The phytochemical constituents, acute toxicity, and antiulcer activity of Duranta erecta fruit extracts were determined in rodents. The antiulcer activity was tested utilizing a pylorus ligation. For the pylorus ligation-induced ulcer model, positive controls received ranitidine 10 mg kg−1. The negative controls were given ethanol (1 mL). The hydroethanolic fruit extracts of D. erecta demonstrated no mortality or notable behavioral effects up to 5000 mg kg−1. The crude ethanol fruit extract of D. erecta demonstrated antiulcer effects on pyloric ligation in rats at 500 and 1000 mg kg−1 doses. The purpose of the present study was to examine if potentiation of mortality occurred after simultaneous administration of several Escherichia coli genotypes, each capable of inducing the E. coli peritonitis syndrome, in comparison with single genotype application. Five groups of productive specified pathogen free White Leghorn hens were housed in isolators. Groups 1–4 consisted of 32 hens each, group 5 of 10 hens. At 32 weeks of age all groups were inoculated intratracheally. Groups 1 and 2 were inoculated with a mix of four E. coli genotypes and groups 3 and 4 with a mix of four other genotypes. Groups 1 and 3 were given 1 median lethal dose (LD50) of each genotype per hen and groups 2 and 4 had a dose of 0.1 LD50 per genotype per hen; group 5 was mock inoculated. The experiment ended one week after inoculations. In Group 5, no mortality occurred and gross lesions were absent at post-mortem examination. Mortality in groups 1 and 3 was 84% and 81%, respectively; in groups 2 and 4 59% and 66%, respectively. Although mortality in groups 1 and 3 exceeded the expected 50%, this could not be due to potentiation as cluster analysis of reisolates showed that in individual hens only one genotype was found, indicating interference between E. coli genotypes. In groups all four or only two genotypes were recovered, showing that not all genotypes will induce colibacillosis in all experimental groups. Therefore, broad protection can be best assessed by challenging with various single genotypes. Snakebite envenoming (SBE) remains a severely neglected public health issue, particularly affecting tropical and subtropical regions, with Africa experiencing an estimated 435,000 to 580,000 snakebites annually, leading to high morbidity and mortality rates, especially across Africa and Asia. Recognized as a Neglected Tropical Disease, SBE management is further complicated by the inadequate efficacy of current antivenom treatments. Of particular concern are cobras (Naja sp.), whose neurotoxins can induce rapid fatal respiratory paralysis. In this study, we investigate the potential of nanobodies as a promising next-generation of immunotherapeutics against cobra venoms. Through a dual strategy of the characterization of venom toxic fractions from cobras captured for the first time in Algeria and Tunisia biotopes, coupled with in vitro assays to evaluate their interactions with acetylcholine receptors, and subsequent immunization of dromedaries to produce specific nanobodies, we identified two lethal fractions, F5 and F6, from each venom, and selected five nanobodies with significant binding and neutralizing of 3DL50 (0.74 mg/kg). The combination of these nanobodies demonstrated a synergistic effect, reaching 100% neutralizing efficacy of 2DL50 lethal venom fraction (0.88 mg/kg) doses in mice. Drug use during pregnancy is an important issue that must be investigated due to its adverse effects on maternal and foetal health. This study aimed to determine the embryotoxic and teratogenic effects of in‐ovo administered metamizole (dipyrone), which can be used when needed during pregnancy and has potent analgesic, antipyretic, anti‐inflammatory, and long bone (tibia and femur) effects. This study used 240 fertile eggs from Atak S breed chickens, divided into eight equal groups: control, vehicle control, and 15.62, 31.25, 62.5, 125, 250 and 500 mg/kg metamizole. The eggs were hatched on the 21st day of incubation, and the chicks' body weights and mortality rates were determined. The right and left femur and tibia bones were resected from the chicks. Anatomical reference points were determined after removing the soft tissues of the bones, and necessary morphometric measures were taken from these points with a 0.01 mm precision using digital callipers. The 100% lethal dose (LD100) was identified in the highest examined dose (500 mg/kg) in the Chicken Embryotoxicity Screening Test (CHEST)‐I stage. The CHEST‐II stage determined the 50% lethal dose (LD50). Low-temperature flooding and ice encasement (LTFIE) cause variable survival of winter wheat (Triticum aestivum L.) in Ontario, which limits the adoption of wheat into crop rotations by growers. The development of novel cultivars capable of withstanding LTFIE is a promising avenue for improvement, but the methods used to assess the survival of winter wheat under LTFIE are restricted. This study developed updated methods to determine the survival of wheat cultivars under LTFIE using controlled environments and, to our knowledge, is the first method since the 1980s to use Canadian eastern soft red winter wheat (CESRW) to conduct cold tolerance studies. Chamber-acclimated plants of AC Carberry (spring wheat control), Branson (CESRW), CM614 (CESRW), and Norstar (hardy Canadian western red winter control) cultivars were used to estimate the days (LD50) and temperature (LT50) to reach 50% mortality under ice and without ice treatments. Norstar had the longest LD50 at 33 days, Branson and CM614 had similar LD50 of 18 and 20 days, and AC Carberry did not reach an LD50 as it died early in both treatments. The LT50 of each cultivar was different; Norstar had the lowest LT50 (−13.6 °C day 0 and −13.2 °C day 7), and AC Carberry had the highest LT50 (−6.6 °C day 0 and −2.7 °C day 7). The detailed methods developed in this study were more reliable compared to older methods based on the more accurate reported LD50 and LT50 of the cultivars, therefore, these methods can be used to screen winter cereals for LTFIE in the future. The investigation of "Inert Minerals and Botanicals Synergism with Spinosad: co-toxicity against Callosobruchus maculatus in Stored Blackgram" was performed in the Laboratory of the Department of Entomology, Agricultural College, Bapatla, during 2020-21. The inert minerals and botanicals were mixed with Spinosad and used for the bioassay. The data were recorded at 24, 48 and 72 hours after the release of the test insect, and probit analysis was performed to calculate the LD50. By using the LD50 of inert minerals and botanicals alone and in combination with spinosad, the co-toxicity coefficient was calculated. Among all the treatments, the highest co-toxicity coefficient (433.33) was recorded for diatomaceous earth, indicating increased efficacy and synergism between diatomaceous earth and Spinosad. Insecticide toxicity to insect herbivores has long been known to vary across different host plants; this phenomenon has been widely documented in both foliage-feeders and sap-feeders. Species-specific phytochemical content of hostplant tissues is assumed to determine the pattern of induction of insect enzymes that detoxify insecticides, but specific phytochemicals have rarely been linked to host plant-associated variation in pesticide toxicity. Moreover, no studies to date have examined the effects of nectar source identity and phytochemical composition on the toxicity of insecticides to pollinators. In this study, we compared LD50 values for the insecticide bifenthrin, a frequent contaminant of nectar and pollen in agroecosystems, in the western honey bee, Apis mellifera, consuming three phytochemically different monofloral honeys: Nyssa ogeche (tupelo), Robinia pseudoacacia (black locust), and Fagopyrum esculentum (buckwheat). We found that bifenthrin toxicity (LD50) values for honey bees across different honey diets is linked to their species-specific phytochemical content. The profiles of phenolic acids and flavonoids of buckwheat and locust honeys are richer than is the profile of tupelo honey, with buckwheat honey containing the highest total content of phytochemicals and associated with the highest bifenthrin LD50 in honey bees. The vector fitting in the ordination analysis revealed positive correlations between LD50 values and two honey phytochemical richness estimates, Chao1 and Abundance-based Coverage Estimator (ACE). These findings suggest unequal effects among different phytochemicals, consistent with the interpretation that certain compounds, including ones that are rare, may have a more pronounced effect in mitigating pesticide toxicity. The purpose of the research is to evaluate some toxicological parameters of antiparasitic selamectin-based drug.Materials and methods. The study drug is a solution for external use, 1 mL of which contains the active component selamectin (120 mg), as well as additives. The experiments used a total of 66 white outbred male rats, 42 white outbred male mice and 3 guinea pigs. The parameters of acute oral toxicity (in mice and rats), acute dermal toxicity (in rats), and dermal and mucous membrane irritation (in rats and guinea pigs) of antiparasitic selamectin-based drug were evaluated. LD50 was calculated and the irritation of the drug was evaluated using established procedures in toxicology.Results and discussion. The maximum tolerated dose of the selamectin-based drug was 2550 mg/kg in the mice and 4250 mg/kg in the rats. The absolutely lethal dose for the mice and the rats was 6800 and 8500 mg/kg, respectively. It was established that the drug belonged to hazard class 3 (moderately hazardous substances) when administered intragastrically to the animals: LD50 for the mice was 4816.7 mg/kg, and LD50 for the rats was 6091.7 mg/kg. In determining acute dermal toxicity in the rats, the drug was classified as hazard class 4 (low-hazardous substances): LD50 more than 8500 mg/kg. The drug irritant effect on the skin of the rats at doses of 4250, 6375 and 8500 mg/kg was not established. The drug had a mild effect on the mucous membrane of the eyes of the guinea pigs with its recovery to normal within 24 hours.

