This study demonstrates the feasibility of extracting lecithin from oil industry byproducts in an eco‐friendly manner, with minimal use of water and without harmful chemicals. Liposomes can be generated directly from this extracted lecithin, enhancing the value of these byproducts and enabling the production of catalytic gold nanoparticles (AuNPs). Thin‐layer chromatography of the extracted lecithin revealed a phospholipid composition primarily consisting of phosphatidylethanolamine and phosphatidylcholine, and surface tension studies demonstrated similar behavior between the extracted and commercial lecithin. Liposome formation using sustainable lecithin (LPn) resulted in structures that were stable for at least 10 days, exhibiting a low polydispersity index (0.395) and uniform size (approximately 214 ± 7 nm). Gold nanoparticles were synthesized successfully in LPn loaded with [HAuCl4] by using different photoreduction methods: ultraviolet (UV) lamp, pulsed laser 355 nm, and sunlight irradiation. The AuNPs exhibited characteristic sizes (ranging from 5.03 to 6.78 nm) and optical properties typical of nanoparticles, including a distinct surface plasmon resonance. As a proof of concept, we also demonstrated that the synthesized AuNPs exhibited catalytic activity in UV‐induced cis‐trans isomerization reactions. Aim Materials and Methods Results Conclusions The aim was to investigate the causal relationship between puberty timing and plasma metabolites, accounting for birth weight, childhood and adulthood adiposity.The meta‐analysis of genome‐wide association studies (GWAS) for puberty timing was extracted from the ReproGen Consortium, involving 329 345 women of European ancestry. Summary data for 174 plasma metabolites were retrieved from a recently conducted cross‐platform GWAS that involved a meta‐analysis of three cohort studies (i.e. the Fenland, European Prospective Investigation into Cancer‐Norfolk and INTERVAL studies) and three publicly available studies and included up to 86 507 participants. We conducted a two‐sample Mendelian randomization (MR) analysis to infer the causal relationship of puberty timing on 174 plasma metabolites, complemented by a two‐step and multivariable Mendelian randomization (MVMR) analysis to assess direct and indirect effects. Additionally, summary‐level data from the UK Biobank were used for our replication analysis.The results of the two‐sample MR provide moderate evidence supporting a causal relationship between puberty timing and 23 of 174 plasma metabolites (i.e. 7 acylcarnitines, 8 amino acids, 2 biogenic amines and 6 lysophosphatidylcholines). Even after single‐nucleotide polymorphisms associated with birth weight and childhood adiposity were excluded, causal effects persisted for 16 metabolites (i.e. 8 acylcarnitines, 4 amino acids, 2 biogenic amines and 2 lysophosphatidylcholines). The two‐step MR analysis provided evidence that the relationship between puberty timing and plasma metabolites was mediated by adulthood adiposity. Additionally, moderate evidence emerged for an independent causal effect of puberty timing on 10 metabolites through an MVMR analysis (i.e. 5 acylcarnitines, 2 amino acids, 1 biogenic amine, 1 lysophosphatidylcholine and 1 phosphatidylcholine). Furthermore, the replication analysis suggested the robustness of our results.In summary, our study provides compelling evidence that puberty timing has a causal influence on certain plasma metabolites, although this influence is largely mediated by adulthood adiposity. Background: Slow transit constipation (STC) is highly prevalent and has rising incidence. Shouhui Tongbian capsule (SHTB) is a traditional Chinese Medicine formula with extensive and highly efficacious usage in STC treatment, however, its mechanism of action, especially the regulation of microbiome and lipid metabolites, remains unclear. Methods: After quality control of SHTB using LC‒MS to obtain its material basis, we tried to elucidate the cohesive modulatory network of SHTB against STC using hyphenated methods from microbiomics, lipidomics, mass spectrometry imaging (MSI) and molecular methods. Results: SHTB could repair intestinal barrier damage, reduce systemic inflammation and increase intestinal motility in a diphenoxylate-induced STC rat model. Based on 16S rDNA sequencing results, SHTB rehabilitated the abnormal changes in Alloprevotella, Coprococcus, Marvinbryantia, etc., which were associated with STC symptoms. Meanwhile, microbial functional prediction showed that lipid metabolism was improved with SHTB administration. The differential lipids, including fatty acids, lysophosphatidylcholine, phosphatidylcholine, sphingomyelin triglyceride and ceramide, that are closely related to STC disease and SHTB efficacy. Furthermore, SHTB significantly reversed the abnormal expression of these key target enzymes in colon samples, including CTP-phosphocholine cytidylyltransferase, CTP-phosphoethanolamine cytidylyltransferase, phosphatidic acid phosphatase, acid sphingomyelinase etc. Conclusions: Combined analysis demonstrated that SHTB reducing lipid accumulation and recovery of intestinal microbial homeostasis was the critical mechanism by which SHTB treats STC. 3D hierarchical superstructures (3DHSs) are key products of nature's evolution and have raised wide interest. However, the preparation of 3DHSs composed of building blocks with different structures is rarely reported, and regulating their structural parameters is challenging. Herein, a simple lecithin‐mediated biomineralization approach is reported for the first time to prepare gold 3DHSs composed of 0D nucleus and 1D protruding dendritic spikes. It is demonstrated that a hydrophobic complex by coordination of disulfiram (DSF) with a share of chloroauric acid is the key to forming the 3DHSs. Under the lecithin mediation, chloroauric acid is first reduced to form the 0D nucleus, followed by the spike growth through the reduction of the hydrophobic complex. The prepared 3DHSs possess well‐defined morphology with a spike length of ≈95 nm. Notably, the hierarchical spike density is systematically manipulated from 38.9% to 74.3% by controlling DSF concentrations. Moreover, the spike diameter is regulated from 9.2 to 12.9 nm by selecting different lecithin concentrations to tune the biomineralization process. Finite‐difference time‐domain (FDTD) simulations reveal that the spikes form “hot spots”. Phosphatidylethanolamine N-methyltransferase (PmtA) catalyzes the biosynthesis of phosphatidylcholine (PC) from phosphatidylethanolamine (PE). Although PC is one of the major phospholipids constituting bilayer membranes in eukaryotes, certain bacterial species encode PmtA, a membrane-associated methyltransferase, to produce PC, which is correlated with cellular stress responses, adaptability to environmental changes, and symbiosis or virulence with eukaryotic hosts. Depending on the organism, multiple PmtAs may be required for producing monomethyl-and dimethyl-PE derivatives along with PC, whereas in organisms such as Rubellimicrobium thermophilum, a single enzyme is sufficient to direct all three methylation steps. In this study, we present the x-ray crystal structures of PmtA from R. thermophilum in complex with dimethyl-PE and S-adenosyl-L-homocysteine, as well as in its lipid-free form. Moreover, we demonstrate that the enzyme associates with the cellular membrane via electrostatic interactions facilitated by a group of critical basic residues and can successively methylate PE and its methylated derivatives, culminating in the production of PC. Introduction: This study aimed to investigate the causal relationship between phosphatidylcholine (PC) levels and dysmenorrhea using Mendelian randomization (MR) analysis. Methods: Weconducted a two-sample MRanalysis using GWAS data on PC levels and dysmenorrhea. Single nucleotide polymorphisms (SNPs) associated with PC levels were used as instrumental variables. MR-Egger regression and inverse variance weighting (IVW) were used to estimate the causal effect of PC levels on dysmenorrhea. Sensitivity analyses were performed to assess the robustness of the results. Results: The IVW analysis revealed a significant positive association between higher PC levels and dysmenorrhea (OR: 1.533, 95% CI: 1.039-2.262, P = 0.031). The MR-Egger regression did not detect pleiotropy. Sensitivity analyses confirmed the robustness of the results. Conclusion: This study provides evidence suggesting a causal link between increased PC levels and dysmenorrhea. In this study, the effects of emulsifiers such as lecithin, AMPs, Palsgaard® Oil‐Binder and GMS on cocoa hazelnut spread rheology were compared under the same process conditions and formulation. Emulsifiers were added to the formulation separately at rates of 0.3%–0.4%–0.5%. Hardness values in cocoa hazelnut spread were examined at 15‐day intervals until the 60th day. In addition, viscosity, rheological