Abstract Background Somatic Symptom Disorder (SSD) is a condition often linked to excessive health anxiety and somatic symptoms. In recent years, studies have found associations between the cerebellum and various mental illnesses, including SSD. However, the microstructure of cerebellar subregions in SSD using diffusion magnetic resonance imaging has not been fully defined. Methods This is a cross-sectional study, that included 30 SSD patients and 30 age- and gender-matched healthy controls to investigate the microstructure of the cerebellum using diffusion magnetic resonance imaging. SSD diagnosis followed DSM-5 criteria, excluding major psychiatric comorbidities, while healthy controls underwent rigorous screening to exclude psychiatric or neurological histories. Clinical evaluations utilized standardized scales to assess depressive, anxiety, and cognitive symptoms. MRI data were acquired using a 3T Siemens Prisma scanner, including T1-weighted and diffusion-weighted imaging (30 directions, b = 1000/2000 s/mm²). Multi-compartment diffusion magnetic resonance imaging metrics from free water elimination diffusion tensor imaging and neurite orientation dispersion and density imaging were used to observe microstructural changes in the cerebellum’s white matter and gray matter subregions in SSD patients. Results Compared to the control group, patients with SSD exhibited significant alterations in white matter microstructure. These changes were characterized by increased free water-eliminated fractional anisotropy and neurite density index, as well as decreased free water-eliminated mean diffusivity and radial diffusivity. Furthermore, the cerebellum displayed varying microstructural changes across 26 gray matter subregions. These changes included reduced mean diffusivity, free water-eliminated axial diffusivity, and free water-eliminated radial diffusivity, alongside increased neurite density index and orientation dispersion index. Importantly, the study identified significant correlations between these microstructural changes and clinical symptoms. Specifically, Vermis X and the left lobule VIIb showed significant associations with both depression and anxiety scores. Conclusions The findings suggest greater neurite density and enhanced diffusion restriction in the cerebellum of patients with SSD, which may indicate possible adaptive changes associated with chronic stress.

Abstract Background This study examines how the prevalence of depression and anxiety symptoms differ by socioeconomic status and explores their associations with diabetes in rural southwest China. Methods Data were collected from a cross-sectional health interview and examination survey of 5,005 adults aged ≥ 35 years in rural southwest China. Height, weight, waist circumference, and fasting blood glucose measurement were taken. Depression and anxiety symptoms were assessed using Zung’s Self-rating Depression Scale (SDS) and Self-Rating Anxiety Scale (SAS), respectively. An individual socioeconomic position (SEP) index was constructed using principal component analysis. Results The overall prevalence of depression, anxiety, comorbid depression and anxiety symptoms, and diabetes was 5.4%, 12.8%, 4.0%, and 9.6%, respectively. Abstract Objectives This study was designed to develop and validate models based on delta intratumoral and peritumoral radiomics features from breast masses on dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) for the prediction of axillary lymph node (ALN) pathological complete response (pCR) after neoadjuvant therapy (NAT) in patients with breast cancer (BC). Methods We retrospectively collected data from 187 BC patients with ALN metastases. Radiomics features were extracted from the intratumoral and 3 mm-peritumoral regions on DCE-MRI at baseline and after the 2nd course of NAT to calculate delta intratumoral and peritumoral radiomics features, respectively. After feature selection, the delta intratumoral radiomics (DIR) model and delta peritumoral radiomics (DPR) model were built using the retained features. An ultrasound model was constructed on the basis of preoperative axillary ultrasound results. All variables were screened by univariate and multivariate logistic regression to construct the combined model. The above models were evaluated and compared. Results In the validation set, the ultrasound model had the lowest AUC, which was lower than those of the DIR, DPR and combined models (0.627 vs 0.825, 0.687, 0.846, respectively). Abstract Background This study aimed to uncover the changing prevalence of pre-diabetes and diabetes as well as diabetes self-management behaviors across socioeconomic gradients in rural southwest China. Methods A two-wave, community-based, cross-sectional survey was conducted. A total of 7,747 and 7,536 individuals aged ≥ 35 years, residing in rural southwest China, were selected using a three-stage stratified random sampling method. Demographic characteristics and self-management behaviors were collected through face-to-face interviews, and physical indicators were measured through on-site examinations. An individual socioeconomic position (SEP) index was constructed using principal component analysis based on three variables: education, annual household income, and access to medical services. 6. Identification of the immune infiltration and biomarkers in ulcerative colitis based on liquid–liquid phase separation-related genes

Abstract Liquid–liquid phase separation (LLPS) associates with immune infiltration in multiple diseases. Nonetheless, the role of LLPS-related genes (LLPS-RGs) in immune infiltration of ulcerative colitis (UC) is still elusive. We identified the hub LLPS-RGs (DE-LLPS-RGs) (HSPB3, SLC16A1, TRIM22, SRI, PLEKHG6, GBP1, PADI2) by machine learning algorithms. Hub genes were screened that displayed high prediction accuracy of UC patients. Both the microarray and scRNA-seq datasets showed a strong correlation with immune cell infiltration and cytokines, especially GBP1, TRIM22, SRI. And qRT-PCR analysis showed that GBP1 play a pro-inflammatory role in UC. Two distinct clusters were identified, in which cluster A displayed higher immune infiltration level compared with the cluster B. The top targeted biological pathways of two clusters were distinct, glutamate receptor antagonist ranked top for cluster A while HDAC inhibitor ranked top in cluster B. External cohort and UC cell model validation indicated the similar immune infiltration levels, gene expression and cytokine expression patterns. We determined the seven high accuracy diagnostic genes of UC patients and provide a new perspective on immunoregulation in UC pathogenesis. And suggest patient stratification and candidate targets for precision treatment based on hub genes screened.

7. A multi-scale rotated ship targets detection network for remote sensing images in complex scenarios

Abstract Detecting ship targets in remote sensing images within complex scenarios faces numerous challenges. The limited feature information of small-scale targets and their random orientation angles often result in missed and false detections. To address these issues, this paper proposes a Multi-Scale Rotated Detection Network (MSRO-Net) for detecting rotated ship targets in remote sensing images. The network adopts a CNN-Transformer hybrid architecture for collaborative feature extraction and integrates our proposed Coordinate-Aware Pyramid Feature Aggregation module (CAPP). This backbone network retains the capability of local feature extraction while also connecting global context and capturing long-range dependencies. The model can gather information from any position in the sequence, extract contextual information of targets at different scales, and enhance global feature representation. Additionally, this paper proposes an Upsampling Feature Reconstruction Pyramid (ARFPN-C), based on Adaptive Rotated Convolution (ARC). This network combines ARC adaptive rotating convolution with a multi-scale feature fusion mechanism to enhance the model’s perceptual capability, addressing the issues of limited target feature information and random orientation angles. The proposed algorithm is validated on two public remote sensing image datasets, HRSC2016 and DOTA. The mAP07, mAP12 on the HRSC2016 dataset, as well as the mAP in the ship category of the DOTA dataset, show a significant advantage over other commonly used object detection algorithms, with accuracies of 90.70%, 98.98%, and 89.46%, respectively. These results further validate the accuracy and effectiveness of the proposed MSRO-Net in object detection.

