1. Fungal Consortia Mediated Bio-Treatment of Organic Matter and Metals Uptake from Sewage Water: Maize Agro-Physiological Assessment

Abstract

The present investigation aims to improve the efficiency of fungal mono- and mixed cultures in removing organic pollutants and metals from sewage water (SW) for further maize plant response assessments. The reduction in the organic load from the SW was harnessed using a co-culture consortium consisting of Aspergillus niger (KB5), Sordariomycetes sp. (D10), and Coniochaetaceae sp. (LB3). The testing results had evinced removal of up to 88% of the organic matter and more than 96%, 91%, 80%, and 47.6%, of removal percentages for Copper (Cu), Nickel (Ni), Cadmium (Cd), and Lead (Pb), respectively, with the developed fungal consortium [KB5 + D10 + LB3]. After treatment and lab experiments, a reuse of treated and untreated SW for plant irrigation was evaluated towards improving maize plant growth. Irrigation was conducted in pot experiments with three types of water: clean water (Control), untreated (USW), and treated SW by fungal consortia (TSW) and by station treatment plant STP (TSWP) using the randomized complete block (RCB) experimental design. Results of the pots trial revealed that the morphological parameters of SW-irrigated plants are slightly improved compared to water-irrigated plants. Data regarding assimilating area attributes indicated that the most significant enlargement of the assimilation area was observed with TSW-D (1/4) irrigation by 1051 cm2, followed by TSWP-D (0) by 953.96 cm2, then USW-D (1/4) by 716.54 cm2, as compared to plants irrigated with clean water (506.91 cm2). On average, the assimilation areas were larger by 51.76%, 46.86%, and 29.25% in TSW, USW, and TSWP-irrigated plants, respectively. Thus, SW irrigation supports the required qualities and quantities of microelements and water for plant growth. Oxidative stress assessment showed that irrigations with treated SW caused a significant decrease in both enzymatic and non-enzymatic antioxidants, depicting that the treatment lowered the stress of sewage water.

Abstract

In an era of connectivity and automation, the vehicle industry is adopting numerous technologies to transform driver-centric vehicles into intelligent mechanical devices driven by software components. Software integration and network connectivity inherit numerous security issues that open the door for malicious attacks. Software security testing is a scalable and practical approach to identify systems’ weaknesses and vulnerabilities at an early stage and throughout their life-cycle. Security specialists recommend fuzz testing to identify vulnerabilities within vehicle software systems. Nevertheless, the randomness and blindness of fuzzing hinder it from becoming a reliable security tool. This article presents a vulnerability-oriented fuzz (VulFuzz) testing framework that utilizes security vulnerability metrics designed particularly for connected and autonomous vehicles to direct and prioritize the fuzz testing toward the most vulnerable components. While most gray-box fuzzing techniques aim solely to expand code coverage, the proposed approach assigns weights to ensure a thorough examination of the most vulnerable components. Moreover, we employ an input structure-aware mutation technique that can bypass vehicle software systems’ input formats to boost test performance and avoid dropped test cases. Such a testing technique will contribute to the quality assurance of vehicle software engineering. We implemented the proposed approach on OpenPilot, a driver assistance system, and compared our results to American fuzzy lop (AFL) and an unguided mutation-based fuzzer. Within 16.8 h, VulFuzz exposed 335 crashes, 41 times more than AFL and two times more than an unguided mutation-based fuzzer. VulFuzz is explicitly efficient for automotive systems, reaching the same code coverage as AFL but with more exposed crashes and fewer dropped messages.

