The conformational analysis of nine functionalized 1,2,3-triazoles was carried out by the correlation of calculated and experimental high-level nuclear magnetic resonance (NMR) chemical shifts. In solution, the studied triazoles are in exchange dynamic equilibrium caused by their prototropic tautomerism of the NH-proton. The experimentally unresolved NMR signals were assigned for most of the compounds. A more thorough survey was conducted for 4- t -butyl-1,2,3-triazole-5-carbaldehyde oxime. The analysis performed within the framework of the DP4+ formalism completely confirmed the hypothesis of the predominance of the 2H-tautomer. Thus, the methodology for estimating stereochemical structures in the absence of some experimental data allowed the most stable conformations for dynamic systems with different tautomeric ratios to be reliably identified.

Substituted acrylamides have found an extensive application in organic and medical chemistry; therefore, it is very important to get insight into their features such as electronic structure, spectral properties, and stereochemical transformations. A correct interpretation of the chemical behavior and biological activity of these heteroatomic systems is impossible without knowledge of the structure of stereodynamic forms and factors determining their relative stability. The structure and peculiarities of stereodynamic behavior of substituted acrylamides and their model compounds were studied by dynamic and multinuclear 1 H, 13 C, and 15 N nuclear magnetic resonance (NMR) spectroscopy in CDCl 3 and DMSO-d 6 solution. It has been established that acrylamides in solution are realized as Z- and E-isomers, with the E-rotamer being somewhat predominant. A family of novel pharmacologically prospective carbohydrate polymer aminomethylpropargyl derivatives has been synthesized by CuI-catalyzed A 3 -coupling reaction of arabinogalactan propargyl ethers with aldehydes and secondary amines in DMSO at 23-65 °С. The arabinogalactan derivatives bearing phenyl, pyridinе, and furan moieties in combination with piperidyne, pyrrolidyne, morpholine fragments are prepared in up to 95 % yield and 0.3-2.0 degree of substitution. Conversion of the propargyl groups of the starting arabinogalactan ethers reaches in most cases 80-100 %. The non-catalyzed aminoalkylation (96-100 °С, degree of substitution up to 1.1) of propargylated AG proceeds both at the ethynyl and hydroxy centers to afford mainly the products of C-substitution. New salicylate derivatives of arabinogalactan have been synthesized in up to 90% yield by cross-coupling of propargyl ethers of arabinogalactan with 5-bromosalicylic acid salts. The reaction proceeds in the presence of the catalytic system Pd(PPh 3 ) 4 /PPh 3 with CuBr (CuI), and piperidine or 1,8-diazabicyclo-[5.4.0]-undec-7-ene (DBU) as a base in DMSO at 70-95 °С. The propargyl groups of the starting arabinogalactan ethers are converted into propinylsalicylate ones with 35 and 50% conversion depending on the nature of the base used. Degree of arabinogalactan substitution with propynyl salicylate fragments reaches 1.0 and 0.7 in the reactions with piperidine and DBU, respectively. In the case of piperidine, along with the main process, intensive unprecedented (for Sonogashira reaction) hydroamination of propargyl groups is also observed, followed by hydrolysis of unstable enamines. The products have been characterized by IR, 1 Н and 13 С NMR, UV spectroscopy as well as elemental analysis and gel permeation chromatography. Salicylate derivatives of arabinogalactan show high activity in prolonging the activated partial thromboplastin time (APTT), whereas neither the arabinogalactan nor propargyl ethers of arabinogalactan nor salicylic acid salts have obvious effects., (Copyright © 2021 Elsevier Ltd. Secondary phosphine chalcogenides react with primary amino alcohols under mild conditions (room temperature, molar ratio of the initial reagents 1 : 1) in a CCl4/Et3N oxidizing system to chemoselectively deliver amides of chalcogenophosphinic acids with free OH groups. Under similar conditions, mono-cross-coupling between secondary phosphine chalcogenides and 1,2- or 1,3-aminophenols proceeds only with the participation of phenolic hydroxyl to give aminophenylchalcogenophosphinic O-esters. The yields of the synthesized functional amides or esters are 60-85%.

