BackgroundDynamic contrast-enhanced MRI (DCE-MRI) parameters have been shown to be biomarkers for treatment response in glioblastoma (GBM). However, variations in analysis and measurement methodology complicate determination of biological changes measured via DCE. The aim of this study is to quantify DCE-MRI variations attributable to analysis methodology and image quality in GBM patients.MethodsThe Extended Tofts model (eTM) and Leaky Tracer Kinetic Model (LTKM), with manually and automatically segmented vascular input functions (VIFs), were used to calculate perfusion kinetic parameters from 29 GBM patients with double-baseline DCE-MRI data. DCE-MRI images were acquired 2-5 days apart with no change in treatment. Repeatability of kinetic parameters was quantified with Bland-Altman and percent repeatability coefficient (%RC) analysis.ResultsThe perfusion parameter with the least RC was the plasma volume fraction (v p ), with a %RC of 53%. The extra-cellular extra-vascular volume fraction (v e ) %RC was 82% and 81%, for extended Tofts-Kety Model (eTM) and LTKM respectively. The %RC of the volume transfer rate constant (K trans ) was 72% for the eTM, and 82% for the LTKM, respectively. Using an automatic VIF resulted in smaller %RCs for all model parameters, as compared to manual VIF.ConclusionsAs much as 72% change in K trans (eTM, autoVIF) can be attributable to non-biological changes in the 2-5 days between double-baseline imaging. Poor K trans repeatability may result from inferior temporal resolution and short image acquisition time. This variation suggests DCE-MRI repeatability studies should be performed institutionally, using an automatic VIF method and following quantitative imaging biomarkers alliance guidelines.

Current diagnostic criteria for myelofibrosis are largely based on bone marrow (BM) biopsy results. However, these have several limitations, including sampling errors. Explorative studies have indicated that imaging might form an alternative for the evaluation of disease activity, but the heterogeneity in BM abnormalities complicates the choice for the optimal technique. In our prospective diagnostic pilot study, we aimed to visualize all BM abnormalities in myelofibrosis before and during ruxolitinib treatment using both PET/CT and MRI. A random sample of patients was scheduled for examinations at baseline and after 6 and 18 months of treatment, including clinical and laboratory examinations, BM biopsies, MRI (T1-weighted, Dixon, dynamic contrast-enhanced (DCE)) and PET/CT ([15O]water, [18F]NaF)). At baseline, all patients showed low BM fat content (indicated by T1-weighted MRI and Dixon), increased BM blood flow (as measured by [15O]water PET/CT), and increased osteoblastic activity (reflected by increased skeletal [18F]NaF uptake). One patient died after the baseline evaluation. In the others, BM fat content increased to various degrees during treatment. Normalization of BM blood flow (as reflected by [15O]water PET/CT and DCE-MRI) occurred in one patient, who also showed the fastest clinical response. Vertebral [18F]NaF uptake remained stable in all patients. In evaluable cases, histopathological parameters were not accurately reflected by imaging results. A case of sampling error was suspected. We conclude that imaging results can provide information on functional processes and disease distribution throughout the BM. Differences in early treatment responses were especially reflected by T1-weighted MRI. Limitations in the gold standard hampered the evaluation of diagnostic accuracy.

Abstract Silicon carbide (SiC) is one of the hardest known materials. Its exceptional mechanical properties combined with its high thermal conductivity make it a very attractive material for a variety of technological applications. Recently, it is discovered that two‐layer epitaxial graphene films on SiC can undergo a pressure activated phase transition into a sp3 diamene structure at room temperature. Here, it is shown that epitaxial graphene films grown on SiC can increase the hardness of SiC up to 100% at low loads (up to 900 µN), and up to 30% at high loads (10 mN). By using a Berkovich diamond indenter and nanoindentation experiments, it is demonstrated that the 30% increase in hardness is present even for indentations depths of 175 nm, almost three hundred times larger than the graphene film thickness. The experiments also show that the yield point of SiC increases up to 77% when the SiC surface is coated with epitaxial graphene. These improved mechanical properties are explained with the formation of diamene under the indenter's pressure.

Quinoa and amaranth are of special interest since they are increasingly used for the development of new bakery products with enhanced nutritional value. The aim of the study was to evaluate the agronomic, microbiological, and nutritional characteristics of quinoa and amaranth seeds grown in Southern Italy. For this reason, quinoa Titicaca and three amaranth accessions (5, 12, and 14) were cultivated in different experimental fields in the Campania Region and analyzed for the cultivation aspects, chemical composition, and microbiological quality of the seeds. All seeds showed a good adaptability to cultivation in the experimental areas of the Mediterranean basin. Quinoa seeds were characterized by their higher protein, fat, and ash content than the amaranth seeds, which were characterized by their higher value in dietary fiber. All seeds, regardless of the geographical area of production, were contaminated with yeasts, moulds, and spore-forming bacteria, mainly Bacillus cereus, B. licheniformis, B. safensis and B. subtilis, as identified by 16S rRNA sequencing analysis. So, the detection of Bacillus spp. must be strongly monitored, as quinoa and amaranth seeds could be used in bread production, where they can cause ropiness, resulting in great economic losses for the industries.

