Recent advancements in polymer science and engineering underscore the importance of creating sophisticated soft materials characterized by well-defined structures and adaptable properties to meet the demands of emerging applications. The primary objective of polymeric composite technology is to enhance the functional utility of materials for high-end purposes. Both the inherent qualities of the materials and the intricacies of the synthesis process play pivotal roles in advancing their properties and expanding their potential applications. Polypyrrole (PPy)-based composites, owing to their distinctive properties, hold great appeal for a variety of applications. Despite the limitations of PPy in its pure form, these constraints can be effectively overcome through hybridization with other materials. This comprehensive review thoroughly explores the existing literature on PPy and PPy-based composites, providing in-depth insights into their synthesis, properties, and applications. Special attention is given to the advantages of intrinsically conducting polymers (ICPs) and PPy in comparison to other ICPs. The impact of doping anions, additives, and oxidants on the properties of PPy is also thoroughly examined. By delving into these aspects, this overview aims to inspire researchers to delve into the realm of PPy-based composites, encouraging them to explore new avenues for flexible technology applications.

Introduction: Doxorubicin (DOX) is a widely used anticancer drug known for its significant cardiotoxic and nephrotoxic effects. Seeking remedies to mitigate these adverse effects is crucial. This study investigates the potential of Emblica officinalis (Amla) extract, a prominent component in Chinese and Indian traditional medicine systems, in alleviating DOX-induced cardiotoxicity and nephrotoxicity. Methods: DOX (20 mg/kg i.p., once) was given to rats to cause acute cardiotoxicity and nephrotoxicity. Rats received 16 similar and cumulative doses of DOX (1.25 mg/kg, i.p.) on alternate days for chronic cardiotoxicity and nephrotoxicity. Biochemical and histological evaluations were done to confirm the onset of cardiotoxicity and nephrotoxicity. Results: The cardioprotective and nephroprotective effects of Amla extract (AE) (150 mg/kg p.o. and 300 mg/kg p.o) were evaluated in comparison to Vitamin E (25 mg/kg p.o.). The treatment with AE (300 mg/kg/day, p.o.) considerably prevented DOX-induced cardiotoxicity, nephrotoxicity, and oxidative stress by positively altering the integrity of glomeruli, restoring the tissue GSH and decreasing serum TBARS. AE (300 mg/kg) was found to be more cardioprotective and nephroprotective than Vitamin E (25 mg/kg p.o.). Discussion: It may be concluded that the induction of cardiotoxicity and nephrotoxicity in rats may be due to DOX-induced oxidative stress, and chronic treatment with AE (300 mg/kg) is an effective way to alleviate the cardiotoxic and nephrotoxic adverse effects of DOX in rats. Moreover, given Amla's historical and contemporary significance in Chinese and Indian traditional medicine systems, its potential therapeutic role merits further exploration in clinical settings.

It is well known that solid hypoxic tumour cells oxidise glucose through glycolysis, and the end product of this pathway is fermented into lactate which accumulates in the tumour microenvironment (TME). Initially, it was proclaimed that cancer cells cannot use lactate; therefore, they dump it into the TME and subsequently augment the acidity of the tumour milieu. Furthermore, the TME acts as a lactate sink with stope variable amount of lactate in different pathophysiological condition. Regardless of the amount of lactate pumped out within TME, it disappears immediately which still remains an unresolved puzzle. Recent findings have paved pathway in exploring the main role of lactate acidosis in TME. Cancer cells utilise lactate in the de novo fatty acid synthesis pathway to initiate angiogenesis and invasiveness, and lactate also plays a crucial role in the suppression of immunity. Furthermore, lactate re-programme the lipid biosynthetic pathway to develop a metabolic symbiosis in normoxic, moderately hypoxic and severely hypoxic cancer cells. For instance: severely hypoxic cancer cells enable to synthesizing poly unsaturated fatty acids (PUFA) in oxygen scarcity secretes excess of lactate in TME. Lactate from TME is taken up by the normoxic cancer cells whereas it is converted back to PUFAs after a sequence of reactions and then liberated in the TME to be utilized in the severely hypoxic cancer cells. Although much is known about the role of lactate in these biological processes, the exact molecular pathways that are involved remain unclear. This review attempts to understand the molecular pathways exploited by lactate to initiate angiogenesis, invasiveness, suppression of immunity and cause re-programming of lipid synthesis. This review will help the researchers to develop proper understanding of lactate associated bimodal regulations of TME.

