Introduction Patients with clinically significant portal hypertension (CSPH) are recommended to be treated with non-selective beta-blockers (ie, carvedilol) to prevent the first hepatic decompensation event by the renewing Baveno VII consensus. CSPH is defined by hepatic venous pressure gradient (HVPG)≥10 mm Hg; however, the HVPG measurement is not widely adopted due to its invasiveness. Liver stiffness (LS)≥25 kPa can be used as a surrogate of HVPG≥10 mm Hg to rule in CSPH with 90% of the positive predicting value in majority aetiologies of patients. A compelling argument is existing for using LS≥25 kPa to diagnose CSPH and then to initiate carvedilol in patients with compensated cirrhosis, and about 5%–6% of patients under this diagnosis criteria may not be benefited from carvedilol and are at risk of lower heart rate and mean arterial pressure. Randomised controlled trial on the use of carvedilol to prevent liver decompensation in CSPH diagnosed by LS remains to elucidate. Therefore, we aimed to investigate if compensated cirrhosis patients with LS≥25 kPa may benefit from carvedilol therapy.Methods and analysis This study is a randomised, double-blind, placebo-controlled, multicentre trial. We will randomly assign 446 adult compensated cirrhosis patients with LS≥25 kPa and without any previous decompensated event and without high-risk gastro-oesophageal varices. Patients are randomly divided into two groups, with 223 subjects in group A and 223 subjects in group B. Group A is a carvedilol intervention group, while group B is a placebo group. All patients in both groups will receive aetiology therapies and are followed up at an interval of 6 months. The 3-year incidences of decompensated events of cirrhosis-related and liver-related death are the primary outcome. The secondary outcomes include development of each complication of portal hypertension individually (ascites, variceal bleeding or overt hepatic encephalopathy), development of spontaneous bacterial peritonitis and other bacterial infections, development of new varices, growth of small varices to large varices, delta changes in LS and spleen stiffness, change in hepatic dysfunction assessed by Child-Pugh and model for end-stage liver disease score, change in platelet count, development of hepatocellular carcinoma, development of portal vein thrombosis and adverse events with a 3-year follow-up. A predefined interim analysis will be performed to ensure that the calculation is reasonable.Ethics and dissemination The study protocol has been approved by the ethics committees of the Sixth People’s Hospital of Shenyang (2023-05-003-01) and independent ethics committee for clinical research of Zhongda Hospital, affiliated to Southeast University (2023ZDSYLL433-P01). The results from this trial will be submitted for publication in peer-reviewed journals and will be presented at international conferences.Trial registration number ChiCTR2300073864.

Seasonal reproduction is a strategy used by many marine fish to adapt to environmental changes. However, the composition of germ cells and environmental cues throughout the reproductive cycle in seasonal breeding fish is not well understood. This study investigated the gonadal development of Nibea albiflora at two ages during its annual reproductive cycle. We observed similar testicular development in 1-year-old and 2-year-old males, with two spermiation periods in spring and autumn. In contrast, 1-year-old females showed non-seasonal ovarian regression, while 2-year-old females had seasonally regulated development, with extended spawning from spring to autumn. Additionally, our correlation analysis between temperature and gonadal development revealed a close association of seasonal gametogenesis with water temperature. Notably, both testes and ovaries stagnated at the regression stage at the lowest winter temperature (10 ℃), resuming gametogenesis as temperatures increased in spring. Moreover, we analyzed and found that the expression levels of vasa, a specific marker for early germ cells, were seasonally regulated with peak levels in spring for the testis and in winter for the ovary. These findings enhance our understanding of germ cell activity under the seasonal reproductive cycle in teleosts, and provide a basis for the aquaculture and conservation management of N. albiflora.

