Background Understanding the variation in vector-borne disease transmission intensity across time and space relies on a thorough understanding of the impact of environmental factors on vectorial capacity traits of mosquito populations. This is driven primarily by variation in larval development and growth, with carryover effects influencing adult traits such as longevity and adult body size. The relationship between body size and longevity strongly affects the evolution of life histories and the epidemiology of vector-borne diseases. This relationship ranges from positive to negative but the reasons for this variability are not clear. Both traits depend on a number of environmental factors, but primarily on temperature as well as availability of nutritional resources. We therefore asked how the larval environment of the mosquito Anopheles gambiae Giles (sensu stricto) (Diptera: Culicidae) affects the relationship between body size and longevity. Methods We reared the larvae of An. gambiae individually at three temperatures (21, 25 and 29 °C) and two food levels (the standard and 50% of our laboratory diet) and measured adult size and longevity. We estimated the direct and indirect (via adult size) effects of food and temperature on longevity with a piecewise structural equation model (SEM). Results We confirmed the direct effects of food and temperature during larval development on body size, as wing length decreased with increasing temperature and decreasing food levels. While the overall relationship between size and longevity was weak, we measured striking differences among environments. At 25 °C there was no clear relationship between size and longevity; at 29 °C the association was negative with standard food but positive with low food; whereas at 21 °C it was positive with standard food but negative with low food. Conclusions The larval environment influences the adult’s fitness in complex ways with larger mosquitoes living longer in some environments but not in others. This confirmed our hypothesis that the relationship between size and longevity is not limited to a positive correlation. A better understanding of this relationship and its mechanisms may improve the modelling of the transmission of vector borne diseases, the evolution of life history traits, and the influence of vector control. The activity of the Dpp morphogen adapts to tissue size in the growing Drosophila wing imaginal disc, and Pentagone, an important secreted feedback regulator of the Dpp pathway, is required for this adaptation., The wing of the fruit fly, Drosophila melanogaster, with its simple, two-dimensional structure, is a model organ well suited for a systems biology approach. The wing arises from an epithelial sac referred to as the wing imaginal disc, which undergoes a phase of massive growth and concomitant patterning during larval stages. The Decapentaplegic (Dpp) morphogen plays a central role in wing formation with its ability to co-coordinately regulate patterning and growth. Here, we asked whether the Dpp signaling activity scales, i.e. expands proportionally, with the growing wing imaginal disc. Using new methods for spatial and temporal quantification of Dpp activity and its scaling properties, we found that the Dpp response scales with the size of the growing tissue. Notably, scaling is not perfect at all positions in the field and the scaling of target gene domains is ensured specifically where they define vein positions. We also found that the target gene domains are not defined at constant concentration thresholds of the downstream Dpp activity gradients P-Mad and Brinker. Most interestingly, Pentagone, an important secreted feedback regulator of the pathway, plays a central role in scaling and acts as an expander of the Dpp gradient during disc growth., Author Summary Scaling, the fitting of pattern to size, manifests itself in numerous examples around us. During development, individual body parts scale up to fit the overall body size. Starved animals form smaller adults with proportionally smaller parts, and amphibian embryos can form normally proportioned adults after extreme surgical operations. How scaling is achieved is not well understood. Here, we establish the Drosophila wing imaginal disc, the precursor tissue of the adult wing, as a model to study scaling quantitatively during growth. In this model, we define scaling as the preservation of proportions of gene expression domains with tissue size during disc growth. The Decapentaplegic (Dpp) morphogen is known to play a central role in Drosophila wing formation and co-coordinately regulates growth and patterning. We found that as the disc grows, the Dpp response expands and scales with the tissue size. Interestingly, scaling is not perfect at all positions in the field. The scaling of the target gene domains is best where they have a function; Spalt, for example, scales best at the position in the anterior compartment where it helps to form one of the anterior veins of the wing. Analysis of mutants for pentagone, a transcriptional target of Dpp that encodes a secreted feedback regulator of the pathway, indicates that Pentagone plays a key role in scaling the Dpp gradient activity.