Abstract Background In the present study, fatty acid synthesis is targeted to combat mammary gland carcinoma by activating prolyl hydroxylase-2 with Voacamine alone and in combination with Tamoxifen. It was hypothesized that the activation of prolyl hydroxylase-2 would inhibit the hypoxia-induced fatty acid synthesis and mammary gland carcinoma. Mammary gland carcinoma was induced with a single dose administration of N-methyl-N-nitrosourea (50 mg/kg,i.p.) and treatment with Voacamine and Tamoxifen 15 days after carcinogen administration. Results At the end of the study, hemodynamic profiling of animals was recorded to assess the cardiotoxic potential of the drug. Blood serum was separated and subjected to nuclear magnetic resonance spectroscopy. Carmine staining and histopathology of mammary gland tissue were performed to evaluate the anti-angiogenic potential of the drug. The antioxidant potential of the drug was measured with antioxidant markers. Western blotting was performed to study the effect of the drug at the molecular level. Conclusion Results of the study have shown that Voacamine treatment stopped further decrease in body weight of experimental animals. The hemodynamic study evidenced that Voacamine at a low dose is safe in cardiac patients. Microscopic evaluation of mammary gland tissue documented the anti-angiogenic potential of Voacamine and Tamoxifen therapy. Perturbed serum metabolites were also restored to normal along with antioxidant markers. Immunoblotting of mammary gland tissue also depicted restoration of proteins of the hypoxic and fatty acid pathway. Conclusively, Voacamine and its combination with Tamoxifen activated prolyl hydroxylase-2 to combat mammary gland carcinoma.

The current study investigated the role of combination therapy with voacamine and vincristine in preventing mammary gland carcinoma through prolyl hydroxylase-2 activation. Prolyl hydroxylase-2 activation leads to the downregulation of hypoxia-inducible factor-1α and fatty acid synthase. Overexpression of hypoxia-inducible factor-1α and fatty acid synthase has been previously reported in solid tumors of the mammary gland. After screening a battery of natural compounds similar to vincristine, voacamine was selected as a possible prolyl hydroxylase-2 activator, and its activity was evaluated using a 7,12-dimethylbenz[a]anthracene-induced rat model. The combination therapy was evaluated for cardiac toxicity using a hemodynamic profile. Angiogenic markers were evaluated by carmine staining. Monotherapy and combination therapy were also evaluated for liver and kidney toxicity using hematoxylin and eosin staining. The antioxidant potential was delineated using oxidative stress markers. The serum metabolomic profile was studied using NMR spectroscopy, and the disruption of fatty acids was evaluated using gas chromatography. Western blotting of proteins involved in hypoxic pathways was performed to decipher the action of therapy at the molecular level. Immunoblotting analysis validated that combination therapy has potential toss with prolyl hydroxylase-2 activity and thus initiates proteolytic degradation of hypoxia-inducible factor-1α and its consequent effects. Combination therapy also stimulated programmed cell death (apoptosis) in rapidly dividing cancer cells. The present study explored the role of voacamine inactivation of prolyl hydroxylase-2, which can decrease the overexpression of hypoxia-inducible factor-1α and fatty acid synthase in mammary gland carcinoma cells.

The novel SARS-CoV-2virus that caused the disease COVID-19 is currently a pandemic worldwide. The virus requires an alveolar type-2 pneumocyte in the host to initiate its life cycle. The viral S1 spike protein helps in the attachment of the virus on toACE-2 receptors present on type-2 pneumocytes, and the S2 spike protein helps in the fusion of the viral membrane with the host membrane. Fusion of the SARS-CoV-2virus and host membrane is followed by entry of viral RNA into the host cells which is directly translated into the replicase-transcriptase complex (RTC) following viral RNA and structural protein syntheses. As the virus replicates within type-2 pneumocytes, the host immune system is activated and alveolar macrophages start secreting cytokines and chemokines, acting as an inflammatory mediator, and chemotactic neutrophils, monocytes, natural NK cells, and CD8+ T cells initiate the local phagocytosis of infected cells. It is not the virus that kills COVID-19 patients; instead, the aberrant host immune response kills them. Modifying the response from the host immune system could reduce the high mortality due to SARS-CoV-2 infection. The present study examines the viral life cycle intype-2 pneumocytes and resultant host immune response along with possible therapeutic targets.