Introduction: The intergovernmental organizations Organisation for Economic Co-operation and Development (OECD) and Interagency Coordinating Committee on the Validation of Alternative Methods (ICCVAM) have developed guidelines for the use of in vitro models for toxicological evaluation of chemicals. However, the presence of manual steps and the requirement of multiple tools for data analysis, apart from being costly and time-consuming, can inadvertently introduce errors by researchers. Objectives: We have developed the SAEDC platform (Technological Solution for Exploratory Data Analysis and Statistics for Cytotoxicity, in Portuguese), which enables analysis of cytotoxicity data from assays following OECD Guideline No. 129. Methodology: In vitro experimental data were used to compare with the analysis methodology suggested in the Guideline. We analyzed 117 data sets covering chemicals from Category I to Unclassified according to GHS classification. Results: The four-parameters of non-linear regression (4PL) calculated by the SAEDC platform showed no significant differences compared to standard methodology in any of the data sets (p > 0.05). The coefficient of determination (R-squared) also demonstrated not only a good fit of the 4PL model to the data but also significant similarity to values obtained by the conventional methodology. Finally, the SAEDC platform predicted LD50 values for the chemicals from IC50, using the Registry of Cytotoxicity (RC) regression models. Conclusion: The comparison with the standard data analysis methodology revealed that SAEDC platform fulfills the requirements for cytotoxicity data analysis, generating reliable and accurate results with fewer steps performed by researchers. The use of SAEDC platform for obtaining toxicity values can reduce analysis time compared to the standard methodology proposed by regulatory agencies. Thus, automation of the analysis using the SAEDC platform has the potential to save time and resources for cytotoxicity researchers and laboratories while generating reliable results.

Enterococcus faecalis is a newly emerged fish pathogen affecting many cultured fish species. The present work aimed to determine the cause of mortalities affecting the cultured Meagre farm and study the infectious agent virulence, disease pathology. Also, to determine the influence of infection on complement 3, cyclooxygenase, and lysozyme gene expression and concentrations in fish serum. The internal organs of diseased fish were severely congested with the presence of petechial haemorrhages on the liver, gonads, and all over the elementary tract. Thirty-one bacterial isolates were recovered from naturally infected fish; seventeen were identified biochemically as E. faecalis. Random E. faecalis isolate was verified for pathogenicity in meager fingerlings and was indeed pathogenic with lethal dose-fifty (LD50) estimated by 4.6 × 106 colony forming unit (CFU) fish−1. The posterior kidney and hepatopancreas of experimentally infected fish showed severe inflammation, necrosis, degeneration, and haemorrhage. Detachment of mucosa lining the stomach and intestine (intestinal villi and gastric glands) was prominent. The examined gastric tissues exhibited inflammation, leucocytic infiltration, and congestion. The expression of C3, Cox2, and Lyz was upregulated 24-h post-infection and then decreased near the baseline at 96 h. Serum C 3 and Lyz concentrations peaked at 24 h and 48 h after that decreased. IgM level gradually increased with time and peaked at 96 h. In conclusion, E. faecalis is assumed pathogenic for cultured meager, experimental infection stimulated immune systems and upregulated the expression of some immune-related genes. The purpose of the research is to evaluate some toxicological parameters of a new combined anthelmintic based on oxantel pamoate, pyrantel pamoate and praziquantel in tablet formulation for dogs and cats.Materials and methods. The studies used 70 outbred male rats and 50 outbred male mice. Acute oral toxicity was studied in male mice and male rats weighing 14–16 and 160–180 g, respectively. The subchronic toxicity study with the drug administered repeatedly, orally for 14 days was conducted on male rats weighing 180–200 g. The study monitored the laboratory animals’ overall health status, behavior and possible death, and any intoxication symptoms were recorded. All studies were performed using conventional techniques for experimental (preclinical) studies of new pharmacological substances using the guidelines edited by R. U. Khabriev (2005) and A. I. Mironov et al. (2012). Additionally, the experiments assessed clinical toxicological, biochemical, and morphological parameters of the laboratory animals.Results and discussion. In evaluation of the drug acute toxicity in the male mice and the female rats, the LD50 exceeded the dose of 6000 mg/kg; the animals showed no signs of intoxication during the entire study. Thus, the drug was classified as substance hazard category 4. Doses 600, 300 and 120 mg/kg of the drug had no negative effect on the organism of the laboratory animals in the subchronic experiment on the male rats; the doses were ineffective (safe). Herbal plants are widely used as traditional medicine for generations. Some of them are bitter, in Indonesian called Sambiloto (Andrographis paniculata), red ginger in Indonesian called Jahe merah (Zingiber officinale var rubrum), and turmeric in Indonesian called Kunyit (Curcuma domestica). This study tested extracts containing a combination of three ingredients, hereinafter named 'SIJAKUN' extract (SambIloto, JAhe, KUNyit)". This combination has good benefits for the body, especially as an anti-inflammatory and antioxidant. This study aims to measure the acute and subacute toxic effects of 'SIJAKUN' extract. This is a true experimental study. In the acute toxicity study, the SIJAKUN extract was given to 5 groups of mice a doses of 