Taploca water sample contained a unique presence of Planctomycetota and Myxococcota at the same time with Szent Jakab water sample which showed an additional presence of Chloroflexi, Berzsenyi contained Campylobacterota at the same time with Polányi kút which had important fractions of Nitrospirota and Desulfobacterota. Finally, Kossuth Lajos water sample was dominated by Fusobacteriota, mainly members of Hypnocyclicus genus.Piricske and especially Nagy-borvíz water samples contained important fractions of Altiarchaeota, moreover, Piricske contained high fraction of Euryarchaeota phylum -most of them belong to Methanobacteriaceae family.Nitrosopumilaceae dominated Taploca and Kiskút in addition to their important presence in the rest of the samples. Diversity indices and cell count showed in general that cell count values are tending to be lower when the diversity is higher. The results of taxon specific PCR reactions showed that water treatment would be essential in some water samples.

A bioaktív komponensek érzékenyek a környezeti hatásokra és a technológiai folyamatok körülményeire, ezért ezeknek az anyagoknak a védelmét biztosítani kell, melynek hatékony módszere a mikrokapszulázás. Kutatásaink célja Magyarországon termesztett, jelentős antioxidáns kapacitással bíró gyógy- és fűszernövények (orvosi citromfű, kerti és spanyol kakukkfű), valamint színanyagokban gazdag gyümölcsféleségek (fekete bodza, szeder, cseresznye és homoktövis) kivonatainak és sűrítményeinek mikrokapszulázása és funkcionális élelmiszerekben való felhasználása. Kísérleteink első eredményeit összegezve megállapítottuk, hogy a két gyógynövény esetében a növények kivonatának felhasználásával előállított, míg a bogyós gyümölcsök esetében a sűrítmény felhasználásával legyártott mikrokapszulák antioxidáns kapacitása kiemelkedő és ezeket a kapszula-változatokat hoszszabb távú eltartási kísérleteknek is érdemes alávetni.

Periglacial and volcanic environments are considered terrestrial analogs of Mars with regard to astrobiological characteristics due to their specific set of extreme features. Ojos del Salado, the highest volcano on Earth (6893 m a.s.l.), is surrounded by several craters, one of which harbors the highest known altitude lake (6480 m a.s.l.), which is influenced by a rare combination of extreme environmental factors, that is, low mean temperature, permafrost, fumarolic activity, acidity, and extreme low organic matter content. To assess the genetic diversity and ecological tolerance of bacteria, samples were taken in February 2016 from the sediments covered with acidic cold (pH 4.88, 3.8°C) and warm (pH 2.08, 40.8°C) water. As a control, a nonvolcanic high-altitude lake (at 5900 m a.s.l.) was also studied by both cultivation-based and next-generation DNA sequencing methods. In the present study 12 water samples of five sampling sites (Tatabánya, Dandár, Szentendre, Szent Flórián and Ciprián groundwaters) known as nutrient-depleted aquatic environments were studied using amplicon sequencing (NGS) and cultivation techniques. Diversity indices and cell counts were determined to assess the species richness in relation to the cell counts within the samples, and the oligocarbophile growth capability of the isolated bacteria was tested in microtiter plates. Altogether, 55 bacterial phyla were identified from the samples by amplicon sequencing. The microbial communities of the different sampling times of the same sites did not differ significantly. Patescibacteria and Proteobacteria were present in all samples. Ciprián sample was dominated by Bacteroidetes, while in Dandár sample a high ratio of Chloroflexi was detected. Rokubacteria and WOR-1 dominated Szent Flórián sample and Tatabánya had a high number of Epsilonbacteraeota. Nine archaeal phyla were also detected, the samples were characterized by the presence of unclassified archaea and Nanoarchaeota, among them Woesearchaeia, as the most dominant. Crenarchaeota and Altiarchaeota were detected in high ratios in Dandár water samples. Among Thaumarchaeota the family Nitrosopumilaceae, and orders of Nitrosotaleales and Nitrososphaerales appeared in Szent Flórián and Tatabánya samples. Key organisms of the different biogeochemical cycles were discovered in these nutrient-depleted environments: methanogenic archaea, methanotrophic bacteria, ammonia oxidizer, nitrate reducers, diazotrophs, sulfate reducers, and sulfur oxidizer. Diversity indices and cell counts of the samples show negative correlation in case of bacteria and positive in case of archaea in Ciprián sample. The high diversity indices in Szentendre samples are connected to low cell counts, most probably due to the vulnerability of the groundwaters to the external environment factors which lead to the infiltration of soil microbes and contaminants to the water. The isolated bacteria were affiliated into four phyla, most of them belonging to Proteobacteria (59%) followed by Actinobacteria (21%), Firmicutes (17%) and Verrucomicrobia (1%). The members of the facultative chemolithotrophic genera of Sphingobium, Sphingomonas, Sphingopyxis were characterizing only Szentendre, Szent Flórián and Tatabánya samples. Only 10% of the isolated species showed an obligate oligocarbophile character. From the samples, a high number of novel bacterial taxa were cultivated. As a conclusion, our results confirmed the predominance of unclassified and unknown taxa in subsurface water, pointing to the importance and necessity of further studies to characterize these microbial populations.

