Manure nitrogen (N) from cattle contributes to nitrous oxide and ammonia emissions and nitrate leaching. Measurement of manure N outputs on dairy farms is laborious, expensive, and impractical at large scales; therefore, models are needed to predict N excreted in urine and feces. Building robust prediction models requires extensive data from animals under different management systems worldwide. Thus, the study objectives were (1) to collate an international database of N excretion in feces and urine based on individual lactating dairy cow data from different continents; (2) to determine the suitability of key variables for predicting fecal, urinary, and total manure N excretion; and (3) to develop robust and reliable N excretion prediction models based on individual data from lactating dairy cows consuming various diets. A raw data set was created based on 5,483 individual cow observations, with 5,420 fecal N excretion and 3,621 urine N excretion measurements collected from 162 in vivo experiments conducted by 22 research institutes mostly located in Europe (n = 14) and North America (n = 5). A sequential approach was taken in developing models with increasing complexity by incrementally adding variables that had a significant individual effect on fecal, urinary, or total manure N excretion. Nitrogen excretion was predicted by fitting linear mixed models including experiment as a random effect. Simple models requiring dry matter intake (DMI) or N intake performed better for predicting fecal N excretion than simple models using diet nutrient composition or milk performance parameters. Simple models based on N intake performed better for urinary and total manure N excretion than those based on DMI, but simple models using milk urea N (MUN) and N intake performed even better for urinary N excretion. The full model predicting fecal N excretion had similar performance to simple models based on DMI but included several independent variables (DMI, diet crude protein content, diet neutral detergent fiber content, milk protein), depending on the location, and had root mean square prediction errors as a fraction of the observed mean values of 19.1% for intercontinental, 19.8% for European, and 17.7% for North American data sets. Complex total manure N excretion models based on N intake and MUN led to prediction errors of about 13.0% to 14.0%, which were comparable to models based on N intake alone. Intercepts and slopes of variables in optimal prediction equations developed on intercontinental, European, and North American bases differed from each other, and therefore region-specific models are preferred to predict N excretion. In conclusion, region-specific models that include information on DMI or N intake and MUN are required for good prediction of fecal, urinary, and total manure N excretion. The objective of this work was to construct a dynamic model of hepatic amino acid metabolism in the lactating dairy cow that could be parameterized using net flow data from in vivo experiments. The model considers 22 amino acids, ammonia, urea, and 13 energetic metabolites, and was parameterized using a steady-state balance model and two in vivo, net flow experiments conducted with mid-lactation dairy cows. Extracellular flows were derived directly from the observed data. An optimization routine was used to derive nine intracellular flows. The resulting dynamic model was found to be stable across a range of inputs suggesting that it can be perturbed and applied to other physiological states. Although nitrogen was generally in balance, leucine was in slight deficit compared to predicted needs for export protein synthesis, suggesting that an alternative source of leucine (e.g. peptides) was utilized. Simulations of varying glucagon concentrations indicated that an additional 5 mol/d of glucose could be synthesized at the reference substrate concentrations and blood flows. The increased glucose production was supported by increased removal from blood of lactate, glutamate, aspartate, alanine, asparagine, and glutamine. As glucose output increased, ketone body and acetate release increased while CO(2) release declined. The pattern of amino acids appearing in hepatic vein blood was affected by changes in amino acid concentration in portal vein blood, portal blood flow rate and glucagon concentration, with methionine and phenylalanine being the most affected of essential amino acids. Experimental evidence is insufficient to determine whether essential amino acids are affected by varying gluconeogenic demands.