1000 mg/kg, 2,000 mg/kg, 3,000 mg/kg, 4,000 mg/kg, and 5,000 mg/kg. After being observed for 24 hours, the number of dead mice from each group was counted to calculate the LD50 of the 'SIJAKUN' extract. Whereas in subacute toxicity, 5 groups of mice were given 0.5 ml of CMCNa solution, 'SIJAKUN' extract at doses of 25 mg/kg, 75 mg/kg, 150 mg/kg, and 25 mg/kg ibuprofen, the test substance was given orally once daily for 28 days. Then an examination was carried out: SGOT, SGPT, BUN, and serum creatinine. the results of subsequent examinations were analyzed statistically. To assess the safety of the SIJAKUN combination, it is necessary to conduct research to determine the acute and sub acut toxicity of SIJAKUN to the liver and kidneys. Based on the results of acut toxixity study conducted, no experimental animals died at one dose of 1000, 2000, 3000, 4000 and 5000 mg/kgbb 'SIJAKUN' extract. Based on these results, the LD50 of 'SIJAKUN' extract was above 5000 mg/kgbb. Therefore, it can be said that 'SIJAKUN' extract is a non-toxic compound. In the subacute toxicity study, examination result of SGOT, SGPT, BUN, and serum creatinin was not change significantly, with P value > 0.05. This result indicating no significant difference between the control group and the treatment group. It can be said that the sub-acute toxicity test found that 'SIJAKUN' extract did not toxic for liver and kidney. In acute and subacute toxicity studies that has been carried out, 'SIJAKUN' extract was classified as a non-toxic compound, so it is safe to use as herbal medicine. [ABSTRACT FROM AUTHOR]

This investigation is intended to isolate, identify, and assess the pathogenicity of Edwardsiella tarda, which originated from diseased clown knifefish. A total of 43 isolates were obtained from infected fish samples in Hau Giang and Dong Thap provinces of the Mekong Delta, Vietnam. Two isolates of DT37 and HG41 were identified as E. tarda by morphological, biochemical, and 16S rRNA gene sequencing. Experimental challenge studies revealed that isolate DT37 leads to 83.33% at a 108 CFU/ml concentration after 60 hours. Meanwhile, in isolate HG41, mortality reached 100% within 48 hours post-injection at the highest concentration of 108 CFU/ml. The challenged clown knifefish exhibited gross signs of abnormal swimming, skin ulcerations, and petechial hemorrhages in the body. Internally, ascites with hemoperitoneum, light-colored nodules on the liver, hemorrhagic kidneys, and splenomegaly were also recorded. The LD50 of two isolates, DT37 and HG41, was 4.89 x 105 and 4.07 x 105 CFU/ml, respectively. The antibiogram result showed that most of the isolates were highly susceptible to ampicillin (65%), enrofloxacin (85%), florfenicol (100%), flumequine (90%), cefotaxime (80%), and trimethoprim and sulfamethoxazole (70%). Chickpea (Cicer arietinum L.) production in Ethiopia has been seriously constrained by the insect pest, Helicoverpa armigera. Insecticides have been used for decades to control this noxious pest. However, the restriction on chemical insecticides necessitates biological pest control alternatives. Therefore, the present study investigated the effectiveness of native Bacillus thuringiensis (Bt) isolates against Helicoverpa armigera 3rd instar larvae. The treatments consisted of Bt concentrations at the LD90 values, whereas the control plots were sprayed with distilled water and a commercial Bt. var. thuringiensis product. The results revealed statistically (p < 0.05) significant differences between the native Bt isolates and the control plots. Post 1st, pre 2nd and post 2ndBt sprayed larval populations counted per plant ranged from 0.8 ± 0.3–3.1 ± 0.1, 0.6 ± 0.1–3.6 ± 0.0 and 0.5 ± 0.1–3.6 ± 0.4, respectively. Pod damage (%) and grain yield (t/ha) ranged between 9.7 ± 3.1 and 47.5 ± 9.5, 1.0 ± 0.0 and 2.6 ± 0.5, respectively compared to 47.5 ± 9.5% pod damage and 1.0 ± 0.0 (t/ha) grain yield from the control plots. These results indicate the potential of these bio-insecticides in the management of chickpea pod borer. Fentanyl overdose is a survivable condition that commonly resolves without chronic overt changes in phenotype. While the acute physiological effects of fentanyl overdose, such as opioid-induced respiratory depression (OIRD) and Wooden Chest Syndrome, represent immediate risks of lethality, little is known about longer-term systemic or organ-level impacts for survivors. In this study, we investigated the effects of a single, bolus fentanyl overdose on components of the cardiopulmonary system up to one week post. SKH1 mice were administered subcutaneous fentanyl at the highest non-lethal dose (62 mg/kg), LD10 (110 mg/kg), or LD50 (135 mg/kg), before euthanasia at 40 min, 6 h, 24 h, or 7 d post-exposure. The cerebral cortex, heart, lungs, and plasma were assayed using an immune monitoring 48-plex panel. The results showed significantly dysregulated cytokine, chemokine, and growth factor concentrations compared to time-matched controls, principally in hearts, then lungs and plasma to a lesser extent, for the length of the study, with the cortex largely unaffected. Major significant analytes contributing to variance included eotaxin-1, IL-33, and betacellulin, which were generally downregulated across time. The results of this study suggest that cardiopulmonary toxicity may persist from a single fentanyl overdose and have wide implications for the endurance of the expanding population of survivors. The article presents a study which examined the presence of Nucleopolyhedrovirus (NPV) infected Spodoptera frugiperda larvae in order to manage the invasive pest the fall armyworm, Spodoptera frugiperda in north India. Topics discussed include isolation and purification of polyhedral occlusion bodies (POBs), evaluation of the most potent virus isolate, and natural incidence of NPV in Spodoptera frugiperda larvae collected from different zones of Jammu and Kashmir, India.

