1. Early time dynamics of laser-ablated silicon using ultrafast grazing incidence X-ray scattering

Abstract

Controlling the morphology of laser-derived nanomaterials is dependent on developing a better understanding of the particle nucleation dynamics in the ablation plume. Here, we utilize the femtosecond-length pulses from an x-ray free electron laser to perform time-resolved grazing incidence x-ray scattering measurements on a laser-produced silicon plasma plume. At 20 ps we observe a dramatic increase in the scattering amplitude at small scattering vectors, which we attribute to incipient formation of liquid silicon droplets. These results demonstrate the utility of XFELs as a tool for characterizing the formation dynamics of nanomaterials in laser-produced plasma plumes on ultrafast timescales.

Abstract

The MST1R gene is overexpressed in pancreatic cancer producing elevated levels of the RON tyrosine kinase receptor protein. While mutations in MST1R are rare, alternative splice variants have been previously reported in epithelial cancers. We report the discovery of a novel RON isoform discovered in human pancreatic cancer. Partial splicing of exons 5 and 6 (P5P6) produces a RON isoform that lacks the first extracellular immunoglobulin-plexin-transcription domain. The splice variant is detected in 73% of xenografts derived from pancreatic adenocarcinoma patients and 71% of pancreatic cancer cell lines. Peptides specific to RON P5P6 detected in human pancreatic cancer specimens by mass spectrometry confirm translation of the protein isoform. The P5P6 isoform is found to be constitutively phosphorylated, present in the cytoplasm, and it traffics to the plasma membrane. Expression of P5P6 in immortalized human pancreatic duct epithelial (HPDE) cells activates downstream AKT, and in human pancreatic epithelial nestin-expressing cells, activates both the AKT and MAPK pathways. Inhibiting RON P5P6 in HPDE cells using a small molecule inhibitor BMS-777607 blocked constitutive activation and decreased AKT signaling. P5P6 transforms NIH3T3 cells and induces tumorigenicity in HPDE cells. Resultant HPDE-P5P6 tumors develop a dense stromal compartment similar to that seen in pancreatic cancer. In summary, we have identified a novel and constitutively active isoform of the RON tyrosine kinase receptor that has transforming activity and is expressed in human pancreatic cancer. These findings provide additional insight into the biology of the RON receptor in pancreatic cancer and are clinically relevant to the study of RON as a potential therapeutic target.

7. Temporal and spatial characterisation of tidal blade load variation for structural fatigue testing

Abstract

To achieve the full potential of tidal stream energy, developers are incentivised to use larger blades on tidal turbines. This requires validation of blade structural designs through full-scale blade fatigue tests to de-risk the engineering process. However, the loading scenarios encountered in testing facilities and those in reality could be significantly different, which induces errors in blade loads and fatigue damage. Here we characterise the unsteady tidal blade load variation through model-scale experiment. It was found that the standard deviations of thrust load range between 200% and 637% of condition without waves. This results in an increase of predicted fatigue damage between 6% and 18%. It was observed that the centre of effort shifts towards the blade root when encountering wave crests of opposing waves, which has not been reported in the literature to date. To reduce errors in fatigue test while the centre of effort is fixed, matching blade shear forces should be sacrificed to match target bending moment at the root. Matching blade shear forces leads to a reduction of predicted fatigue damage ranges from 17% to 25%, which can induce errors in fatigue testing. We anticipate our findings would facilitate the development of fatigue testing of tidal turbine blades.

Ipilimumab (IPI) achieved a borderline survival advantage in a large randomized trial. SIP-T and IPI are potentially synergistic. Patients and Methods: Nine men with progressive metastatic castrate-resistant prostate cancer (mCRPC) were treated prospectively with SIP-T followed immediately by IPI with one of the following doses of IPI: 1 mg/kg at 1 week after SIP-T; 1 mg/kg at 1 and 4 weeks after SIP-T; or 1 mg/kg at 1, 4, and 7 weeks after SIP-T. Three patients were evaluated at each level. Cancer-specific immunoglobulins directed at granulocyte-macrophage-colony-stimulating factor/prostatic acid phosphatase (PAP) fusion protein (PA2024) and PAP were measured prior to SIP-T, after SIP-T, 1 week after IPI, every other month for 5 months, then every 3 months for an additional 12 months. Results: Adverse events of SIP-T were consistent with previous reports. IPI only caused a transient grade 1 rash in one patient. Median age, Gleason score, and number of previous hormonal interventions were 77 years, 8, and 3, respectively. Eight men had bone metastases and one had lymph node metastasis. Statistically significant increases in serum immunoglobulin G (IgG) and IgG-IgM specific for PA2024 and PAP occurred after SIP-T. An additional statistically significant increase in the aforementioned immunoglobulins – above the levels achieved by SIP-T – occurred after IPI. Median clinical follow-up was 36 months (range: 26–40). Three patients died from progressive disease after 9, 18, and 20 months. Out of the remaining six patients, five of them needed further treatment that included abiraterone acetate, enzalutamide, radium-223 dichloride, and spot radiation. One patient had an undetectable PSA, who did not receive any other treatment except spot radiation. Median PSA at last follow-up for the surviving patients was 3.8 (range: 0.6–7.47). Conclusion: In this small trial, the addition of IPI to SIP-T was well tolerated. IPI increased immunoglobulins specific for the PA2024 protein and PAP above the level achieved with SIP-T alone. 17. Measurement of the reliability of pharmacy staff and simulated patient reports of non-prescription medicine requests in community pharmacies

