Abstract

The Aria project consists of a plant, hosting a 350 m cryogenic isotopic distillation column, the tallest ever built, which is currently in the installation phase in a mine shaft at Carbosulcis S.p.A., Nuraxi-Figus (SU), Italy. Aria is one of the pillars of the argon dark-matter search experimental program, lead by the Global Argon Dark Matter Collaboration. Aria was designed to reduce the isotopic abundance of $^{39}$Ar, a $\beta$-emitter of cosmogenic origin, whose activity poses background and pile-up concerns in the detectors, in the argon used for the dark-matter searches, the so-called Underground Argon (UAr). In this paper, we discuss the requirements, design, construction, tests, and projected performance of the plant for the isotopic cryogenic distillation of argon. We also present the successful results of isotopic cryogenic distillation of nitrogen with a prototype plant, operating the column at total reflux.

Abstract

The Aria cryogenic distillation plant, located in Sardinia, Italy, is a key component of the DarkSide-20k experimental program for WIMP dark matter searches at the INFN Laboratori Nazionali del Gran Sasso, Italy. Aria is designed to purify the argon, extracted from underground wells in Colorado, USA, and used as the DarkSide-20k target material, to detector-grade quality. In this paper, we report the first measurement of argon isotopic separation by distillation with the 26 m tall Aria prototype. We discuss the measurement of the operating parameters of the column and the observation of the simultaneous separation of the three stable argon isotopes: $${}^{36}\hbox {Ar}$$ 36 Ar , $${}^{38}\textrm{Ar}$$ 38 Ar , and $${}^{40}\textrm{Ar}$$ 40 Ar . We also provide a detailed comparison of the experimental results with commercial process simulation software. This measurement of isotopic separation of argon is a significant achievement for the project, building on the success of the initial demonstration of isotopic separation of nitrogen using the same equipment in 2019.

Abstract

Aria is a plant hosting a ${350}\,\hbox {m}$ cryogenic isotopic distillation column, the tallest ever built, which is being installed in a mine shaft at Carbosulcis S.p.A., Nuraxi-Figus (SU), Italy. Aria is one of the pillars of the argon dark-matter search experimental program, lead by the Global Argon Dark Matter Collaboration. It was designed to reduce the isotopic abundance of ${^{39}\hbox {Ar}}$ in argon extracted from underground sources, called Underground Argon (UAr), which is used for dark-matter searches. Indeed, ${^{39}\hbox {Ar}}$ is a $\beta $-emitter of cosmogenic origin, whose activity poses background and pile-up concerns in the detectors. In this paper, we discuss the requirements, design, construction, tests, and projected performance of the plant for the isotopic cryogenic distillation of argon. We also present the successful results of the isotopic cryogenic distillation of nitrogen with a prototype plant. The Aria project consists of a plant, hosting a 350 m cryogenic isotopic distillation column, the tallest ever built, which is currently in the installation phase in a mine shaft at Carbosulcis S.p.A., Nuraxi-Figus (SU), Italy. Abstract

Aria is a plant hosting a $${350}\,\hbox {m}$$ 350 m cryogenic isotopic distillation column, the tallest ever built, which is being installed in a mine shaft at Carbosulcis S.p.A., Nuraxi-Figus (SU), Italy. Aria is one of the pillars of the argon dark-matter search experimental program, lead by the Global Argon Dark Matter Collaboration. It was designed to reduce the isotopic abundance of $${^{39}\hbox {Ar}}$$ 39 Ar in argon extracted from underground sources, called Underground Argon (UAr), which is used for dark-matter searches. Indeed, $${^{39}\hbox {Ar}}$$ 39 Ar is a $$\beta $$ β -emitter of cosmogenic origin, whose activity poses background and pile-up concerns in the detectors. In this paper, we discuss the requirements, design, construction, tests, and projected performance of the plant for the isotopic cryogenic distillation of argon. We also present the successful results of the isotopic cryogenic distillation of nitrogen with a prototype plant.