This study marks the first occasion that Streptococcus iniae has been isolated, identified, and characterized as the causative pathogen in spotted sea bass (Lateolabrax maculates). Infected fish exhibited a range of external symptoms, including scale loss, bleeding from the jaw, anus, and tail, among other signs, as well as internal manifestations such as congested liver, splenomegaly, branchial anemia, yellow fat syndrome, and intestinal edema. Notably, exophthalmia and meningoencephalitis—typical symptoms associated with previous S. iniae infections—were not observed. A predominant bacterial isolate (designated 10S01) was recovered from the pure culture of spleen of a diseased spotted sea bass in Zhuhai, China. The strain was then subjected to Gram staining, biochemical profiling, and molecular confirmation through 16S rRNA and gyrB gene, corroborating its identity as S. iniae. Pathogenicity was assessed by intraperitoneal injection challenge in spotted sea bass weighing approximately 13 g/fish, revealing a LD50 of 74 cfu/g-fish. The 10S01 strain demonstrated the ability to colonize various organs, including the spleen, liver, kidney, and brain, with a relatively higher affinity for the spleen. Furthermore, antimicrobial susceptibility testing indicated that the 10S01 strain was sensitive to 14 tested antibiotics, particularly chloramphenicol, ciprofloxacin, clarithromycin, florfenicol, ofloxacin, rifampicin, and trimethoprim/sulfamethoxazole, highlighting these as preferred treatments for S. iniae infections in spotted sea bass. These findings contribute significantly to our understanding of S. iniae pathogenesis and inform the prompt and appropriate antibiotic treatment of S. iniae infections. [ABSTRACT FROM AUTHOR]

RESEARCH HIGHLIGHTSThe pathogenicity of avian pathogenic Escherichia coli (APEC) can be reliably determined using a biological model with 1-day-old chicks. However, public concerns regarding the use of vertebrates in research have led to the exploration of other biological models. Alternative infection models, including Galleria mellonella larvae, have also been studied. This study aimed to evaluate whether G. mellonella larvae are a viable in vivo model for evaluating the pathogenicity of APEC isolates and to determine the best protocol for this model. First, the pathogenicity index (PI) in vivo was determined for 40 APEC isolates from 1-day-old chicks. The same isolates were then used to establish the best protocol for the determination of PI in larvae of G. mellonella by determining the LD50, melanization, cytotoxicity test, total haemocyte count, mortality rate, construction of survival curves, and larval disease score. Cytotoxicity tests and total haemocyte counts were not suitable for distinguishing the pathogenicity groups. LD50 was a useful tool for differentiating between high and low pathogenicity but not for intermediate pathogenicity. Similarly, melanization could be used as a variable to characterize the pathogenicity of APEC isolates but should be associated with other variables. Mortality rates and survival curves were useful tools for characterizing nonpathogenic and highly pathogenic isolates. Streptococcosis, due to Streptococcus agalactiae, has become a serious challenge for aquaculture around the world. Therefore, the main purpose of this work was to confirm the etiological agent that caused "dark body" disease in snakeskin gourami (Trichogaster pectoralis) cultured in the Mekong Delta, Vietnam. Infected fish displayed clinical signs, including anorexia, lethargic swimming on the water surface, corneal opacity, and hemorrhage in the base of the fin. Remarkably, abnormal black coloration on the body and serious hemorrhage at the base of the pectoral fin, and on the body were found in infected fish. In total, 75 bacterial strains were isolated from different diseased fish farms. Based on morphological and physiological characteristics, the API 20 Strep test, and 16S rRNA sequence analysis, the result illustrated that the bacterial isolates were identified as S. agalactiae. Additionally, antibiotic sensitivity testing revealed that all the S. agalactiae isolates were sensitive to amoxicillin, rifampicin, clarithromycin, erythromycin, doxycycline, cephalexin, novobiocin, and sulfamethoxazole-trimethoprim. Interestingly, S. agalactiae strains were only resistant to gentamycin in this study. Two strains, SRT41 and SRT43, carried out an experimental challenge with concentrations from 104 to 107 CFU/ml in healthy snakeskin gourami, and the LD50 values of the isolates, SRĐT41 and SRĐT43, were determined to be 2.15 × 105 and 3.59 × 103 CFU/ml, respectively, after 7 days. Enrofloxacin is a broad-spectrum synthetic antimicrobial drug widely used in veterinary medicine. The present study aimed to determine the effective enrofloxacin dose for treating Pseudomonas aeruginosa and Enterococcus faecalis infection in Oreochromis niloticus. P. aeruginosa and E. faecalis isolates were verified using selective differential media and biochemically using the Vitek 2 test. Bacterial isolates were virulent for O. niloticus with LD50 equal to 2.03 × 106 and 2.22 × 107 CFU fish−1 for P. aeruginosa and E. faecalis, respectively. Infected fish suffered from decreased feed intake followed by off-food, tail erosion, darkening of the external body surface, exophthalmia, ascites, and loss of escape reflex. Internally, congested hemorrhagic hepatopancreas with engorged distended gall bladder were dominant. The posterior kidney was congested with enlarged spleen, and empty elementary tract. Pathologically, severe degenerative changes were dominant in the hepatopancreas, posterior kidney, spleen, stomach, and gills of infected fish. Antimicrobial sensitivity test indicated the high susceptibility of P. aeruginosa and E. faecalis to enrofloxacin with MIC estimated at 1 and 0.0625 µg/mL, respectively. Enrofloxacin effectively protected O. niloticus against E. faecalis and P. aeruginosa infection when used with medicated feed at doses of 10 and 20 mg kg−1 body weight. [ABSTRACT FROM AUTHOR]

The use of disinfectants is a crucial aspect of preventive and health improvement measures for infectious diseases in farm and domestic animals. Regulatory documents require the determination of toxicity to macroorganisms, including the establishment of lethal doses and toxicity groups, during the development and registration of antimicrobial agents. This study aimed to investigate the histological changes in the internal organs of laboratory animals when determining lethal doses of innovative disinfectants. The experiments used domestic disinfectants containing glutaraldehyde as the active ingredient. Histological studies were conducted on the internal organs (kidney, liver, stomach, intestines, and spleen) of 75 laboratory animals using an Axioskop 40/40FL microscope (Carl Zeiss, Germany) with video microscopic photography. The methods used were under current standards. Hemodynamic disorders were observed in the renal tissue under the influence of lethal doses of aldehyde disinfectants. These disorders were characterized by capillary dilation and blood filling. Glomerular capillary dilation and overflow with blood cells were also detected. Additionally, stasis was observed in the lumen of the microcirculatory vessels throughout the entire length. The examination of histological sections from animal liver samples revealed a significant expansion of Dissé spaces, variations in the size of hepatocyte nuclei, beam decomposition and fragmentation, small acute perivascular hemorrhages, leukostasis in sinusoids, and hemodynamic disorders. The structure of the organ's beams was also disturbed, and a significant number of venous vessels were dilated and excessively filled with blood cells. Minor changes were detected in the stomach, including desquamation of the epithelial cells of the glands and their exfoliation into the gastric lumen, as well as circulatory disorders. Epithelial desquamation, blood vessel dilation, and signs of connective tissue edema were observed in the intestine. The kidneys exhibited signs of acute venous hemorrhage and stasis in vessels of various calibers, with the development of small acute parenchymal hemorrhages and localized lymphoid cell death in the white pulp. The prospect of further research is to investigate the histomorphological changes in the internal organs of laboratory animals when exposed to modern complex disinfectants with different active ingredients.

S. agalactiae infection is one of the most important bacterial diseases imparting mass mortality in the aquaculture sector, worldwide. It is responsible for posing both economic burdens and risks of cross-infectivity and zoonotic transmission. In this aspect, this study examined the virulence gene profiles, molecular serotyping and pathogenicity of S. agalactiae strains isolated from tilapia farms in Kerala, India in 2022. Following sample collection and processing, three S. agalactiae strains (KSA/01, KSA/02, and KSA/03) were isolated and confirmed the bacterial identity using 16S rRNA sequencing. In pathogenicity test, the LD50 of the S. agalactiae was found to be 1 × 106 CFU/ml. An increase in the salinity was found to affect the pathogenicity of S. agalactiae as well as the hematological and serological profile of tilapia. Thus the presence of S. agalactiae, a highly virulent and zoonotic pathogen in tilapia farms, underscores the imperative of continuous monitoring in aquaculture facilities.