A method for the synthesis of pharmacologically prospective arabinogalactan (AG) imidazole- and benzimidazole derivatives in a yield of up to 97 % via Au(III)-catalyzed hydroamination of AG propargyl ethers has been developed. It is found that in the presence of 5 mol% HAuC1 4 and 10-fold excess imidazole relative to the propargyl groups, the hydroamination successfully competes with cross-linking processes to afford products soluble in DMSO and aqueous HC1 solutions (degree of substitution of imidazolylpropenyl fragments 0.5-1.8, yield 62-97 %). It is established that under the conditions of hydroamination Au(III) is reduced to give mainly Au(0) and minor amounts of Au(I), which are contained in AG imidazole derivatives as particles of 190-640 nm in size. Hydrochlorides of Au-containing AG imidazole derivatives show high bacteriostatic activity with respect to test gram-positive microorganisms and thus confirming their prospects as new AG-derived bioactive agents., (Copyright © 2020 Elsevier Ltd. Novel pharmacologically prospective derivatives of arabinogalactan (a polysaccharide from larch wood) containing acetylenic and thiophene moieties have been obtained in up to 90% yield by cross-coupling of propargylic ethers of arabinogalactan with 2-bromothiophene. The reaction proceeds in the presence of the catalytic system Pd(Ph3P)4/CuBr/LiBr and piperidine in DMSO at 80-85°С. An advantageous feature of the synthesis is that it requires 5-25 times lesser catalytic loading than in common Sonogashira protocols thus making the reaction particularly beneficial to synthesize pharmaceutically-oriented polysaccharides., (Copyright © 2016 Elsevier Ltd. Polysaccharide arabinogalactan (AG) has been modified by triazolyl functions via the copper-catalyzed 1,3-dipolar addition of azides to propargyl ethers. A range of new AG triazolo derivatives bearing benzyl, 4-vinylbenzyl, 1-naphthylmethyl, (1-vinylimidazol-2-yl)methyl, (1-ethylimidazol-2-yl)methyl, (1-vinylbenzimidazol-2-yl)methyl, allyl, carboxymethyl (as Na-salt) substituents is prepared by "one-pot" approach from organic azides generated in situ and AG propargyl ethers. The latter (degree of substitution 2.0-2.2) are converted into 1,2,3-triazolo AGs in DMSO/water mixture in the presence of CuSO4·5H2O/sodium ascorbate/Et3N in 82-94% yields and with 60-100% conversion., (Copyright © 2014 Elsevier Ltd. New arabinogalactan propargyl ethers with degree of substitution (DS) up to 2.8 have been obtained by propargylation of arabino-3,6-galactan (AG) with propargyl bromide (PB) in the two-phase system 30-60% КОН aqueous solution/toluene in the presence of triethylbenzylammonium chloride (TEBAС) or without catalyst (ambient temperature, 1-24 h) in 20-87% yields. The highest yields have been reached using TEBAС as phase-transfer catalyst, though the non-catalytic version proves to be also efficient (DS 2.8, 70% yield). The propargylation of AG is less effective in the systems MОН (M=Na, K)/DMSO. DS of propargyl AG reaches 1.8 (70% yield) when propargyl chloride is used as propargylating agent, while with PB under the same conditions DS value does not exceed 0.4-0.6, the yields being 44-65%. Under the action of t-BuOK/DMSO system, the AG propargyl ethers have been isomerized to the corresponding unstable allenyl ethers. AG propargyl ethers with DS 2.0 have been almost exhaustively metallated with Ag2O/NH3 system to give AG-Ag acetylides derivatives with Ag/C≡C ratio equaling 0.95., (Copyright © 2013 Elsevier Ltd. The effects of intramolecular and intermolecular coordination on (31)P nuclear shielding have been investigated in the series of tetracoordinated, pentacoordinated and hexacoordinated N-vinylpyrazoles and intermolecular complexes of N-vinylimidazole and 1-allyl-3,5-dimethylpyrazole with phosphorous pentachloride both experimentally and theoretically. It was shown that either intramolecular or intermolecular coordination involving phosphorous results in a dramatic (31)P nuclear shielding amounting to approximately 150 ppm on changing the phosphorous coordination number by one. A major importance of solvent effects on (31)P nuclear shielding of intramolecular and intermolecular complexes involving N → P coordination bond has been demonstrated. It was found that the zeroth-order regular approximation-gauge-including atomic orbital-B1PW91/DZP method was sufficiently accurate for the calculation of (31)P NMR chemical shifts, provided relativistic corrections are taken into account, the latter being of crucial importance in the description of (31)P nuclear shielding., (Copyright © 2012 John Wiley & Sons, Ltd.)