analyses, color, spreadability, stability tests, and sensory analyses were performed. In the production of cocoa hazelnut spread, lecithin and AMP have less hardness and lower viscosity, greater fluent consistency, better spreadability, and lower "work of shear" values compared with other emulsifiers. The emulsifier type/ratio difference did not affect the color value statistically. It was determined that the use of Oil‐Binder and GMS significantly protected the stability compared with other emulsifiers. During the 60‐day storage period, lecithin preserved its hardness properties better than other emulsifiers. When sensory properties were examined, the use of lecithin and AMP in cocoa hazelnut spread samples scored high in brightness, spreadability, mouthfeel, and taste parameters. Summary: The study explored the combination effect between of green tea extract (GTE) and black rice extract (BRE) and the encapsulation process of the co‐extract. A 2:1 (w/w) ratio of GTE to BRE in citric aqueous solution at pH 6 showed the highest antioxidant activity (IC50 = 0.306 mg mL−1) with the strongest incremental effect (combination index = 0.547). It exhibited potent anti‐inflammatory effects, with sirtuin 1 enzyme‐stimulating activity (28.593 of fluorescent intensity) and NF‐κB protein suppression in RAW 264.7 macrophages (57.76% at 100 μg mL−1). Freeze‐dried double emulsion encapsulated powder (CEP) with the highest encapsulation efficiency (73.38%) and total polyphenol content (218.28 mg GAE g−1) was prepared using a lecithin to O phase ratio of 2:25 (w/w) and a chitosan‐carboxymethyl cellulose to W1/O emulsion phase ratio of 1:1 (w/w). Summary: This study delves into the influence of soy lecithin addition aimed at augmenting the structural recovery and enhancing textural similarities between protein oleogels and solid fats. The co‐oleogelation of soy protein isolate (SPI) and xanthan gum (XG) with lecithin was explored to structure sunflower oil. Oleogels incorporating lecithin concentrations ranging from 4% to 12% were examined for their textural properties and structural recovery performance. Results indicate that these oleogel formulations exhibit comparable textural properties to solid fats, signifying the potential of lecithin as a co‐oleogelator. Moreover, the incorporation of lecithin enhanced the structural recovery of the SPI‐XG oleogel, leading to no oil release when put under pressure, further emphasising its role in improving the overall quality of protein‐based oleogel systems. This study aimed to optimize a liposomal formulation to enhance the stability of a phenolic-rich extract from green walnut husk. Liposomes were prepared using varying concentrations of phosphatidylcholine (0.15–2% w/v), extract (0–1.3% v/v), and sodium laurate (0–0.2% w/v) via ethanol injection and sonication. Characterization included visual appearance, particle size, polydispersity index, surface charge, encapsulation efficiency, and morphology. Stable liposomes were achieved at 0.15% and 0.3% w/v phosphatidylcholine, although with low encapsulation efficiency (<40%). The addition of 0.2% (w/v) sodium laurate improved the stability, especially at higher phosphatidylcholine concentrations, enhancing the electrostatic repulsion. Optimal concentrations of 2% w/v phosphatidylcholine, 0.2% w/v sodium laurate, and 0.6% v/v extract were determined. The liposomes exhibited a spherical unilamellar morphology with a size of 97.5 ± 0.9 nm and a negative surface charge of –39.8 ± 0.9 mV. These nanoliposomes showed 79.7 ± 0.7% encapsulation efficiency and remained stable under pH, temperature, ionic strength, and storage time variations. Background: Observational studies have suggested that plasma lipidome play a pivotal role in the occurrence of Parkinson's disease (PD). However, it remains unknown which lipids among plasma lipidome affect PD and how they exert their influence. Clarity is lacking regarding the causal relationship between plasma lipidome and PD, as well as whether circulating inflammatory proteins serve as mediators. Methods: Single nucleotide polymorphisms (SNPs) significantly associated with 179 plasma lipidome were selected as instrumental variables to assess their causal impact on PD. PD data, serving as the outcome, were sourced from the International Parkinson's Disease Genomics Consortium, which boasts the largest sample size to date. The inverse variance weighted (IVW), Weighted median method, MR-Egger method, Simple mode method, Weighted mode method and MR-PRESSO were employed to evaluate the influence of the 179 plasma lipidome on PD. Heterogeneity, pleiotropy tests, and reverse causality analyses were conducted accordingly. Additionally, we analyzed the causal relationship between 91 circulating inflammatory proteins and PD, exploring whether these proteins serve as mediators in the pathway from plasma lipidome to PD. Results: Among the 179 plasma lipidome, three were found to be associated with a reduced risk of PD: Phosphatidylcholine (14:0_18:2) (IVW, OR = 0.877; 95%CI, 0.787-0.978; p = 0.018), Phosphatidylcholine (16:0_16:1) levels (IVW, OR = 0.835; 95%CI, 0.717-0.973; p = 0.021), and Phosphatidylcholine (O-17:0_17:1) levels (IVW, OR = 0.854; 95%CI, 0.779-0.936; p = 0.001). Meanwhile, Sphingomyelin (d38:1) was linked to an increased risk of PD (IVW, OR = 1.095; 95%CI, 1.027-1.166; p = 0.005). Among the 91 circulating inflammatory proteins, three were associated with a lower PD risk: Fibroblast growth factor 21 levels (IVW, OR = 0.817; 95%CI, 0.674-0.990; p = 0.039), Transforming growth factor-alpha levels (IVW, OR = 0.825; 95%CI, 0.683-0.998; p = 0.048), and Tumor necrosis factor receptor superfamily member 9 levels (IVW, OR = 0.846; 95%CI, 0.744-0.963; p = 0.011). Two were associated with a higher risk of PD: Interleukin-17A levels (IVW, OR = 1.285; 95%CI, 1.051-1.571; p = 0.014) and TNF-beta levels (IVW, OR = 1.088; 95%CI, 1.010-1.171; p = 0.026). Additionally, a positive correlation was observed between Phosphatidylcholine (14:0_18:2) levels and Fibroblast growth factor 21 levels (IVW, OR = 1.125; 95%CI, 1.006-1.257; p = 0.038), suggesting that Fibroblast growth factor 21 levels may serve as a mediating factor in the pathway between Phosphatidylcholine (14.0_18.2) levels and PD. The mediation effect was estimated to be -0.024, accounting for approximately 18% of the total effect. Conclusion: Both plasma lipidome and circulating inflammatory proteins demonstrate a causal relationship with PD. Additionally, circulating inflammatory proteins may serve as mediators in the pathway from plasma lipidome to PD. Phosphatidylinositol is a precursor of various phosphoinositides, which play crucial roles in intracellular signaling and membrane dynamics and have impact on diverse aspects of cell physiology. Phosphoinositide synthesis and turnover occur in the cytoplasmic leaflet of the organellar and plasma membranes. P4-ATPases (lipid flippases) are responsible for translocating membrane lipids from the exoplasmic (luminal) to the cytoplasmic leaflet, thereby regulating membrane asymmetry. However, the mechanism underlying phosphatidylinositol translocation across cellular membranes remains elusive. Here, we discovered that the phosphatidylcholine flippases ATP8B1, ATP8B2, and ATP10A can also translocate phosphatidylinositol at the plasma membrane. To explore the function of these phosphatidylinositol flippases, we used cells depleted of CDC50A, a protein necessary for P4-ATPase function and ATP8B1 and ATP8B2, which express in HeLa cells. Upon activation of the Gq-coupled receptor, depletion of phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2] was accelerated in CDC50A knockout (KO) and ATP8B1/8B2 double KO cells compared with control cells, suggesting a decrease in PtdIns(4,5)P2 levels within the plasma membrane of the KO cells upon stimulation. These findings highlight the important role of P4-ATPases in maintaining phosphoinositide homeostasis and suggest a mechanism for asymmetry of phosphatidylinositol in the cytoplasmic leaflet of the plasma membrane. Objective: To evaluate the causal relationship between lipids and ulcerative colitis (UC) through Mendelian Randomization (MR), and to further investigate the involvement of immune cells in mediating this process. Methods: Utilizing summary statistics from genome-wide association studies (GWAS) of individuals with European ancestry, we analyzed the causal link between 179 lipid types and UC (2,569 UC cases and 453,779 controls) through Two-sample Mendelian randomization (2SMR) and Bayesian-weighted MR (BWMR). Based on this, a mediation screening of 731 immune cell phenotypes was conducted to identify exposure and mediator factors. Lastly, the role and proportion of immune cells in mediating the causal effects of lipids on UC were assessed via reverse MR (RMR) and two-step MR. Results: The results of MR showed that there was a causal relationship between the six genetically predicted