In the past decade, a number of mechanistic, dynamic simulation models of several components of the dairy production system have become available. However their use has been limited due to the detailed technical knowledge and special software required to run them, and the lack of compatibility between models in predicting various metabolic processes in the animal. The first objective of the current study was to integrate the dynamic models of [Brit. J. Nutr. 72 (1994) 679] on rumen function, [J. Anim. Sci. 79 (2001) 1584] on methane production, [J. Anim. Sci. 80 (2002) 248] on N partition, and a new model of P partition. The second objective was to construct a decision support system to analyse nutrient partition between animal and environment. The integrated model combines key environmental pollutants such as N, P and methane within a nutrient-based feed evaluation system. The model was run under different scenarios and the sensitivity of various parameters analysed. A comparison of predictions from the integrated model with the original simulation models showed an improvement in N excretion since the integrated model uses the dynamic model of [Brit. J. Nutr. 72 (1994) 679] to predict microbial N, which was not represented in detail in the original model. The integrated model can be used to investigate the degree to which production and environmental objectives are antagonistic, and it may help to explain and understand the complex mechanisms involved at the ruminal and metabolic levels. A part of the integrated model outputs were the forms of N and P in excreta and methane, which can be used as indices of environmental pollution.

A model of mammary metabolism has been constructed and parameterized, with milk protein synthesis represented as a function of five essential amino acids (EAA) (Hanigan et al., 2001). Herein the model is evaluated using both the data used to construct the model (reference data) and an independent data set (literature data), and sensitivity to inputs and parameter estimates is assessed. The model predicted metabolite removal well for the reference data with exceptions for glutamate, glucose, and acetate. However, predictions of milk protein synthesis exhibited significant mean positive bias, which apparently was associated with the representation of milk protein synthesis. Adjustment of model parameters removed the mean bias, however, prediction accuracy was still inadequate. Simulation of the single reference experiment containing all critical inputs resulted in predictions of milk protein output that explained 53% of the observed variation, suggesting that the limited accuracy of the model when applied to the entire reference data set was due to assumptions regarding missing inputs. Mammary removal of glutamate, isoleucine, lysine, phenylalanine, tyrosine, valine, glycerol, β -hydroxybutyrate (BHBA), and acetate were predicted less accurately when simulations of the independent data set were conducted. Twenty-five percent of the observed variation in milk protein yields for the independent data set was explained by the model. Refitting parameters for removal of isoleucine, lysine, phenylalanine, tyrosine, valine, glycerol, BHBA, and acetate raised the variation explained to 43%. Sensitivity analysis indicated that milk protein synthesis was responsive to only the five EAA used in its determination, with sensitivity to any single EAA falling to zero as supply of the EAA exceeded protein synthetic needs. Similarly, milk protein synthesis was readily affected by parameters associated with removal and metabolism of the five EAA. Milk lactose was found to be sensitive to glucose input as well as to similar parameters and inputs as milk protein. It is concluded that representation of the milk protein synthesis process as a function of a single limiting EAA may not be adequate and might be better represented by simultaneous consideration of multiple EAA. Additional work on the description of energy metabolism is also suggested.

The effect of a mixed beta 1/beta 2-adrenergic agonist, isoproterenol, on hindlimb metabolism was studied in growing wether lambs using arteriovenous difference and blood flow rate techniques. Isoproterenol (48 micrograms kg-1 d-1), or saline, was infused into a jugular vein of five wether lambs (30 to 35 kg) for five days and samples taken on the fifth day of treatment. Infusion of isoproterenol significantly increased blood flow, oxygen uptake and tyrosine uptake across the hindlimb. Hindlimb non-esterified fatty acid uptake was increased but not significantly (P = 0.11) and arterial growth hormone concentration was not altered by isoproterenol infusion. Results suggest that beta-adrenergic agonists promote lean tissue deposition by increased muscle blood flow rate and amino acid uptake.

