Aging involves complex processes that impact the structure, function, and metabolism of the human brain. Declines in both structural and functional integrity along with reduced local inhibitory tone in the motor areas, as indicated by reduced γ-aminobutyric acid (GABA) levels, are often associated with compromised motor performance in elderly adults. Using multimodal neuroimaging techniques including magnetic resonance spectroscopy (MRS), diffusion magnetic resonance imaging (MRI), functional MRI as well as transcranial magnetic stimulation to assess short-interval intracortical inhibition (SICI), this study explores whether these age-related changes can be mitigated by motor learning. The investigation focused on the effects of long-term balance learning (3 months) on intracortical inhibition, metabolism, structural, and functional connectivity in the cortical sensorimotor network among an elderly cohort. We found that after 3 months of balance learning, subjects significantly improved balance performance, upregulated sensorimotor cortical GABA levels and ventral sensorimotor network functional connectivity (VSN-FC) compared to a passive control group. Furthermore, correlation analysis suggested a positive association between baseline VSN-FC and balance performance, between baseline VSN-FC and SICI, and between improvements in balance performance and upregulation in SICI in the training group, though these correlations did not survive the false discovery rate correction. These findings demonstrate that balance learning has the potential to counteract aging-related decline in functional connectivity and cortical inhibition on the "tonic" (MRS) and "functional" (SICI) level and shed new light on the close interplay between the GABAergic system, functional connectivity, and behavior., (© 2024 The Author(s). Objective: The breathing technique is a determining factor for the sound quality of the singers' vocal production and thus crucial for the choral sound. The aim of this study was to examine choral singers' awareness of breathing techniques and habits while singing and to identify potential correlations between the breathing technique and the preparatory gestures of the conductor., Methods: In our prospective, quasi-experimental study, 720 choral singers answered a 25-item anonymous online survey which focused on demographic information, musical and choral experience, information on choir (size, type, repertoire) and conductor (age, gender, level of expertise). As the central issue we examined the subjects' general knowledge of breathing techniques and habits and their own use thereof in choral practice. Finally, subjects were asked to watch videos showing two different preparatory conducting gestures and to assess the meaning of these gestures., Results: The results of the study show that the majority of choral singers (68.1%) inhale not before and not after, but during the conductor's preparatory gesture. The most well-known (94.7%) and allegedly most frequently (81.8%) used type of inhalation is abdominal breathing. Six times more singers associate abdominal breathing with an outward gesture rather than with the more frequently occurring inward-upward gesture., Conclusion: The conducting literature does not discriminate between out- and inward preparatory gestures and describe them as equivalent. However, according to our data, a vast majority of singers assign these gestures to different types of inhalation which influences singing technique and sound quality. ABSTRACT The purpose of this study was to investigate the effect of a supplementation with specific collagen peptides (SCP) combined with resistance training (RT) on changes in structural properties of the patellar tendon. Furthermore, tendon stiffness as well as maximal voluntary knee extension strength and cross-sectional area (CSA) of the rectus femoris muscle were assessed. In a randomized, placebo-controlled study, 50 healthy, moderately active male participants completed a 14-week resistance training program with three weekly sessions (70-85% of 1 repetition maximum [1RM]) for the knee extensors. While the SCP group received 5g of specific collagen peptides daily, the other group received the same amount of a placebo (PLA) supplement. The SCP supplementation led to a significant greater ( p  < 0.05) increase in patellar tendon CSA compared with the PLA group at 60% and 70% of the patellar tendon length starting from the proximal insertion. Both groups increased tendon stiffness ( p  < 0.01), muscle CSA ( p  < 0.05) and muscular strength ( p  < 0.001) throughout the intervention without significant differences between the groups. The current study shows that in healthy, moderately active men, supplementation of