Abstract Background Human parainfluenza virus-1 (HPIV-1) is a notable pathogen instigating acute respiratory tract infections in children. The article is to elucidate the epidemiological and genetic characteristics of HPIV-1 circulating in Hangzhou during the period of 2021–2022. Methods A cohort of 2360 nasopharyngeal swabs were amassed and subsequently examined via RT-PCR, with HPIV-1 positive samples undergoing P gene sequencing. Results The highest HPIV-1 infection rates were found in children aged between 3 and 6 years. A pronounced positive rate persisted through the latter half of 2021, with a notable decline observed in the initial half of 2022. All HPIV-1 strains could be clustered into 2 groups: Cluster 1, with strains similar to those found in Japan (LC764865, LC764864), and Cluster 2, with strains similar to the Beijing strain (MW575643). Conclusion In conclusion, our study contributes to the comprehensive data on the epidemiological and genetic characteristics of HPIV-1 in pediatric patients from Hangzhou, post the COVID-19 peak.

Abstract Background This study aims to evaluate the efficacy and clinical outcomes of Nice knot-assisted fixation in the management of comminuted and displaced clavicle fractures. Methods A systematic search was conducted across multiple electronic databases, including PubMed, Embase, Cochrane Library, China Biology Medicine (CBM) database, CNKI, Wanfang, and VIP database, to identify studies comparing Nice knot-assisted fixation with traditional surgical treatment for displaced midshaft clavicle fractures. The primary outcomes assessed included fracture healing times, hospitalization days, complications, and functional outcomes. Secondary outcomes such as intraoperative blood loss, operative time, incision length, and fluoroscopy time were also evaluated. Data were analyzed using random-effects models, and summary statistics including Mean Difference (MD), risk ratios (RRs) and theirs’ 95% confidence intervals (CIs) were calculated. Results Screening of the literature yielded a total of 11 studies meeting the inclusion criteria, involving 754 patients. Meta-analysis of the pooled data demonstrated a significant advantage of Nice knot-assisted fixation over traditional surgical treatment in terms of operative time (MD = -11.53, 95% CI: -18.16 to -4.91, p = 0.0006) and blood loss (MD = -14.19, 95% CI: -20.93 to -7.45, p = 0.00001). Additionally, Nice knot-assisted fixation was associated with reduced fracture healing time (MD = -0.63, 95% CI: -1.12 to -0.14, p = 0.01) rather than hospitalization days (MD = -0.47, 95% CI: -1.14 to 0.21, p = 0.18) and complications (RR = 0.48, 95% CI: 0.23 to 1.01, p = 0.05) compared to traditional surgical treatment. Moreover, nice knot-assisted fixation was associated with increased constant-murley score and Neer score, and reduced visual analogue scale (VAS) score compared to traditional surgical treatment. Conclusion This study highlights that Nice knot assistance offers advantages in reducing intraoperative blood loss, shortening operation time, and achieving favorable postoperative outcomes.

Abstract Background Homocysteine (Hcy) is a pro-inflammatory molecule that has the potential to induce oxidative damage to cells and stimulate the release of inflammatory mediators. Hcy has been observed to enhance the production of inflammatory agents in vascular endothelial cells. However, the impact of Hcy on intestinal mucosal inflammation remains largely unexplored. Therefore, the objective of this study was to examine the potential of Hcy to stimulate the synthesis of inflammatory mediators and elucidate the underlying mechanisms in the intestinal mucosa. Methods A total of 99 patients diagnosed with inflammatory bowel disease (IBD) and 10 healthy individuals were included in this study to assess the impact of homocysteine (Hcy) on the levels of leukotriene E4 (LTE4) and prostaglandin E2 (PGE2). The underlying mechanism responsible for the generation of LTE4 and PGE2 induced by Hcy was investigated using colitis rats and Caco-2 cells. 32 Sprague–Dawley rats were categorized into four groups: normal control, TNBS model, normal with Hcy injection, and TNBS model with Hcy injection. The mRNA expressions of 5-LOX, COX-2, and NF-κB were assessed using reverse transcription quantitative polymerase chain reaction (RT-qPCR). Caco-2 cells were subjected to treatment with varying concentrations (10, 20, 50, 100 μmol/L) of Hcy and incubated for different durations (1, 3, 6 h). The alterations in NF-κB activity, as well as the levels of Hcy, LTE4, and PGE2, were measured using enzyme-linked immunosorbent assay (ELISA). Results The excretion of Hcy, LTE4, and PGE2 in urine exhibited significant increases in patients with inflammatory bowel disease (IBD), Crohn's disease (CD), and ulcerative colitis (UC). In addition, Hcy demonstrated a significant increase in the expression of 5-LOX, COX-2, and NF-κB, as well as elevated levels of LTE4 and PGE2 in rats with colitis. Furthermore, Hcy was found to induce NF-κB activation and nuclear translocation, thereby contributing to the enhanced synthesis of LTE4 and PGE2 in Caco-2 cells. Conclusions Hcy was found to enhance the expression of 5-LOX and COX-2 by activating NF-κB, thereby augmenting the production of LTE4 and PGE2, which ultimately exacerbates colonic inflammation in individuals with inflammatory bowel disease (IBD).

Abstract Background Preoperative smoking cessation may reduce postoperative complications in patients with lung cancer. However, the optimal duration of short-term preoperative smoking cessation remains unclear. Methods Three databases, PubMed, Embase, and the Cochrane Library, were searched for studies published up to April 5, 2024. The Newcastle–Ottawa scale was used to assess the risk of bias. The included studies compared the incidence of postoperative complications between patients with different preoperative smoking cessation times and those with persistent preoperative smoking. A meta-analysis of postoperative complications and events such as pneumonia was performed in patients with lung cancer. Results Fourteen studies met the inclusion criteria and included a total of 50,741 patients who had undergone pulmonary resection. The meta-analysis showed that preoperative smoking cessation of > 2 weeks and 1 month was effective in reducing the incidence of postoperative complications (OR 0.72; 95% CI 0.63–0.83; P 1 month is effective in reducing complications and pneumonia after pulmonary resection in patients with lung cancer, especially as video-assisted thoracoscopic surgery (VATS) and robotic-assisted surgery become more common.

Abstract Intra-tumor immune infiltration plays a pivotal role in the interaction with tumor cells in hepatocellular carcinoma (HCC). However, its phenotype and related spatial structure remained elusive. To address these limitations, we conducted a comprehensive study combining spatial data (38,191 spots from eight samples) and single-cell data (56,022 cells from 20 samples). Our analysis revealed two distinct infiltration patterns: immune exclusion and immune activation. Plasma cells emerged as the primary cell type within intra-tumor immune clusters. Notably, we observed the co-location of CCL19+ fibroblasts with plasma cells, which secrete chemokines and promote T-cell activation and leukocyte migration. Conversely, in immune-exclusion samples, this co-location was primarily observed in the adjacent normal area. This co-localization correlated with T cell infiltration and the formation of tertiary lymphoid structures, validated by multiplex immunofluorescence conducted on twenty HCC samples. Both CCL19+ fibroblasts and plasma cells were associated with favorable survival outcomes. In an immunotherapy cohort, HCC patients who responded favorably exhibited higher infiltration of CCL19+ fibroblasts and plasma cells. Additionally, we observed the accumulation of DKK1+ tumor cells within the tumor area in immune-exclusion samples, particularly at the tumor boundary, which inhibited the infiltration of CCL19+ fibroblasts and plasma cells into the tumor area. Furthermore, in immune-exclusion samples, the SPP1 signaling pathway demonstrated the highest activity in communication between tumor and immune clusters, and CCL19-CCR7 played a pivotal role in the self-communication of immune clusters. This study elucidates immune exclusion and immune activation patterns in HCC and identifies relevant factors contributing to immune resistance.

