22. Fame, Feminism, and Failure: Lessons from the Rise and Fall of Time's Up.

Abstract

In the wake of #MeToo, the non-profit feminist organization, Time's Up, was started by a group of celebrity women in 2018, only to dissolve four years later, mired in scandal. Using Boorstin's theorization of the pseudo-event, this project analyzes the rise and fall of Time's Up through a multimodal analysis of celebrities' publicization of its launch alongside an analysis of reports detailing its demise. The findings show how the illusory, image-focused logics of social media and celebrity culture as well as the organization's prioritization of the powerful subverted its goals of promoting gender equality. As of the few examples of large-scale feminist mobilization within celebrity circles, Time's Up demonstrates the difficulty of converting stars' publicity into sustained activism, providing lessons about the pros and cons of celebrity's deployment of solidarity capital in social justice work. 38. Novel blended SNRPE‐related spliceosomopathy phenotype characterized by microcephaly and congenital atrichia.

Abstract

Variants in genes encoding core components of the spliceosomes are associated with craniofacial syndromes, collectively called craniofacial spliceosomopathies. SNRPE encodes a core component of pre‐mRNA processing U‐rich small nuclear ribonuclear proteins (UsnRNPs). Heterozygous variants in SNRPE have been reported in six families with isolated hypotrichosis simplex in addition to one case of isolated non syndromic congenital microcephaly. Here, we report a patient with a novel blended phenotype of microcephaly and congenital atrichia with multiple congenital anomalies due to a de novo intronic SNRPE deletion, c.82‐28_82‐16del, which results in exon skipping. As discussed within, this phenotype, which we propose be named SNRPE‐related syndromic microcephaly and hypotrichosis, overlaps other craniofacial splicesosomopathies. [ABSTRACT FROM AUTHOR]

The microbiome from fecal specimens with that obtained from normal mucosa in children with FAP and from fecal specimens in healthy controls was compared in addition to biopsies from an adenoma to synchronous normal mucosa. Observations including significant preferential expression of probiotic candidate bacteria in control stool vs. decreased potentially protective bacterial subtypes in polyp compared with normal mucosa were made along with a reduction in alpha- and beta-diversities in polyp compared with apparently normal mucosa. Machine learning further distinguished bacterial populations between patients with and without polyposis. Changes are indicated through up or down arrows, respectively. 46. Recurrent FOXP4 nonsense variant in two unrelated patients: Association with neurodevelopmental disease and congenital diaphragmatic hernia.

Abstract

De novo variants in FOXP4 were recently associated with a neurodevelopmental disorder characterized by speech and language delay, growth abnormalities, hypotonia, and variable congenital abnormalities, including congenital diaphragmatic hernia, cervical spine abnormalities, strabismus, cryptorchidism, and ptosis. The variant spectrum in this small cohort was limited to de novo missense except for one frameshift, the inheritance of which was unknown. Variants tested in vitro exhibited reduced repressor transcriptional activity, indicating loss of function is the likely mechanism of disease, but only one frameshift variant was reported. Here, we report four affected individuals from two unrelated families heterozygous for a nonsense variant, c.1893C > G, p.Tyr631*, in FOXP4. The phenotype of the affected children includes developmental delay, feeding difficulties in infancy, and similar facial features. In both cases, the variant was inherited from a parent with mild or even subclinical features. Interestingly, one patient presented with congenital diaphragmatic hernia, as reported in two other FOXP4 patients. This report implicates FOXP4 truncating variants in human disease and highlights the wide phenotypic spectrum and variable expressivity. [ABSTRACT FROM AUTHOR]

49. Gas Powered Reefs: Exploring the Nature and Variability of the Coral Volatilome

Abstract

Biogenic volatile organic compounds (BVOCs) are a diverse class of chemicals well studied in terrestrial ecosystems, where they play important ecological and physiological roles, while influencing atmospheric processes and climate. As the health of tropical coral reefs rapidly deteriorates, BVOCs can potentially mitigate stress events directly by functioning as antioxidants and indirectly through atmospheric chemistry. Yet, the composition of the coral volatilome (total BVOCs) and its potential importance for coral reef functioning is understudied. I investigated the BVOC capacity of corals systematically – from microbial symbiont to coral holobiont. I examined Symbiodiniaceae, the photosynthetic microbes that provide nutrients to corals. Across six species, I detected 82 BVOCs and using a targeted thermal stress experiment on two key species, Cladocopium goreaui and Durusdinium trenchii, I identified significant changes in specific BVOCs, with the majority of these significantly increasing following stress. Given that Symbiodiniaceae are associated with complex and abundant bacterial communities, I described the microbiome of 18 species of Symbiodiniaceae. Three bacterial genera were consistently present across all Symbiodiniaceae species: Labrenzia, Marinobacter and an unclassified Chromatiaceae. I then characterised the volatilome of Labrenzia sp. 21p and Marinobacter adhaerens HP15 and detected 35 BVOCs between them. The composition of the Labrenzia sp. volatilome significantly changed following incubation in Symbiodiniaceae exudate and additional changes were detected in the quantities of individual BVOCs in both bacterial species. This indicates the ability of Symbiodiniaceae-associated bacteria to alter their BVOC production in the presence of Symbiodiniaceae-derived chemicals. I finally characterised the volatilomes of two common, heat-sensitive, reef-building corals (Acropora intermedia and Pocillopora damicornis) during a simulated heat stress event. I detected 88 BVOCs from these holobionts however, the BVOC richness of both species decreased following stress (by 41% in A. intermedia and 62% in 𝘗. damicornis) and the abundance of multiple BVOCs significantly decreased. As such, this study revealed that thermal stress influences the coral holobiont by decreasing the richness and abundance of the volatilome. From microbe to holobiont, I consistently detected a wide range of BVOCs, many of which are implicated in stress response, signalling, antimicrobial defence or potentially impacting local climate. I demonstrated that corals and therefore potentially coral reefs, are significant sources of numerous BVOCs that should receive increased attention to determine their biological functions. This work highlights the diversity of BVOCs produced by corals and their constituents, and provides important new knowledge for the successful management and conservation of these threatened ecosystems.
