Direct encounters, in which two or more individuals are physically close to one another, are a topic of increasing interest as more and better movement data become available. Recent progress, including the development of statistical tools for estimating robust measures of changes in animals’ space use over time, facilitates opportunities to link direct encounters between individuals with the long-term consequences of those encounters. Working with movement data for coyotes (Canis latrans) and grizzly bears (Ursus arctos horribilis), we investigate whether close intraspecific encounters were associated with spatial shifts in the animals’ range distributions, as might be expected if one or both of the individuals involved in an encounter were seeking to reduce or avoid conflict over space. We analyze the movement data of a pair of coyotes in detail, identifying how a shift in home range location resulting from altered movement behavior was apparently a consequence of a close intraspecific encounter. With grizzly bear movement data, we approach the problem from the perspective of a set of encounter pairs within a population. We find support for the hypotheses that 1) close intraspecific encounters between bears are, on average, associated with subsequent shifts in range distributions and 2) encounters defined at finer spatial scales are followed by greater changes in space use. Our results suggest that animals can undertake long-term, large-scale spatial shifts in response to close intraspecific encounters that have the potential for conflict. These results lend support for existing theory on the evolution of territories and space use (e.g., Maynard-Smith’s bourgeois strategy regarding low-conflict coexistence). Protein-truncating variants protective against human disease provide in vivo validation of therapeutic targets. Here we used targeted sequencing to conduct a search for protein-truncating variants conferring protection against inflammatory bowel disease exploiting knowledge of common variants associated with the same disease. Through replication genotyping and imputation we found that a predicted protein-truncating variant (rs36095412, p.R179X, genotyped in 11,148 ulcerative colitis patients and 295,446 controls, MAF=up to 0.78%) in RNF186, a single-exon ring finger E3 ligase with strong colonic expression, protects against ulcerative colitis (overall P=6.89 × 10(-7), odds ratio=0.30). We further demonstrate that the truncated protein exhibits reduced expression and altered subcellular localization, suggesting the protective mechanism may reside in the loss of an interaction or function via mislocalization and/or loss of an essential transmembrane domain.

- Abstract
This paper explores the possibilities of using a pedagogical model for working with adolescent girls in physical education as a means of balancing the challenge of external prescription from outside the school with teacher and pupil agency. We report data from a study involving four schools in Glasgow. We note that the national curriculum for Scotland, Curriculum for Excellence, is a broad and bold type that provides teachers with 'spaces for manoeuvre' in order to shape local curricula that best meet the needs and interests of girls. This is particularly the case in physical education, which in the Basic General Education phase for 12-15 years olds there is no well-established assessment regime. We identify four spaces for manoeuvre for teachers and pupils within an activist model: new forms of communication based on authorising pupil voice; offering choices and opening up learning possibilities; the co-construction of a safe class environment; opportunities to rethink traditional structures based on the multi-activity curriculum form. We conclude that an activist pedagogical model provided teachers and pupils with spaces to explore alternative practices to traditional forms of physical education.

- Abstract
COVID-19 lockdowns in early 2020 reduced human mobility, providing an opportunity to disentangle its effects on animals from those of landscape modifications. Using GPS data, we compared movements and road avoidance of 2300 terrestrial mammals (43 species) during the lockdowns to the same period in 2019. Individual responses were variable with no change in average movements or road avoidance behavior, likely due to variable lockdown conditions. However, under strict lockdowns 10-day 95th percentile displacements increased by 73%, suggesting increased landscape permeability. Animals’ 1-hour 95th percentile displacements declined by 12% and animals were 36% closer to roads in areas of high human footprint, indicating reduced avoidance during lockdowns. Overall, lockdowns rapidly altered some spatial behaviors, highlighting variable but substantial impacts of human mobility on wildlife worldwide.

- Abstract
An assay was developed to study plant receptor kinase activation and signaling mechanisms. The extracellular leucine-rich repeat (LRR) and transmembrane domains of the Arabidopsis receptor kinase BRI1, which is implicated in brassinosteroid signaling, were fused to the serine/threonine kinase domain of XA21, the rice disease resistance receptor. The chimeric receptor initiates plant defense responses in rice cells upon treatment with brassinosteroids. These results, which indicate that the extracellular domain of BRI1 perceives brassinosteroids, suggest a general signaling mechanism for the LRR receptor kinases of plants. This system should allow the discovery of ligands for the LRR kinases, the largest group of plant receptor kinases.

- Abstract
Background: Anti-tumour necrosis factor (TNF) drugs such as infliximab are associated with attenuated antibody responses after COVID-19 vaccination. It is unknown how infliximab impacts vaccine-induced serological responses against highly transmissible Omicron variants, which possess the ability to evade host immunity and are now the dominating variants causing current waves of infection. Methods: CLARITY IBD is a prospective, multicentre, observational cohort study investigating the impact of infliximab and vedolizumab on SARS-CoV-2 infection and vaccination in patients with inflammatory bowel disease (IBD). In the current work, the primary outcome was neutralising antibody responses against SARS-CoV-2 wild-type and Omicron BA.1 and BA.4/5 variants after three doses of COVID-19 vaccination in 1288 patients with IBD without prior COVID-19 infection, who were established on either infliximab (n=871) or vedolizumab (n=417). Cox proportional hazards models were constructed to investigate the risk of breakthrough infection in relation to neutralising antibody titres. Findings: Following three doses of COVID-19 vaccine, neutralising titre NT50 (half-inhibitory neutralising titre) was significantly diminished in patients treated with infliximab as compared to patients treated with vedolizumab, against wild-type (geometric mean [95% CI], 1990 [1781, 2223] vs 3212 [2780, 3712], p