Abstract Understanding phase transformations in 2D materials can unlock unprecedented developments in nanotechnology, since their unique properties can be dramatically modified by external fields that control the phase change. Here, experiments and simulations are used to investigate the mechanical properties of a 2D diamond boron nitride (BN) phase induced by applying local pressure on atomically thin h‐BN on a SiO2 substrate, at room temperature, and without chemical functionalization. Molecular dynamics (MD) simulations show a metastable local rearrangement of the h‐BN atoms into diamond crystal clusters when increasing the indentation pressure. Raman spectroscopy experiments confirm the presence of a pressure‐induced cubic BN phase, and its metastability upon release of pressure. Å‐indentation experiments and simulations show that at pressures of 2–4 GPa, the indentation stiffness of monolayer h‐BN on SiO2 is the same of bare SiO2, whereas for two‐ and three‐layer‐thick h‐BN on SiO2 the stiffness increases of up to 50% compared to bare SiO2, and then it decreases when increasing the number of layers. Up to 4 GPa, the reduced strain in the layers closer to the substrate decreases the probability of the sp2‐to‐sp3 phase transition, explaining the lower stiffness observed in thicker h‐BN.

Today, irrigated agriculture is even more influenced by climate change with consequent negative effects on food security. The Mediterranean area is most affected by climate change, leading to greater exposure to uncertainty and production risks. In these environments, water stress, rainfall variability, and soil salinization have been accentuated. Improving crop productivity by minimizing such effects is possible through intelligent climate farming practices (CSFP). Towards resilient and sustainable integrated agro-ecosystems through appropriate climate-smart farming practices (TRUSTFATM) is a project funded by the European Union’s Horizon 2020 research and innovation program with the aim to design integrated agro-ecosystems by conserving natural resources and using the principles of the circular economy for developing climate-resilient production systems in Egypt, Morocco, Italy, France, and Senegal. The Department of Agricultural and Environmental Science (DISAAT) of the University of Bari is responsible for coordinating the activities (starting in 2022) related to the introduction of new crop varieties and management of water and efficient irrigation systems, such as deficit irrigation, use of marginal quality (saline water) irrigation water, and introduction of abiotic stress-tolerant crops.

Agriculture in south Europe is facing the negative effect of abiotic stresses such as salinity that mostly affect the seed production and seed quality of traditional crops. Under these conditions, quinoa represent a good alternative to ensure the production of high-protein-quality seeds thanks to its tolerance to abiotic stresses. In 2015–2017, a sweet variety of quinoa, “Vikinga”, was tested in Italy within the PROTEIN2FOOD project (EU Horizon2020) as high-quality-protein crop to enhance food protein production in Europe. A field trial was carried out at the experimental farm of CNR-ISAFOM in South Italy, to evaluate the combined effect of drought and salinity on quinoa Vikinga; both freshwater and saline water were used for irrigation. The plots were arranged in a randomized complete block design. The main yield parameters (seed yield, aboveground dry biomass 1000 seed weight), the protein content and other quality traits were analyzed at harvest, to evaluate the effect of applied treatments. The results showed that, in general, different treatments did not affect the main production and quality traits of quinoa “Vikinga”.

Introduction: The novel coronavirus has spread rapidly around the world with particularly high mortality in the elderly. Care and nursing homes have become the sites of greatest concentration of cases. We intended to review 30-day mortality of COVID-19 patients with hip fractures. This is relevant given the disease impact in this age group. Methods: Medline and Embase databases were searched for relevant studies linked to mortality and morbidity in COVID-19 patients who have undergone non-elective hip surgeries using the keywords “COVID-19”OR “SARS-cov-2”OR “Coronavirus Infections”; AND “Surgery”OR “Hip”OR “Fracture”OR “Orthopedics.” We included all patients with hip fractures, but excluded pathological fractures and other non-traumatic hip pathologies. Four-hundred and eighty-one articles were identified for screening, in addition to an unpublished case-series of 67 patients that have 3 cases turned positive for COVID 19, yielding a total of 50patients for the final review. Results: The study included 4 articles published until May 9th 2020 and a case-series: 26(52%) patients were females; the median age was 86years; hypertension(53.6%), diabetes mellitus type II(28.6%), and coronary artery heart disease(25%) were the most common comorbidities; 34(68%) patients had intertrochanteric hip fracture and 16(32%) patients had femoral neck fractures; 22(59.5%) patients underwent cephalomedullary nail fixation, 12(32.4%) patients had hemiarthroplasty, and for 3(8.1%) patients, the type of surgery was not documented; 20(40%) patients died(12patients died before the surgery and 8died after surgery at a median time of 3days), 29(76.3%) patients had an unremarkable course throughout hospitalization and were discharged including 1(2.6%) patient managed non-operatively, and 1(2.6%) patient was admitted to the ICU after the surgery but eventually discharged. Conclusion: COVID-19 infected elderly patients have a higher 30-days mortality rate compared to non-COVID-19 infected cases. Further studies are warranted to look at the morbidity and mortality rates in COVID-19 positive patients with hip fractures and to investigate how these outcomes can be improved. Lentil (Lens culinaris Medik.) is a popular legume crop in the Mediterranean region, widely grown for its nutritious seeds and improving soil fertility. Lentil yield is a critical and challenging trait for crop genetic improvement because it is influenced by various factors that have detrimental effects on seed yields and seed quality traits. This research was carried out in Italy between 2017 and 2019 to identify high-performing stable genotypes presenting multiple desirable traits and to assess the seed quality of 13 lentil accessions in the field. According to the results of the multi-trait stability index (MTSI), (1) only three accessions (Altamura, Easton, and Caltagirone) fared better in various environmental conditions, and (2) the selected accessions had strength toward seed yield (SY), above-ground biomass (AGB), and 1000-seed weight (THS). The genotype × environment interaction (GEI) effects were highly significant for all traits. During the third growing season, most lentil accessions were sensitive to frost. There was no correlation between lentil seed yield and protein concentration. The MTSI is a useful tool for breeders interested in selecting accessions based on their mean performance and stability, as well as desirable traits and minimum multicollinearity issues.

