- Abstract
Stroke is a devastating disease. Endovascular mechanical thrombectomy is dramatically changing the management of acute ischemic stroke, raising new challenges regarding brain outcome and opening up new avenues for brain protection. In this context, relevant experiment models are required for testing new therapies and addressing important questions about infarct progression despite successful recanalization, reversibility of ischemic lesions, blood–brain barrier disruption and reperfusion damage. Here, we developed a minimally invasive non-human primate model of cerebral ischemia (Macaca fascicularis) based on an endovascular transient occlusion and recanalization of the middle cerebral artery (MCA). We evaluated per-occlusion and post-recanalization impairment on PET-MRI, in addition to acute and chronic neuro-functional assessment. Voxel-based analyses between per-occlusion PET-MRI and day-7 MRI showed two different patterns of lesion evolution: "symptomatic salvaged tissue" (SST) and "asymptomatic infarcted tissue" (AIT). Extended SST was present in all cases. AIT, remote from the area at risk, represented 45% of the final lesion. This model also expresses both worsening of fine motor skills and dysexecutive behavior over the chronic post-stroke period, a result in agreement with cortical-subcortical lesions. - Abstract
To cause disease, a microbial pathogen must adapt to the challenges of its host environment. The leading fungal pathogen Candida albicans colonizes nutrient-poor bodily niches, withstands attack from the immune system, and tolerates treatment with azole antifungals, often evolving resistance. To discover agents that block these adaptive strategies, we screened 300,000 compounds for inhibition of azole tolerance in a drug-resistant Candida isolate. We identified a novel indazole derivative that converts azoles from fungistatic to fungicidal drugs by selective inhibition of mitochondrial cytochrome bc1. We synthesized 103 analogs to optimize potency (half maximal inhibitory concentration 0.4 μM) and fungal selectivity (28-fold over human). In addition to reducing azole resistance, targeting cytochrome bc₁ prevents C. albicans from adapting to the nutrient-deprived macrophage phagosome and greatly curtails its virulence in mice. Inhibiting mitochondrial respiration and restricting metabolic flexibility with this synthetically tractable chemotype provides an attractive therapeutic strategy to limit both fungal virulence and drug resistance., National Institutes of Health (U.S.) (GM46059)

International audience; To choose the most suitable RF coil available for an MRI study, we propose a procedure which uses a calibrated phantom, a 3D gradient-echo sequence, and an automatic post-processing tool available on the web. This tool generates a report which contains the measurement of a SNR with uniform volumes located in the depth. The post-processing could be done on MR images acquired on most main MRI vendors (Siemens, GE, Philips and Bruker) with prior verification of applied scaling or filtering. RF coil characterization results performed on at 4.7T and 7T were compared. The tool can be used for quality control.

National audience; But de l'étudeLes antennes radiofréquences jouent un rôle crucial dans la détection du signal IRM. Elles connaissent une évolution permanente pour augmenter leur efficacité. Aujourd'hui, il existe un grand nombre d'antennes différentes et il peut être difficile de choisir celle qui est la plus adaptée à un examen inhabituel. De plus, ce choix peut lourdement se complexifier lorsqu'on a accès à des systèmes IRM de différents fabricants et opérant à différents champs statiques. Le protocole de caractérisation que nous proposons ici a pour but de comparer les antennes en terme de SNR et de volume uniforme. Il peut être utilisé pour comparer des systèmes IRM dans le cadre d’étude multicentrique ou encore comme outil de contrôle qualité.Matériels et MéthodeLe protocole peut se décomposer en trois étapes:1- Utiliser un fantôme chargé pour reproduire les conditions in-vivo. Dans le cadre multicentrique, la plateforme PILoT propose différents fantômes pour le clinique et le préclinique (pilot@creatis.insa-lyon.fr ).2- Acquérir les images RMN en utilisant une séquence écho de gradient 3D disponible sur tous les systèmes. 3-Traiter les acquisitions de manière automatique en utilisant notre programme déployé sur la plateforme d'imagerie virtuelle (VIP) [1] : https://vip.creatis.insa-lyon.fr et disponible en libre accès. Le programme génère un rapport au format PDF qui inclut: une mesure du SNR; des représentations de volumes uniformes à 10, 50 et 90% de variation; un tableau comparatif de toutes les antennes analysées ainsi qu'une figure des profils d'intensité superposés. Un effort particulier a été fourni pour s'assurer de la mesure du SNR en appliquant deux tests basés sur la loi de distribution du bruit dans l'air des images enmagnitude [2]. Enfin nous proposons un SNR corrigé (SNRc) prenant en compte des variations des paramètres d'acquisition ainsi que des temps de relaxation [3].RésultatsLa figure 1 montre la superposition des profils d'intensité de l'ensemble des antennes disponibles sur un IRM Bruker précliniques à 4.7T. Le fantôme utilisé simule l'abdomen d'une souris de taille moyenne. L'intersection des courbes donne une information sur la profondeur d’exploration relative par rapport aux autres antennes. Le tableau 1 présente une partie des résultats obtenus précédemment et comparés à ceux d'un IRM Bruker précliniques à 7T pour des antennes similaires. La bobine en quadrature nommée "Quadrature" provenant du même constructeur donne un rapport des SNR proche de la théorie (R = 1.5). Ce qui n'est pas le cas pour des antennes de constructeurs différents: "Loop_rat" (capteurs de configurations différentes) et "BirdCage" (bobines de générations différentes).DiscussionLa comparaison des caractéristiques mesurées par rapport à la région d’intérêt à explorer permet de choisir l'antenne avec le SNRc le plus élevé. Ainsi, on confirme par exemple dans le cas d’un examen du foie que le capteur "Loop1H_rat" est adapté à une mesure de spectroscopie localisée tandis que la bobine "Quad1H_30mm" est à privilégier pour une IRM du foie entier de souris.