Abstract

La résistance aux antipaludéens et la persistance de l’endémie palustre constituent un problème majeur de santé publique en Afrique. Outre la production des radicaux libres, l’infection palustre s’accompagne d’une dépression générale des antioxydants totaux plasmatiques pouvant contribuer à la morbidité et mortalité dues à la malaria. Ce travail a consisté à étudier l’Enantia olivacea, une espèce végétale consommée par les bonobos, Pan paniscus. Le screening chimique de l’extrait éthanolique (80 %) a été effectué. Nous avons réalisé des mesures semi-quantitatives de l’activité antioxydante par le radical 1,1-diphényl-2-picrylhydrazyle (DPPH) d’une solution méthanolique à 2 % (m/v) sur chromatographie en couche mince (CCM), et à 0,004 % une mesure quantitative sur microplaque. L’activité antiplasmodiale in vitro a été réalisée sur une souche clinique de Plasmodium falciparum. Les résultats obtenus ont montré la présence des flavonoïdes, des leucoanthocyanes, des tanins galliques, des alcaloïdes et des coumarines. L’analyse de l’activité antioxydante sur CCM a révélé les composés actifs aux Rf 1 et 0,67 et la concentration inhibitrice de 50 % (CI50) déterminée était de 21,3 ± 6,89 μg/ml face aux molécules de références utilisées dans notre test qui étaient l’acide ascorbique et la quercétine dont les CI50 étaient respectivement de 2,13 ± 0,21 et de 0,69 ± 0,02μg/ml. L’activité antiplasmodiale in vitro de l’extrait a donné une CI50 de 0,002 ± 0,0007 μg/ml relativement identique à la quinine. Il est donc possible que les bonobos se nourrissent de cette plante pour se faire guérir de la malaria. Ce travail montre une piste pour trouver de nouvelles molécules face à cette émergence de résistance du P. falciparum aux antipaludéens existants.

A case of aortic dissection extending to an aberrant right subclavian artery seen at CT and MRI is reported. The clinical and imaging patterns of this rare disease are described and compared with those of the literature. Multiplanar views are, in this particular case, very useful for a precise assessment of the involved aberrant artery.

Abstract

Background: Regular consumption of produce is a challenge for families with young children in low food access areas., Objective: A community partnership formed to evaluate feasibility of and interest in a child care center-based program for produce delivery from an online grocery vendor., Methods: Surveys were collected from caregivers across three child care centers, including produce program participants. Descriptive statistics summarize household characteristics and participants' experience with the program., Results: Challenges related to online payment and difficulty planning delivery times led to implementation of a modified intervention. Survey results revealed factors related to food access and storage that may impact interest and feasibility of online grocery in some communities., Conclusions: Online grocery vendors may increase accessibility to fresh produce for families, but barriers to their use still exist. Trusted community partners such as child care centers may offer some of the resources needed for success.

Abstract

Introduction: Rates of obesity amongst children and teens in the United States have tripled since the 1970s, and 18.5% are now considered obese. With smartphone use among children and teens becoming the norm, smartphone applications may be a cost-effective solution to support patients and families motivated to change lifestyle behaviors and reduce obesity. The purpose of this quality-improvement project was (1) to develop an app evaluation tool and star rating system based on scientific evidence and current clinical practice guidelines in managing childhood obesity, and (2) to determine whether an in-service training can increase provider knowledge and efficacy in the use of smartphone apps in managing childhood obesity., Methods: An app evaluation tool (Ped-WHAT) was developed that includes evidence-based behavior modification strategies (BMS) and the American Academy of Pediatrics (AAP) guidelines for healthy weight management in pediatrics. Apps were given a rating based on the number of these criteria included in the app. An educational in-service training was developed to assist health care providers in using the app evaluation tool with patients. This training was tested with providers working in a pediatric weight management clinic. Pre- and post-training surveys were administered. Results were analyzed using t tests to assess differences., Results: Sixteen commercially available apps were evaluated using the Ped-WHAT tool. Those that included the greatest number of BMS and AAP criteria received the highest rating on a 0-5 scale. Statistically significant improvements in provider knowledge and confidence were found after the in-service training., Discussion: In this current digital environment, commercially available weight management apps should undergo evaluation by health care providers to ensure that they follow the standard of care and current practice guidelines/recommendations., (Copyright © 2020. Abstract