The synergistic alkali metals fixation potential of Oxygen Carrier Aided Combustion (OCAC) technology has attracted increasing attention. Using the tailings from natural ilmenite after titanium extraction and its individual components Fe2TiO5 and TiO2 as absorbents, and KCl and CH3COOK as simulated alkali sources, the high-temperature absorption characteristics of alkali metal potassium by ilmenite tailings were studied in a static absorption experimental setup. The effects of various factors such as time, particle size, temperature, mass ratio, and gas-solid components on the absorption performance were systematically investigated. Techniques such as X-ray diffraction, inductively coupled plasma optical emission spectrometry scanning electron microscopy and X-ray energy dispersive spectroscopy were used to reveal the micro-mechanisms of K enrichment on the surface and bulk phase migration within ilmenite. The results show that ilmenite tailings possess good potassium absorption capabilities at high temperatures, with the absorption rate displaying a trend of rapid increase followed by a slow decline over time. Under certain conditions, reducing particle size, increasing reaction temperature, and the concentration of K in the gas phase can further enhance the alkali fixation performance of ilmenite tailings. At the temperatures and alkali metal concentrations like those of actual industrial flue gases, ilmenite tailings can achieve a K capture rate of over 40%. The primary phase of ilmenite tailings has a unique lamellar structure composed of iron-rich and titanium-rich layers arranged alternately. Compared to single-component model substances, it has a more complex morphology and superior alkali fixation capabilities. The process involves an initial deposition of K on the ilmenite surface, forming an enriched layer, followed by a solid-phase diffusion inward and migration deep into the particle along the titanium-rich layers. Fe in the titanium-rich layers migrates synchronously outward, doubly impacting the absorption of K in ilmenite. Additionally, Cl can further promote the absorption of K in ilmenite by reacting with Fe on the surface to form volatile FeCl2. Minerals such as almandine calcian in ilmenite tailings also show a significant synergistic absorption of K. The findings further confirm the synergistic alkali metals fixation potential of OCAC technology, while also offering new insights into the resource utilization of low-grade ilmenite.

Soil tensile strength is a critical parameter governing the initiation and propagation of tensile cracking. This study proposes an eco-friendly approach to improve the tensile behavior and crack resistance of clayey soils. To validate the feasibility and efficacy of the proposed approach, direct tensile tests were employed to determine the tensile strength of the compacted soil with different W-OH treatment concentrations and water contents. Desiccation tests were also performed to evaluate the effectiveness of W-OH treatment in enhancing soil tensile cracking resistance. During this period, the effects of W-OH treatment concentration and water content on tensile properties, soil suction and microstructure were investigated. The tensile tests reveal that W-OH treatment has a significant impact on the tensile strength and failure mode of the soil, which not only effectively enhances the tensile strength and failure displacement, but also changes the brittle failure behavior into a more ductile quasi-brittle failure behavior. The suction measurements and mercury intrusion porosimetry (MIP) tests show that W-OH treatment can slightly reduce soil suction by affecting skeleton structure and increasing macropores. Combined with the microstructural analysis, it becomes evident that the significant improvement in soil tensile behavior through W-OH treatment is mainly attributed to the W-OH gel's ability to provide additional binding force for bridging and encapsulating the soil particles. Moreover, desiccation tests demonstrate that W-OH treatment can significantly reduce or even inhibit the formation of soil tensile cracking. With the increase of W-OH treatment concentration, the surface crack ratio and total crack length are significantly reduced. This study enhances a fundamental understanding of eco-polymer impacts on soil mechanical properties and provides valuable insight into their potential application for improving soil crack resistance.

Liparis gigantea is a Chinese traditional medicinal herb in the Orchidaceae family. It is a rare and special Liparis species that exhibits relatively large flowers. To illuminate its phylogenetic status and augment genomic resources, the complete chloroplast (cp) genome of L. gigantea was first sequenced and assembled using whole genome next-generation sequencing in this study. The cp genome size is 158,462 bp with a total GC content of 36.9%. Characterized by a quadripartite structure, the genome consists of a large single-copy (LSC) region of 86,032 bp, a small single-copy (SSC) region of 18,322 bp, which is separated by a pair of 27,054 bp inverted repeat regions (IRs). A total of 133 genes were annotated, including 87 protein-coding genes, 38 tRNA genes, and 8 rRNA genes. Phylogenetic analysis strongly supported L. gigantea as the sister to two closely related terrestrial species, Liparis nervosa and L. vivipara. The results of this study provide genomic information for future research and application of this medicinal herb.

Abstract Spread through air spaces (STAS) is a recognized aggressive pattern in lung cancer, serving as a crucial risk factor for postoperative recurrence. However, its phenotype and related spatial structure have remained elusive. To address these limitations, we conducted a comprehensive study based on spatial data, analyzing over 30,000 spots from 14 non-STAS samples and one STAS sample. We observed increased proliferation activities and angiogenesis in STAS, identifying S100P as a potential biomarker for STAS. Furthermore, our investigation into the heterogeneity of STAS tumor cells revealed a subset identified as S100P + TFF1 +, exhibiting a negative impact on patients' survival in public datasets. This subtype exhibited the highest activities in the TGFb and hypoxia, suggesting its potential pro-tumor role within the tumor microenvironment. To assess the role of S100P + TFF1 + tumor cells in therapy response, we included data from two clinical trial cohorts (BPI-7711 for EGFR-TKI therapy and ORIENT-3 for immunotherapy). The presence of S100P + TFF1 + tumor cells correlated with worse responses to both EGFR-TKI therapy and immunotherapy. Notably, TFF1 emerged as a serum marker for predicting EGFR-TKI response. Cell–cell communication analysis revealed that the TGFb signaling pathway was the most activated in S100P + TFF1 + tumor cells, with TGFB2-TGFBR2 identified as the main ligand-receptor pair. This was further validated by multiplex immunofluorescence performed on twenty NSCLC samples. In summary, our study identified S100P as the biomarker for STAS and highlighted the adverse role of S100P + TFF1 + tumor cells in survival outcomes.

Abstract Background Apnea is common in preterm infants and can be accompanied with severe hypoxic damage. Early assessment of apnea risk can impact the prognosis of preterm infants. We constructed a prediction model to assess apnea risk in premature infants for identifying high-risk groups. Methods A total of 162 and 324 preterm infants with and without apnea who were admitted to the neonatal intensive care unit of Xiamen University between January 2018 and December 2021 were selected as the case and control groups, respectively. Demographic characteristics, laboratory indicators, complications of the patients, pregnancy-related factors, and perinatal risk factors of the mother were collected retrospectively. The participants were randomly divided into modeling (n = 388) and validation (n = 98) sets in an 8:2 ratio. Least Absolute Shrinkage and Selection Operator (LASSO) and multivariate logistic regression analyses were used to independently filter variables from the modeling set and build a model. A nomogram was used to visualize models. The calibration and clinical utility of the model was evaluated using consistency index, receiver operating characteristic (ROC) curve, calibration curve, and decision curve, and the model was verified using the validation set. Results Results of LASSO combined with multivariate logistic regression analysis showed that gestational age at birth, birth length, Apgar score, and neonatal respiratory distress syndrome were predictors of apnea development in preterm infants. The model was presented as a nomogram and the Hosmer-Lemeshow goodness of fit test showed a good model fit (χ 2 =5.192, df=8, P=0.737), with Nagelkerke R 2 of 0.410 and C-index of 0.831. The area under the ROC curve and 95% CI were 0.831 (0.787-0.874) and 0.829 (0.722-0.935), respectively. Delong's test comparing the AUC of the two data sets showed no significant difference (P=0.976). The calibration curve showed good agreement between the predicted and actual observations. The decision curve results showed that the threshold probability range of the model was 0.07-1.00, the net benefit was high, and the constructed clinical prediction model had clinical utility. Conclusions Our risk prediction model based on gestational age, birth length, Apgar score 10 min post-birth, and neonatal respiratory distress syndrome was validated in many aspects and had good predictive efficacy and clinical utility.

Osteoid plays a crucial role in directing cell behavior and osteogenesis through its unique characteristics, including viscoelasticity and liquid crystal (LC) state. Thus, integrating osteoid-like features into 3D printing scaffolds proves to be a promising approach for personalized bone repair. Despite extensive research on viscoelasticity, the role of LC state in bone repair has been largely overlooked due to the scarcity of suitable LC materials. Moreover, the intricate interplay between LC state and viscoelasticity in osteogenesis remains poorly understood. Here, we developed innovative hydrogel scaffolds with osteoid-like LC state and viscoelasticity using digital light processing with a custom LC ink. By utilizing these LC scaffolds as 3D research models, we discovered that LC state mediates high protein clustering to expose accessible RGD motifs to trigger cell-protein interactions and osteogenic differentiation, while viscoelasticity operates via mechanotransduction pathways. Additionally, our investigation revealed a synergistic effect between LC state and viscoelasticity, amplifying cell-protein interactions and osteogenic mechanotransduction processes. Furthermore, the interesting mechanochromic response observed in the LC hydrogel scaffolds suggests their potential application in mechanosensing. Our findings shed light on the mechanisms and synergistic effects of LC state and viscoelasticity in osteoid on osteogenesis, offering valuable insights for the biomimetic design of bone repair scaffolds.