This work includes the synthesis of new ester compounds containing two 1,3,4- oxadiazole rings, 15a-c and 16a-c. This was done over seven steps, starting with pacetamido-phenol 1 and 2-mercaptobenzoimidazole 2. The structure of the products was determined using FT-IR, ¹H NMR, and mass spectroscopy. The evaluation of the antimicrobial activities of some prepared compounds was achieved against four types of bacteria (two types of gram-positive bacteria; Staphylococcus aureus and Bacillus subtilis, and two types of gram-negative bacteria, Pseudomonas aeruginosa and E. Coli), as well as against one types of fungus (C. albino). The results show moderate activit against the study bacteria, and the theoretical analysis of the toxicity demonstrates non-toxicity with acute oral human toxicity. The impact of Origanum compactum essential oil (EO) at 80 mg kg−1 on Triton WR-1339-induced hyperlipidemia in rats was investigated.The EO's phytochemical composition revealed the predominance of carvacrol (35.71%), p-cymene (13.74%), and carvacrol methyl ether (11.71%). Vibrio harveyi, a recently discovered pathogenic bacterium isolated from American eels (Anguilla rostrata), poses uncertainties regarding its pathogenesis in American eel and the molecular mechanisms underlying host defense against V. harveyi infection. This study aimed to determine the LD50 of V. harveyi in American eel and assess the bacterial load in the liver, spleen, and kidney post-infection with the LD50 dose. The results showed that the LD50 of V. harveyi via intraperitoneal injection in American eels over a 14d period was determined to be 1.24 × 103 cfu/g body weight (6.2 × 104 cfu/fish). The peak bacterial load occurred at 36 h post-infection (hpi) in all three organs examined. Histopathology analysis revealed hepatic vein congestion and thrombi, tubular vacuolar degeneration, and splenic bleeding. Moreover, quantitative reverse transcription polymerase chain reaction (qRT-PCR) results indicated significant up or downregulation of 18 host immune- or anti-infection–related genes post 12 to 60 hpi following the infection. Additionally, RNA sequencing (RNA-seq) unveiled 7 hub differentially expressed genes (DEGs) and 11 encoded proteins play crucial roles in the anti-V. harveyi response in American eels. This study firstly represents the comprehensive report on the pathogenicity of V. harveyi to American eels and RNA-seq of host's response to V. harveyi infection. Mosquitofish, Gambusia affinis, are eponymous larval mosquito predators. Their ability to colonize and survive in habitats that are uninhabitable by other potential predators allows them to naturally manage larval mosquito populations in most ground pools they are present in. However, effluent from residential onsite wastewater treatment systems (OWTSs) appears to limit the presence of fish predators. This is especially problematic in Louisiana, where regulations allow the discharge of OWTS effluent into open drainage conveyances. To determine the effect of effluent on the capacity of mosquitofish for biocontrol in contaminated areas, we assessed the body condition metrics of populations from two effluent-exposed sites and two sites not exposed to effluent, determined the lethal effect of effluent-contaminated drainage water on fish, and measured the prey consumption rates in the presence of effluent. Female fish collected from effluent-impacted sites had a reduced somatic body condition and most females examined displayed masculinized anal fins resembling the male gonopodium structure. This trait was not seen in fish collected from the control sites and has not yet been documented in association with OWTSs or in the state of Louisiana. Fish from the control sites survived at effluent-contaminated water levels < 70%, and the prey clearance rates increased with dilution. Onsite wastewater treatment system effluent has significant effects on both the short- and long-term persistence of mosquitofish, their body composition, reproductive health, and larval mosquito consumption. In order to have a deep understanding of rabbit Bordetella bronchiseptica (Bb), the strain identification, growth curve determination, pathogenicity study, drug susceptibility test, drug resistance test and gene detection were expected to understand the growth pattern of the bacteria and lay a foundation for clinical drug use and prevention and control. The isolates were identified by Gram staining, microscopy, isolation and culture, 16S rDNA sequencing analysis; the growth curve, LD50, drug resistance of the isolates were studied and the isolates were tested for β-lactamase resistance gene. A strain of Gramnegative Bacillus, named P201215, was isolated, and the 16S rDNA gene sequence of the isolate had 99% homology with that of Bordetella bronchiseptica (accession number: NR113628.1) in GenBank. The results showed that the logarithmic growth period of the isolate was 2-9 h, and it entered the plateau period after 9 h. The LD50 of the isolate was 1.19× 107 CFU in KM mice, and the results of the drug sensitivity test showed that the isolate was resistant to some cephalosporins and sensitive to most β-lactams, quinolones, tetracyclines and aminogly-cosides, and no extended-spectrum β -lactamase resistance gene was detected by PCR. The present study focuses on investigating the interaction between radiation and biological systems, specifically examining the detrimental effects of radiation. This approach has proven to be effective in determining the optimal tolerance range of radiation on plants, which can lead to the development of beneficial characteristics and help improve the narrow genetic base. The study was conducted on Salvia hispanica L., a medicinally important plant. Inbred seeds of Salvia hispanica L. were subjected to five different doses of gamma rays (50, 100, 150, 200, 250 Gy) using a Co-60 source with a gamma irradiator of 7.247 k rate. The seeds were sown in triplicate, along with a control set. Various characteristics were assessed, including germination percentage, survival percentage, presence of chlorophyll mutants, and plant height. The results indicate that lower doses of gamma irradiation (LD50) have detrimental effects on several plant attributes, such as seed germination, survivability, inflorescence size, and pollen fertility. Chromosomal abnormalities were also observed, with an increase in their occurrence corresponding to higher doses of gamma radiation. Germination percentage and survivability were found to decrease as the dose of gamma radiation increased. The dose of 50 Gy resulted in the highest level of genetic variability. Additionally, compared to the control group, a significant percentage of chromosomal abnormalities, particularly stickiness and scattering, was observed at a dose of 250 Gy. Lower doses of gamma radiation (specifically 50 and 100 Gy) showed statistically significant (p>0.5) positive responses in Salvia plants, including plant height, leaf area, leaf mutants, inflorescence axis, and seed size. On the other hand, higher doses of radiation proved to be fatal for the plants. In summary, this study provides valuable insights into the effects of gamma radiation on Salvia hispanica L., highlighting the optimal dose range for inducing beneficial changes in plant characteristics while also emphasizing the harmful consequences associated with higher doses of radiation. Determining the optimum dose of a mutagen that causes a high frequency of favourable mutations with minimum damage to the plant is the vital criterion for the success of a mutation breeding programme. We evaluated the comparative effectiveness, radiosensitivity and relative biological effectiveness (RBE) of three physical mutagens, viz., electron beams, X-rays and gamma-rays, on twenty indigenous aromatic Joha rice cultivars of Assam. We exposed dry uniform seeds of 12.4% moisture content to 16 different mutagens doses ranging from 50 to 800 Gy at an interval of 50 Gy. Key results: Germination percentage and the seedling traits registered a decreasing trend towards increasing the mutagen doses in all the cultivars, showing differential radiosensitivities. The LD50 values ranged from 515 to 615 Gy for electron beams and 421 to 537 Gy for X-rays and 414 to 481 Gy for gamma-rays. The