Abstract

Ampelocissus africana (Lour) Merr (Vitaceae) is a plant used in traditional medicine in Burkina Faso in the treatment of wounds, edema, drops, infections. This study focused on pharmacological investigations effects of methanolic and aqueous extracts from Ampelocissus africana (Lour) Merr rhizomes.The acute toxicity of the extracts from Ampelocissus africana was performed under OECD 423 guidelines. The anti-inflammatory activity was evaluated in vivo using carrageenan-induced paw edema of mice at the doses to 50, 100, 200, 400, 600 mg/kg body weight (bw). The antinociceptive activity of the extracts was determined using acetic acid, formalin nociception models in mice at the doses to 50, 100, 200, 400 and 600 mg/kg bw. The involvement of ATP-sensitive K+ channel pathway and opioid system were tested using glibenclamide, and naloxone respectively at the unique dose (400 mg/kg).No mortality of mice were observed at dose of 2000 mg/kg b.w. The lethal dose (LD50) value estimated to 5000 mg/kg b.w. The tests were showed that the extracts exerted significant dose-dependent anti-inflammatory responses in the paw induced by carrageenan (from 37 to 72.90 percent inhibition), antinociceptives in acetic-induced abdominal contractions (from 25.10 to 63.08%), and in formaldehyde-induced paw licking (from 9.27 to 71.77%) tests. The pretreatment with naloxone and glibenclamide did not affect the antinociceptives effects.The results suggested that the extracts from the rhizomes of Ampelocissus africana possessed anti-inflammatory and antinociception effects in mice. These effects could justify the uses of this plant in traditional medicine.

Abstract

Balanites aegyptiaca (L.) Del (Balanitaceae) is traditionally used for the treatment of various ailments such as syphilis, jaundice and liver disorders, epilepsy, ... This study was designed to evaluate acute toxicity and hepatoprotective effect of aqueous extract of Balanites aegyptiaca on CCl4 induced hepatotoxicity in rats.Methods: Acute toxicity was assessed with the extract at a dose of 2000 mg / kg bw. The extract at doses of 25, 50 and 100 mg / kg b.w. was orally administered respectively to CC14-induced hepatotoxicity (0.5 ml / kg) animals. Silymarin (100 mg / kg) was given as a reference. Biochemical parameters such as ALT, AST, PT, ALB and ALP were assayed as well as enzymatic antioxidant activities SOD, CAT and MDA. Nitrogen monoxide (NO) involved in inflammation was also measured.Results: Activities of liver marker enzymes, ALT, AST and ALP, total protein, albumin and showed a significant hepatoprotective effect. Regarding antioxidant enzymatic activities in vivo (SOD, CAT and MDA) of aqueous extract exhibited a significant effect showing increasing levels of SOD, CAT and reducing malondialdehyde (MDA) levels. The production of NO is significantly reduced compared to the batch intoxicated by CCl4.Conclusion: Balanites aegyptiaca is endowed with hepatoprotective properties that can be attributed to antioxidant potential which could justify its use in traditional medicine in liver disorders.

Abstract

Introduction. Le bilan lipidique correspond à un ensemble d’analyses permettant de mettre en évidence des anomalies du métabolisme des lipides et d’optimiser la prise en charge diététique et/ou le besoin thérapeutique pour le suivi, la surveillance et l’évaluation du risque athérogène. L’objectif de cette étude est d’évaluer la place du jeûne dans les recommandations pré analytiques du dosage des paramètres lipidiques. Matériels et Méthodes. Notre étude prospective a été réalisée sur 27 prélèvements sanguins sur tube hépariné, des patients en deux temps : à jeun et non à jeun après la prise d’un repas consistant. Le dosage des deux paramètres : Triglycérides et le Cholestérol total, ont été réalisé par la méthode colorimétrique enzymatique. Résultats. Nos résultats ont été classés en deux groupes pour chaque paramètre. L’étude statistique a été réalisée sur le logiciel Graph Pad Prism7. Abstract