lipid types and UC (P <0.05), and the four immune cell phenotypes were identified as mediators of the association between lipids and UC. Notably, Phosphatidylcholine (PC) (16:0_0:0) served as the exposure factor, and myeloid cells CD11b on CD33+ HLA DR+ CD14dim acted as the mediator. Mediation analysis showed that CD11b on CD33+ HLA DR+ CD14dim had a mediation effect of -0.0205 between PC (16:0_0:0) and UC, with the mediation effect ratio at 15.38%. Conclusion: Our findings elucidate the causal effect of lipids on UC and identify the significant mediating role of myeloid cells CD11b on CD33+ HLA DR+ CD14dim in regulating UC through PC (16:0_0:0), offering new pathways and strategies for UC clinical treatment. Objective: The causal relationship and mechanisms between lipids and glioblastoma (GBM) remain unclear. This study aims to investigate the independent causal relationship between liposomal phosphatidylcholine 16:0_22:6 (PC16) and GBM, and to identify the potential mediating role of the inflammatory factor-fibroblast growth factor 21(FGF21). Methods: Utilizing summary statistics from genome-wide association studies (GWAS) of lipids (179 types in 7174 Finnish individuals), GBM (243 cases and 287137 controls), and inflammatory factors (91 types in 14824 European individuals), a two-sample Mendelian Randomization (MR) approach was employed to establish the causal link between liposomal PC16 and GBM. Additionally, a two-step MR method was used to quantify the proportion of the causal effect of PC16 on GBM that is mediated by the inflammatory factor FGF21. Results: MR analyses revealed a strong causal relationship between PC16 and GBM (OR=1.72, 95% CI: 1.11-2.68, P=0.016), but no reverse causality was observed from GBM to PC16 (OR=1.01, 95% CI: 0.99-1.02, P=0.38). Mediation analysis showed a strong causal relationship between PC16 and the FGF21 (OR = 0.94, 95% CI: 0.89-0.99, P=0.018) as well as between FGF21 and GBM (OR = 0.42, 95% CI: 0.25-0.71, P=0.001), with the mediation effect accounting for 9.78% of the total effect. This suggests that the causal relationship between PC16 and GBM is likely mediated by the intermediary factor FGF21. No evidence of pleiotropy was found in the sensitivity analysis of these positive results. Conclusion: In summary, the findings of this study suggest that liposomal PC16 may increase the risk of GBM occurrence, and FGF21 may play a significant mediating role in this causal relationship. Summary: Objective: This study identified metabolite modules associated with adiposity and body fat distribution in childhood using gold‐standard measurements. Methods: We used cross‐sectional data from 329 children at mid‐childhood (age 5.3 ± 0.3 years; BMI 15.7 ± 1.5 kg/m2) from the Genetics of Glucose regulation in Gestation and Growth (Gen3G), a prospective pre‐birth cohort. We quantified 1038 plasma metabolites and measured body composition using the gold‐standard dual‐energy x‐ray absorptiometry (DXA), in addition to skinfold, waist circumference, and BMI. We applied weighted‐correlation network analysis to identify a network of highly correlated metabolite modules. Spearman's partial correlations were applied to determine the associations of adiposity with metabolite modules and individual metabolites with false discovery rate (FDR) correction. Results: We identified a 'green' module of 120 metabolites, primarily comprised of lipids (mostly sphingomyelins and phosphatidylcholine), that showed positive correlations (all FDR p < 0.05) with DXA estimates of total and truncal fat (ρadjusted = 0.11–0.19), skinfold measures (ρadjusted = 0.09–0.26), and BMI and waist circumference (ρadjusted = 0.15 and 0.18, respectively). These correlations were similar when stratified by sex. Within this module, sphingomyelin (d18:2/14:0, d18:1/14:1)*, a sphingomyelin sub‐specie that is an important component of cell membranes, showed the strongest associations. The objective of the present study was to prepare flavonosomes loaded with kaempferol and curcumin in order to improve the bioavailability of the flavonoids. The flavonosomes loaded with kaempferol and curcumin were prepared using bulk co-loading method using lecithin as the lipid molecule. The particle size of the flavonosomes was determined using zeta sizer and ranged from 497.3 nm to 167.1 nm. The results indicated that increase the ratio of lipid to extract was able to reduce the particle size of the flavonosomes. The flavonosomes prepared using 4:1 ratio of lipid to flavonoids were found to be rigid, spherical as seen in SEM with smooth regular surface. Sharp and distinct endothermic peaks in DSC revealed the formulation of stable flavonosomes due to molecular interactions between the extract and lecithin. The entrapment efficiency of flavonosome preparation F4 was determined by measurement of the non-entrapped flavonoids and subtracting from the total flavonoids used to obtain the flavonoids entrapped in the formulation. The entrapment efficiency of F4 has been calculated to be 94.12%. The flavonosome F4 was studied for release of the entrapped flavonoids by dialysis method. The amount of flavonoids released at 1, 2, 3, 4, 6, 8, 10 and 12 hours was determined using UV spectrophotometry. After the first hour, the entrapped flavonoids released in a steady manner from the flavonosome depicting and almost complete release (90.52% for kaempferol and 94.67% for curcumin) from the formulation. The flavonosomes were found to be stable under the storage conditions. The best formulation with respect to particle size and anti-inflammatory action was F4 that contained 4:1 ratio of lecithin: flavonoids. DPPH radical scavenging assay was used to determine the antioxidant action of the individual flavonoids and the flavonosome formulation. It was found that the formulation F4 was having higher antioxidant activity in comparison to kaempferol and curcumin. The IC50 of kaempferol, curcumin and flavonosome were found to be 228.20 µg/mL, 175.51 µg/mL and 158.19 µg/mL respectively. [ABSTRACT FROM AUTHOR]

In the present study, the olive mill wastewater (OMW) phenolics were extracted with soy lecithin using the cloud point extraction method, and this enriched lecithin (OMW-L) was converted into spray-dried powders using maltodextrin (MD) and whey protein concentrate to be utilized as a dry food ingredient in a cake premix. The inlet temperature of 170 °C and a 3 mL/min feed flow rate yielded the highest powder yield (70.88 ± 2.12%) with a moisture content of 3.78 ± 0.03% when the mass ratio of lecithin to MD was 1:3 (w:w). The hydroxytyrosol and tyrosol contents of the powder were 42.60 ± 4.51 mg/100 g and 15.48 ± 2.50 mg/100 g, respectively. Vanillic acid, caffeic acid, 3-hydroxybenzoic acid, catechin, and rutin were also identified in the powders. The spray-dried OMW-L powder with a higher loading of polyphenols was then used in a cake premix, replacing 1% and 3% of wheat flour. This substitution significantly reduced the K value of the cake batter, as determined by rheological analyses. The addition of spray-dried OMW-L powder to the cake samples, particularly at higher concentrations (3%), influenced both crust and crumb color, causing changes in L*, a*, and b* values. The hardness values of the cake samples did not alter when blank or OMW-enriched lecithin powders were added; rather, the hardness value was influenced by the powder content. In-demand fresh-cut fruits are highly perishable and require shelf stability. Starch, such as sago, is a naturally available polysaccharide with good gas barrier properties. The study aimed to develop an edible coating and examine the effect of its application on the quality parameters of fresh-cut apples. The coating solution was prepared with sago and soy oil in concentrations of 3, 4, and 5% and 0, 0.25, and 0.50%, respectively. Lecithin (0.50%) was used as an emulsifier and glycerol (1.5%) as a plasticizer. Coated fresh-cut apples were evaluated for physicochemical properties (TSS, pH, non-enzymatic browning index, moisture content, weight loss, headspace gas, and color attributes) during a 12-day shelf-life study at 4 °C. Sensory analysis was also performed to assess consumer acceptability, and microbial analysis to investigate its inhibiting effect against yeast and mold. Compared to the control, developed coatings reduced browning, respiration rate, moisture, weight loss, and microbial load in fresh-cut apples. The study indicates that the blend of 5% sago and 0.5% soy oil produced the best coatings which were most effective for retaining the original quality attributes and in extending the shelf life of apple slices for 12 days in storage at 4 °C. The dynamics of water physicochemical properties and hydrochemical characteristics in the Dubna River was studied from April 18, 2021 to July 4, 2021. The study showed an increase in pH with increasing water temperature and considerable variation in the concentrations of ammonium, nitrite, and