Several representatives of natural flavonoids and their synthetic nitro-derivatives have been investigated by polarography and electron paramagnetic resonance (EPR) spectroscopy under electrochemical reduction in acetonitrile, dimethylformamide (DMF), dimethylsulfoxide (DMSO) or 1,2-dimethoxyethane. All the compounds studied are reduced in the first stage by one-electron transfer, apart from flavanone, which accepts two electrons simultaneously. However, the primary radical anions were detected by EPR spectroscopy only for 4'-nitroflavone. It was shown that radical anions of other flavonoids quickly dimerized. The analysis of the temperature dependence of the hyperfine interaction constants and broadening of lines in EPR spectra of 4'-nitroflavone radical anions has shown that the distribution of spin density is due to both the change of polarity of the medium and rotation of the nitrophenyl moiety. The structure of novel phosphorus-containing N-vinylazoles prepared by action of phosphorus pentachloride has been studied by multinuclear (1)H, (13)C, (15)N, (31)P and two-dimensional (2D) NMR spectroscopy. N-vinyl-substituted 1,2-diazoles and 1,2,3-triazoles have undergone phosphorylation, exclusively, on double bond. N-vinylazoles-based hexa-coordinated phosphorus compounds have been synthesized for the first time. Novel 2-acetylamino-4-acetyl-5-aryl(heteryl)-1,3,4-thiadiazolines, 2-acetylamino-5-aryl(heteryl)-1,3,4-thiadiazoles, and some of their salts were prepared and studied by multinuclear 1H, 13C, 15N, 19F and 2D NMR spectroscopy. Configurational assignment of five carbon, silicon and germanium containing propynal oximes has been carried out by means of experimental measurements and high-level ab initio calculations of their 13C-1H, 13C-13C and 15N-1H spin-spin coupling constants. Carbon-carbon coupling constants have been experimentally measured using the INADEQUATE pulse sequence in the series of N-[1,2-bis(dialkylamino)-2-arylethylidene]arylsulfonamides obtained from N-(1-aryl-2,2,2-trichloroethyl)amides of arylsulfonic acids. Comparison of the experimental J(C,C) in this series with those calculated at the SOPPA (Second-Order Polarization Propagator Approach) level in the model aminosulfonylamidine provided an unambiguous assignment at the C=N bond of the eight-title aminosulfonylamidines to E-configuration, while the unknown J(C,C) couplings in their inaccessible diverse Z isomers have been predicted with high reliability. The established marked difference between J(C,C) of the corresponding carbon-carbon bonds in cis and trans orientations to the nitrogen lone pair in aminosulfonylamidines provides a powerful tool in the configurational assignment at the C=N bond in a wide series of the related systems containing the C=N-SO2R moiety.

The results obtained in the study of antibiotic and sulfamide sensitivity of 197 Neisseria meningitidis strains of groups A, B and C, isolated from the spinal fluid and blood of patients with meningococcal infection hospitalized in the 2nd Clinico-Infectious Hospital, Moscow, in 1984-1989 and studied with the use of the disc diffusion method and the method of serial dilutions of antibiotics in solid culture media, are presented. As revealed in this study, N. meningitidis strains retained their high sensitivity to penicillin and ampicillin (MIC50 = 0.016 and 0.032 micrograms/ml respectively). Sensitivity to tetracycline decreased (MIC50 = 0.5 micrograms/ml) and to rifampicin increased (MIC50 = 0.063 micrograms/ml). 48.5% of strains were resistant to streptomycin. In recent years the proportion of N. meningitidis, resistant to sulfanilamide preparations, significantly decreased and MIC50 was equal to 2.5 micrograms/ml in comparison with 5-10 micrograms/ml in the preceding period. The results of testing sensitivity to antibiotics by both methods coincided. Still the disc diffusion method can be used in epidemiological surveillance on meningococcal infection, while for more exact differentiation of N. meningitidis strains the use of the method of serial dilutions is necessary.