With the ultra-thin thickness and excellent performance, metalens are bi-dimensional optical elements composed of nanopatterned structures that demonstrate significant potential for various applications such as holography, planar imaging and tomography. However, the small size of metalens makes it challenging to precisely quantify their wavefront aberration, thus impeding their broader utilization. To address this challenge, we propose an innovative method for measuring wavefront aberration in metalens using phase-shift interferometry. Furthermore, we have developed the Small Aperture Interferometric Imaging Detection System with variable fields of view, specifically designed for metalens. This system can accurately measure the wavefront aberration of metalens with different fields of view with PV accuracy of 1/20λ, further enabling the acquisition of imaging performance parameters, including Modulation Transfer Function (MTF) and Point Spread Function (PSF). This innovative measurement approach not only enables quantitative evaluation of performance of metalens but also assists designers and manufacturers in optimizing process flows and identifying potential defects. As a result, this approach aids in promoting the industrial application of metalens.

Abstract Recovery of skin function remains a significant clinical challenge for deep burns owing to the severe scar formation and poor appendage regeneration, and stem cell therapy has shown great potential for injured tissue regeneration. Here, a cell‐free therapy system for deep burn skin was explored using mesenchymal stem cell paracrine proteins (MSC‐PP) and polyethylene glycol (PEG) temperature‐sensitive hydrogels. A three‐dimensional (3D) dynamic culture system for MSCs' large‐scale expansion was established using a porous gelatin microcarrier crosslinked with hyaluronic acid (PGM‐HA), and the purified MSC‐PP from culture supernatant was characterized by mass spectrometric analysis. The results showed the 3D dynamic culture system regulated MSCs cell cycle, reduced apoptosis, and decreased lactic acid content, and the MSC‐PP produced in 3D group can promote cell proliferation, migration, and adhesion. The MSC‐PP + PEG system maintained stable release in 28 days of observation in vitro. The in vivo therapeutic efficacy was investigated in the rabbit's third‐degree burn model, and saline, PEG, MSC‐PP, and MSC‐PP + PEG treatments groups were set. The in vivo results showed that the MSC‐PP + PEG group significantly improved wound healing, inhibited scar formation, and facilitated skin appendage regeneration. In conclusion, the MSC‐PP + PEG sustained‐release system provides a potentially effective treatment for deep burn skin healing.

Diseases are a significant impediment to aquaculture’s sustainable and healthy growth. The aquaculture industry is suffering significant financial losses as a result of the worsening water quality and increasing frequency of aquatic disease outbreaks caused by the expansion of aquaculture. Drug control, immunoprophylaxis, ecologically integrated control, etc. are the principal control strategies for fish infections. For a long time, the prevention and control of aquatic diseases have mainly relied on the use of various antibiotics and chemical drugs. However, long-term use of chemical inputs not only increases pathogenic bacteria resistance but also damages the fish and aquaculture environments, resulting in drug residues in aquatic products, severely impeding the development of the aquaculture industry. The development and use of aquatic vaccines are the safest and most effective ways to prevent aquatic animal diseases and preserve the health and sustainability of aquaculture. To give references for the development and implementation of aquatic vaccines, this study reviews the development history, types, inoculation techniques, mechanisms of action, development prospects, and challenges encountered with aquatic vaccines.

