Despite many interventions, science education remains highly inequitable throughout the world. Internet-enabled experimental learning has the potential to reach underserved communities and increase the diversity of the scientific workforce. Here, we demonstrate the use of lab-on-a-chip (LoC) technologies to expose Latinx life science undergraduate students to introductory concepts of computer programming by taking advantage of open-loop cloud-integrated LoCs. We developed a context-aware curriculum to train students at over 8000 km from the experimental site. Through this curriculum, the students completed an assignment testing bacteria contamination in water using LoCs. We showed that this approach was sufficient to reduce the students' fear of programming and increase their interest in continuing careers with a computer science component. Altogether, we conclude that LoC-based internet-enabled learning can become a powerful tool to train Latinx students and increase the diversity in STEM.

The demand for rapid, high-quality, and controlled mixing at the microscale has led to the development of various types of micromixers. Micromixers are commonly categorised as active, or passive based on whether they utilise external energy to enhance mixing. Passive micromixers utilise a complex geometry to enhance the diffusion coefficient at lower Reynolds numbers and induce chaotic advection at higher Reynolds numbers for effectively mixing fluids without external energy. Active micromixers, on the other hand, achieve precise, fast, and controllable mixing by employing external energy sources such as pressure, electric, magnetic, or acoustic fields. Some active methods such as magnetic field-driven micromixers need fluids with specific properties. Others, such as acoustic field-driven micromixers apply to various types of fluids. Bubbles can be used as membranes or stirrers in microfluidic devices for both passive and active micromixers. They are easy to use, compatible with microfluidic systems, low cost, and effective. Improvements in manufacturing methods, notably, 3D printing have emerged as promising methods for the development of new micromixer designs. In this paper, a wide range of micromixer types is reviewed and the main mechanism for enhanced mixing is investigated. This study aims to guide researchers proposing innovative designs. Furthermore, it is shown that combining different methods can lead to the development of more effective micromixers, promising further advancements in microscale mixing technology. Multicellular model organisms, such as Drosophila melanogaster (fruit fly), are frequently used in a myriad of biological research studies due to their biological significance and global standardization. However, traditional tools used in these studies generally require manual handling, subjective phenotyping, and bulk treatment of the organisms, resulting in laborious experimental protocols with limited accuracy. Advancements in microtechnology over the course of the last two decades have allowed researchers to develop automated, high-throughput, and multifunctional experimental tools that enable novel experimental paradigms that would not be possible otherwise. We discuss recent advances in microtechnological systems developed for small model organisms using D. melanogaster as an example. We critically analyze the state of the field by comparing the systems produced for different applications. Additionally, we suggest design guidelines, operational tips, and new research directions based on the technical and knowledge gaps in the literature. This review aims to foster interdisciplinary work by helping engineers to familiarize themselves with model organisms while presenting the most recent advances in microengineering strategies to biologists. Detecting foodborne contamination is a critical challenge in ensuring food safety and preventing human suffering and economic losses. Contaminated food, comprising biological agents (e.g. bacteria, viruses and fungi) and chemicals (e.g. toxins, allergens, antibiotics and heavy metals), poses significant risks to public health. Microfluidic technology has emerged as a transformative solution, revolutionizing the detection of contaminants with precise and efficient methodologies. By manipulating minute volumes of fluid on miniaturized systems, microfluidics enables the creation of portable chips for biosensing applications. Advancements from early glass and silicon devices to modern polymers and cellulose‐based chips have significantly enhanced microfluidic technology, offering adaptability, flexibility, cost‐effectiveness and biocompatibility. Microfluidic systems integrate seamlessly with various biosensing reactions, facilitating nucleic acid amplification, target analyte recognition and accurate signal readouts. As research progresses, microfluidic technology is poised to play a pivotal role in addressing evolving challenges in the detection of foodborne contaminants. In this short review, we delve into various manufacturing materials for state‐of‐the‐art microfluidic devices, including inorganics, elastomers, thermoplastics and paper. Additionally, we examine several applications where microfluidic technology offers unique advantages in the detection of food contaminants, including bacteria, viruses, fungi, allergens and more. Cancer continues to be a significant global health issue with one in six deaths linked to the disease despite advancements in cancer detection and treatment. Recently, Helicobacter pylori (H. pylori) was identified as a risk factor for cancer development. This gram-negative bacterium is associated with gastric conditions, including stomach cancer. Although the exact transmission methods of this bacterium are still unclear, studies suggest that waterborne transmission is possible. This study focuses on the development of a colorimetric nanomaterial-based paper biosensor for specific H. pylori detection using H. pylori extracellular proteases as biomarkers. The biosensor utilizes a unique substrate labeled with magnetic nanobeads and bound to a gold sensing platform. The biosensor's limit of detection (LOD) of 100 CFU/mL, selectivity, stability, and ability to detect H. pylori in clinical specimens were evaluated, demonstrating promising results in terms of sensitivity and specificity. In comparison to traditional methods, this biosensor offers advantages in simplicity and ease of use, making it appropriate for on-site detection in both environmental and clinical settings.

