Today’s small rotor helicopter vehicles mostly use electric motors and batteries as power sources. Most helicopter power configurations are either front powered or rear powered, causing the center of the helicopter to be offset. This kind of offset should be overcome by structural compensation or by adjusting the angle of attack of the blades, but some lift of the angle of attack will be sacrifice at the same time. This paper proposes a new type of electric helicopter powertrain design. The advantage of new type helicopter design is setting the centre of gravity at the centre position of helicopter. The main shaft of the helicopter through the power transmission method of the electric motor built into the internal gear of the star gear, which improves the design of the center offset of the helicopter. That is improved the flight stability of the helicopter. At the same time, in terms of power transmission, the motor power consumed by the primary gear power of the star gear, while the traditional helicopter power transmission method has three or four gear power losses. The new concept of electric motor with star gear structure proposed in this paper can effectively reduce the transmission power loss of the gear. In addition, the distributed power control system proposed in this paper also reduces the possibility of helicopter crash due to motor failure.

Generally, the occurrence of F region irregularities associated with Equatorial Plasma Bubbles is considered difficult to predict due to its day-to-day variability. Recent investigation shows that the dominant period of variability of EPB can be > 25 days (quasi 27 day), presumably associated with solar rotation period. VHF scintillation recorded from Pingtung, Taiwan in 2015 indicated the coexistence of planetary scale variability (4-8 days), 10-15 days variability and quasi 27 day variability. Lomb-Scargle power spectrum of S4 time series indicated that the larger period variability (quasi 27 days) is much more dominant than the other scales (which includes day-to-day variability). Interestingly, the dominant period of S4 variability corresponded exactly with the dominant period of high-latitude geomagnetic variability. Continuous operation of Equatorial Atmosphere Radar (EAR) in 2012 also indicated similar observation. Dominant period of variability of EPB in EAR observations corresponded with the high-latitude geomagnetic variability. These observations reveal that EPB are controlled predominantly by geomagnetic activity through large period variability (quasi- 27 days).

The F region evening electrodynamics in the equatorial region is characterized by a pre-reversal enhancement (PRE) in the zonal eastward electric field. Although the theoretical mechanisms for PRE are known, its variability, particularly day-to-day variability is not fully resolved. PRE is a large scale phenomenon driven by the F region dynamo after the sunset hours. This paper investigates whether the variability of the E region conductivity (particularly the one associated with the sporadic E, Es) has any influence on the F region dynamo and hence on the PRE of zonal electric field. Interestingly, ionosonde observations have indicated a higher occurrence of the blanketing type Es (Esb) over the low latitude on days with highly suppressed PRE of zonal electric field in comparison with the days with significantly larger PRE. Observational evidences presented in this paper suggests that the formation of the Esb in the evening hours is a sovereign process, not always controlled by the sheared F region vertical electric field of equatorial origin, mapping along the magnetic field line on to the low latitude E region. Model computations of the PRE suppression based on the measured Es densities have further substantiated the observational findings presented in this paper. These results clearly indicate that the low latitude Es has the potential to suppress the PRE of zonal electric field and possibly can play a vital role in explaining the PRE variability, particularly the day-to-day variability. Results have been discussed in light of earlier reports on PRE mechanisms and E-F region coupling processes.

To improve the accurate rate of mapping multi-spectral remote sensing images, in this paper we construct a class of HyperRectangular Composite Neural Networks (HRCNNs), integrating the paradigms of neural networks with the rule-based approach. The supervised decision-directed learning (SDDL) algorithm is also adopted to construct a two-layer network in a sequential manner by adding hidden nodes as needed. Thus, the classification knowledge embedded in the numerical weights of trained HRCNNs can be extracted and represented in the form of If-Then rules. The rules facilitate justification on the responses to increase accuracy of the classification. A sample of remote sensing image containing forest land, river, dam area, and built-up land is used to examine the proposed approach. The accurate recognition rate reaching over 99% demonstrates that the proposed approach is capable of dealing with image mapping.