15. TFEB signaling attenuates NLRP3-driven inflammatory responses in severe asthma

Abstract

Background: NLRP3-driven inflammatory responses by circulating and lung-resident monocytes are critical drivers of asthma pathogenesis. Autophagy restrains NLRP3-induced monocyte activation in asthma models. Yet, the effects of autophagy and its master regulator, transcription factor EB (TFEB), on monocyte responses in human asthma remain unexplored. Here, we investigated whether activation of autophagy and TFEB signaling suppress inflammatory monocyte responses in asthmatic individuals. Methods: Peripheral blood CD14+ monocytes from asthmatic patients (n = 83) and healthy controls (n = 46) were stimulated with LPS/ATP to induce NLRP3 activation with or without the autophagy inducer, rapamycin. ASC specks, caspase-1 activation, IL-1β and IL-18 levels, mitochondrial function, ROS release, and mTORC1 signaling were examined. Autophagy was evaluated by LC3 puncta formation, p62/SQSTM1 degradation and TFEB activation. In a severe asthma (SA) model, we investigated the role of NLRP3 signaling using Nlrp3−/− mice and/or MCC950 administration, and the effects of TFEB activation using myeloid-specific TFEB-overexpressing mice or administration of the TFEB activator, trehalose. Results: We observed increased NLRP3 inflammasome activation, concomitant with impaired autophagy in circulating monocytes that correlated with asthma severity. SA patients also exhibited mitochondrial dysfunction and ROS accumulation. Autophagy failed to inhibit NLRP3-driven monocyte responses, due to defective TFEB activation and excessive mTORC1 signaling. NLRP3 blockade restrained inflammatory cytokine release and linked airway disease. TFEB activation restored impaired autophagy, attenuated NLRP3-driven pulmonary inflammation, and ameliorated SA phenotype. Conclusions: Our studies uncover a crucial role for TFEB-mediated reprogramming of monocyte inflammatory responses, raising the prospect that this pathway can be therapeutically harnessed for the management of SA. © 2022 European Academy of Allergy and Clinical Immunology and John Wiley & Sons Ltd.

Abstract

Background: Real-world data on non-vitamin K oral anticoagulants (NOACs) are essential in determining whether evidence from randomised controlled clinical trials translate into meaningful clinical benefits for patients in everyday practice. RIVER (RIVaroxaban Evaluation in Real life setting) is an ongoing international, prospective registry of patients with newly diagnosed non-valvular atrial fibrillation (NVAF) and at least one investigator-determined risk factor for stroke who received rivaroxaban as an initial treatment for the prevention of thromboembolic stroke. The aim of this paper is to describe the design of the RIVER registry and baseline characteristics of patients with newly diagnosed NVAF who received rivaroxaban as an initial treatment. Methods and results: Between January 2014 and June 2017, RIVER investigators recruited 5072 patients at 309 centres in 17 countries. The aim was to enroll consecutive patients at sites where rivaroxaban was already routinely prescribed for stroke prevention. Each patient is being followed up prospectively for a minimum of 2-years. The registry will capture data on the rate and nature of all thromboembolic events (stroke / systemic embolism), bleeding complications, all-cause mortality and other major cardiovascular events as they occur. Data quality is assured through a combination of remote electronic monitoring and onsite monitoring (including source data verification in 10% of cases). Patients were mostly enrolled by cardiologists (n = 3776, 74.6%), by internal medicine specialists 14.2% (n = 718) and by primary care/general practice physicians 8.2% (n = 417). The mean (SD) age of the population was 69.5 (11.0) years, 44.3% were women. Mean (SD) CHADS2 score was 1.9 (1.2) and CHA2DS2-VASc scores was 3.2 (1.6). Almost all patients (98.5%) were prescribed with once daily dose of rivaroxaban, most commonly 20 mg (76.5%) and 15 mg (20.0%) as their initial treatment; 17.9% of patients received concomitant antiplatelet therapy. Most patients enrolled in RIVER met the recommended threshold for AC therapy (86.6% for 2012 ESC Guidelines, and 79.8% of patients according to 2016 ESC Guidelines). Conclusions: The RIVER prospective registry will expand our knowledge of how rivaroxaban is prescribed in everyday practice and whether evidence from clinical trials can be translated to the broader cross-section of patients in the real world. Rapid eye movement (REM) sleep behaviour disorder (RBD) is a rare parasomnia that may predict the later occurrence of alpha-synucleinopathies. Variants in the gene encoding for the lysosomal enzyme glucocerebrosidase, GBA, strongly increase the risk of RBD. In a GBA1-mouse model recently shown to mimic prodromal stages of α-synucleinopathy, we now demonstrate striking REM and NREM sleep abnormalities accompanied by distinct structural changes in the more widespread sleep neurocircuitry.