The intensification of electromagnetic (EM) pollution and the development of military detection technology have increased the requirements for EM functional materials. In this study, a molybdenum diselenide@reduced graphene oxide (MoSe2@rGO)-assembled architecture is constructed, where the MoSe2 nanosheets grow uniformly on the rGO sheets. By regulating the contributions of conduction genes and polarization genes, adjustable EM functions of MoSe2@rGO hybrids can be achieved. The reflection loss (RL) of the sample can reach −68.7 dB at a thickness of 2.32 mm, and the maximum effective absorption bandwidth can reach 5.04 GHz. When conduction genes dominate, the MoSe2@rGO hybrids exhibit a 98.7% electromagnetic interference (EMI) shielding efficiency. The design of the EM energy conversion device and the results of the radar cross section (RCS) simulation demonstrate the practical application potential of the material. This work provides inspiration for designing multifunctional EM materials.

We report two cases of sarcoidosis presenting with skin nodules as the first manifestation, without prominent respiratory symptoms. Chest CT showed multiple pulmonary nodules and lymphadenopathy. Case 1, a 47-year-old female, presented with nodules on the elbows and fingers. Dermatological examination revealed demarcated, asymptomatic red and skin-colored nodules on the right elbow, and the index finger and the little finger of the left hand, sized 0.3 to 0.9 cm in diameter. Histopathology of the left pinky finger showed normal epidermis and epithelia granuloma with fibrous encapsulated naked tubercles in the dermis. Chest CT revealed bronchitis and multiple nodules in the both lungs, mediastinal and hilar lymphadenopathy. The diagnosis was sarcoidosis. Case 2, a 44-year-old female, complained of a 3-month history of nodules on the left lower limb. Dermatological examination showed two dark red and skin-colored nodules, sized 2.0 cm×2.0 cm and 3.0 cm×2.0 cm, respectively, on the anterior tibia of the left lower leg. A demarcated asymptomatic skin-colored nodule was observed on the posterior left lower leg. Histopathology of the left shin revealed mild epidermal hyperplasia and superficial dermal infiltrates of epithelioid cells and multinucleated giant cells. Chest CT revealed multiple nodules in the lower lobe of the left lung, upper and middle lobes of the right lung, mosaic perfusion of both lungs, a few fibers and calcifications in the middle and lower lobes of the right lung, mediastinal lymphadenopathy with calcification. Patients was diagnosed with sarcoidosis. Both patients were treated with oral prednisone acetate at 30 mg/d for 2 months, followed by a gradual reduction in glucocorticoid dose. No new rash was found after one-year follow-up.

Abstract Background and Objective Tinengotinib, a novel multi-target small molecule kinase inhibitor, is currently undergoing phase II clinical trial in the USA and China. The purpose of this open-label study was to investigate the absorption, metabolism, and excretion of [14C]tinengotinib following a single oral dose in healthy subjects. Methods Six healthy male subjects received a single oral dose of [14C]tinengotinib capsules at 10 mg/100 µCi, and blood, urine, and feces samples were collected. Phenotyping experiments were further conducted to confirm the enzymes involved in its metabolism. Results Tinengotinib was rapidly absorbed in plasma with a time to peak drug concentration (T max) of 1.0–4.0 h post-dose and a long terminal half-life (t ½) of 23.7 h. Blood-to-plasma radioactivity concentration ratios across timepoints ranged from 0.780 to 0.827, which indicated minimal association of radioactivity with blood cells. The mean cumulative excreted radioactivity was 99.57% of the dose, including 92.46% (68.65% as unchanged) in feces and 7.11% (0.28% as unchanged) in urine. In addition to unchanged tinengotinib, a total of 11 radioactive metabolites were identified in plasma, urine, and feces. The most abundant circulating radioactivity was the parent drug in plasma, which comprised 88.23% of the total radioactivity area under the concentration–time curve (AUC). Metabolite M410-3 was a major circulating metabolite, accounting for 5.38% of the parent drug exposure and 4.75% of the total drug-related exposure, respectively. All excreted metabolites accounted for less than 5.10% and 1.82% of the dose in feces and urine, respectively. In addition, no unique metabolites were observed in humans. Tinengotinib was metabolized mainly via CYP3A4. Conclusions Overall, tinengotinib demonstrated a complete mass balance with limited renal excretion, no disproportionate blood metabolism, and slow elimination, primarily through the fecal route. The results of this study provide evidence to support the rational use of tinengotinib as a pharmacotherapeutic agent. Registration ChinadrugTrials.org.cn identifier: CTR20212852.

Pogostemon cablin essential oil (PEO), extracted from P. cablin, has anti-oxidant, anti-inflammatory, and anti-stress properties, as well as the ability to improve gastrointestinal digestion. This study aims to evaluate the effects of PEO on the performance, rumen epithelial morphology, and barrier function in heat-stressed beef cattle. Thirty-six male Jingjiang cattle at 18 months old were randomly assigned into four groups and fed a diet containing PEO at 0 (control), 50, 100, or 150 mg/kg in the feed concentrate (n = 9). All experimental cattle were fed under high temperature and humidity in summer for 60 days. The results indicated that 50 mg/kg of PEO treatment enhanced the average daily gain of beef cattle compared with the control group (P = 0.032). All PEO treatments reduced the diamine oxidase activity (P = 0.004) and malondialdehyde content (P = 0.008) in serum. In addition, the content of 70 kDa heat shock protein in the 100 mg/kg group was increased, and the activity of glutathione peroxidase and total antioxidant capacity in both 100 mg/kg and 150 mg/kg groups were enhanced compared to the control group (P

Abstract Background Anaplastic lymphoma kinase (ALK) test in advanced non‐small cell lung cancer (NSCLC) can help physicians provide target therapies for patients harboring ALK gene rearrangement. This study aimed to investigate the real‐world test patterns and positive rates of ALK gene rearrangements in advanced NSCLC. Methods In this real‐world study (ChiCTR2000030266), patients with advanced NSCLC who underwent an ALK rearrangement test in 30 medical centers in China between October 1, 2018 and December 31, 2019 were retrospectively analyzed. Interpretation training was conducted before the study was initiated. Quality controls were performed at participating centers using immunohistochemistry (IHC)‐VENTANA‐D5F3. The positive ALK gene rearrangement rate and consistency rate were calculated. The associated clinicopathological characteristics of ALK gene rearrangement were investigated as well. Results The overall ALK gene rearrangement rate was 6.7% in 23,689 patients with advanced NSCLC and 8.2% in 17,436 patients with advanced lung adenocarcinoma. The quality control analysis of IHC‐VENTANA‐D5F3 revealed an intra‐hospital consistency rate of 98.2% (879/895) and an inter‐hospital consistency rate of 99.2% (646/651). IHC‐VENTANA‐D5F3 was used in 53.6%, real‐time polymerase chain reaction (RT‐PCR) in 25.4%, next‐generation sequencing (NGS) in 18.3%, and fluorescence in‐situ hybridization (FISH) in 15.9% in the adenocarcinoma subgroup. For specimens tested with multiple methods, the consistency rates confirmed by IHC‐VENTANA‐D5F3 were 98.0% (822/839) for FISH, 98.7% (1,222/1,238) for NGS, and 91.3% (146/160) for RT‐PCR. The overall ALK gene rearrangement rates were higher in females, patients of ≤ 35 years old, never smokers, tumor cellularity of > 50, and metastatic specimens used for testing in the total NSCLC population and adenocarcinoma subgroup (all P < 0.05). Conclusions This study highlights the real‐world variability and challenges of ALK test in advanced NSCLC, demonstrating a predominant use of IHC‐VENTANA‐D5F3 with high consistency and distinct clinicopathological features in ALK‐positive patients. These findings underscore the need for a consensus on optimal test practices and support the development of refined ALK test strategies to enhance diagnostic accuracy and therapeutic decision‐making in NSCLC.