To control illegal wildlife-product trade and protect endangered species of animals, unambiguous identification of the animal specimens is vitally important. Genetic approaches were adopted to identify animal species for conservation and to prevent their fraudulent misidentification in Taiwan, especially for samples of animal residues, powders, and processed products. PCR or nested PCR based on the nature of DNA was used for amplification of cyt b, COI, CHD, and D-loop DNA fragments. Sequences of these fragments were compared with those registered in DNA databases and phylogenetic analysis was performed. The established methods were applied in forensic cases for support of conservation efforts and they were proved to be robust. For conservation animal identification, various samples seized by law enforcement agents have been identified by our systems as rhinoceros horns, Indian sawback turtles, shahtoosh, ivories, dolphins, whales, etc. The systems were also successfully used in investigating the illegal trade of commercial turtle shells and the fraudulent misidentification of food contents on product labels in Taiwanese markets. This review summarizes the work conducted in our laboratory and describes the Taiwan experience.

Background: The complementary DNA (cDNA) encoding for Blo t 11, a 102 kD allergen from Blomia tropicalis (Bt) was isolated, expressed and characterized previously. This study aimed to isolate the native Blo t 11 allergen and compare its allergenicity with the recombinant forms. Methods: Native Blo t 11 (nBlo t 11) was isolated from crude Bt extract by immuno-affinity chromatography, analyzed by sodium dodecyl sulfatepolyacrylamide gel electrophoresis (SDS-PAGE) and Western blot, and verified by MALDI-TOF MS. Recombinant full-length Blo t 11 (rFL-Blo t 11) and its immunodominant peptide (fD) were expressed as glutathione S-transferase (GST)-fusion proteins in Escherichia coli. Immunoglobulin E (IgE) reactivity of the Blo t 11 allergens were determined by enzyme-linked immunosorbent assay (ELISA) and skin prick test. The inhibition capacity of the nBlo t 11 against fD and vice versa was determined by absorption studies. Results: Affinity purified nBlo t 11 was susceptible to degradation with the major degraded product resolved at 66 kD. The nBlo t 11 was confirmed by immunoblot analysis and MALDI-TOF MS that generated 13 peptides with complete identity to the deduced amino acid sequence of Blo t 11. Comparative in vitro and in vivo allergenicity tests and the cross inhibition studies between the native and recombinant Blo t 11 showed that recombinant fD, but not the rFL-Blo t 11, has comparable IgE reactivity with the native counterpart. Conclusions: This comparative study confirmed that the recombinant peptide fD contains the main immunodominant region of Blo t 11. This recombinant peptide, instead of the full-length protein, is a good candidate for diagnostic and therapeutics development for mite allergy.

Based on product function density analysis, this article chooses cellular phones as a research subject among all types of smart products, and investigates the effects of on-screen menu searching aids and function key layouts to the first-time users. The results show that among three types of on-screen menu searching aids, the tree structure and list structure aids are superior to the digit aid. In terms of function key layouts, symbol labeling is better than text labeling while no significant difference was found between parallel layouts and cross layouts. For first-time users, on-screen menu searching aids provide structural information for building mental model toward using a cellular phone. Thus, if the searching aids are designed compatibly, the learning efficiency for menu searching tasks can be increased.

Background: A 98-kDa mite paramyosin (Der f 11) from Dermatophagoides farinae (Df) is highly allergenic, and its cDNA (Df642) has been cloned. This paper describes the sequence characteristics and the mapping of the immunodominant human IgE and IgG epitopes of Der f 11. Methods: The protein sequence analysis was performed with a combination of FASTA, GCG, and CLUSTAL W computing packages. The whole cDNA insert and its PCR-derived DNA fragments were generated and expressed in E. coli. These overlapping recombinant peptides (F1 to F5) were used for B-cell epitope mapping with 18 mite-allergic sera by dot immunoassays. Results: Df642 cDNA encodes a partial sequence that contains the 2nd to 26th 28-residue repeats and lacks the N-terminus and the C-terminus. The sequence identity of the Der f 11 with other known paramyosins is 34–60%. The dominant IgE epitopes are located in peptides F1 and F4, whereas the dominant IgG epitopes are located in peptides F1 and F2. These peptides are more reactive than whole rDf642. Conclusions: Mite paramyosin is very similar to other known paramyosins. The human IgE and IgG epitopes are scattered throughout the entire molecule. Data also indicate the presence of unique IgE and IgG epitopes in Der f 11.