Introduction. Les mutations délétères (classe 5) des gènes BRCA1 et BRCA2 sont des mutations inactivatrices, qui entraînent une perte de la fonction de la protéine. Ces mutations constitutionnelles entrainent une augmentation du risque tumoral de cancer du sein et /ou de cancer de l’ovaire. Dans certaines situations la présence simultanée de deux variants de classe 1 (POLYMORPHISME) des gènes BRCA1 et ou BRCA2, sont responsables d’une augmentation du risque tumoral ovarien. Matériels et Méthodes. Nous avons réalisé le test génétique BRCA1 et BRCA2 par séquençage direct Sanger ; chez 88 cas index de cancer familiaux sein et ou ovaire provenant de tout le territoire national. Résultats. Trois situations d’association de variants polymorphes de classe 1 ont été identifiées. Il s’agit de l’association des deux polymorphismes : (c.2612C>T ; p.Pro871Leu) et (c.1067A>G ; p.Gln356Arg). Conclusion. Cette association dangereuse de variants polymorphes doit être prise en considération par le clinicien afin d’assurer une surveillance étroite, adaptée, dans le but d’un diagnosticprécoceducancerdel’ovaire ou de proposer une chirurgie ovarienne prophylactique aux femmesquiportentcesvariations.

Abstract

Lantana rhodesiensis Moldenke an herbaceous, belonging to the Verbenaceae family is widely used in Africa folk medicine for the treatment of cancer, measles, malaria, smallpox, strength, coughs, fever, rheumatism, body pains, diabetes mellitus arrhythmia, parasitic diseases, arterial hypertension, old diarrhoeas. Many studies have been conducted on the chemical composition of the whole plant of L. rhodesiensis as well as biological activities. The aim of the present review was to give a detailed literature survey on its traditional uses, phytochemistry and therapeutical properties of L. rhodesiensis.

Abstract

Lantana rhodesiensis Moldenke, Lippia chevalieri Moldenke, Vitex diversifolia Bak and Vitex doniana Sweet are four species of Verbenaceae family widely used in traditional medicines in Burkina. The aim of this investigation was to study the chromatographic profile of phenols acids and that of the flavonoids as well as the antioxidants activities. Then, three types of extraction were made: acetonic (100%), decoction, ethanol-water(80/20 + 20/80); and fractions of ethanol-water which arehexane fraction (HF), ethyl acetate fraction (EAF), butanol fraction (BF) and aqueous fraction (AqF) of each species. These evaluations were supplemented by the quantification of the metal ions and the characterization by atomic absorption and HPLC-DAD methods of totals phenols acids and totals flavonoids.The whole of these proportioning was directed against the antioxidant activity through the anti-DPPH* (free radical scavenging activity), anti-TAC (total antioxidant capacity) and anti-IRP(iron reducing power).36 flavonoids on 41 were highlighted by the HPLC-DAD method. A derivative glycosylate of quercetin was detected in the V. diversifolia extracts. L. chevalieri presented the strongest value in terms of ions content with a 55.88 g/kg of calcium.In terms of antioxidant activity, L. rhodesiensis and L. chevalieri presented the best activities on total antioxidant activity and anti-DPPH, and iron reducing power respectively.It’s arise that decoction extract have the best’s activities and can justify the traditional uses.

Abstract

Le diabète a atteint des proportions épidémiques dans le monde entier. Des éléments de preuves suggèrent qu’il y a une interaction complexe entre les gènes et l'environnement pouvant ainsi jouer un rôle majeur dans la pathogénie multifactorielle de cette maladie et ses complications évoquant ainsi, un résultat de l'implication des facteurs épigénétiques. De récentes études montrent que les facteurs épigénétiques, y compris la méthylation de l'ADN et les modifications de l’histone, peuvent affecter la sensibilité ainsi que l’évolution des complications du diabète de type 2. Toutefois, nos connaissances sur les mécanismes moléculaires reliés aux facteurs environnementaux et le diabète de type 2 restent limitées.

Abstract

Background: A quality system in a clinical laboratory involves supervision of all the phases of the total testing process: pre-analytical, analytical, and post-analytical. Misinterpretations during the last phase may cause a wrong diagnosis. Aim: Improvement of the interpretation of test results in the laboratory of biochemistry - CHU Batna, by the introduction of computer software ensuring better delivery and interpretation of results. Method: The updating of the reference values was based on the biochemistry analysis guides and equipment method sheets used in the biochemistry laboratory. The development of the software used two programs, Visual Basic 6 for the interface and Access 2007 for setting up the database. The first version of the software includes only general biochemistry. Results and Discussion: The analysis report obtained by the software developed presents specific reference values to each patient, and allows a record of results as an electronic file that can be archived and/or sent directly to the requesting service in the case of the presence of an intranet. In the future, the development of this first version of the software will focus on the introduction of tumor markers and hormonology with regular reviews of the reference values.

