Introduction: Getah virus (GETV) is a zoonotic virus transmitted via a mosquito-vertebrate cycle. While previous studies have explored the epidemiology and pathogenicity of GETV in various species, its molecular mechanisms remain largely unexplored. Methods: This study investigated the impact of GETV infection and associated molecular mechanisms on reactive oxygen species (ROS) and autophagy levels in mouse Leydig cells both in vivo and in vitro. The male mice and TM3 cells were treatment with N-acetylcysteine (NAC) to reduce cellular ROS levels. Rapamycin (Rapa) and 3-Methyladenine (3- MA) were used to change autophagy in both infected and uninfected TM3 cells. Results and Discussion: The findings revealed that GETV infection in mouse testes speciffcally targeted Leydig cells and induced oxidative stress while enhancing autophagy in testicular tissue. Using TM3 cells as an in vitro model, the study confirmed GETV replication in this cell line, triggering increased ROS and autophagy levels. Treatment with N-acetylcysteine (NAC) to reduce cellular ROS levels markedly reduced autophagy in testicular tissue and TM3 cells infected with GETV. Interestingly, the use of rapamycin (Rapa) and 3-Methyladenine (3- MA) led to autophagy change in both infected and uninfected TM3 cells, with no signiffcant alterations in cellular ROS levels. These results indicate that GETV infection elevates ROS levels, subsequently inducing autophagy in mouse Leydig cells. We also found that autophagy plays an important role in GETV replication. When autophagy levels were reduced using NAC and 3-MA, a corresponding decrease in TCID50 was observed. Conversely, upregulation of autophagy using Rapa resulted in an increase in TCID50 of GETV. Therefore, we speculate that GETV may exploit the autophagy pathway to facilitate its replication. These ffndings illuminate the interplay between GETV and host cells, providing valuable insights for therapeutic strategies targeting autophagy in GETV infections. [ABSTRACT FROM AUTHOR]

Facets engineering of high entropy alloy (HEA) nanocrystals might be achieved via shape‐controlled synthesis, which is promising but remains challenging in designing Ir‐based catalysts towards efficient and robust oxygen evolution reaction (OER) in acidic medium. Herein, icosahedra nanocrystals featured with PdCu core and IrPdCuFeNiCoMo shell were prepared by wet‐chemical reduction in one‐pot, ascribing to the initial formation PdCu core and subsequent deposition and diffusion of IrPdCuFeNiCoMo HEA shell. Sequential selective chemical etching of PdCu core results in IrPdCuFeNiCoMo HEA nanocages, delivering an overpotential of 235 mV at 10 mA cm−2, 51.0 mV dec−1, and 1624 A gIr−1 at 1.50 V vs reversible hydrogen electrode in a conventional three electrode cell. In a proton exchange membrane water electrolyzer, it delivers a low cell voltage of 1.65 and 1.77 V at a current density of 1.0 and 2.0 A cm−2, respectively, and maintains stable over 900 h at 500 mA cm−2. Theoretical calculations attribute the enhanced intrinsic activity to the broad distribution of the binding energy for OER intermediates on IrPdCuFeNiCoMo HEA, which breaks the linear scaling relationship and accelerates the OER process. The engineering of the spin state and axial coordination of the metal center of single‐atom catalyst (SAC) represents an effective strategy for regulating the catalytic activity, selectivity, and stability toward electrocatalytic reduction of CO2 (ECO2R). However, rational design and deliberate fabrication of SACs with axial coordination of specified atoms remain challenging. Herein, Ni single atoms with axial coordination of phosphorus (NiP−N4−C) and four planar nitrogen atoms are fabricated, which induces reorientation of the 3d orbitals of the Ni atom and shift of the spin state from low (S = 0) to high (S = 2). The enhanced d−p orbital coupling between the Ni and the adsorbents enhances CO2 activation and reduces energy barrier for formation of *COOH, a key intermediate in the ECO2R to produce CO, enabling the high activity and near unity selectivity of CO in ECO2R in a broad potential range of 600 mV (−0.4–−1.0 V vs reversible hydrogen electrode vs