Abstract

International audience; Ultrasmall but multifunctional: Rigid imaging particles that are smaller than 5 nm in size can be obtained in a top-down process starting from a core–shell structure (core=gadolinium oxide; shell=polysiloxane). They represent the first multifunctional silica-based particles that are sufficiently small to escape hepatic clearance and enable animal imaging by four complementary techniques.

Abstract

International audience; We describe a setup for LIBS mapping of nanoparticles and trace metallic elements in biological organ as well as the followed experimental procedure. Mapping was performed for metallic elements such as Gd, Si, Ca and Fe, with a resolution of 100 μm on kidney slices sampled from a mouse 24 h after intravenous injection of a solution of gadolinium-based nanoparticles. An approach for quantifying Gd in the tissue is also presented with a good agreement with measurement performed by ICP-OES. We demonstrate that LIBS offers a simple and robust method to study the distribution of gadolinium-based nanoparticles in biological samples, without any labeling of the nanoparticles. The used bench-top instrumentation is fully compatible with the standard optical microscopy, which shows its large potential use in Biology and Medicine as a tool for complementary observation of trace metallic elements with respect to the classical optical observations which are generally based on the responses of biomolecules or cells.

Abstract

International audience; Gadolinium based Small Rigid Plaforms (SRPs) have previously demonstrated their efficiency for multimodal imaging and radiosensitization. Since the RGD sequence is well-known to be highly selective for αvβ3 integrins, a cyclic pentapeptide containing the RGD motif (cRGDfK) has been grafted onto the SRP surface. An appropriate protocol led to the grafting of two targeting ligands per nano-object. The resulting nanoparticles have demonstrated a strong association with αvβ3 integrins in comparison with cRADfK grafted SRPs as negative control. Flow cytometry and fluorescence microscopy have also been used to highlight the ability of the nanoparticles to target efficiently HEK293(β3) and U87MG cells. Finally the grafted radiosensitizing nanoparticles were intravenously injected into Nude mice bearing subcutaneous U87MG tumors and the signal observed by optical imaging was twice as high for SRP-cRGDfK compared to their negative analogue.

International audience; Ultrasmall gadolinium based particles have been functionalized with positively charged pyridinium quaternary ammonium and labelled with 111In. Evidence of their active targeting properties towards proteoglycans has been demonstrated in vivo after intravenous injection into rats opening thus a route to cancer imaging and therapy.

Abstract

International audience; Small Rigid Platforms (SRPs) are sub-5 nanometre gadolinium based nanoparticles that have been developed for multimodal imaging and theranostic applications. They are composed of a polysiloxane network surrounded by gadolinium chelates. A covalent coupling with quinoxaline derivatives has been performed. Such derivatives have proven their affinity for melanin frequently expressed in primary melanoma cases. Three different quinoxaline derivatives have been synthesised and coupled to the nanoparticles. The affinity of the grafted nanoparticles for melanin has then been shown in vitro by surface plasmon resonance on a homemade melanin grafted gold chip.

Abstract

International audience; Emission spectroscopy of laser-induced plasma from a thin section of mouse kidney successfully detected inorganic elements, Na, Ca, Cu, and Gd, naturally contained in the organ or artificially injected in the form of Gd-based nanoparticle. A two-dimensional scan of the sample allowed the laser beam to explore its surface with a resolution of 100 μm, resulting in a quantitative elemental mapping of the organ with sub-mM sensitivity. The compatibility of the setup with standard optical microscopy emphasizes the potential to provide multiple images of a same biological tissue with different types of response which can be elemental, molecular, or cellular.

Abstract

International audience; Amplifying local electromagnetic fields by engineering optical interactions between individual constituents of an optical antenna is considered fundamental for efficient nonlinear wavelength conversion in nanometer-scale devices. In contrast to this general statement we show that high field enhancement does not necessarily lead to an optimized nonlinear activity. In particular, we demonstrate that second-harmonic responses generated at strongly interacting optical gap antennas can be significantly suppressed. Numerical simulations are confirming silencing of second-harmonic in these coupled systems despite the existence of local field amplification. We then propose a simple approach to restore and amplify the second-harmonic signal by changing the manner in which electrically-connected optical antennas are interacting in the charge-transfer plasmon regime. Our observations provide critical design rules for realizing optimal structures that are essential for a broad variety of nonlinear surface-enhanced characterizations and for realizing the next generation of electrically-driven optical antennas.