Abstract

Le cancer représente la deuxième cause de mortalité dans le monde. Sa prise en charge est multidisciplinaire dont la part du biologiste est non négligeable. Le dosage des marqueurs tumoraux sériques permet d’identifier dans le sang des substances pouvant indiquer la présence d’un cancer. En pratique médicale, on assiste souvent à des prescriptions fréquentes du dosage de marqueurs tumoraux et qui ne sont pas toujours appropriées, entrainant ainsi un surcout financier important. Dans cet article, sont passés en revue les points particuliers concernant les dosages des marqueurs tumoraux sériques : classification, indications et intérêt de la prescription, ainsi que les bonnes pratiques que chaque médecin doit connaitre et bien maitriser.

Abstract

Lippia chevalieri Moldenke is an herbaceous, belonging to the Verbenaceae family largely used in folk medicine in Burkina Faso and in certain African countries. The aim of the present review was to give a detailed literature survey on its traditional uses, and phytochemistry and therapeutical properties in order to systematize achievements and direct further research. Composition and biological activities of its essential oil have been largely investigated. Literature shows that this species of plant is used mostly for the treatment of malaria, mental disorder, liver pathologies, hepatitis, anemia and respiratory disorders. We note that the most published studies on this plant are concerned to its content of essential oil.

Abstract

In the present study, the antioxidant and antibacterial activities of methanolic extracts of two Verbenaceae (Lantana camara L. and Lippia chevalieri Moldenke: aerial part) were investigated. Their polyphenolic composition in the ethyl acetate and aqueous fractions were characterized by HPLC-DAD. The antioxidant capability of the methanolic extracts was assessed by the Ferric Reducing Antioxidant Power (FRAP) and the scavenging activity of the free radical 2,2-diphenyl-1-picrylhydrazyl (DPPH). The Minimum Inhibitory Concentrations (MIC) of the methanolic extracts (25 µg mL-1), and the minimum bactericidal concentrations (MBC) (12.5 µg mL-1) against 13 pathogenic bacteria and four serotyped bacteria from the American Type Culture Collection (ATCC) were also determined by the agar-well diffusion method. The results indicated that the L. chevalieri extracts are rich in phenolic compounds (among the 27 polyphenolic compounds detected, 20 belong to L. chevalieri) and showed the highest antioxidant activities, simultaneously on iron (III) to iron (II)-reducing activity and the radical scavenging activity. However, L. camara displayed the best and the broadest antimicrobial spectrum, especially on Shigella flexneri and Pantoea sp. (two Gram-negative strains of bacteria). The nature of polyphenolics compounds detected (phenol acid and flavone) in the L. camara can justify this activity.Key words: Verbenaceae, HPLC-DAD, antioxidant activity, antimicrobial activity, polyphenolic compounds

Abstract

Our aim in this study concerning the ethyl acetate and dichloromethane fractions was to provide a scientific basis for the treatment of hepatitis B in Burkina Faso of these three ethnomedicinal plants. As a result, we evaluated polyphenol content, antioxidant and anti-inflammatory evaluated by lipoxygenase (LOX) inhibitory and Xanthine Oxidase (XO) activities of aqueous acetone bioactive fractions from three species of Malvaceae (Sida cordifolia, Sida rhombifolia, S. urens). Folin-ciocalteu; AlCl3 methods and tannic acid respectively were used for polyphenol content research. The antioxidant activity of the samples was evaluate using three separate methods, inhibition of free radical 2,2-diphenyl-1-picrylhydramzyl (DPPH), ABTS radical cation decolorization assay, Iron (III) to iron (II) reduction activity (FRAP). For anti-inflammatoty activity, lypoxygenase and xanthine oxidase inhibitory activities were used. Finally, in this study, the ethyl acetate fraction has shown the best results comparatively to the dichloromethane fraction. Abstract

La vitamine D a des effets sur l’os qui sont bien connus. Ces dernières années, d’autres effets ont été mis en évidence et les études observationnelles ont noté des associations avec de nombreuses pathologies. Le déficit en cette vitamine est très fréquent. L’évaluation du statut en vitamine D repose sur le dosage de la 25-OHD qui a très récemment fait l’objet d’une standardisation. Alors que l’intérêt de la supplémentation en vitamine D dans le cadre des pathologies osseuses est clairement établi, son intérêt dans le cadre des maladies métaboliques et immunitaires n’est pas encore démontré.