Microgripper is a small‐miniaturized robotic device employed to maneuver sub‐millimeter‐sized particles or objects that are too small to be manipulated by human hands or conventional tools. The design and fabrication of the microgripper pose challenges due to the integration of actuation components into their structure. To address this challenge, the authors in this work presented a novel magnetic microgripper wherein the functions of the actuation components are executed by the two pairs of microgripper arms made up of magnetic material. A custom‐made electromagnetic system consisting of eight coils is employed to externally control these arms, enabling precise execution of gripping up to a maximum of two microparticles per working cycle. By varying the magnetic field strength from 20 to 120 mT, a wide range of gripper‐arms opening distance is achieved, spanning from 0.3 to 1.3 mm. Consequently, the microgripper is demonstrated to grip different sizes of microparticles ranging from 0.3 to 1.2 mm. As part of the motion control capabilities, the microgripper is demonstrated to orient and navigate across a 360‐degree range within the microfluidic environment. The development of optical and photonic applications using soft-matter droplets holds great scientific and application importance. The machining of droplet structures is expected to drive breakthroughs in advancing frontier applications. This review highlights recent advancements in micro–nanofabrication techniques for soft-matter droplets, encompassing microfluidics, laser injection, and microfluidic 3D printing. The principles, advantages, and weaknesses of these technologies are thoroughly discussed. The review introduces the utilization of a phase separation strategy in microfluidics to assemble complex emulsion droplets and control droplet geometries by adjusting interfacial tension. Additionally, laser injection can take full advantage of the self-assembly properties of soft matter to control the spontaneous organization of internal substructures within droplets, thus providing the possibility of high-precision customized assembly of droplets. Microfluidic 3D printing demonstrates a 3D printing-based method for machining droplet structures. Its programmable nature holds promise for developing device-level applications utilizing droplet arrays. Finally, the review presents novel applications of soft-matter droplets in optics and photonics. The integration of processing concepts from microfluidics, laser micro–nano-machining, and 3D printing into droplet processing, combined with the self-assembly properties of soft materials, may offer novel opportunities for processing and application development. This article has tried to provide an overview of the most sophisticated microfluidic biosensors for identifying nucleic acids and proteins at the site of treatment. Microfluidics, which also automates sample preparation and reduces processing time and reagent consumption, enables the analysis of small sample quantities. Microfluidics and biosensor technologies collaborate to provide diagnostics at the point of care with high throughput analysis, portability, and disposability. The high sensitivity and selectivity requirements, false response errors, and integration with other essential modules are some of the challenges posed by this merger. The broad categories of protein-based and DNA-based biosensor technology are covered in this review. Also, recent advancements in coupling the biosensors to microfluidics, the main challenges and potential solutions in deploying microfluidic biosensors for point-of-care diagnostics, and the most recent developments in these areas have been discussed.

The strength of cell adhesion is important in understanding the cell's health and in culturing them. Quantitative measurement of cell adhesion strength is a significant challenge in bioengineering research. For this, the present study describes a system that can measure cell adhesion strength using acoustic streaming induced by Lamb waves. Cells are cultured on an ultrasound transducer using a range of preculture and incubation times with phosphate-buffered saline (PBS) just before the measurement. Acoustic streaming is then induced using several Lamb wave intensities, exposing the cells to shear flows and eventually detaching them. By relying upon a median detachment rate of 50 %, the corresponding detachment force, or force of cell adhesion, was determined to be on the order of several nN, consistent with previous reports. The stronger the induced shear flow, the more cells were detached. Further, we employed a preculture time of 8 to 24 h and a PBS incubation time of 0 to 60 min, producing cell adhesion forces that varied from 1.2 to 13 nN. Hence, the developed system can quantify cell adhesion strength over a wide range, possibly offering a fundamental tool for cell-based bioengineering.