Abstract

Nε-lysine acetylation is recognized as a prevalent post-translational modification (PTM) that regulates proteins across all three domains of life. In Bacillus subtilis, the histone-like protein HBsu is acetylated at seven sites, which regulates DNA compaction and the process of sporulation. In Mycobacteria, DNA compaction is a survival strategy in response antibiotic exposure. Acetylation of the HBsu ortholog HupB decondenses the chromosome to escape this drug-induced, non-growing state, and in addition, regulates the formation of drug-tolerant subpopulations by altering gene expression. We hypothesized that the acetylation of HBsu plays similar regulatory roles. First, we measured nucleoid area by fluorescence microscopy and in agreement, we found that wild-type cells compacted their nucleoids upon kanamycin exposure, but not exposure to tetracycline. We analyzed a collection of HBsu mutants that contain lysine substitutions that mimic the acetylated (glutamine) or unacetylated (arginine) forms of the protein. Our findings indicate that some level of acetylation is required at K3 for a proper response and K75 must be deacetylated. Next, we performed time-kill assays of wild-type and mutant strains in the presence of different antibiotics and found that interfering with HBsu acetylation led to faster killing rates. Finally, we examined the persistent subpopulation and found that altering the acetylation status of HBsu led to an increase in persister cell formation. In addition, we found that most of the deacetylation-mimic mutants, which have compacted nucleoids, were delayed in resuming growth following removal of the antibiotic, suggesting that acetylation is required to escape the persistent state. Together, this data adds an additional regulatory role for HBsu acetylation and further supports the existence of a histone-like code in bacteria.

Abstract

Variants in ATP-binding cassette transporter type A4 (ABCA4) have been linked to several forms of inherited retinal diseases (IRDs) besides the classically defined Stargardt disease (STGD), known collectively as ABCA4 retinopathies. ABCA4 is a sizable locus harboring 50 exons; thus, its analysis has revealed over 2,400 variants described, of which more than 2,000 are causal. Due to the clinical and genetic heterogeneity, diagnosing ABCA4 retinopathies is challenging. To date, no ABCA4-related retinopathy has been detected in Lebanon. Using next-generation sequencing, we analyzed our IRDs' cohort retrospectively (61 families) and identified five with ABCA4-related retinopathies, making it a relatively abundant cause of IRDs (about 8 %). Three families were diagnosed with rod-cone dystrophy (RCD), two with STGD, and one with cone-rod dystrophy (CRD). In conclusion, our study showed the presence of ABCA4 variants with a high degree of heterogeneity in Lebanon.

Software integration in modern vehicles is continuously expanding. This is due to the fact that vehicle manufacturers are always trying to enhance and add more innovative and competitive features that may rely on complex software functionalities. However, these features come at a cost. They amplify the security vulnerabilities in vehicles and lead to more security issues in today's automobiles. As a result, the need for identifying vulnerable components in a vehicle software system has become crucial. Security experts need to know which components of the vehicle software system can be exploited for attacks and should focus their testing and inspection efforts on it. Nevertheless, it is a challenging and costly task to identify these weak components in a vehicle's system. In this paper, we propose some security vulnerability metrics for connected vehicles that aim to assist software testers during the development life-cycle in order to identify the frail links that put the vehicle at highsecurity risks. Vulnerable function assessment can give software testers a good idea about which components in a connected vehicle need to be prioritized in order to mitigate the risk and hence secure the vehicle. The proposed metrics were applied to OpenPilot - a software that provides Autopilot feature - and has been integrated with 48 different vehicles.. The application shows how the defined metrics can be effectively used to quantitatively measure the vulnerabilities of a vehicle software system.