Background The important dust mite allergens identified to date are of molecular weights ranging from 14 to 60 kDa. Our previous protein study indicated that the 98-kDa native paramyosin in Dermatophagoides farinae mite showed IgE reactivity with 82% of the mite-sensitive asthmatic patients suggesting that it is a novel major mite allergen. This study described the isolation and characterization of the cDNA clone encoding the 98-kDa mite allergen. Methods A Dermatophagoides farinae cDNA library was constructed in lambda ZAPII vector and the library was immunoscreened with a monoclonal antibody 642. The cDNA insert was sub-cloned into M13 sequencing vector for single-stranded sequencing. The whole cDNA insert was expressed in pGEX-2T Escherichia coli expression system as a fusion protein with GST. The allergenicity of the recombinant peptides was tested by skin tests and IgE immunoassay. The IgE and IgG immunoassays were performed with sera from 20 mite-allergic patients. Results The cDNA clone Df642 was 2134 bp long, coding for a polypeptide of 711 amino acid residues. Protein sequence analysis and alignment confirmed that the deduced polypeptide is a mite paramyosin which is truncated slightly at the N- and C-terminuses. In vivo skin tests and in vitro IgE-binding study showed that 62% (13/21) and 50% (10/20) of the mite-sensitive asthmatic patients reacted positively with the recombinant Dermatophagoides farinae paramyosin, respectively. Conclusion The study indicated that 98-kDa mite paramyosin is an important allergen.

We report a detailed investigation of the “positive” persistent photoconductivity (PPC) and “negative” persistent photoconductivity (NPPC) in semimetallic AlxGa1−xSb/InAs quantum wells. The studies of the NPPC and PPC effects have been performed under various conditions, such as different photon energy of excitation, different temperature, different Al composition x, and different well width. It is found that all the previously proposed mechanisms fail to explain several of our observations. We suggest that the NPPC and PPC effects are produced principally by two competing processes. At a high temperature, the photoconduction is dominated by the photogenerated electrons in the InAs well, in which the relaxation of the excess electrons is prohibited by an energy barrier due to the trapping of photoexcited holes by deep defects in the InAs well. Cytokeratin-19 is an intermediate filament protein associated with the integrity of cell structure, and its elevated expression has been reported to correlate with the disease progression of oesophagus and lung cancers. In this study, we examined the level of cytokeratin-19 in five cervical cancer cell lines by immunobinding and Western blotting analyses. Compared with two control cell lines, FS-4 (foreskin cell line) and G9T (glioma cell line), all five cervical carcinoma cell lines (Caski, CC7T, ME180, HeLa and SIHA) showed higher cytokeratin-19 expression. By double-staining flow cytometry, expression of cytokeratin-19 in cervical cancer cells was suggested to be in a cell cycle-independent manner. Furthermore, we could specifically localize the SIHA cell-derived tumours in nude mice by injecting with cytokeratin-19-recognized radiolabelled MAb Cx-99 antibody, suggesting the possibility of using cytokeratin-19 as a marker of cervical carcinoma. A clinical investigation was therefore performed on 19 patients (11 patients with cervical carcinoma and eight patients with benign neoplasia). In the 11 patients having cervical carcinoma, all eight patients with advanced stages and one out of three patients with early stage diseases showed higher cytokeratin-19 protein contents than the other 10 patients with benign neoplasia. This suggested that elevation of cytokeratin-19 level was associated with cervical cancer staging. In addition, we have studied the biological significance of elevated cytokeratin-19 level in malignant cervical cancer. The apoptotic rate of cervical carcinoma cells in response to cisplatin was increased if their cellular cytokeratin-19 level was reduced by specific antibody MAb Cx-99. These results indicated that elevation of cytokeratin-19 expression could associate with the apoptotic resistance and malignant progression of cervical carcinoma.

