1. Multiple Myeloma Risk and Outcomes Are Associated with Pathogenic Germline Variants in DNA Repair Genes.

Abstract

First-degree relatives of patients with multiple myeloma are at increased risk for the disease, but the contribution of pathogenic germline variants (PGV) in hereditary cancer genes to multiple myeloma risk and outcomes is not well characterized. To address this, we analyzed germline exomes in two independent cohorts of 895 and 786 patients with multiple myeloma. PGVs were identified in 8.6% of the Discovery cohort and 11.5% of the Replication cohort, with a notable presence of high- or moderate-penetrance PGVs (associated with autosomal dominant cancer predisposition) in DNA repair genes (3.6% and 4.1%, respectively). PGVs in BRCA1 (OR = 3.9, FDR < 0.01) and BRCA2 (OR = 7.0, FDR < 0.001) were significantly enriched in patients with multiple myeloma when compared with 134,187 healthy controls. Five of the eight BRCA2 PGV carriers exhibited tumor-specific copy number loss in BRCA2, suggesting somatic loss of heterozygosity. PGVs associated with autosomal dominant cancer predisposition were associated with younger age at diagnosis, personal or familial cancer history, and longer progression-free survival after upfront high-dose melphalan and autologous stem-cell transplantation (P < 0.01). Significance: Our findings suggest up to 10% of patients with multiple myeloma may have an unsuspected cancer predisposition syndrome. Given familial implications and favorable outcomes with high-dose melphalan and autologous stem-cell transplantation in high-penetrance PGV carriers, genetic testing should be considered for young or newly diagnosed patients with a personal or family cancer history. Abstract

In the central region of the galaxy cluster Abell 2199 (A2199) resides the cD galaxy NGC 6166, which spatially coincides with the 3C 338 radio source. Lobes, jets, and a more detached southern structure (similar to a jet labelled as ridge) are seen at kiloparsec-scale images of 3C 338. This unusual radio morphology has led to the proposition of different hypotheses about its physical origin in the literature. In this work, we study the feasibility of a dynamical scenario where NGC 6166 moves around the X-ray inferred centre of A2199 from the point of view of three-dimensional hydrodynamic simulations. The physical characteristics of the intra-cluster medium in which the jet propagates are constrained to those derived from X-ray observations in the vicinity of NGC 6166. Possible orbits for the jet inlet region are derived from the estimated radial velocity of NGC 6166, while the jet parameters are constrained by parsec-scale interferometric radio observations and the estimated jet power of 3C 338 obtained from radio and X-ray data. Our results show that the hypothesis of NGC 6166 has been moving around the centre of A2199 during the last tens of million of years is compatible with the general radio morphology of 3C 338. Furthermore, the proposed dynamic scenario for the motion of NGC 6166 may be linked to gravitational perturbations induced by the passage of a sub-cluster of galaxies hundreds of millions of years ago., Comment: 26 pages, 24 figures

Abstract

We study a pickup-and-delivery problem that arises when customers randomly submit requests over the course of a day from a choice of vendors on a collaborative e-commerce portal. Based on the attributes of a customer request, a dispatcher dynamically schedules the delivery service on either a dedicated vehicle or a crowdshipper, both of whom experience time dependent travel times. While dedicated vehicles are available throughout the day, the availability of crowdshippers is unknown a priori and they appear randomly for only portions of the day. With an objective of minimizing the sum of routing costs, piece-rate crowdshipper payments, and lateness charges, we model the uncertainty in request arrivals and crowdshipper appearances as a Markov decision process. To determine an action at each decision epoch, we employ a heuristic that partially destroys the existing routes and repairs them guided by a parameterized cost function approximation that accounts for the remaining temporal capacity of delivery vehicles. Through a set of computational experiments, we demonstrate the improvement of our approach over a myopic approach in several key performance metrics. In addition, we conduct computational experiments demonstrate the impact of inserting wait time in the route scheduling and the benefit of explicitly modeling time-dependent travel times. Through our computational testing, we also investigate the effect of demand management mechanisms that facilitate many-to-one request bundles or one-to-many request bundles on reducing the cost to service requests.