GR50 values based on seedling vigour index I ranged from 125 to 304 Gy for electron beams, 191 to 313 Gy for X-rays and 178 to 267 Gy for gamma-rays. The optimum dose ranges for electron beams, X-rays and gamma-rays are 241 to 337 Gy, 228 to 324 Gy and 250 to 346 Gy, respectively. The cultivar Soru Joha-Tinsukia was the most radio tolerant, followed by Keteki Joha and Local Joha, while the cultivar Kon Joha-Moran showed the least tolerance. Joha rice cultivars showed more radiosensitivity for X-rays, followed by electron beams and gamma-rays. Similarly, a high RBE was evident for X-rays, followed by electron beams and gamma-rays, indicating their decreasing penetration capacity and lethality trend. Background: Kashmir Himalaya hosts the most diverse and rich flora in the world, which serves as grazing land for millions of small ruminants in the area. While most plant species are beneficial, some can be poisonous, causing economic losses and animal health issues for livestock. Consequently, this study is the first comprehensive report on the traditional phyto-toxicological knowledge in District Muzaffarabad and the assessment of its authenticity through experimental studies in rats. Methods: The data regarding traditional knowledge was gathered from 70 key respondents through semi-structured interviews, which was quantitatively analyzed and authenticated through plant extract testing on Wistar female rats and comparison with published resources. Results: A total of 46 poisonous plant species belonging to 23 families and 38 genera were reported to be poisonous in the study area. Results revealed that leaves were the most toxic plant parts (24 species, 52.1%), followed by the whole plant (18 species, 39.1%), stem (17 species, 36.9%), and seeds (10 species, 21.7%). At the organ level, liver as most susceptible affected by 13 species (28.2%), followed by the gastrointestinal tract (15 species, 32.6%), nervous system (13 species, 8.2%), dermis (8 species, 17.3%), renal (7 species, 15.2%), respiratory (4 species, 8.7%), cardiovascular system (3 species, 6.5%), and reproductive system (2 species, 4.3%). The poisonous plant species with high Relative frequency citation (RFC) and fidelity level (FL) were Nerium oleander (RFC, 0.6; FL, 100), Lantana camara (RFC, 0.6; FL, 100), and Ricinus communis (RFC, 0.6; FL, 100). Experimental assessment of acute toxicity assay in rats revealed that Nerium oleander was the most toxic plant with LD50 of (4,000 mg/kg), trailed by Ricinus communis (4,200 mg/kg), L. camara (4,500 mg/kg), and Datura stramonium (4,700 mg/kg); however, other plants showed moderate to mild toxicity. The major clinical observations were anorexia, piloerection, dyspnea, salivation, tachypnea, constipation, diarrhea, tremor, itchiness, and dullness. Conclusion: This study showed that numerous poisonous plants pose a significant risk to the livestock industry within Himalayan territory, leading to substantial economic losses. Zika virus (ZIKV) is a positive-sense single-stranded virus member of the Flaviviridae family. Among other arboviruses, ZIKV can cause neurological disorders such as Guillain Barré syndrome, and it can have congenital neurological manifestations and affect fertility. ZIKV nonstructural protein 5 (NS5) is essential for viral replication and limiting host immune detection. Herein, we performed virtual screening to identify novel small-molecule inhibitors of the ZIKV NS5 methyltransferase (MTase) domain. Compounds were tested against the MTases of both ZIKV and DENV, demonstrating good inhibitory activities against ZIKV MTase. Extensive molecular dynamic studies conducted on the series led us to identify other derivatives with improved activity against the MTase and limiting ZIKV infection with an increased selectivity index. Preliminary pharmacokinetic parameters have been determined, revealing excellent stability over time. Preliminary in vivo toxicity studies demonstrated that the hit compound 17 is well tolerated after acute administration. Hafnium is a promising element as contrast agent for diagnostic and therapeutic radiology. Currently there are no suitable hafnium drugs with renal excretion which can be used in radiology. In this work two new hafnium complexes with nitrilotriacetic acid (Hf-NTA) and 1,2-diaminocyclohexanetetraacetic acid (Hf-CDTA) were synthesized and studied for acute toxicity and biodistribution. Inorganic chemistry methods were used for Hf-NTA and Hf-CDTA synthesis. Acute toxicity was studied in female mice C57Bl/6. Median lethal dose (LD50) of Hf-CDTA was assessed as 408 ± 64 mg Hf/kg and of Hf-NTA less than 120 mg Hf/kg. Biodistribution and contrast properties studies of Hf-CDTA were performed in vivo using micro-CT in mice with transplanted subcutaneous mammary adenocarcinoma Ca755. CT imaging of mice intravenously injected with Hf-CDTA confirmed its renal excretion and contrast capability. CT imaging of tumor region with single intratumoral administration of Hf-CDTA showed promising hafnium concentration and retention in tumor for use in contrast-enhanced radiotherapy. Hf-CDTA showed acceptable toxicity and biodistribution in mice with subcutaneous tumors for biomedical application in radiology and radiotherapy. For diagnostic clinical application, the Hf-CDTA formulation must be improved to increase water solubility and decrease toxicity. Background: Avocado (Persea americana) fruit has a high oil content, so it is widely used in the pharmaceutical and cosmetic industries. Objective: This study aims to determine the toxicity of avocado oil in mice using the lethal dose (LD50) method so that it can be used as a reference for further testing. Methods: This study used a total of 20 DDY strain female mice, which were divided into 5 groups: one control group and four treatment groups that were fed with avocado oil with 5, 10, 15, and 20 g/kg BW doses orally. The mortalities of experimental mice were observed for 14 days after treatment. Other parameters observed in this study were physiological response, body weight, absolute organ weight, and relative organ weight. Results: There was a change in behavior, and the obtained LD50 value was 25.4 g/kg BW. Observation of physiological responses, body weight, absolute organ weights, and relative organ weights showed no significant differences. Conclusion: It was concluded that avocado oil is considered relatively harmless and safe to use.

The bacterium Francisella halioticida, known to induce francisellosis in abalone and Yesso scallop, is suspected of being involved in the blue mussel mortalities observed in France. Recently, several isolates of F. halioticida were obtained from moribund mussels and categorized into two type strains, FR21 and FR22. Two other strains, AG1 and AG3, determined to belong to the genus Francisella were isolated. To determine the virulence of these isolates, juvenile and adult blue mussels were injected with bacterial solutions at high dose and monitored for 11 days. FR22c and FR22d were found to induce 80% mortality in less than seven days. The isolates AG1 and AG3 led to over 50% mortality in adult mussels but only AG1 led to significant mortality in juveniles (41%). FR22c and FR22d, the most virulent isolates, were selected to determine their respective lethal dose at 50% (LD50) in juveniles and adults. This analysis was performed with bacterial solutions ranging from 102 to 106 CFU/mussel and monitored for 30 days. The isolate FR22c was found to be the most virulent. Observed LD50 for the isolate FR22c was 4.14 ×103 CFU/juvenile and 3.45 ×103 CFU/adult and for the isolate FR22d, 1.89 ×104 CFU/juvenile and 1.52 ×104 CFU/adult. To confirm Koch’s postulate, a selection of moribund, surviving and control animals were plated on specific media. The isolates were reisolated from moribund animals but not from surviving or controls. To confirm the proliferation, a specific real-time PCR was performed. All moribund individuals were positive by PCR. The main Ct values were lower for moribund compared to surviving animals and a dose effect was observed in DNA bacterial load. This study shows that some F. halioticida isolates are able to induce francisellosis in mussels and lead to high mortality, highlighting differences in virulence among the strains.