O objetivo do estudo foi obter dados que pudessem sugerir desenvolvimento de resistência dos mosquitos à cafeína. MÉTODOS: Foi avaliada a produção de adultos em gerações sucessivas, a partir de ovos produzidos na geração anterior e a taxa de oviposição em cada geração, utilizando meios contendo cafeína a 200 e 500 µg/ml e água de torneira proveniente de poço artesiano como controle. Os experimentos foram conduzidos em São José do Rio Preto, entre 2002 e 2005. Nos testes estatísticos foram utilizados a análise exploratória de dados e algoritmos de alisamento. RESULTADOS: Ocorreu redução crescente da produção de adultos, nas duas concentrações, ao longo das gerações, mas apenas no experimento a 200 µg/ml os dados foram estatisticamente significantes. Quanto à oviposição, a análise dos números mostra redução crescente e acentuada na média de ovos por fêmea, no experimento tratado. CONCLUSÕES: Não houve evidência de resistência ao longo das gerações devido ao tratamento com cafeína. Os resultados encontrados podem reforçar a indicação da cafeína como uma alternativa aos principais agentes de controle do Ae. aegypti atualmente usados, contra os quais os mosquitos têm desenvolvido resistência. OBJECTIVE: Previous experiments showed that caffeine blocks the development of Aedes aegypti (Diptera, Culicidae) in the larval stage, consequently inhibiting the production of adults. This study aimed at obtaining data suggestive of caffeine resistance by these mosquitoes. METHODS: Experiments were carried out in successive generations to assess adult production from eggs laid in previous generation and oviposition rate in every generation using 200 and 500 µg/mL caffeine. Tap water was used as control. Experiments were conducted in the city of São José do Rio Preto, Southeastern Brazil between 2002 and 2005. Statistical tests consisted of exploratory data analysis and smoothing algorithms. RESULTS: Increasing reduction in productivity of adults occurred among generations at both caffeine concentrations but the differences were only significant at 200µg/mL caffeine. As for the oviposition rate, there was a decrease in the mean number of eggs per female over generations at both caffeine concentrations. CONCLUSIONS: There was no evidence of caffeine resistance over generations. The study results corroborate caffeine as an alternative as an important Ae. Aegypti control agent to avoid resistance. 0092-8674 (Print) Journal Article Research Support, Non-U.S. Gov't; A critical determinant of insect body size is the time at which the larva stops feeding and initiates wandering in preparation for metamorphosis. No genes have been identified that regulate growth by contributing to this key developmental decision to terminate feeding. We show here that mutations in the DHR4 orphan nuclear receptor result in larvae that precociously leave the food to form premature prepupae, resulting in abbreviated larval development that translates directly into smaller and lighter animals. In addition, we show that DHR4 plays a central role in the genetic cascades triggered by the steroid hormone ecdysone at the onset of metamorphosis, acting as both a repressor of the early ecdysone-induced regulatory genes and an inducer of the betaFTZ-F1 midprepupal competence factor. We propose that DHR4 coordinates growth and maturation in Drosophila by mediating endocrine responses to the attainment of critical weight during larval development.