Summary Background The group 7 mite allergens react with IgE in 50% of sera from allergic patients. Objective To determine the molecular and antigenic characteristics and heterogeneity of Der f 7 in mite extracts. Methods Monoclonal antibodies (MoAbs) produced from mice immunized with recombinant Der f 7 were examined for crossreactivity to Der p 7 and then used for immunoblotting of 1 and 2-D gel electrophoresis. Deglycosylation was studied with N-glycosidase-F and N-terminal sequencing by Edman degradation. The epitopes of the monoclonal antibodies were compared by cross-inhibitory immunoassays. Results Immunoblotting of D. farinae extracts with all the anti Der f 7 MoAbs showed major reactivities at 31, 30 and 25 kDa. The strongest immunostaining was at 25 kDa which contrasted with Der p 7 where the 31 and 30 kDa bands were strongest. The relative strength of staining however varied between extracts. The 31 and 30 kDa components were glycosylation products of the 25 kDa form which had the N-terminal sequence predicted from cDNA analysis. Two MoAbs stained an 18 kDa band consistent with a degradation product. The 2-D gels showed that different components with pls from 5.6–6.4. Both species-specific and Der p 7 crossreactive MoAbs were produced and a two-site ELISA assay for detecting group 7 allergen was developed with MoAbs recognizing different epitopes. Conclusions Der f 7 has been defined by its natural N-terminal sequence and MoAbs. It apparently exists as different glycosylation and degradation products in mite extracts, the relative abundance of which differs with different preparations. A two-site ELISA to measure the allergen was developed.

The Met121Glu azurin mutant has been crystallized and the structure determined at a resolution of 2.3 A. In the crystal structure a carboxyl oxygen of Met121Glu is coordinated to the metal at a distance of 2.2 A. Single-crystal resonance Raman spectroscopy was used to show that the glutamic acid residue in the copper site was in the protonated state. Titration of this residue gives rise to a number of unusual, pH-dependent properties: as the pH is increased from 4 to 8, the S(Cys)-Cu ligand-to-metal charge transfer bands are blue shifted and their intensity ratio is reversed, the EPR signal changes from type 1 copper to a new form of protein-bound copper, and the redox potential changes from 370 to 180 mV. The spectroscopic changes in this pH interval are consistent with a two-state model. From the pH dependence of the optical and EPR spectra, pKa = 5.0 for the glutamic acid in the oxidized protein was determined.

Conservation of the endangered species is a global trend and an important issue. Many countries have legislated against illegal trade of the endangered species. For forensic scientists, unambiguous identification of the animal species is one of the strategies to investigate the illegal trade and protect the endangered species. In tradition, the morphological and chemical approaches are adopted to identify the animal species. However, molecular DNA tool is one of the alternative and most effective methods presently. DNA is more stable and polymorphic than the other previous used markers. The DNA analysis methods are usually sensitive. Therefore, a small quantity of specimen is sufficient for the following analysis. The DNA analysis methods, such as RFLP, PCR–RFLP, RAPD, AFLP, PCR (or nested PCR)-sequencing of the specific loci, and STR typing, have been widely used in genetic analysis of animals (including the endangered species) for species identification, genealogy analysis (e.g., paternity test) and population monitoring on forensic science, phylogenetic analysis, population genetics, and ecology. Validation of the DNA identification methods is absolutely important to ensure the efficacy and accuracy.

The structure of the azurin mutant nickel-Trp48Met from Pseudomonas aeruginosa has been determined by difference Fourier synthesis using phases from the wild-type azurin model. The final crystallographic R value is 0.170 for 17 394 reflections to a resolution of 2.2 A. The mutant crystallized in the orthorhombic space group P2(1)2(1)2(1), a = 57.4, b = 80.4, c = 110.3 A. The four molecules in the asymmetric unit are packed as a dimer of dimers. The nickel metal site of this mutant structure is similar to the zinc metal site in the azurin Asp47 mutant. The site-specific mutation was performed at residue Trp48, which is located in the center of the protein in a highly hydrophobic environment, to investigate its suggested role in the long-range electron-transfer pathway between the disulfide bond on one side of the protein to the Cu centre. The structure around the mutation site Met48 showed no significant change compared with the wild-type structure.

Radioimmunodetection (RAID) is more sensitive and specific than conventional diagnostic methods. In this study, a monoclonal antibody against cervical carcinoma antigen, MAb Cx-99, was labeled with 111Indium (111In). This immunoconjugate was intravenously injected into athymic nude mice bearing cervical squamous cell carcinoma (SCC) xenografts. The tissue distribution study showed that the xenograft tumor had higher binding activity than most other tissues after 48 h from injection, demonstrated by localization ratio of tumor of tissues (c.p.m./g) against blood (c.p.m./g). However, this localization ratio was also high in the liver, spleen and kidney. The imaging study by immunoscintigraphy also showed that the tumor and liver were distinct from other background tissues 2 days after injection. This preliminary study showed that 111In-labeled MAb Cx-99 may have potential for RAID of cervical cancer, especially for tumors in the pelvis.