Rice (Oryza sativa L.) plays a pivotal role in global food security, necessitating continuous efforts to enhance yield potential and grain quality. Mutation breeding, employing physical (electron beam) and chemical (ethyl methane sulphonate, EMS) mutagens, offers a promising avenue for creating genetic variability. This study assessed the sensitivity of rice variety ASD 16 and landrace Norungan to varying doses of electron beam (100 Gy to 500 Gy) and EMS (30 mM to 70 mM). The genotypes exhibited dose-dependent responses in germination, seedling survival, shoot and root length, seedling height, pollen fertility and spikelet fertility. The LD50 values for electron beam under in vivo and in vitro conditions were 314.24 Gy and 348.21 Gy respectively for ASD 16 and 445.07 Gy and 461.88 Gy for Norungan whereas for EMS, the LD50 values for ASD 16 were 49.17 mM and 55.37 mM; 53.53 mM and 65.12 mM for Norungan under in vivo and in vitro conditions based on probit analysis. ASD 16 showed a GR50 value of 309.02 Gy for the electron beam and 51.60 mM for EMS while Norungan exhibited a GR50 value of 538.79 Gy for electron beam and 98.71 mM for EMS. The variety, ASD 16 demonstrated higher sensitivity, proved by greater growth reduction percentages and lower lethal dose 50 (LD50) values compared to landrace Norungan. Electron beam treatments induce significant reductions in various growth traits, underlining the potential deleterious effects at higher doses. EMS, as a chemical mutagen, also impacted germination and growth characteristics. Both mutagens affected pollen and spikelet fertility with notable influence on yield contributing traits. This finding emphasizes the significance of precise dosage selection in mutation breeding to induce desirable traits and maintaining complete plant viability and productivity. Understanding genotype-specific responses is crucial for successful mutation breeding programmes. Overall, this study offers valuable insights into the application of electron beam and EMS mutagenesis in rice, contributing to the optimization of mutagenic protocols for crop improvement.

Snakebite envenoming (SBE) remains a severely neglected public health issue, particularly affecting tropical and subtropical regions, with Africa experiencing an estimated 435,000 to 580,000 snakebites annually, leading to high morbidity and mortality rates, especially across Africa and Asia. Recognized as a Neglected Tropical Disease, SBE management is further complicated by the inadequate efficacy of current antivenom treatments. Of particular concern are cobras (Naja sp.), whose neurotoxins can induce rapid fatal respiratory paralysis. In this study, we investigate the potential of nanobodies as a promising next-generation of immunotherapeutics against cobra venoms. Through a dual strategy of the characterization of venom toxic fractions from cobras captured for the first time in Algeria and Tunisia biotopes, coupled with in vitro assays to evaluate their interactions with acetylcholine receptors, and subsequent immunization of dromedaries to produce specific nanobodies, we identified two lethal fractions, F5 and F6, from each venom, and selected five nanobodies with significant binding and neutralizing of 3DL50 (0.74 mg/kg). The combination of these nanobodies demonstrated a synergistic effect, reaching 100% neutralizing efficacy of 2DL50 lethal venom fraction (0.88 mg/kg) doses in mice. Additionally, our findings highlighted the complex mechanism of cobra venom action through the lethal synergism among its major toxins.

Annual ryegrass (Lolium rigidum Gaudin) is a problematic weed in most of the cropping regions in Australia. Evolved herbicide resistance to multiple modes of action groups, the lack of new herbicidal molecules, and the emergence of late cohorts in winter season crops necessitate the use of existing herbicides in an innovative way to control L. rigidum. Pendimethalin as pre-emergence (PRE) is used for L. rigidum control; however, it is unable to control late cohorts of L. rigidum in winter season crops. Pot studies were conducted from 2019 to 2021 to evaluate the effect of a post-emergence (POST) application of pendimethalin with six doses (0, 220, 440, 880, 1,760, and 3,520 g a.i. ha-1) on L. rigidum control in addition to crop safety in wheat, barley, and sorghum. POST application of pendimethalin was found to be safe for wheat and barley, and the biomass reduction in both crops at varied rates (220 to 3,520 g ha-1) of pendimethalin ranged from 10% to 20%. A dose-response study revealed that the pendimethalin dose required for 50% mortality and growth reduction of L. rigidum was 179 and 144 g ai ha-1, respectively. Pendimethalin at 880 g ai ha-1 completely killed late cohorts of L. rigidum. These results suggest that the POST application of pendimethalin can be used for the effective control of late cohorts of L. rigidum in wheat and barley crops. POST application of pendimethalin suppressed the biomass of sorghum plants compared with the non-treated control treatment, and biomass reduction in sorghum at different pendimethalin doses ranged from 32% to 40%. Results implied that POST applications of pendimethalin in wheat and barley can diversify herbicide programs in managing L. rigidum and may help in delaying the evolution of resistance in this weed. The use of disinfectants is a crucial aspect of preventive and health improvement measures for infectious diseases in farm and domestic animals. Regulatory documents require the determination of toxicity to macroorganisms, including the establishment of lethal doses and toxicity groups, during the development and registration of antimicrobial agents. This study aimed to investigate the histological changes in the internal organs of laboratory animals when determining lethal doses of innovative disinfectants. The experiments used domestic disinfectants containing glutaraldehyde as the active ingredient. Histological studies were conducted on the internal organs (kidney, liver, stomach, intestines, and spleen) of 75 laboratory animals using an Axioskop 40/40FL microscope (Carl Zeiss, Germany) with video microscopic photography. The methods used were under current standards. Hemodynamic disorders were observed in the renal tissue under the influence of lethal doses of aldehyde disinfectants. These disorders were characterized by capillary dilation and blood filling. Glomerular capillary dilation and overflow with blood cells were also detected. Additionally, stasis was observed in the lumen of the microcirculatory vessels throughout the entire length. The examination of histological sections from animal liver samples revealed a significant expansion of Dissé spaces, variations in the size of hepatocyte nuclei, beam decomposition and fragmentation, small acute perivascular hemorrhages, leukostasis in sinusoids, and hemodynamic disorders. The structure of the organ's beams was also disturbed, and a significant number of venous vessels were dilated and excessively filled with blood cells. Minor changes were detected in the stomach, including desquamation of the epithelial cells of the glands and their exfoliation into the gastric lumen, as well as circulatory disorders. Epithelial desquamation, blood vessel dilation, and signs of connective tissue edema were observed in the intestine. The kidneys exhibited signs of acute venous hemorrhage and stasis in vessels of various calibers, with the development of small acute parenchymal hemorrhages and localized lymphoid cell death in the white pulp. The prospect of further research is to investigate the histomorphological changes in the internal organs of laboratory animals when exposed to modern complex disinfectants with different active ingredients. 