8. The effect of cluster dynamical state on ram-pressure stripping

Abstract

Theoretical and observational studies have suggested that ram-pressure stripping by the intracluster medium can be enhanced during cluster interactions, boosting the formation of the "jellyfish" galaxies. In this work, we study the incidence of galaxies undergoing ram-pressure stripping in 52 clusters of different dynamical states. We use optical data from the WINGS/OmegaWINGS surveys and archival X-ray data to characterise the dynamical state of our cluster sample, applying eight different proxies. We then compute the number of ram-pressure stripping candidates relative to the infalling population of blue late-type galaxies within a fixed circular aperture in each cluster. We find no clear correlation between the fractions of ram-pressure stripping candidates and the different cluster dynamical state proxies considered. These fractions also show no apparent correlation with cluster mass. To construct a dynamical state classification closer to a merging "sequence", we perform a visual classification of the dynamical states of the clusters, combining information available in optical, X-ray, and radio wavelengths. We find a mild increase in the RPS fraction in interacting clusters with respect to all other classes (including post-mergers). This mild enhancement could hint at a short-lived enhanced ram-pressure stripping in ongoing cluster mergers. However, our results are not statistically significant due to the low galaxy numbers. We note this is the first homogeneous attempt to quantify the effect of cluster dynamical state on ram-pressure stripping using a large cluster sample, but even larger (especially wider) multi-wavelength surveys are needed to confirm the results., Comment: 18 pages, 8 figures. Abstract

Paneth cells (PCs), a specialized secretory cell type in the small intestine, are increasingly recognized as having an essential role in host responses to microbiome and environmental stresses. Whether and how commensal and pathogenic microbes modify PC composition to modulate inflammation remain unclear. Using newly developed PC-reporter mice under conventional and gnotobiotic conditions, we determined PC transcriptomic heterogeneity in response to commensal and invasive microbes at single cell level. Infection expands the pool of CD74+ PCs, whose number correlates with auto or allogeneic inflammatory disease progressions in mice. Similar correlation was found in human inflammatory disease tissues. Infection-stimulated cytokines increase production of reactive oxygen species (ROS) and expression of a PC-specific mucosal pentraxin (Mptx2) in activated PCs. A PC-specific ablation of MyD88 reduced CD74+ PC population, thus ameliorating pathogen-induced systemic disease. A similar phenotype was also observed in mice lacking Mptx2. Thus, infection stimulates expansion of a PC subset that influences disease progression.

Abstract

A ciência moderna foi historicamente estabelecida como atividade masculina, concepção mantida até o século XX, a despeito da crescente participação das mulheres nas atividades científicas. Considerando a importância dos livros didáticos na educação escolar, este trabalho investiga como os livros didáticos de ciências apresentam as mulheres nas ciências no recorte temporal 1980–2022. Foram contabilizadas em coleções didáticas de três grandes editoras brasileiras todas as citações textuais e imagéticas de mulheres e homens cientistas, e analisadas via semiótica peirceana as imagens das cientistas mulheres. Resultados apontaram incremento nas referências de mulheres cientistas a partir dos anos 2000, porém sempre em número muito menor que as citações de homens. A análise semiótica das imagens permitiu identificar quatro categorias de representação de mulheres cientistas: reais (imagens de cientistas devidamente identificadas); anônimas (fotografias sem identificação); fictícias (representações figurativas de personagens criadas) e ocultadas (imagens fotográficas/pictóricas de cientistas reais sem identificação nominal). Cerca de metade das imagens situa-se na categoria reais (49%); as demais distribuem-se em: anônimas (31%), fictícias (15%) e ocultadas (5%). Apesar da crescente presença de mulheres cientistas nos livros didáticos do século XXI, o baixo número de citações, assim como as formas como são apresentadas, colabora para a manutenção da concepção masculina da ciência.

Abstract

T-cell redirection therapy using chimeric antigen receptor (CAR) T cells and bispecific antibodies (BiAbs) has shown promising efficacy in heavily pretreated patients with relapsed/refractory multiple myeloma (RRMM), leading to the approval of 2 CAR T-cell products and numerous BiAb trials. Data on the outcomes after relapse following BiAbs are urgently required to develop strategies for sequencing salvage therapies. We identified 58 patients progressing after a BiAb trial at Mount Sinai Hospital. Progression-free survival (PFS) to the first salvage (PFS1), second salvage therapy (PFS2), and overall survival (OS) were estimated using the Kaplan-Meier method. The median age of the patients was 67 years, and 78% had high-risk cytogenetics. They had a median of 6 prior therapy lines, 89% were triple-class refractory, and 44% were penta-drug refractory. After the BiAb trial, patients were followed for a median of 30.5 months and received a median of 2 additional salvage therapies (range, 1-9). The most common first salvage was T-cell redirection in 19 patients (10 BiAb and 9 CAR T cells). Ten patients underwent T-cell redirection as a second salvage treatment. T-cell redirection therapy as first or second salvage was feasible and associated with a median PFS1 of 28.9 months, PFS2 of 30.9 months, and an OS of 62% at 2 years. The sequential use of different T-cell redirection therapies is possible and may lead to deep and durable responses following the relapse after BiAb therapy in RRMM.

