22. CaM kinase I[alpha]--induced phosphorylation of Drp1 regulates mitochondrial morphology

Abstract

Mitochondria are dynamic organelles that frequently move, divide, and fuse with one another to maintain their architecture and functions. However, the signaling mechanisms involved in these processes are still not well characterized. In this study, we analyze mitochondrial dynamics and morphology in neurons. Using time-lapse imaging, we find that [Ca.sup.2+] influx through voltage-dependent [Ca.sup.2+]channels (VDCCs) causes a rapid halt in mitochondrial movement and induces mitochondrial fission. VDCC-associated [Ca.sup.2+]signaling stimulates phosphorylation of dynamin-related protein 1 (Drp1) at serine 600 via activation of [Ca.sup.2+]/calmodulin-dependent protein kinase I[alpha] (CaMKle [alpha]). In neurons and HeLa cells, phosphorylation of Drp1 at serine 600 is associated with an increase in Drp 1 translocation to mitochondria, whereas in vitro, phosphorylation of Drp 1 results in an increase in its affinity for Fis 1. CaMKl[alpha] is a widely expressed protein kinase, suggesting that [Ca.sup.2+] is likely to be functionally important in the control of mitochondrial dynamics through regulation of Drp1 phosphorylation in neurons and other cell types.

Abstract

A dual-wavelength laser system with wideband tunable wavelength spacing and more than 2 W output power is proposed and demonstrated. The operation principle of the laser system is based on the saturation gain characteristic of a cladding-pumped Er3+/Yb3+ co-doped fibre amplifier combining with a wideband tunable fibre laser with flat power spectrum. The tuning range of the wavelength spacing is continuous within a 35 nm spectral range, and the power difference between both lasing wavelengths can be easily adjusted and controlled by a variable optical attenuator. The total output power can approximately keep unchangeable when the wavelength spacing and the power difference between both lasing wavelengths are continuously tuned and changed. The maximum total output power of the laser system is about 2.22 W.

43. Dual effects of cortical polarization on peripheral motor activity in the rabbit

Abstract

Anodal direct currents at intensities ranging from 0.3 to 30.0 microA were unilaterally applied for 30 min once a day to the premotor area of the rabbit cerebral cortex. The anodal polarization was repeated 10 times at intervals of 2-3 days, and the effect on the motor activity of the forelimbs during and after each polarization trial was compared with that before polarization. Peripheral motor activity was classified as either gentle flexion of forelimbs or struggle with violent movement of forelimbs. A current of 0.3 microA caused no change in motor behavior. Flexion of the forelimb contralateral to the polarized cortex was clearly increased when a polarizing current of 1.0 or 3.0 microA was applied, and peak flexion was observed between the third and seventh polarization trials. A current of 10 or 30 microA had no effect on forelimb flexion. Conversely, forelimb struggle on both sides was decreased when 10.0 or 30.0 microA, but not 1.0 or 3.0 microA, was applied. These results show that anodal polarization of the cerebral cortex exerts dual effects on peripheral motor activity, probably through changes in cortical excitability associated with the current intensity.
