2. Metal-Free Synthesis of 6-Phosphorylated Phenanthridines: Synthetic and Mechanistic Insights

Abstract

A novel and efficient method for the generation of phosphinoyl radicals from the combination of diphenyliodonium salt (Ph2I+, -OTf) with triethylamine (Et3N) in the presence of secondary phosphine oxides is reported. By employing this practical and simple approach, a large variety of 6-phosphorylated phenanthridines have been synthesized through the addition of phosphinoyl radicals to isonitriles as, radical acceptors. The reaction works smoothly in the absence of any transition metal or photocatalyst. Abstract

Phosphonated benzoxazole derivatives are good candidates as ligands for metal complexes. Two regioisomers were synthesized with the phosphonate function at the C-4 and C-7 positions. Different alkyl chains were also introduced at the C-2 position to functionalize these two regioisomers and increase their complexation properties. The formation of coordination complexes of selected benzoxazoles with M2+ and M3+ metal cations has been screened in solution using HRMS (high-resolution mass spectrometry) as the analytical tool.

Abstract

Functionalized aromatic alkylphosphonates, hemi-phosphonates and phosphonic acids are good candidates to elaborate water-soluble building blocks. The key step of the synthesis developed here consisted of the introduction of a phosphoryl group by an ortho-metallation reaction from protected ortho-anisidine. A practical route to phosphonated benzoxazoles was thus discovered. Chemoselective deprotections were investigated and mono-, bis-, and ter-deprotected aromatic derivatives were obtained. © Georg Thieme Verlag Stuttgart. New York.