Abstract Background Alzheimer’s disease (AD) is a prevalent neurodegenerative disorder worldwide, and microglia are thought to play a central role in neuroinflammatory events occurring in AD. Chemerin, an adipokine, has been implicated in inflammatory diseases and central nervous system disorders, yet its precise function on microglial response in AD remains unknown. Methods The APP/PS1 mice were treated with different dosages of chemerin-9 (30 and 60 µg/kg), a bioactive nonapeptide derived from chemerin, every other day for 8 weeks consecutively. The primary mouse microglia were stimulated by amyloid beta 42 (Aβ42) oligomers followed by treatment with chemerin-9 in vitro. ChemR23 inhibitor α-NETA was further used to investigate whether the effects of chemerin-9 were ChemR23-dependent. Results We found that the expression of chemerin and ChemR23 was increased in AD. Intriguingly, treatment with chemerin-9 significantly ameliorated Aβ deposition and cognitive impairment of the APP/PS1 mice, with decreased microglial proinflammatory activity and increased phagocytic activity. Similarly, chemerin-9-treated primary microglia showed increased phagocytic ability and decreased NLRP3 inflammasome activation. However, the ChemR23 inhibitor α-NETA abolished the neuroprotective microglial response of chemerin-9. Conclusions Collectively, our data demonstrate that chemerin-9 ameliorates cognitive deficits in APP/PS1 transgenic mice by boosting a neuroprotective microglial phenotype. Graphical Abstract

9. Association between cytokines and progression-free survival in ovarian cancer following CRS/HIPEC treatment

Abstract Background The benefit of cytoreduction with hyperthermic intraperitoneal chemotherapy (CRS/HIPEC) for epithelial ovarian cancer (EOC) remains uncertain. This study investigated the relationship between serum cytokines, particularly monocyte chemoattractant protein-1 (MCP-1), a key inflammatory mediator, and recurrence risk in EOC patients undergoing CRS/HIPEC. Methods From January 2018 to January 2023, serum cytokine levels were analyzed in 34 EOC patients (17 primary, 17 recurrent) before and after CRS/HIPEC using MILLIPLEX Magnetic Bead Panels. Cox proportional hazards regression calculated adjusted hazard ratios (HRs) after controlling for clinical variables. Immunohistochemical (IHC) staining was performed on tissue microarrays from 19 patients. Results Higher 1-unit increment of MCP-1_Baseline were associated with increased recurrence risk within the first year post-CRS/HIPEC (HR: 1.010, 95% CI: 1.000-1.021). After one year, higher 1-unit increments of MCP-1_Post and MCP-1_Change were associated with increased recurrence risk. Lower IL-13 change and higher GROα change were associated with better progression-free survival (PFS) (p = 0.002 and p = 0.025, respectively). IHC analysis showed a trend towards worse PFS within the first year for patients with MCP-1 expression in tumor tissue (HR: 3.252, p = 0.264). Conclusion Cytokines, particularly MCP-1, may help predict PFS following CRS/HIPEC in EOC patients and could inform postoperative treatment decisions. Further research is needed to confirm these findings and explore clinical applications.

10. Constructing a Student Engagement and Learning Development Model in Mobile Learning by SEM

This study aimed to build a model to detect the factors to enhance student engagement and learning development in mobile learning during the COVID-19 Pandemic. Data from a total of 400 junior-high-school students were collected in China in the fall semester of 2020, and a large proportion of students preferred accessing their study with cellphones (67.0%) than with computers (11.8%), laptops (11.3%) or pads (10.0%). Exploratory factor analysis and structural equation modeling (SEM) were used for data analysis. The fitness of the items for each scale of the student engagement and learning development indicated a sufficient fit (X[superscript 2][subscript (19)] = 41.252, GIF = 0.974, AGFI = 0.951, CFI = 0.986, RMSEA = 0.054, NNFI = 0.979, IFI = 0.986). The results of SEM analysis show that emotional engagement is the most important factor (r[superscript 2] = 0.859) in the model, and student engagement has a significant positive impact on learning development in mobile learning. The findings of this study provide a good reference for enhancing student engagement or fostering students' learning development in mobile learning. [For the full proceedings, see ED639391.]

11. Effect of Cs+ on Electrochemical Behavior of La3+ in LiCl-KCl

The main objective of developing molten salt electrolysis refining technology is to achieve the effective separation of Ln/An elements. The examination of the electrochemical behavior of lanthanides, with a particular focus on La3+ during electrolytic refining, provides valuable insights into the impact of accumulated cationic fission products, notably Cs+, on the electrochemical dynamics within a LiCl-KCl molten salt matrix. This investigation employed a comprehensive approach, utilizing advanced simulation techniques alongside experimental methodologies such as cyclic voltammetry, chronoamperometry, linear polarization, and electrochemical impedance spectroscopy, to elucidate the intricate interactions at play in this electrochemical environment. Preliminary evaluations of the electrochemical properties of La3+ ions were conducted on both tungsten(W) and liquid gallium(Ga) electrodes using cyclic voltammetry. The results reveal a notable shift in the reduction potential of La3+ towards more negative values as the concentration of Cs+ increases. This behavior can be ascribed to competitive ion interactions and modifications in the electrochemical milieu, which impede the reduction efficiency of La3+ ions. Subsequent experiments employing chronoamperometry were utilized to elucidate the diffusion behavior of La3+ ions within the molten salt matrix. The results demonstrate a marked decrease in the diffusion coefficient from 1.08×10−5 cm2/s to 4.80×10−6 cm2/s as Cs+ concentration escalated. This decline suggests a notable impediment to ion mobility, likely due to the increased viscosity and ion crowding within the electrolyte as Cs+ accumulates. Further kinetic analyses of the La3+ reduction on liquid Ga cathode were conducted utilizing linear polarization and electrochemical impedance spectroscopy. The investigation reveales a substantial decrease in the exchange current density(i0) from 0.302 A/cm2 to 0.084 5 A/cm2 with increasing Cs+ concentrations, indicating a marked deceleration in the kinetics of the reduction reaction. Concurrently, the activation energy(Ea) for the reduction process increases from 26.77 kJ/mol to 39.24 kJ/mol, suggesting a heightened energetic barrier for the electrochemical transformation. In addition, equivalent circuit modeling of the alternating current impedance spectra indicates that the physical properties of the electrolyte are significantly altered by elevated Cs+ levels. Notably, an increase in solution resistance is observed, further supporting the conclusion that Cs+ presence considerably modifies the electrolyte characteristics, thus impacting the overall electrochemical behavior of lanthanides during refining processes. In summary, the accumulation of cationic fission products, particularly Cs+, within a LiCl-KCl molten salt matrix profoundly influences the electrochemical properties and reaction kinetics associated with lanthanide reduction. This finding underscores the critical importance of thoroughly assessing these interactions to inform the design, optimization, and overall efficacy of electrolytic refining techniques employed in the separation and recovery of valuable lanthanides.