National audience; Le CERMEP (Centre d'Etude et de Recherche Multimodal Et Pluridisciplinaire) est un centre d'imagerie in vivomultimodale dédié à la recherche biomédicale fondamentale et clinique. A ce titre, le CERMEP fait partie du Noeud Lyonnais de l’infrastructure nationale France Life Imaging (FLI).La plate-forme d’imagerie préclinique ANIMAGE est un département du CERMEP qui a pour objectif de proposer aux chercheurs académiques ou industriels des méthodes d’imagerie in vivo pour l’exploration anatomique, moléculaire et fonctionnelle du petit animal de laboratoire.En termes d’équipement d’imagerie, ANIMAGE dispose d’une IRM BRUKER BioSpec 7T, d’une caméra TEP/TDM SIEMENS Inveon, d’un scanner SKYSCAN-1076 aux rayons X et d’un échographe VISUALSONICS Vevo 770. Une vingtaine de radiotraceurs TEP sont accessibles grâce à la proximité du département de radiochimie et radiopharmacie du CERMEP. Des explorations autoradiographiques in vitro de ces radiotraceurs sont également possibles sur le site. Tous les imageurs sont équipés d’anesthésie gazeuse, d’un poste de contrôle et de suivi physiologique qui servent d’outils de monitoring des animaux tout au long des expérimentations, ponctuelles ou longitudinales. Une salle de salle de chirurgie est mise à la disposition des expérimentateurs, à proximité immédiate des imageurs. Equipée de deux paillasses, disposant de postes d'anesthésie gazeuse, microscopes, etc., elle permet la préparation des modèles animaux ou des journées de formations. Une animalerie conventionnelle (type A1 avec un niveau de confinement L2) permet d'héberger les animaux pendant la durée du protocole d'imagerie. Ces locaux sont agréés par les services vétérinaires pour cinq espèces animales (porcs, lapins, chats, rats, souris). La structure « Bien-Etre Animal » interne au CERMEP accompagne les investigateurs dans leur demande d’autorisation de projet utilisant des animaux à des fins scientifiques (APAFIS – Comité d’éthique). L’essentiel des applications visées aujourd’hui se situe dans le domaine des neurosciences mais la plateforme est ouverte à d’autres thématiques (oncologie, cardiologie, métabolisme, etc.).Les membres d’ANIMAGE accompagnent les équipes de recherche tout au long du protocole de recherche, selon leurs besoins :- définition de la question biologique/physiopathologique/pharmacologique à laquelle pourra répondre l’imagerie,- adéquation de la modalité d’imagerie et du modèle expérimental,- organisation du protocole expérimental,- préparation du modèle animal,- acquisition des images in vivo,- analyse des images et aide à la valorisation scientifique (rapport, article...).

The copper-catalyzed asymmetric allylic alkylation (AAA) is one of the most powerful methods to introduce a simple alkyl group in allylic position. Copper is particularly known to induce a high γ-selectivity stemming from a SN2' process. This feature has been mainly applied to the alkylation of linear prochiral substrates leading to the formation of stereogenic centers. In sharp contrast, the use of racemic substrates has been scarcely studied. We will present here that this range of substrates could be interesting reaction partners leading to the development of new valuable resolution processes.

46. Exercise Does Not Protect against Peripheral and Central Effects of a High Cholesterol Diet Given Ad libitum in Old ApoE-/- Mice.

Aim: Advanced atherosclerosis increases inflammation and stroke risk in the cerebral vasculature. Exercise is known to improve cardio-metabolic profiles when associated with a caloric restriction, but it remains debated whether it is still beneficial without the dietary control. The aim of this study was to determine both the peripheral and central effects of exercise training combined with a cholesterol-rich diet given ad libitum in old ApoE
-/- mice. Methods : Forty-five-weeks old obese ApoE-/- mice fed with a high cholesterol diet ad libitum were divided into Exercise-trained (EX; running wheel free access) and Sedentary (SED) groups. Insulin tolerance and brain imaging were performed before and after the twelve-weeks training. Tissue insulin resistance, oxidative stress, and inflammation markers in plasma, aorta, and brain were then assessed. Results : In EX ApoE-/- mice, no beneficial effect of exercise was observed on weight, abdominal fat, metabolic parameters, oxidative stress, or inflammation compared to SED. Despite the regular exercise training in ApoE-/- EX mice (mean of 12.5 km/week during 12 weeks), brain inflammation imaging score was significantly associated with increased blood brain barrier (BBB) leakage evaluated by imaging follow-up (r2 = 0.87; p = 0.049) with a faster evolution compared to SED ApoE-/- mice. Conclusion : We conclude that in a context of high cardio-metabolic risk, exercise does not provide any protective effect in old ApoE-/- -/- animals under high cholesterol diet given ad libitum. Peripheral (insulin sensitivity and oxidative/inflammatory status) but also central features (BBB preservation and protection against inflammation) did not show any benefits of exercise. Indeed, there was a fast induction of irreversible brain damage that was more pronounced in exercise-trained ApoE-/- mice. [ABSTRACT FROM AUTHOR]- Published