phosphate ions in it. Promising approaches were shown to be the use of mathematical processing of UV-spectra of water samples by Gauss method and model biological systems based on natural lipids (low-temperature oxidation of lecithin, and its ability to spontaneously aggregate in a polar environment) for assessing the quality of natural water and its effect on the regulation of oxidation processes in biological objects. Thus, analysis of UV-spectra revealed the presence in water of hydrophobic organic compounds throughout the observation period, compounds with conjugated double bonds and N-containing compounds in late April, free fatty acids, organic compounds with a carbonyl group and P-containing compounds at the end of May and an increase in the diversity of components in water samples in late June. Water samples taken in summer, increased the rate of lecithin oxidation by a factor of more than 1.8, and, throughout the observation period, they caused staged changes in the size of aggregates formed by it and produced a considerable effect on their dzeta-potential. In this article we studied chocolate with 100% cocoa without lecithin and traces of milk in the form of chips. The data obtained are in accordance with those obtained from the specialized literature. This article proposes two relations of dependence of log dynamic viscosity on log shear rate. In recent years, there has been a growing interest in regulating lipid digestion through the construction of various interfacial structures. In the present work, a series of complex interfacial structures were designed by combining Tween 80 in the aqueous phase and lecithin in the oil phase at different concentration ratios. The emulsification properties, the roles in regulating lipid digestion, and the interfacial dilatational rheological properties of the composite emulsifying systems were characterized. The results showed that the combination of Tween 80 and lecithin at different ratios could effectively modulate the rate of lipid digestion. The polyoxyethylene chains of Tween 80 formed a network, that provided a spatial obstacle for the adsorption of bile salts and lipases. Thus, Tween 80 significantly delayed the lipid digestion. The introduction of lecithin gradually replaced Tween 80 molecules at the interface, thus providing space for the adsorption of bile salts and lipases. In addition, as the ratio of lecithin concentration to Tween 80 increased, lecithin gradually became the dominant factor in the interfacial properties. As a result, the rate of lipid digestion was accelerated. Therefore, by compounding different ratios of lecithin and Tween 80, a series of emulsions with different lipid digestion rates were obtained. Many parameters are effective in obtaining optimum rheological and sensory properties in chocolate. Lecithin, ammonium phosphatides (AMP), and less commonly glycerol monopalmitate (GMP) are typically used as emulsifiers to achieve the desired viscosity in chocolate. In addition to these emulsifiers, polyglycerol polyricinoleate (PGPR) is used as a complement to regulate the yield value in chocolate. In this review, the properties of these additives, their interactions with each other, and the mechanism of their structural improvement functions in chocolate production are discussed. Depending on the type and amount of emulsifier, adjustments can be made to the flow behavior of molten chocolate, textural, melting character, tempering and tempering index, color and other properties of the chocolate. In addition to the use of emulsifiers, other parameters such as particle size distribution, moisture and fat content should also be taken into account, which can affect all these quality parameters. In this regard, the correct selection of the type of emulsifiers used in chocolate production and their combined usage rates will ensure that the demands of producers and consumers are met. The caviar and lecithin complex of freshwater fish can be widely used for the production of various food products, including those of functional orientation. The main technological operations in lecithin technology should include the processes of extraction of the initial caviar raw material and raffinate obtained after the first stage of selective extraction. The purpose of the study is to identify the rational mass ratio of extractant and raw material (module), on which the extraction efficiency depends, as well as the final concentration of extracted substances in the extract to determine its current value in the process of mass transfer to study the kinetics of extraction. The object of the study is the caviar-anchovy complex of fathead carp, carp, pikeperch and catfish, as well as the extract and raffinate from it. Summarizing the experimental data on revealing the rational module, as well as the final concentration of extracted substances in the extracts obtained, we can conclude that for caviar of the fish species under study, the rational hydromodule for its extraction with acetone is the ratio of dry caviar : extractant 1 : 4, and for its extraction with ethyl alcohol is the ratio of dry caviar : extractant 1 : 8. If the mass ratio refers to the fresh caviar of the studied fish species, then its rational indicator at its extraction with acetone is the ratio of fresh caviar : extractant 1 : 1.5, and at its extraction with ethyl alcohol is the ratio of fresh caviar : extractant 1 : 3. The average final concentration of extracted substances from dry caviar of the studied fish species in the extracts obtained, if acetone was used as a solvent under rational conditions of mass transfer, corresponds to the value of 1.3 %, and if ethyl alcohol was used as a solvent under rational conditions of mass transfer, corresponds to the value of 1.0 %.

Hole cleaning is one of the most important aspects of extractive drilling operations. When drill cuttings combine with drilling fluids, cutting bed gel is created, which becomes extremely challenging to remove while drilling. One of the functions of drilling fluid when the flow is static is to maintain the drill bits' suspension. However, the settling of drill bits makes this nearly impossible. In this paper, the influence of fluid viscosity on hole cleaning performance was critically studied through a series of laboratory experiments. The effects of different viscosity levels on hole cleaning were investigated. This paper evaluated the use of lecithin and carboxymethyl hydroxyethyl cellulose (CMHEC) as a viscosifier and fluid loss agent in oil-based drilling fluid. The rheological properties of the formulated drilling fluids were determined at various temperatures according to the API 13B-2 specifications in order to identify the most suitable mud type for high-temperature drilling operations. From this study, it was discovered that Formulation B has the best rheology, with a plastic viscosity (PV) of 12, 7, and 7 cP at 30, 40, and 50 °C, respectively, while its yield point (YP) was found to be 30, 24, and 16, respectively. These values were compared with the generic oil-based formulation (A), which had a plastic viscosity (VP) of 43, 39, and 22 cP at 30, 40, and 50 °C, respectively, while its yield point (YP) was found to be 38, 20, and 26, respectively. The findings show that formulation B has a significant resistance to high temperatures, which means that it can be applied to drilling operations in reservoirs with high temperatures. It is more environmentally friendly. Furthermore, this paper explores the implications of the results in terms of the efficiency of hole cleaning and the drilling process relative to the viscosity of oil-based drilling fluid. This paper concludes by providing suggestions for improving drilling efficiency through the selection of an appropriate drilling fluid viscosity.

This study examined the effects of fat supplementation with high palmitic acid enriched with lecithin on the performance, egg quality, and fatty acid profile of quails. This study applied a completely randomized design to 330 female quails (Cortunix cortunix japonica) which were allocated into 5 different groups with 3 replicates of 22 birds. The groups were assigned as follows: T0 = commercial diet (control), T1 = commercial diet + 2.5% fat + 6% lecithin of added fat, T2 = commercial diet + 2.5% commercial fat, T3 = commercial diet + 5% fat + 6 % lecithin of added fat, T4 = commercial diet + 5% commercial fat. The results showed that the treatment group did not give effects on performances. However, the treatment group with fat level of 2.5% and 5% increased the score of egg yolk and eggshell thickness. Moreover, the treatment group decreased fat content in terms of saturated fatty acids and n-6/n-3 ratio of quail eggs. In conclusion, fat supplementation containing high palmitic acid enriched with lecithin proves effective in improving egg quality. However, the performance remains relatively similar across the groups.