Abstract

International audience; Several populations of Cu spherical nanoparticles grown in a silicate glass at different temperatures with respect to the glass transition temperature were studied using high-resolution, low-frequency Raman scattering and optical absorption. The analysis of the spectra shows that the annealing of the doped glass at temperatures close to T-g leads to the formation of metallic copper nanoparticles with high crystallinity, whereas lower-temperature and higher-temperature annealings result in the formation of poorer nanoparticle assemblies in terms of size distribution and/or nanocrystallinity. It is also shown that in the case where the optical data do not unambiguously reveal the presence of metallic Cu nanoparticles, the Raman probe enables us to probe their presence as a result of the resonance process.

Abstract

cited By 6; International audience; Phosphorus (P) removal in constructed wetlands (CWs) has received particular attention during the last decades by using specific materials which promote P adsorption/precipitation phenomena. Recent studies have shown an interest in using apatite materials to promote P precipitation onto their surface. About ten years of experiments at different scales (laboratory, pilot, full scale) have passed. The process is now open in France. This paper presents the first design recommendations according to the apatite mineral quality and treatment outlet requirements. Kinetics and their evolution with the material saturation degree are presented based on the k-C* model. The process can achieve 1 mgP.l-1 for long term while using surface P removal filter area of 0.5 m2.p.e.-1. With a low land footprint and high P removal efficiency, the interest of the process also result in the orthophosphates accumulation on a phosphate mineral which makes easier to reuse phosphorus from wastewater once saturated. © IWA Publishing 2011.

Abstract

High resolution low frequency Raman scattering measurements from embedded AgAu nanoparticles unveil efficient scattering by harmonics of both the quadrupolar and the spherical modes. Comparing the experimental data with theoretical calculations that account for both the embedding medium and the resonant Raman process enables a very complete description of the observed multiple components in terms of harmonics of both the quadrupolar and spherical modes, with a dominating Raman response from the former ones. It is found that only selected harmonics of the quadrupolar mode contribute significantly to the Raman spectra in agreement with earlier theoretical predictions., Comment: 11 pages, 4 figures

Abstract

International audience; Alkanethiol passivated silver nanoparticles were spread at an air/water interface to form a single monolayer film. The surface pressure isotherms and the UV-visible absorbance spectra of the film were recorded as a function of compression, whereas the dynamic behaviour was investigated by reflectance correlation spectroscopy. The film is shown to be inhomogeneous, formed by domains of particles separated by large areas of low particle density. Two distinct motions were observed: Brownian diffusion of the domains and their translational flow due to convection. From the characteristic diffusion time of the domains, and using a Stokes-Einstein analysis, the domain size is etermined as a function of surface compression. The domains start to form and grow once a fixed average article density is reached. Above this density threshold, the attractive van der Waals forces between the particles are dominating compared with the repulsion forces due to the alkanethiol chains.

Abstract

We investigate the fundamental properties of films resulting from the deposition of Eu3+ doped Gd2O3 clusters. We first report that the crystalline structure of clusters with diameter as low as 2.8 nm is still BCC (as in the bulk phase at ambient temperature and pressure). No phase transition to monoclinic structure is observed. We show that contamination from air (formation of hydroxide) is important and leads to the modification of the luminescence of the Eu3+ ions. Furthermore, films protected from this contamination have been fabricated and show a new feature (broad peak at 625 nm). It means that contamination is not the only mechanism responsible for the modification of the light emission spectrum. The crystal field symmetry breaking induced by the surface also plays a major role. Eventually, we show that the widening of the optical gap continues for these very small clusters. We discuss this effect in the frame of the quantum confinement model.

Abstract

The vibrational radial modes of silver nanospheres embedded in a glass matrix are investigated using a high sensitivity femtosecond pump-probe technique. The results yield evidence for coherent launching of the fundamental and higher order radial modes in agreement with a sphere dilation mediated excitation model. The results are consistent with low-frequency Raman scattering experiments.

Abstract

International audience; (C60)mNan clusters are produced in a tandem laser vaporization source and analyzed by photoionization and photofragmentation time-of-flight mass spectroscopy. At low sodium coverage, the special behavior of (C60)m=1,2Nan clusters is consistent with a significant electron transfer from the first six adsorbed atoms towards each of the C60 fullerenes and an ionic-like bonding in this size range. However, the stability of the (C60)Na3+ cation is found much more pronounced than the one of (C60)Na7+ predicted to be a magic size under the hypothesis of a full charge transfer from the metal atoms to the C60 molecule. When more sodium atoms are present, metal-metal bonds tend to become preponderant and control the cluster properties. Relative to the number of sodium atoms, an odd-even alternation in their stability is explained by the high dissociation rates for even-numbered clusters. The even clusters evaporate neutral sodium atoms whereas odd ones prefer to evaporate Na2 molecules. The hypotheses for the growth of a sodium droplet that does not wet the fullerene surface or for the formation of a concentric metallic layer are discussed in the light of this study.

