Der Sammelband stellt aktuelle Ansätze zum digital unterstützten beruflichen Lernen dar. Die Beiträge geben Einblicke in die dynamische Entwicklung der Schnittstellen von Erwerbsarbeit und beruflicher Aus-, Fort- und Weiterbildung im Kontext der Digitalisierung Arbeits- und Lernmitteln. Der Band schließt damit an die 2019 ebenfalls im Universitätsverlag der Technischen Universität Berlin erschienene Publikation „Berufsbildung am Bau digital“ (hrsg. von Bernd Mahrin und Johannes Meyser) an. Das erste Kapitel erörtert grundsätzliche didaktische Fragen zu digital unterstütztem Lernen und Arbeiten einschließlich der Rahmenbedingungen. Im zweiten Kapitel schließen sich Beiträge zur Kapazitätsentwicklung, zu Standards und zu digitalen Werkzeugen an. Das dritte Kapitel widmet sich konkreten Einzellösungen mit starkem Praxisbezug und hohem Transferpotenzial zum digitalisierten Arbeiten und Lernen im Bausektor und im Metallbereich. Das abschließende vierte Kapitel präsentiert übergreifend nutzbare und frei zugängliche Online-Angebote wie einen Medienpool für Bildungszwecke, eine Lernmedien-Datenbank und ein hybrides Lernsystem mit virtuellem 3D-Gebäudemodell. Das Buch ist entstanden im Rahmen des durch das Bundesministerium für Bildung und Forschung und den Europäischen Sozialfonds geförderten Projektes DigiBAU – Digitales Bauberufliches Lernen und Arbeiten. The first chapter discusses fundamental didactic questions about digitally supported learning and working, including the framework conditions. The second chapter picks contributions on capacity development, standards, and digital tools out as central themes. The third chapter is dedicated to concrete specific solutions with strong practical relevance and high transfer potential for digitized work and learning in the construction sector and in the metal sector. The final fourth chapter presents comprehensive and freely accessible online offers such as a media pool for educational purposes, a learning media database and a hybrid learning system with a virtual 3D building model. The book was created as part of the DigiBAU project - digital vocational learning and working in the field of construction - funded by the German Federal Ministry of Education and Research and the European Social Fund. Der Beitrag fokussiert die Entwicklung inklusive Lernumgebungen an überbetrieblichen Berufsbildungsstätten der Bauwirtschaft im Projekt mein Beruf Bau vor dem Hintergrund des Stands der Forschung zur Inklusion mithilfe digitaler Medien in der gewerblich-technischen Berufsbildung.

Curcumin, the extract of the rhizome of Curcuma longa, is known for its health-promoting properties in traditional medicine. It has anti-inflammatory, antitumor and antioxidant properties and stimulates appetite. In the present study, we investigated the stability of curcumin and its effect on cytotoxicity, apoptosis and melanin content in melanoma cells and the effect on atrophic C2C12 muscle cells. Cytotoxicity of curcumin was dose-dependent and the EC50 for 24-h incubation was 69 µM. Saturation was reached at 30 µM for a 48-h incubation. The EC50 for 24-h incubation with degraded curcumin solution was 116 ?M and that for 48-h was 94 µM. Curcumin induced a strong increase in caspase-3/7 activity at 30-40 µM. Electrical impedance measurements showed that sub-toxic doses of curcumin counteracted atrophy in an in vitro model system. These findings indicate not only the positive effects of curcumin on melanoma cells in vitro, but also that curcumin was able to considerably trigger anti-cachectic effects in vitro. However, the importance of the stability of curcumin and its tumoricidal and anti-cachectic potential might play a pivotal role in its use in the nutrition and health industrie since it degrades rapidly in aqueous solutions.