Aim: Crohn's anal fistula (CAF) is a complex condition, with no agreement on which patient characteristics should be routinely reported in studies. The aim of this study was to develop a core descriptor set of key patient characteristics for reporting in all CAF research.Method: Candidate descriptors were generated from published literature and stakeholder suggestions. Colorectal surgeons, gastroenterologists and specialist nurses in inflammatory bowel disease took part in three rounds of an international modified Delphi process using nine-point Likert scales to rank the importance of descriptors. Feedback was provided between rounds to allow refinement of the next ratings. Patterns in descriptor voting were assessed using principal component analysis (PCA). Resulting PCA groups were used to organize items in rounds two and three. Consensus descriptors were submitted to a patient panel for feedback. Items meeting predetermined thresholds were included in the final set and ratified at the consensus meeting.Results: One hundred and thirty three respondents from 22 countries completed round one, of whom 67.0% completed round three. Ninety seven descriptors were rated across three rounds in 11 PCA-based groups. Forty descriptors were shortlisted. The consensus meeting ratified a core descriptor set of 37 descriptors within six domains: fistula anatomy, current disease activity and phenotype, risk factors, medical interventions for CAF, surgical interventions for CAF, and patient symptoms and impact on quality of life.Conclusion: The core descriptor set proposed for all future CAF research reflects characteristics important to gastroenterologists and surgeons. This might aid transparent reporting in future studies.

- Abstract
For the past several years, the JET scientific programme (Pamela et al 2007 Fusion Eng. Des. 82 590) has been engaged in a multi-campaign effort, including experiments in D, H and T, leading up to 2020 and the first experiments with 50%/50% D-T mixtures since 1997 and the first ever D-T plasmas with the ITER mix of plasma-facing component materials. For this purpose, a concerted physics and technology programme was launched with a view to prepare the D-T campaign (DTE2). This paper addresses the key elements developed by the JET programme directly contributing to the D-T preparation. This intense preparation includes the review of the physics basis for the D-T operational scenarios, including the fusion power predictions through first principle and integrated modelling, and the impact of isotopes in the operation and physics of D-T plasmas (thermal and particle transport, high confinement mode (H-mode) access, Be and W erosion, fuel recovery, etc). This effort also requires improving several aspects of plasma operation for DTE2, such as real time control schemes, heat load control, disruption avoidance and a mitigation system (including the installation of a new shattered pellet injector), novel ion cyclotron resonance heating schemes (such as the three-ions scheme), new diagnostics (neutron camera and spectrometer, active Alfven eigenmode antennas, neutral gauges, radiation hard imaging systems...) and the calibration of the JET neutron diagnostics at 14 MeV for accurate fusion power measurement. The active preparation of JET for the 2020 D-T campaign provides an incomparable source of information and a basis for the future D-T operation of ITER, and it is also foreseen that a large number of key physics issues will be addressed in support of burning plasmas.

This dissertation addresses social differentiation among rural residents of Chunhuayum, an ancient Maya village in northwest Yucatan, from the Late Preclassic to the Late Early Classic (300 B.C. ��� A.D. 600/630). The three axes of social differentiation investigated are household wealth, occupation, and social connectivity to external networks. Using a practice theory approach, my research seeks to identify how material and social practices shaped and expressed social differentiation among Chunhuayum households, as well as how these may have shaped the particular history of Chunhuayum within its regional context. Throughout Chunhuayum���s occupation, residential architecture was the most salient marker of wealth disparities, although nuanced variation found among households��� possessions, including decorated and serving ware, obsidian, and shell, suggest households also differed in occupation, social connectivity, and local authority. I argue wealth inequalities emerged during the Preclassic, likely based on household size as larger labor pools could produce and transmit greater social and material resources to later generations. Chunhuayum���s Early Classic residents continued shaping locally meaningful differences through both habituated uses of everyday objects and innovative action. The greater resources accumulated by two households, N148 and N141, enabled them to engage in locally innovative strategies of socio-material wellbeing during the late Early Classic���shell crafting and group oriented ritual orchestration���that ultimately had different outcomes. N148 hosted village-wide, group oriented, rituals within their household compound starting in the Late Preclassic through the Early Classic. These rituals not only benefited the N148 household in establishing greater authority and wealth within the village, but also likely supported the community���s longevity by fostering inter-household obligations and a shared sense of place that would continue into the Postclassic. Crafters at N141 made disks out of conch shell using some of the large amounts of obsidian they procured, which they likely traded outside of Chunhuayum. N141���s more ���open��� livelihood strategy enabled this household to increase their social connectivity and portable wealth during the Early Classic, yet did not enable the household���s to maintain their wealth or social connectivity into the Late Classic. This dissertation adds to the growing literature underlining the rich and multifaceted lives of ancient rural populations and supports a number of points concerning ancient Maya rurality. Notwithstanding their shared commonalities, people were active and at times innovative in shaping social differences locally and beyond. While Chunhuayum lacks the archaeological proxies often associated with elite social identity, rural residents shaped locally intelligible differences through the materials, skills, ritual spaces, and social relations available to them. Moreover, following the disintegration of the Uc. polity during the Early Classic, residents actively engaged in increasingly complex relations within and beyond their village and strengthened local forms of authority, suggesting that local processes are inherent to rural complexity, which cannot be simply understood as a trickling-down of normative hierarchical structures. Finally, Chunhuayum presents an example of how most rural communities were neither wholly dependent nor fully isolated from larger centers but instead practiced a combination of open and closed-community strategies. The varied practices and relationships of rural residents, including strategies in facing new circumstances, led to diverging outcomes for both households and the community at large.