Abstract

Background: Numerous surveys studied individuals’ decision to receive COVID-19 vaccine but the motives behind accepting or refusing COVID-19 vaccines are not yet fully understood. We aimed to more qualitatively explore the views and perceptions toward COVID-19 vaccines in Saudi Arabia to provide recommendations to mitigate the vaccine hesitancy issue. Methods: Open-ended interviews were conducted between October 2021-January 2022. The interview guide included questions about beliefs in vaccine efficacy and safety, and previous vaccination history. The interviews were audio-recorded, transcribed verbatim, and the content was analyzed using thematic analysis. Nineteen participants were interviewed. Results: All of the interviewees were vaccine acceptors; however, three participants were hesitant as they felt they were forced to receive it. Several themes emerged as the reasons to accept or refuse the vaccine. The key reasons behind vaccine acceptance were the sense of obligation to fulfill a governmental command, trust in the government decisions, vaccine availability, and the impact of family/friends. The main reason behind vaccine hesitancy was doubts regarding vaccine efficacy and safety and that vaccines were pre-invented, and the pandemic is made-up. Participants’ sources of information included social media, official authorities, and family/friends. Conclusion: Findings from this study show that the convenience of receiving the vaccine, the abundance of credible information from the Saudi authorities, and the positive influence of family/friends were among the major factors that encouraged the public in Saudi Arabia to get vaccinated against COVID-19. Such results may inform future policies regarding encouraging the public to receive vaccines in cases of pandemic.

Abstract Inherited retinal dystrophies (IRDs) are a major cause of vision loss. Altogether are highly heterogeneous genotypically and phenotypically, exhibiting substantial differences worldwide. To shed more light on these conditions, we investigated the genetic and phenotypic landscape of IRDs in the Arabs globally and per country. We analyzed 1,621 affected individuals from 16 Arabic countries reported in 198 articles. At the phenotypic level, rod-cone dystrophy (RCD) and Usher syndrome were the most prevalent conditions among non-syndromic and syndromic IRDs. At the gene level, TULP1, ABCA4, RP1, CRB1, MYO7A, RPE65, KCNV2, and IMPG2 were the most mutated genes. Interestingly, all except CRB1 were highly prevalent because they harbored founder mutations, implying that consanguinity is a major determinant in Arab countries. Of note, ~ 93% of the investigated individuals carried homozygous mutations. The country analysis for the IRDs conditions and their associated genotypes revealed that whereas Leber Congenital Amaurosis, RCD, and USHER syndrome were widely distributed, bestrophinopathies and non-syndromic hearing loss were restricted to specific countries (till now). This study could be a starting point for initiating suitable health policies towards IRDs in the Arab world. The high degree of homozygosity urges the need for genetic counsellors to provide personalized information and support the affected individuals.

We describe three distinct approaches to visualization for multiscale materials modelling research. These have been developed with the framework of the SimPhoNy FP7 EU-project, and complement each other in their requirements and possibilities. All have been integrated via wrappers to one or more of the simulation approaches within the SimPhoNy project. In this manuscript we describe and contrast their features. Together they cover visualization needs from electronic to macroscopic scales and are suited to simulations made on personal computers, workstations or advanced High Performance parallel computers. Examples as well as recommendations for future calculations are presented.