The Controlled transport of tiny particles in a microfluidic environment has attracted the attention of numerous researchers in the field of lab-on-a-chip. In this work, for the first time, a fully operational microfluidic chip composed of asymmetric magnetic tracks that unidirectionally transport multiple magnetic particles synced with a general tri-axial magnetic field is proposed. In this innovative chip, the particle motion is analogous to the electron transport in electrical diodes, with similar controllability and automation levels not seen in other single-particle manipulation systems. The vertical bias component of the magnetic field by providing a repulsive force between the particles and preventing undesired cluster formation, makes the proposed chip even more similar to the electrical circuits. Additionally, the chip functions as a highly sensitive biosensor capable of detecting extremely low levels of DNA fragments using ligand-functionalized magnetic beads. The uniqueness of the proposed sensor lies in the introduction of a novel particle/analyte concentrator based on the proposed diodes, which enhances its detection sensitivity. This sensitivity is even further enhanced by a single-particle and pair detection image processing code. Furthermore, the background noise is reduced by eliminating the unwanted bead cluster formation commonly observed in previous works. The proposed device serves as a high-throughput unidirectional transport system at the single-particle resolution, offering sensitive bio-detection with many applications in biomedicine. The incorporation of biomacromolecules onto silicon waveguiding microstructures constitutes a growing trend that pushes towards compact and miniaturized biosensing systems. This paper presents the integration of one-dimensional periodic nanostructures of proteins on the surface of micrometric silicon waveguides for transducing binding events between biomacromolecules. The study demonstrates this new bioanalytical principle by experimental results and theoretical calculations, and proves that rib waveguides (1-–1.6-µm width) together with protein gratings (495-–515-nm period) display suitable spectral responses for this optical biosensing system. Protein assemblies of bovine serum albumin are fabricated on the surface of silicon nitride waveguides, characterized by electron microscopy, and their response is measured by optical frequency domain reflectometry along the fabrication process and the subsequent stages of the biorecognition assays. Detection and quantification limits of 0.3 and 3.7 µg·mL−1, respectively, of specific antibodies are inferred from experimental dose–response curves. Statistical analysis of the properties of single microparticles, such as cells, bacteria or plastic slivers, has attracted increasing interest in recent years. In this regard, field flow cytometry is considered the gold standard technique, but commercially available instruments are bulky, expensive, and not suitable for use in point-of-care (PoC) testing. Microfluidic flow cytometers, on the other hand, are small, cheap and can be used for on-site analyses. However, in order to detect small particles, they require complex geometries and the aid of external optical components. To overcome these limitations, here, we present an opto-fluidic flow cytometer with an integrated 3D in-plane spherical mirror for enhanced optical signal collection. As a result, the signal-to-noise ratio is increased by a factor of six, enabling the detection of particle sizes down to 1.5 µm. The proposed optofluidic detection scheme enables the simultaneous collection of particle fluorescence and scattering using a single optical fiber, which is crucial to easily distinguishing particle populations with different optical properties. The devices have been fully characterized using fluorescent polystyrene beads of different sizes. This work proposes a multi-objective polydimethylsiloxane (PDMS) micro-optofluidic (MoF) device suitably designed and manufactured through a 3D-printed-based master–slave approach. It exploits optical detection techniques to characterize immiscible fluids or microparticles in suspension inside a compartment specifically designed at the core of the device referred to as the MoF chamber. In addition, we show our novel, fast, and cost-effective methodology, dual-slit particle signal velocimetry (DPSV), for fluids and microparticle velocity detection. Different from the standard state-of-the-art approaches, the methodology focuses on signal processing rather than image processing. This alternative has several advantages, including the ability to circumvent the requirement of complex and extensive setups and cost reduction. Additionally, its rapid processing speed allows for real-time sample manipulations in ongoing image-based analyses. For our specific design, optical signals have been detected from the micro-optics components placed in two slots designed ad hoc in the device. To show the devices' multipurpose capabilities, the device has been tested with fluids of various colors and densities and the inclusion of synthetic microparticles. Additionally, several experiments have been conducted to prove the effectiveness of the DPSV approach in estimating microparticle velocities. A digital particle image velocimetry (DPIV)-based approach has been used as a baseline against which the outcomes of our methods have been evaluated. The combination of the suitability of the micro-optical components for integration, along with the MoF chamber device and the DPSV approach, demonstrates a proof of concept towards the challenge of real-time total-on-chip analysis. A fabrication strategy for high-efficiency passive three-dimensional microfluidic valves with no mechanical parts fabricated in silicon and glass substrates is presented. 3D diffuser-nozzle valve structures were produced and characterized in their added value in comparison to conventional diffuser-nozzle valve designs with rectangular cross sections. A grayscale lithography approach for 3D photoresist structuring combined with a proportional transfer by reactive ion etching allowed to transfer 3D resist valve designs with high precision into the targeted substrate material. The efficiency with respect to the rectification characteristics or so-called diodicity of the studied valve designs is defined as the ratio of the pressure drops in backward and forward flow directions. The studied valve designs were characterized experimentally as well as numerically based on finite element simulations with overall matching results that demonstrate a significantly improved flow rectification of the 3D valves compared to the corresponding conventional structure. Our novel 3D valve structures show, for instance, even without systematic optimization a measured diodicity of up to 1.5 at low flow rates of only about 10 μl/s. [ABSTRACT FROM AUTHOR]