SCP in combination with RT leads to greater increase in patellar tendon CSA than RT alone. Since underlying mechanisms of tendon hypertrophy are currently unknown, further studies should investigate potential mechanisms causing the increased morphology adaptions following SCP supplementation. Purpose: Recent evidence indicates that low-load blood flow restriction (LL-BFR) training elicits an anabolic response in tendinous tissue. The purpose of the present study was to investigate the hypertrophic pattern induced in the Achilles tendon by LL-BFR, in comparison with the regional hypertrophy typically observed with conventional high-load (HL) resistance training., Methods: N = 40 male participants were randomly and concealed allocated to one of two groups: LL-BFR training (20-35% one-repetition maximum/1RM) or HL training (70-85% 1RM). The training was completed three times per week for a total of 14 weeks. Before and after the training period, Achilles tendon morphology was assessed using magnetic resonance imaging along the entire tendon length. Additionally, dynamic strength measures of the plantar flexors were evaluated., Results: In line with previous findings, dynamic plantar flexion strength was improved to a comparable extent in both groups (LL-BFR: 43.6%; HL: 43.5%). The results also confirmed significant increases in Achilles tendon cross-sectional area with LL-BFR (+5.2%). Moreover, they revealed that the hypertrophic pattern obtained with LL-BFR was similar to regional changes seen with conventional HL training., Conclusion: The present findings point towards the notion that despite the low loads being applied, LL-BFR training induces Achilles tendon hypertrophy by potentiating anabolic responses in the same regions as with conventional high-load training. Future studies are needed to (i) focus on the potential mechanisms underlying these tendon morphology changes and (ii) apply and evaluate LL-BFR training in clinical populations to validate these results in rehabilitative settings., (© 2023 The Authors. During movements, neural signals are translated into muscle fibre shortening, lengthening or they remain isometric. This study investigated cortical excitatory and inhibitory processes in relation to muscle fascicle dynamics during fixed-end rapid contractions. Fourteen adults performed submaximal and maximal ankle dorsiflexions. Single and paired pulse transcranial magnetic stimulation over the cortical representation projecting to the tibialis anterior (TA) was applied during rest, the activation and deactivation phase of contractions to test for short- (SICI) and long-interval intracortical inhibition (LICI) and intracortical facilitation (ICF). Ultrasound images were taken to measure muscle fascicle dynamics of the superficial (TA SF ) and deep (TA DP ) TA compartments. The results show significantly greater maximal shortening velocities (p = 0.003, d = 0.26, CI [4.89, 18.52]) and greater maximal fascicle shortening (p = 0.003, d = 0.86, CI [0.29, 3.13]) in TA SF than TA DP during submaximal dorsiflexions. Significantly lower SICI levels during activation compared to deactivation (p = 0.019, d = 1.12, CI [19.82, 1.76]) and at rest (p < 0.0001) were observed. ICF was significantly greater during activation (p = 0.03) than during rest while LICI did not modulate significantly. Maximal TA SF but not TA DP shortening velocity correlated with SICI levels at activation (p = 0.06) and with the rate of torque development (p = 0.02). Objective: The breathing technique is a determining factor for the singer's sound quality and consequently crucial for the choral sound. However, very little is known about possible influences of the conductor's preparatory gesture on the way choral singers inhale before the beginning of a piece (respectively every subsequent phrase). The conducting literature does not discriminate between out- and inward preparatory gestures and even describes them as equivalent, but previous studies suggest that singers assign different types of inhalation to different preparatory gestures. It may therefore be assumed that the type of preparatory gesture has a direct influence on the singer's inhalation and tone production, and the aim of this study is hence to examine possible effects of two contrasting preparatory gestures on the singer's inhalation type and the resulting tone quality., Methods: In our within-subjects study design, 18 healthy choral singers (9 male/ 9 female) were recruited to participate in a laboratory experiment. The participants were asked to sing a tone suitable for their voice register in response to different video stimuli. These consisted of two conducting-videos, each showing a different preparatory gesture, and two control conditions