12. The USP11/Nrf2 positive feedback loop promotes colorectal cancer progression by inhibiting mitochondrial apoptosis

Abstract Abnormal antioxidant capacity of cancer is closely related to tumor malignancy. Modulation of oxidative stress status is a novel anticancer therapeutic target. Nrf2 is a key regulator of various antioxidant enzymes, but the mechanism of its deubiquitination remains largely unclear. This study unveiled that Nrf2 received post-transcriptional regulation from a proteasome-associated deubiquitinating enzyme, USP11, in colorectal cancer (CRC). It was found that USP11 was overexpressed in CRC tissues acting as an oncogene by inhibiting mitochondrial apoptosis, and USP11 managed to maintain balance in the production and elimination of reactive oxygen species (ROS). Mechanistically, we identified a feedback loop between USP11 and Nrf2 maintaining the redox homeostasis. USP11 stabilized Nrf2 by deubiquitinating and protecting it from proteasome-mediated degradation. Interestingly, we also map that Nrf2 could bind to the antioxidant reaction element (ARE) in the USP11 promoter to promote its transcription. Hence, USP11/Nrf2 positive feedback loop inhibited mitochondrial apoptosis of CRC cells by activating Nrf2/ARE signaling pathway, thus promoting CRC progression. Schematic diagram of the mechanism by which USP11/Nrf2 positive feedback loop inhibited mitochondrial apoptosis in CRC cells. This study found that USP11 was highly expressed in colorectal cancer (CRC) tissue and was associated with poor prognosis. In CRC, the inhibition of USP11 expression could promote the ubiquitination degradation of Nrf2, thereby inhibiting the Nrf2/ARE signaling pathway. This led to an increase in reactive oxygen species in the cell, causing mitochondrial apoptosis. In addition, Nrf2 could bind to the promoter region of USP11 to promote its transcription, both of which formed positive feedback loop.

Abstract Objectives Cardiovascular disease (CVD) is the leading cause of death in China. This study compared ethnic disparities and lifestyle determinants in the prevalence of CVD (hypertension, coronary heart disease [CHD], and stroke) among older adults of the Han majority and Ha Ni ethnic minority in rural southwest China, to provide evidence for preventing and controlling CVD among older-adult minority communities. Methods A multi-stage stratified random sampling method was used to select 1,413 Han majority participants and 1,402 Ha Ni ethnic minority participants aged ≥ 60 years in rural Southwest China. Data on general demographic characteristics, behavioral lifestyle, and self-reported diagnostic information for patients with CHD and stroke were collected using a standardized questionnaire. The height, weight, waist circumference, and blood pressure of each participant were recorded. The relationship between lifestyle factors and CVD was analyzed using multivariate logistic regression. Abstract Few aerobic hyperthermophilic microorganisms degrade polysaccharides. Here, we describe the genome-enabled enrichment and optical tweezer-based isolation of an aerobic polysaccharide-degrading hyperthermophile, Fervidibacter sacchari, previously ascribed to candidate phylum Fervidibacteria. F. sacchari uses polysaccharides and monosaccharides for growth at 65–87.5 °C and expresses 191 carbohydrate-active enzymes (CAZymes) according to RNA-Seq and proteomics, including 31 with unusual glycoside hydrolase domains (GH109, GH177, GH179). Fluorescence in-situ hybridization and nanoscale secondary ion mass spectrometry confirmed rapid assimilation of 13C-starch in spring sediments. Purified GHs were optimally active at 80–100 °C on ten different polysaccharides. Finally, we propose reassigning Fervidibacteria as a class within phylum Armatimonadota, along with 18 other species, and show that a high number and diversity of CAZymes is a hallmark of the phylum, in both aerobic and anaerobic lineages. Our study establishes Fervidibacteria as hyperthermophilic polysaccharide degraders in terrestrial geothermal springs and suggests a broad role for Armatimonadota in polysaccharide catabolism.

Four new lactones, including hypomonacid A (1) and hypomonone A–C (4–6), as well as nine known polyketide analogues (2–3 and 7–13) were obtained from endophytic fungus Hypomontagnella sp. TX-09 derived from Santalum album. Their planar structures were extensively established by analysing HRESIMS and NMR spectroscopic data. Stereochemistry of new compounds was determined by X-ray diffraction analysis and modified Mosher’s method in combination with quantum-chemical ECD calculation. In addition, compounds 1 and 2 showed anti-inflammatory activity by inhibition of lipopolysaccharide (LPS)-induced NO production in RAW264.7 cells at 50 μmol/L without cytotoxicity. Among them, compound 1 inhibited the production of LPS-stimulated inflammation in mouse macrophage RAW264.7 cells by suppressing the expression of iNOS, TNF-α, IL-1β, and IL-6.

This study utilizes VR technology to explore the influences of the number of bystanders and gender on the moral courage process (i.e., emotions, perceived personal responsibility, perceived self-efficacy in intervention, and cost-benefit analysis of taking action) and bystander behaviors among junior high schools in Taiwan. Employing a 2 (gender: boys vs. girls) x 3 (number of bystanders: 0 vs.2 vs. 4) experimental design, 209 middle school students from midwestern Taiwan experienced VR bullying scenarios and completed questionnaires. MANCOVAs and ANCOVAs results indicated no significant effect of the number of bystanders on the moral courage process and bystander behaviors. However, gender effects were observed, with girls considering more cost evaluation and displaying more defending behaviors, while boys showed more pro-bullying behaviors. Regression analysis revealed gender differences in the mechanisms underlying bystander behaviors. For boys, guilt, defender self-efficacy, and perceived personal responsibility positively predicted defending behaviors. For girls, empathy and defender self-efficacy positively predicted defending behaviors. Boys' empathy negatively predicted outsider behaviors, but cost evaluation showed the positive effect. Girls’ guilt and defender self-efficacy negatively predicted outsider behaviors, while a greater number of bystanders and perceived personal responsibility negatively predicted their pro-bullying behaviors. Quality data reveal that some students declared their intention to intervene based on personal ethics or obligations regardless of the presences of other bystanders, while others reported being more willing to intervene when more bystanders were present. Findings partially support moral courage model. This study demonstrates the potential of VR as an educational tool for developing bystander intervention skills among high school students.