RHE), achieving a turnover frequency of 37.2 s−1 at −1.0 V versus RHE. As a bifunctional cathode electrocatalyst, NiP−N4−C demonstrates a peak power density of 18.5 mW cm−2 and maintains cycling durability over 70 h in rechargeable Zn−CO2 batteries. Senecavirus A (SVA)-associated porcine idiopathic vesicular disease (PIVD) and Pseudorabies (PR) are highly contagious swine disease that pose a significant threat to the global pig industry. In the absence of an effective commercial vaccine, outbreaks caused by SVA have occurred in many parts of the world. In this study, the PRV variant strain PRV-XJ was used as the parental strain to construct a recombinant PRV strain with the TK/gE/gI proteins deletion and the VP3 protein co-expression, named rPRV-XJ-ΔTK/gE/gI-VP3. The results revealed that PRV is a suitable viral live vector for VP3 protein expressing. As a vaccine, rPRV-XJ-ΔTK/gE/gI-VP3 is safe for mice, vaccination with it did not cause any clinical symptoms of PRV. Intranasal immunization with rPRV-XJ-ΔTK/gE/gI-VP3 induced strong cellular immune response and high levels of specific antibody against VP3 and gB and neutralizing antibodies against both PRV and SVA in mice. It provided 100% protection to mice against the challenge of virulent strain PRV-XJ, and alleviated the pathological lesion of heart and liver tissue in SVA infected mice. rPRV-XJ-ΔTK/gE/gI-VP3 appears to be a promising vaccine candidate against PRV and SVA for the control of the PRV variant and SVA. • This study reports the first-time construction of a recombinant live attenuated PRV virus, denoted as rPRV-XJ-ΔTK/gE/gI-VP3, which incorporated the VP3 gene of SVA and deleted the TK/gE/gI genes of PRV. • PRV proves to be an ideal vehicle for the persistent and stable expression of SVA VP3. • rPRV-XJ-ΔTK/gE/gI-VP3 exhibited both safety and efficacy in mice, eliciting robust specific humoral and cellular immune responses against SVA and PRV. • Intranasal immunization with rPRV-XJ-ΔTK/gE/gI-VP3 provided full protection against the PRV-XJ strain in the mice. Furthermore, vaccination with this recombinant virus reduced the virus copies and alleviated pathological inflammatory changes in the heart and liver of mice after SVA infection. Porcine reproductive and respiratory syndrome virus (PRRSV) causes a highly contagious disease that threatens the global swine industry. Recent studies have focused on the damage that PRRSV causes to the reproductive system of male pigs, although pathological research is lacking. Therefore, we examined the pathogenic mechanisms in male piglets infected with PRRSV. Gross and histopathological changes indicated that PRRSV affected the entire reproductive system, as confirmed via immunohistochemical analysis. PRRSV infected Sertoli cells and spermatogonia. To test the new hypothesis that PRRSV infection in piglets impairs blood - testis barrier (BTB) development, we investigated the pathology of PRRSV damage in the BTB. PRRSV infection significantly decreased the quantity and proliferative capacity of Sertoli cells constituting the BTB. Zonula occludens-1 and β-catenin were downregulated in cell - cell junctions. Transcriptome analysis revealed that several crucial genes and signalling pathways involved in the growth and development of Leydig cells, Sertoli cells, and tight junctions in the testes were downregulated. Apoptosis, necroptosis, inflammatory, and oxidative stress-related pathways were activated, whereas hormone secretion-related pathways were inhibited. Many Sertoli cells and spermatogonia underwent apoptosis during early differentiation. Infected piglets exhibited disrupted androgen secretion, leading to significantly reduced testosterone and anti-Müllerian hormone levels. A cytokine storm occurred, notably upregulating cytokines such as tumour necrosis factor-α and interleukin-6. Markers of oxidative-stress damage (i.e. H 2 O 2 , malondialdehyde, and glutathione) were upregulated, whereas antioxidant-enzyme activities (i.e. superoxide dismutase, total antioxidant capacity, and catalase) were downregulated. Our results demonstrated that PRRSV infected multiple organs in the male reproductive system, which impaired growth in the BTB.