Abstract The mechanisms underlying mesenchymal stem cells’ (MSC) suppressive potency are largely unknown. We here show that highly suppressive human adipose tissue-derived MSC (AdMSC) display and induce a differential immunologic profile, upon ongoing AdMSC suppressive activity, promoting: (i) early correlated inhibition of IFN-γ and TNF-α production, along IL-10 increase, (ii) CD73+Foxp3+Treg subset expansion, and (iii) specific correlations between gene expression increases, such as: MMP9 correlated with CCL22, TNF, FASL, RUNX3, and SEMAD4 in AdMSC and, in T cells, MMP9 upregulation correlated with CCR4, IL4 and TBX21, among others, whereas MMP2 correlated with BCL2 and LRRC31. MMP9 emerged as an integrating molecule for both AdMSC and T cells in molecular networks built with our gene expression data, and we confirmed upregulation of MMP9 and MMP2 at the protein level, in AdMSC and T cells, respectively. MMP2/9 inhibition significantly decreased AdMSC suppressive effect, confirming their important role in suppressive acitivity. We conclude that MMP9 and 2 are robust new players involved in human MSC immunoregulatory mechanisms, and the higher suppressive activity correlates to their capacity to trigger a coordinated action of multiple specific molecules, mobilizing various immunoregulatory mechanisms.

PurposePharmacokinetic models facilitate assessment of properties of the micro-vascularization based on DCE-MRI data. However, accurate pharmacokinetic modeling in the liver is challenging since it has two vascular inputs and it is subject to large deformation and displacement due to respiration.MethodsWe propose an improved pharmacokinetic model for the liver that (1) analytically models the arrival-time of the contrast agent for both inputs separately; (2) implicitly compensates for signal fluctuations that can be modeled by varying applied flip-angle e.g. due to B1-inhomogeneity. Orton's AIF model is used to analytically represent the vascular input functions. The inputs are independently embedded into the Sourbron model. B1-inhomogeneity-driven variations of flip-angles are accounted for to justify the voxel's displacement with respect to a pre-contrast image.ResultsThe new model was shown to yield lower root mean square error (RMSE) after fitting the model to all but a minority of voxels compared to Sourbron's approach. Furthermore, it outperformed this existing model in the majority of voxels according to three model-selection criteria.ConclusionOur work primarily targeted to improve pharmacokinetic modeling for DCE-MRI of the liver. However, other types of pharmacokinetic models may also benefit from our approaches, since the techniques are generally applicable.

Chickpea (Cicer arietinum L.) is an important cool-season food legume crop that is mainly cultivated as a rainfed crop. This study was conducted in Italy between 2017 and 2019 to evaluate the stability of seed yield (SY), biomass (AGB) and 1000 seed weight (THS), and to assess the seed quality of 12 kabuli chickpea accessions under field conditions. The likelihood-ratio test revealed significant effects of genotype only for the SY and THS. The environment and genotype × environment interaction (GEI) effects were highly significant for all variables. We found that the environment (year) and GEI explain 55.72% and 20.87% of the total seed yield variation, respectively. Most chickpea accessions showed sensitivity to frost conditions in the third growing season. No relationship was observed between the yield and the protein content in Kabuli chickpea. Among the accessions, Ares and Reale showed the best performance under all environmental conditions, and the Reale was the most stable chickpea.

Faba beans (Vicia faba L.), also known as fava beans, like other crops, are influenced by several factors: their genotype, environment, and management, as well as the interaction between these, have an important impact on seed yielding and seed quality traits. This study was conducted at three locations in South Italy between 2017 and 2019 to evaluate the sowing date effect on yield and yield components of three Vicia faba L., originating from cool climates. The results showed that seed yield (SY) and yield components declined with sowing delay. The crop’s environment (year × site) and management (sowing date) were found to explain 34.01% and 42.95% of the total seed yield variation, respectively. The data showed that the tested genotypes were positively influenced by the environment with sandy loam soil and early winter sowing date, resulting in either a greater number of SY and THS than in the other environment. The three faba bean genotypes showed tolerance to winter frost conditions in the two growing seasons.