The laboratory examination, by microscopic or bacteriological methods and by counterimmunoelectrophoresis, of CSF and blood from 2653 patients with purulent bacterial meningitis, including those with clinically diagnosed meningococcal infection, was carried out between June 1980 and June 1984. The results showed three main etiological agents: meningococci (79.9%), pneumococci (10.8%) and Haemophilus influenzae type b (5.25%). Out of 488 Neisseria meningitidis strains isolated from the CSF or blood, 58.2% belonged to serogroup A, 17.2% to serogroup B, and 14% to serogroup C; infection due to the B serogroup reached nearly 59% among children in 1984. Serotypes were determined in 131 out of 151 strains of Streptococcus pneumoniae and the most frequent were types 1, 19 and 3; type 34, which was isolated from 4 patients, is not a component of the pneumococcal vaccine. The age groups at high risk were children under 5 years old (for meningococcal infection), adults and babies in the first year (pneumococcal meningitis), and children under 3 years, especially between 6 months and 2 years old (H.influenzae type b infection).

Antagonistic and bacteriocinogenic activity was studied in 169 strains of meningococci of various serological groups and with different localization in the human organism at the time of isolation. Bacteriocinogenic activity was revealed in all the meningococcus strains (by delayed antagonism method), and, in addition, antagonistic activity was found in 100 strains. The inhibitory activity was the greatest in meningococci isolated from the cerebrospinal fluid. The data obtained suggest an important inhibitory activity of meningococci, along with their resistance to the antagonistic activity of the nasopharyngeal microorganisms, in the manifestation of pathogenic properties in them.

Comparative study of the meningococcus sensitivity of various serological groups with different localization in the human organism to the antagonistic activity of normal microbes of the nasopharynx and the antibiotics it was found that strains isolated from the cerebrospinal fluid of patients suffering from cerebrospinal meningitis, chiefly of serological group A, had the greatest resistance to the antagonists. Taking into consideration the leading epidemiological role of the cultures belonging to the serological group A in the USSR, it can be supposed that meningococcus sensitivity to the nasopharyngeal antagonists was of significance for the manifestation of their pathogenic properties. No association of the antibiotic sensitivity of the same strains with reference to a definite serological group or the site of the microbe localization was revealed.

Complexes of surface soluble antigens were obtained in grown meningococci of serological groups A, B and C in fluid nutrient media. In studying the chemical composition and biological properties of these complexes released of the nutrient medium components it was revealed that they consisted of proteins, carbohydrates and lipids, possessed marked group-specific properties in the PHAT, were toxic (LD50 about 0.15 mg) and immunogenic.

A study was made of the duration of meningococcus carrier state, immunological indices and group-specific properties of meningococci isolated from them. The periods of meningococcus discharge were studied in 738 persons. Three categories of the carrier state were revealed: a single discharge (67% of the carriers), of average duration (up to 4 weeks), and prolonged. Greater indices of group-specific antibodies (in the passive hemagglutination test) were revealed in the carriers with prolonged presence of meningococci in the nasopharynx. Dynamics of immunological indices and periods of the carrier state formation in the foci of infection permitted to characterize the prolonged carrier state as a latent form of meningococcus infection. A study of the group-specific properties in 1845 strains and comparison of the group-specific pattern of the circulating strains with the epidemic situation indicated that meningococci of group A were not only epidemic, but also more virulent.

The results of studies carried out with the use of the unified, specially developed methods in eight regions of the USSR are generalized. Basing on the analysis of the results, the leading role of serogroup A meningococci in the rise of morbidity is emphasized, the relation of the morbidity level to the serogroup composition of meningococci circulating among healthy population is revealed, the necessity of carrying out selective bacteriological surveys with a view to establish the serogroups of meningococci circulating among the population is shown. The expedient criteria to be used in the system of epidemiological surveillance of meningococcal infection are presented.