with an animated bar and an arrow indicating the desired breathing type. The singers reacted to 10 sets of videos, each set consisting of the four stimuli in randomized order. For evaluation of the breathing behavior and vocal output during the different experimental conditions, chest wall kinematics of upper rib cage, abdominal rib cage and abdomen were measured via 3D motion capture and voice samples were recorded. The obtained data were filtered and compared using the repeated measures analysis of variance and post hoc Tukey test for significant results. The level of significance was set at P < 0.05., Results: The results of the study show significant differences in volume of the abdomen between the two different gestures (F 1,17  = 24.04, η 2  = 0.59, P = 0.0001), which can be validated by the two control measurements (F 1,17  = 21.12, η 2  = 0.55, P = 0.0002). An outward preparatory gesture evoked an abdominal breathing type while an inward-upward movement led to an inhalation with a higher portion of clavicular breathing. Furthermore, significant differences in timbre and loudness of the produced tone could be observed. The maximum sound pressure level of the outward preparatory gesture was significantly higher than in case of the inward-upward movement (F 1,17  = 20.4, η 2  = 0.56, P = 0.0004)., Conclusions: In contrast to the existing conducting literature, which does not discriminate between out- and inward preparatory gestures, the results of this study show that the conductor's choice of trajectory direction and form of the preparatory gestures elicit spontaneous, gesture-specific reactions in singers' breathing behavior as well as the corresponding loudness and sound quality., (Copyright © 2022. The transcriptome of human immune cells rapidly reacts to altered gravity in a highly dynamic way. We could show in previous experiments that transcriptional patterns show profound adaption after seconds to minutes of altered gravity. To gain further insight into these transcriptional alteration and adaption dynamics, we conducted a highly standardized RNA-Seq experiment with human Jurkat T cells exposed to 9xg hypergravity for 3 and 15 min, respectively. We investigated the frequency with which individual exons were used during transcription and discovered that differential exon usage broadly appeared after 3 min and became less pronounced after 15 min. Additionally, we observed a shift in the transcript pool from coding towards non-coding transcripts. Thus, adaption of gravity-sensitive differentially expressed genes followed a dynamic transcriptional rebound effect. The general dynamics were compatible with previous studies on the transcriptional effects of short hypergravity on human immune cells and suggest that initial up-regulatory changes mostly result from increased elongation rates. The shift correlated with a general downregulation of the affected genes. All chromosome bands carried homogenous numbers of gravity-sensitive genes but showed a specific tendency towards up- or downregulation. Altered gravity affected transcriptional regulation throughout the entire genome, whereby the direction of differential expression was strongly dependent on the structural location in the genome. A correlation analysis with potential mediators of the early transcriptional response identified a link between initially upregulated genes with certain transcription factors. The purpose of this study was to investigate the effect of specific collagen peptides (SCP) combined with resistance training (RT) on changes in tendinous and muscular properties. In a randomized, placebo-controlled study, 40 healthy male volunteers (age: 26.3 ± 4.0 years) completed a 14 weeks high-load resistance training program. One group received a daily dosage of 5g SCP while the other group received 5g of a placebo (PLA) supplement. Changes in Achilles tendon cross-sectional area (CSA), tendon stiffness, muscular strength, and thickness of the plantar flexors were measured. The SCP supplementation led to a significantly (p = 0.002) greater increase in tendon CSA (+11.0%) compared with the PLA group (+4.7%). Moreover, the statistical analysis revealed a significantly (p = 0.014) greater increase in muscle thickness in the SCP group (+7.3%) compared with the PLA group (+2.7%). Finally, tendon stiffness and muscle strength increased in both groups, with no statistical difference between the groups. In conclusion, the current study shows that the supplementation of specific collagen peptides combined with RT is associated with a greater hypertrophy in tendinous and muscular structures than RT alone in young physically active men. Introduction: Low-load resistance training with blood flow restriction (LL-BFR) has