Abstract

We investigate the binding of Si and C 60 theoretically (by ab initio calculations within the local-density approximation to the density functional theory) and confront the predictions to experimental X-ray absorption results from Si-C 60 films synthesized by the cluster beam deposition technique. The calculations predict that Si preferentially binds to hexagon-hexagon edges. The geometry with Si bound to a pentagonal face is metastable. The binding energy is in each case higher than the typical van der Waals binding energy. Extended X-ray absorption fine structure measurements reveal that in C 60 -Si obtained from (C 60 ) m Si n clusters deposition, Si is bound to the pentagonal face of two C 60 molecules. Both theoretical and experimental investigations go to show that the polymerisation of C 60 -Si clusters is possible, leading to nanostructured C 60 -based materials with high binding energy.

Abstract

Low-frequency Raman scattering experiments have been performed on thin films consisting of nickel-silver composite nanoparticles embedded in alumina matrix. It is observed that the Raman scattering by the quadrupolar modes, strongly enhanced when the light excitation is resonant with the surface dipolar excitation, is mainly governed by the silver electron contribution to the plasmon excitation. The Raman results are in agreement with a core-shell structure of the nanoparticles, the silver shell being loosely bonded to the nickel core., 3 figures. To be published in Phys. Rev. B

We review the properties of covalent clusters-based materials in relation to free cluster properties, namely carbon, silicon and mixed carbon clusters. These properties are understood in terms of quantum size especially the so called rehybridization effect. International audience; We report the three-dimensional packing of C60 clusters stabilized by the addition of Si. X-ray absorption spectroscopy reveals that Si atoms are in an unusual environment: between two C60, with ten or more carbon neighbors. According to ab initio calculations, the cohesive energy is about 2 eV per Si atom, much higher than the Van der Waals binding energy between two C60. Experiment and calculations both indicate a charge transfer from Si to C60. Eventually, the film may have a local decahedral symmetry.

Abstract

International audience; (C60)nCq+ cationic clusters are produced in a laser vaporization source by quenching the vapors from C60 and graphite targets. They are analyzed in the gas phase by abundance and photofragmentation time-of-flight mass spectroscopy. Among the large number of expected isomers, the present experiments give evidence for the stability of special arrangements that may result from the attachment of preformed carbon rings to the fullerene cage. The particular case of C10 will be discussed, considering the results obtained on complexes with one, two or even three C60 molecules. This study reveals that the incorporation of large molecules into carbon cages could be an essential mechanism during the growth process of larger fullerenes and that the polymerization of C60 molecules is promoted by carbon links involving a single atom or a dimer.

Abstract

International audience; The kinetic energy release distribution (KERD) in the vibrational autodetachment (VAD) from sulfur hexafluoride anion SF6- has been measured in a velocity map imaging spectrometer for delays in the range of a few tens of microseconds. The experimental KERD is analyzed within the framework of the detailed-balance: first using the standard Langevin model and subsequently using a more refined and realistic model based on the experimental attachment cross section. A discussion on the processes involved in the attachment and the VAD is presented based on an empirical fit of the attachment cross section. The lifetime derived from the model is in good agreement with the experimental time window, strengthening this theoretical approach for this model system.

Abstract

International audience; Naphthalene dimer cations [C10H8]2+ have been produced by using an electron cyclotron resonance plasma ion source and stored in a compact electrostatic ion storage ring. We show that the radiative cooling of these cations is much slower than the isolated monomer naphthalene cations. We also report on photo-dissociation studies in the gas phase of naphthalene dimer cations at high internal energy. The dissociation energy is estimated to 0.5 eV in close agreement with previous measurements but a factor of 2 smaller than recent (density functional theory (DFT) and ab initio) theoretical studies. As uncertainties on theory as well as on the experiment cannot be as large as this difference, we conclude that this discrepancy may be due to temperature effects with possible isomerization. As an interpretation of the photo-dissociation spectrum of naphthalene dimer cations, we propose a tentative simple analytical model based on effective Morse potentials. These effective potentials are expected to “average” temperature effects that would apparently result in a smaller energy difference between the fundamental and dissociation states due to the twisting vibration modes of the naphthalene dimer cations.