The crystal structure of azurin mutant Phe114Ala from Pseudomonas aeruginosa has been solved by molecular replacement. The final crystallographic R value is 0.185 for 9832 reflections to a resolution of 2.6 A. The root-mean-square deviation for main-chain atom positions is 0.020 A between the four independent monomers in the asymmetric unit. The mutant Ala114 crystallized from PEG 4000 in a new crystal form and the crystals are monoclinic, P2(1), a= 51.0, b = 83.6, c= 66.4 A and beta = 110.5 degrees. The four molecules in the asymmetric unit are packed as a dimer of dimers and are related by an approximate twofold axis. The dimer packing and the dimer contact region are very similar to that of the Alcaligenes denitrificans azurin dimer. The mutation was performed at residue Phe114, which exhibits a pi-electron overlap with the copper ligand His117, to investigate its suggested role in the electron self-exchange reaction. Removal of steric constrains from the phenylalanine side chain created a somewhat different geometry around the copper site with an increased mobility of His117 resulting in an enlarged Cu-N length which may be responsible for the slight differences obtained in the spectral properties of the mutant versus the wild-type protein.

The sealing characteristics of an air-cooled gas turbine disk cavity have been studied using laser sheet flow visualization. Experiments were performed on a simplified half-scale model of an actual gas turbine disk cavity. This type of rotor–stator geometry with a double-toothed-rim (DTR) seal at the outer periphery and a labyrinth seal at the inner periphery of the cavity has been tested for its ability in preventing ingress of hot mainstream gases. The results show good agreement with previously esimated design data. Experiments were conducted for various labyrinth seal flow rates and rotational Reynolds numbers up to 1.52 × 106. The effects of rotor eccentricity on minimum purge flows have also been discussed.

The Pseudomonas aeruginosa azurin mutant Asn47Asp has been isolated, its spectroscopic and kinetic properties characterized, and the X-ray crystal structure of its zinc derivative determined. While the optical and electron paramagnetic resonance spectra as well as the electron-transfer activity of the mutant are very similar to the wild-type values, the Asn47Asp reduction potential is slightly increased by 20 mV. The mutant crystallized in the orthorhombic space group P2(1)2(1)2(1) with cell dimensions a = 57.8, b = 81.5 and c = 112.6 A. There are four molecules in the asymmetric unit, packed as a tetramer which consists of two independent dimers. The zinc site of this mutant structure is similar to the wild-type zinc azurin and, in particular, the metal-binding site is almost identical to the site found in the wild-type zinc-azurin structure [Nar, Huber, Messerschmidt, Filippou, Barth, Jaquinod, Kamp & Canters (1992). Eur. J. Biochem. 205, 1123-1129]. The Asp47 side chain at that mutation site takes on a very similar orientation to Asn47 in the wild-type structure preserving the two hydrogen bonds with the neighbouring Thr113 NH and O(gamma)H. Therefore, the increased reduction potential of the mutant is probably a result of an altered charge distribution close to the metal site.

Experimental data from a constellation of five NOAA POES satellites were used for studying the penetration of solar energetic particles (SEP) to high latitudes during long-lasting SEP events on December 5 to 15, 2006. We determined cutoff latitudes for electrons with energies >100 keV and > 300 keV, and protons with energies from 240 keV to >140 MeV. The large number of satellites allowed us to derive snap shots of the cutoff boundaries with 1-hour time resolution. The boundaries were fitted well by ellipses. Based on the elliptical approach, we developed a model of cutoff latitudes for protons and electrons in the northern and southern hemispheres. The cutoff latitude is represented as a function of rigidity, R, of particles, MLT, geomagnetic indices: Dst, Kp, AE, and dipole tilt angle PS. The model predicts tailward and duskward shifting of the cutoff boundaries in relation to intensification of the cross-tail current, field-aligned currents, and symmetrical and asymmetrical parts of the ring current. The model was applied for prediction of a polar cap absorption (PCA) effects observed at high latitudes by CADI network of ionosondes. It was found that the PCA effects are related mainly to intense fluxes of >2.5 MeV protons and >100 keV electrons, which contribute mostly to the ionization of ionospheric D-layer at altitudes of ~75 to 85 km. This finding was confirmed independently by FORMOSAT-3/COSMIC observations of the SEP-associated enhancements of electron content at altitudes of ~80 km. Every year, approximately 80,000 to 150,000 ACL tears occur.[1,2] Post-injury, ACL reconstruction (ACLR) is often recommended to restore functional stability and prevent long-term joint degradation. However, while surgical techniques have improved, individuals with ACLR have been shown to have a higher risk of developing osteoarthritis (OA) and a higher rate of re-injury.[3] The higher incidence and earlier onset of knee OA in individuals who have undergone ACLR may be a result of a post-injury movement strategy that utilizes a higher degree of muscle co-contraction of the hamstrings and the quadriceps muscles during landing to increase joint stability.[4] This increased co-contraction leads to a “stiff” landing pattern, decreases shock absorption, and increases the ground reaction forces. These changes in lower extremity biomechanics also may result in higher forces which likely place the knee cartilage under greater stress and higher risk of OA development and re-injury.Copyright © 2010 by ASME