Nasopharyngeal carcinoma (NPC) is a malignant tumor predominantly influenced by Epstein-Barr virus infection and genetic factors. The transforming growth factor-beta (TGF-β) superfamily is implicated in various cellular processes, including tumorigenesis. This study aimed to detect the role of one TGF-β superfamily member activin receptor type IIB (ACTRIIB) in NPC. This study analyzed NPC datasets, including GSE12452, GSE102349, and GSE53819. ACTRIIB expression and N-glycosylation levels were assessed by western blot, real-time PCR, immunofluorescence, and immunohistochemistry in NPC cells and tissues. As indicated by the datasets, ACTRIIB was significantly upregulated in NPC tissues, and the up-regulation was associated with poor prognosis. This study confirmed the N-glycosylation of ACTRIIB primarily at the forty-second amino acid, an asparagine. The N-glycosylation of ACTRIIB promoted the localization of ACTRIIB to the cell membrane and prevented the degradation of the protein by lysosomes, through which ACTRIIB activated the downstream Smard1/2 to promote tumor cell proliferation and invasion. Inhibition of N-glycosylation or knockdown of ACTRIIB resulted in reduced cell proliferation and invasion and increased the cell sensitivity to docetaxel. In conclusion, N-glycosylation of ACTRIIB was a critical post-translational modification that enhanced protein stability and induced membrane localization, which facilitates the functions of ACTRIIB in cell proliferation and invasion in NPC. Inhibition of ACTRIIB N-glycosylation could potentially serve as a therapeutic strategy to improve the efficacy of chemotherapy in NPC.

18. Older adults’ Internet use behavior and its association with accelerometer-derived physical activity

ObjectiveThe aging population is thriving worldwide, and it is critical to improve the health of older adults through physical activity (PA). Although the Internet can promote PA in older adults, limited studies have used objective tools to measure it. Thus, we aimed to investigate the association between the frequency of Internet use and PA levels in older adults.MethodsFor this cross-sectional study, we employed convenience sampling. The participants were 172 adults aged 65 and older without cognitive impairment who could walk independently. We measured PA using a triaxial accelerometer, step counts, and moderate-to-vigorous PA (MVPA). We measured Internet use via a self-report questionnaire, separated by frequency of use into high, moderate, and low or no use. We performed a one-way analysis of variance (ANOVA) and multiple logistic regression to examine the relationship between Internet use and PA.ResultsWe included a total of 172 older adults (72.0 ± 5.5 years; 78.5% female). The group that used the Internet with moderate frequency exhibited higher daily step counts than the group that used the Internet with low frequency or not at all (moderate frequency: 7,888 steps > low frequency or no use: 6,070.6 steps). Compared to the group that used the Internet at a moderate frequency, older adults with a low frequency or those who did not use the Internet were less likely to meet the recommendations of the World Health Organization (WHO) regarding PA [odds ratio (OR): 0.242; 95% confidence interval (CI): 0.077–0.751].ConclusionOlder adults with a low frequency of Internet use or those who did not use the Internet (i.e., those who used the Internet less than once a week or not at all, respectively) were less likely to meet the WHO’s recommended levels for PA than older adults who used the Internet with moderate frequency. The findings of this study can inform efforts to reduce age-related health risks and promote strategies for encouraging PA.

Abstract Background Numerous reports indicate that both obesity and type 2 diabetes mellitus (T2DM) are factors associated with cognitive impairment (CI). The objective was to assess the relationship between abdominal obesity as measured by waist-to-hip ratio adjusted for body mass index (WHRadjBMI) and CI in middle-aged and elderly patients with T2DM. Methods A cross-sectional study was conducted, in which a total of 1154 patients with T2DM aged ≥ 40 years were included. WHRadjBMI was calculated based on anthropometric measurements and CI was assessed utilizing the Montreal Cognitive Assessment (MoCA). Participants were divided into CI group (n = 509) and normal cognition group (n = 645). Correlation analysis and binary logistic regression were used to explore the relationship between obesity-related indicators including WHRadjBMI, BMI as well as waist circumference (WC) and CI. Meanwhile, the predictive power of these indicators for CI was estimated by receiver operating characteristic (ROC) curves. Results WHRadjBMI was positively correlated with MoCA scores, independent of sex. The Area Under the Curve (AUC) for WHRadjBMI, BMI and WC were 0.639, 0.521 and 0.533 respectively, and WHRadjBMI had the highest predictive power for CI. Whether or not covariates were adjusted, one-SD increase in WHRadjBMI was significantly related to an increased risk of CI with an adjusted OR of 1.451 (95% CI: 1.261–1.671). Abstract Monoterpenoid indole alkaloid (MIA) biosynthesis in Catharanthus roseus is a paragon of the spatiotemporal complexity achievable by plant specialized metabolism. Spanning a range of tissues, four cell types, and five cellular organelles, MIA metabolism is intricately regulated and organized. This high degree of metabolic differentiation requires inter-cellular and organellar transport, which remains understudied. Here, we have characterized a vacuolar importer of secologanin belonging to the multidrug and toxic compound extrusion (MATE) family, named CrMATE1. Phylogenetic analyses of MATEs suggested a role in alkaloid transport for CrMATE1, and in planta silencing in two varieties of C. roseus resulted in a shift in the secoiridoid and MIA profiles. Subcellular localization of CrMATE1 confirmed tonoplast localization. Biochemical characterization was conducted using the Xenopus laevis oocyte expression system to determine substrate range, directionality, and rate. We can confirm that CrMATE1 is a vacuolar importer of secologanin, translocating 1 mM of substrate within 25 min. The transporter displayed strict directionality and specificity for secologanin and did not accept other secoiridoid substrates. The unique substrate-specific activity of CrMATE1 showcases the utility of transporters as gatekeepers of pathway flux, mediating the balance between a defense arsenal and cellular homeostasis.