The presence of heavy metal ions in soil, air and water constitutes an important global environmental threat, as these ions accumulate throughout the food chain, contributing to the rise of chronic diseases, including, amongst others, cancer and kidney failure. To date, many efforts have been made for their detection, but there is still a need for the development of sensitive, low-cost, and portable devices able to conduct on-site detection of heavy metal ions. In this work, we combine microfluidic technology and electrochemical sensing in a plastic chip for the selective detection of heavy metal ions utilizing DNAzymes immobilized in between platinum nanoparticles (PtNPs), demonstrating a reliable portable solution for water pollution monitoring. For the realization of the microfluidic-based heavy metal ion detection device, a fast and easy-to-implement fabrication method based on the photolithography of dry photosensitive layers is proposed. Oil production is a complex process that can be made more efficient by applying gas enhanced oil recovery (EOR) methods. Thus, it is essential to know the minimum miscibility pressure (MMP) and minimum miscibility enrichment (MME) of gas in oil. Conventional slim-tube experiments for the measurement of MMP require hundreds of millilitres of real or recombined oil and last over 30 days. Advances in microfluidic technology allow the reduction of the amount of fluid and the time required in determining MMP (or MME), hence making the process rapid. In this study, we developed a microfluidic model with a stochastically distributed pore network, porosity of 74.6% and volume of 83.26 nanolitres. Although the volume was six orders of magnitude smaller than the slim tube, it retained the same proportions, guaranteeing a proper comparison between the tests. This microfluidic chip allowed the study of the MMP of n-decane with carbon dioxide at two different temperature conditions. The experimental results coincided with the results received both from conventional and microfluidic experiments. Furthermore, a numerical simulation of a section of the microfluidic model under the experimental conditions presented results within acceptable margins of the experimental ones. The results of the presented methodology indicate the potential to replace conventional technology for the measurement of MMP with microfluidic technology. Immunoassays, which use antigen–antibody reactions, are the primary techniques used to selectively quantify specific disease markers in blood. Conventional immunoassays, such as the microplate-based enzyme-linked immunosorbent assay (ELISA) and paper-based immunochromatography, are widely used, but they have advantages and disadvantages in terms of sensitivity and operating time. Therefore, in recent years, microfluidic-chip-based immunoassay devices with high sensitivity, rapidity and simplicity, which are compatible with whole blood assays and multiplex assays, have been actively investigated. In this study, we developed a microfluidic device using gelatin methacryloyl (GelMA) hydrogel to form a wall-like structure in a microfluidic channel and perform immunoassays inside the wall-like structure, which can be used for rapid and highly sensitive multiplex assays with extremely small sample amounts of ~1 μL. The characteristics of GelMA hydrogel, such as swelling rate, optical absorption and fluorescence spectra, and morphology, were carefully studied to adapt the iImmunowall device and immunoassay. Using this device, a quantitative analysis of interleukin-4 (IL-4), a biomarker of chronic inflammatory diseases, was performed and a limit of detection (LOD) of 0.98 ng/mL was achieved with only 1 μL sample and 25 min incubation time. The superior optical transparency over a wide range of wavelengths and lack of autofluorescence will help to expand the application field of the iImmunowall device, such as to a simultaneous multiple assay in a single microfluidic channel, and provide a fast and cost-effective immunoassay method. Lab-on-a-chip has recently become an alternative for in situ monitoring for its portability and simple integration with an electrochemical immunoassay. Here, we present an electrochemical cell-on-a-chip configured in a three-electrode system to detect benzo(a)pyrene (BaP) in water. 11-Mercaptoundecanoic acid (MUA), a self-assembled monolayer (SAM), was used to modify a gold chip surface to reduce the randomness of antibody binding. A carboxylic acid group was activated with -ethyl-3-(3-dimethylaminopropyl) (EDC) in combination with N-hyrodsuccinimide (NHS) before antibody immobilisation. The mechanism of the electrochemical reactions on a gold surface and SAM formation were investigated by cyclic voltammetry and contact angle measurements. The data revealed a lower contact angle in the modified chip and a scan rate of 50 mV/s. Through the addition of modification layers and thiol end groups to the SAM, our design allowed the chip surface to became more insulated. All were tested by amperometric detection using the developed Q-sense system. This novel technique detected multiple samples, and completed the analysis reasonably quickly. While the integrated system proved successful in a lab setting, the aim of the research is to use this system for in situ analysis, which can be brought into a water environment to carry out tests with existing processes. In this way, any issues that may arise from an environmental setting can be rectified in an efficient manner.