جُمع ثلاث أنواع من منطقة اللاذقية تابعة لجنس اللوف (Arum L. )، وهي: A. palaestinum، A. dioscoridis، A. italicum. وشملت هذه الدراسة الشكل الحياتي للنبات، الدرنات، الأوراق، النورة، الكفرى، الأزهار ( المذكرة والمؤنثة و العقيمة)، الثمرة، البذرة. كما حددت أنماط حبيبات النشاء للدرنات باستخدام معايير شكلية مميزة لها ( الشكل الخارجي – توضع السرة – وجود الصفائح – وجود التجويف المركزي – الشقوق – الأبعاد) وتباينت بين الشكل الكروي – نصف الكروي – المستطيل – مثلثي- معيني – شبه المنحرف – متعدد الأضلاع. كانت السرة مركزية في حبيبات لنشاء للأنواع المدروسة، ووضحت الشقوق في الشكل الكروي فقط عند كل من Arum palaestinum, A.dioscoridis على شكل حرف Y، أما التجويف المركزي وجد في الشكل الكروي عند Arum palaestinum . توزعت أحجام حبيبات النشاء في نمطين هما : النمط A وسجل أكبر حجم لحبيبة النشاء بلغ 12*14 µm للشكل شبه المنحرف عند Arum palaestinum ، والنمط B فكانت أصغر حبيبة للنشاء وبلغت 6* 7 µm ميزت الشكل المثلثي عند Arum palaestinum .

Abstract Non-syndromic rod-cone dystrophy (RCD) is the most common condition in inherited retinal diseases. The aim of this study was to evaluate the research output and productivity related to RCD genetics per countries as classified by the human development index (HDI), by analyzing publication frequency and citations, the choice of journals and publishers, since 2000 to date. We have also analyzed the use of next-generation sequencing (NGS) in publications originating from countries with different HDIs. One thousand four hundred articles focusing on non-syndromic RCD were downloaded and analyzed. Citations and published articles were adjusted per one million individuals. The research output is significantly higher in very high HDI countries (86% of the total publications and 95% of the citations) than countries with lower HDIs in all aspects. High and medium HDI countries published together 13.6% of the total articles worldwide and received 4.6% of the citations. On the publication level, the USA (26%), United Kingdom (10%), and Japan (7%) were the top 3 among very high HDI countries, while China (6%) and India (2%) ranked first in high and medium HDI countries respectively. Fire is one the main risks in the construction sector. Consequences as material losses, stoppage and possible personal damages cause the fire risk assessment and knowledge of building behaviour in fire situation are essential to prevent and minimize the consequences. Currently there are sets of conventional scalar methods that quantify the risk level and provide different protection standards. At the same time, procedures for Modelling and Computer Simulation of Fire based on vector methods are under development, which provide multidimensional information about the fire. The aim of this work is to establish an integration model of design and assessment with scalar and vector technics for increase the knowledge about the fire risk (both development and evolution processes), supply full information on the development of fire in all lifecycle stage’s installation. This model will support the task of Project Manager and Experts in prevention of occupational Risks. Los incendios son uno de los principales riesgos en el ámbito constructivo. Consecuencias como pérdidas materiales, interrupción de actividad o daños personales hacen que la evaluación del riesgo de incendio y el conocimiento del comportamiento de la construcción ante el fuego sean imprescindibles a la hora de disminuir la probabilidad de ocurrencia, prevenir efectos y minimizar consecuencias. Actualmente existen un conjunto de métodos convencionales (escalares) que cuantifican el nivel de riesgo y aportan diferentes medidas de protección. Además se encuentran en desarrollo métodos (vectoriales) de Modelado y Simulación Computacional que aportan información multidimensional. El presente trabajo establece un modelo para la integración de métodos escalares y vectoriales para conocer el desarrollo y evolución del incendio junto al nivel de riesgo no solo en la etapa de diseño de la instalación, sino en su ciclo de vida facilitando las tareas de ingenieros de proyectos y técnicos de prevención de riesgos laborales.

إن مشكلة التلوث بأشكاله المختلفة عالمية, يعاني منها الجميع، ويعد التلوث بالنفايات الطبية أحد أهم هذه الأشكال, والنفايات الطبية تنتج عن مؤسسات الرعاية الصحية, وقد تم تقسيمها عالمياً لعدة أنواع. بحثنا هذا دراسة إحصائية لكيفية إدارة النفايات الطبية التي تأخذ شكلا سائلا عند التخلص منها في بعض المراكز الصحية العامة في محافظة اللاذقية. تبين أن هناك قلة معرفة بكيفية التمييز بين النفايات الطبية وخاصة السائلة منها, مما أدى لأخطاء في تصنيفها, وبالتالي أدى لوصولها للمصادر الطبيعية نتيجة هذه الأخطاء, لذلك لابد من تأهيل وتدريب العاملين في القطاع الصحي لاستخدام الطرق المتبعة عالمياً للتعامل معها, والتوعية أكثر بها وبمخاطرها.