Abstract Objective To investigate the impact of vitamin A (VA), vitamin D (VD), and homocysteine (Hcy) on cardiometabolic multimorbidity (CMM). Methods This study is a cross-sectional study conducted in Ningxia Province, China. A total of 5000 participants aged 25–74 were recruited and divided into two groups based on the definition of cardiometabolic multimorbidity: the CMM group and the Non CMM group. Demographic, lifestyle, and laboratory data were collected to investigate the correlation between vitamin A, D, Hcy levels and CMM risk. The association was analyzed using multiple logistic regression and restricted cubic spline method. Results CMM incidence increased with age, being higher in females (20.05%) compared to males, Hypertension was present in 96.20% of CMM cases. Reduced VD levels correlated with an elevated CMM risk (OR = 1.799, 95% CI: 1.466–2.238), showing an inverse dose-response relationship, even after adjusting for confounders (OR = 1.553, 95% CI: 1.233–1.956). However, VA and Hcy levels were not significantly associated with CMM risk. Abstract Background The Atribacterota are widely distributed in the subsurface biosphere. Recently, the first Atribacterota isolate was described and the number of Atribacterota genome sequences retrieved from environmental samples has increased significantly; however, their diversity, physiology, ecology, and evolution remain poorly understood. Results We report the isolation of the second member of Atribacterota, Thermatribacter velox gen. nov., sp. nov., within a new family Thermatribacteraceae fam. nov., and the short-term laboratory cultivation of a member of the JS1 lineage, Phoenicimicrobium oleiphilum HX-OS.bin.34TS, both from a terrestrial oil reservoir. Physiological and metatranscriptomics analyses showed that Thermatribacter velox B11T and Phoenicimicrobium oleiphilum HX-OS.bin.34TS ferment sugars and n-alkanes, respectively, producing H2, CO2, and acetate as common products. Comparative genomics showed that all members of the Atribacterota lack a complete Wood-Ljungdahl Pathway (WLP), but that the Reductive Glycine Pathway (RGP) is widespread, indicating that the RGP, rather than WLP, is a central hub in Atribacterota metabolism. Ancestral character state reconstructions and phylogenetic analyses showed that key genes encoding the RGP (fdhA, fhs, folD, glyA, gcvT, gcvPAB, pdhD) and other central functions were gained independently in the two classes, Atribacteria (OP9) and Phoenicimicrobiia (JS1), after which they were inherited vertically; these genes included fumarate-adding enzymes (faeA; Phoenicimicrobiia only), the CODH/ACS complex (acsABCDE), and diverse hydrogenases (NiFe group 3b, 4b and FeFe group A3, C). Finally, we present genome-resolved community metabolic models showing the central roles of Atribacteria (OP9) and Phoenicimicrobiia (JS1) in acetate- and hydrocarbon-rich environments. Conclusion Our findings expand the knowledge of the diversity, physiology, ecology, and evolution of the phylum Atribacterota. This study is a starting point for promoting more incisive studies of their syntrophic biology and may guide the rational design of strategies to cultivate them in the laboratory. Video Abstract

Abstract Background Although several studies have found the relationship between essential elements and diabetes, the studies about the association of essential elements with diabetes diagnosed according to an oral glucose tolerance test (OGTT) and glycated hemoglobin (HbA1c) in a sex- and age-specific manner were limited. To investigate the linear and nonlinear relationship of five essential elements including iron (Fe), copper (Cu), Zinc (Zn), magnesium (Mg), and calcium (Ca) with diabetes, fasting plasma glucose (FPG), 2-h postprandial plasma glucose (PPG), and HbA1c and to evaluate the sex- and age-specific heterogeneities in these relationships. Methods A total of 8392 community-dwelling adults were recruited to complete a questionnaire and undergo checkups of anthropometric parameters and serum levels of five metals (Fe, Cu, Zn, Mg, and Ca). The multivariable logistic and linear regression, the restricted cubic spline (RCS) analysis, and subgroup analysis were applied to find the associations between the essential elements and the prevalence of diabetes as well as FPG, PPG, and HbA1c. Abstract Background Kidney Renal Clear Cell Carcinoma (KIRC) is a common malignant tumor of the urinary system, and its incidence is increasing. ERBB3 binding protein (EBP1) is upregulated in various cancers. However, the connection between EBP1 and KIRC has not been reported. Methods The expression of EBP1 in normal kidney tissue and KIRC tissue was analyzed through database and tissue microarray. EBP1 was knocked down in KIRC cell lines, and its impact on KIRC proliferation was assessed through CCK-8, soft agar assay, and flow cytometry. Scratch and transwell assays were used to evaluate the influence of EBP1 on KIRC invasion and migration. Nude mice tumor experiment were conducted to examine the effect of EBP1 on tumor tissue. Database analysis explored potential pathways involving EBP1, and validation was performed through Western blot experiments and p38 inhibitor. Results EBP1 is upregulated in KIRC and significantly correlates with clinical staging, pathological grading, and lymph node metastasis in patients. The mechanism research showed that knocking down EBP1 inhibited KIRC proliferation, invasion, and migration and inhibited p38 phosphorylation and the expression of hypoxia-inducible factor-1α (HIF-1α) in KIRC. p-38 inhibitor (SB203580) inhibits p38 phosphorylation and HIF-1α expression and suppresses cell viability in a concentration-dependent manner, but has no effect on EBP1 expression. HEK 293T cells overexpressing EBP1 showed increased expression of phosphorylated p38 and HIF-1α and enhanced cell viability, however, SB203580 inhibited this effect of EBP1. Conclusion EBP1 may promote the occurrence and development of KIRC by regulating the expression of p38/HIF-1α signaling pathway. Graphical Abstract