Short-chain halogenated aliphatic hydrocarbons (e.g. perchloroethene, trichloroethene) are among the most toxic environmental pollutants. Perchloroethene and trichloroethene can be dechlorinated to non-toxic ethene through reductive dechlorination by Dehalococcoides sp. Bioaugmentation, applying cultures containing organohalide-respiring microorganisms, is a possible technique to remediate sites contaminated with chlorinated ethenes. Application of site specific inocula is an efficient alternative solution. Our aim was to develop site specific dechlorinating microbial inocula by enriching microbial consortia from groundwater contaminated with trichloroethene using microcosm experiments containing clay mineral as solid phase. Our main goal was to develop fast and reliable method to produce large amount (100 L) of bioactive agent with anaerobic fermentation technology. Polyphasic approach has been applied to monitor the effectiveness of dechlorination during the transfer process from bench-scale (500 mL) to industrial-scale (100 L). Gas chromatography measurement and T-RFLP (Terminal Restriction Fragment Length Polymorphism) revealed that the serial subculture of the enrichments shortened the time-course of the complete dechlorination of trichloroethene to ethene and altered the composition of bacterial communities. Complete dechlorination was observed in enrichments with significant abundance of Dehalococcoides sp. cultivated at 8 °C. Consortia incubated in fermenters at 18 °C accelerated the conversion of TCE to ethene by 7–14 days. Members of the enrichments belong to the phyla Bacteroidetes, Chloroflexi, Proteobacteria and Firmicutes. According to the operational taxonomic units, main differences between the composition of the enrichment incubated at 8 °C and 18 °C occurred with relative abundance of acetogenic and fermentative species. Organic and inorganic precipitates are both characteristic in the active hypogenic karst area of Buda Thermal Karst in Hungary. As an active system, it is a good natural laboratory to study ongoing precipitation processes. Because of anthropogenic influence and the complexity of spring environments, it is challenging to reveal all the governing factors in the process of precipitation. In situ experiments, i.e. artificially controlled natural systems simplify the complexity by adding, excluding or stabilizing influencing parameters during the experiment. CO2 degassing drives changes in the physicochemical parameters of spring waters from the discharge along their flow path. The rate and spatial extension of these changes depend on local hydrogeological, geological, climatic, topographical etc. factors, affecting precipitation processes. In this study, two one-day-long in situ experiments were executed to examine the physicochemical parameter changes of thermal water in a tunnel. The integration of the results with reactive transport models revealed the physicochemical processes of ingassing and degassing and predicted CaCO3 precipitation along the flow path. Small-scale roughness of the channel surface seemed to further influence pH and concentration of HCO3−. After 6 weeks of thermal water flowing, organic precipitate (biofilm) formed close to the discharge and then, with a sharp change, inorganic precipitate (calcite) dominates a bit further from the discharge. In situ experiments and connected numerical simulations revealed the role of CO2 degassing and calcite precipitation in the changes of physicochemical parameters, but organic precipitates also have to be considered near the discharge.

Winter conditions in aquatic habitats of the temperate zone markedly differ from those present in warmer seasons, nevertheless, relatively scarce information is available on planktonic microbial composition, as sites are not easily accessible and it was supposed traditionally that microbial activity is low during this cold period. Since microorganisms could have great impact on the ecosystem even during winter, we explored various sites in the Eastern Carpathians regarding the abundance and taxonomic composition of planktonic microorganisms. Although many of the studied environments were extreme habitats, planktonic microbial communities were abundant and mostly diverse with the presence of previously unidentified taxa.

