtropics, and their role as carbon sequestrators has been widely recognized. However, there is little understanding of the silicaceous structures (phytoliths) of ferns, rate of phytolith turnover, the consequences for organic matter sequestered in phytoliths and consequences for other soil properties. In the study reported here, high‐resolution X‐ray tomographic microscopy and chemical characterization were applied to examine the traits of phytoliths of the fern Dicranopteris linearis (Burm.f.) Underw. (D. linearis), with a focus on their dissolution properties and accumulation in northern Vietnamese soils in relation to soil properties. Tomographic images revealed an inter‐embedding structure of silica and organic matter, especially in leaf‐derived material. We propose that organic matter and silica can preserve each other against decomposition. In batch experiments, there was a relatively small rate of dissolution of phytoliths with dry ashing and subsequent H2O2 treatment. Silicon (Si) dissolution for D. linearis phytolith samples was much less than that for rice phytoliths. Despite the fact that the aluminum (Al) content was large in D. linearis leaves, batch dissolution data did not confirm a relation between Al and the slow rate of phytolith dissolution. The soil phytolith content varied from 0.9 to 7.5 g kg−1 in the topsoil across the mountainous areas in northern Vietnam, whereas it tended to be smaller in the subsoil. The data indicate a relation between phytolith and soil organic matter, clay content, oxalate‐soluble Al and electrical conductivity, suggesting that these soil properties are among the important factors affecting the size of the soil phytolith Si pool. The nano-Fe-Fe3O4-graphene oxide (GO) composite was successfully synthesized by the precipitation method and followed by chemical reduction using FeCl3 as iron sources and NaBH4 as reducing agent. The sample was characterized by XRD, FTIR, TEM, BET, XPS, and VMS. From XRD results, it reveals the formation of both zero valent iron (Fe) and Fe3O4 particles on GO surface. From TEM images, it showed that both Fe3O4-GO and Fe-Fe3O4-GO had a small particle size of 10-20 nm. The as-prepared Fe3O4-GO and Fe-Fe3O4-GO nanocomposites were tested in the photocatalytic degradation of phenol, which exhibited excellent catalytic activity. After 90 min of reaction under the simulated sunlight irradiation, the conversion reached the value of 87% for Fe-Fe3O4-GO and 75% for Fe3O4-GO composite, respectively. The catalyst also showed the high stability. Thus, after three cycles, the conversion of 84% still remained. The high photocatalytic activity of Fe-Fe3O4-GO composite was rationalized by synergistic effect of Fe° and Fe2+. [ABSTRACT FROM AUTHOR]

Abstract: Ag3PO4 nanoparticles have been successfully synthesized by precipitation of AgNO3 and H3PO4 in the presence of oleylamine. The products were characterized by XRD, UV–vis and FE-SEM. It showed that the particle sizes of products ranged from 5 to 10nm depending on the synthesis conditions. UV–vis result revealed a strong light absorbance in the visible light region at 470–475nm. Photocatalytic activities of TiO2 and Ag3PO4 nanoparticles were tested in photocatalytic degradation of Rhodamine B (RhB) under visible light irradiation. In comparison to nano-TiO2, nano-sized Ag3PO4 samples exhibited much higher photocatalytic activity in the RhB degradation. After 3h of reaction, the conversion reached the value of 96%. Obesity is associated with increased incidence and metastasis of triple-negative breast cancer, an aggressive breast cancer subtype. The extracellular matrix (ECM) is a major component of the tumor microenvironment that drives metastasis. To characterize the temporal effects of age and high-fat diet (HFD)-driven weight gain on the ECM, we injected allograft tumor cells at 4-week intervals into mammary fat pads of mice fed a control or HFD, assessing tumor growth and metastasis and evaluating the ECM composition of the mammary fat pads, lungs, and livers. Tumor growth was increased in obese mice after 12 weeks on HFD. Liver metastasis increased in obese mice only at 4 weeks, and elevated body weight correlated with increased metastasis to the lungs but not the liver. Whole decellularized ECM coupled with proteomics indicated that early stages of obesity were sufficient to induce changes in the ECM composition. Obesity led to an increased abundance of the proinvasive ECM proteins collagen IV and collagen VI in the mammary glands and enhanced the invasive capacity of cancer cells. Cells of stromal vascular fraction and adipose stem and progenitor cells were primarily responsible for secreting collagen IV and collagen VI, not adipocytes. Longer exposure to HFD increased the invasive potential of ECM isolated from the lungs and liver, with significant changes in ECM composition found in the liver with short-term HFD exposure. Together, these data suggest that changes in the breast, lungs, and liver ECM underlie some of the effects of obesity on triple-negative breast cancer incidence and metastasis.