2,2-Dibromo-3-nitrilopropionamide (DBNPA) is a widely used biocide with potential environmental implications due to its toxicity. This study aimed to investigate the in vivo toxicity of DBNPA in zebrafish (Danio rerio), a model organism in environmental toxicology. Both adult and larval zebrafish were exposed to varying concentrations of DBNPA, and significant morphological changes and mortality rates were observed. The study found that even relatively low concentrations of DBNPA can have detrimental effects on zebrafish embryonic development, and high concentrations resulted in rapid mortality in adult zebrafish and larvae. The LC50 values calculated from this study were 9.3 ppm for adults and 9.1 ppm for larvae, indicating the high toxicity of DBNPA to these organisms. Background: AB-CHMINACA is a cannabimimetic indazole derivative. In 2013, it was reported in different countries as a substance of abuse. Purpose: This study evaluated the subacute toxic effects of AB-CHMINACA on the liver and kidneys and measured its blood level in adult male mice. Methods: The histological and biochemical subacute toxic effects on the liver and kidneys were assessed after four weeks of daily intraperitoneal injections of one of the following doses: 0.3 mg/kg, 3 mg/kg, or 10 mg/kg as the highest dose in adult male albino mice. In addition, the blood concentration level of AB-CHMINACA was determined by GC–MS-MS. Results: The histological effects showed congestion, hemorrhage, degeneration, and cellular infiltration of the liver and kidney tissues. Considering the control groups as a reference, biochemical results indicated a significant increase in the serum AST only in the highest dose group, while the ALT and creatinine levels did not significantly change. The mean values of AB-CHMINACA blood levels were 3.05 ± 1.16, 15.08 ± 4.30, and 54.43 ± 8.70 ng/mL for the three treated groups, respectively, one hour after the last dose of intraperitoneal injection. The calibration curves were linear in the 2.5–500 ng/mL concentration range. The intra-assay precision and accuracy of the method were less than 7.0% (RSD) and ± 9.2% (Bias). Conclusion: This research supports the available case reports on AB-CHMINACA toxicity that it has low lethality; still, the chronic administration causes evident liver and kidney histotoxic effects even at low doses with unnoticeable clinical effects in mice. Background: Our study evaluated the effect of Chrysanthemum coronarium L. (CC) leaves on ethanol-induced acute gastric ulcers in Wistar rats. Materials and Methods: The organic extract of CC was obtained by Soxhlet extraction with methanol and then divided into two dose groups: 250 mg/kg and 500 mg/kg. Omeprazole was used as a positive control at 20 mg/kg. Our extract was subjected to the separation of bioactive compounds by High-Performance Liquid Chromatography (HPLC). Lethality tests (LD50) were carried out using standard procedures. Gastric protection was assessed by measuring gastric juice volume, total acidity, and free acidity. Gastric mucosal damage was assessed by histopathological examination. Results: Chromatographic analysis of the Methanolic Extract of Chrysanthemum coronarium L. (MECC) identified the presence of 19 phenolic compounds, representing 46.47% of the total sample. The dominant components were o-coumaric acid (9.55%), chlorogenic acid (6%), myricetin (4.19%), and benzoic acid (2.87%). Oral LD50 value was more than 5000 mg/kg in rat. In the present study, the methanolic extract of CC decreased total and free gastric acidity (53.80±7.038 and 17.8±2.375 respectively) for the 500 mg/kg dose and (60.40±4.490 and 24.8±1.855 respectively) for the 250 mg/kg dose. Omeprazole also decreased free and total gastric acidity (54.40±3.092 and 20±2.449 respectively), compared with the ethanol groups (19.40±2.909 and 4.4±0.678 respectively). In the histological study, we found that the gastric mucosal barrier could be significantly strengthened when the rats were pretreated with 500 mg/kg of the MECC and showed almost normal histology compared with the ethanol-ulcerated groups. Conclusion: Based on the present results, we can conclude that CC leaves could be a promising food for the protection of the gastric mucosa against ethanol-induced lesions. [ABSTRACT FROM AUTHOR]

Being a pulse crop, green gram has huge scope in crop improvement in terms of productivity and other yield related parameters. Genetic variability is a preferable option for breeders in breeding programme for varietal development programme and others crop improvement aspect. The present investigation was framed to create the variability in mung bean var. GAM 8 during the seedling period through gamma (γ) irradiation. The significant variation was observed in germination percentage and it was found that dose dependent relationship between the germination rate and dose. Significantly minimum germination percentage (22.38 %) was observed in 700 Gy but optimal lethal dose (LD50) calculated through Probit analysis based on germination percentage was revealed at 540.26 Gy. Data revealed that γ irradiation had significantly reduced the seedling growth parameters such as shoot length (10.05 cm), shoot dry weight (19.68 mg), seedling length (13.90 cm), vigour index I (311.01) and II (509.01) was registered in 700 Gy while minimum root length (3.83 cm) was occurred in 600 Gy γ irradiation. Growth reduction (GR) 50 and 30 with respect to the seedling length was occurred respectively at 1093.79 and 1469.74 Gy. Therefore, this finding as the source of genetic variability would be used in future breeding and crop improvement programme like enhancement of yield potentiality and stress management in mung bean var. Currently, only Apis mellifera is used in environmental regulation to evaluate the hazard of pesticides to pollinators. The low representativeness of pollinators and bee diversity in this approach may result in insufficient protection for the wild species. This scenario is intensified in tropical environments, where little is known about the effects of pesticides on solitary bees. We aimed to calculate the medium lethal dose (LD50) and medium lethal concentration (LC50) of the insecticide dimethoate in the Neotropical solitary bee Centris analis, a cavity‐nesting, oil‐collecting bee distributed from Brazil to Mexico. Males and females of C. analis were exposed orally to dimethoate for 48 h under laboratory conditions. Lethality was assessed every 24 h until 144 h after the beginning of the test. After the LD50 calculation, we compared the value with available LD50 values in the literature of other bee species using the species sensitivity distribution curve. In 48 h of exposure, males showed an LD50 value 1.33 times lower than females (32.78 and 43.84 ng active ingredient/bee, respectively). Centris analis was more sensitive to dimethoate than the model species A. mellifera and the solitary bee from temperate zones, Osmia lignaria. However, on a body weight basis, C. analis and A. mellifera had similar LD50 values. Ours is the first study that calculated an LD50 for a Neotropical solitary bee. Besides, the results are of crucial importance for a better understanding of the effects of pesticides on the tropical bee fauna and will help to improve the risk assessment of pesticides to bees under tropical conditions, giving attention to wild species, which are commonly neglected. Callinina georgiana (banded mystery snail) and Cipangopaludina japonica (Japanese mystery snail) are two exotic viviparid snail species that have been introduced to North America. These snail species harbor parasites, predate on the eggs of native fish fauna and are prolific in their ability to spread across various freshwater environments. While several recent studies have explored the population structure and range expansion of the species in the United States, none have formally addressed control or eradication strategies. In this study, we attempted to do this by carrying out a series of assays to determine the efficacy of the popular molluscicide, niclosamide monohydrate against both species under both acute and chronic exposure. Our results show that the lethal dose required to achieve 50 % mortality (LD50) for C. georgiana and C. japonica was 1.509 mg/L and 1.296 mg/L respectively. In acute exposure experiments we observed that at higher concentrations, all C. georgiana specimens in experimental treatment tanks were dead but C. japonica never reached 100% mortality even at the highest concentration. In chronic exposure experiments, C. georgiana did not survive more than a week while C. japonica did not survive more than 11 days in the experimental tanks. We conclude that niclosamide is an effective chemical control agent for eradication of invasive mystery snails in North America. However, further studies exploring the effect of the molluscide on non-target species, especially local native fauna, should be investigated prior to any scaled up field applications.