BackgroundAflatoxin B1 (AFB1), a potent carcinogen produced by Aspergillus species, is a prevalent contaminant in oil crops, with prolonged exposure associated with liver damage. Home-made peanut oil (HMPO) produced by small workshops in Guangzhou is heavily contaminated with AFB1. Despite the enactment of the Small Food Workshops Management Regulations (SFWMR), no quantitative assessment has been conducted regarding its impact on food contamination and public health. The study aims to assess the impact of SFWMR on AFB1 contamination in HMPO and liver function in the population.MethodAFB1 contamination in HMPO were quantified using high-performance liquid chromatography and liver function data were obtained from the health center located in a high-HMPO-consumption area in Guangzhou. Interrupted time series and mediation analyses were employed to assess the relationship between the implementation of SFWMR, AFB1 concentrations in HMPO, and liver function among residents.ResultThe AFB1 concentrations in HMPO were 1.29 (0.12, 6.58) μg/kg. The average daily intake of AFB1 through HMPO for Guangzhou residents from 2010 to 2022 ranged from 0.25 to 1.68 ng/kg bw/d, and the Margin of Exposure ranged from 238 to 1,600. The implementation of SFWMR was associated with a significant reduction in AFB1 concentrations in HMPO, showing an immediate decrease of 2.865 μg/kg (P = 0.006) and a sustained annual reduction of 2.593 μg/kg (P = 0.034). Among residents in the high-HMPO-consumption area, the implementation of SFWMR was significantly associated with a reduction in the prevalence of liver function abnormality (PR = 0.650, 95% CI: 0.469–0.902). Subgroup analysis revealed that this reduction was significantly associated with the implementation of SFWMR in the female (PR = 0.484, 95% CI: 0.310–0.755) and in individuals aged ≥ 60 years (PR = 0.586, 95% CI: 0.395–0.868). Mediation analysis demonstrated that AFB1 concentrations in HMPO fully mediated the relationship between the implementation of SFWMR and the liver function abnormality (PR = 0.981, 95% CI: 0.969–0.993).ConclusionIn Guangzhou, the public health issue arising from AFB1 intake through HMPO warrants attention. The implementation of SFWMR had a positive impact on the improvement of AFB1 contamination in HMPO and the liver function. Continued efforts are necessary to strengthen the enforcement of the regulations. The exposure risks to AFB1 among high-HMPO-consumption groups also demand greater focus.

AbstractBackground We aimed to investigate the associations between thyroid hormone sensitivity indices and diabetes in euthyroid adults in the United States and China.Methods 2296 euthyroid adults from the NHANES in the United States and 8319 euthyroid adults from the SPEED-Shunde in China were involved. The thyroid sensitivity indices, namely TFQIFT4 and TFQIFT3, were calculated. Multivariable logistic regression, restricted cubic spline analysis, and general ordinal logit regression were utilized.Results In the NHANES, compared with participants in quartile 1st (Q1), those in Q4 of TFQIFT3 (OR 2.12, 95% CI (1.18, 3.81)) and those in Q3 of TFQIFT4 (OR 2.31, 95% CI (1.18, 4.53)) (both P for trend < 0.05) were associated with a greater prevalence of diabetes. In the SPEED-Shunde, compared with participants in Q1, those in Q4 of TFQIFT3 had a greater prevalence of diabetes (OR 1.36, 95% CI (1.11, 1.66) (P for trend < 0.05), while no significant associations between TFQIFT4 and diabetes were found.Conclusions TFQIFT3 was associated with a higher prevalence of diabetes both in the United States and China. However, TFQIFT4 was only associated with a higher prevalence of diabetes in the United States, not in China. Further prospective cohort studies are necessary to validate these findings.

ABSTRACTThe gut commensal bacteria Christensenellaceae species are negatively associated with many metabolic diseases, and have been seen as promising next-generation probiotics. However, the cultured Christensenellaceae strain resources were limited, and their beneficial mechanisms for improving metabolic diseases have yet to be explored. In this study, we developed a method that enabled the enrichment and cultivation of Christensenellaceae strains from fecal samples. Using this method, a collection of Christensenellaceae Gut Microbial Biobank (ChrisGMB) was established, composed of 87 strains and genomes that represent 14 species of 8 genera. Seven species were first described and the cultured Christensenellaceae resources have been significantly expanded at species and strain levels. Christensenella strains exerted different abilities in utilization of various complex polysaccharides and other carbon sources, exhibited host-adaptation capabilities such as acid tolerance and bile tolerance, produced a wide range of volatile probiotic metabolites and secondary bile acids. Cohort analyses demonstrated that Christensenellaceae and Christensenella were prevalent in various cohorts and the abundances were significantly reduced in T2D and OB cohorts. At species level, Christensenellaceae showed different changes among healthy and disease cohorts. C. faecalis, F. tenuis, L. tenuis, and Guo. tenuis significantly reduced in all the metabolic disease cohorts. The relative abundances of C. minuta, C. hongkongensis and C. massiliensis showed no significant change in NAFLD and ACVD. and C. tenuis and C. acetigenes showed no significant change in ACVD, and Q. tenuis and Geh. tenuis showed no significant change in NAFLD, when compared with the HC cohort. So far as we know, this is the largest collection of cultured resource and first exploration of Christensenellaceae prevalences and abundances at species level.

Objective The association of appendicular skeletal muscle mass (ASM), grip strength and fat-to-muscle ratio (FMR) and the progression of metabolic dysfunction-associated steatotic liver disease (MASLD) are not well known.Materials and methods This study included participants older than 40 years who underwent bioelectrical impedance assessment in Prevalence of Metabolic Diseases and Risk Factors in Shunde (SPEED-Shunde). We measured grip strength with an electronic grip strength metre. ASM and grip strength were adjusted by dividing body mass index (BMI). FMR was calculated as total fat mass to total muscle mass. Liver steatosis and liver fibrosis were evaluated by vibration-controlled transient elastography. Multifactorial logistic regression was used to analyse the relationship between ASM, grip strength, FMR, and MASLD or MASLD-associated liver fibrosis. We performed subgroup analyses according to sex, age and BMI. Interaction tests and linear trend tests were also conducted.Results This study included a total of 3277 participants. FMR was positively associated with MASLD (OR: 1.89, 95% CI: 1.66–2.15) and MASLD-associated liver fibrosis (OR: 1.70, 95% CI: 1.22–2.37). While ASM/BMI (OR: 0.59, 95% CI: 0.52–0.67) or grip strength/BMI (OR: 0.72, 95% CI: 0.66–0.78) were negatively associated with MASLD. Interactions were observed between ASM/BMI and age, grip strength and sex in MASLD, as well as FMR and MASLD-associated liver fibrosis.Conclusion In a middle-to-elderly aged population, FMR was positively associated with the risk of MASLD and MASLD-associated liver fibrosis, and muscle mass and grip strength were negatively associated with MASLD, rather than MASLD-associated liver fibrosis.

