Abstract

The science guiding the EUREC4A campaign and its measurements is presented. EUREC4A comprised roughly 5 weeks of measurements in the downstream winter trades of the North Atlantic - eastward and southeastward of Barbados. Through its ability to characterize processes operating across a wide range of scales, EUREC4A marked a turning point in our ability to observationally study factors influencing clouds in the trades, how they will respond to warming, and their link to other components of the earth system, such as upper-ocean processes or the life cycle of particulate matter. This characterization was made possible by thousands (2500) of sondes distributed to measure circulations on meso- (200 km) and larger (500 km) scales, roughly 400 h of flight time by four heavily instrumented research aircraft; four global-class research vessels; an advanced ground-based cloud observatory; scores of autonomous observing platforms operating in the upper ocean (nearly 10 000 profiles), lower atmosphere (continuous profiling), and along the air-sea interface; a network of water stable isotopologue measurements; targeted tasking of satellite remote sensing; and modeling with a new generation of weather and climate models. In addition to providing an outline of the novel measurements and their composition into a unified and coordinated campaign, the six distinct scientific facets that EUREC4A explored - from North Brazil Current rings to turbulence-induced clustering of cloud droplets and its influence on warm-rain formation - are presented along with an overview of EUREC4A's outreach activities, environmental impact, and guidelines for scientific practice. Abstract

ObjectiveDespite recent progress in caring for patients born with esophageal atresia (EA), undernutrition and stunting remain common. Our study objective was to assess nutritional status in the first year after birth with EA and to identify factors associated with growth failure.Study designWe conducted a population-based study of all infants born in France with EA between 2010 and 2016. Through the national EA register, we collected prenatal to 1 year follow-up data. We used body mass index and length-for-age ratio Z scores to define patients who were undernourished and stunted, respectively. Factors with P < 0.20 in univariate analyses were retained in a logistic regression model.ResultsAmong 1,154 patients born with EA, body mass index and length-for-age ratio Z scores at 1 year were available for about 61%. Among these, 15.2% were undernourished and 19% were stunted at the age of 1 year. There was no significant catch-up between ages 6 months and 1 year. Patients born preterm (41%), small for gestational age (17%), or with associated abnormalities (55%) were at higher risk of undernutrition and stunting at age 1 year (P < 0.05). Neither EA type nor surgical treatment was associated with growth failure.ConclusionUndernutrition and stunting are common during the first year after birth in patients born with EA. These outcomes are significantly influenced by early factors, regardless of EA type or surgical management. Identifying high-risk patient groups with EA (i.e., those born preterm, small for gestational age, and/or with associated abnormalities) may guide early nutritional support strategies.

Abstract

Although the field has a long collaborative tradition, immunology has made less use than genetics of 'consortium biology', wherein groups of investigators together tackle large integrated questions or problems. However, immunology is naturally suited to large-scale integrative and systems-level approaches, owing to the multicellular and adaptive nature of the cells it encompasses. Here, we discuss the value and drawbacks of this organization of research, in the context of the long-running 'big science' debate, and consider the opportunities that may exist for the immunology community. We position this analysis in light of our own experience, both positive and negative, as participants of the Immunological Genome Project. © 2012 Macmillan Publishers Limited. All rights reserved.

Abstract

International audience; Ecohydrology is a growing field of science that aims to quantify the interplay between water as a nutrient carrier and bio-assimilation processes (Zalewski 2001). Its basic assumption relies on the fact that water pollution can be processed efficiently by biota along the water pathways by regulating abiotic and biotic controlling factors at key locations. The most important feature to design the so called dual regulation is the non linear time response of biota to controlling factors. Biota refer either to aquatic vegetation or micro-organisms. We here develop a method to analyze and model the response dynamic of biota to abiotic factors evolution at a reach scale. In a first step a natural growing function is fitted to observed biotic data. Then residues to this model are processed in a second step with a memory analysis technique that helps to identify key abiotic factors. In a third step, a dynamic model is build by combining the natural growth functionl with its abiotic controlling factors. Model is then run at a daily time step. Data come from a reach of the river Dordogne. Reach was 250 m long and 100 m wide. Vegetation species and abundance, water level, velocity and temperature were sampled during five years several times a year. Most abundant taxonomic groups of aquatic macrophytes in our samplings were phanerogams and algae. The good model fitting over years with observed data confirms the mixing methodology of biotic and abiotic factors can serve the dynamic process analysis. An equivalent N uptake function is then processes considering the macrophyte biomass. Memory analysis results indicate that Phanerogam are mainly linked to water temperature and water depth variations that occurred in the past 10 days but also depend on a seasonal time scale. Algae variations are closely linked to the antecedent day(s) variations in flow velocity. First conclusions are drawn on the methodology and a discussion is open on controlling factors of the nitrogen uptake that would be regulated to increase the yearly overall natural N uptake at the reach scale.

33. Amylin inhibits bone resorption while the calcitonin receptor controls bone formation in vivo

Abstract

Amylin is a member of the calcitonin family of hormones cosecreted with insulin by pancreatic beta cells. Cell culture assays suggest that amylin could affect bone formation and bone resorption, this latter function after its binding to the calcitonin receptor (CALCR). Here we show that Amylin inactivation leads to a low bone mass due to an increase in bone resorption, whereas bone formation is unaffected. In vitro, amylin inhibits fusion of mononucleated osteoclast precursors into multinucleated osteoclasts in an ERK1/2-dependent manner. Although Amylin +/- mice like Amylin-deficient mice display a low bone mass phenotype and increased bone resorption, Calcr +/- mice display a high bone mass due to an increase in bone formation. Moreover, compound heterozygote mice for Calcr and Amylin inactivation displayed bone abnormalities observed in both Calcr +/- and Amylin +/- mice, thereby ruling out that amylin uses CALCR to inhibit osteoclastogenesis in vivo. Thus, amylin is a physiological regulator of bone resorption that acts through an unidentified receptor.

Abstract

The mouse pre-T-cell receptor alpha (pTα) chain is a 33 000 M r glycoprotein expressed on the surface of immature thymocytes as a disulfide-linked heterodimer with the T-cell receptor beta (TCRβ) chain, and in association with proteins of the CD3 complex. The cDNA for pTα, isolated previously, encodes a type I transmembrane protein that is a member of the immunoglobulin (Ig) superfamily. Here we report the complete nucleotide sequence, the exon/intron structure, and the chromosomal location of the pTa gene. The gene spans about 8.4 kilobases (kb) and consists of four exons. Exon 1 encodes the 5′ untranslated region, the leader peptide, and the first three amino acids of the mature protein. This exon is followed by a relatively long intron of 4.9 kb that contains many short interspersed repeats (SINEs) of the B1 and B2 family. The second exon encodes the extracellular Ig-like domain and exon 3 with just 45 base pairs the connecting peptide (CP), including the cysteine required for heterodimer formation. A similar exon/intron structure encoding corresponding parts of the mature polypeptide is found both in the Tcra and Tcrd constant region genes. The last exon encodes the transmembrane portion, the cytoplasmic tail, and about 540 nucleotides of 3′ untranslated sequence, including a B2 repetitive element. In situ hybridization maps the pTa gene to the D/E1 region of mouse chromosome 17.