Effect of carrier texture and type on biofilm development and maturation were studied on an eight-reactors-containing pilot-scale cascade system which treated municipal wastewater under aerobic conditions and various system operational characteristics. Dissimilarities in the biofilm structures grown on four types of polymer fiber-based carriers placed in the reactors having different role during wastewater purification and compositional changes of bacterial communities during biofilm maturation were clearly observable. In the first 2–3 weeks, taxonomic diversity of biofilm bacterial communities increased and reached their maximum values. Carrier type and texture had comparable effect on the weight, activity and composition of biofilms as the wastewater matrix. Biofilm community grown on the carrier having the best colonization properties were dominated by Proteobacteria, Synergistetes, Bacteroidetes and Firmicutes. In the nascent 7-days-old biofilm, aerobic chemoorganotrophic genera, while in the mature 45-days-old biofilm anaerobes, such as sulfate-reducer and fermentative genera were the major members of the bacterial communities.

Due to the increasing amount of persistent organic pollutants (POPs) in general and pharmaceutical residues in particular in municipal wastewater, the efficiency of water treatment technologies should be improved. Following the biological treatment of wastewater nanofiltration offers a possible way for the removal of POPs. In this study β-cyclodextrin containing nanofilters having different chemical composition and thickness (1.5–3.5 mm) were investigated. For their characterization, their adsorption capacity was determined applying ibuprofen containing model solution and total organic carbon (TOC) analyzer. It could be established that the regeneration of nanofilters with ethanol and the application of inorganic additives (NaCl, NaHCO3, NH4HCO3) increased the adsorption capacity of nanofilters. The best results were achieved with chemical composition of 30 m/m% β-cyclodextrin polymer beads and 70 m/m% ultra-high molecular weight polyethylene in the presence of 12 mmol ammonium hydrogen carbonate/nanofilter.

The effect of vertical physico-chemical stratification on the planktonic microbial community composition of the deep, hypersaline and heliothermal Lake Ursu (Sovata, Romania) was examined in this study. On site and laboratory measurements were performed to determine the physical and chemical variables of the lake water, and culture-based and cultivation-independent techniques were applied to identify the members of microbial communities. The surface of the lake was characterized by a low salinity water layer while the deepest region was extremely saline (up to 300 g/L salinity). Many parameters (e.g. photosynthetically active radiation, dissolved oxygen concentration, pH, redox potential) changed dramatically from 2 to 4 m below the water surface in conjunction with the increasing salinity values. The water temperature reached a maximum at this depth. At around 3 m depth, there was a water layer with high (bacterio) chlorophyll content dominated by Prosthecochloris vibrioformis, a phototrophic green sulfur bacterium. Characteristic microbial communities with various prokaryotic taxa were identified along the different environmental parameters present in the different water layers. Some of these bacteria were known to be heterotrophic and therefore may be involved in the decomposition of lake organic material (e.g. Halomonas, Idiomarina and Pseudoalteromonas) while others in the transformation of sulfur compounds (e.g. Prosthecochloris). Eukaryotic microorganisms identified by molecular methods in the lake water belonged to genera of green algae (Mantionella and Picochlorum), and were restricted mainly to the upper layers.

Biofilm containing fix-bed systems are frequently applied for intensifying biological treatment in wastewater purification. In these operational units a lot of different biofilm carriers have been used and tested; among them, however, the polymer fiber-based materials are barely investigated. The authors studied four carriers: three types of polypropylene (PP) fibers and a polyester (PES) fiber were compared relating their physical–chemical characteristics and biofilm colonization properties. For the physical–chemical characterization surfactants release, wettability, methylene blue bromophenol blue and protein (bovine serum albumin) adsorption were measured as well as the surface charges of the biofilm carriers were determined by zeta potential examinations. Colonization experiments were carried out in a pilot plant scale biological municipal wastewater treatment system consisting of eight cascade reactors for 18 days. Each of the four tested fibers were placed into the reactors, and biofilm colonization was followed by measuring the dry biomass content and TTC (triphenyl tetrazolium chloride) enzyme activity of the biofilms adhered to the polymer fibers. During the colonization experiments pH, chemical oxygen demand (COD), concentration of total suspended solids (TSS), various nitrogenous compounds and dissolved oxygen were also monitored in the water phase of the reactors. Among the four different fibers one of the PP carriers turned to be the most favorable in colonization tests. This carrier, having surface and wetting properties differing from the other ones colonized the maximum biofilm mass, and the biofilm grown on this fiber had also the highest total enzyme activity all along the purification process, both in organic material degrading and in nitrifying reactors.