NATO-Advanced-Study-Institute Meeting on Nano- Structures for Optics and Photonics - Nano-Optics - Principles Enabling Basic Research and Applications, Erice, ITALY, JUL 04-19, 2015; International audience; The main goal of this research is the localization of Yb3+ and Er3+ rare earth ions and Co2+ transition metal ion as dopants in a glass ceramics composed of MgAl2O4 spinel nano-crystals of 10-20 nm size embedded in SiO2 glass. This is the first step of a compact self-Q-switched microchip laser. We conjugate, that is rather rare, both TEM-EDX and optical spectroscopy techniques. The use of TEM-EDX technique associated with both the elemental mapping of each dopant and the direct visualization of the heavier rare earth ions is unique way in luminescent materials which has led to the result that Er3+ and Yb3+ rare earth ions are preferentially located in the spinel nano-crystals. Regarding the Co2+ low concentration, this technique was not enough accurate and finally absorption spectroscopy technique have probed the main presence of Co2+ ions in the spinel nano-crystals. The use of site selective spectroscopy technique applied to Yb3+ structural probes allows to identify the 0-phonon broad line at 975 nm with both that of the disordered glass and that of the spinel inverted phases. A new Yb3+ radiationless center has been pointed out by the presence of a strong absorption line at 970 nm which has been assigned to the strongly perturbed area of the spinel nano-crystallite surface. This dopant characterization worthwhile to be shown in this School by applying both TEM-EDX and optical spectroscopy techniques as a pedagogical case in the research of rare earth ions and transition metal ions localization.

Abstract

International audience; Sub-5 nm multimodal nanoparticles have great potential for theranostic applications due to their easy renal elimination combined with complementary imaging properties and therapeutic facilities. Their potential clinical use requires the full characterization of not only the nanoparticle but also all its possible degradation products. We have recently proposed new ultrasmall gadolinium-based nanoparticles for multimodal imaging and radiosensitization. The aim of this article is to describe an analytical tool to characterize degradation products in a highly diluted medium. We demonstrate that HPLC coupled to electrospray ionization mass spectrometry (ESI-MS) can be used in order to determine precisely the composition of nanoparticles and their degradation fragments during aging.

Abstract

International audience; Absolute extinction measurements on individual silver nanoparticles under illumination show a steady evolution of their localized surface plasmon resonance. Their progressive transformation during light exposure and the influence of various parameters such as the nature of stabilizers, the local environment (oxygen rate), the spectral range of the incident light, and the shape of the nanoparticle (spheres or nanocubes) have been carefully investigated in correlation with transmission electron microscopy imaging. A combination of optics and electron microscopy gives evidence that photoaging mainly consists of the progressive formation of an oxide shell around a metallic silver core during light illumination. Moreover, in the case of nanocubes, the metallic core not only decreases in volume but also changes morphologically since edges and corners are rounded off during the photo-oxidation process. The generalized Mie theory and finite element method, used to calculate the optical extinction cross-section of core/shell Ag@AgxO nanoparticles, well account for the observed time evolutions of the absolute extinction spectra of the silver nanospheres and nanocubes. Furthermore, the calculated electromagnetic field at the nanocube surface, enhanced on edges and corners, can explain the higher efficiency of the photo-oxidation on edges and corners and the rounding increase under illumination.

Abstract

International audience; Here we report on the visible luminescence of single gold nanospherical particles diffusing in water excited by a pulsed-laser at 488 nm. The signal studied by fluorescence correlation spectroscopy does not display the expected characteristics. The main deviation is obtained for the diffusion time that depends on the laser irradiance. Additional advanced methods of measurements have been implemented to further characterize the emission. These methods have allowed us to demonstrate that the luminescence does not blink even in the picosecond domain and that it does not photobleach either. The comparison between the signals obtained under pulsed and continuous excitations at the same wavelength suggests that the increase in the particle temperature plays a role in the non-linear increase in the luminescence intensity with the excitation power. As in the case of two-photon excited luminescence, it implies that a model describing the luminescence process for a single particle diffusing in liquid must take into account the whole system composed of the gold particle, the capping ligands and the surrounding water.

Abstract

International audience; The stable structures of cationic neon clusters containing up to 57 atoms have been located using a diatomic-in-molecules potential energy surface and basin-hopping hierarchical optimization. The effects of vibrational delocalization were included either in the harmonic approximation, or by performing Langevin molecular dynamics simulations coupled to a quantum thermal bath at T=0. For most clusters, zero-point motion is sufficiently high to blur the picture of a single well-defined structure. However, structural diversity of the ground state wavefunction is found to be lower at sizes 14, 21, and 56, which correspond to special stabilities in experimental mass spectra.