emerged as a viable alternative to conventional high-load (HL) resistance training regimens. Despite increasing evidence confirming comparable muscle adaptations between LL-BFR and HL resistance exercise, only very little is known about tendinous mechanical and morphological adaptations after LL-BFR. Therefore, the aim of the present study was to examine the effects of 14 wk of LL-BFR and HL training on patellar tendon adaptations., Methods: Twenty-nine recreationally active male participants were randomly allocated into the following two groups: LL-BFR resistance training (20%-35% one-repetition maximum (1RM)) or HL resistance training (70%-85% 1RM). Both groups trained three times per week for 14 wk. One week before and after the intervention, patellar tendon mechanical and morphological properties were assessed via ultrasound and magnetic resonance imaging. In addition, changes in muscle cross-sectional area were quantified by magnetic resonance imaging and muscle strength via dynamic 1RM measurements., Results: The findings demonstrated that both LL-BFR and HL training resulted in comparable changes in patellar tendon stiffness (LL-BFR: +25.2%, P = 0.003; HL: +22.5%, P = 0.024) without significant differences between groups. Similar increases in tendon cross-sectional area were observed in HL and LL-BFR. Muscle mass and strength also significantly increased in both groups but were not statistically different between HL (+38%) and LL-BFR (+34%), except for knee extension 1RM where higher changes were seen in LL-BFR., Conclusions: The present results support the notion that both HL and LL-BFR cause substantial changes in patellar tendon properties, and the magnitude of changes is not significantly different between conditions. Explosive strength and balance training are commonly applied to enhance explosive strength and balance performance. Even though both training methods are frequently implemented, ordering effects have largely been neglected. Therefore, the present study aimed to investigate ordering effects of balance and explosive strength training on explosive strength and balance performance as well as changes in short-interval intracortical inhibition (SICI). Two groups of subjects either participated in 4 weeks of balance training followed by 4 weeks of explosive strength training (BT-ET) or vice versa (ET-BT). Before, after 4 and 8 weeks, balance performance, as well as explosive strength, was tested. Additionally, SICI was tested during rest as well as during balance perturbations and explosive contractions. The results show a training specific increase in performance with an increase in balance control followed by an increase in explosive strength in the BT-ET, while the ET-BT increased its balance and explosive strength in the opposite order. There were no significant ordering effects. Both groups showed a significant decrease in SICI during the explosive contractions after the eight weeks of training. When SICI was tested during the balance perturbations, SICI initially increased after the first 4 weeks of training but returned to baseline until the end of the eight weeks. It is suggested that the decrease in SICI with prolonged training might show a disengagement of the motor cortex during the balance task. Background: Numerous studies have demonstrated that the addition of blood flow restriction (BFR) to low-load (LL) resistance exercise leads to elevated levels of muscle hypertrophy and strength gains. In terms of main underlying mechanisms, metabolic accumulation and increased neuromuscular adaptations seem to play a primary role. However, this evidence is largely based on dynamic exercise conditions. Therefore, the main objective was to investigate the acute physiological adaptations following isometric LL-BFR exercise., Methods: Fifteen males participated in this cross-over trial and completed the following sessions in a random and counterbalanced order: isometric LL-BFR exercise (20% maximum voluntary contraction, MVC) and load matched LL exercise without BFR. Lactate levels, muscle activation as well as muscle swelling were recorded during the whole exercise and until 15 min post completion. Additionally, changes in maximal voluntary torque and ratings of perceived exertion (RPE) were monitored., Results: During exercise, EMG amplitudes (72.5 ± 12.7% vs. 46.3 ± 6.7% of maximal EMG activity), muscle swelling and RPE were significantly higher during LL-BFR compared to LL (p < 0.05). Lactate levels did not show significant group differences during exercise but revealed higher increases 15 min after completion in the LL-BFR condition (LL-BFR: + 69%, LL: + 22%) (p < 0.05). Additionally, MVC torque significantly decreased immediately post exercise only in LL-BFR (~ - 11%) (p < 0.05) but recovered after 15 min., Conclusions: The present results demonstrate that isometric LL-BFR causes increased metabolic, neuromuscular as well as perceptual responses compared to LL alone. These adaptations are similar to dynamic exercise and therefore LL-BFR represents a valuable type of exercise where large joint movements are contraindicated (e.g. rehabilitation after orthopedic injuries).