MXenes, a novel family of 2D transition metal carbide, nitride and carbonitride materials, have been gaining tremendous interest in recent days as potential electrocatalysts for various electrochemical reactions, including hydrogen evolution reaction (HER). MXenes are characterized by their etchable metal layers, excellent structural stability, versatility for heteroatoms doping, excellent electronic conductivity, unique surface functional groups and admirable surface area, suitable for the role of electrocatalyst/support in electrochemical reactions, such as HER. In this review article, we summarized recent developments in MXene-based electrocatalysts synthesis and HER performance in terms of the theoretical and experimental point of view. We systematically evaluated the superiority of the MXene-based catalysts over traditional Pt/C catalysts in terms of HER kinetics, Tafel slope, overpotential and stability, both in acidic and alkaline electrolytic environments. We also pointed out the motives behind the electro catalytic enhancements, the effect of synthesis conditions, heteroatom doping, the effect of surface terminations on the electrocatalytic active sites of various MXenes families. At the end, various possible approaches were recommended for a deeper understanding of the active sites and catalytic improvement of MXenes catalysts for HER.

The distribution of internal resistance in most microbial electrolysis cells (MECs) remains unclear, which hinders the optimization and scaling up of the technology. In this study, a method for quantifying the effects of design and operation decisions on internal resistance was applied for the first time to MECs. In typical single chamber MECs with carbon cloth electrodes, the internal resistance was distributed as follows: 210 Ω cm2 for anode, 77 Ω cm2 for cathode, and 11 Ω cm2 M for solution. Biomass can be a sustainable choice for bioenergy production worldwide. Biohydrogen production using fermentative conversion of biomass has gained great interest during the last decade. Besides being an efficient transportation fuel, biohydrogen can also be also be a low-carbon source of heat and electricity. Microbes assisted conversion (bioconversion) can be take place either in presence or absence of light. This is called photo-fermentation or dark-fermentation respectively. This review provides an overview of approaches of fermentative hydrogen production. This includes: dark, photo and integrated fermentative modes of hydrogen production; the molecular basis behind its production and diverse range of its applicability industrially. Mechanistic understanding of the metabolic pathways involved in biomass-based fermentative hydrogen production are also reviewed. Bioactive flavonoids derived from Asplenium nidus (fern) possess potent tumoricidal, anti-bacterial, anti-oxidant activities that are responsible for their chemo-preventive potential against multidrug-resistant (MDR) pathogens, Proteus mirabilis, Proteus vulgaris and Pseudomonas aeruginosa. Fractionation and recognition of the flavonoids were attained through gas chromatography and mass spectrometry (GC/MS). Twelve known and three unknown compounds were revealed by fractions 1 & 3, representing 13.12% and 2.61% out of the total composition (15.12%). Gliricidin7-O-hexoside was found 3.83% followed by quercetin-7-O-rutinoside (3.09%) keampferol-3-O-rutinoside (0.19%) and myricetin-3-O-rhamnoside (1.10%). The anti-bacterial and anti-oxidant activities of present fractions along with pure bioactive constituents were tested against three MDR pathogens through microbroth dilution: MIC50 ranges for amoxicillin, gliricidin7-O-hexoside and quercetin 7-O-rutinoside were 0.0003–0.06 μg/mL, 0.004–0.06 μg/mL and 0.005–6.0 μg/mL, respectively. Gliricidin-7-O-hexoside and quercetin-7-O-rutinoside’s inhibitory activities were comparable to standard antibiotic, amoxicillin (p > 0.05). Herein we report for the first time a number of flavonoids that may act as the source of therapeutically useful compounds against MDR pathogens. In addition, the most extracted flavonoids (gliricidin-7-O-hexoside (78.1%) and quercetin-7-O-rutinoside (69.2%) showed a significant in vitro antioxidant activity i.e. DPPH radical scavenging activity. Both fractions of A. nidus showed obvious cytotoxic effects on human hepatoma HepG2 and human carcinoma HeLa cells. Moreover, the anti-cancer activity generally enhanced with ameliorating antioxidant and antibacterial potential of fern’s flavonoids. These findings illustrate the potential of this fern as a probable source of bioactive constituents and provide a scientific basis for its folklore or ethno-medicinal uses for infectious diseases and cancers. Microbial fuel cells (MFCs) are biochemical systems having the benefit of producing green energy through the microbial degradation of organic contaminants in wastewater. The efficiency of MFCs largely depends on the cathode oxygen reduction reaction (ORR). A preferable ORR catalyst must have good oxygen reduction kinetics, high conductivity and durability, together with cost-effectiveness. Platinum-based electrodes are considered a state-of-the-art ORR catalyst. However, the scarcity and higher cost of Pt are the main challenges for the commercialization of MFCs; therefore, in search of alternative, cost-effective catalysts, those such as doped carbons and transition-metal-based electrocatalysts have been researched for more than a decade. Recently, perovskite-oxide-based nanocomposites have emerged as a potential ORR catalyst due to their versatile elemental composition, molecular mechanism and the scope of nanoengineering for further developments. In this article, we discuss various studies conducted and opportunities associated with perovskite-based catalysts for ORR in MFCs. Special focus is given to a basic understanding of the ORR reaction mechanism through oxygen vacancy, modification of its microstructure by introducing alkaline earth metals, electron transfer pathways and the synergistic effect of perovskite and carbon. At the end, we also propose various challenges and prospects to further improve the ORR activity of perovskite-based catalysts.