The results of the laboratory examination of 2034 patients with meningococcal infection and purulent meningitides, hospitalized during the period of June 1980 to October 1983, revealed that three main etiological agents were responsible for these diseases: meningococci, pneumococci and Haemophilus influenzae. The susceptibility of the patients to different etiological agents was found to depend on their age. Children aged up to 3 years constituted 75% of the patients with meningitis caused by H. influenzae; 50% of the patients with meningococcal infection were children aged up to 5 years; pneumococcal meningitis occurred more frequently in adults. Serogroup A meningococci were found to prevail in patients with meningococcal infection. Besides, in children serogroup C meningococci could be isolated in 24% of cases. Since 1983 the cases of the isolation of strains belonging to serogroup B increased in number. Among the pneumococci responsible for the disease serotypes 1, 19, 6 and in children serotype 12 occurred most frequently.

The results obtained in 1987 in the study of the immunostructure of the population of Yaroslavl with respect to meningococcal polysaccharides, groups, A, B, C, and lipopolysaccharide are presented in comparison with earlier results obtained in 1976. The regulating role of the immunological factor in the evolution of the epidemic process of meningococcal infection has been confirmed. The level of antibodies to meningococcal polysaccharides, groups A and B, has been found to reflect the intensity of the circulation of the infective agent among the population. The comparison of the results of investigations carried out in 1976 and 1987 has revealed the essential role of the lipopolysaccharide antigen in the formation of the postinfection immunity of the population to meningococcal infection, irrespective of the group of the infective agent.

As the result of laboratory examination of 2165 patients with virulent bacterial meningitides, including cases of meningococcal infection, the etiological diagnosis was confirmed in 1407 patients (65.0%), the number of cases confirmed by the laboratory examination being significantly greater among adults than among children: 67.5 +/- 1.37% and 63.1 +/- 1.53%, respectively, (t = 2.1). Meningococcal infection was confirmed in 1111 (70.6%) out of 1572 patients under examination. Of the patients with purulent meningitides, pneumococcal etiology was determined in 27.4%, type b of Haemophilus influenzae in 13.5%, other infective agents in 10.0%. The comparison of the results obtained in the examination, carried out by different methods, of 946 children and 770 adults with meningococcal infection revealed a considerable difference in the number of positive results yielded by the bacteriological method and countercurrent immunoelectrophoresis (CIE). Among adults meningococci were isolated twice as frequently (41.1 +/- 2.5% - 19.4 +/- 1.6%), and the results yielded by CIE were predominantly positive (55.1 +/- 2.5% and 40.1 +/- 2.5%). CIE and the immunoenzyme assay were shown to have advantages in the diagnosis of the disease. Under the conditions of intensive antibiotic therapy the methods based on the detection of specific antigens in body fluids can greatly assist in diagnosis.

The materials on the development and use of the test system, based on the enzyme-linked immunosorbent assay (ELISA) and intended for the detection of specific group A and C meningococcal polysaccharides and type b Haemophilus influenzae polysaccharide in the spinal fluid of patients, are presented. In this work commercial preparations manufactured in the USSR were used, and all parameters of the assay were developed on the basis of these preparations. The study was made on the samples of spinal fluid from 410 patients; of these, 203 had meningococcal infection, 57 had purulent bacterial meningitides and 150 had other diseases (acute respiratory diseases, influenza, etc.). As demonstrated by the results of this study, ELISA proved to be a highly specific and sensitive technique. In the investigation of the spinal fluid samples from the patients with meningococcal infection the use of ELISA with bacteriological techniques increased the number of positive results to 67%; with countercurrent electrophoresis, to 78%; and with bacterioscopy, to 83.8%. ELISA is recommended for practical use as an auxiliary laboratory technique and as a rapid method for the diagnosis of meningococcal infection.

The coagglutination (COA) test is used for the serogrouping of streptococci, group B, and meningococci. The trial of a Soviet commercial preparation (staphylococcal reagent) has shown good prospects for its use in the COA test and high specificity of this test method. The availability of highly specific immune sera permits making standard kits for the identification of different infective agents in the COA test.