article i nfo Investigations were performed along the Danube river at Budapest (Hungary) by collecting water and sediment samples simultaneously for 1 year in order to clarify the possible hazard of selected acidic pharmaceuticals (ibuprofen, naproxen, ketoprofen, and diclofenac) on the water supply used for the production of drinking water by bank filtration. In the case of water samples, the sample preparation procedure included solid phase extraction (SPE), meanwhile, in the case of sediment samples, microwave- assisted extraction (MAE) followed by dispersive matrix extraction (DME) for pre-cleaning as well as SPE for enrichment. The quantification was carried out using gas chromatography-mass spectrometry (GC-MS). The calculated recoveries were 97-99% (±7%) for the water and 95-103% (±12%) for the sediment samples. In the river water, ketoprofen concentration was always below the limit of quantification (LOQ) level; ibuprofen, naproxen and diclofenac could be quantified in the range of 8-50, 2-30, 7-90 ng/L. In sediments, only naproxen and diclofenac were found in the range of 2-20 and 5-38 ng/g, respectively. According to the obtained results, the concentration ratios of the two phases linearly depended on the total organic carbon content (TOC) of the sediments at each sampling date. The linear regressions were 0.925 and 0.946 for naproxen and diclofenac, respectively. © 2010 Published by Elsevier B.V.

In this study, scanning electron microscopy (SEM) and 16S rRNA gene-based phylogenetic approach were applied to reveal the morphological structure and genetic diversity of thermophilic prokaryotic communities of a thermal karst well located in Budapest (Hungary). Bacterial and archaeal diversity of the well water (73.7 °C) and the biofilm developed on the inner surface of an outflow pipeline of the well were studied by molecular cloning method. According to the SEM images calcium carbonate minerals serve as a surface for colonization of bacterial aggregates. The vast majority of the bacterial and archaeal clones showed the highest sequence similarities to chemolithoautotrophic species. The bacterial clone libraries were dominated by sulfur oxidizer Thiobacillus (Betaproteobacteria) in the water and Sulfurihydrogenibium (Aquificae) in the biofilm. A relatively high proportion of molecular clones represented genera Thermus and Bellilinea in the biofilm library. The most abundant phylotypes both in water and biofilm archaeal clone libraries were closely related to thermophilic ammonia oxidizer Nitrosocaldus and Nitrososphaera but phylotypes belonging to methanogens were also detected. The results show that in addition to the bacterial sulfur and hydrogen oxidation, mainly archaeal ammonia oxidation may play a decisive role in the studied thermal karst system.

Lake Red is one of the saline lakes which were formed as a consequence of salt massif dissolution at the foot of the Gurghiu Mountains (Central Romania) at the end of the nineteenth century. The lake water had approximately 15 % w/v salt content. Phylogenetic diversity of prokaryotes inhabiting the water and sediment of the lake was studied using cultivation and cultivation-independent methods following a sampling in spring 2009. According to the results of 16S rRNA gene-based denaturing gradient gel electrophoresis (DGGE), the richness of Bacteria was higher than Archaea on the basis of the number and position of dominant bands in the gel. Sequences from DGGE bands were affiliated with Gammaproteobacteria (Halomonas and Alkalilimnicola) and Bacteroidetes (Psychroflexus) as well as Euryarchaeota. Cultivation from five different saline media resulted in 101 bacterial strains of which Gammaproteobacteria (Halomonas, Marinobacter and Salinivibrio) were the most abundant. Firmicutes (Bacillus) and Alphaproteobacteria (Aurantimonas and Roseovarius) were also identified among the isolated strains. The 16S rRNA genes from 82 bacterial and 95 archaeal clones were also phylogenetically analyzed. Bacterial clones were related to various genera of Gammaproteobacteria (Alkalilimnicola, Alkalispirillum, Arhodomonas, Halomonas, Saccharospirillum), Bacteroidetes (Gracilimonas, Psychroflexus) and Alphaproteobacteria (Oceanicola, Roseinatronobacter, Roseovarius). All of the archaeal clones sequenced corresponded to a homologous cluster affiliated with Halopelagius.

In April 2014, dual bloom of green algae and purple bacteria occurred in a shallow, alkaline soda pan (Kiskunság National Park, Hungary). The water was only 5 cm deep, in which an upper green layer was clearly separated from a near-sediment purple one. Based on microscopy and DNA-based identification, the upper was inhabited by a dense population of the planktonic green alga, Oocystis submarina Lagerheim, while the deeper layer was formed by purple, bacteriochlorophyll-containing bacteria, predominated by Thiorhodospira and Rhodobaca. Additional bacterial taxa with a presumed capability of anoxygenic phototrophic growth belonged to the genera Loktanella and Porphyrobacter. Comparing the bacterial community of the purple layer with a former blooming event in a nearby soda pan, similar functional but different taxonomic composition was revealed. Members from many dominant bacterial groups were successfully cultivated including potentially new species, which could be the result of the application of newly designed media.