Abstract To investigate the dispersion process of the underground toxic gas carbon monoxide (CO) into the refuge chamber during a mine disaster and enhance the survival rate of trapped miners, a simplified model of an underground refuge chamber and the main roadway was constructed. The impact of temperature and pressurized air volume on CO dispersion into the refuge chamber has been examined through both analog experiments and numerical simulations, and the reliability of the simulation results was verified. The results indicate that CO dispersion into the refuge chamber through the top of the protective isolation door occurs when the temperature in the refuge chamber is lower than that of the toxic gas. When the temperature of the toxic gas is higher, it tends to enter the refuge chamber through the bottom of the protective isolation door. The evolution of CO concentration in the transition chamber can be divided toxic survival chamber can be categorized into a sudden decline stage and a stable stage. And a flexible isolation door designed to control the entry of toxic gases into the refuge chamber was implemented, and its impact on CO dispersion has been compared and analyzed. When the temperature of the main roadway is 50 °C and the temperature of the refuge chamber is 20 °C, the required pressurized air volume to maintain the CO concentration within the safe threshold (24 ppm) is reduced to 69.6% of that needed without the isolation door, thereby significantly reducing the infiltration of harmful gases from the main roadway into the refuge chamber.

Abstract Background The shoot apical meristem (SAM), from which all above-ground tissues of plants are derived, is critical to plant morphology and development. In maize (Zea mays), loss-of-function mutant studies have identified several SAM-related genes, most encoding homeobox transcription factors (TFs), located upstream of hierarchical networks of hundreds of genes. Results Here, we collect 46 transcriptome and 16 translatome datasets across 62 different tissues or stages from the maize inbred line B73. We construct a dynamic regulome for 27 members of three SAM-related homeobox subfamilies (KNOX, WOX, and ZF-HD) through machine-learning models for the detection of TF targets across different tissues and stages by combining tsCUT&Tag, ATAC-seq, and expression profiling. This dynamic regulome demonstrates the distinct binding specificity and co-factors for these homeobox subfamilies, indicative of functional divergence between and within them. Furthermore, we assemble a SAM dynamic regulome, illustrating potential functional mechanisms associated with plant architecture. Lastly, we generate a wox13a mutant that provides evidence that WOX13A directly regulates Gn1 expression to modulate plant height, validating the regulome of SAM-related homeobox genes. Conclusions The SAM-related homeobox transcription-factor regulome presents an unprecedented opportunity to dissect the molecular mechanisms governing SAM maintenance and development, thereby advancing our understanding of maize growth and shoot architecture.

Abstract Background The existing predictive models for metabolic-associated fatty liver disease (MAFLD) possess certain limitations that render them unsuitable for extensive population-wide screening. This study is founded upon population health examination data and employs a comparison of eight distinct machine learning (ML) algorithms to construct the optimal screening model for identifying high-risk individuals with MAFLD in China. Methods We collected physical examination data from 5,171,392 adults residing in the northwestern region of China, during the year 2021. Feature selection was conducted through the utilization of the Least Absolute Shrinkage and Selection Operator (LASSO) regression. Additionally, class balancing parameters were incorporated into the models, accompanied by hyperparameter tuning, to effectively address the challenges posed by imbalanced datasets. This study encompassed the development of both tree-based ML models (including Classification and Regression Trees, Random Forest, Adaptive Boosting, Light Gradient Boosting Machine, Extreme Gradient Boosting, and Categorical Boosting) and alternative ML models (specifically, k-Nearest Neighbors and Artificial Neural Network) for the purpose of identifying individuals with MAFLD. Furthermore, we visualized the importance scores of each feature on the selected model. Results The average age (standard deviation) of the 5,171,392 participants was 51.12 (15.00) years, with 52.47% of the participants being females. MAFLD was diagnosed by specialized physicians. 20 variables were finally included for analyses after LASSO regression model. Following ten rounds of cross-validation and parameter optimization for each algorithm, the CatBoost algorithm exhibited the best performance, achieving an Area Under the Receiver Operating Characteristic Curve (AUC) of 0.862. The ranking of feature importance indicates that age, BMI, triglyceride, fasting plasma glucose, waist circumference, occupation, high density lipoprotein cholesterol, low density lipoprotein cholesterol, total cholesterol, systolic blood pressure, diastolic blood pressure, ethnicity and cardiovascular diseases are the top 13 crucial factors for MAFLD screening. Conclusion This study utilized a large-scale, multi-ethnic physical examination data from the northwestern region of China to establish a more accurate and effective MAFLD risk screening model, offering a new perspective for the prediction and prevention of MAFLD.

Abstract Background Müllerian duct anomalies (MDAs) are congenital developmental disorders that present as a series of abnormalities within the reproductive tracts of females. Genetic factors are linked to MDAs and recent advancements in whole-exome sequencing (WES) provide innovative perspectives in this field. However, relevant mechanism has only been investigated in a restricted manner without clear elucidation of respective observations. Methods Our previous study reported that 2 of 12 patients with MDAs harbored the CHD1L variant c.348-1G>C. Subsequently, an additional 85 MDAs patients were recruited. Variants in CHD1L were screened through the in-house database of WES performed in the cohort and two cases were identified. One presented with partial septate uterus with left renal agenesis and the other with complete septate uterus, duplicated cervices and longitudinal vaginal septum. The pathogenicity of the discovered variants was further assessed by molecular dynamics simulation and various functional assays. Results Ultimately, two novel heterozygous CHD1L variants, including a missense variant c.956G>A (p.R319Q) and a nonsense variant c.1831C>T (p.R611*) were observed. The variants were absent in 100 controls. Altogether, the contribution yield of CHD1L to MDAs was calculated as 4.12% (4/97). All three variants were assessed as pathogenic through various functional analysis. The splice-site variant c.348-1G>C resulted in a 11 bp sequence skipping in exon 4 of CHD1L and led to nonsense mediated decay of its transcripts. Unlike WT CHD1L, the truncated R611* protein mislocalized to the cytoplasm, abolish the ability of CHD1L to promote cell migration and failed to interact with PARP1 owing to the loss of macro domain. The R319Q variant exhibited conformational disparities and showed abnormal protein recruitment behavior through laser microirradiation comparing with the WT CHD1L. All these variants impaired the CHD1L function in DNA damage repair, thus participating in MDAs. Conclusions The current study not only expands the mutational spectrum of CHD1L in MDAs but determines three variants as pathogenic according to ACMG guidelines with reliable functional evidence. Additionally, the impairment in DNA damage repair is an underlying mechanism involved in MDAs.

Abstract Background Healthy dietary patterns have been negatively associated with methylation-based measures of biological age, yet previous investigations have been unable to establish the relationship between them and biological aging assessed through blood chemistry-based clinical biomarkers. We sought to assess the associations of 4 dietary metrics with 4 measures of biological age. Methods Among 16,666 participants in NHANES 1999–2018, 4 dietary metrics [Dietary inflammatory index (DII), Dietary approaches to stop hypertension index (DASH), Alternate mediterranean diet score (aMED), and Healthy eating index-2015 (HEI-2015)] were calculated through the ‘dietaryindex’ R package. Twelve blood chemistry parameters were utilized to compute 4 indicators of biological age [homeostatic dysregulation (HD), allostatic load (AL), Klemera–Doubal method (KDM), and phenotypic age (PA)]. Binomial logistic regression models and restricted cubic spline (RCS) regression were employed to evaluate the associations. Results All 4 dietary metrics were significantly associated with biological age acceleration or deceleration. In comparison to the lowest DII, the odds ratios (ORs) for accelerated HD, AL, KDM, and PA were 1.25 (1.08,1.45), 1.29 (1.11,1.50), 1.34 (1.08,1.65), and 1.61 (1.39,1.87) for the highest. The multivariable-adjusted ORs of the highest quartile of DASH, aMED, and HEI-2015 were 0.85 (0.73,0.97), 0.88 (0.74,1.04), and 0.84 (0.74,0.96) for HD, 0.64 (0.54,0.75), 0.61 (0.52,0.72), and 0.70 (0.59,0.82) for AL, 0.68 (0.54,0.85), 0.62 (0.50,0.76), and 0.71 (0.58,0.87) for KDM, and 0.50 (0.42,0.59), 0.64 (0.54,0.76), and 0.51 (0.44,0.58) for PA when compared with the lowest level. The findings were validated by the best-fitting dose-response curves for the associations. Among participants consuming dietary supplements (P interaction < 0.05), the positive effects of a healthy dietary pattern on biological aging were more pronounced. Systemic immune inflammation index (SII) and atherogenic index of plasma (AIP) were identified as being involved in and mediating the associations. Conclusions Biological aging assessed through blood chemistry-based clinical biomarkers is negatively associated with diet quality. The anti-aging benefits of improving the diet may be due to its ability to reduce inflammation and lower blood lipids.