Granzymes (Gzm) are serine proteases, contained into the secretory granules of cytotoxic cells, responsible for the cell-mediated cytotoxicity (CMC) against tumor cells and intracellular pathogens such as virus and bacteria. In fish, they have received little attention to their existence, classification or functional characterization. Therefore, we aimed to identify and evaluate their functional and transcriptomic relevance in the innate CMC activity of two relevant teleost fish species, gilthead seabream and European sea bass. Afterwards, we wanted to focus on their regulation upon nodavirus (NNV) infection, a virus that causes great mortalities to sea bass specimens while seabream is resistant. In this study, we have identified genes encoding GzmA and GzmB in both seabream and sea bass, as well as GzmM in seabream, which showed good phylogenetic relation to their mammalian orthologs. In addition, we found enzymatic activity related to tryptase (GzmA and/or GzmK), aspartase (GzmB), metase (GzmM), or chymase (GzmH) in resting head-kidney leucocytes (HKLs), with the following order of activity: GzmA/K ~ GzmM >> GzmH >>> GzmB. In addition, during innate CMC assays consisting on HKLs exposed to either mock- or NNV-infected target cells, though all the granzyme transcripts were increased only the tryptase activity did. Thus, our data suggest a high functional activity of GzmA/K in the innate CMC and a marginal one for GzmB. Moreover, GzmB activity was detected into target cells during the CMC assays. However, the percentage of target cells with GzmB activity after the CMC assays was about 10-fold lower than the death target cells, demonstrating that GzmB is not the main inductor of cell death. Moreover, in in vivo infection with NNV, gzm transcription is differently regulated depending on the fish species, genes and tissues. However, the immunohistochemistry study revealed an increased number of GzmB stained cells and areas in the brain of seabream after NNV infection, which was mainly associated with the lesions detected. Further studies are needed to ascertain the molecular nature, biological function and implication of fish granzymes in the CMC activity, and in the antiviral defense in particular.

With the improvement of China’s electrification level, the relationship between electricity power consumption and social economy development is getting closer. Under this background, in this paper, the business cycle analysis method is applied to the electricity power industry in Hebei Province, and the link between electric power generation and economic development is determined through the prosperity index. First of all, the steps and methods of business cycle analysis were introduced, and the time-difference grey correlation method considering the consistency of time series to screen indicators was put forward, thus to reduce the number of indicators. Next, Hebei Province was taken as an example, and its monthly data related to power generation and economic growth in the 10 years from 2009 to 2018 to was selected to build the business cycle indicator system, at last the composite prosperity index of Hebei Province in electric power industry was calculated. This research results manifest that it is feasible to apply the business cycle analysis to the power industry.

Monitoring the characteristic components in tipping paper is important to ensure the quality of cigarette products. This study establishes a method based on gas chromatography (GC) for determining cooling agents in tipping paper. The tipping paper was cut into pieces and extracted by ethanol under specific temperatures in a shaker. Then, the characteristic cooling agents of (-)-menthone, L-menthol, and (-)-menthyl lactate in extract were determined using GC coupled with a flame ionization detector (GC-FID). The limits of detection (LODs) and limits of quantification (LOQs) for the three cooling agents ranged from 0.15~0.32 [micro]g/mL and 0.49~1.06 [micro]g/mL, respectively. Results demonstrated a good linear relationship with high correlation coefficients for the three tested cooling agents. The extraction conditions of the cooling agents were optimized through a single-factor experiment as well as an orthogonal experiment. As the second most widely consumed eggs, duck eggs are made into preserved eggs, salted duck eggs, and roasted duck eggs to extend their shelf-life. To investigate the differences in potent odorants (POs) between salted duck egg yolk (SDEY) and roasted duck egg yolk (RDEY), the volatiles in SDEY and RDEY were extracted through solvent extraction coupled with solvent-assisted flavor evaporation and were assayed with gas chromatography-mass spectrometry-olfactometry. A total of 45 volatiles were identified in two samples, 24 odor-active compounds (OACs) were screened, and more OACs were in RDEY. The flavor-dilution (FD) factors of OACs were obtained by aroma extract dilution analysis and ranged from 3 to 6561. Twenty-two OACs with FD factors ≥ 9 were quantitated, and the results indicated the concentrations of OACs in yolk increased greatly after salted duck eggs were roasted. Based on the concentrations and thresholds, odor activity values (OAVs) were determined; 17 odorants with OAVs ≥ 1 were determined as POs. Acetoin was the most PO in SDEY; there were more POs in RDEY, including 2-ethyl-3,6-dimethylpyrazine, acetoin, 2-acetyl-3-methylthiophene, dihydro-4-hydroxy-2(3H)-furanone, etc. The outcomes obtained have reference values for making better use of duck eggs in the food industry.
