The Central Composite Design (CCD) within the Response Surface Methodology (RSM) was applied to optimize the enzymatic hydrolysis process. This process used Alcalase® to hydrolyze Bactronophorus thoracites protein with the goal of maximizing its antimicrobial effects. Four distinct parameters were identified as independent variables: pH (A: 8.5-10.5), temperature (B: 45-65 °C), hydrolysis time (C: 120-360 min), and enzyme-to-substrate ratio (D: 1.45%-2.65% w/v). Meanwhile, the antimicrobial activity was chosen as the response variable, specifically against Pantoea ananatis (Y1) and Pantoea stewartii (Y2). According to the findings, the constructed quadratic polynomial model showed a significant correlation with the experimental data, as evidenced by the coefficient of determination (R²) values for antimicrobial activity: Y1 being 0.9893 (p < 0.0001) and Y2 at 0.9848 (p < 0.0001). Optimal antimicrobial activity for Bactronophorus thoracites protein hydrolysates (BTPH) was recorded at 46.748% against P. ananatis and 40.768% against P. stewartii. This result was observed under the optimal conditions of pH 9.5, temperature 55°C, hydrolysis duration of 240 minutes, and 2.05% w/v enzyme-to-substrate ratio. There was a notable alignment between the actual and predicted values from our models, with the Residual Standard Error (RSE) values falling under 5%. Furthermore, the established Minimum Inhibitory Concentration (MIC) was 250µg/mL, and the Minimum Bactericidal Concentration (MBC) was 500µg/mL for both P. ananatis and P. stewartii. In this study, we aims to determine the factors that affect the consumers’ food consumption behaviour and further to examine the impact of consumers’ food consumption behaviour toward food waste in urban area. This study collected 400 questionnaires from the Klang Valley, Malaysia. The findings showed that there are three latent factors from the factor analysis which related to the negative food consumption behaviour, which were consumers’ negative attitude on food demand, perceived behavioural control on food waste, and subjective norm. Multiple regression analysis showed that the negative food consumption attitude and perceived behavioural control on food waste have significant impact on the negative food consumption behaviour. Furthermore, the chi-square test showed that the percentage of food waste related to the difference level of negative consumption behaviour on food. Besides that, the Pearson Chi-square also showed that over demand for food and did not consume the food before expired date are highly significant associated with the number of food waste created by a household. In conclusion, consumers’ proper planning on food consumption, cooking, and food deliver order playing a significant role to reduce food waste in Malaysia.

The TBX1 gene plays a critical role in the development of 22q11.2 deletion syndrome (22q11.2DS), a complex genetic disorder associated with various phenotypic manifestations. In this study, we performed in-silico analysis to identify potentially deleterious non-synonymous single nucleotide polymorphisms (nsSNPs) within the TBX1 gene and evaluate their functional and structural impact on 22q11.2DS. A comprehensive analysis pipeline involving multiple computational tools was employed to predict the pathogenicity of nsSNPs. This study assessed protein stability and explored potential alterations in protein-protein interactions. The results revealed the rs751339103(C>A), rs780800634(G>A), rs1936727304(T>C), rs1223320618(G>A), rs1248532217(T>C), rs1294927055 (C>T), rs1331240435 (A>G, rs1601289406 (A>C), rs1936726164 (G>A), and rs911796187(G>A) with a high-risk potential for affecting protein function and stability. These nsSNPs were further analyzed for their impact on post-translational modifications and structural characteristics, indicating their potential disruption of molecular pathways associated with TBX1 and its interacting partners. These findings provide a foundation for further experimental studies and elucidation of potential therapeutic targets and personalized treatment approaches for individuals affected by 22q11.2DS.