Abstract Background Poorly supported transitions from pediatric to adult healthcare can lead to negative health outcomes for youth and their families. To better understand the current landscape of healthcare transition care across Canada, the Canadian Health Hub in Transition (the “Transition Hub”, established in 2019) identified a need to: (1) describe programs and services supporting the transition from pediatric to adult healthcare across Canada; and (2) identify strengths, barriers, and gaps affecting the provision of transition services. Methods Our project included two iterative steps: a national survey followed by a qualitative descriptive study. Service providers were recruited from the Transition Hub and invited to complete the survey and participate in the qualitative study. The survey was used to collect program information (e.g., setting, clinical population, program components), and semi-structured interviews were used to explore providers’ perspectives on strengths, barriers, and gaps in transition services. Qualitative data were analyzed using the Framework Method. Results Fifty-one surveys were completed, describing 48 programs (22 pediatric, 19 bridging, and 7 adult) across 9 provinces. Almost half of the surveyed programs were in Ontario (44%) and most programs were based in hospital (65%) and outpatient settings (73%). There was wide variation in the ages served, with most programs focused on specific diagnostic groups. Qualitative findings from 23 interviews with service providers were organized into five topics: (1) measuring transition success; (2) program strengths; (3) barriers and gaps; (4) strategies for improvement; and (5) drivers for change. Conclusions While national transition guidelines exist in Canada, there is wide variation in the way young people and their families are supported. A national strategy, backed by local leadership, is essential for instigating system change toward sustainable and universally accessible support for healthcare transition in Canada.

Abstract Rapid synthesis of high-entropy alloy nanoparticles (HEA NPs) offers new opportunities to develop functional materials in widespread applications. Although some methods have successfully produced HEA NPs, these methods generally require rigorous conditions such as high pressure, high temperature, restricted atmosphere, and limited substrates, which impede practical viability. In this work, we report laser solid-phase synthesis of CrMnFeCoNi nanoparticles by laser irradiation of mixed metal precursors on a laser-induced graphene (LIG) support with a 3D porous structure. The CrMnFeCoNi nanoparticles are embraced by several graphene layers, forming graphene shell-encapsulated HEA nanoparticles. The mechanisms of the laser solid-phase synthesis of HEA NPs on LIG supports are investigated through theoretical simulation and experimental observations, in consideration of mixed metal precursor adsorption, thermal decomposition, reduction through electrons from laser-induced thermionic emission, and liquid beads splitting. The production rate reaches up to 30 g/h under the current laser setup. The laser-synthesized graphene shell-encapsulated CrMnFeCoNi NPs loaded on LIG-coated carbon paper are used directly as 3D binder-free integrated electrodes and exhibited excellent electrocatalytic activity towards oxygen evolution reaction with an overpotential of 293 mV at the current density of 10 mA/cm2 and exceptional stability over 428 h in alkaline media, outperforming the commercial RuO2 catalyst and the relevant catalysts reported by other methods. This work also demonstrates the versatility of this technique through the successful synthesis of CrMnFeCoNi oxide, sulfide, and phosphide nanoparticles.

Abstract Introduction Since 2015, the Guanghua School of Stomatology has established an elective dental basic research course (EDBRC). To make all students benefit from the dental basic research course without causing excessive academic burden, the “flexible” compulsory dental basic research course (CDBRC) was settled in 2020. This study intends to introduce the “flexible” compulsory teaching module and assessment system of CDBRC, and analyze its effectiveness over 3 years. Materials and methods The grade point average (GPA), course pressure, level of basic research knowledge and skills, and students’ research achievements were collected and analyzed between EDBRC and CDBRC. The unpaired t test was used to analyze the difference. Results The “flexible” CDBRC has been successfully constructed with compulsory teaching module and hierarchical assessment system. The CDBRC has not caused significant course pressure to students compared with the EDBRC. Besides, the “flexible” CDBRC can improve the students’ GPA, basic research knowledge, and research achievements. Conclusions The “flexible” CDBRC can improve students’ academic performance and basic research abilities without causing significant course pressure, which can be conducted in dental schools with similar backgrounds. Mesembryanthemum cordifolium, a perennial plant with crassulacean acid metabolism (CAM) in the Aizoaceae family, has significant ornamental and medicinal values. In this study, we reported the first complete chloroplast genome sequence of this species. The total genome size was 153,734 bp in length, including a large single-copy (LSC) region of 85,692 bp, a small single-copy (SSC) region of 18,212 bp, and a pair of inverted repeat (IR) regions of 24,915 bp by each. The overall GC content of the M. cordifolium chloroplast genome was 37.08%. The genome encodes 131 genes, comprising 87 protein-coding genes (PCGs), 36 transfer RNA genes (tRNAs), and eight ribosomal RNA genes (rRNAs). Phylogenetic analysis shows this species was relatively close to M. crystallinum. This chloroplast genome sequence will be valuable for species discrimination and for understanding phylogenetic relationships within the genus Mesembryanthemum.

This study examines the microstructural evolution, elemental distribution, phase composition, and hardness variations in multi-material samples fabricated using Laser Powder Bed Fusion (LPBF) with Invar and Ti6Al4V with compositional gradients mediated by copper. A range of compositional transitions from Invar to Ti6Al4V with different copper fractions are reported. Defects such as gas and keyhole pores were detected in well-deep keyhole mode areas, while lack of fusion defects were observed in samples with pure Cu interlayers due to poor wettability and processing parameter variations. Crack formation between interlayer gradient zones and Invar regions was linked to FeNi and CuNi intermetallic phases. Intermetallic compounds (e.g., FeNi, CuNi, TiFe, TiNi) significantly influenced the microstructure and mechanical performance. Vickers hardness testing shows varying hardness across different builds, with the highest hardness being observed in the gradient zone enabled by pure copper (approximately 620 HV). This study also underscores the need for tailored component design linking build processing parameters and compositional transitions in multi-material LPBF to achieve desired properties and component structural behaviour.

Optical imaging in the second near-infrared (NIR-II; 900–1880 nm) window is currently a popular research topic in the field of biomedical imaging. This study aimed to explore the application value of NIR-II fluorescence imaging in foot and ankle surgeries. A lab-established NIR-II fluorescence surgical navigation system was developed and used to navigate foot and ankle surgeries which enabled obtaining more high-spatial-frequency information and a higher signal-to-background ratio (SBR) in NIR-II fluorescence images compared to NIR-I fluorescence images; our result demonstrates that NIR-II imaging could provide higher-contrast and larger-depth images to surgeons. Three types of clinical application scenarios (diabetic foot, calcaneal fracture, and lower extremity trauma) were included in this study. Using the NIR-II fluorescence imaging technique, we observed the ischemic region in the diabetic foot before morphological alterations, accurately determined the boundary of the ischemic region in the surgical incision, and fully assessed the blood supply condition of the flap. NIR-II fluorescence imaging can help surgeons precisely judge surgical margins, detect ischemic lesions early, and dynamically trace the perfusion process. We believe that portable and reliable NIR-II fluorescence imaging equipment and additional functional fluorescent probes can play crucial roles in precision surgery.

Abstract Objectives Accurate segmentation of focal cortical dysplasia (FCD) lesions from MR images plays an important role in surgical planning and decision but is still challenging for radiologists and clinicians. In this study, we introduce a novel transformer-based model, designed for the end-to-end segmentation of FCD lesions from multi-channel MR images. Methods The core innovation of our proposed model is the integration of a convolutional neural network-based encoder-decoder structure with a multiscale transformer to augment the feature representation of lesions in the global field of view. Transformer pathways, composed of memory- and computation-efficient dual-self-attention modules, leverage feature maps from varying depths of the encoder to discern long-range interdependencies among feature positions and channels, thereby emphasizing areas and channels relevant to lesions. The proposed model was trained and evaluated on a public-open dataset including MR images of 85 patients using both subject-level and voxel-level metrics. Results Experimental results indicate that our model offers superior performance both quantitatively and qualitatively. It successfully identified lesions in 82.4% of patients, with a low false-positive lesion cluster rate of 0.176 ± 0.381 per patient. Furthermore, the model achieved an average Dice coefficient of 0.410 ± 0.288, outperforming five established methods. Conclusion Integration of the transformer could enhance the feature presentation and segmentation performance of FCD lesions. The proposed model has the potential to serve as a valuable assistive tool for physicians, enabling rapid and accurate identification of FCD lesions. Critical relevance statement This multiscale transformer-based model performs segmentation of focal cortical dysplasia lesions, aiming to help radiologists and clinicians make accurate and efficient preoperative evaluations of focal cortical dysplasia patients from MR images. Key Points The first transformer-based model was built to explore focal cortical dysplasia lesion segmentation. Integration of global and local features enhances the segmentation performance of lesions. A valuable benchmark for model development and comparative analyses was provided. Graphical Abstract