Abstract

The present study was conducted to evaluate the pharmacological and toxic effects of aqueous acetone extract of Sida alba L. a Malvaceae species, in mice Swiss and albinos Wistar rats. In acute toxicity test, mice received doses of this extract by intraperitoneal route with LD50 value of 3200 mg/kg. In sub-acute toxicity test, albinos Wistar rats were treat by gavage during 28 days with different doses of aqueous acetone extracts of Sida alba L., (75, 100 and 150 mg/kg). About to the pharmacological properties, the results varied widely in dose of extract and weight of rats and did not show clinical correlations. We undertook this study of extracts in order to provide a scientific basis for the traditional use of Sida alba L., in traditional medicine particularly to treat hepatitis B. Our results of this study appeared to show the safety of acute and sub-acute toxicities of extract from Sida alba L., which can therefore be continuously used with safety in traditional medicine. Statistical studies revealed that there is a low significant difference in body and organ weights, and biological parameters between control group and the treated assay groups (p<0.01 or p<0.05).

Abstract

International audience; Lipopolysaccharide (LPS) is a key player for innate immunity activation. It is therefore a prime target for sepsis treatment, as antibiotics are not sufficient to improve outcome during septic shock. An extracorporeal removal method by polymyxin (PMX) B direct hemoperfusion (PMX-DHP) is used in Japan, but recent trials failed to show a significant lowering of circulating LPS levels after PMX-DHP therapy. PMX-DHP has a direct effect on LPS molecules. However, LPS is not present in a free form in the circulation, as it is mainly carried by lipoproteins, including LDLs. Lipoproteins are critical for physiological LPS clearance, as LPSs are carried by LDLs to the liver for elimination. We hypothesized that LDL apheresis could be an alternate method for LPS removal. First, we demonstrated in vitro that LDL apheresis microbeads are almost as efficient as PMX beads to reduce LPS concentration in LPS-spiked human plasma, whereas it is not active in PBS. We found that PMX was also adsorbing lipoproteins, although less specifically. Then, we found that endogenous LPS of patients treated by LDL apheresis for familial hypercholesterolemia is also removed during their LDL apheresis sessions, with both electrostatic-based devices and filtration devices. Finally, LPS circulating in the plasma of septic shock and severe sepsis patients with gram-negative bacteremia was also removed in vitro by LDL adsorption. Overall, these results underline the importance of lipoproteins for LPS clearance, making them a prime target to study and treat endotoxemia-related conditions.

Abstract

Background and ObjectivesKappa free light chains (KFLC) seem to efficiently diagnose MS. However, extensive cohort studies are lacking to establish consensus cut-offs, notably to rule out non-MS autoimmune CNS disorders. Our objectives were to (1) determine diagnostic performances of CSF KFLC, KFLC index, and KFLC intrathecal fraction (IF) threshold values that allow us to separate MS from different CNS disorder control populations and compare them with oligoclonal bands' (OCB) performances and (2) to identify independent factors associated with KFLC quantification in MS.MethodsWe conducted a retrospective multicenter study involving 13 French MS centers. Patients were included if they had a noninfectious and nontumoral CNS disorder, eligible data concerning CSF and serum KFLC, albumin, and OCB. Patients were classified into 4 groups according to their diagnosis: MS, clinically isolated syndrome (CIS), other inflammatory CNS disorders (OIND), and noninflammatory CNS disorder controls (NINDC).ResultsOne thousand six hundred twenty-one patients were analyzed (675 MS, 90 CIS, 297 OIND, and 559 NINDC). KFLC index and KFLC IF had similar performances in diagnosing MS from nonselected controls and OIND (p= 0.123 andp= 0.991 for area under the curve [AUC] comparisons) and performed better than CSF KFLC (p< 0.001 for all AUC comparisons). A KFLC index of 8.92 best separated MS/CIS from the entire nonselected control population, with better performances than OCB (p< 0.001 for AUC comparison). A KFLC index of 11.56 best separated MS from OIND, with similar performances than OCB (p= 0.065). In the multivariate analysis model, female gender (p= 0.003), young age (p= 0.013), and evidence of disease activity (p< 0.001) were independent factors associated with high KFLC index values in patients with MS, whereas MS phenotype, immune-modifying treatment use at sampling, and the FLC analyzer type did not influence KFLC index.DiscussionKFLC biomarkers are efficient tools to separate patients with MS from controls, even when compared with other patients with CNS autoimmune disorder. Given these results, we suggest using KFLC index or KFLC IF as a criterion to diagnose MS.Classification of EvidenceThis study provides Class III evidence that KFLC index or IF can be used to differentiate patients with MS from nonselected controls and from patients with other autoimmune CNS disorders.