It is economically desirable to develop a material that can simultaneously detect and recover uranium. Herein, a CC-bridged two-dimensional metal-covalent organic framework (Cu-BTAN-AO MCOF) was constructed by condensation of metal single crystals with a rigid structure (Cu3(PyCA)3) and cyano monomers (BTAN) via Knoevenagel reaction for simultaneous detection and adsorption of uranium. The amidoxime group within the pore and the presence of unsaturated Cu(I) in the framework facilitate the adsorption of uranyl ions onto the amidoxime group, leading to fluorescence quenching via the photoinduced electron transfer (PET) mechanism, achieving a detection limit of as low as 167 nM uranyl ions. Furthermore, Cu-BTAN-AO demonstrates exceptional efficiency in capturing uranium from wastewater characterized by rapid kinetics and superior selectivity. It is noteworthy that Cu-BTAN-AO is the first example of simultaneous detection, adsorption and chemical reduction of uranium using metal centers and functional groups in MCOF, indicating that Cu-BTAN-AO has great potential for the detection and recovery of uranium-containing wastewater. This design strategy may also be applicable to advancing sensing and energy materials for other important metal ions.

Several transcription factors intricately participate in the initiation and progression of associated processes. Among these, nuclear factor kappa B (NF-κB), a pleiotropic transcription factor, assumes a crucial role in regulating oxidative stress, inflammatory response, endothelial dysfunction, myocardial fibrosis and hypertrophy, and cardiomyocyte apoptosis in DCM. However, the precise role of NF-κB in DCM remains a subject of ongoing investigation. This article provides a comprehensive review of NF-κB's involvement in several processes associated with DCM.

As a natural emulsifier, egg white protein (EWP) has great interfacial characteristics and high security, and has broad development prospects. This study explored the impact of ultrasound synergized citral (CI) treatment on the microstructure, molecular flexibility and emulsifying property of EWP, and predicted the interaction between CI and ovalbumin (the main protein in EWP) through molecular docking. The decrease in free amino content and the growth in molecular weight of EWP suggested that CI and proteins were successfully grafted. The results of physicochemical properties revealed that UCEWP (ultrasound synergized citral-treated EWP) had smaller particle size and larger ζ-potential absolute value, which meant that the stability of UCEWP system was enhanced. From the perspective of interfacial characteristics, UCEWP had lower interfacial tension, which remarkably improved its emulsifying property. The emulsifying activity index (EAI) and emulsifying stability index (ESI) of UCEWP were 1.99 times and 3.19 times higher than that of natural EWP (NEWP). Analysis of Fourier transform infrared spectroscopy (FT-IR) and fluorescence spectroscopy illustrated that the secondary and tertiary structures of UCEWP were more disordered and stretched than those of EWPs. Protein microstructure demonstrated that UCEWP presented loose small particle distribution, and correlation analysis reflected that the improvement of molecular flexibility was positively correlated with the enhancement of emulsifying property. These results elucidated that ultrasound synergized CI treatment is an effective mean to improve the molecular flexibility and emulsifying property of EWP, which provides a valuable reference for further application of EWP.

Fasting blood glucose, blood pressure, height, weight and waist circumference (WC) were measured for each participant. SEM was employed to assess the relationships between demographic characteristics (sex, age and ethnicity), socioeconomic position (SEP; annual household income, education level and access to medical services), lifestyle factors (obesity status (body mass index and WC) and physical inactivity), hypertension, hyperlipidaemia and family history of diabetes.Setting This study was conducted in rural Yunnan Province of China.Participants 7536 individuals aged ≥35 years consented to participate in the study.Results The overall prevalence of diabetes in the present study was 8.3%. Prevalence did not differ by gender (prevalence for both men and women was 8.3% (p>0.05)). 38. Individualised cumulative cisplatin dose for locoregionally advanced nasopharyngeal carcinoma patients based on induction chemotherapy response and tumour volume

Background and objectives: To evaluate the prognostic value of an integrated model consisting of tumour response to induction chemotherapy (IC) and gross tumour volume (GTV) after IC in nasopharyngeal carcinoma (NPC) and elucidate optimal cumulative cisplatin dose (CCD) in concurrent chemoradiotherapy (CCRT) for different subgroups. Design and methods: This retrospective study enrolled 896 patients with NPC diagnosed from 2010 to 2017 receiving IC plus radiotherapy. The primary endpoint was disease-free survival (DFS). Cut-off points for GTV were combined with IC response to develop an integrated model. Propensity score matching (PSM) was used to adjust for potential confounders. Survival outcomes and acute toxicity were compared between the different CCD groups. Results: Unsatisfactory IC response and large GTV after IC were correlated with poor survival outcomes; the AUC increased to 0.668 when these factors were incorporated. The integrated model classified patients into three groups. After PSM, radiotherapy alone and CCRT demonstrated similar efficacy in the low-risk group (complete response (CR)/partial response (PR) and GTV 200 mg/m 2 and 101–200 mg/m 2 increased the 5-year DFS rates (83.7% vs 81.1% vs 65.3%, p = 0.042). In the high-risk group (stable disease/progressive disease and any GTV), the use of different CCDs did not result in significantly different survival outcomes ( p = 0.793). Additionally, high CCD was significantly associated with increased incidence of grade 1–4 acute toxicity. Conclusion: The integrated model incorporating IC response and GTV after IC demonstrates satisfactory value in risk stratification and the potential to guide individualised decision-making in CCD selection. Balancing toxicity and efficacy, RT alone seems to be the optimal treatment for patients in low-risk groups and 200 mg/m 2 might be the optimal dose for intermediate-risk groups. Moreover, increasing CCD does not benefit patients in high-risk groups, and treatment options for these patients require further consideration.

Isolation and identification of acid-producing bacteria from environmental samples is still one of important and effective approaches to obtain strains producing lactic acid with high optical purity. In this study, an isolate strain named HL64-1, producing D-lactic acid with high optical purity, was isolated from rotten fruit. And HL64-1 was characterized as Leuconostoc pseudomesenteroides based on morphology, 16S rDNA sequence analysis and sequence similarity Blast analysis. After fermentation for 24 h, 62.18 g/L D-lactic acid was produced with an optical purity of 99.90%(ee) in basic fermentation culture. The acid production rate was 2.59 g/(L·h). Scale-up fermentation of L. pseudomesenteroides HL64-1 in 5-L fermenter for 72 h produced 78.74 g/L D-lactic acid by replenishment of carbon sources, and the average acid production rate was 1.09 g/(L·h). Moreover, L. pseudomesenteroides HL64-1 was found to be capable of using effectively cheap raw materials, such as agricultural by-products peanut cake meal and cottonseed meal, as alternative nitrogen sources, leading a reduction of fermentation cost. In addition, L. pseudomesenteroides HL64-1 could metabolize xylose into D-lactic acid. Co-fermentation of glucose and xylose mixtures showed that glucose significantly enhanced the utilization efficiency of xylose, making L. pseudomesenteroides HL64-1 to be a promising strain for pure D-lactic acid industrial production.