Food quality and safety are important to protect consumers from foodborne illnesses. Currently, laboratory scale analysis, which takes several days to complete, is the main way to ensure the absence of pathogenic microorganisms in a wide range of food products. However, new methods such as PCR, ELISA, or even accelerated plate culture tests have been proposed for the rapid detection of pathogens. Lab-on-chip (LOC) devices and microfluidics are miniaturized devices that can enable faster, easier, and at the point of interest analysis. Nowadays, methods such as PCR are often coupled with microfluidics, providing new LOC devices that can replace or complement the standard methods by offering highly sensitive, fast, and on-site analysis. This review's objective is to present an overview of recent advances in LOCs used for the identification of the most prevalent foodborne and waterborne pathogens that put consumer health at risk. In particular, the paper is organized as follows: first, we discuss the main fabrication methods of microfluidics as well as the most popular materials used, and then we present recent literature examples for LOCs used for the detection of pathogenic bacteria found in water and other food samples. Lab-on-a-chip (LOC) systems are miniaturized devices aimed to perform one or several analyses, normally carried out in a laboratory setting, on a single chip. LOC systems have a wide application range, including diagnosis and clinical biochemistry. In a clinical setting, LOC systems can be associated with the Point-of-Care Testing (POCT) definition. POCT circumvents several steps in central laboratory testing, including specimen transportation and processing, resulting in a faster turnaround time. Provider access to rapid test results allows for prompt medical decision making, which can lead to improved patient outcomes, operational efficiencies, patient satisfaction, and even cost savings. These features are particularly attractive for healthcare settings dealing with complicated patients, such as those affected by chronic kidney disease (CKD). CKD is a pathological condition characterized by progressive and irreversible structural or functional kidney impairment lasting for more than three months. The disease displays an unavoidable tendency to progress to End Stage Renal Disease (ESRD), thus requiring renal replacement therapy, usually dialysis, and transplant. Cardiovascular disease (CVD) is the major cause of death in CKD, with a cardiovascular risk ten times higher in these patients than the rate observed in healthy subjects. The gradual decline of the kidney leads to the accumulation of uremic solutes, with negative effect on organs, especially on the cardiovascular system. The possibility to monitor CKD patients by using non-invasive and low-cost approaches could give advantages both to the patient outcome and sanitary costs. Despite their numerous advantages, POCT application in CKD management is not very common, even if a number of devices aimed at monitoring the CKD have been demonstrated worldwide at the lab scale by basic studies (low Technology Readiness Level, TRL). The reasons are related to both technological and clinical aspects. In this review, the main technologies for the design of LOCs are reported, as well as the available POCT devices for CKD monitoring, with a special focus on the most recent reliable applications in this field. Electrochemical microfluidic technology has been extensively studied in many fields due to its significant advantages. Despite the increasing number of publications on the subject, there are no studies based on bibliometric analysis in this area. In this study, we performed a bibliometric analysis of 457 publications related to electrochemical microfluidic technology during 2012–2021 through the Web of Science core database. Results show that the hot topics include the development of label-free immunosensors, biosensors, microfluidic device performance, and low cost of equipment, and the major contributors to these publications are from China, USA, Spain, India, and Germany. In addition, applications of electrochemical microfluidics in biomedicine, food safety, and environmental monitoring are summarized and analyzed. Finally, the future challenges as well as strategies for future research are discussed. Paper-based microfluidic systems have emerged as one of the most promising technologies for developing point-of-care diagnostic platforms (POCT) for detecting and monitoring various diseases. Saliva is a non-invasive biofluid easily collected, transported, and stored. Due to its accessibility and connection to systemic diseases, saliva is one of the best candidates for medical advancement at the point of care, where people can easily monitor their health. However, saliva is a complex mixture of DNA, RNA, proteins, exosomes, and electrolytes. Thus, nucleic acid separation from the salivary components is essential for PCR applications. Paper membranes are a highly porous and foldable structure capable of transporting fluids without pumps and sophisticated systems. The current work presents an insight into simulations for nucleic acid extraction on three types of porous paper membranes for use in point-of-care devices. The flow fluid model is solved on a COMSOL Multiphysics 5.3 free version platform, and the results are compared with experimental assays. The results show that pore uniformity, wet strength, porosity, and functional groups of MF1™ and Fusion 5™ paper membranes are vital parameters affecting nucleic acid extraction and PCR amplification efficiency. Electronic communication in natural systems makes use, inter alia, of molecular transmission, where electron transfer occurs within networks of redox reactions, which play a vital role in many physiological systems. In view of the limited understanding of redox signaling, we developed an approach and an electrochemical–optical lab-on-a-chip to observe cellular responses in localized redox environments. The developed fluidic micro-system uses electrogenetic bacteria in which a cellular response is activated to electrically and chemically induced stimulations. Specifically, controlled environments for the cells are created by using microelectrodes to generate spatiotemporal redox gradients. The in-situ cellular responses at both single-cell and population levels are monitored by optical microscopy. The elicited electrogenetic fluorescence intensities after 210 min in response to electrochemical and chemical activation were 1.3 × 108±0.30 × 108 arbitrary units (A.U.) and 1.2 × 108±0.30 × 108 A.U. per cell population, respectively, and 1.05 ± 0.01 A.U. and 1.05 ± 0.01 A.U. per-cell, respectively. We demonstrated that redox molecules' mass transfer between the electrode and cells – and not the applied electrical field – activated the electrogenetic cells. Specifically, we found an oriented amplified electrogenetic response on the charged electrodes' downstream side, which was determined by the location of the stimulating electrodes and the flow profile. We then focused on the cellular responses and observed distinct subpopulations that were attributed to electrochemical rather than chemical stimulation, with the distance between the cells and the stimulating electrode being the main determinant. These observations provide a comprehensive understanding of the mechanisms by which diffusible redox mediators serve as electron shuttles, imposing context and activating electrogenetic responses. This work proposes a multi-objective polydimethylsiloxane (PDMS) micro-optofluidic (MoF) device suitably designed and manufactured through a 3D-printed-based master–slave approach. It exploits optical detection techniques to characterize immiscible fluids or microparticles in suspension inside a compartment specifically designed at the core of the device referred to as the MoF chamber. In addition, we show our novel, fast, and cost-effective methodology, dual-slit particle signal velocimetry (DPSV), for fluids and microparticle velocity detection. Different from the standard state-of-the-art approaches, the methodology focuses on signal processing rather than image processing. This alternative has several advantages, including the ability to circumvent the requirement of complex and extensive setups and cost reduction. Additionally, its rapid processing speed allows for real-time sample manipulations in ongoing image-based analyses. For our specific design, optical signals have been detected from the micro-optics components placed in two slots designed ad hoc in the device. To show the devices’ multipurpose capabilities, the device has been tested with fluids of various colors and densities and the inclusion of synthetic microparticles. Additionally, several experiments have been conducted to prove the effectiveness of the DPSV approach in estimating microparticle velocities. A digital particle image velocimetry (DPIV)-based approach has been used as a baseline against which the outcomes of our methods have been evaluated. The combination of the suitability of the micro-optical components for integration, along with the MoF chamber device and the DPSV approach, demonstrates a proof of concept towards the challenge of real-time total-on-chip analysis.

