Abstract

Forty-two cervical biopsies with cervical intraepithelial neoplasia were compared with respect to the expression of human papillomavirus (HPV) structural proteins and the expression of the cellular structural protein involucrin, a marker of suprabasal squamous differentiation. HPV structural protein and involucrin expression displayed an inverse correlation with the severity of dysplasia. Both of these proteins were detected in 11 of 28 cases (39%) of mild and moderate dysplasia, but in only two of 14 (14%) cases of severe dysplasia. This difference was statistically significant (p less than 0.001). The presence of HPV was also associated with expression of involucrin in the full thickness of the epithelium, including the basal layer, and an altered staining pattern in the more superficial cells, particularly the koilocytotic cells. These findings support the hypothesis that squamous differentiation is required for the expression of viral structural proteins and that HPV infection begins in the basal epithelium. The study also demonstrates the utility of involucrin staining in differentiating virus-induced cytologic atypia from true neoplasia.

Abstract

Abstract Background Chronic inflammation and autoimmunity likely contribute to the pathogenesis of abdominal aortic aneurysms (AAAs). The aim of this study was to investigate the role of autoimmunity in the etiology of AAAs using a genetic association study approach with HLA polymorphisms. Methods HLA-DQA1, -DQB1, -DRB1 and -DRB3-5 alleles were determined in 387 AAA cases (180 Belgian and 207 Canadian) and 426 controls (269 Belgian and 157 Canadian) by a PCR and single-strand oligonucleotide probe hybridization assay. Results We observed a potential association with the HLA-DQA1 locus among Belgian males (empirical p = 0.027, asymptotic p = 0.071). Specifically, there was a significant difference in the HLA-DQA1*0102 allele frequencies between AAA cases (67/322 alleles, 20.8%) and controls (44/356 alleles, 12.4%) in Belgian males (empirical p = 0.019, asymptotic p = 0.003). In haplotype analyses, marginally significant association was found between AAA and haplotype HLA-DQA1-DRB1 (p = 0.049 with global score statistics and p = 0.002 with haplotype-specific score statistics). Conclusion This study showed potential evidence that the HLA-DQA1 locus harbors a genetic risk factor for AAAs suggesting that autoimmunity plays a role in the pathogenesis of AAAs.

Abstract

Hepatitis B virus (HBV) and certain human papillomaviruses (HPV) have been suspected of playing a role in human malignancy. The long latency period (decades) and the fact that only a small proportion of infected individuals subsequently have cancer suggests that these viruses probably contribute in an indirect manner to the development of cancer. Analysis of viral gene function at the molecular level supports this conclusion. More studies are needed to understand the subtle interactions between host cell and viral functions and the genetic and environmental factors that may contribute to malignancy.

Abstract

Thirty years ago it was recognized that nuclear and cytoplasmic changes in exfoliated cervical cells (koilocytes) were pathognomonic for infection by the human papillomavirus (HPV). The observation that HPV cytology detected preneoplastic changes and infection (which led to the reduction of cancer incidence in industrialized countries), led to the hypothesis that the virus would be involved in the etiology of cervical cancer. Subsequent identification of HPV antigens in cells coilocitóticas coexisting with dysplasias, added strength to this hypothesis. The cloning of HPV genomes in cells from genital lesions provided enough weapons that were used in retrospective and prospective studies of the natural history of cervical cancer and its precursors. Hace treinta años se reconoció que los cambios nucleares y citoplasmáticos en células exfoliadas del cuello uterino (coilocitos), eran patognomónicos para infección por virus del papiloma humano (VPH). La observación que la citología detecta cambios preneoplásicos e infección por VPH (lo cual llevó a la reducción de la incidencia de cáncer en países industrializados), condujo a la hipótesis que el virus estaría involucrado en la etiología de cáncer cervical. La subsecuente identificación de antígenos VPH en las células coilocitóticas, coexistiendo con displasias, añadieron fuerza a dicha hipótesis. La clonación de genomas VPH en células provenientes de lesiones genitales proveyó las armas suficientes que fueron utilizadas en estudios retrospectivos y prospectivos acerca de la historia natural del cáncer de cuello uterino y sus precursores.

24. Analysis of Genes Differentially Expressed in a Human Ovarian Cancer Model

Abstract

The objectives of this study are to: (1) analyze gene expression in a model of human ovarian carcinogenesis from a benign to a malignant phenotype in organotypic culture and (2) to confirm gene expression patterns by RNA analysis. Sufficient RNA for microarray analysis has been isolated and purified and we changed from cDNA arrays to oligonucleotide arrays. All RNAs from one cell line (96.9.18) have queried the Affymetrix human U133 A and B chips in triplicate. All RNAs from the second cell line (1.24.96) have queried the Affymetrix U133A plus chip in triplicate. Data mining is underway by the Bioinformatics Core at Wayne State University. Analysis of the queries of the U133 A and B chips indicate that 231 genes are differentially expressed between benign and malignant cells when data transformation is done by ANOVA at p-<0.1 with an expression level of >1.2-fold change. Quantitative real time polymerase chain reaction validated the expression levels of all genes tested. About 60% of the genes we have identified have previously been shown to be dysregulated in cancer and about 50% of those have been shown to be dysregulated in ovarian cancer thus validating our model.

Abstract

Most epithelial cell cancers (cervix, colon, skin, prostate, breast, etc.) develop from precursor lesions resulting from an accumulation of mutations in growth regulatory genes. Such precursor lesions have not been identified for OVCA but it has been proposed that OVCAs arise by a multistep process through increasingly aggressive stages. We have shown that immortalized human ovarian surface epithelial (HOSE) cells undergo stepwise progression to the malignant phenotype in vitro. We now hypothesize that this phenotypic presentation reflects changes in the expression of genes in biochemical pathways required for transition from benign cells to malignantly transformed cells. The long-range goal of these studies is to identify aberrantly expressed genes in NOSE cells at various stages along the path to the malignant phenotype for the pur%)ose of characterizing biochemical pathways whose expression is dysregulated.