Porcine reproductive and respiratory syndrome virus (PRRSV) causes significant economic losses in the swine industry. Frequent mutations and recombinations account for PRRSV immune evasion and the emergence of novel strains. In this study, we isolated and characterized two novel PRRSV-2 strains from Southwest China exhibiting distinct recombination patterns. They were designated SCABTC-202305 and SCABTC-202309. Phylogenetic results indicated that SCABTC-202305 was classified as lineage 8, and SCABTC-202309 was classified as lineage 1.8. Amino acid mutation analysis identified unique amino acid substitutions and deletions in ORF5 and Nsp2 genes. The results of the recombination analysis revealed that SCABTC-202305 is a recombinant with JXA1 as the major parental strain and NADC30 as the minor parental strain. At the same time, SCABTC-202309 is identified as a recombinant with NADC30 as the major parental strain and JXA1 as the minor parental strain. In this study, we infected piglets with SCABTC-202305, SCABTC-202309, or mock inoculum (control) to study the pathogenicity of these isolates. Although both isolated strains were pathogenic, SCABTC-202305-infected piglets exhibited more severe clinical signs and higher mortality, viral load, and antibody response than SCABTC-202309-infected piglets. SCABTC-202305 also caused more extensive lung lesions based on histopathology. Our findings suggest that the divergent pathogenicity observed between the two novel PRRSV isolates may be attributed to variations in the genetic information encoded by specific genomic regions. Elucidating the genetic determinants governing PRRSV virulence and transmissibility will inform efforts to control this devastating swine pathogen.IMPORTANCEPorcine reproductive and respiratory syndrome virus (PRRSV) is one of the most critical pathogens impacting the global swine industry. Frequent mutations and recombinations have made the control of PRRSV increasingly difficult. Following the NADC30-like PRRSV pandemic, recombination events involving PRRSV strains have further increased. We isolated two novel field PRRSV recombinant strains, SCABTC-202305 and SCABTC-202309, exhibiting different recombination patterns and compared their pathogenicity in animal experiments. The isolates caused higher viral loads, persistent fever, marked weight loss, moderate respiratory clinical signs, and severe histopathologic lung lesions in piglets. Elucidating correlations between recombinant regions and pathogenicity in these isolates can inform epidemiologic tracking of emerging strains and investigations into viral adaptive mechanisms underlying PRRSV immunity evasion. CuMnO 2 is a prototype ABO 2 -type crednerite compound featured by transition metal ions of variable valence states essential for creating novel properties and optimum performance. However, the phase stabilization region of CuMnO 2 has not yet been well established, restricting one's ability in comprehending this unique structure for functional applications. Here, layered Cu 1- z Mn 1+ z O 2 crednerite was systematically synthesized and characterized by accurately regulating the reaction parameters of hydrothermal conditions, which led to a first demonstration of the phase diagram for CuMnO 2 crednerite. The pure phase layered structure was uncovered to be stabilized under hydrothermal conditions as the temperature varies between 85 and 175 °C and the molar ratio of Cu to (Cu + Mn) varies between 0.45 and 0.55. For Cu 1- z Mn 1+ z O 2 , there appeared non-stoichiometric occupation of transition metal ions. Strikingly, different from many other layered oxides, the samples at a molar ratio of Cu:(Cu + Mn) = 0.55 showed a special structure, in which excess Cu 2+ occupied the position of the Mn 3+ site to form a Cu 2+ (3d 9 )/Mn 4+ (3d 3 ) ionic pair and traces of corresponding cationic ordered phases. Such a configuration (3d 9 /3d 3 ionic pair) gives rise to an optimum super-capacitor performance, as represented by a highest mass specific capacitance of 428.4 F/g at a current density of 1 A/g. The strategy reported in this work for mapping the phase diagram of layered CuMnO 2 crednerite is fundamentally important, which may offer guidance to explore the potentials of other ABO 2 -type compounds for functional applications.