Abstract

Contrary to the claims in a recent letter by Michaelianand Santamar´ia-Holek (MSH) [1], heat capacities can benegative in atomic clusters. This effect has been care-fully examined many times in the past [2–7]. Negativeheat capacities can be found in noncanonical ensembles,and have been extensively investigated in microcanoni-cal ensembles, whereas they are always positive in thecanonical ensemble. The finite size of the system is a suffi-cient condition for statistical ensembles to be inequivalent.This inequivalence has also been proved for systems in thethermodynamic limit [8], providing various examples withan “S-bend” in the caloric curve. The Gibbs equationinvoked by MSH does not hold in the microcanonicalensemble.On the other hand, both true non-ergodicity andthe failure to achieve ergodicity due to a finite obser-vation time may also cause S-bends in caloric curves.Intermediate states with the solid and liquid phases incontact can be thermodynamically unfavourable becauseof the free-energy cost of introducing the interface [5].The related energy barriers between the phases increasewith system size. Abstract

International audience; The present paper describes the successful one-pot synthesis of self-stabilized particles composed of amphiphilic block copolymers based on poly(methacrylic acid) (PMAA) obtained by polymerization-induced self-assembly. First, controlled radical polymerization of MAA is performed in water using the RAFT process by taking advantage of our recent results showing the successful RAFT polymerization of MAA in water [Chaduc Macromolecules 2012, 45, 1241−1247]. The so-formed hydrophilic macroRAFT agents are then chain-extended in situ with a hydrophobic monomer to form amphiphilic block copolymer chains of controlled molar mass that self-assemble into stable nanoparticles. Various parameters such as the pH, the molar mass and the concentration of the PMAA segments or the nature of the hydrophobic block have been investigated.

Abstract

International audience; Manipulation for simplifying or increasing the observed charge state distributions of proteins can be highly desirable in mass spectrometry experiments. In the present work, we implemented a vapor introduction technique to an Agilent Jet Stream ESI (Agilent Technologies, Santa Clara, CA, USA) source. An apparatus was designed to allow for the enrichment of the nitrogen sheath gas with basic vapors. An optical setup, using laser-induced fluorescence and a pH-chromic dye, permits the pH profiling of the droplets as they evaporate in the electrospray plume. Mechanisms of pH droplet modification and its effect on the protein charging phenomenon are elucidated. An important finding is that the enrichment with basic vapors of the nitrogen sheath gas, which surrounds the nebulizer spray, leads to an increase in the spray current. This is attributed to an increase in the electrical conductivity of water-amine enriched solvent at the tip exit. Here, the increased current results in a generation of additional electrolytically produced OH– ions and a corresponding increase in the pH at the tip exit. Along the electrospray plume, the pH of the droplets increases due to both droplet evaporation and exposure to basic vapors from the seeded sheath gas. The pH evolution in the ESI plume obtained using pure and basic seeded sheath gas was correlated with the evolution of the charge state distribution observed in mass spectra of proteins, in the negative ion mode. Taking advantage of the Agilent Jet Stream source geometry, similar protein charge state distributions and ion intensities obtained with basic initial solutions, can be obtained using native solution conditions by seeding the heated sheath gas with basic vapors.

Abstract

A theoretical study in the framework of the density functional theory is performed to investigate the stability, the structural and electronic properties of both neutral and cationic copper bromide clusters (CuBr) n (+), n = 1–6. The most stable isomers are found to be cyclic arrangements. Calculated infrared frequencies are compared with the available experimental spectra. The nature of the ionio-covalent bonding is characterized. The fragmentation, the ionization potentials are also investigated.

Abstract

International audience; Achieving (bio) macromolecular structural assignment from the interpretation of ion mobility spectrometry (IMS) experiments requires successful comparison with computer modeling. Replica-exchange molecular dynamics simulations with suitable force fields not only offer a convenient framework to locate relevant conformations, especially in the case of multiple-funnel energy landscapes, but they are also well suited to statistical analyses. In the present paper, we discuss two extensions of the method used to improve its efficiency in the context of IMS. Two doubly-protonated polyalanines [RA(4)XA(4)K + 2H](2+) with X=V and D appear as favorable cases for which the calculated collision cross-section distributions naturally agree with the measurements, providing reliable candidate structures. For these compounds, a careful consideration of other order parameters based on the weighted histogram method resolves several otherwise hidden underlying conformational families. In the case of a much larger peptide exhibiting bistability, assignment is more difficult but could be achieved by guiding the sampling with an umbrella potential using the square gyration radius as the biasing coordinate. Applied to triply protonated bradykinine, the two presented methods indicate that different conformations compatible with the measurements are very close in energy.