Aristolochene synthase (ATAS) is a high-fidelity terpenoid cyclase that converts farnesyl diphosphate exclusively into the bicyclic hydrocarbon aristolochene. Previously determined crystal structures of ATAS complexes revealed trapped active site water molecules that could potentially interact with catalytic intermediates: water "w" hydrogen bonds with S303 and N299, water molecules "w1" and "w2" hydrogen bond with Q151, and a fourth water molecule coordinates to the Mg(2+)C ion. There is no obvious role for water in the ATAS mechanism because the enzyme exclusively generates a hydrocarbon product. Thus, these water molecules are tightly controlled so that they cannot react with carbocation intermediates. Steady-state kinetics and product distribution analyses of eight ATAS mutants designed to perturb interactions with active site water molecules (S303A, S303H, S303D, N299A, N299L, N299A/S303A, Q151H, and Q151E) indicate relatively modest effects on catalysis but significant effects on sesquiterpene product distributions. X-ray crystal structures of S303A, N299A, N299A/S303A, and Q151H mutants reveal minimal perturbation of active site solvent structure. Seven of the eight mutants generate farnesol and nerolidol, possibly resulting from addition of the Mg(2+)C-bound water molecule to the initially formed farnesyl cation, but no products are generated that would suggest enhanced reactivity of other active site water molecules. However, intermediate germacrene A tends to accumulate in these mutants. Thus, apart from the possible reactivity of Mg(2+)C-bound water, active site water molecules in ATAS are not directly involved in the chemistry of catalysis but instead contribute to the template that governs the conformation of the flexible substrate and carbocation intermediates.

Abstract

Aristolochene synthase, a metal-dependent sesquiterpene cyclase from Aspergillus terreus, catalyzes the ionization-dependent cyclization of farnesyl diphosphate (FPP) to form the bicyclic eremophilane (+)-aristolochene with perfect structural and stereochemical precision. Here, we report the X-ray crystal structure of aristolochene synthase complexed with three Mg(2+) ions and the unreactive substrate analogue farnesyl-S-thiolodiphosphate (FSPP), showing that the substrate diphosphate group is anchored by metal coordination and hydrogen bond interactions identical to those previously observed in the complex with three Mg(2+) ions and inorganic pyrophosphate (PPi). Moreover, the binding conformation of FSPP directly mimics that expected for productively bound FPP, with the exception of the precise alignment of the C-S bond with regard to the C10-C11 π system that would be required for C1-C10 bond formation in the first step of catalysis. We also report crystal structures of aristolochene synthase complexed with Mg(2+)3-PPi and ammonium or iminium analogues of bicyclic carbocation intermediates proposed for the natural cyclization cascade. Various binding orientations are observed for these bicyclic analogues, and these orientations appear to be driven by favorable electrostatic interactions between the positively charged ammonium group of the analogue and the negatively charged PPi anion. Surprisingly, the active site is sufficiently flexible to accommodate analogues with partially or completely incorrect stereochemistry. Although this permissiveness in binding is unanticipated, based on the stereochemical precision of catalysis that leads exclusively to the (+)-aristolochene stereoisomer, it suggests the ability of the active site to enable controlled reorientation of intermediates during the cyclization cascade. Taken together, these structures illuminate important aspects of the catalytic mechanism.

Aim: The triage forms filled in 2006 by nursing personnel dedicated to the triage in an Emergency Department were analysed., Methods: The 37.323 triage forms were scrutinized to identify any discrepancies in severity codes assigned by the nurse and by the medical doctor, after the clinical and instrumental assessment., Results: Four-hundred-forty discrepancies were identified (13% of those initially classified as discrepancies): 221 (50.2%) severity codes were overestimated and 219 underestimated. The percentage of discrepancies increased during night hours and at the morning shift change. Nurses who performed the triage less often, made an higher number of over or underestimates. Overall, the number of discrepancies was 0.5 per shift., Conclusions: The overall performance can be considered very good. A model for analysis of discrepancies can be proposed. A number of proposals have been advanced to further improve the nurses performance.

Two known lignans, liriodendrin and syringaresinol mono-beta-D-glucoside, have been isolated from the methanol extract of the roots of Boerhaavia diffusa L. (Nyctaginaceae), and the former compound was found to exhibit a significant calcium (Ca2+) channel antagonistic effect in frog heart single cells using the whole-cell voltage clamp method. Reexamination of the carbon-13 nuclear magnetic resonance (13C-NMR) spectra of these compounds was also carried out by the use of two-dimensional NMR techniques including a 1H-detected heteronuclear multiple bond connectivity (HMBC) experiment, and it was found that the previous signal assignments for C-1' and C-4' have to be revised.
