Plasticity among cell lineages is a fundamental, but poorly understood, property of regenerative tissues. In the gut tube, the small intestine absorbs nutrients, whereas the colon absorbs electrolytes. In a striking display of inherent plasticity, adult colonic mucosa lacking the chromatin factor SATB2 is converted to small intestine. Using proteomics and CRISPR-Cas9 screening, we identify MTA2 as a crucial component of the molecular machinery that, together with SATB2, restrains colonic plasticity. MTA2 loss in the adult mouse colon activated lipid absorptive genes and functional lipid uptake. Mechanistically, MTA2 co-occupies DNA with HNF4A, an activating pan-intestinal transcription factor (TF), on colonic chromatin. MTA2 loss leads to HNF4A release from colonic chromatin, and accumulation on small intestinal chromatin. SATB2 similarly restrains colonic plasticity through an HNF4A-dependent mechanism. Our study provides a generalizable model of lineage plasticity in which broadly-expressed TFs are retained on tissue-specific enhancers to maintain cell identity and prevent activation of alternative lineages, and their release unleashes plasticity.

Non-submerged ultrafiltration(NSUF) technology was used to purify coal mine water. The field tests were carried out on the operating parameters and performance of NSUF ceramic membrane, membrane pollution removal, ultrafiltration concentrated water flocculation with chemicals, and sludge pressure filtration dehydration performance. Under the condition of controlling the ultrafiltration water yield of 70%, the combination of NSUF “membrane surface flow” operation mode and on-line physical cleaning could remove the filter layer blockage and membrane pollution, restore the membrane flux and transmembrane pressure difference, and realize the continuous and stable operation of NSUF device. After adding PAC and PAM, the sedimentation ratio of 30% concentrated water produced by ultrafiltration concentration reached 4.3%, and the clarified liquid returned to the ultrafiltration unit. Flocculated sludge was dewatered by plate-and-frame filter and the water content of filter cake was about 75%. After the combined process of three-stage NSUF-flocculation sedimentation-sludge pressure filtration, the total water production rate of the system could reach 98%.

Abstract Background High-flow nasal cannula (HFNC) has emerged as a promising noninvasive method for delivering oxygen to critically ill patients, particularly those with sepsis and acute lung injury. However, uncertainties persist regarding its therapeutic benefits in this specific patient population. Methods This retrospective study utilized a propensity score-matched cohort from the Medical Information Mart in Intensive Care-IV (MIMIC-IV) database to explore the correlation between HFNC utilization and mortality in patients with sepsis-induced acute lung injury. The primary outcome was 28-day all-cause mortality. Results In the propensity score-matched cohort, the 28-day all-cause mortality rate was 18.63% (95 out of 510) in the HFNC use group, compared to 31.18% (159 out of 510) in the non-HFNC group. Postharvest rot caused by various fungal pathogens is a damaging disease affecting kiwifruit production and quality, resulting in significant annual economic losses. This study focused on isolating the strain P3-1W, identified as Diaporthe eres, as the causal agent of ‘Hongyang’ postharvest rot disease in China. The investigation highlighted cell wall degrading enzymes (CWDEs) as crucial pathogenic factors. Specially, the enzymatic activities of cellulase, β-galactosidase, polygalacturonase, and pectin methylesterases peaked significantly on the second day after infection of D. eres P3-1W. To gain a comprehensive understanding of these CWDEs, the genome of this strain was sequenced using PacBio and Illumina sequencing technologies. The analysis revealed that the genome of D. eres P3-1W spans 58,489,835 bp, with an N50 of 5,939,879 bp and a GC content of 50.7%. A total of 15,407 total protein-coding genes (PCGs) were predicted and functionally annotated. Notably, 857 carbohydrate-active enzymes (CAZymes) were identified in D. eres P3-1W, with 521 CWDEs consisting of 374 glycoside hydrolases (GHs), 108 carbohydrate esterase (CEs) and 91 polysaccharide lyases (PLs). Additionally, 221 auxiliary activities (AAs), 91 glycosyltransferases (GTs), and 108 carbohydrate binding modules (CBMs) were detected. These findings offer valuable insights into the CAZymes of D. eres P3-1W.

To achieve all-organic polymer with high dielectric performances, we have designed a novel side-chain liquid crystalline polymer (P7) with strong polar mesogen of (Z)-4-(2-cyano-2-phenylvinyl)benzonitrile (CSCN) attached to polycyclooctene backbone. The bis-cyano-substituted CSCN is board-shaped and exhibits a large dipole moment (8.54 D) which tilts ∼34.2° away from its molecular long axis. Consequently, CSCN shows unique dual molecular anisotropy: one from biaxial shape anisotropy and the other from polarization anisotropy. The complex phase behaviors of P7 were investigated employing mainly the techniques of differential scanning calorimetry and X-ray diffraction. Four liquid crystal (LC) phases are identified as K0, K1, K2 and K3, which are SmA, highly-ordered biaxial SmA, B5-like and B7-like, respectively, with the thermal stability increased in sequence. The experimental results indicate that the different LC phases are arisen from the competition and balance between π-π stacking and dipole-dipole interaction. While the face-to-face π-π stacking is dominant in K0 and K1, optimizing the dipole-dipole interaction causes the CSCN mesogens within the smectic layer to tilt and rotate, resulting in K2 and K3. We further investigated the dielectric properties of P7 films using polarization-electric field loops test. The dielectric constant (εr) of P7 is found to be LC structure dependent, which is increased when the LC phase is varied from K0 to K3. With an average εr of 9.7 achieved in K3 and the low dielectric loss (tan δ = 0.001), P7 film offers a promising material in advanced applications like energy storage and electronic devices.

The transient receptor potential mucolipin proteins (TRPMLs) have been shown to be strongly associated with virus infection and other physiological processes in humans, but their tissue distribution and responses to PRRSV in pigs remain unknown.

The Internet era resulted in the rise and advancement of MOOK, WeChat, and mobile networks, making it possible to expand English teaching methods. However, the English teaching industry has the problem of not valuing students’ personalized cognition, and the accuracy of teaching resource delivery is low. Therefore, the research uses the noise gate analysis method to design a cognitive diagnostic model for students and designs an English teaching resource recommendation model in view of a convolutional joint probability matrix (JPM) decomposition algorithm. The research results showed that the cognitive diagnostic model designed in the study had a higher accuracy. Compared to traditional algorithms, the overall recommendation effect of the English teaching resource recommendation model had an average improvement of 11.63% and compared to the JPM algorithm combined with cognitive diagnosis (CD), the overall recommendation effect value had an average improvement of 1.977%. When recommending complex teaching resources, the recommendation effect value had an average improvement of 11.54% compared to traditional algorithms, and the overall average improvement was 1.877% compared to the JPM algorithm combined with CD. In the experimental group, with the assistance of the research algorithm, students’ grades improved by an average of 2.38 points, which was significantly higher than the 0.89 points in the control group. The experiment showcases that the CD and recommendation model designed by the research has higher accuracy, can help improve the efficiency of teaching resource recommendation, reduces teaching costs, and has certain application value.