Statistical analysis of the properties of single microparticles, such as cells, bacteria or plastic slivers, has attracted increasing interest in recent years. In this regard, field flow cytometry is considered the gold standard technique, but commercially available instruments are bulky, expensive, and not suitable for use in point-of-care (PoC) testing. Microfluidic flow cytometers, on the other hand, are small, cheap and can be used for on-site analyses. However, in order to detect small particles, they require complex geometries and the aid of external optical components. To overcome these limitations, here, we present an opto-fluidic flow cytometer with an integrated 3D in-plane spherical mirror for enhanced optical signal collection. As a result, the signal-to-noise ratio is increased by a factor of six, enabling the detection of particle sizes down to 1.5 µm. The proposed optofluidic detection scheme enables the simultaneous collection of particle fluorescence and scattering using a single optical fiber, which is crucial to easily distinguishing particle populations with different optical properties. The devices have been fully characterized using fluorescent polystyrene beads of different sizes. As a proof of concept for potential real-world applications, signals from fluorescent HEK cells and Escherichia coli bacteria were analyzed.

Nature consists of numerous solutions to overcome challenges in designing artificial systems. Various actuation mechanisms have been implemented in microrobots to mimic the motion of microorganisms. Such bio-inspired designs have contributed immensely to microscale developments. Among the actuation mechanisms, magnetic actuation is widely used in bio-inspired microrobotic systems and related propulsion mechanisms used by microrobots to navigate inside a magnetic field and are presented in this review. In addition, the considered robots are in microscale, and they can swim inside a fluidic environment with a low Reynolds number. In relation to microrobotics, mimicry of bacteria flagella, sperm flagella, cilia, and fish are significant. Due to the fact that these biological matters consist of different propulsion mechanisms, the effect of various parameters was investigated in the last decade and the review presents a summary that enhances understanding of the working principle of propulsion mechanisms. In addition, the effect of different parameters on the various speeds of the existing microrobots was analyzed to identify their trends. So, the swimming speeds of the microrobots show an upward trend with increasing body length, frequency, magnetic flux density, and helix angle. Microfabrication techniques play a significant role in the microscale because the device designs are highly dependent on the availability of the techniques. The presented microrobots were manufactured by 3D/4D photolithography and rapid prototyping techniques. Proper materials enable effective fabrication of microrobots using the mentioned techniques. Therefore, magnetically active material types, matrix materials, biocompatible and biodegradable materials are presented in this study. Utilizing biocompatible and biodegradable materials avoids adverse effects to the organs that could occur otherwise. In addition, magnetic field generation is significant for the propulsion of such microrobots. Rapid bacterial identification (ID) and antibiotic susceptibility testing (AST) are in great demand due to the rise of drug-resistant bacteria. Conventional culture-based AST methods suffer from a long turnaround time. By necessity, physicians often have to treat patients empirically with antibiotics, which has led to an inappropriate use of antibiotics, an elevated mortality rate and healthcare costs, and antibiotic resistance. Recent advances in miniaturization and automation provide promising solutions for rapid bacterial ID/AST profiling, which will potentially make a significant impact in the clinical management of infectious diseases and antibiotic stewardship in the coming years. In this review, we summarize and analyze representative emerging micro- and nanotechnologies, as well as automated systems for bacterial ID/AST, including both phenotypic (e.g., microfluidic-based bacterial culture, and digital imaging of single cells) and molecular (e.g., multiplex PCR, hybridization probes, nanoparticles, synthetic biology tools, mass spectrometry, and sequencing technologies) methods. We also discuss representative point-of-care (POC) systems that integrate sample processing, fluid handling, and detection for rapid bacterial ID/AST. Finally, we highlight major remaining challenges and discuss potential future endeavors toward improving clinical outcomes with rapid bacterial ID/AST technologies.

