Abstract

National audience; L’utilisation des indicateurs de changements écologiques pour suivre l’état des relations populations/ environnement est un préalable à une bonne gestion des populations d'ongulés sauvages. Cependant, la mise en application d’une démarche de type gestion adaptative nécessite de respecter un certain nombre d’étapes structurantes : définition d’objectifs, synthèse des connaissances, choix de modalités de gestion mise en oeuvre, suivis de ces modalités et retour d’expériences. Cette dernière étape correspond à une analyse critique de la gestion mise en oeuvre et le cas échéant du processus de concertation ayant permis de la définir. Elle permet une adaptation progressive des pratiques. Hors, l’analyse des pratiques, à l’appui de certains projets de recherches récents (Cosson, Dysperse) montre à l’évidence des insuffisances. Parmi celles-ci, on note le plus souvent un manque de consensus sur des objectifs communs et partagés entre les différents acteurs (forestiers, chasseurs...) voire au sein d’un même groupe d’acteurs (forestiers publics/forestiers privés par exemple). Une revue des besoins sera menée au cas particulier de la Sologne où se cristallisent des points de désaccords.

Abstract

The article presents a study on the biochemical and hematological profiles of newly weaned rats submitted to experimental poisoning with T. serrulatus venom. The researchers observed 15 recently weaned male Wistar rats with a mean weight of 130 grams and used electronic counter to perform erythrocyte and leukocyte counts. They found that envenomation by T.serrulatus caused relative lymphocytosis, leukocytosis and polycythemia.

Abstract

Objective: Lactoferrin is an iron-binding protein that has been implicated in protection against infections and allogeneic recognition reactions and in the control of cell growth. We studied the biochemical characteristics and expression of the unique lactoferrin epitopes (LF(1)) in human placentas.Study Design: Immunohistologic studies of normal human term placentas were done by using monoclonal antibodies to LF(1). Double-antibody experiments were done by using monoclonal antibodies to markers of inflammation (macrophages, human leukocyte antigen [HLA-DR]). LF(1) was studied immunochemically by using lactoferrin fragments produced by the reaction of lactoferrin with trypsin or N-glycanase.Results: Anti-LF(1) monoclonal antibodies reacted with most interstitial cytotrophoblasts in the basal plate and with villous cytotrophoblasts of some but not all chorionic villi. Cytotrophoblasts expressing LF(1) were associated with large numbers of HLA-DR-reactive macrophages. Anti-LF(1) monoclonal antibodies reacted with 2 distinct tryptic fragments of lactoferrin, and these reactivities were not affected by treatment with N-glycanase.Conclusion: Placental cytotrophoblasts express unique epitopes of lactoferrin (LF(1)). Such expression is increased in the presence of activated macrophages. This expression could be an extraembryonic response to inflammation and maternal allogeneic recognition as an effort to protect trophoblastic cells. The LF(1) epitopes might represent conserved polypeptide epitopes on 2 homologous lobes of lactoferrin. [ABSTRACT FROM AUTHOR]

43. LacSwitch®II Regulation of Connexin43 cDNA Expression Enables Gap-Junction Single-Channel Analysis

Abstract

Metabolic and electrical coupling through gap junction channels is implicated in cell differentiation, tissue homeostasis, and electrotonic propagation of signals in excitable tissues. The characterization of gating properties of these channels requires electrophysiological recordings at both single- and multiple-channel levels. Hence, a system that is able to control connexin expression by external means would provide a useful tool. To regulate the expression of connexins in cells, plasmids encoding a transactivator and/or a lac-operon IPTG response-dependent Cx43 target gene were transfected into communication-deficient N2a neuroblastoma cells. Immunoblotting, dye coupling, and electrophysiological methods revealed that expression of Cx43 in selected clones could be tightly regulated. After 15–20 h of acute induction with IPTG, cell-to-cell communication reached its peak with junctional conductances of 15–30 nS. Chronic induction at specific doses of IPTG produced constant, controlled levels of Cx43 expression, which were reflected by predictable junctional coupling levels. These conditions allowed prolonged recordings from either lowly or highly coupled cells, making lac operon an ideal regulatory system for channel gating studies at a single-channel level.