The visual and refractive outcomes were assessed preoperatively and 6 weeks after the surgery. Specifically, monocular uncorrected distance visual acuity (UDVA), corrected distance visual acuity (CDVA), uncorrected intermediate visual acuity (UIVA) at 80 and 60 cm and uncorrected near visual acuity at (UNVA) at 40 cm were evaluated. The rotational stability of the lens was also assessed.Results: Sixty-one eyes (98.39%) were within ± 1.00D and 55 eyes (88.71%) were within ± 0.50 D of spherical equivalent, with a mean value of 0.09± 0.39 D. 51 (82.26%) and 61 (98.39%) eyes had a UDVA of ≥ 20/20 and ≥ 20/25, respectively, and for CDVA these values were as follows: 59 (95.16%) and 62 eyes (100%), respectively. The mean UDVA and CDVA were 0.01± 0.06 and − 0.01± 0.04logMAR, respectively. Greater than or equal to unaided 20/20 vision was achieved at 40 cm in 42 (67.74%), UIVA at 60 cm in 42 (67.74%) and 50 eyes (80.65%) at 80 cm. Those achieving ≥ 20/25 were 56 (90.32%, 40 cm), 59 (95.16%, 60 cm), and 62 eyes (100%, 80 cm). Postoperative mean values were 0.04± 0.07, 0.03± 0.07, and 0.00± 0.07logMAR for UCNVA, UIVA at 60 cm, and UIVA at 80 cm, respectively. The mean rotation of the IOL was 5.8 degrees.Conclusion: This hydrophobic trifocal toric IOL provides good refractive outcomes with excellent visual acuity across multiple distances, providing a full range of focus.Keywords: astigmatism, trifocal, toric, multifocal, cataract

Abstract

Off-axis collisions between galaxy clusters may induce the phenomenon of sloshing, causing dense gas to be dragged from the cool core of a cluster, resulting in a spiral of enhanced X-ray emission. Abell 2199 displays signatures of sloshing in its core and it is possible that the orbital plane of the collision is seen nearly edge-on. We aim to evaluate whether the features of Abell 2199 can be explained by a sloshing spiral seen under a large inclination angle. To address this, we perform tailored hydrodynamical $N$-body simulations of a non-frontal collision with a galaxy group of $M_{200}=1.6\times10^{13}\,{\rm M_{\odot}}$. We obtain a suitable scenario in which the group passed by the main cluster core 0.8 Gyr ago, with a pericentric separation of 292 kpc. Good agreement is obtained from the temperature maps as well as the residuals from a $\beta$-model fit to the simulated X-ray emission. Abstract

Computer vision plays a crucial role in Advanced Assistance Systems. Most computer vision systems are based on Deep Convolutional Neural Networks (deep CNN) architectures. However, the high computational resource to run a CNN algorithm is demanding. Therefore, the methods to speed up computation have become a relevant research issue. Even though several works on architecture reduction found in the literature have not yet been achieved satisfactory results for embedded real-time system applications. This paper presents an alternative approach based on the Multilinear Feature Space (MFS) method resorting to transfer learning from large CNN architectures. The proposed method uses CNNs to generate feature maps, although it does not work as complexity reduction approach. After the training process, the generated features maps are used to create vector feature space. We use this new vector space to make projections of any new sample to classify them. Our method, named AMFC, uses the transfer learning from pre-trained CNN to reduce the classification time of new sample image, with minimal accuracy loss. Our method uses the VGG-16 model as the base CNN architecture for experiments; however, the method works with any similar CNN model. Using the well-known Vehicle Image Database and the German Traffic Sign Recognition Benchmark, we compared the classification time of the original VGG-16 model with the AMFC method, and our method is, on average, 17 times faster.