Abstract Shade avoidance helps plants maximize their access to light for growth under crowding. It is unknown, however, whether a priming shade avoidance mechanism exists that allows plants to respond more effectively to successive shade conditions. Here, we show that the shade-intolerant plant Arabidopsis can remember a first experienced shade event and respond more efficiently to the next event on hypocotyl elongation. The transcriptional regulator PHYTOCHROME-INTERACTING FACTOR 7 (PIF7) and the histone H3K27-demethylase RELATIVE OF EARLY FLOWERING 6 (REF6) are identified as being required for this shade avoidance memory. RNA-sequencing analysis reveals that shade induction of shade-memory-related genes is impaired in the pif7 and ref6 mutants. Based on the analyses of enrichments of H3K27me3, REF6 and PIF7, we find that priming shade treatment induces PIF7 accumulation, which further recruits REF6 to demethylate H3K27me3 on the chromatin of certain shade-memory-related genes, leading to a state poised for their transcription. Upon a second shade treatment, enhanced shade-mediated inductions of these genes result in stronger hypocotyl growth responses. We conclude that the transcriptional memory mediated by epigenetic modification plays a key role in the ability of primed plants to remember previously experienced shade and acquire enhanced responses to recurring shade conditions.

Lilium saccatum is a species of ornamental plant found in southeastern Xizang, China. In the present study, the complete chloroplast (cp) genome of L. saccatum was sequenced using next-generation sequencing (NGS). The de novo assembled cp genome was 151,839 bp in length, including a pair of inverted repeat regions (IRs; 26,421 bp), a small single-copy region (SSC; 17,528 bp), and a large single-copy region (LSC; 81,469 bp). The cp genome encodes 113 unique genes, including 79 protein-coding genes (PCGs), 30 tRNA genes, and four rRNA genes. The total GC content of the cp genome was 37.0%. Phylogenetic analysis of 24 cp genomes revealed that L. saccatum was closely related to L. souliei. This study could provide fundamental information for the phylogenomics and utilization of Lilium.

Automated pavement condition survey is of critical importance to road network management. There are three primary tasks involved in pavement condition surveys, namely data collection, data processing and condition evaluation. Artificial intelligence (AI) has achieved many breakthroughs in almost every aspect of modern technology over the past decade, and undoubtedly offers a more robust approach to automated pavement condition survey. This article aims to provide a comprehensive review on data collection systems, data processing algorithms and condition evaluation methods proposed between 2010 and 2023 for intelligent pavement condition survey. In particular, the data collection system includes AI-driven hardware devices and automated pavement data collection vehicles. The AI-driven hardware devices including right-of-way (ROW) cameras, ground penetrating radar (GPR) devices, light detection and ranging (LiDAR) devices, and advanced laser imaging systems, etc. These different hardware components can be selectively mounted on a vehicle to simultaneously collect multimedia information about the pavement. In addition, this article pays close attention to the application of artificial intelligence methods in detecting pavement distresses, measuring pavement roughness, identifying pavement rutting, analyzing skid resistance and evaluating structural strength of pavements. Based upon the analysis of a variety of the state-of-the-art artificial intelligence methodologies, remaining challenges and future needs with respect to intelligent pavement condition survey are discussed eventually.

Abstract Background In 2022, our team launched the pioneering national proficiency testing (PT) scheme for the pathological diagnosis of breast cancer, rapidly establishing its credibility throughout China. Aiming to continuously monitor and improve the proficiency of Chinese pathologists in breast pathology, the second round of the PT scheme was initiated in 2023, which will expand the number of participating institutions, and will conduct a nationwide investigation into the interpretation of HER2 0, 1+, and 2+/FISH- categories in China. Methods The methodology employed in the current round of PT scheme closely mirrors that of the preceding cycle in 2022, which is designed and implemented according to the “Conformity assessment—General requirements for proficiency testing”(GB/T27043—2012/ISO/IEC 17043:2010). More importantly, we utilized a statistics-based method to generate assigned values to enhance their robustness and credibility. Results The final PT results, published on the website of the National Quality Control Center for Cancer ( http://117.133.40.88:3927 ), showed that all participants passed the testing. However, a few institutions demonstrated systemic biases in scoring HER2 0, 1+, and 2+/FISH- with accuracy levels below 59%, considered unsatisfactory. Especially, the concordance rate for HER2 0 cases was only 78.1%, indicating challenges in distinguishing HER2 0 from low HER2 expression. Meanwhile, areas for histologic type and grade interpretation improvement were also noted. Conclusions Our PT scheme demonstrated high proficiency in diagnosing breast cancer in China. But it also identified systemic biases in scoring HER2 0, 1+, and 2+/FISH- at some institutions. More importantly, our study highlighted challenges in the evaluation at the extreme lower end of the HER2 staining spectrum, a crucial area for further research. Meanwhile, it also revealed the need for improvements in interpreting histologic types and grades. These findings strengthened the importance of robust quality assurance mechanisms, like the nationwide PT scheme conducted in this study, to maintain high diagnostic standards and identify areas requiring further training and enhancement.

Addressing peripheral nerve defects remains a significant challenge in regenerative neurobiology. Autografts emerged as the gold-standard management, however, are hindered by limited availability and potential neuroma formation. Numerous recent studies report the potential of wireless electronic system for nerve defects repair. Unfortunately, few has met clinical needs for inadequate electrode precision, poor nerve entrapment and insufficient bioactivity of the matrix material. Herein, we present an advanced wireless electrical nerve stimulator, based on water-responsive self-curling silk membrane with excellent bioabsorbable and biocompatible properties. We constructed a unique bilayer structure with an oriented pre-stretched inner layer and a general silk membrane as outer layer. After wetting, the simultaneous contraction of inner layer and expansion of outer layer achieved controllable super-contraction from 2D flat surface to 3D structural reconfiguration. It enables shape-adaptive wrapping to cover around nerves, overcomes the technical obstacle of preparing electrodes on the inner wall of the conduit, and prevents electrode breakage caused by material expansion in water. The use of fork capacitor-like metal interface increases the contact points between the metal and the regenerating nerve, solving the challenge of inefficient and rough electrical stimulation methods in the past. Newly developed electronic stimulator is effective in restoring 10 mm rat sciatic nerve defects comparable to autologous grafts. The underlying mechanism involves that electric stimulation enhances anterograde mitochondrial transport to match energy demands. This newly introduced device thereby demonstrated the potential as a viable and efficacious alternative to autografts for enhancing peripheral nerve repair and functional recovery.

As the number of high-density buildings has increased, the management of property with complex condominium ownership has become an ongoing challenge in property registration and management. The three-dimensional (3D) modeling of condominium ownership has emerged as an effective means of meeting this challenge and has attracted great attention from fields such as geographical information science, urban planning and management, and property administration. Much progress has been made in building 3D models of condominium ownership; however, existing studies are all on a case-by-case basis and have left some critical issues unsolved, such as vague ownership boundaries, spatial rights without physical counterparts, and the unfixed spatial extent. The purpose of this study is to construct a 3D building ownership model with multiple levels of detail in the context of Chinese law to overcome the defects of 3D models above. This 3D model is presented in a case study of China by subdividing ownership boundaries based on clarifying the internal structure of condominium ownership, embedding the apportionment mechanism, and integrating the semantics, attributes, and geometry associated with the physical and legal entity of the condominium. The proposed 3D model is implemented by extending Building Information Modeling (BIM) based on the Industry Foundation Classes (IFC) and inheriting legal information from Land Administration Domain Model (LADM). In this study, examples of condominium ownership from three real legal dispute cases in China are analyzed and tested. The study clearly demonstrates that the proposed model can provide a better cognitive understanding of the legal space of property by rendering unambiguous ownership boundaries and presenting the spatial internal structure of ownership, which offers solid technical support for dealing with property registration and many legal dispute cases about condominium ownership.