Even though the take-off in ski jumping is decisive, athletes only have a very limited number of training trials on the actual ski jump to practice under real ski jump conditions. Hence, various imitation jumps aiming to mimic the hill jump are performed during daily training. These imitation jumps should therefore mimic the kinematic pattern of hill jumps appropriately. This study aimed to identify imitation jumps that resemble hill jumps regarding four performance-related biomechanical criteria: maximal vertical take-off velocity, maximal knee extension velocity, maximal forward-directed angular momentum and anterior shift of the center of mass. Therefore, a three-dimensional analysis of the take-off during six different modalities of imitation jumps as well as hill jumps for validation was carried out in nine professional ski jumpers. Imitation jumps from a rolling platform show better agreement than stationary jumps and three out of the four parameters were best resembled via an imitation jump that included ski jumping boots. Thus, non-hill take-off training should be performed with complex imitation jumps to mimic the actual ski jump. Except for the vertical take-off velocity, we could identify one imitation jump type that is not statistically different to the hill. Consequently, the individual deficiencies of the athletes can be addressed and specifically trained using the appropriate imitation jump. These information about the similarity between imitation jumps and real hill jumps are highly relevant for trainers and athletes in order to effectively design their training programs., (© 2020 The Authors. Abstract: Kurz, A, Lauber, B, Franke, S, and Leukel, C. Balance training reduces postural sway and improves sport-specific performance in visually impaired cross-country skiers. J Strength Cond Res 35(1): 247-252, 2021-Balance training is highly effective in reducing sport injuries and causes improvements in postural stability and rapid force production. So far, the positive effects of balance training have been described for healthy athletes. In the present experiments, we questioned whether athletes with disabilities of the visual system can also benefit from balance training. Fourteen visually impaired cross-country skiers participated in this randomized controlled study. The intervention group (N = 7) completed 8 sessions of balance training over a period of 4 weeks (2 times per week), whereas a waiting control group (N = 7) received no training during that time. After training, postural sway was significantly reduced in the intervention group but not in the waiting control group. In addition, sport-specific performance, which was assessed by a standardized Cooper's 12-minute test on roller skis or rollerblades, increased in the intervention group. The change in postural sway from the premeasurement to the postmeasurement correlated with the change in sport-specific performance in all subjects. Our results indicate that balance training is useful for improving postural stability and sport-specific performance in visually impaired cross-country skiers. In contrast to other rhythmic tasks such as running, the preferred movement rate in cycling does not minimize energy consumption. It is possible that neurophysiological mechanisms contribute to the choice of cadence, however this phenomenon is not well understood. Eleven participants cycled at a fixed workload of 125 W and different cadences including a freely chosen cadence (FCC, ∼72), and fixed cadences of 70, 80, 90 and 100 revolutions per minute (rpm) during which transcranial magnetic stimulation (TMS) was used to measure short interval intracortical inhibition (SICI) and intracortical facilitation (ICF). There was a significant increase in SICI at 70 (P = 0.004), 80 (P = 0.008) and 100 rpm (P = 0.041) compared to FCC. ICF was significantly reduced at 70 rpm compared to FCC (P = 0.04). Inhibition-excitation ratio (SICI divided by ICF) declined (P = 0.014) with an increase in cadence. The results demonstrate that SICI is attenuated during FCC compared to fixed cadences. Motor cortical contribution was shown to be important for balance control and for ballistic types of movements. However, little is known about the role of cortical inhibitory mechanisms and even less about long(er)-term adaptations