Abstract

A 9-yr-old, female Bactrian camel (Camelus bactrianus) developed severe pruritus in June 1993. During the next 8 yr, the camel exhibited annual episodes of pruritus and epiphora, generally beginning in June and subsiding in October of each year. These signs could usually be controlled with topical agents and fly repellents, although pruritus flare-ups were effectively controlled with intermittent injections of diphenhydramine and corticosteroids. There were no signs during the colder months. The pruritus became more severe and difficult to control when the camel reached its 18th year. Histopathologic descriptions of skin biopsies taken from several sites suggested a hypersensitivity reaction with secondary changes because of bacterial infection or corticosteroid administration (or both). An intradermal test (IDT) performed using 62 allergens regionalized to Northern California resulted in 17 positive test sites. Hyposensitization therapy was initiated using allergens chosen on the basis of exposure, availability, and the results of the IDT, using standard protocols. The clinical signs of pruritus were markedly reduced in the 2 yr after the hyposensitization injections were initiated.

Abstract

This is the first report of a rapid technique for the isolation of elephant heterophils form ethylene diamine tetra-acetic acid (EDTA) anticoagulated whole blood using hetastarch sedimentation and Percoll discontinuous gradient centrifugation modified from equine neutrophil isolation techniques (Sedgwick et al. 1986; Pycock 1987). Heterophil purity and viability was greater than 98% and 99%, respectively. The integrity of the isolated heterophils was evaluated by light microscopy and flow cytometric forward (size of cell) and orthogonal light-scattering (granularity) properties. In addition, functional properties including phagocytosis and oxidative product formation were evaluated and compared to heterophils within a buffy coat (centrifuged blood) population. Light microscopic morphological features including toxic or pyknotic changes, general size, cellular membrane and cytoplasmic granule integrity of heterophils from peripheral blood, buffy coats and isolated heterophils were similar. Flow cytometric forward and orthogonal light-scattering properties were unchanged. No significant differences between isolated cells and heterophils within buffy coat preparations were evident for phagocytosis or oxidative product formation. Variation in percentage phagocytosis (66% to 86%) and mean channel fluorescence intensity (2288 to 3625) in isolated heterophils was observed among individual elephants. This may be attributed to functional heterogeneity of heterophils among elephants.

Abstract

Personnel at The Marine Mammal Center (The Center) treated 1,446 stranded marine mammals recovered from the central and northern California (USA) coast from 1984 through 1990, including California sea lions (Zalophus californianus), northern elephant seals (Mirounga angustirostris), Pacific harbor seals (Phoca vitulina richardsi), northern fur seals (Callorhinus ursinus), Steller sea lions (Eumetopias jubatus), and Guadalupe fur seals (Arctocephalus townsendi). The primary disease findings in stranded California sea lions were renal disease, renal disease complicated by severe verminous pneumonia, verminous pneumonia, seizures of unknown etiology, and renal disease complicated by severe pneumonia of unknown etiology. Stranded elephant seals included pups, yearlings with dermatological problems, and neonates. Most harbor seals admitted to The Center were underweight and premature pups. Stranded northern fur seals included animals with seizures of unknown etiology and emaciated pups. Stranded Steller sea lions included underweight pups and aged adult females with pneumonia. Two Guadalupe fur seals had hemorrhagic gastroenteritis. Incidental findings at the time of stranding among the six species included verminous pneumonia and pneumonia of unknown etiology, renal disease, internal parasitism, ophthalmologic problems, gastrointestinal disorders, otitis externa, and external wounds.

Abstract

A captive adult female American badger (Taxidea taxus) suffered periodic episodes of agitation and self-mutilation over the course of its lifetime. Initially environmental enrichment curtailed the aberrant behavior; however, intensifying clinical signs periodically required the use of diazepam for amelioration of the problem. When diazepam treatment failed to effectively manage a series of escalating behavioral problems, alternative therapy with buspirone, an azaperone anxiolytic, was initiated. The badger was treated with 10 mg buspirone p.o. b.i.d. for over 18 mo, during which time no undesirable behaviors or noticeable side effects were observed.

37. Preliminary findings of restriction fragment differences in mitochondrial DNA among killer whales (Orcinus orca)

Abstract

A preliminary assessment of mitochondrial DNA restriction patterns in the killer whale (Orcinus orca) was conducted using 10 captive North Atlantic killer whales from the southeastern coast of Iceland, a captive-born offspring of one of these whales, and 9 North Pacific killer whales. No restriction pattern variation was seen among these whales, using the enzymes BamH I, Bgl II, Hinf I, Kpn I, or Pvu II. Restriction pattern variation was found using the enzyme Hae III. This restriction endonuclease distinguished the North Atlantic killer whales (type 1) from the North Pacific killer whales. The North Pacific killer whales were further differentiated into two groups: those originating from the "resident" communities of the Vancouver Island region (type 2), and those from the "transient" community of Vancouver Island, as well as those stranded along the Oregon coast (type 3). The observed Hae III restriction pattern differences suggest that mitochondrial DNA analysis will be a valuable technique for investigating regional and local distributions of maternal lineages among killer whale pods, especially in the North Pacific.