A group of six mouse monoclonal antibodies (MoAbs) with the unusual lambda-type light chain were generated by fusion of NS-1 cells with splenic cells derived from BALB/c mice immunized with crude extracts of Bermuda grass pollen (BGP). Four of them were IgG1, one was IgG2b, and one was IgG3. Binding inhibition assay showed that they recognized the same (or very similar) epitope. Using sera from BGP-allergic patients, it was found that the specific binding between the IgE antibodies and the MoAb 26-11-fixed antigen could be blocked by MoAb 26-11 itself and another MoAb 9-13 in a dose-dependent manner. It appears that the epitope recognized by the lambda-type MoAbs is a human IgE-binding antigenic determinant. Further physico-chemical analyses showed that this epitope was stable under heat but sensitive to treatments of sodium periodate and proteinase K. Results from these studies indicate that this unique epitope which leads to the generation of lambda-type MoAbs is part of a glycoprotein.

[1] We studied ionization effects produced by solar energetic particles (SEP) in the northern polar ionosphere and atmosphere during December 2006 (local night). Height profiles of specific ionization produced by the SEP in the polar cap were calculated using POES data on energetic particles. The SEP ionization dynamics reveals three intensifications at heights from 50 to 100 km in 7 to 8, 13 and 14 December. At the same time a 3-D tomography of electron content (EC) in the ionosphere and upper atmosphere was performed on the base of COSMIC/FORMOSAT-3 sounding data. We found pretty close temporal and spatial patterns for the EC enhancements and for the SEP ionization. The discrepancies might be attributed to three effects: unaccounted contribution of low-energy electrons to the ionization of lower ionosphere, fast growth of electron losses with the depth in atmosphere, and strong contribution from storm-time ionospheric disturbances.

Cytokeratin-19 is an intermediate filament protein associated with the integrity of cell structure, and its elevated expression has been reported to correlate with the disease progression of oesophagus and lung cancers. In this study, we examined the level of cytokeratin-19 in five cervical cancer cell lines by immunobinding and Western blotting analyses. In the 11 patients having cervical carcinoma, all eight patients with advanced stages and one out of three patients with early stage diseases showed higher cytokeratin-19 protein contents than the other 10 patients with benign neoplasia. This suggested that elevation of cytokeratin-19 level was associated with cervical cancer staging. In addition, we have studied the biological significance of elevated cytokeratin-19 level in malignant cervical cancer. The apoptotic rate of cervical carcinoma cells in response to cisplatin was increased if their cellular cytokeratin-19 level was reduced by specific antibody MAb Cx-99. These results indicated that elevation of cytokeratin-19 expression could associate with the apoptotic resistance and malignant progression of cervical carcinoma.

To determine the prognostic values of tissue polypeptide antigen (TPA), squamous cell carcinoma antigen (SCC-Ag), and carcinoembryonic antigen (CEA) in the sera of cervical carcinoma patients, especially in those with a poor prognosis.In this retrospective study, the preoperative serum SCC-Ag, TPA, and CEA were analyzed in 779 patients with cervical squamous cell carcinoma of stage Ib-IIa who received radical hysterectomy and pelvic lymph node dissection (RAH-PLND) between 1984 and 1994.Due to poor predictive value and poor correlation between serum CEA and clinico-pathological factors, CEA was abandoned in this study. Elevated TPA and SCC-Ag levels, pelvic lymph node metastasis (PLNM), lymphvascular space involvement (LVSI) and deep stromal invasion (DSI) were associated with poor survival time by univariate analysis. The correlation study showed that elevated serum TPA was significantly related to PLNM, LVSI, and DSI (p = 0.004, 0.008, and 0.021, respectively), and SCC-Ag was related to PLNM and bulky tumor size (p = 0.001 and 0.02, respectively). In the multivariate analysis, only PLNM and LVSI remained independently significant indicating poor survival. Further stratification studies by PLNM and LVSI showed that elevated TPA levels could even indicate higher recurrence rates in patients with PLNM (p = 0.045), as well as SCC-Ag in patients with LVSI (p = 0.038).The results suggest that both elevated TPA and SCC-Ag levels depicting poor prognosis in stage Ib-IIa cervical SCC, especially indicates a group of high-risk patients who may need more aggressive therapy.