of these inhibitory processes. Therefore, the aim of the present study was to investigate the role of intracortical inhibition before and after four weeks of explosive or balance training. Two groups of subjects participated for four weeks either in an explosive training programme of the plantar flexor muscles or in a balance training programme on unstable devices. Adaptations in short-interval intracortical inhibition (SICI) were assessed by applying paired-pulse TMS to the soleus muscle during dynamic plantar flexions, balance perturbations and at rest. Furthermore, SICI was assessed for the untrained tibialis anterior muscle. The results show task-, muscle- and group-specific adaptations in SICI after the training (p = .021) with significantly increased SICI after balance training in the balance task and decreased SICI after explosive training in the ballistic task. The training also caused task- and group-specific behavioural adaptations indicated by improved balance performance after balance training and increased ballistic performance after explosive training. There were no changes in SICI when measured at rest or in the untrained tibialis anterior muscle. Cellular processes are influenced in many ways by changes in gravitational force. In previous studies, we were able to demonstrate, in various cellular systems and research platforms that reactions and adaptation processes occur very rapidly after the onset of altered gravity. In this study we systematically compared differentially expressed gene transcript clusters (TCs) in human Jurkat T cells in microgravity provided by a suborbital ballistic rocket with vector-averaged gravity (vag) provided by a 2D clinostat. Additionally, we included 9× g centrifuge experiments and rigorous controls for excluding other factors of influence than gravity. We found that 11 TCs were significantly altered in 5 min of flight-induced and vector-averaged gravity. Among the annotated clusters were G3BP1 , KPNB1 , NUDT3 , SFT2D2 , and POMK . Our results revealed that less than 1% of all examined TCs show the same response in vag and flight-induced microgravity, while 38% of differentially regulated TCs identified during the hypergravity phase of the suborbital ballistic rocket flight could be verified with a 9× g ground centrifuge. In the 2D clinostat system, doing one full rotation per second, vector effects of the gravitational force are only nullified if the sensing mechanism requires 1 s or longer. Due to the fact that vag with an integration period of 1 s was not able to reproduce the results obtained in flight-induced microgravity, we conclude that the initial trigger of gene expression response to microgravity requires less than 1 s reaction time. Additionally, we discovered extensive gene expression differences caused by simple handling of the cell suspension in control experiments, which underlines the need for rigorous standardization regarding mechanical forces during cell culture experiments in general., Competing Interests: The authors declare no conflict of interest.

This study examined the effect of drop height on neuromechanical control of the plantarflexors in drop jumps (DJs) before and during ground contact (GC). The effect of anticipation on muscle mechanical configurations was investigated in 22 subjects in three conditions (20, 30, and 40 cm): (i) known, (ii) unknown, or (iii) cheat falling heights (announced 40 cm, but actual drop height was 20 cm). Electromyographic (EMG) activity of the m. gastrocnemius medialis (GM) and other shank muscles was recorded and analyzed before GC and during GC separately for the short-, medium-, and long-latency responses (SLR, MLR, and LLR). Changes in GM fascicle length (L M ) were determined via B-mode ultrasound, and muscle-tendon unit length (L MTU ) was estimated. Peak force (P < .001), rate of force development (RFD) (P = .001) and GM EMG activity prior to (P = .003) and during GC (P = .007) was reduced in the unknown compared with the known conditions (P < .05). The amount of shortening in L MTU during GC in unknown and cheat was less compared with the known conditions (P = .005; P = .049). Changes in L MTU lengthening negatively correlated with changes in GM activity around SLR and MLR (P = .006; P = .02) in known and unknown conditions. Taken together, it seems that the central nervous system applies a protective strategy in the unknown condition by reducing muscle activity to result in a lower muscular stiffness and increased tendinous lengthening prior to and during GC. This might be a mechanism to absorb greater elastic energy in the tendon and reduce the magnitude and rate of muscle lengthening and subsequent stretch-induced muscle damage., (© 2019 John Wiley & Sons A/S.
