Based on a review of literature and a few case studies, this paper summarizes the state of the art on the dating of folds and thrusts, then presents and discusses how recent advances in K–Ar illite and U–Pb calcite geochronology applied to brittle structures in fold-and-thrust belts have helped better constrain the timing, sequence, duration and rates of deformation.

By demonstrating that extensional inheritance plays a decisive role in the formation of orogens, recent studies have questioned the ability of a unique, complete Wilson cycle model to explain the diversity of collisional orogens. For 5 years, the OROGEN Research Project had therefore the ambition to challenge this classical Wilson cycle model. By focusing on the diffuse Africa-Europe plate boundary in the Biscay-Pyrenean-Western Mediterranean system, the project questioned the preconceived “Orogen singularity” assumption and investigated the role of divergent and convergent maturities in orogenic and post-orogenic processes. This work led us to rethink the development of collisional orogens in a genetic (or process-driven) way and to propose an updated version of the ” classical Wilson cycle”, the Wilson Cycle 2.0, and the ORO-Genic ID concept presented in this paper. The particularity of the Wilson Cycle 2.0 is to take into account the divergence and convergence maturity reached during extensional and orogenic processes in proposing different tectonic tracks associated with different ORO-Genic ID numbers. The ORO-Genic ID is composed of a letter (or track), corresponding to the maturity of divergence reached and a number corresponding to the maturity of convergence reached during the formation of the orogen. This new concept relies on the observed pre- and syn- convergent tectono- stratigraphic and magmatic record and deformation history and can be identified in using diagnostic criteria presented in this paper. It represents therefore a powerful tool that can be used to characterize the evolution and the architectural type of an orogenic system. Moreover, as a mappable concept, it can be easily used worldwide and can help us to explain differences in the style of deformation at crustal scale between orogens.

Immergés en hiver, les marais saisonniers (merjas) de la plaine du Gharb au Maroc étaient traditionnellement exploités pour le pâturage en été, à côté d’autres usages productifs. Pourtant, ils étaient considérés par l’administration coloniale comme des milieux insalubres et peu valorisés, malgré leur abondance en ressources naturelles. Au cours du XXe siècle, la plaine a fait l’objet d’un aménagement hydro-agricole, incorporant les merjas, pour contribuer aux objectifs nationaux de sécurité alimentaire et d’exportation de produits agricoles. Cet article interroge le regard porté par les acteurs locaux et institutionnels sur les merjas et les bénéfices qu’elles procurent, à travers une analyse des services écosystémiques. Si les institutions les considèrent comme des terres vierges à aménager et des zones tampon de régulation des crues pour protéger les secteurs aménagés et les villes, les collectivités riveraines les considèrent comme des espaces productifs, défavorisés par les crues. L’étude montre des visions contrastées des services écosystémiques, où chaque acteur a une vision utilitariste des merjas. Malgré le débat international sur la biodiversité et le stockage de carbone dans les zones humides, les dimensions écologiques sont peu citées. L’approche par les services écosystémiques dévoile les antagonismes entre acteurs, mais peut être un moyen de négociation sur le devenir des merjas dans une vision territoriale.

Les agriculteurs utilisant les eaux des khettaras, galeries souterraines drainant la nappe phréatique et donnant un accès collectif à l’eau souterraine dans les oasis, font aujourd’hui face à un dilemme. Les khettaras se tarissent sous l’effet conjugué du pompage pour l’eau potable et de l’irrigation des exploitations agricoles des nouvelles extensions, et d’une recharge décroissante de la nappe. Leurs khettaras menacées de disparition, certains collectifs ont choisi d’installer des puits ou des forages alimentés par l’énergie solaire. Cela permet de renforcer le débit des khettaras et ainsi de maintenir l’accès collectif à l’eau souterraine, mais ces installations contribuent aussi à sa surexploitation. Dans cet article, nous mettons en discussion ce choix cornélien des communautés oasiennes dans le sud du Maroc. Des observations de terrain, l’analyse des images satellites, et des enquêtes avec les agriculteurs ont permis de comprendre le contexte d’émergence d’un dispositif associant la khettara au pompage par énergie solaire, d’analyser sa conception technique et de mettre en évidence la capacité des oasiens à intervenir sur les règles de gestion pour superposer ce dispositif technique et institutionnel nouveau au système traditionnel des khettaras. Cet article contribue à une réflexion sur la durabilité de l’accès à l’eau souterraine dans ce contexte présaharien.