The peroxisome proliferator-activated receptor γ coactivator 1 (PGC-1) proteins are key regulators of cellular bioenergetics and are accordingly expressed in tissues with a high energetic demand. For example, PGC-1α and PGC-1β control organ function of brown adipose tissue, heart, brain, liver and skeletal muscle. Surprisingly, despite their prominent role in the control of mitochondrial biogenesis and oxidative metabolism, expression and function of the PGC-1 coactivators in the retina, an organ with one of the highest energy demands per tissue weight, are completely unknown. Moreover, the molecular mechanisms that coordinate energy production with repair processes in the damaged retina remain enigmatic. In the present study, we thus investigated the expression and function of the PGC-1 coactivators in the healthy and the damaged retina. We show that PGC-1α and PGC-1β are found at high levels in different structures of the mouse retina, most prominently in the photoreceptors. Furthermore, PGC-1α knockout mice suffer from a striking deterioration in retinal morphology and function upon detrimental light exposure. Gene expression studies revealed dysregulation of all major pathways involved in retinal damage and apoptosis, repair and renewal in the PGC-1α knockouts. The light-induced increase in apoptosis in vivo in the absence of PGC-1α was substantiated in vitro, where overexpression of PGC-1α evoked strong anti-apoptotic effects. Finally, we found that retinal levels of PGC-1 expression are reduced in different mouse models for retinitis pigmentosa. We demonstrate that PGC-1α is a central coordinator of energy production and, importantly, all of the major processes involved in retinal damage and subsequent repair. Together with the observed dysregulation of PGC-1α and PGC-1β in retinitis pigmentosa mouse models, these findings thus imply that PGC-1α might be an attractive target for therapeutic approaches aimed at retinal degeneration diseases., PLoS ONE, 7 (2), ISSN:1932-6203

The initial sensing of invading potential pathogenic microorganisms by pattern recognition receptors (PRRs) of the innate immune system is pivotal for the survival of the host. Nucleotide binding and oligomerization domain (NOD)-like receptors (NLRs) are a family of cytoplasmic PRRs that recognize conserved microbe-associated molecular patterns (MAMPs) or endogenous danger signals. Upon activation, these proteins trigger the release of pro-inflammatory cytokines and antimicrobial peptides (AMPs) via a variety of signal transduction cascades in mammals. All pathways have in common the initial formation of a multimeric complex of the activated NLR and downstream signalling proteins through homophilic interactions of effector domains. Many genetic variants of NLRs have been linked to chronic inflammatory disorders of the human epithelial barrier organs. Although these receptors are of fundamental importance for understanding the maintenance of the human barrier integrity not much is known about their evolutionary roots. This thesis involved screening the genomes of a variety of animal species with a focus on basal Metazoa and particularly on the freshwater polyp Hydra for NLR orthologues. It was subsequently demonstrated that the family of NLRs is evolutionary conserved and that frequently throughout animal evolution species-specific gene-amplifications occurred. Hydra is continually surrounded by potential pathogenic microorganisms and must rely on an effective epithelial defence machinery. Using Hydra as a model organism for epithelial defence, the expression profile of one of the identified Hydra NLRs, HyNLR type 1 was analysed in detail. This gene is predominantly expressed in the endodermal epithelium and is subject of alternative splicing. A co-expressed putative HyNLR-interactome has been identified as well. First functional in vitro studies using an artificially activatable chimeric protein including the N-terminal part of HyNLR type 1 could show that Hydra caspases may link NLR signalling to apoptosis as an ancient mechanism of innate immune defence. As well as a genetic background, environmental changes and an altered composition of the host’s microbiota are proposed to contribute to the development of barrier diseases. A tumour bearing Hydra strain with an altered microbial composition was analysed using histological and molecular biological