A novel nomenclature for the hypervariable microsatellite DNA, APOAI1 locus, is proposed. The complex nature of the repeat unit in this locus results in alleles separated by a single base. Polymerase chain reaction (PCR) products amplified from this locus were separated by single-strand conformation polymorphism (SSCP) electrophoresis. All the single-stranded DNA bands on the SSCP gel were removed from the gel and a second amplification performed. Homozygous DNA fragments amplified from single-stranded DNA were sequenced. From the 100 individuals studied, 30 alleles and 73 genotypes were found. A system of nomenclature for the APOAI1 locus is provided that is logical and in line with previous models. Using the primers described, the locus can be amplified and alleles designated on the basis of size. This system of nomenclature will assist in the exchange of data between laboratories for this locus.

The site-specific mutant alanine 114 of the blue copper protein azurin from Pseudomonas aeruginosa in Escherichia coli has been crystallized from PEG 4000 in a new crystal form compared to the wild type utilizing the hanging-drop procedure. The crystals are blue well-formed prisms. Monoclinic, P2(1), a = 51.03 (5), b = 83.36 (5), c = 66.30 (6) A and beta = 111.0 (1) degrees. 14,875 reflections up to 2.7 A have been collected using a modified Syntex P2(1) automated four-circle diffractometer.

Dermatophagoides pteronyssinus (Dp) and Dermatophagoides farinae (Df), the major components of house dust, are considered to be etiologic factors of extrinsic asthma. The relationships between immunoglobulin (Ig) G subclass antibodies specific for Dp (or Df) were compared in specific IgE-positive and -negative asthmatic children.Serum levels of IgG and IgE subclass antibodies specific for Dp and Df were studied in 52 children (age, 3-13 years; mean age, 8.4 years) with asthma using enzyme-linked immunosorbent assays. The skin prick test was also used in diagnosis of the reactivity of allergic disease.The levels of serum-specific IgG1 and IgG2 to Dp and Df in mite-specific IgE-(or skin-test) positive asthmatic children were significantly higher than those in mite-specific IgE- (or skin test) negative children (p0.01). Significant correlations between the level of the specific IgE and IgG1 (r = 0.6067; p = 0.0001) or IgG2 (r = 0.5851; p = 0.0002) to Dp, and IgG1 (r = 0.3823; p = 0.0214) or IgG2 (r = 0.5057; p = 0.0017) to Df were found. The specific IgE level and skin test reactivity showed a high correlation of greater than 96% (50/52).The levels of mite-specific IgG subclasses were partially compatible to specific IgE levels and skin test reactivity. We conclude that house dust mite allergy is significantly associated with specific IgG1, IgG2 and IgE responses.

The ABO locus on chromosome 9 contains many more alleles than are currently used routinely in forensic science. The use of single-strand conformation polymorphism (SSCP) can separate sequence polymorphisms that differ by only one base. The SSCP process used allows for both single- and double-stranded polymerase chain reaction (PCR) products to be visualized. The six ABO genotypes can be differentiated by the allele-specific B and O SSCP patterns. The double-stranded DNA produced 'hybrid' bands due to heterozygous samples and allowed sequence diversity to be detected between alleles of heterozygotes. These 'hybrid' bands are valid markers to confirm genotypes of specimens.