Abstract

Gold(I)– and silver(I)–thiolate oligomers generated under nanoparticle growth conditions have been suggested to play an important role in the growth mechanism of thiolate-protected noble metal clusters. In this work, we explore the formation of isolated noble metal–glutathione complexes by complementing electrospray mass spectrometry and optical action spectroscopy with TDDFT calculations. We have isolated and recorded action spectra of [Au+GSH–2H]−, [Ag+GSH–2H]−, and [3Ag+2GSH–4H]− complexes. Competition between photofragmentation and photodetachment channels related to electron binding energies was observed. Our findings show that, although structural properties of silver and gold metal–glutathione oligomers are similar, their optical properties in the UV range differ substantially. Abstract

International audience; We have studied the photodissociation of gas-phase deprotonated caerulein anions by vacuum ultraviolet (VUV) photons in the 4.5 to 20 eV range, as provided by the DESIRS beamline at the synchrotron radiation facility SOLEIL (France). Caerulein is a sulphated peptide with three aromatic residues and nine amide bonds. Electron loss is found to be the major relaxation channel at every photon energy. However, an increase in the fragmentation efficiency (neutral losses and peptide backbone cleavages) as a function of the energy is also observed. The oxidized ions, generated by electron photodetachment were further isolated and activated by collision (CID) in a MS3 scheme. The branching ratios of the different fragments observed by CID as a function of the initial VUV photon energy are found to be independent of the initial photon energy. Thus, there is no memory effect of the initial excitation energy on the fragmentation channels of the oxidized species on the time scale of our tandem MS experiment. We also report photofragment yields as a function of photon energy for doubly deprotonated caerulein ions, for both closed-shell ([M - 2H](2-)) non-radical ions and open-shell ([M - 3H](2-center dot)) radical ions. These latter ions are generated by electron photodetachment from[M - 3H](3-) precursor ions. The detachment yield increases monotonically with the energy with the appearance of several absorption bands. Spectra for radical and non-radical ions are quite similar in terms of observed bands; however, the VUV fragmentation yield is enhanced by the presence of a radical in caerulein peptides.

Abstract

International audience; Characterization of end-groups in poly(methacrylic acid) (PMAA) was achieved using tandem mass spectrometry after activated-electron photodetachment dissociation (activated-EPD). In this technique, multiply deprotonated PMAA oligomers produced in the negative-ion mode of electrospray ionization were oxidized into radical anions upon electron photodetachment using a 220 nm laser wavelength, and further activated by collision. In contrast to conventional collision induced dissociation of negatively charged PMAA, which mainly consists of multiple dehydration steps, fragmentation of odd-electron species is shown to proceed via a radical-induced decarboxylation, followed by reactions involving backbone bond cleavages, giving rise to product ions containing one or the other oligomer termination. A single radical-induced mechanism accounts for the four main fragment series observed in MS/MS. The relative position of the radical and of the anionic center in distonic precursor ions determines the nature of the reaction products. Experiments performed using PMAA sodium salts allowed us to account for relative abundances of product ions in series obtained from PMAA, revealing that ion stability is ensured by hydrogen bonds within pairs of MAA units.

International audience; A laser-induced fluorescence set-up profiling physico-chemical parameters of droplets and their changes in an electrospray plume is coupled to an Agilent single quadrupole mass spectrometer equipped with an Agilent Jet Stream electrospray source. This coupling allows investigating the evolution of the pH of droplets as they evaporate in the electrospray plume by measuring emission spectra of a pH-chromic dye. Simultaneous measurement of mass spectra and optical pH profiling permits to study the relation between the charge states of peptide anions in mass spectra and pH changes in the spray plume. In particular, due to changes in droplets evaporation rates, the source parameters (i.e., sheath gas flow and temperature) influence the pH of the droplets and relative intensities of charge states in peptide mass spectra.

Abstract

International audience; The acoustic vibrations of metal nanoparticles encapsulated in a dielectric shell (Ag@SiO2) were investigated using a time-resolved femtosecond technique. The measured vibration periods significantly differ from those predicted for the bare metal cores and, depending on the relative core and shell sizes, were found to be either larger or smaller than them. These results show that the vibration of the whole core-shell particle is excited and detected. Moreover, vibrational periods are in excellent agreement with the predictions of a model based on continuum thermoelasticity. However, such agreement is obtained only if a good mechanical contact of the metal and dielectric parts of the core-shell particle is assumed, providing a unique way to probe this contact in multimaterial or hybrid nano-objects.

Abstract

International audience; Laser ablation coupled to inductively coupled plasma optical emission spectrometry (LA-ICP-OES) and laser-induced breakdown spectroscopy (LIBS) were investigated for the determination of Ca, Mg, Zn and Na in milk samples. The accuracy of both methods was evaluated by comparison of the concentration found using LA-ICP-OES and LIBS with classical wet digestion associated with ICP-OES determination. The results were not fully acceptable, with biases from less than 1% to more than 60%. Matrix effects were also investigated. The sample matrix can influence the temperature, electron number density (n (e)) and other excitation characteristics in the ICP. These ICP characteristics were studied and evaluated during ablation of eight milk samples. Differences in n (e) (from 8.9 to 13.8 × 10(14) cm(-3)) and rotational temperature (ranging from 3,400 to 4,400 K) occurred with no correlation with trueness. LIBS results obtained after classical external calibration procedure gave degraded accuracy, indicating a strong matrix effect. The LIBS measurements clearly showed that the major problem in LA-ICP was related to the ablation process and that LIBS spectroscopy is an excellent diagnostic tool for LA-ICP techniques.