Abstract

International audience; Background and aims: The identification of underlying genes of genetic conditions has expanded greatly in the past decades, which has broadened the field of genes responsible for inherited neuromuscular diseases. We aimed to investigate mutations associated with neuromuscular disorders phenotypes in 2 Moroccan families.Material and methods: Next-generation sequencing combined with Sanger sequencing could assist with under-standing the hereditary variety and underlying disease mechanisms in these disorders. Results: Two novel homozygous mutations were described in this study. The SIL1 mutation is the first identified in the Moroccan population, the mutation was identified as the main cause of Marinesco-Sjogren syndrome in one patient. While the second mutation identified in the fatty acid 2-hydroxylase gene (FA2H) was associated with the Spastic paraplegia 35 in another patient, both transmitted in an autosomal recessive pattern.Discussion and conclusions: These conditions are extremely rare in the North African population and may be underdiagnosed due to overlapping clinical characteristics and heterogeneity of these diseases. We have reported in this study mutations associated with the diseases found in the patients. In addition, we have narrowed the phenotypic spectrum, as well as the diagnostic orientation of patients with neuromuscular disorders, who might have very similar symptoms to other disease groups.

Abstract

IntroductionDuring peritonitis, lipopolysaccharides (LPS) cross the peritoneum and pass through the liver before reaching the central compartment. The aim of the present study was to investigate the role of lipoproteins and phospholipid transfer protein (PLTP) in the early stages of LPS detoxification.Material and MethodsPeritonitis was induced by intra-peritoneal injection of LPS in mice. We analyzed peritoneal fluid, portal and central blood. Lipoprotein fractions were obtained by ultracentrifugation and fast protein liquid chromatography. LPS concentration and activity were measured by liquid chromatography coupled with mass spectrometry and limulus amoebocyte lysate. Wild-type mice were compared to mice knocked out for PLTP.ResultsIn mice expressing PLTP, LPS was able to bind to HDL in the peritoneal compartment, and this was maintained in plasma from portal and central blood. A hepatic first-pass effect of HDL-bound LPS was observed in wild-type mice. LPS binding to HDL resulted in an early arrival of inactive LPS in the central blood of wild-type mice.ConclusionPLTP promotes LPS peritoneal clearance and neutralization in a model of peritonitis. This mechanism involves the early binding of LPS to lipoproteins inside the peritoneal cavity, which promotes LPS translocation through the peritoneum and its uptake by the liver.

Abstract

International audience; The typing of proteinuria is one of the complementary examinations carried out during the exploration of proteinuria. It aims to separate and identify the different proteins, or fractions of proteins, that make up proteinuria. The nature and relative importance of the proteins present reflect the location of the renal involvement and help to determine the etiology. The typing of a proteinuria also allows the detection of a monoclonal component in urine and its quantification. Finally, it allows highlighting the existence of a proteinuria of overload that can occur in the absence of kidney damage. Many methods allow the typing of proteinuria, and these have benefited in recent years from technological advances. The purpose of this review is to summarize typing methods currently used, their benefits and limitations, and the help that these diagnostic tools can provide to the management of patients.; Le typage des protéinuries constitue un des examens complémentaires mis en œuvre au cours de l’exploration d’une protéinurie. Il vise à séparer et à identifier les différentes protéines, ou fractions de protéines, qui composent la protéinurie. La nature et l’importance relative des protéines présentes reflètent la localisation de l’atteinte rénale et contribuent à en déterminer l’étiologie. Le typage d’une protéinurie permet également la détection d’un composant monoclonal urinaire ainsi que sa quantification. Enfin, elle permet de mettre en évidence l’existence d’une protéinurie de surcharge autre que les chaînes légères libres d’immunoglobulines pouvant survenir en l’absence de lésion rénale. De nombreuses méthodes permettent le typage des protéinuries, et celles-ci ont bénéficié ces dernières années d’avancées technologiques. L’objet de cette revue est de faire la synthèse des méthodes de typage actuellement utilisées, de leurs avantages et de leurs limites, et de l’aide que ces analyses peuvent apporter à la prise en charge des malades.

Abstract

International audience; Chronic stimulation by infectious pathogens or self-antigen glucosylsphingosine (GlcSph) can lead to monoclonal gammopathy of undetermined significance (MGUS) and multiple myeloma (MM). Novel assays such as the multiplex infectious antigen microarray (MIAA) and GlcSph assays, permit identification of targets for >60% purified monoclonal immunoglobulins (Igs). Searching for additional targets, we selected 28 purified monoclonal Igs whose antigen was not represented on the MIAA and GlcSph assays; their specificity of recognition was then analyzed using microarrays consisting of 3760 B-cell epitopes from 196 pathogens. The peptide sequences PALTAVETG and PALTAAETG of the VP1 coat proteins of human poliovirus 1/3 and coxsackievirus B1/B3, respectively, were specifically recognized by 6/28 monoclonal Igs. Re-analysis of patient cohorts showed that purified monoclonal Igs from 10/155 MGUS/SM (6.5%) and 3/147 MM (2.0%) bound to the PALTAVETG or PALTAAETG epitopes. Altogether, PALTAV/AETG-initiated MGUS are not rare and few seem to evolve toward myeloma.