The recursion formula of the Gompertz function is an established method for the analysis of growth processes. In the present study the recursion formula of the Gompertz survival function 1n S(t + s) = a + b x ln S(t) is introduced for the analysis of survival data, where S(t) is the survival fraction at age 1, s is the constant age increment between two consecutive measurements of the survival fraction and a and b are parameters. With the help of this method--and provided stroboscopial measurements of rates of survival are available--the Gompertz survival function, instead of the corresponding mortality function, can be determined directly using linear regression analysis. The application of the present algorithm is demonstrated by analysing two sets of data taken from the literature (survival of Drosophila imagoes and of female centenarians) using linear regression analysis to fit survival or mortality rates to the corresponding models. In both cases the quality of fit was superior by using the algorithm presently introduced. Moreover, survival functions calculated from the fits to the mortality law only poorly predict the survival data. On the contrary, the results of the present method not only fit to the measurements, but, for both sets of data the mortality parameters calculated by the present method are essentially identical to those obtained by a corresponding application of a non-linear Marquardt-Levenberg algorithm to fit the same sets of data to the explicit form of the Gompertz survival function. Taking into consideration the advantages of using a linear fit (goodness-of-fit test and efficient statistical comparison of survival patterns) the method of the recursion formula of the Gompertz survival function is the most preferable method to fit survival data to the Gompertz function. The TGF-β homolog Decapentaplegic (Dpp) acts as a secreted morphogen in the Drosophila wing disc, and spreads through the target tissue in order to form a long range concentration gradient. Despite extensive studies, the mechanism by which the Dpp gradient is formed remains controversial. Two opposing mechanisms have been proposed: receptor-mediated transcytosis (RMT) and restricted extracellular diffusion (RED). In these scenarios the receptor for Dpp plays different roles. In the RMT model it is essential for endocytosis, re-secretion, and thus transport of Dpp, whereas in the RED model it merely modulates Dpp distribution by binding it at the cell surface for internalization and subsequent degradation. Here we analyzed the effect of receptor mutant clones on the Dpp profile in quantitative mathematical models representing transport by either RMT or RED. We then, using novel genetic tools, experimentally monitored the actual Dpp gradient in wing discs containing receptor gain-of-function and loss-of-function clones. Gain-of-function clones reveal that Dpp binds in vivo strongly to the type I receptor Thick veins, but not to the type II receptor Punt. Importantly, results with the loss-of-function clones then refute the RMT model for Dpp gradient formation, while supporting the RED model in which the majority of Dpp is not bound to Thick veins. Together our results show that receptor-mediated transcytosis cannot account for Dpp gradient formation, and support restricted extracellular diffusion as the main mechanism for Dpp dispersal. The properties of this mechanism, in which only a minority of Dpp is receptor-bound, may facilitate long-range distribution., Author Summary Morphogens are signaling molecules that trigger specific responses in cells in a concentration-dependent manner. The formation of morphogen gradients is essential for the patterning of tissues and organs. Decapentaplegic (Dpp) is the Drosophila homolog of the bone morphogenic proteins in vertebrates and forms a morphogen gradient along the anterior-posterior axis of the Drosophila wing imaginal disc, a single-cell layered epithelium. Dpp determines the growth and final size of the wing disc and serves as an ideal model system to study gradient formation. Despite extensive studies the mechanism by which morphogen gradients are established remains controversial. In the case of Dpp two mechanisms have been postulated, namely extracellular diffusion and receptor-mediated transcytosis. In the first model Dpp is suggested to move by diffusion through the extracellular matrix of a tissue, whereas in the latter model Dpp is transported through the cells by receptor-mediated uptake and re-secretion. In this work we combined novel genetic tools with mathematical modeling to discriminate between the two models. Our results suggest that the Dpp gradient forms following the extracellular diffusion mechanism. Moreover, our data suggest that the majority of the extracellular Dpp is free and not bound to its receptor, a property likely to play a role for the long-range gradient formation.

SummaryCaste differentiation and division of labor are the hallmarks of social insect colonies [1, 2]. The current dogma for female caste differentiation is that female eggs are totipotent, with morphological and physiological differences between queens and workers stemming from a developmental switch during the larval stage controlled by nutritional and other environmental factors (e.g., [3–8]). In this study, we tested whether maternal effects influence caste differentiation in Pogonomyrmex harvester ants. By conducting crossfostering experiments we identified two key factors in the process of caste determination. New queens were produced only from eggs laid by queens exposed to cold. Moreover, there was a strong age effect, with development into queens occurring only in eggs laid by queens that were at least two years old. Biochemical analyses further revealed that the level of ecdysteroids was significantly lower in eggs developing into queens than workers. By contrast, we found no significant effect of colony size or worker exposure to cold, suggesting that the trigger for caste differentiation may be independent of the quantity and quality of resources provided to larvae. Altogether these data demonstrate that the developmental fate of female brood is strongly influenced by maternal effects in ants of the genus Pogonomyrmex.