Therefore, concerted efforts are made to prevent or manage disruptions from normal functioning of the cardiovascular system. Disruption in lipid metabolism is a major contributor to cardiovascular dysfunction. This review examines how lecithin impacts lipid metabolism and cardiovascular health. It emphasizes lecithin's ability to reduce excess low-density lipoproteins (LDL) while specifically promoting the synthesis of high-density lipoprotein (HDL) particles, thus contributing to clearer understanding of its role in cardiovascular well-being. Emphasizing the importance of lecithin cholesterol acyltransferase (LCAT) in the reverse cholesterol transport (RCT) process, the article delves into its contribution in removing surplus cholesterol from cells. This review aims to clarify existing literature on lipid metabolism, providing insights for targeted strategies in the prevention and management of atherosclerotic cardiovascular disease (ASCVD). As an acne sequela, post-acne scarring (PSA) has huge negative impact on sufferers' quality of life because of aesthetical embarrassment. Transdermal delivery of botulinum toxin-A (BTXA) is a promising strategy for PAS treatment, but currently reported approaches are far from satisfactory. In this work, phosphatidylcholine/cholesterol (PC/Chol) nanoliposomes were utilized for encapsulation and transdermal delivery of BTXA. The composition, structure, morphology, size, size distribution, etc. of as-prepared BTXA@liposome nanoparticles were investigated in detail. Simulated transdermal delivery assay indicated that the diffusion depth of the BXTA@liposome nanoparticles was nearly 8 times that of pure BTXA and reached 380 μm. 12 facial PSA patients were recruited to evaluate the curative effect of the BTXA@liposome nanoparticles on PSA. Through ECCA (échelle d'évaluation clinique des cicatrices d'acné) scoring and self-evaluation of patients, the resultant data indicated that compared to hyaluronic acid (HA) hydrogel treatment the BTXA@liposome/HA hydrogel treatment could better relieve PSA to some extent but didn't show significant advantage. Background: Multiple sclerosis (MS) is an irreversible progressive CNS pathology characterized by the loss of myelin (i.e. demyelination). The lack of myelin is followed by a progressive neurodegeneration triggering symptoms as diverse as fatigue, motor, locomotor and sensory impairments and/or bladder, cardiac and respiratory dysfunction. Even though there are more than fourteen approved treatments for reducing MS progression, there are still no cure for the disease. Thus, MS research is a very active field and therefore we count with different experimental animal models for studying mechanisms of demyelination and myelin repair, however, we still lack a preclinical MS model assembling demyelination mechanisms with relevant clinical-like signs. Results: Here, by inducing the simultaneous demyelination of both callosal and cerebellar white matter fibers by the double-site injection of lysolecithin (LPC), we were able to reproduce CNS demyelination, astrocyte recruitment and increases levels of proinflammatory cytokines levels along with motor, locomotor and urinary impairment, as well as cardiac and respiratory dysfunction, in the same animal model. Single site LPC-injections either in corpus callosum or cerebellum only, fails in to reproduce such a complete range of MS-like signs. Conclusion: We here report that the double-site LPC injections treatment evoke a complex MS-like mice model. Background: Previous studies have linked sports-related concussions and repeated subconcussive head impacts in contact sport athletes to elevated brain injury biomarkers. Docosahexaenoic acid (DHA), the primary omega-3 (n-3) highly unsaturated fatty acid (HUFA) in the brain, has shown neuroprotective effects in animal models after brain injury, but clinical research has shown mixed results. Methods: We conducted a randomized, double-blind, placebo-controlled study on 29 Division 1 collegiate American football players, exploring the impact of DHA (2.5 g) and eicosapentaenoic acid (EPA) (1.0 g) supplied as ethyl esters, on levels of plasma lipids shown to cross the blood-brain barrier. Dietary intake data was collected using food frequency questionnaires (FFQ). Complex lipids and unesterified fatty acids were isolated from plasma, separated via reversed-phase liquid chromatography and analyzed by targeted lipidomics analysis. Results: FFQ results indicated that participants had low dietary n-3 HUFA intake and high omega-6 (n-6):n-3 polyunsaturated fatty acids (PUFA) and HUFA ratios at baseline. After DHA + EPA supplementation, plasma lysophosphatidylcholine (LPC) containing DHA and EPA significantly increased at all timepoints (weeks 17, 21, and 26; p < 0.0001), surpassing placebo at Weeks 17 (p < 0.05) and 21 (p < 0.05). Phosphatidylcholine (PC) molecular species containing DHA or EPA, PC38:6 PC36:6, PC38:7, PC40:6, and PC40:8, increased significantly in the DHA + EPA treatment group at Weeks 17 (and 21. Lipid nanoparticles often contain a phosphatidylcholine with a long chain fatty acid, 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC). However, their preparation often encounters difficulties such as the inability to yield <20 nm nanoparticles due to the aggregation-prone behavior of DSPC. High-density lipoproteins (HDLs) are ∼10 nm protein-bound lipid nanoparticles in our body, and microfluidic preparations of HDL-mimicking nanoparticles (μHDL) have been reported. Herein, we report a new microfluidic mixing mode that enables preparation of μHDL with DSPC in high yield (≥90% on a protein basis). The critical mechanism of this mode is a spontaneous asymmetric distribution of the ethanol flow injected in a symmetric manner followed by turbulent mixing in a simple rectangular parallelepiped-shaped chip. Background: The most popular and well-designed nano-carriers for drug delivery targeting are liposomes. In order to overcome barriers to cellular and tissue uptake and increase medication bio-distribution to target areas in vivo, they have improved medicines for a variety of biomedical applications. Objectives: The diffusion method is used to release the medication from liposomes. The major goals of this medication delivery system were to increase contact resistance duration in the eye and bioavailability. Materials and Methods: Liposomes were effectively produced by reverse phase evaporation technique and their shape, particle size, drug entrapment efficiency and content were examined. Liposomes were prepared by using drug, egg lecithin, chloroform, and diethyl ether. Results: This dosage form bears considerable promise for drug release, according to in vitro experiments. As a result, the design of the liposomes revealed virtually spherical vesicles. Up to 88.9% and 92.1%, respectively, of the EE% and DC% were possible. Studies on medication release from liposomes in vitro demonstrate that an increase in lipid concentration delays drug release. Conclusion: The drug is known as dorzolamide in accordance with the conclusions of the investigations stated above. The medicine is stable in all pH, it was found after the Preformulation experiment was over. The article aimed to formulate an MLX binary ethosome hydrogel for topical delivery to escalate MLX solubility, facilitate dermal permeation, avoid systemic adverse events, and compare the permeation flux and efficacy with the classical type. MLX ethosomes were prepared using the hot method according to the Box–Behnken experimental design. The formulation was implemented according to 16 design formulas with four center points. Independent variables were (soya lecithin, ethanol, and propylene glycol concentrations) and dependent variables (vesicle size, dispersity index, encapsulation efficiency, and zeta potential). The design suggested the optimized formula (MLX–Ethos–OF) with the highest desirability to perform the best responses formulated and validated. It demonstrates a 169 nm vesicle size, 0.2 dispersity index, 83.1 EE%, and −42.76 mV good zeta potential. MLX–Ethos–OF shows an amorphous form in PXRD and a high in vitro drug release of >90% over 7 h by diffusion and erosion mechanism. MLX–Ethos–OF hyaluronic acid hydrogel was fabricated and assessed. It shows an elegant physical appearance, shear thinning system rheological behavior, good spreadability, and skin-applicable pH value. The ex vivo permeation profile shows a flux rate of 70.45 μg/cm2/h over 12 h. The in vivo anti-inflammatory effect was 53.2% ± 1.3 over 5 h. compared with a 10.42 flux rate and 43% inflammatory inhibition of the classical ethosomal type. The receptive phase of the uterus is marked by structural and functional maturation of the endometrium. During this limited time span, the blastocyst competency is superimposed on the receptive endometrium. It is a well-known fact that lipid signalling in early-stage pregnancy has a crucial role in successful embryogenesis. In our