Antarctic krill (Euphausia superba) is a vital species in the Antarctic ecosystem, boasting substantial reserves and high nutritional value. It has emerged as a significant commercial fishery resource in the Southern Ocean. However, because of its exceptionally high autolytic enzyme activity, it is susceptible to autolysis and blackening during processing, storage, and transportation, resulting in a decline in the sensory quality and nutritional value of raw materials. Shrimp melanosis is caused by a series of enzymatic reactions mediated by phenoloxidase (PO) within the body. PO facilitates the oxidation of phenolic substances, leading to the formation of highly unstable quinone compounds, which further react to produce melanin. The accumulation of melanin significantly affects the appearance of Antarctic krill, thereby reducing its commercial value. On account of their cost-effectiveness and potent anti-blackening effects, sulfites are commonly used as shrimp anti-black agents. Sodium metabisulfite is one of the compounds whose use is allowed for shrimp preservation under GB 2760-2014 regulations. It not only inhibits PO activity, but also reduces quinone compounds to prevent blackening. However, excessive sulfite use can lead to discomfort and health issues such as gastrointestinal problems, liver disturbances, dyspnea, vomiting, diarrhea, and headaches. Therefore, GB 2760-2014 regulations stipulate that the residual SO2 content should not exceed 0.1 g/kg. Currently, the use of sodium metabisulfite in production is widespread but lacks standardized requirements, with limited guidance available on how to regulate its usage. Therefore, to assess the impact of sodium metabisulfite on blackening control in Antarctic krill and refine the treatment process, experiments were conducted to measure the changes in SO2 residue and PO activity in Antarctic krill subjected to various concentrations of sodium metabisulfite solutions and soaking times. Furthermore, observations and analyses were conducted to monitor the blackening of Antarctic krill during cold storage and frozen storage. The results demonstrated a positive correlation between the SO2 residue in Antarctic krill and the concentration of sodium metabisulfite solution. A concentration of 1 g/L sodium metabisulfite solution resulted in a SO2 residue of 0.065 g/kg in Antarctic krill, which is below the national standard limit of 0.1 g/kg. However, treatment with a 2 g/L solution led to a SO2 residue of 0.166 g/kg in shrimp, exceeding the national standard limit. Treatment with a 3 g/L solution resulted in a SO2 residue of 0.254 g/kg in shrimp, which significantly surpassed the national standard limit. Consequently, we recommended using 1 g/L or 2 g/L sodium metabisulfite solutions to prevent blackening in Antarctic krill while maintaining the SO2 residue below 0.1 g/kg. Under these conditions, the treatment time for a 1 g/L sodium pyrosulfite solution could be extended to 300 s, with a relative PO activity of 21.2%. For a 2.0 g/L sodium pyrosulfite solution, a treatment time of 30 s was suitable, yielding a relative PO activity of 34.5%. During storage, the control group exhibited noticeable blackening after storage at 4 ℃ for 24 h and frozen storage for 6 months, predominantly in the head, chest, abdomen, and tail sections, and slight variations were observed in the blackening process among individuals. Compared with the control group, the sodium metabisulfite treatment group displayed significantly lower blackening scores during storage (P < 0.05). Specifically, the 1 g/L sodium metabisulfite solution with 300 s treatment group and the 2 g/L sodium metabisulfite solution with 30 s treatment group showed obvious blackening at 48 h and 72 h of cold storage, respectively, and after 12 months of frozen storage, indicating that higher concentrations of sodium metabisulfite solution during treatment led to a better control effect on the blackening of Antarctic krill during storage. Notably, PO activity is just one of the factors influencing the melanosis process. Although the 1 g/L sodium metabisulfite solution with treatment for 300 s could better inhibit PO activity, the anti-blackening effect of this group was not as pronounced as that of the 2 g/L sodium metabisulfite solution with treatment for 30 s during storage. In summary, soaking in 1 g/L sodium metabisulfite solution for 300 s and 2 g/L sodium metabisulfite solution for 30 s resulted in relative PO enzyme activities of 21.2% and 34.5%, respectively. Simultaneously, the SO2 residues in shrimp were measured at 0.094 g/kg and 0.096 g/kg, respectively, meeting the GB 2760-2014 limit of SO2 residue not exceeding 0.1 g/kg. Compared with the control group, sodium metabisulfite treatment effectively delayed the blackening process of Antarctic krill during cold storage and frozen storage, with the 2 g/L sodium metabisulfite solution soaking for 30 s exhibiting superior anti-blackening effects. This suggests that a high concentration of sodium metabisulfite solution and a short treatment period had a better blackening inhibition effect. These findings offer a theoretical foundation and technical guidance for maintaining the quality of Antarctic krill during storage and transportation.

Through the creation and characterization of novel poly (L-lactic acid) (PLLA)-based nanocomposites containing graphene oxide (GO), nanocrystalline hydroxyapatite (nano-HAP), and nickel oxide (NiO) nanoparticles, this study seeks to improve the performance of bone tissue scaffolds. Utilizing a casting procedure to create nanocomposites, wet chemical methods were used to synthesize nano-HAP. X-ray diffraction, transmission electron microscopy, and Fourier transform infrared spectroscopy were used to characterize the materials. The nanocomposite comprising 5 % GO + 5 % NiO + 10 % nano-HAP showed a 110 % increase in tensile modulus, a 38 % increase in extension, and a 29 % increase in load-bearing capacity compared to plain PLLA. The results showed a considerable increase in mechanical strength. On the surfaces of the nanocomposite, osteoblast adhesion and proliferation increased by 185 %, according to biological tests. Studies on degradation revealed regulated rates that matched bones' natural mending process while preserving a pH environment that was steady. According to these results, adding nanostructures to PLLA scaffolds improves their mechanical and biological properties considerably, which may lead to therapeutic uses in bone tissue engineering in the future.

Both the physical features and social functions of urban green spaces (UGSs) are crucially important to the ecological and social benefits of urban residents. Increasing attention has been focused on exploring how UGS social functions affect the ecological and social benefits of urban residents, but the social functional classification of UGSs has rarely been studied, and related efficient classification methods are urgently needed. Thus, a novel methodological framework for accurately mapping UGS social functional zones was proposed by integrating remote sensing images, crowd-sourced geospatial data (i.e. point of interest data, the OpenStreetMap road network, and Baidu Map boundary), and a deep learning algorithm. A sequence of combination experiments and ablation experiments were designed for performance validation and for quantifying the contributions of individual crowd-sourced geospatial data to UGS social functional classification. The results showed that the proposed methodological framework can precisely and effectively map UGS social functional zones and that all kinds of crowd-sourced geospatial data contributed to improving the accuracy of UGS social functional classification. This study can assist planners and government departments in the rapid monitoring, effective management, and scientific planning of UGS social functional zones by providing accurate data sources and an effective mapping tool.

Purpose – The aim of this work is to research and design an expert diagnosis system for rail vehicle driven by data mechanism models. Design/methodology/approach – The expert diagnosis system utilizes statistical and deep learning methods to model the real-time status and historical data features of rail vehicle. Based on data mechanism models, it predicts the lifespan of key components, evaluates the health status of the vehicle and achieves intelligent monitoring and diagnosis of rail vehicle. Findings – The actual operation effect of this system shows that it has improved the intelligent level of the rail vehicle monitoring system, which helps operators to monitor the operation of vehicle online, predict potential risks and faults of vehicle and ensure the smooth and safe operation of vehicle. Originality/value – This system improves the efficiency of rail vehicle operation, scheduling and maintenance through intelligent monitoring and diagnosis of rail vehicle.

Layered Bi2MoO6/Bi2WO6 composites were prepared by a one pot hydrothermal method and Ag3PO4 was deposited on its surface through in-situ growth with it as a supporter. A new type of Bi2MoO6/Bi2WO6/Ag3PO4 composite dual Z-scheme heterojunction photocatalyst was designed. The catalytic activity of the composite catalyst for the degradation of RhB under visible light was investigated. Experiments showed that when the mass fraction of Ag3PO4 in the complex was 5%, the composite catalysts exhibited optimal photocatalytic activity under visible light, with a degradation rate of 99.15% for RhB within 120 minutes. The synergistic effect of Bi2MoO6, Bi2WO6, and Ag3PO4 effectively improved the absorption ability of visible light. The constructed double Z-scheme heterojunction could change the transmission path strength of photo generated electrons, which promoting the separation of photo generated electron holes. As a result, significant photocatalytic activity was obtained. Based on capture experiments and band theory analysis, a dual Z-scheme photocatalytic mechanism was proposed.