Abstract

Gas-phase structures of mass-selected singly- and doubly charged angiotensin ions have been determined by means of infrared multiple photon dissociation (IRMPD) spectroscopy and ion mobility spectrometry. Simulation of IRMPD spectra at the DFT level provides the location of the proton on the Arg side-chain in the case of the singly charged species. Interpretation of the ion mobility data suggests that the structures of singly- and doubly charged species are rather similar except for an internal proton transfer.

Abstract

International audience; The conformations of a number of M2TMP-(22-46) sequence variants have been investigated using ion mobility spectrometry (IMS). Substantial conformational changes were evidenced by IMS upon the variation of a single amino acid in the peptide sequence, with two main drift time signatures. Replica-exchange molecular dynamics simulations were used to help assign the structures of the different identified conformers. Even though one-on-one agreement with experiment was found for only two variants, the simulations generally confirmed the existence of two structural families. Based on these results, most of the triply protonated variants, including the wild-type peptide, were found to display collision cross sections in agreement with compact conformations in the gas phase, whereas they tend to form extended alpha-helices in the condensed phase, as confirmed by circular dichroism and previously reported NMR measurements. The destabilization of alpha-helices in vacuo upon amino acid substitution is interpreted as being driven by the solvation pattern of the charges.

Abstract

International audience; Mineral elements contained in commercially available milk powders, including seven infant formulae and one adult milk, were analyzed with inductively coupled plasma atomic emission spectrometry (ICP-AES) and laser-induced breakdown spectroscopy (LIBS). The purpose of this work was, through a direct comparison of the analytical results, to provide an assessment of the performance of LIBS, and especially of the procedure of calibration-free LIBS (CF-LIBS), to deal with organic compounds such as milk powders. In our experiments, the matrix effect was clearly observed affecting the analytical results each time laser ablation was employed for sampling. Such effect was in addition directly observed by determining the physical parameters of the plasmas induced on the different samples. The CF-LIBS procedure was implemented to deduce the concentrations of Mg and K with Ca as the internal reference element. Quantitative analytical results with CF-LIBS were validated with ICP-AES measurements and nominal concentrations specified for commercial milks. The obtained good results with the CF-LIBS procedure demonstrate its capacity to take into account the difference in physical parameters of the plasma in the calculation of the concentrations of mineral elements, which allows a significant reduction of the matrix effect related to laser ablation. We finally discuss the way to optimize the implementation of the CF-LIBS procedure for the analysis of mineral elements in organic materials

International audience; The development of strategies based on mass spectrometry to help for deep structural analysis of acidic oligosaccharides remains topical. We thus examined the dissociation behavior of deprotonated ions of heparin-derived di- to tetra-saccharides under UV irradiation at 220 nm. Depending on the ionization state of the carboxylic groups, an oxidized species issued from electron photodetachment was observed in complement to photoinduced fragmentation of precursor ions. The influence of the charge location in the oligosaccharide dianions on the balance between photodissociation and electron photodetachment is examined and a way to direct the relaxation pathways, (i.e., dissociation versus electron detachment), is proposed using sodium adducts. The oxidized species was subjected to activated-electron photodetachment (activated-EPD) leading to complementary informative fragment ions to those issued from photodissociation. Directed photoinduced dissociation at 220 nm and activated-EPD should complement the more conventional CAD and IRMPD activation modes for deeper structural analysis of acidic oligosaccharides-derived anions.

Abstract

Gabedit is a freeware graphical user interface, offering preprocessing and postprocessing adapted (to date) to nine computational chemistry software packages. It includes tools for editing, displaying, analyzing, converting, and animating molecular systems. A conformational search tool is implemented using a molecular mechanics or a semiempirical potential. Input files can be generated for the computational chemistry software supported by Gabedit. Some molecular properties of interest are processed directly from the output of the computational chemistry programs; others are calculated by Gabedit before display. Molecular orbitals, electron density, electrostatic potential, nuclear magnetic resonance shielding density, and any other volumetric data properties can be displayed. It can display electronic circular dichroism, UV-visible, infrared, and Raman-computed spectra after a convolution. Gabedit can generate a Povray file for geometry, surfaces, contours, and color-coded planes. Output can be exported to a selection of popular image and vector graphics file formats; the program can also generate a series of pictures for animation. Quantum mechanical electrostatic potentials can be calculated using the partial charges on atoms, or by solving the Poisson equation using the multigrid method. The atoms in molecule charges can also be calculated. Gabedit is platform independent. The code is distributed under free open source X11 style license and is available at http://gabedit.sourceforge.net/.