In theaters and the filmmaking industry, video streams, images, audio streams and scalar data are commonly used. In these fields, one of the most important magnitudes to be collected and controlled is the light intensity in different scene spots. So, it is extremely important to be able to deploy a network of light sensors which are usually integrated in a more general Wireless Multimedia Sensor Network (WMSN). If many light measurements have to be acquired, the simpler and cheaper the sensor, the more affordable the WMSN will be. In this paper we propose the use of a set of very cheap light sensors (photodiodes) and to spectrally and directionally correct their measurements using mathematical methods. Abstract

Dopamine (DA) administration in sepsis is used to modulate the hypotensive condition and to normalize the blood vessels perfusion. However, whether this administration of DA has an effect on the release of cytokines in vivo deserves investigation.Pre-exposure of DA (1 µg/ml) to whole blood enhanced IL-10 (30%) production level following LPS stimulation. This IL-10 enhancement became statistically significant (p0.001) upon the addition of D2-DA receptor (DAR) antagonists, Clozapine or Haloperidol. Furthermore, systemic administration of DA (0.5-50 mg/kg) in mice suppressed significantly LPS-induced TNF-α levels in blood, liver, spleen, brain, and lungs; IL-10 levels in blood, brain and liver; and IFN-γ levels in blood, liver, brain, and lungs. On the other hand, DA enhanced significantly LPS-induced IL-10 production in the lungs and spleen, and IFN-γ levels in the spleen. Administration of Clozapine (54 mg/kg) or Haloperidol (62 mg/kg) with LPS (1 µg) and DA (5 mg/kg) reversed DA suppressive effects on LPS-induced cytokines in blood, IFN-γ in brain and lungs, and enhanced significantly LPS-induced IL-10 production in blood, spleen, liver, and lungs.These results indicate that DA modulatory effect on LPS-induced blood cytokines-producing cells is mediated mainly by D2-DAR (D2/ D3/D4) through enhancing immune cells migration and extravasation into tissues. Furthermore, DA selectivity on cytokines modulation is tissue specific, mediated by the type of DAR expressed and on the immune cells lodged in each tissue.

50. Novel Missense and Splice Site Mutations in USH2A, CDH23, PCDH15, and ADGRV1 Are Associated With Usher Syndrome in Lebanon

Abstract

The purpose of this study was to expand the mutation spectrum by searching the causative mutations in nine Lebanese families with Usher syndrome (USH) using whole-exome sequencing. The pathogenicity of candidate mutations was first evaluated according to their frequency, conservation, and in silico prediction tools. Then, it was confirmed via Sanger sequencing, followed by segregation analysis. Finally, a meta-analysis was conducted to calculate the prevalence of USH genes in the Lebanese population. Three missense mutations, two splice site mutations, and one insertion/deletion were detected in eight of the families. Four of these variants were novel: c.5535C > A; p.(Asn1845Lys) in exon 41 of CDH23, c.7130G > A; p.(Arg2377Gln) in exon 32 of ADGRV1, c.11390-1G > A in USH2A, and c.3999–6A > G in PCDH15. All the identified mutations were shown to be likely disease-causing through our bioinformatics analysis and co-segregated with the USH phenotype. The mutations were classified according to the ACMG standards. Finally, our meta-analysis showed that the mutations in ADGRV1, USH2A, and CLRN1 are the most prevalent and responsible for approximately 75% of USH cases in Lebanon. Of note, the frequency USH type 3 showed a relatively high incidence (23%) compared to the worldwide prevalence, which is around 2–4%. In conclusion, our study has broadened the mutational spectrum of USH and showed a high heterogeneity of this disease in the Lebanese population.