17 páginas, 3 figuras, 3 tablas., Grass-based diets are of increasing social-economic importance in dairy cattle farming, but their low supply of glucogenic nutrients may limit the production of milk. Current evaluation systems that assess the energy supply and requirements are based on metabolisable energy (ME) or net energy (NE). These systems do not consider the characteristics of the energy delivering nutrients. In contrast, mechanistic models take into account the site of digestion, the type of nutrient absorbed and the type of nutrient required for production of milk constituents, and may therefore give a better prediction of supply and requirement of nutrients. The objective of the present study is to compare the ability of three energy evaluation systems, viz. the Dutch NE system, the agricultural and food research council (AFRC) ME system, and the feed into milk (FIM) ME system, and of a mechanistic model based on Dijkstra et al. [Simulation of digestion in cattle fed sugar cane: prediction of nutrient supply for milk production with locally available supplements. J. Agric. Sci., Cambridge 127, 247-60] and Mills et al. [A mechanistic model of whole-tract digestion and methanogenesis in the lactating dairy cow: model development, evaluation and application. J. Anim. Sci. 79, 1584-97] to predict the feed value of grass-based diets for milk production. The dataset for evaluation consists of 41 treatments of grass-based diets (at least 0.75 g ryegrass/g diet on DM basis). For each model, the predicted energy or nutrient supply, based on observed intake, was compared with predicted requirement based on observed performance. Assessment of the error of energy or nutrient supply relative to requirement is made by calculation of mean square prediction error (MSPE) and by concordance correlation coefficient (CCC). All energy evaluation systems predicted energy requirement to be lower (6-11%) than energy supply. The root MSPE (expressed as a proportion of the supply) was lowest for the mechanistic model (0.061), followed by the Dutch NE system (0.082), FIM ME system (0.097) and AFRCME system(0.118). For the energy evaluation systems, the error due to overall bias of prediction dominated the MSPE, whereas for the mechanistic model, proportionally 0.76 of MSPE was due to random variation. CCC analysis confirmed the higher accuracy and precision of the mechanistic model compared with energy evaluation systems. The error of prediction was positively related to grass protein content for the Dutch NE system, and was also positively related to grass DMI level for all models. In conclusion, current energy evaluation systems overestimate energy supply relative to energy requirement on grass-based diets for dairy cattle. The mechanistic model predicted glucogenic nutrients to limit performance of dairy cattle on grass-based diets, and proved to be more accurate and precise than the energy systems. A comparison of the models of Vitti et al. (2000, J. Anim. Sci. 78, 2706-2712) and Fernandez (1995c, Livest. Prod. Sci. 41, 255-261) was carried out using two data sets on growing pigs as input. The two models compared were based on similar basic principles, although their aims and calculations differed. The Vitti model employs the rate:state formalism and describes phosphorus (P) flow between four pools representing P content in gut, blood, bone and soft tissue in growing goats. The Fernandez model describes flow and fractional recirculation between P pools in gut, blood and bone in growing pigs. The results from both models showed similar trends for P absorption from gut to blood and net retention in bone with increasing P intake, with the exception of the 65 kg results from Date Set 2 calculated using the Fernandez model. Endogenous loss from blood back to gut increased faster with increasing P intake in the Fernandez than in the Vitti model for Data Set 1. However, for Data Set 2, endogenous loss increased with increasing P intake using the Vitti model, but decreased when calculated using the Fernandez model. Incorporation of P into bone was not influenced by intake in the Fernandez model, while in the Vitti model there was an increasing trend. The Fernandez model produced a pattern of decreasing resorption in bone with increasing P intake, with one of the data sets, which was not observed when using the Vitti model. The pigs maintained their P homeostasis in blood by regulation of P excretion in urine.

Previous efforts to simulate mammary metabolism have focused on energy, mostly considering amino acids (AA) in aggregate. The main objective of this work was to build a model of mammary metabolism, based on data from arterio-venous difference studies, which considered AA in sufficient detail to predict yields of milk solids. The model contains 19 state variables and considers the removal of 37 metabolites from blood, including 22 AA. It is driven by blood flow and arterial concentrations, and outputs include milk protein, milk lactose, and three classes of milk fat (by chain length). The model was parameterized using a balance version of it and the mean observations from four arterio-venous difference experiments, with a limited number of assumptions, and evaluated against these experiments. In assembling the balance model, milk protein output was not predicted satisfactorily, as some essential AA were not present in quantities great enough to support the rates of milk protein synthesis observed experimentally. Tryptophan showed the greatest deficit, followed by tyrosine plus phenylalanine, methionine, and histidine. In addition, significant quantities of pyruvate were needed to synthesize serine, glycine, and alanine. The supply of alpha-ketoglutarate plus glutamate to synthesize proline and glutamine was provided in part by catabolism of arginine; the remainder was derived from catabolism of other AA and energetic substrates.