study, CD-1 mouse uteri after normal and in vitro fertilization (IVF) were investigated at 6.5, 8.5, and 10.5 days of pregnancy. Matrix-assisted laser desorption/ionization time-of-flight imaging mass spectrometry and liquid chromatography coupled tandem mass spectrometry were used for identification of phosphatidylcholine (PC) lipid structures. In the embryonal tissues, PC 32:0 and PC 34:0 were increased, while in the antemesometrial (AM) decidua the two 20:4-containing PCs, PC 36:4 and PC 38:4 were increased. In transferred uterus samples, higher expressions of PC 34:0, PC 34:1, PC 34:2, PC 36:1, and PC 36:2 in mesometrial decidua were seen, whereas the two 20:4-containing PCs, PC 36:4 and PC 38:4 showed increased expression in the AM and lateral decidua. The elucidation of the interaction mechanism between phospholipids and milk proteins within emulsions is pivotal for comprehending the properties of infant formula fat globules. In this study, multispectral methods and molecular docking were employed to explore the relationship between phosphatidylcholine (PC) and whey protein isolate (WPI). Observations indicate that the binding constant, alongside thermodynamic parameters, diminishes as temperature ascends, hinting at a predominantly static quenching mechanism. Predominantly, van der Waals forces and hydrogen bonds constitute the core interactions between WPI and PC. This assertion is further substantiated by Fourier transform infrared spectroscopy, which verifies PC's influence on WPI's secondary structure. A detailed assessment of thermodynamic parameters coupled with molecular docking reveals that PC predominantly adheres to specific sites within α‐lactalbumin, β‐lactoglobulin, and bovine serum albumin, propelled by a synergy of hydrophobic interactions, hydrogen bonding, and van der Waals forces, with binding energies noted at −5.59, −6.71, and −7.85 kcal/mol, respectively. An increment in PC concentration is observed to amplify the emulsification properties of WPI whilst concurrently diminishing the zeta potential. Familial lecithin:cholesterol acyltransferase (LCAT) deficiency (FLD) is an ultra-rare autosomal recessive disease characterized by very low high-density lipoprotein cholesterol (HDL-C) levels, corneal opacity, anemia, and progressive renal disease. The rate and severity of renal disease are variable across FLD patients and the biomarkers and risk factors for disease progression are poorly understood. Here we report a 30 year-long comparative analysis of the clinical and laboratory biomarkers in an FLD patient with accelerated renal decline, who underwent two kidney and one liver transplantations. Results show that elevated triglyceride and non-HDL-C levels may promote the formation of LpX and accelerate renal function decline, whereas markers of anemia may be early predictors. Conversely, corneal opacity progresses at a steady rate and does not correlate with lipid, hematologic, or renal biomarkers. Our study suggests that monitoring of markers of anemia may aid the early detection and timely management of kidney disease with conservative therapies. Introduction: Nowadays, the bioeconomy and sustainability trends drive the food sector to replace chemical preservatives with bioactive compounds recovered from biomass such as essential oils (EOs) and their derivatives. Moreover, nanotechnology trend drives food sector in the nanoencapsulation of such bioactive compounds in novel edible formulations such as nanoemulsions to enhance their controlled release properties, their bioactivity and their biocompatibility. Methods: In this study the development and characterization of novel casein/ lecithin (CSN/LCN) based nanoemulsions (NEs) with carvacrol (CV), cinnamaldehyde (CI), citral (CT), and eugenol (EG) as nanoencapsulated oil phase is presented as well as the application of such NEs as active coatings for the preservation of fresh pork tenderloin. Results and discussion: DLS measurements as well as TEM images showed an average particle size distribution of 20--40 nm for all obtained NEs, while z-potential results indicate their physicochemical stability. All the obtained NEs needed at least 5 times less amount of EOs derivatives to scavenge 50% of radicals and demonstrated a higher antibacterial activity against Escherichia coli and Listeria monocytogenes compared to free EOs derivatives. Pork fresh tenderloin meat treated with NEs demonstrated 30% higher efficacy in obtained lipid oxidation values than uncoated pork meat samples and 12% higher efficacy in obtained lipid oxidation values than pork meat samples treated with EOs derivatives. From a microbiological point of view NEs manage to extend pork tenderloin meat shelf life by 6 days compared to uncoated pork tenderloin meat and by 3 days compared to tenderloin pork meat coated with free EOs derivatives. Lab* colorimetry analysis as well as sensory analysis revealed that such edible CSN/LCN based NEs succeeded to preserve the texture, appearance, and color of pork tenderloin meat in higher acceptance limits compared to the corresponding characteristics of pork tenderloin meat coated with free EOs derivatives. This study was designed to compare the addition of lecithin (LC) and lecithin nanoparticles (LC-NPs) to a semen extender (control group – egg yolk, EY) during a storing period in relation to its effectiveness on buffalo sperm cryopreservation. Ejaculates were collected from six mature, healthy Egyptian buffalo bulls using an artificial vagina, and diluted with nine extenders. LC-NPs and LC were tested at concentrations of 0.25, 0.5, 1.00, and 1.50 µg/ml compared to the egg yolk extender. The results indicated an overall enhancement in sperm quality: increased progressive motility and improved hypo-osmotic swelling test (HOST), reduced instances of dead sperm, abnormal morphology and chromatin damage in the LC-NPs group compared to both LC and the control groups (P < 0.05), apart from group 0.25 µg/ml. Furthermore, LC-NPs demonstrated superior viability at a concentration of 1 µg/ml. This concentration exhibited lower rates of early and late apoptotic events, as well as reduced necrosis compared to lower concentrations. The results showed significantly increased (P < 0.05) antioxidant indices, including superoxide dismutase (SOD), glutathione peroxidase, and total antioxidant capacity, with the inclusion LC-NPs, particularly notable at concentrations of 1.00 and 1.50 µg/ml, surpassing the effects observed in other groups. In contrast, the levels of malondialdehyde decreased significantly (P < 0.05) with LC-NPs, particularly at 1.00 µg/ml in comparison to the LC and control groups. Moreover, the fortification of cryopreserved spermatozoa with 1 or 1.5 µg/ml LC-NPs resulted in the preservation of acrosomal and plasma membrane integrity, as well as normal ultrastructure. So, the addition of LC-NPs, particularly of 1 or 1.5 µg/ml, significantly enhanced sperm quality by improving antioxidant indices, reducing apoptosis, and preserving ultrastructure integrity of buffalo sperm after the post-thawing process. Background: Ezetimibe, which lowers cholesterol by blocking the intestinal cholesterol transporter Niemann-Pick C1 like 1, is reported to reduce hepatic steatosis in humans and animals. Here, we demonstrate the changes in hepatic metabolites and lipids and explain the underlying mechanism of ezetimibe in hepatic steatosis. Methods: We fed Otsuka Long-Evans Tokushima Fatty (OLETF) rats a high-fat diet (60 kcal % fat) with or vehicle (control) or ezetimibe (10 mg kg-1) via stomach gavage for 12 weeks and performed comprehensive metabolomic and lipidomic profiling of liver tissue. We used rat liver tissues, HepG2 hepatoma cell lines, and siRNA to explore the underlying mechanism. Results: In OLETF rats on a high-fat diet, ezetimibe showed improvements in metabolic parameters and reduction in hepatic fat accumulation. The comprehensive metabolomic and lipidomic profiling revealed significant changes in phospholipids, particularly phosphatidylcholines (PC), and alterations in the fatty acyl-chain composition in hepatic PCs. Further analyses involving gene expression and triglyceride assessments in rat liver tissues, HepG2 hepatoma cell lines, and siRNA experiments unveiled that ezetimibe's mechanism involves the upregulation of key phospholipid biosynthesis genes, CTP:phosphocholine cytidylyltransferase alpha and phosphatidylethanolamine N-methyl-transferase, and the phospholipid remodeling gene lysophosphatidylcholine acyltransferase 3. Conclusion: This study demonstrate that ezetimibe improves metabolic parameters and reduces hepatic fat accumulation by influencing the composition and levels of phospholipids, specifically phosphatidylcholines, and by upregulating genes related to phospholipid biosynthesis and remodeling. Introduction: Traditional clinical risk factors are insufficient to estimate the residual risk of large-vessel ischemic stroke. Non-targeted lipidomic techniques provide