Abstract

International audience; Diet shapes the gut microbiota which impacts hepatic lipid metabolism. Modifications in liver fat content are associated with metabolic disorders. We investigated the extent of dietary fat and fiber-induced alterations in the composition of gut microbiota and hepatic fatty acids (FAs). Mice were fed a purified low-fat diet (LFD) or high-fat diet (HFD) containing non-soluble fiber cellulose or soluble fiber inulin. HFD induced hepatic decreases in the amounts of C14:0, C16:1n-7, C18:1n-7 and increases in the amounts of C17:0, C20:0, C16:1n-9, C22:5n-3, C20:2n-6, C20:3n-6, and C22:4n-6. When incorporated in a LFD, inulin poorly affected the profile of FAs. However, when incorporated in a HFD, it (i) specifically led to an increase in the amounts of hepatic C18:0, C22:0, total polyunsaturated FAs (PUFAs), total n-6 PUFAs, C18:3n-3, and C18:2n-6, (ii) exacerbated the HFD-induced increase in the amount of C17:0, and (iii) prevented the HFD-induced increases in C16:1n-9 and C20:3n-6. Importantly, the expression/activity of some elongases and desaturases, as well as the gut microbiota composition, were impacted by the dietary fat and fiber content. To conclude, inulin modulated gut microbiota and hepatic fatty acid composition, and further investigations will determine whether a causal relationship exists between these two parameters.

Abstract

International audience; Recent studies have highlighted an important role for lysophosphatidylcholine acyltransferase 3 (LPCAT3) in controlling the PUFA composition of cell membranes in the liver and intestine. In these organs, LPCAT3 critically supports cell-membrane-associated processes such as lipid absorption or lipoprotein secretion. However, the role of LPCAT3 in macrophages remains controversial. Here, we investigated LPCAT3's role in macrophages both in vitro and in vivo in mice with atherosclerosis and obesity. To accomplish this, we used the LysMCre strategy to develop a mouse model with conditional Lpcat3 deficiency in myeloid cells (Lpcat3KOMac). We observed that partial Lpcat3 deficiency (approximately 75% reduction) in macrophages alters the PUFA composition of all phospholipid (PL) subclasses, including phosphatidylinositols and phosphatidylserines. A reduced incorporation of C20 PUFAs (mainly arachidonic acid [AA]) into PLs was associated with a redistribution of these FAs toward other cellular lipids such as cholesteryl esters. Lpcat3 deficiency had no obvious impact on macrophage inflammatory response or endoplasmic reticulum (ER) stress; however, Lpcat3KOMac macrophages exhibited a reduction in cholesterol efflux in vitro. In vivo, myeloid Lpcat3 deficiency did not affect atherosclerosis development in LDL receptor deficient mouse (Ldlr-/-) mice. Lpcat3KOMac mice on a high-fat diet displayed a mild increase in hepatic steatosis associated with alterations in several liver metabolic pathways and in liver eicosanoid composition. We conclude that alterations in AA metabolism along with myeloid Lpcat3 deficiency may secondarily affect AA homeostasis in the whole liver, leading to metabolic disorders and triglyceride accumulation.

Abstract

Introduction KRAS mutations are detected in 20% to 30% of NSCLC. However, KRAS mutation subtypes may differently influence the outcome of patients with advanced NSCLC. Methods In the Biomarkers France study, 4894 KRAS mutations (26.2%) were detected in 4634 patients from the 17,664 enrolled patients with NSCLC. Survival and treatment data on noncurative stage III to IV NSCLC were available for 901 patients. First- and second-line treatment effects on progression-free survival and overall survival were analyzed according to the KRAS mutations subtype. Results Over 95% of patients with KRAS mutation were smokers or former smokers who were white (99.5%), presenting with adenocarcinoma (82.5%). The most common KRAS mutation subtype was G12C (374 patients; 41.5%), followed by G12V (168; 18.6%), G12D (131; 14.5%), G12A (62; 6.9%), G13C (45; 5.0%), G13D (31; 3.4%), and others (10; 1%). Approximately 21% of patients had transition mutation and 68.2% had a transversion mutation. G12D and transition mutations were predominant in never-smokers. The median overall survival for patients with KRAS-mutated NSCLC was 8.1 months (95% confidence interval [CI]: 7.5–9.5), without any differences according to the different KRAS subtypes mutations. The median progression-free survival was 4.6 months (95% CI: 4.2–5.1) for first-line treatment and 4.8 months (95% CI: 4.3–6.8) for second-line treatment, without any differences according to the different KRAS subtypes mutations. Conclusions KRAS mutation subtypes influenced neither treatment responses nor outcomes. The KRAS G12C mutation was detected in 41.5% of patients, who are now eligible for potent and specific G12C inhibitors.