The contribution to casein biosynthesis of peptides derived from blood was examined in late lactation goats (254 to 295 d in milk). Ratios of mammary uptake of free amino acids (AA) in blood to output of AA in milk protein and ratios of the enrichments of Phe, Tyr, Met, and Lys at isotopic plateau in secreted milk casein to the free AA in arterial and mammary vein blood were monitored during the last 5h of a 30-h continuous i.v. infusion of [1- 13 C]Phe, [ 2 H 4 ]Tyr, [5- 13 CH, 3 ]Met, and [2- 15 N]Lys on two occasions: before (control) and on d 6 of an i.v. infusion of Phe (6 g/d). During the control, uptakes of free Phe and Met were less than their output in milk. This result was comparable with the labeling kinetic results, suggesting that vascular peptides contributed 5 to 11% of Phe and 8 to 18% of Met. Free Tyr and Lys uptakes during the control were sufficient for milk output; however, the labeling kinetics indicated that 13 to 25% of the Tyr and 4 to 13% of the Lys were derived from peptides. Infusion of Phe increased the uptake of free AA but reduced the contribution of peptides toward Phe (0 to 3%) and Tyr (8 to 14%) supply for casein synthesis. Whole body hydroxylation of Phe to Tyr increased from 10 to 18% with the infusion of Phe; within the mammary gland, this conversion was lower (3 to 5%). Results suggest that the mammary utilization of peptides containing Phe and Tyr appears to depend on the supply of free AA in blood.

Research in Brazil has been carried out to study mineral metabolism in sheep and cattle, especially phosphorus, by using the isotope dilution technique (Vitti et al., 2000). However there is very little information on calcium metabolism in goats. The proposal of the present study is to measure the intensity of the different processes that occur in goats fed different amounts of calcium, by using radioactive calcium, and to propose a model of Ca metabolism.

Agriculture is one of the major sources of nitrogen (N) pollution. Dairy production causes losses of N in faeces and urine that contribute to environmental pollution with an estimated annual output of 320 kt N and 80 kt ammonia in the U.K. alone. Therefore, improving N utilization in dairy cows and especially reducing N output in excreta is desirable to reduce environmental N pollution, particularly as ammonia. Mathematical models have been used to predict potential N excretion from dairy cows. However, these models generally are empirical in nature, hence not process based and therefore there is a need to develop a model that can describe biological processes in the animal. The objective of the present study was to develop a dynamic N model to predict the amount and form of N excreted by dairy herds and seek to make appropriate recommendations that will reduce N excretion from dairy cows.

Dietary intervention to reduce methane emissions from lactating dairy cattle is both environmentally and nutritionally desirable due to the importance of methane as a causative agent in global warming and as a significant loss of feed energy. This investigation involved the development of a dynamic mechanistic model of whole rumen function (Dijkstra et al. 1992), with the objective to simulate whole-animal methane emissions for a range of dietary inputs.

A non-invasive method is proposed for determining the extent of degradation in the rumen, based on the gas production technique and mathematical modelling. The exercise involves developing both a statistical model and a kinetic model (France et al., 2000). The statistical model shifts (or maps) the gas accumulation profile obtained using a faecal inoculum to a rumen gas profile, thus obviating the need for rumen sampling. The kinetic model determines the extent of degradation in the rumen from the shifted profile. It is presented as a generalised mathematical function, allowing any one of a number of alternative equation forms to be selected.

Phosphorus (P) is a key mineral in energy metabolism and is essential in nearly every biochemical aspect of dairy cow metabolism. Therefore, P needs to be supplied in sufficient quantity to optimize animal performance. However, dairy cows only use 30 – 45% of their dietary P intake and the rest is excreted mainly in faeces. Excess faecal excretion can lead to P accumulation and saturation in the soil and filter into groundwater or remain in surface water (Tamminga, 1996), which is known to cause eutrophication. It is therefore desirable to formulate P rations according to the requirement of the animals and thereby reduce P pollution. The objective of the present study was to develop a dynamic model of P metabolism in dairy cows and use that to identify and quantify trends of P excretion as a function of P intake and investigate effects of energy supplementation on P utilisation.

