Abstract

The sequences required for full repression of the Escherichia coli deoP1 and P2 promoters by the deoR repressor (DeoR) have been analyzed in vivo. Using recombinant techniques, we have constructed a set of deo‐lacZ fusions which contain different parts of the sequences involved in the regulation of deo expression on low copy number fusion vectors. Since these vectors are present in only one copy per chromosome at temperatures below 37 degrees C, this vector system allows very accurate studies of gene control signals. Our results show that three DeoR operator sites exist in the deoP1‐P2 regulatory region. Two of these loci overlap the initiation sites for deoP1 (O1) and deoP2 (O2) transcription located 599 bp apart, whereas the third site (OE) is present approximately 270 bp upstream of P1. DeoR repression of both P1 and P2 transcription is weak on promoter fragments which only contain one operator site (O1 or O2). Enhanced repression by deoR is observed on promoter fragments containing two operator sites. However, all three sites are needed for full repression. These findings are discussed with respect to upstream and downstream control regions of eukaryotic genes.

Abstract

This publication updates some of the tables presented in the original volumes. It provides historical statistics relating to the world aluminium industry and covers the areas of production, consumption, trade and prices, for bauxite, alumina and aluminium. Forecasts are also given for future world productive capacity. Although in general the original text did not warrant a major revision, an overview of the world aluminium industry has been included in this issue which discusses significant developments in the past few years and summarises the outlook for the immediate future. 26. Region and Field Level Distributions of Aster Yellows Phytoplasma in Small Grain Crops.

Abstract

Aster yellows (AY), a disease of small grain crops caused by aster yellows phytoplasma (AYp), produces disease symptoms similar to barley yellow dwarf (BYD). From 2003 to 2005, small grain production fields in Minnesota and North Dakota were surveyed to determine the incidences of AY and BYD. In-field spatial patterns of AY-infected plants also were investigated. Plants collected along a five-point transect line were tested for AYp using nested polymerase chain reaction (PCR) and quantitative real-time PCR assays, and extracted plant sap was tested for serotypes PAV and RPV of Barley yellow dwarf virus (BYDV) using enzyme-linked immunosorbent assays. During 2003, 2004, and 2005, AYp was detected in plants from 49, 15, and 7% of tested fields, respectively, whereas BYDV was found in plants from 2, 0, and 5% of fields, respectively. Average amplicon count number indicated an in-field spatial trend for greater incidence of AYp and increased populations of AYp in plants located near field edges, with comparably low copy numbers at transect point locations toward the direction of field center. AY is likely a common but largely undetected disease on small grain crops in the Upper Midwest. [ABSTRACT FROM AUTHOR]