an opportunity to evaluate these risks. Methods: Plasma samples were collected from 113 hypertensive individuals, including 55 individuals at high risk of ischemic stroke and 58 matched individuals, in a prospective nested case-control cohort. To identify dysregulated lipid metabolites, we conducted multivariate and univariate analyses. A classifier based on a cross-validated procedure was employed to select the optimal combination of lipid species and their ratios. Results: We identified 23 dysregulated lipid species in patients with and without ischemic stroke, including 16 (69.6%) up-regulated and 7 (30.4%) down-regulated lipid species. Through internal cross-validation, the optimal combination of two lipid features (phosphatidylcholine 34:2 and triglyceride 18:1/18:1/22:1 / phosphatidylcholine 34:2, referred to as ischemic stroke-related 2 lipid features - IS2LP) was selected, leading to a more precise prediction probability for ischemic stroke within 3.9 years. In the comparison of different risk factors, the traditional risk score, the IS2LP risk score, and the combination of the traditional risk score with IS2LP yield AUC values of 0.613(95% CI:0.509– 0.717), 0.833(95% CI:0.755– 0.911), and 0.843(95% CI:0.777– 0.916), respectively. The combination of the traditional risk score and IS2LP exhibited significantly improved discriminative performance, with an integrated discrimination improvement (IDI) of 0.31 (p< 0.001) and a continuous net reclassification improvement (NRI) of 1.06 (p < 0.001) compared to the traditional risk score. Conclusion: We identified new lipidomic biomarkers associated with the futural event of large-vessel ischemic stroke. Background: Lipid droplet (LD)-laden microglia is a key pathological hallmark of multiple sclerosis. The recent discovery of this novel microglial subtype, lipid-droplet-accumulating microglia (LDAM), is notable for increased inflammatory factor secretion and diminished phagocytic capability. Lipophagy, the autophagy-mediated selective degradation of LDs, plays a critical role in this context. This study investigated the involvement of microRNAs (miRNAs) in lipophagy during demyelinating diseases, assessed their capacity to modulate LDAM subtypes, and elucidated the potential underlying mechanisms involved. Methods: C57BL/6 mice were used for in vivo experiments. Two weeks post demyelination induction at cervical level 4 (C4), histological assessments and confocal imaging were performed to examine LD accumulation in microglia within the lesion site. Autophagic changes were observed using transmission electron microscopy. miRNA and mRNA multi-omics analyses identified differentially expressed miRNAs and mRNAs under demyelinating conditions and the related autophagy target genes. The role of miR-223 in lipophagy under these conditions was specifically explored. In vitro studies, including miR-223 upregulation in BV2 cells via lentiviral infection, validated the bioinformatics findings. Immunofluorescence staining was used to measure LD accumulation, autophagy levels, target gene expression, and inflammatory mediator levels to elucidate the mechanisms of action of miR-223 in LDAM. Results: Oil Red O staining and confocal imaging revealed substantial LD accumulation in the demyelinated spinal cord. Transmission electron microscopy revealed increased numbers of autophagic vacuoles at the injury site. Multi-omics analysis revealed miR-223 as a crucial regulatory gene in lipophagy during demyelination. It was identified that cathepsin B (CTSB) targets miR-223 in autophagy to integrate miRNA, mRNA, and autophagy gene databases. In vitro, miR-223 upregulation suppressed CTSB expression in BV2 cells, augmented autophagy, alleviated LD accumulation, and decreased the expression of the inflammatory mediator IL-1β. Conclusion: These findings indicate that miR-223 plays a pivotal role in lipophagy under demyelinating conditions. By inhibiting CTSB, miR-223 promotes selective LD degradation, thereby reducing the lipid burden and inflammatory phenotype in LDAM. This study broadens the understanding of the molecular mechanisms of lipophagy and proposes lipophagy induction as a potential therapeutic approach to mitigate inflammatory responses in demyelinating diseases. Dietary emulsifiers are linked to various diseases. The recent discovery of the role of gut microbiota–host interactions on health and disease warrants the safety reassessment of dietary emulsifiers through the lens of gut microbiota. Lecithin, sucrose fatty acid esters, carboxymethylcellulose (CMC), and mono- and diglycerides (MDG) emulsifiers are common dietary emulsifiers with high exposure levels in the population. This study demonstrates that sucrose fatty acid esters and carboxymethylcellulose induce hyperglycemia and hyperinsulinemia in a mouse model. Lecithin, sucrose fatty acid esters, and CMC disrupt glucose homeostasis in the in vitro insulin-resistance model. MDG impairs circulating lipid and glucose metabolism. All emulsifiers change the intestinal microbiota diversity and induce gut microbiota dysbiosis. Lecithin, sucrose fatty acid esters, and CMC do not impact mucus–bacterial interactions, whereas MDG tends to cause bacterial encroachment into the inner mucus layer and enhance inflammation potential by raising circulating lipopolysaccharide. Our findings demonstrate the safety concerns associated with using dietary emulsifiers, suggesting that they could lead to metabolic syndromes. Common dietary emulsifiers such as lecithin, sucrose fatty acid esters, carboxymethylcellulose, and mono- and diglycerides can promote metabolic disorders and intestinal microbiota dysbiosis in mice. Neutral conditions are conducive for the yeast to mycelium transformation of C. albicans and inducing biofilm formation. Alternatively, an acidic environment can promote the growth of C. albicans yeast and phospholipase expression, and obviously affect the antifungal activity of ketoconazole (KET). In this study, KET-loaded nanostructure hybrid lipid capsules (KET-nHLCs) comprised of soybean lecithin, HS15 and ethyl α-linolenate were prepared using the thin film hydration method. Additionally, the effect of pH on the release and antifungal activity of KET-nHLCs was investigated. The encapsulation efficiency of KET-nHLCs was determined to be 91.53% ± 1.24%, and the water solubility of KET in the KET-nHLC group was enhanced by 158-fold compared to the crude KET. KET-nHLCs could control the in vitro drug release at pH 4.5/7.4, while its slow-release effect on KET was more obvious at pH 4.5. The MIC50 values of KET-nHLCs and KET were 0.5 and 0.25 μg mL−1 at pH 7.4 and 2 and 4 μg mL−1 pH 4.5, respectively. Interestingly, KET-nHLCs more effectively inhibited the yeast-hyphae transformation and had better antibiofilm activity than crude KET. [Display omitted] Conjugating biomolecules, such as antibodies, to bioconjugate moieties on lipid surfaces is a powerful tool for engineering the surface of diverse biomaterials, including cells and nanoparticles. We developed supported lipid bilayers (SLBs) presenting well-defined spatial distributions of functional moieties as models for precisely engineered functional biomolecular-lipid surfaces. We used quartz crystal microbalance with dissipation (QCM-D) and atomic force microscopy (AFM) to determine how vesicles containing a mixture of 1,2-dipalmitoyl- sn - glycero -3-phosphatidylcholine (DPPC) and 1,2-distearoyl- sn - glycero -3-phosphoethanolamine-N-[azido(polyethylene glycol)-2000] (DSPE-PEG-N 3) form SLBs as a function of the lipid phase transition temperature (T m). Above the DPPC T m , DPPC/DSPE-PEG-N 3 vesicles form SLBs with functional azide moieties on SiO 2 substrates via vesicle fusion. Below this T m , DPPC/DSPE-PEG-N 3 vesicles attach to SiO 2 intact. Intact DPPC/DSPE-PEG-N 3 vesicles on the SiO 2 surfaces fuse and rupture to form SLBs when temperature is brought above the DPPC T m. AFM studies show uniform and complete DPPC/DSPE-PEG-N 3 SLB coverage of SiO 2 surfaces for different DSPE-PEG-N 3 concentrations. As the DSPE-PEG-N 3 concentration increases from 0.01 to 6 mol%, the intermolecular spacing of DSPE-PEG-N 3 in the SLBs decreases from 4.6 to 1.0 nm. The PEG moiety undergoes a mushroom to brush transition as DSPE-PEG-N 3 concentration varies from 0.1 to 2.0 mol%. Via copper-free click reaction, IgG was conjugated to SLB surfaces with 4.6 nm or 1.3 nm inter-DSPE-PEG-N 3 spacing. QCM-D and AFM data show; 1) uniform and complete IgG layers of similar mass and thickness on the two types of SLB; 2) a higher-viscosity/less rigid IgG layer on the SLB with 4.6 nm inter-DSPE-PEG-N 3 spacing. The low rehydration properties of commercial soy protein powder (SPI), a major plant−based food ingredient, have limited the development of plant−based foods. The present study proposes a treatment of soy lecithin modification combined with Alcalase hydrolysis to improve the rehydration of soy protein powder, as well as other processing