This paper discusses an active droplet generation system, and the presented droplet generator successfully performs droplet generation using two fluid phases: continuous phase fluid and dispersed phase fluid. The performance of an active droplet generation system is analysed based on the droplet morphology using vision sensing and digital image processing. The proposed system in the study includes a droplet generator, camera module with image pre-processing and identification algorithm, and controller and control algorithm with a workstation computer. The overall system is able to control, sense, and analyse the generation of droplets. The main controller consists of a microcontroller, motor controller, voltage regulator, and power supply. Among the morphological features of droplets, the diameter is extracted from the images to observe the system performance. The MATLAB-based image processing algorithm consists of image acquisition, image enhancement, droplet identification, feature extraction, and analysis. RGB band filtering, thresholding, and opening are used in image pre-processing. After the image enhancement, droplet identification is performed by tracing the boundary of the droplets. The average droplet diameter varied from ~3.05 mm to ~4.04 mm in the experiments, and the average droplet diameter decrement presented a relationship of a second-order polynomial with the droplet generation time. We are beginning a new era of Smart Diagnostics—integrated biosensors powered by recent innovations in embedded electronics, cloud computing, and artificial intelligence (AI). Universal and AI-based in vitro diagnostics (IVDs) have the potential to exponentially improve healthcare decision making in the coming years. This perspective covers current trends and challenges in translating Smart Diagnostics. We identify essential elements of Smart Diagnostics platforms through the lens of a clinically validated platform for digitizing biology and its ability to learn disease signatures. This platform for biochemical analyses uses a compact instrument to perform multiclass and multiplex measurements using fully integrated microfluidic cartridges compatible with the point of care. Image analysis digitizes biology by transforming fluorescence signals into inputs for learning disease/health signatures. The result is an intuitive Score reported to the patients and/or providers. This AI-linked universal diagnostic system has been validated through a series of large clinical studies and used to identify signatures for early disease detection and disease severity in several applications, including cardiovascular diseases, COVID-19, and oral cancer. The utility of this Smart Diagnostics platform may extend to multiple cell-based oncology tests via cross-reactive biomarkers spanning oral, colorectal, lung, bladder, esophageal, and cervical cancers, and is well-positioned to improve patient care, management, and outcomes through deployment of this resilient and scalable technology. Current study proposes a novel mechanism for micro-scale droplet generation. To this end, a new micro pump consisting of an expansion chamber with two nozzles on its left and right sides is introduced, which is capable of creating a unidirectional ow. In the current study, for the first time, thermo-viscose expansion method is used to actuate the ow by assuming that the uid is heated using a laser beam source, leading to expansion and movement of the uid. Two-phase ow analysis using the Volume Of Fraction (VOF) method shows that the proposed micro pump can consistently generate droplets with uniform and micro-scale dimensions. [ABSTRACT FROM AUTHOR]

Tissue engineering research fundamentally relies on experiments to advance knowledge, utilizing various models for research on both humans and animals. With scientific progress, experimental models have become increasingly complex over time. This complexity sometimes blurs the distinction between categories, making terminology less consistent. In biomedical research, three overarching terms are commonly used to characterize experimental environments: in vitro , ex vivo , and in vivo . While in vitro translates from Latin as "in glass," referring historically to experimental conditions in a test tube or petri dish, in vivo experiments occur within a living organism's natural environment. Conversely, ex vivo originates from living tissue outside its host environment while striving to maintain conditions as close to the host surroundings as possible. In the tissue engineering and regenerative medicine (TE&RM) community, there needs to be more clarity between in vitro and ex vivo terminology, with historical definitions sometimes disregarded and new terms often introduced without rigorous scientific justification. At this juncture, the question arises of when to refer to experiments as in vitro or ex vivo or whether the terms may be used synonymously in some instances. Moreover, what criteria must ex vivo experiments meet to be legitimately defined as such? This perspective is intended to address questions that would assist the TE&RM community in better understanding the differences between in vitro and ex vivo models. Impact Statement In the tissue engineering & regenerative medicine literature, the terms "in vitro" and "ex vivo" are often used interchangeably to describe experiments. This interchangeable usage can lead to a compromised interpretation of research results and, consequently, misleading scientific conclusions and teachings. This perspective aims to provide clarity on the various definitions of experimental designs. It also highlights the issue of using terms with inconsistent meanings that have origins dating back to the distant past. It's important to note that scientific definitions constantly evolve, and there is a scientifically rooted responsibility to evaluate and rethink the current state of affairs critically.

High-risk human papillomavirus (HPV) screening is crucial for cervical cancer prevention. However, laboratory-based nucleic acid amplification tests (NAATs) require costly equipment, designated lab space, and skilled personnel. Additionally, cervical swabs collected by healthcare professionals can be inconvenient, uncomfortable, and reduce privacy, limiting broader application and patient compliance. A SlipChip-based Integrated Point-of-Care (SIPOC) system featuring an injection-molded SlipChip is presented with preloaded reagents for nucleic acid extraction and a portable four-channel real-time quantitative PCR instrument for detection. This system incorporates a self-sampling method that allows participants to collect their own vaginal swabs, with the β-Globin gene as a control. After testing 130 participants for HPV-16 and HPV-18, 97.7% of the self-collected samples are valid. Among valid samples, 25 tested positive for HPV-16 and 9 for HPV-18. Compared to Roche's standard HPV PCR test, the SIPOC system shows 100% positive predictive value (PPV) for both HPV-16 and HPV-18 and negative predictive values (NPVs) of 99.0% and 99.1%, respectively. The strength of cell adhesion is important in understanding the cell’s health and in culturing them. Quantitative measurement of cell adhesion strength is a significant challenge in bioengineering research. For this, the present study describes a system that can measure cell adhesion strength using acoustic streaming induced by Lamb waves. Cells are cultured on an ultrasound transducer using a range of preculture and incubation times with phosphate-buffered saline (PBS) just before the measurement. Acoustic streaming is then induced using several Lamb wave intensities, exposing the cells to shear flows and eventually detaching them. By relying upon a median detachment rate of 50 %, the corresponding detachment force, or force of cell adhesion, was determined to be on the order of several nN, consistent with previous reports. The stronger the induced shear flow, the more cells were detached. Further, we employed a preculture time of 8 to 24 h and a PBS incubation time of 0 to 60 min, producing cell adhesion forces that varied from 1.2 to 13 nN. Hence, the developed system can quantify cell adhesion strength over a wide range, possibly offering a fundamental tool for cell-based bioengineering.