Abstract

We report a theoretical work on the electronic excitation spectrum of two heparins like disaccharides. Time dependent density functional theory (TD-DFT) calculated spectra for these sulfated and carboxylated disaccharides are presented. The orbitals involved in the observed electronic excitations are analyzed. ππ ∗ excitations involving the acidic functions lead to a strong optical absorption of these compounds between 270 and 220 nm, which is in agreement with experimental spectra reported previously [16] .

Abstract

A colloidal solution of Au25(SG)18-protected metal gold clusters was electrosprayed. [Au25(SG)18-6H]7− was selected, stored in a quadrupole ion trap, and irradiated with a UV−vis tunable laser. The excess charges are due to the deprotonation of six COOH moieties and a net charge of −1 on the Au core. Photoexcitation of this species results in electron detachment. The action spectrum obtained by recording the electron detachment yield as a function of the laser wavelength displays a strong decrease as the photon energy decreases and the onset of a band at around 650 nm. This spectrum is compared with the absorption spectrum recorded for Au25(SG)18 in solution. We also show that electron detachment is a one-photon process and likely involves, at low photon energy, ionization by tunneling through a repulsive coulomb barrier.

Abstract

International audience; The impact of quantum confinement on the width of the surface plasmon resonance of a metal nanoparticle is theoretically investigated in a model system formed by a silver nanosphere in different environments. Calculations are performed using the time-dependent local density approximation (TDLDA) for nanoparticle diameters up to about 11 nm, permitting precise quantification of the surface plasmon broadening due to size reduction. As expected, this is found to be inversely proportional to the particle diameter, but with an amplitude strongly depending on the environment (increasing by a factor of 4 when changing from vacuum to alumina). This is ascribed to the fact that damping is governed by the electronic surface spill-out (inherent in any quantum model) and thus strongly depends on the surface profile of the confining potential, that is, on the particle surface conditions.

Abstract

I investigated the structural and electronic properties of both Na(n)C(60) and Li(n)C(60) (nor = 12) clusters in the framework of density-functional theory. The behavior of the alkali atoms over the C(60) surface is analyzed. The hypotheses for either an homogeneous coating of the C(60) surface by the alkali atoms or the growth of an alkali droplet not wetting the fullerene surface are discussed. Lithium atoms are found to coat homogeneously the fullerene on the C(60) surface via pentagonal sites, contrary to sodium atoms, which prefer to form 4-atom islands on the surface. The charge transfer, the energetics, and the dipole moments are discussed in relation with available results. The adsorption of alkali atoms on the C(60) surface considerably enhances the capacity of C(60) to accept electrons. The arrangement of lithium atoms seems more favorable for the hydrogen storage than that of sodium atoms.

Abstract

Chemical shifts δ have been calculated for the 1 H attached to carbon atoms of sarcosine. Eight levels of theory within the DFT approach were used, mixing the four functionals B3LYP, PBE, OPBE, PBE0 and the two basis sets 6-311++G∗∗ and pcJ2. Boltzmann weighted isomer effects have been evaluated. By comparison of the 1 H NMR spectrum simulated from the calculated δ and the experimental one that we acquired at 300 MHz, the B3LYP/6-311++G∗∗ calculation was seen to be a good compromise between accuracy and cost. Zero-point vibrational corrections, estimated using a second-order perturbation approach, increase the agreement with experiment.

Abstract

Summary Survival following pancreatic cancer remains poor despite incremental advances in surgical and adjuvant therapy, and new strategies for treatment are needed. Oncolytic virotherapy is an attractive approach for cancer treatment. In this study, we have evaluated the effectiveness of the Lister vaccine strain of vaccinia virus armed with the endostatin-angiostatin fusion gene (VVhEA) as a novel therapeutic approach for pancreatic cancer. The Lister vaccine strain of vaccinia virus was effective against all human pancreatic carcinoma cells tested in vitro, especially those insensitive to oncolytic adenovirus. The virus displayed inherently high selectivity for cancer cells, sparing normal cells both in vitro and in vivo, with effective infection of tumors after both intravenous (IV) and intratumoral (IT) administration. The expression of endostatin-angiostatin fusion protein was confirmed in a pancreatic cancer model both in vitro and in vivo, with evidence of inhibition of angiogenesis. This novel vaccinia virus demonstrated significant antitumor potency in vivo against the Suit-2 model by IT administration. The present study suggests that the novel Lister strain of vaccinia virus armed with the endostatin-angiostatin fusion gene is a potential therapeutic agent for pancreatic cancer.