Abstract

International audience; Chronic stimulation by infectious or self-antigens initiates subsets of monoclonal gammopathies of undetermined significance (MGUS), smoldering multiple myeloma (SMM), or multiple myeloma (MM). Recently, glucosylsphingosine (GlcSph) was reported to be the target of one third of monoclonal immunoglobulins (Igs). In this study of 233 patients (137 MGUS, 6 SMM, 90 MM), we analyzed the GlcSph-reactivity of monoclonal Igs and non-clonal Igs. The presence of GlcSph-reactive Igs in serum was unexpectedly frequent, detected for 103/233 (44.2%) patients. However, GlcSph was targeted by the patient’s monoclonal Ig for only 37 patients (15.9%); for other patients (44 MGUS, 22 MM), the GlcSph-reactive Igs were non-clonal. Then, the characteristics of patients were examined: compared to MM with an Epstein-Barr virus EBNA-1-reactive monoclonal Ig, MM patients with a GlcSph-reactive monoclonal Ig had a mild presentation. The inflammation profiles of patients were similar except for moderately elevated levels of 4 cytokines for patients with GlcSph-reactive Igs. In summary, our study highlights the importance of analyzing clonal Igs separately from non-clonal Igs and shows that, if autoimmune responses to GlcSph are frequent in MGUS/SMM and MM, GlcSph presumably represents the initial pathogenic event for ~16% cases. Importantly, GlcSph-initiated MM appears to be a mild form of MM disease.

Abstract

International audience; Previous studies showed that monoclonal immunoglobulins G (IgGs) of “monoclonal gammopathy of undetermined significance” (MGUS) and myeloma were hyposialylated, thus presumably pro-inflammatory, and for about half of patients, the target of the monoclonal IgG was either a virus—Epstein–Barr virus (EBV), other herpes viruses, hepatitis C virus (HCV)—or a glucolipid, lysoglucosylceramide (LGL1), suggesting antigen-driven disease in these patients. In the present study, we show that monoclonal IgAs share these characteristics. We collected 35 sera of patients with a monoclonal IgA (6 MGUS, 29 myeloma), and we were able to purify 25 of the 35 monoclonal IgAs (6 MGUS, 19 myeloma). Monoclonal IgAs from MGUS and myeloma patients were significantly less sialylated than IgAs from healthy volunteers. When purified monoclonal IgAs were tested against infectious pathogens and LGL1, five myeloma patients had a monoclonal IgA that specifically recognized viral proteins: the core protein of HCV in one case, EBV nuclear antigen 1 (EBNA-1) in four cases (21.1%of IgA myeloma). Monoclonal IgAs from three myeloma patients reacted against LGL1. In summary, monoclonal IgAs are hyposialylated and as described for IgG myeloma, significant subsets (8/19, or 42%) of patients with IgA myeloma may have viral or self (LGL1) antigen-driven disease.