Inflammatory bowel disease (IBD) comprises a group of idiopathic intestinal disorders, including ulcerative colitis and Crohn’s disease, significantly impacting the quality of life for affected individuals. The effective management of these conditions remains a persistent challenge. The NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome, a complex molecular structure, regulates the production of pro-inflammatory cytokines such as interleukin-1β. Abnormal activation of the NLRP3 inflammasome plays a pivotal role in the development of IBD, making it a compelling target for therapeutic intervention. Our research revealed that cinnamaldehyde (CA), a major bioactive compound found in the leaves of Cinnamomum osmophloeum kaneh, demonstrated a remarkable ability to alleviate colitis induced by dextran sulfate sodium (DSS) in a mouse model. This effect was attributed to CA’s ability to downregulate the activation of the NLRP3 inflammasome and reduce the expression of pro-inflammatory mediators in the colon. In the mechanism study, we observed that CA inhibited the NLRP3 inflammasome in macrophages, at least partially, by enhancing the autophagic response, without reducing mitochondrial damage. These findings collectively suggest that CA holds significant potential as a therapeutic agent for enhancing the management of IBD, offering a promising avenue for further research and development.

43. Prevalence and determinants of nicotine dependence among rural elderly of four ethnic groups in Yunnan province: a cross-sectional study with structural equation modeling analysis

ObjectiveTo understand the prevalence and influencing factors of nicotine dependence among rural elderly of Han, Dai, Hani, and Bai ethnicity in Yunnan province, to provide a reference for developing strategies to alleviate nicotine dependence symptoms in the population. MethodsA face-to-face survey was conducted among 5 800 residents aged ≥ 60 years recruited by multistage stratified random sampling from the above four ethnic groups in rural areas of Yunnan province from July 2021 to May 2022. The Fagerström Test of Nicotine Dependence (FTND), the Social Support Rating Scale (SSRS) developed by Chinese scholars, and a self-developed questionnaire were used in the study. A composite socioeconomic status (SES) score was constructed using principal component analysis, and the factors influencing nicotine dependence among these ethnic groups were analyzed using structural equation modeling (SEM). ResultsOf the 5 642 participants finally included in the analysis, 935 (16.57%) were classified as having nicotine dependence, and the prevalence of nicotine dependence among participants from Han, Dai, Hani, and Bai ethnic groups was 17.34%, 11.64%, 16.41%, and 20.87%, respectively, with a significant difference in prevalence among ethnic groups (χ2 = 44.410, P < 0.001). The results of the SEM analysis indicated that gender, ethnicity, SES, and smoking cessation attempt had both direct and indirect effects on nicotine dependence, with overall standardized path coefficients of – 0.33, – 0.03, 0.08, and – 0.06, respectively. Gender had an indirect effect through number of cigarettes smoked per day, while ethnicity and attempt to quit smoking influenced nicotine dependence indirectly through social support, and SES had indirect effects through both social support and number of cigarettes smoked per day, with standardized path coefficients of – 0.07, – 0.01, – 0.01, and 0.03, respectively. Social support and number of cigarettes smoked per day had direct effects on nicotine dependence, with standardized path coefficients of – 0.06 and 0.18, respectively. ConclusionThere are significant ethnic differences in nicotine dependence among rural elderly in Yunnan province, especially among males, those with a higher number of cigarettes smoked per day, at a higher SES, having a lower score of social support, those who have not tried to quit smoking, and the Bai minority group, highlighting the need for targeted prevention and control measures.

49. Dihydroartemisinin Modulates Enteric Glial Cell Heterogeneity to Alleviate Colitis

Abstract The precise mechanism underlying the therapeutic effects of dihydroartemisinin (DHA) in alleviating colitis remains incompletely understood. A strong correlation existed between the elevation of glial fibrillary acidic protein (GFAP)+/S100 calcium binding protein B (S100β)+ enteric glial cells (EGCs) in inflamed colonic tissues and the disruption of the intestinal epithelial barrier (IEB) and gut vascular barrier (GVB) observed in chronic colitis. DHA demonstrated efficacy in restoring the functionality of the dual gut barrier while concurrently attenuating intestinal inflammation. Mechanistically, DHA inhibited the transformation of GFAP+ EGCs into GFAP+/S100β+ EGCs while promoting the differentiation of GFAP+/S100β+ EGCs back into GFAP+ EGCs. Furthermore, DHA induced apoptosis in GFAP+/S100β+ EGCs by inducing cell cycle arrest at the G0/G1 phase. The initial mechanism is further validated that DHA regulates EGC heterogeneity by improving dysbiosis in colitis. These findings underscore the multifaceted therapeutic potential of DHA in ameliorating colitis by improving dysbiosis, modulating EGC heterogeneity, and preserving gut barrier integrity, thus offering promising avenues for novel therapeutic strategies for inflammatory bowel diseases.

50. Pediatric Sedation Assessment and Management System (PSAMS) for Pediatric Sedation in China: Development and Implementation Report

Abstract BackgroundRecently, the growing demand for pediatric sedation services outside the operating room has imposed a heavy burden on pediatric centers in China. There is an urgent need to develop a novel system for improved sedation services. ObjectiveThis study aimed to develop and implement a computerized system, the Pediatric Sedation Assessment and Management System (PSAMS), to streamline pediatric sedation services at a major children’s hospital in Southwest China. MethodsPSAMS was designed to reflect the actual workflow of pediatric sedation. It consists of 3 main components: server-hosted software; client applications on tablets and computers; and specialized devices like gun-type scanners, desktop label printers, and pulse oximeters. With the participation of a multidisciplinary team, PSAMS was developed and refined during its application in the sedation process. This study analyzed data from the first 2 years after the system’s deployment. Implementation (Results)From January 2020 to December 2021, a total of 127,325 sedations were performed on 85,281 patients using the PSAMS database. Besides basic variables imported from Hospital Information Systems (HIS), the PSAMS database currently contains 33 additional variables that capture comprehensive information from presedation assessment to postprocedural recovery. The recorded data from PSAMS indicates a one-time sedation success rate of 97.1% (50,752/52,282) in 2020 and 97.5% (73,184/75,043) in 2021. The observed adverse events rate was 3.5% (95% CI 3.4%‐3.7%) in 2020 and 2.8% (95% CI 2.7%-2.9%) in 2021. ConclusionsPSAMS streamlined the entire sedation workflow, reduced the burden of data collection, and laid a foundation for future cooperation of multiple pediatric health care centers.