Abstract

The use of simulated patients (SPs) in pharmacy practice research has become an established method to observe practice. The reliability of data reported using this method in comparison to pharmacy staff self-reported behaviour has yet to be ascertained.To compare the inter-rater agreement of pharmacy staff and SP-reported data to researcher-reported data from audio recordings of SP encounters.A dataset of 352 audio-recorded SP encounters was generated in March-October 2015 by 61 undergraduate pharmacy students completing SP visits to 36 community pharmacies in Sydney, Australia. Post-visit scores were recorded on data collection forms by SPs. Staff completed self-assessments on identical forms immediately after the encounter. Two-hundred-and-seventy visits were randomly selected as the sample for this study, where the researcher independently scored encounters via audio recordings. Inter-rater agreement was calculated through intra-class correlation (ICC) and weighted kappa analyses.Analysis of staff scores returned ICC values of 0.48 (95% CI:0.38-0.56; p 0.001) for information gathering and 0.63 (95% CI:0.55-0.70; p 0.001) for total score. Weighted kappa for information rating was 0.30 (95% CI:0.21-0.38; p 0.001) and 0.43 (95% CI:0.34-0.51; p 0.001) for overall outcome. ICC values for SPs were 0.91 (95% CI:0.88-0.93; p 0.001) and 0.90 (95% CI:0.87-0.92; p 0.001) for information gathering and total scores respectively. Weighted kappa values were 0.44 (95% CI:0.37-0.52; p 0.001) for information rating and 0.63 (95% CI:0.55-0.70; p 0.001) for overall outcome.Pharmacy staff self-reported their behaviour with a poor degree of reliability. Conversely, SPs had a high level of agreement with the researcher scoring from audio recordings. Disagreement for both groups of raters was most apparent in rating the information provided and overall appropriateness of outcome. Future research should investigate this discrepancy between staff-reported behaviour and actual behaviour and consider the implications of this discrepancy in the interpretation of self-reported data.

25. Inwardly rectifying potassium channels mediate polymyxin-induced nephrotoxicity

Abstract

Polymyxin antibiotics are often used as a last-line defense to treat life-threatening Gram-negative pathogens. However, polymyxin-induced kidney toxicity is a dose-limiting factor of paramount importance and can lead to suboptimal treatment. To elucidate the mechanism and develop effective strategies to overcome polymyxin toxicity, we employed a whole-genome CRISPR screen in human kidney tubular HK-2 cells and identified 86 significant genes that upon knock-out rescued polymyxin-induced toxicity. Specifically, we discovered that knockout of the inwardly rectifying potassium channels Kir4.2 and Kir5.1 (encoded by KCNJ15 and KCNJ16, respectively) rescued polymyxin-induced toxicity in HK-2 cells. Furthermore, we found that polymyxins induced cell depolarization via Kir4.2 and Kir5.1 and a significant cellular uptake of polymyxins was evident. All-atom molecular dynamics simulations revealed that polymyxin B1 spontaneously bound to Kir4.2, thereby increasing opening of the channel, resulting in a potassium influx, and changes of the membrane potential. Consistent with these findings, small molecule inhibitors (BaCl2 and VU0134992) of Kir potassium channels reduced polymyxin-induced toxicity in cell culture and mouse explant kidney tissue. Our findings provide critical mechanistic information that will help attenuate polymyxin-induced nephrotoxicity in patients and facilitate the design of novel, safer polymyxins.

Abstract

The “Development and Scaling of Sustainable Feeds for Resilient Aquatic Food Systems in Africa (FASA)” project is a 5-year project funded by NORAD and implemented by WorldFish. The project is focused in three Sub-Saharan African (SSA) countries, where the aquaculture sector is expanding and demand for fish is increasing. The growth of the aquaculture sector has led to dependence on importing ingredients for fish feed supply and reliance on informal, small-scale feed millers that provide inconsistent quality. The reliance on imports and inconsistent quality food can be attributed to limited knowledge of in-country ingredients and potential processing techniques for improvement. For the FASA to achieve project goals and outcomes, it is necessary to have a Monitoring, Evaluation and Learning (MEL) plan to ensure effective implementation and performance measurement. This document serves as a framework that will be used for monitoring, evaluation, and learning throughout the life of the project. The project activities, outputs, and 2 Monitoring, Evaluation and Learning (MEL) Plan outcomes are linked through the results framework, and show how the individual project activities cumulate to end goals. Each activity, output, and outcome has defined indicators and data sources. By organizing each activity with its intended output and overarching goal, this framework will aid in tracking progress during the project and show whether the intended outcomes of the project have been achieved as the project ends.

Abstract

Large scale cell based blood flow simulations are expensive, both in time and resource requirements. HemoCell can perform such simulations on high performance computing resources by dividing the simulation domain into multiple blocks. This division has a performance impact caused by the necessary communication between the blocks. In this paper we implement an efficient algorithm for computing the mechanical model for HemoCell together with an improved communication structure. The result is an up to 4 times performance increase for blood flow simulations performed with HemoCell.