Abstract

T790M mutations inEGFR-mutated non-small cell lung cancer (NSCLC) account for nearly 50% of acquired resistance mechanisms to EGFR-TKIs. Earlier studies suggested that tumor T790M could also be detected in TKI-naïve EGFR-mutated NSCLC. The aim of the study is to assess the prevalence and clinical significance of quantification of tumor pre-treatment T790M subclones.We analyzed 366 EGFR-mutated NSCLC patients of the real-life IFCT Biomarkers France study with available pre-treatment formalin-fixed paraffin-embedded (FFPE) tumor DNA before treatment by first/second-generation EGFR-TKI. We used ultra-sensitive Droplet Digital Polymerase Chain Reaction (ddPCR) QX200 (BIO-RAD®, Hercules, CA, USA). All samples were tested in duplicate.ddPCR identified T790M in 19/240 specimens (8%). T790M-positive and T790M-negative populations were not different for clinical baseline characteristics. T790M Variant Allele Frequency (VAF) was 0.01% 0.1%, 0.1% 1%, 1% 10%, and10% in five (26.3%), six (31.6%), six (31.6%), and two (10.5%) patients, respectively. T790M VAF was0.1% in 11/13 (84%) patients with rapid (3 months) or usual progression (3-20 months) compared to 0/3 with low progression (20 months) (p = 0.02). In a Cox model, T790M mutation positivity was correlated with overall survival (OS) and progression-free survival (PFS) for 10%VAF1% (hazard ratio [HR] = 2.83, 95% confidence interval [CI] 1.13-7.07, p = 0.03; HR=3.62, 95%CI 1.43-4.92, p = 0.007, respectively) and for VAF10% (HR = 19.14, 95%CI 4.35-84.26, p 0.001; HR = 17.89, 95%CI 2.21-144.86, p = 0.007, respectively).Ultra-sensitive detection of tumor T790M mutation concerned 8% of EGFR-mutated TKI-naïve NSCLC patients and has a negative prognostic value only for T790M VAF over 1%.

Abstract

Immune checkpoint inhibitors (ICIs) have dramatically modified the prognosis of several advanced cancers, however many patients still do not respond to treatment. Optimal results might be obtained by targeting cancer cell metabolism to modulate the immunosuppressive tumor microenvironment. Here, we identify sphingosine kinase-1 (SK1) as a key regulator of anti-tumor immunity. Increased expression of SK1 in tumor cells is significantly associated with shorter survival in metastatic melanoma patients treated with anti-PD-1. Targeting SK1 markedly enhances the responses to ICI in murine models of melanoma, breast and colon cancer. Mechanistically, SK1 silencing decreases the expression of various immunosuppressive factors in the tumor microenvironment to limit regulatory T cell (Treg) infiltration. Accordingly, a SK1-dependent immunosuppressive signature is also observed in human melanoma biopsies. Altogether, this study identifies SK1 as a checkpoint lipid kinase that could be targeted to enhance immunotherapy., There are many patients who do not respond to immune checkpoint inhibitor (ICI) immunotherapy. Here, the authors show a significant negative correlation between sphingosine kinase-1 (SK1) expression and survival for ICI-treated melanoma patients, and further show that targeting SK1 improves response to ICI in mouse cancer models.

Abstract

Structure and function of the retina mainly rely on its fatty acid (FA) composition. Evidence from epidemiological studies and from animal experiments indicates that FA composition of the retina is influenced by the diet. Mice under chronic high-fat diet (HFD) develop metabolic syndrome, a risk factor for diabetes that is associated with structural and functional alterations of the retina. Here, we studied the impact of chronic exposure of mice to HFD on retinal FA composition. C57BL/6 J male mice were fed either a chow diet or a HFD for 11 weeks. As expected, HFD induced weight gain, adiposity, hyperglycemia and dyslipidemia. The retinal FA composition was determined by gas chromatography coupled to flame ionization detection. No significant change in the relative abundance of total saturated FAs (SFAs), total monounsaturated FAs (MUFAs) or total polyunsaturated FAs (PUFAs) was observed. However, retinas of HFD-fed mice displayed decreased amounts of C24:0 (p = 0.0231), C16:1n-7 (p < 0.0001), C18:1n-7 (p < 0.0001), C20:3n-9 (p = 0.0425) and C20:3n-6 (p = 0.0008), and an increased amount of C20:2n-6 (p < 0.0001). In addition, the ratio of linoleic acid (C18:2n-6) to alpha-linolenic acid (C18:3n-3) was increased in the retinas of HFD-fed mice (15.0 +/- 0.8 versus 11.8 +/- 0.6 in HFD and CD, respectively, p = 0.0045). No modification in the contents of arachidonic acid (C20:4n-6, AA) and docosahexaenoic acid (C22:6n-3, DHA) were observed. Analysis of dimethylacetals (DMA), which are residues of plasmalogens (Pls), revealed that the amount of Pls containing octadecanal-aldehydes (DMA C18:0) was significantly increased in HFD-fed mice (p = 0.0447). This increase was, at least in part, balanced by a decrease in Pls containing 7-octadecanal-aldehydes (DMA C18:1n-7) (p = 0.0007). In conclusion, HFD had an impact on the relative proportion of essential dietary fatty acids linoleic acid and alpha-linolenic acid that are incorporated in the retina. However, this imbalance in PUFA precursors did not alter the content of the two major retinal long-chain PUFAs, AA and DHA. HFD consumption also led to alterations in the retinal SFAs, MUFAs and Pls profiles.