The presence of heavy metal ions in soil, air and water constitutes an important global environmental threat, as these ions accumulate throughout the food chain, contributing to the rise of chronic diseases, including, amongst others, cancer and kidney failure. To date, many efforts have been made for their detection, but there is still a need for the development of sensitive, low-cost, and portable devices able to conduct on-site detection of heavy metal ions. In this work, we combine microfluidic technology and electrochemical sensing in a plastic chip for the selective detection of heavy metal ions utilizing DNAzymes immobilized in between platinum nanoparticles (PtNPs), demonstrating a reliable portable solution for water pollution monitoring. For the realization of the microfluidic-based heavy metal ion detection device, a fast and easy-to-implement fabrication method based on the photolithography of dry photosensitive layers is proposed. As a proof of concept, we demonstrate the detection of Pb2+ ions using the prototype microfluidic device.

Oil production is a complex process that can be made more efficient by applying gas enhanced oil recovery (EOR) methods. Thus, it is essential to know the minimum miscibility pressure (MMP) and minimum miscibility enrichment (MME) of gas in oil. Conventional slim-tube experiments for the measurement of MMP require hundreds of millilitres of real or recombined oil and last over 30 days. Advances in microfluidic technology allow the reduction of the amount of fluid and the time required in determining MMP (or MME), hence making the process rapid. In this study, we developed a microfluidic model with a stochastically distributed pore network, porosity of 74.6% and volume of 83.26 nanolitres. Although the volume was six orders of magnitude smaller than the slim tube, it retained the same proportions, guaranteeing a proper comparison between the tests. This microfluidic chip allowed the study of the MMP of n-decane with carbon dioxide at two different temperature conditions. The experimental results coincided with the results received both from conventional and microfluidic experiments. Furthermore, a numerical simulation of a section of the microfluidic model under the experimental conditions presented results within acceptable margins of the experimental ones. The results of the presented methodology indicate the potential to replace conventional technology for the measurement of MMP with microfluidic technology. Its promise lies in accelerating laboratory tests and increasing the reliability of experimental results and, subsequently, the quality of field gas EOR operations.

Immunoassays, which use antigen–antibody reactions, are the primary techniques used to selectively quantify specific disease markers in blood. Conventional immunoassays, such as the microplate-based enzyme-linked immunosorbent assay (ELISA) and paper-based immunochromatography, are widely used, but they have advantages and disadvantages in terms of sensitivity and operating time. Therefore, in recent years, microfluidic-chip-based immunoassay devices with high sensitivity, rapidity and simplicity, which are compatible with whole blood assays and multiplex assays, have been actively investigated. In this study, we developed a microfluidic device using gelatin methacryloyl (GelMA) hydrogel to form a wall-like structure in a microfluidic channel and perform immunoassays inside the wall-like structure, which can be used for rapid and highly sensitive multiplex assays with extremely small sample amounts of ~1 μL. The characteristics of GelMA hydrogel, such as swelling rate, optical absorption and fluorescence spectra, and morphology, were carefully studied to adapt the iImmunowall device and immunoassay. Using this device, a quantitative analysis of interleukin-4 (IL-4), a biomarker of chronic inflammatory diseases, was performed and a limit of detection (LOD) of 0.98 ng/mL was achieved with only 1 μL sample and 25 min incubation time. The superior optical transparency over a wide range of wavelengths and lack of autofluorescence will help to expand the application field of the iImmunowall device, such as to a simultaneous multiple assay in a single microfluidic channel, and provide a fast and cost-effective immunoassay method.

Micromixers are widely used in lab-on-a-chip devices for analytical chemistry, bioengineering, and biomedicine to achieve rapid mixing and analysis of samples. However, the existing micromixers are mostly two-dimensional structures with low mixing efficiency. Even three-dimensional (3D) micromixers with complex structures have low mixing efficiency in the low Reynolds number range. In this paper, a 3D split-and-recombination (SAR) micromixer inspired by the horseshoe transform principle is proposed to further improve the mixing efficiency. There 3D SAR micromixers with different subchannel sizes were designed and tested in the Reynolds numbers range of 0.1â€"100. The optimal size of the micromixer was revealed through computational fluid dynamics simulations and experimental test results. A minimum mixing index of 91% is achieved in the range of Reynolds numbers from 0.1 to 100. Especially, for Re â©ľ 20, the mixing index is higher than 99%. The results obtained indicate that this 3D SAR micromixer with an asymmetric structure shows a satisfactory choice in the fluid mixing process of microfluidic systems, and has a potential application in the field of microchip-based biochemical analysis.