The mammary gland of lactating ruminants (Guinard & Rulquin 1994) does not appear to extract sufficient quantities of free amino acids (AA) to account for their output as milk protein. Based upon application of a precursor (blood or plasma free AA):product (casein) labelling technique (Backwell et al. 1996) in goats, results suggest that blood peptides or proteins taken up by the gland probably account for this deficit. However, the deficiency appears to be alleviated when supplemental protein or AA are infused (Guinard & Rulquin 1994), suggesting that uptake of peptide bound AA is reduced while that of the free AA is increased. The objective of the current study was to corroborate these findings, thus the precursonproduct labelling technique was applied in lactating goats to determine whether arterial free phenylalanine supply affects the contribution of peptide bound phenylalanine and tyrosine to casein synthesis and compare the results to the net uptake (NU) method.

Publisher Summary This chapter describes the effects of close-arterial infusion of cimaterol into the hind-limb of growing lambs. The β-agonist cimaterol has been shown to increase the amount of lean tissue in the carcass of farm animals. The direct effects of cimaterol on hind-limb muscle metabolism were studied in growing wether lambs by close-arterially infusing either saline; a low dose of cimaterol or a high dose of cimaterol into one femoral artery for 5 days. The other leg was close-arterially infused with saline for 5 days as a control. Hind-limb muscle metabolism was studied in both limbs on day 5 of treatment. Early observations on blood flow, oxygen uptake, and carbon dioxide output across the hind-limb muscle are shown. Close-arterially infused cimaterol, at a low dose, had a local direct effect on hind-limb muscle, significantly stimulating blood flow and oxygen uptake. The other leg did not show this effect. In the high dose treatment, recirculation of the agonist appears to have occurred as shown by the increased values in both treated and saline legs compared to saline only infusions. Thus it appears that direct effects of cimaterol on muscle are important in its action.

Publisher Summary This chapter elaborates the effect of growth hormone on muscle enzyme activity and lactate dehydrogenase isoenzyme pattern. Anabolic agents have been shown to both increases the proportion of red to white type muscle fibers and to promote protein deposition to a greater extent in red type muscle fibers. Red type muscle fibers differ from white type by having a higher oxidative capacity, higher citric acid cycle enzymes and a lactate dehydrogenase isoenzyme pattern similar to heart. This study examines the effect of growth hormone on muscle LDH isoenzyme pattern and the activities of glycolytic and citric acid cycle enzymes. Six growing wether lambs were treated with either saline or ovine GH for 5 days. Muscle biopsy samples were taken from a red type muscle on the last day of treatment. GH treatment caused a 52% increase in the activity of the citric acid cycle enzyme, citrate synthase but had no effect on the activities of the glycolytic enzymes phosphofructokinase, pyruvate kinase and LDH. There was a small but significant increase in the proportion of LDH 1, the isoenzyme which predominates in heart muscle. It is concluded that a short period of GH administration to growing lambs causes an increase in the oxidative capacity of a red type skeletal muscle and a shift towards the heart muscle enzyme pattern.

Publisher Summary This chapter discusses the effect of growth hormone on hind-limb muscle metabolism in growing lambs. Growth hormone (GH) has been shown to have lipolytic and diabetogenic, as well as growth promoting biological actions. The purpose of this experiment was to establish whether the diabetogenic actions of GH on muscle metabolism were related to a change in the supply of oxidizable substrates as a result of the increase in lipolytic rate. Hind-limb muscle metabolism was measured using arterio-venous difference and blood flow rate procedures in animals which had catheters inserted into the carotid artery and deep femoral vein. Growing wether lambs were treated with either saline or ovine GH for 11 days. Muscle metabolism was measured in 3 control and 3 GH treated lambs on day 11 of treatment. GH treatment significantly decreased hind-limb muscle glucose uptake without any change in arterial glucose concentration or lactate output. Plasma NEFA concentrations were increased by GH treatment, but this did not lead to a major change in either NEFA or 3-hydroxybutyrate uptake by the hind limb.
