Abstract

The genome of phage mv4 and its association with the Lactobacillus delbruekii subsp. bulgaricus chromosome were characterized. Mature mv4 homoduplexes, hybridization studies and restriction enzyme analysis showed that the mv4 genome is terminally redundant and circularly permuted. Characterization of the attP site showed that mv4 could integrate in the chromosome by the model of Campbell. The pac site was also localized to a SalI-A-SalI-E junction. Transduction studies showed that mv4 is capable of transferring a limited set of bacterial DNA. The lysogenic presence of mv4 results in resistance to mv4-like virulent phages.

Abstract

Y-autosome translocations are rare in humans and pigs. In both species, these rearrangements can be responsible for meiotic arrest and subsequent infertility. Chromosome pairing abnormalities on the SSCX, SSCY and SSC1 chromatin domains were identified by analyzing pachytene spermatocytes from a boar carrying a (Y;1) translocation by immunolocalization of specific meiotic protein combined with FISH. Disturbance of the meiotic sex chromosome inactivation (MSCI) was observed by Cot-RNA-FISH and analysis of ZFY gene expression by sequential RNA- and DNA-FISH on spermatocytes. We hypothesized that the meiotic arrest observed in this boar might be due to the silencing of critical autosomal genes and/or the reactivation of some sex chromosome genes. Copyright © 2011 S. Abstract

A comparative map of human chromosome 12 (HSA 12) and pig chromosome 5 (SSC 5) was constructed using ten pig expressed sequence tags (ESTs). These ESTs were isolated from primary granulosa cell cultures by differential display (EST b10b), or from a granulosa cDNA library (VIIIE1, DRIM, N*9, RIIID2 and RVIC1) or from a small intestine cDNA library (ATPSB, ITGB7, MYH9, and STAT2). Also used were two Traced Orthologous Amplified Sequence Tags (TOASTs) (LALBA, TRA1), one microsatellite-associated gene (IGF1) and finally five human YACs selected for their cytogenetic position, with a view to increasing the number of informative markers for the comparison. Large-insert clones were obtained by screening a pig bacterial artificial chromosome (BAC) library with specific primers for each EST and TOAST and for IGF1. These BACs were used as probes for fluorescent in situ hybridisation (FISH) both on porcine and human metaphases. In addition, the human YACs were FISH mapped on pig chromosomes. This allowed us to refine and, in some cases, to correct the previous mapping obtained with a somatic cell hybrid panel. While these data confirm chromosome painting results showing that the distal part of SSC 5p arm is conserved on HSA 22, while the rest of the chromosome corresponds to HSA 12, they also demonstrate gene-order differences between human and pig. A review of the present status of the porcine gene map is given with references. A total of 84 loci have now been studied, and genes have been assigned to 17 chromosomes. Among them, six chromosomes are defined by only one marker. No loci have been attributed yet to three chromosomes.

Abstract

It has been shown that a major gene, called RN, is responsible for the RTN technological yield, a meat quality porcine trait. Experimental families informative for the segregation of RNgene were constituted from animals belonging to the Laconie composite line. We have previously mapped the RNgene to Chromosome (Chr) 15 (Milan et al. Genet. Sel. Evol. 27, 195-199, 1995). A Chr 15 map was established with 16 markers. The RNgene was found to be located between markers Sw120and Sw936, at 2 cM from Sw936(LOD = 38.1). In addition, by localizing Sw936at 15q21–22 using DISC-PCR, we also located RNon the physical map.

Abstract

To explore the relationship between gene activity and nuclear position, genomic imprinting leading to parental-specific expression offers a good model. In one cell, it is possible to compare the nuclear environment of the two alleles for a given locus and search for a potential correlation between their nuclear position and expression status. Using 3D RNA-DNA fluorescence in situ hybridization (FISH) in porcine fetal liver cells, we focused on the imprinted region of Insulin-like growth factor 2(IGF2), a paternally expressed gene located on porcine chromosome 2. We investigated the interchromosomal interactions implicating IGF2. Through a 2D FISH screening, imprinted genes from the Imprinted Gene Network (Varrault et al., 2006) were tested for interactions in liver cells. The locus DLK1/MEG3showed the highest rate of colocalization with IGF2. By 3D RNA-DNA FISH combined to confocal microscopy, we demonstrated a preferential implication of the expressed paternal IGF2allele in a transassociation with DLK1/MEG3region (chromosome 7). We showed that this colocalization occurs also in fetal muscle and demonstrated that it occurs preferentially between the expressed IGF2, DLK1, and MEG3alleles. We are extending this analysis through an interdisciplinary approach to develop large functional mappingstudies focused on the mechanisms involved in the transcriptional regulation of genes expressed in muscle during late fetal development of pig. From a transcriptomic analysis carried on fetal muscle of two extreme genetic lines to study maturity, we identified 2000 genes differentially expressed that characterize its establishment (Voillet et al., 2014). We are now constructing, by in silico processes, networks of co-regulated genes with IGF2as starting point. We are also developing a Hi-C approach to construct interaction maps on a genome-wide scale. A set of key genes belonging to these networks and interaction maps will be selected to study by 3D FISH their position in the nuclear space in cells of the two genotypes, and to determine if co-regulated genes implicated in the same biological function co-localize in the nucleus. These data should allow us to determine if these interactions are genotype and expression pattern dependent. This will open interesting questions to study the possible link between nuclear architecture and control of gene expression in muscle in an animal model for which extreme genotypes for maturity at birth are available.