The purposes of this study were to investigate the effects of operating parameters, such as electrode material, current density, percentage of hydrogen peroxide and amount of polialuminum chloride (PAC) on chemical oxygen demand (COD) removal of palm oil mill effluent (POME). The current density was varied between 30–80 mA cm−2, PAC (1–3 g L−1) as coagulant-aid and 1 and 2% of hydrogen peroxide as an oxidizing agent. As for the performance of electrode type, iron was more effective than aluminum. It appeared that the removal of COD increased with the increased of current density. When PAC and H2O2 increased, the percent of COD removal was increasing as well. The overall results demonstrate that electrocoagulation is very efficient and able to achieve more than 70% COD removal in 180 min at current density 30–80 mA cm−2 reliant upon the concentration of H2O2 and PAC.

There are a number of experimental and theoretical studies on the energy conversion of oil palm derivative biomass. Moreover, the potential of this abundant biomass residue for renewable energy in major producing countries in Southeast Asia has been well documented. In this study, the results of an equilibrium model of downdraft gasification of oil palm fronds (OPF), developed using the Aspen Plus chemical process simulator software, and its validation are presented. In addition, an optimization of the major output parameter of importance (i.e., the higher heating value of syngas) with respect to the main operating parameters (i.e., temperature, equivalence ratio (ER), and moisture content) was performed. The response surface method (RSM) was used to determine the mathematical relationship between the response of interest, which was the heating value of syngas, and the operating conditions. This method was used to further determine the conditions that would lead to optimum higher heating values of syngas. Optimum values identified by RSM were: oxidation zone temperature of 1000 °C, moisture content in the range of 4%, and an equivalence ratio of 0.35. These optimum operating conditions and the corresponding higher heating value of syngas were found to correspond with the experimental results.

The present study is a pursuit to define implications of dual cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LOX) (DuCLOX-2/5) inhibition on various aspects of cancer augmentation and chemoprevention. The monotherapy and combination therapy of zaltoprofen (COX-2 inhibitor) and zileuton (5-LOX inhibitor) were validated for their effect against methyl nitrosourea (MNU) induced mammary gland carcinoma in albino wistar rats. The combination therapy demarcated significant effect upon the cellular proliferation as evidenced through decreased in alveolar bud count and restoration of the histopathological architecture when compared to toxic control. DuCLOX-2/5 inhibition also upregulated levels of caspase-3 and caspase-8, and restored oxidative stress markers (GSH, TBARs, protein carbonyl, SOD and catalase). The immunoblotting and qRT-PCR studies revealed the participation of the mitochondrial mediated death apoptosis pathway along with favorable regulation of COX-2, 5-LOX. Aforementioned combination restored the metabolic changes to normal when scrutinized through 1H NMR studies. Henceforth, the DuCLOX-2/5 inhibition was recorded to import significant anticancer effects in comparison to either of the individual treatments.