Worldwide there is a tendency towards deregulation in many policy sectors - this, for example, includes liberalization and privatization of drinking-water management. However, concerns about the negative impacts this might have on human health call for prospective health impact assessment (HIA) on the management of drinking-water. On the basis of an established generic 10-step HIA procedure and on risk assessment methodology, this paper aims to produce quantitative estimates concerning health effects from increased exposure to carcinogens in drinking-water. Using data from North Rhine-Westphalia in Germany, probabilistic estimates of excess lifetime cancer risk, as well as estimates of additional cases of cancer from increased carcinogen exposure levels are presented. The results show how exposure to contaminants that are strictly within current limits could increase cancer risks and case-loads substantially. On the basis of the current analysis, we suggest that with uniform increases in pollutant levels, a single chemical (arsenic) is responsible for a large fraction of expected additional risk. The study also illustrates the uncertainty involved in predicting the health impacts of changes in water quality. Future analysis should include additional carcinogens, non-cancer risks including those due to microbial contamination, and the impacts of system failures and of illegal action, which may be increasingly likely to occur under changed management arrangements. If, in spite of concerns, water is privatized, it is particularly important to provide adequate surveillance of water quality.

Abstract Background Pontocerebellar hypoplasia (PCH) is a heterogeneous group of diseases characterized by lack of development and/or early neurodegeneration of cerebellum and brainstem. According to clinical features, seven subtypes of PCH have been described, PCH type 2 related to TSEN54 mutations being the most frequent. PCH is most often autosomal recessive though de novo anomalies in the X-linked gene CASK have recently been identified in patients, mostly females, presenting with intellectual disability, microcephaly and PCH (MICPCH). Methods Fourteen patients (12 females and two males; aged 16 months-14 years) presenting with PCH at neuroimaging and with clinical characteristics unsuggestive of PCH1 or PCH2 were included. The CASK gene screening was performed using Array-CGH and sequencing. Clinical and neuroradiological features were collected. Results We observed a high frequency of patients with a CASK mutation (13/14). Ten patients (8 girls and 2 boys) had intragenic mutations and three female patients had a Xp11.4 submicroscopic deletion including the CASK gene. All were de novo mutations. Phenotype was variable in severity but highly similar among the 11 girls and was characterized by psychomotor retardation, severe intellectual disability, progressive microcephaly, dystonia, mild dysmorphism, and scoliosis. Other signs were frequently associated, such as growth retardation, ophthalmologic anomalies (glaucoma, megalocornea and optic atrophy), deafness and epilepsy. As expected in an X-linked disease manifesting mainly in females, the boy hemizygous for a splice mutation had a very severe phenotype with nearly no development and refractory epilepsy. We described a mild phenotype in a boy with a mosaic truncating mutation. We found some degree of correlation between severity of the vermis hypoplasia and clinical phenotype. Conclusion This study describes a new series of PCH female patients with CASK inactivating mutations and confirms that these patients have a recognizable although variable phenotype consisting of a specific form of pontocerebellar hypoplasia. In addition, we report the second male patient to present with a severe MICPCH phenotype and a de novo CASK mutation and describe for the first time a mildly affected male patient harboring a mosaic mutation. In our reference centre, CASK related PCH is the second most frequent cause of PCH. The identification of a de novo mutation in these patients enables accurate and reassuring genetic counselling.