properties (emulsification, viscosity). The results show that the soy protein–soy lecithin complex powder, which is hydrolyzed for 30 min (SPH–SL−30), has the smallest particle size, the smallest zeta potential, the highest surface hydrophobicity, and a uniform microstructure. In addition, the value of the ratio of the α−helical structure/β−folded structure was the smallest in the SPH–SL−30. After measuring the rehydration properties, emulsification properties, and viscosity, it was found that the SPH–SL−30 has the shortest wetting time of 3.04 min, the shortest dispersion time of 12.29 s, the highest solubility of 93.17%, the highest emulsifying activity of 32.42 m2/g, the highest emulsifying stability of 98.33 min, and the lowest viscosity of 0.98 pa.s. This indicates that the treatment of soy lecithin modification combined with Alcalase hydrolysis destroys the structure of soy protein, changes its physicochemical properties, and improves its functional properties. Lysophosphatidylcholine (LPC) is present in various foods and contains a choline moiety such as in glycerophosphocholine (GPC). However, the potential of LPC as a choline source remains unclear. This study investigated the single-dose pharmacokinetics of 480 mg soy-derived LPC in 12 healthy men compared with that of either soy oil with the same lipid amount (placebo) or GPC with the same choline amount. Both LPC and GPC supplementation increased plasma choline, serum phospholipid, and serum triglyceride concentrations, but neither of them significantly elevated plasma trimethylamine N -oxide concentration. In addition, although the intake of LPC slightly increased plasma LPC16:0, LPC18:2, and total LPC concentrations, their concentrations remained within physiological ranges. No adverse events were attributed to the LPC supplementation. Background: This study aimed to assess the response patterns of circulating lipids to exercise and diet interventions in nonalcoholic fatty liver disease (NAFLD). Methods: The 8.6-month four-arm randomized controlled study comprised 115 NAFLD patients with prediabetes who were assigned to aerobic exercise (AEx; n = 29), low-carbohydrate diet (Diet; n = 28), AEx plus low-carbohydrate diet (AED; n = 29), and nonintervention (NI, n = 29) groups. Hepatic fat content (HFC) was quantified by proton magnetic resonance spectroscopy. Serum lipidomic analytes were measured using liquid chromatography-mass spectrometry. Results: After intervention, the total level of phosphatidylcholine (PC) increased significantly in the AEx group (P = 0.043), whereas phosphatidylethanolamine (PE) and triacylglycerol decreased significantly in the AED group (P = 0.046 and P = 0.036, respectively), and phosphatidylserine decreased in the NI group (P = 0.002). Changes of 21 lipid metabolites were significantly associated with changes of HFC, among which half belonged to PC. Most of the molecules related to insulin sensitivity belonged to sphingomyelin (40 of 79). Controlling for the change of visceral fat, the significant associations between lipid metabolites and HFC remained. In addition, baseline serum lipids could predict the response of HFC to exercise and/or diet interventions (PE15:0/18:0 for AED, area under the curve (AUC) = 0.97; PE22:6(4Z,7Z,10Z,13Z,16Z,19Z)/0:0 for AEx, AUC = 0.90; and PC14:1(9Z)/19:1(9Z) for Diet, AUC = 0.92). Conclusions: Changes of lipidome after exercise and/or diet interventions were associated with HFC reductions, which are independent of visceral fat reduction, particularly in metabolites belonging to PC. Importantly, baseline PE could predict the HFC response to exercise, and PC predicted the response to diet. Lysophosphatidylcholine (LPC) is immunomodulatory in nonruminants; however, the actions of LPC on immunity in cattle are undefined. Our objective was to study the effects of LPC administration on measures of immunity, liver health, and growth in calves. Healthy Holstein heifer calves (n = 46; age 7 ± 3 d) were randomly assigned to 1 of 4 treatments (n = 10 to 11 calves/treatment): a milk replacer diet unsupplemented with lecithin in the absence (CON) or presence of subcutaneously (s.c.) administered mixed (mLPC; 69% LPC-16:0, 25% LPC-18:0, 6% other) or pure LPC (pLPC; 99% LPC-18:0), or a milk replacer diet supplemented with 3% lecithin enriched in lysophospholipids containing LPC in the absence of s.c.-administered LPC (LYSO) for 5 wk. Calves received 5 s.c. injections of vehicle (10 mL of phosphate-buffered saline containing 20 mg of bovine serum albumin/mL; CON and LYSO) or vehicle containing mLPC or pLPC to provide 10 mg of total LPC per kilogram of BW per injection every 12 h during wk 2 of life. Calves were fed a milk replacer containing 27% crude protein and 24% fat at 1.75% of BW per day (dry matter basis) until wk 6 of life (start of weaning). Starter grain and water were provided ad libitum. Body measurements were recorded weekly, and clinical observations were recorded daily. Blood samples were collected weekly before morning feeding and at 0, 5, and 10 h, relative to the final s.c. injection of vehicle or LPC. Data were analyzed using a mixed model, with repeated measures including fixed effects of treatment, time, and their interaction. Dunnett's test was used to compare treatments to CON. Peak rectal temperatures were higher in mLPC or pLPC, relative to CON. Plasma LPC concentrations were greater in mLPC and LYSO calves 5 h and 10 h after the final injection, relative to CON. Calves receiving mLPC and pLPC also had higher circulating serum amyloid A concentrations, relative to CON. Calves receiving mLPC had greater serum aspartate aminotransferase, γ-glutamyltransferase, and glutamate dehydrogenase concentrations, relative to CON. Calves provided mLPC experienced lower average daily gain (ADG) after weaning, relative to CON. The LYSO treatment did not modify rectal temperatures, ADG, or measures of liver health, relative to CON. Anthocyanin is one of the bioactive compounds, which is the main pigment of many fruits and vegetables. Since anthocyanins have low thermal stability during food processing, the use of these compounds as natural pigments in foods is associated with challenges. Therefore, microencapsulation of anthocyanin compounds with liposomes is important. Nanoencapsulation of bioactive compounds with liposomes is an effective and efficient way to increase the stability of polyphenolic compounds. Liposomes are polar lipid vesicles that form bilayer structures in polar solvents such as water. In this research, nanoliposomes in ratios of 9-1, 8-2, 7-3 and 6-4 lecithin-cholesterol were prepared using the solvent injection method. Then, the size and zeta potential tests were conducted to determine the characteristics of the produced particles. The average particle size (average hydrodynamic diameter) and particle size distribution for different lecithin-cholesterol ratios were in the range of 132-740 nm and 0.47-0.41, respectively. Zeta potential values were also obtained in the range of -26 to -42 mv. After determining the efficiency of Nanoencapsulation, FTIR test was performed to investigate possible reactions between anthocyanins and nanoliposome wall materials. The morphology of anthocyanin-loaded lecithin-cholesterol nanoliposomes with a ratio of 9-1 was shown by scanning electron microscopy (SEM). The stability of the liposomal sample with a ratio of 9-1 lecithin-cholesterol was evaluated by calculating the amount of release of encapsulated anthocyanin during 60 days of storage at ambient temperature. Samples with 9-1 lecithin-cholesterol ratio were used in Kombucha beverage formulation. The prevalence of Alzheimer's disease (AD) in developed and developing nations is increasing day by day. The monotherapy doesn't fit best for AD due to chronic treatment. This work aimed to prepare the liposomes coloaded with Donepezil and pine oil intended for adjuvant effect. Pine oil was selected due to its amyloid disaggregation and AChE inhibition property. The thin film hydration method with sonication was selected to formulate ultra-small-sized unilamellar vesicles. Apart from this formulated vesicle were subject to particle size, morphological, release, nasal ciliotoxicity and cellular uptake studies. The PDI, particle size, and zeta potential of the final egg lecithin-based liposome were found to be 0.19, 129 nm, and -27.5 mV, respectively, stable at 4° and 25°C. SEM microscopy of oil-loaded liposomes exhibits a crystalline cubic shape and is compared with alone drug-loaded liposomes that are spherical. AChE inhibition activity was found to be higher in combination when compared with others. The prepared liposomes were safe when studied for nasal ciliotoxicity studies and rapid uptake by C6 cells with complete internalization. This study demonstrated the development and potential of the vesicular system that carries valuable characteristics and overcomes patient compliance concerns that prove to be novel therapeutics for managing AD.