techniques. The tumour was shown to be composed of female germline-determined interstitial stem cells. The tumour-like structure shows numerous parallels to oogenesis in Hydra, including lipid storage, the appearance of nurse cells and the expression of germline-specific genes. This first detailed description of tumour formation in a basal eumetazoan animal provides new possibilities for the analysis of an imbalanced tissue homeostasis. Das initiale Erkennen eindringender potenziell pathogener Mikroorganismen durch Muster-Erkennungsrezeptoren (pattern recognition receptors (PRRs)) des angeborenen Immunsystems ist entscheidend für das Überleben des Wirts. Nucleotide binding and oligomerization domain (NOD)-like Rezeptoren (NLRs) stellen eine Familie von zytoplasmatischen PRRs dar, die konservierte Mikroben-assoziierte molekulare Muster (microbe-associated molecular patterns (MAMPs) oder endogene Gefahrsignale erkennen. Nach ihrer Aktivierung leiten diese Proteine in Säugetieren die Freisetzung proinflammatorischer Zytokine und antimikrobieller Peptide (antimicrobial peptides (AMPs)) über eine Vielfalt von Signaltransduktionskaskaden ein. Allen Signalwegen gemein ist die initiale Bildung eines multimeren Komplexes aus dem aktivierten NLR und nachgeschalteten Signalproteinen durch homophile Interaktionen ihrer Effektordomänen. Viele genetische Varianten der NLRs sind mit chronisch entzündlichen Erkrankungen der menschlichen Barriereorgane in Verbindung gebracht worden. Obwohl diese Rezeptoren von fundamentaler Bedeutung für das Verständnis der Aufrechterhaltung der humanen Barriere Integrität sind, ist nicht viel über ihre evolutiven Wurzeln bekannt. Diese Arbeit umfasste das Überprüfen der Genome zahlreicher Tierarten mit einem besonderen Fokus auf basalen Metazoen und insbesondere auf dem Süßwasserpolypen Hydra auf Orthologe für NLRs. Es konnte weiterhin gezeigt werden, dass die Familie der NLRs evolutionär konserviert ist und dass häufig während der Evolution der Tiere artspezifische Gen-Amplifikationen aufgetreten sind. Hydra ist stets von potenziell pathogenen Mikroorganismen umgeben und auf effektive epitheliale Abwehrmechanismen angewiesen. Hydra wurde als Modellorganismus für epitheliale Abwehr benutzt und das Expressionsprofil eines der identifizierten Hydra NLRs, HyNLR type 1, detailliert untersucht. Dises Gen wird vor allem im endodermalen Epithel exprimiert und unterliegt alternativem Spleißen. Ein koexprimiertes putatives Interaktom für HyNLRs konnte ebenfalls identifiziert werden. Erste funktionelle in vitro Studien mit einem künstlich aktivierbaren chimären Protein, das den N-terminalen Bereich von HyNLR type 1 enthält, konnten zeigen, dass Hydra Caspasen die NLR Signalgebung mit Apoptose als einer alten Form der angeborenen Immunantwort verknüpfen können. Es wird vermutet, dass nicht nur ein genetischer Hintergrund, sondern auch Veränderungen der Lebensumstände und eine veränderte Komposition der Wirt-Mikrobiota an der Entstehung von Barriere-Erkrankungen beteiligt sind. Ein spontan aufgetretener Tumor in einer Hydra-Linie, die eine veränderte mikrobielle Zusammensetzung aufwies, wurde histologisch und molekularbiologisch untersucht. Es konnte gezeigt werden, dass der Tumor von interstitiellen Stammzellen, die auf die weibliche Keimbahn festgelegt sind, gebildet wird. Die Tumor-artige Struktur zeigt viele Parallelen zur Hydra-Oogenese auf. Dies beinhaltet die Speicherung von Lipiden, das Auftreten von Nährzellen und die Expression von Keimbahn-spezifischen Genen. Diese erste detaillierte Beschreibung einer Tumorbildung in einem basalen Eumetazoen bietet neue Möglichkeiten für die Analyse einer gestörten Gewebehomöostase.

Kassel : kassel university press 2008, 237, [47] S. - (Zugl.: Kassel, Univ., Diss., 2008), Dietrich Bonhoeffer (1906-1945) gehört für Schüler und Schülerinnen heute zu der Generation ihrer Urgroßeltern. Hat dieser Theologe ihnen trotzdem noch etwas zu sagen? Das ist die Ausgangs- und Leitfrage dieses Buches, die durch eine empirische Untersuchung von Curricula und Religionsbüchern und didaktische und methodische Überlegungen zur unterrichtlichen Praxis versucht werden soll zu beantworten. Die Arbeit gelangt zu dem Fazit, dass Bonhoeffer auch heute Jugendliche noch erreichen kann, wenn es gelingt, ihn von seinem "Podest" herunterzuholen und in die Lebenswelt der Schülerinnen und Schüler hineinzubringen.