Abstract Background Individuals infected with human immunodeficiency virus (HIV) have an increased risk of progression to active tuberculosis following Mycobacterium tuberculosis infection. The objective of the study was to determine IFN-γ responses for the detection of latent tuberculosis infection (LTBI) with QuantiFERON-TB GOLD In Tube (QFT-G-IT) and T-SPOT.TB in HIV patients, and evaluate the influence of CD4 cell count on tests performance. Methods We studied 75 HIV patients enrolled for ongoing studies of LTBI with T-SPOT.TB, QFN-G-IT and TST. Mean CD4 cell counts ± standard deviation was 461.29 ± 307.49 cells/μl. Eight patients had a BCG scar. Results T-SPOT.TB, QFN-G-IT and TST were positive in 7 (9.3%), 5 (6.7%) and 9 (12%) cases, respectively. Global agreement between QFN-G-IT and T-SPOT.TB was 89% (κ = 0.275). The overall agreement of T-SPOT.TB and QFN-G-IT with TST was 80.8% (κ = 0.019) and 89% (κ = 0.373), respectively. We have found negative IFN-γ assays results among 2 BCG-vaccinated HIV-infected individuals with a positive TST. In non BCG-vaccinated patients, QFN-G-IT and TST were positive in 5 cases (7.5%) and T-SPOT.TB in 7 (10.4%). In contrast, in BCG-vaccinated patients, only TST was positive in 4/8 (50%) of the cases. The differences obtained in the number of positive results between TST and both IFN-γ assays in BCG vaccinated patients were significant (95% CI 3-97%, p = 0.046), however, the confidence interval is very wide given the small number of patients. In patients with CD4< 200, we obtained only one (5%) positive result with T-SPOT.TB; however, QFN-G-IT and TST were negative in all cases. On the contrary, percentages of positive results in patients with CD4> 200 were 10.9% (6/55), 9.1% (5/55) and 16.4% (9/55) with T-SPOT.TB, QFN-G-IT and TST, respectively. Conclusions IFN-γ tests have the benefit over TST that are less influenced by BCG vaccination, consequently they are more specific than TST. Although our number of patients with advance immunosuppression is limited, our study suggests that IFN-γ assays are influenced with level of immunosuppression. The use of IFN-γ assays could be a helpful method for diagnosing LTBI in HIV population.

Abstract Background The plant miRNAs represent an important class of endogenous small RNAs that guide cleavage of an mRNA target or repress its translation to control development and adaptation to stresses. MiRNAs are nuclear-encoded genes transcribed by RNA polymerase II, producing a primary precursor that is subsequently processed by DCL1 an RNase III Dicer-like protein. In rice hundreds of miRNAs have been described or predicted, but little is known on their genes and precursors which are important criteria to distinguish them from siRNAs. Here we develop a combination of experimental approaches to detect novel miRNAs in rice, identify their precursor transcripts and genes and predict or validate their mRNA targets. Results We produced four cDNA libraries from small RNA fractions extracted from distinct rice tissues. By in silico analysis we selected 6 potential novel miRNAs, and confirmed that their expression requires OsDCL1. We predicted their targets and used 5'RACE to validate cleavage for three of them, targeting a PPR, an SPX domain protein and a GT-like transcription factor respectively. In addition, we identified precursor transcripts for the 6 miRNAs expressed in rice, showing that these precursors can be efficiently processed using a transient expression assay in transfected Nicotiana benthamiana leaves. Most interestingly, we describe two precursors producing tandem miRNAs, but in distinct arrays. We focus on one of them encoding osa-miR159a.2, a novel miRNA produced from the same stem-loop structure encoding the conserved osa-miR159a.1. We show that this dual osa-miR159a.2-osa-miR159a.1 structure is conserved in distant rice species and maize. Finally we show that the predicted mRNA target of osa-miR159a.2 encoding a GT-like transcription factor is cleaved in vivo at the expected site. Conclusion The combination of approaches developed here identified six novel miRNAs expressed in rice which can be clearly distinguished from siRNAs. Importantly, we show that two miRNAs can be produced from a single precursor, either from tandem stem-loops or tandemly arrayed in a single stem-loop. This suggests that processing of these precursors could be an important regulatory step to produce one or more functional miRNAs in plants and perhaps coordinate cleavage of distinct targets in the same plant tissue.