Abstract Hybrid immunity against SARS-CoV-2 has not been well studied in pregnancy. We conducted a comprehensive analysis of neutralizing antibodies (nAb) and binding antibodies in pregnant individuals who received mRNA vaccination, natural infection, or both. A third vaccine dose augmented nAb levels compared to the two-dose regimen or natural infection alone; this effect was more pronounced in hybrid immunity. There was reduced anti-Omicron nAb, but the maternal-fetal transfer efficiency remained comparable to that of other variants. Vaccine-induced nAbs were transferred more efficiently than infection-induced nAbs. Anti-spike receptor binding domain (RBD) IgG was associated with nAb against wild-type (Wuhan-Hu-1) following breakthrough infection. Both vaccination and infection-induced anti-RBD IgA, which was more durable than anti-nucleocapsid IgA. IgA response was attenuated in pregnancy compared to non-pregnant controls. These data provide additional evidence of augmentation of humoral immune responses in hybrid immunity in pregnancy.

Abstract Human enteroviruses are highly prevalent world-wide. Up to more than 100 subtypes of enteroviruses can cause several diseases, including encephalitis, meningitis, myocarditis, hand-foot-mouth disease, conjunctivitis, respiratory diseases, and gastrointestinal diseases, thus posing a great threat to human health. This study aimed to investigate the epidemiological characteristics of enterovirus in children in Hangzhou, China before and after the COVID-19 outbreak. Systematic monitoring of enterovirus infections was performed by collecting samples from the children admitted to the inpatient wards and outpatient departments in the Children’s Hospital, Zhejiang University School of Medicine, between January 2019 and May 2023. A commercial real-time RT PCR kit was utilized to detect enteroviruses. Among the 34,152 samples collected, 1162 samples, accounting for 3.4% of the samples, were tested positive for enteroviruses. The annual positive rates of the enteroviruses were 5.46%, 1.15%, 4.43%, 1.62%, and 1.96% in 2019, 2020, 2021, 2022, and May 2023, respectively. The positivity rate of the enteroviruses was highest among children aged 3–5 years and 5–7 years. Moreover, the monthly positivity rate of enterovirus infection ranged from 0.32% to 10.38%, with a peak in June and July. Serotypes, especially EV71 and CA16, causing severe symptoms such as HFMD, were decreasing, while the proportion of unidentified serotypes was on the rise. The incidence of enteroviruses in Hangzhou was higher in children aged 1–3 years and 7–18 years.

Objective Summarize the clinical and genetic characteristics of atypical Rett syndrome and developmental epileptic encephalopathy caused by IQSEC2 gene variation. Methods and Results From May 2020 to April 2022, Anhui Provincial Children's Hospital diagnosed and treated 4 children with atypical Rett syndrome and developmental epileptic encephalopathy caused by IQSEC2 gene variation, including 2 males and 2 females were a pair of identical twins. They all had comprehensive developmental delay before onset. At the age of 2 years, all cases gradually exhibited clinical manifestations of atypical Rett syndrome, such as frequent clapping, biting, sleep disorders (increased sleep or difficulty falling asleep), and grinding teeth, followed by developmental regression and seizures. The initial age of epilepsy was from 2 years and 2 months to 2 years and 10 months. All cases started with generalized tonic-clonic seizure, with epileptic spasm occurring between 2 and 11 months of course. Case 2, Case 3 and Case 4 were also accompanied by focal seizures. Four cases with VEEG background of 4-6 Hz θ wave, the VEEG during the interictal phase was a broad multifocal sharp slow complex wave. In Case 2, Case 3 and Case 4,MRI was abnormal, mainly with increased depth of cerebral hemispheric sulcus and gyrus. The whole exome sequencing suggested pathogenicity and possible pathogenic variations in the IQSEC2 gene, Case 1 and Case 2 were frameshift mutations of c. 608dup (p. Gln204Profs*35), while Case 3 and Case 4 were nonsense mutations of c.2231C > A (p.Ser744Ter) and c.2521C > T (p.Gln841Ter), respectively. The four mutation sites have not been reported domestically or internationally. All cases received treatment with multiple antiepileptic seizure medicine. The last follow-up age was from 4 years and 3 months to 6 years and 3 months. All cases were unable to walk alone and had no active language. There were no seizures in Case 1 for 3 years, occasional seizures in Case 2 and Case 4, and frequent seizures in Case 3. Conclusions IQSEC2 gene variation can manifest as atypical Rett syndrome, which can be accompanied by refractory epileptic spasms. Both males and females have severe phenotypes, and the severity of clinical phenotypes at the same mutation site varies. Our report enriches the variation spectrum and clinical phenotype spectrum of the IQSEC2 gene, expands the genetic spectrum of Rett syndrome and developmental epileptic encephalopathy, and provides value for the clinical diagnosis, treatment and subsequent research of this disease.

Abstract. Interpreting genes of interest is essential for identifying molecular mechanisms, but acquiring such information typically involves tedious manual retrieval. To streamline this process, the fanyi package offers tools to retrieve gene information from sources like National Center for Biotechnology Information (NCBI), significantly enhancing accessibility. Additionally, understanding the latest research advancements and sharing achievements are crucial for junior researchers. However, language barriers often restrict knowledge absorption and career development. To address these challenges, we developed the fanyi package, which leverages artificial intelligence (AI)-driven online translation services to accurately translate among multiple languages. This dual functionality allows researchers to quickly capture and comprehend information, promotes a multilingual environment, and fosters innovation in academic community. Abstract Bone marrow mesenchymal stem cell-derived exosomes (BMSC-Exos) have been shown to promote angiogenesis after ischemic stroke, in which microRNAs (miRs) are believed to play an important role in exosome-mediated therapeutic effects, though the mechanism is still not clear. In this study, a series of molecular biological and cellular assays, both in vitro and in vivo, were performed to elucidate the role of exosomal miR-486 in angiogenesis following cerebral ischemic and its molecular mechanisms. Our results revealed that BMSC-Exos significantly improved neurological function and increased microvessel density in ischemic stroke rats. In vitro assays showed that BMSC-Exos promoted the proliferation, migration, and tube formation ability of oxygen–glucose deprivation/reoxygenation (OGD/R) injured rat brain microvascular endothelial cells (RBMECs). Importantly, BMSC-Exos increased the expression of miR-486 and phosphorylated protein kinase B (p-Akt) and down-regulated the protein level of phosphatase and tensin homolog (PTEN) in vivo and in vitro. Mechanistic studies demonstrated that transfection with miR-486 mimic enhanced RBMECs angiogenesis and increased p-Akt expression, while inhibited PTEN expression. On the other hand, the miR-486 inhibitor induced an opposite effect, which could be blocked by PTEN siRNA. It was thus concluded that exosomal miR-486 from BMSCs may enhance the functional recovery by promoting angiogenesis following cerebral ischemic injury, which might be related to its regulation of the PTEN/Akt pathway.

Abstract Background Recognizing the predictors of poor short-term prognosis after first-line immunotherapy in patients with anti-N-methyl-D-aspartate receptor (anti-NMDAR) encephalitis is essential for individualized treatment strategy. The objective of this study was to ascertain the factors that forecast short-term prognosis in patients with anti-NMDAR encephalitis, develop a prognostic prediction model, and authenticate its efficacy in an external validation cohort. Further, all patients were followed-up long-term to assess the factors of long-term outcome and relapses. Methods A prospective enrollment of patients diagnosed with anti-NMDAR encephalitis was conducted across five clinical centers in China from June 2014 to Mar 2022. The enrolled patients were divided into the derivation and validation sets based on enrollment time. The short-term prognostic model was visualized using a nomogram. Further, all patients were followed-up long-term to assess the factors of long-term outcome. Abstract Cross-linked polymers with covalent adaptable networks (CANs) can be reprocessed under external stimuli owing to the exchangeability of dynamic covalent bonds. Optimization of reprocessing conditions is critical since increasing the reprocessing temperature costs more energy and even deteriorates the materials, while reducing the reprocessing temperature via molecular design usually narrows the service temperature range. Exploiting CO2 gas as an external trigger for lowering the reprocessing barrier shows great promise in low sample contamination and environmental friendliness. Herein, we develop a type of CANs incorporated with ionic clusters that achieve CO2-facilitated recyclability without sacrificing performance. The presence of CO2 can facilitate the rearrangement of ionic clusters, thus promoting the exchange of dynamic bonds. The effective stress relaxation and network rearrangement enable the system with rapid recycling under CO2 while retaining excellent mechanical performance in working conditions. This work opens avenues to design recyclable polymer materials with tunable dynamics and responsive recyclability.