The effect of retinoic acid and dexamethasone on alkaline phosphatase (AP) expression was investigated in human breast cancer MCF-7 cells. Cellular AP activity was induced significantly by retinoic acid or dexamethasone in a time-dependent and dose-dependent fashion. A marked synergistic induction of AP activity was observed when the cells were incubated with both agents simultaneously. Two AP isozymes, tissue-nonspecific (TNAP) and intestinal (IAP), were shown to be expressed in MCF-7 cells as confirmed by the differential rate of thermal inactivation of these isozymes and RT-PCR. Based on the two-isozyme thermal-inactivation model, the specific activities for TNAP and IAP in each sample were analyzed. TNAP activity was induced only by retinoic acid and IAP activity was induced only by dexamethasone. Whereas dexamethasone conferred no significant effect on TNAP activity, retinoic acid was shown to inhibit IAP activity by approximately 50%. Interestingly, TNAP was found to be the only isozyme activity superinduced when the cells were costimulated with retinoic acid and dexamethasone. Northern blot and RT-PCR analysis were then used to demonstrate that the steady-state TNAP mRNA level was also superinduced, which indicates that the superinduction is regulated at the transcriptional or post-transcriptional levels. In the presence of the glucocorticoid receptor antagonist RU486, the dexamethasone-mediated induction of IAP activity was blocked completely as expected. However, the ability of RU486 to antagonize the action of glucocorticoid was greatly compromised in dexamethasone-mediated superinduction of TNAP activity. Furthermore, in the presence of retinoic acid, RU486 behaved as an agonist, and conferred superinduction of TNAP gene expression in the same way as dexamethasone. Taken together, these observations suggest that the induction of IAP activity by dexamethasone and the superinduction of TNAP by dexamethasone were mediated through distinct regulatory pathways. In addition, retinoic acid plays an essential role in the superinduction of TNAP gene expression by enabling dexamethasone to exert its agonist activity, which otherwise has no effect.

Cyn d 1, the major allergen of Bermuda grass pollen, consists of a number of isoforms.To examine the extent of sequence variation of Cyn d 1 isoforms at the molecular level.A Bermuda grass pollen lambdaZAP II cDNA expression library was immunoscreened with anti-Cyn d 1 monoclonal antibodies. The reactive clones were isolated, subcloned into Escherichia coli, and sequenced. Some of them were expressed in the yeast Pichia pastoris to obtain recombinant Cyn d 1 proteins.Ten cDNA clones were obtained, all these clones encode the full length of Cyn d 1 protein. Their deduced mature proteins can be grouped into: the long ones with 246 amino acids, and the short ones with 244 amino acids. The last two amino acids (AG) of the long Cyn d 1 are deleted in the short Cyn d 1. The remaining amino acid sequences share more than 98% identity; a total of nine amino acid variations were observed. Two recombinant Cyn d 1 proteins (rCyn d 3-2 and rCyn d 5-4) with three amino acid substitutions showed differential IgE-binding profiles.The present study extended our understanding of the primary structure of isoforms of Cyn d 1.

Human foods are usually prepared by cooking. Boiling of chicken egg-white (EW) led to decreased allergenicity, and abrogated intestinal uptake of immunoreactive ovalbumin (OVA) when fed to mice. Therefore, the effects of oral administration of boiled EW were examined further in BALB/c mice. Specific IgE, IgG1 and IgG antibody responses were suppressed by raw EW, but not by EW boiled for 5 or 60 min, fed prior to sensitization with 10 microg OVA or 1 microg DNP-OVA in alum. Similar results were obtained when mice were sensitized with 10 microg conalbumin, ovomucoid or lysozyme in alum. BALB/c spleen cell proliferation and secretion of Th2 cytokines IL-4 and IL-5 during in vitro stimulation with OVA were also suppressed by feeding raw EW, but not by boiled EW. Although heat denaturation of proteins can minimize allergenicity, the present results suggest that over-cooking of proteins may affect their intestinal antigen processing and thus prevent the induction of oral tolerance.

The effects of estradiol, tamoxifen, and retinoic acid on the proliferation of breast and cervical cancer cells were investigated. Estrogen stimulated only MCF-7 cell growth, whereas tamoxifen and retinoic acid inhibited the proliferation of all cells studied. Northern blot analysis indicated that estradiol up-regulates c-myc mRNA level in all cell lines studied regardless of the estrogen receptor status in the cells. On the contrary, tamoxifen inhibits c-myc gene expression in all cell lines studied except in MCF-7 cells where the c-myc transcript was not affected. The inhibitory effect of tamoxifen on c-myc gene expression and cell proliferation in estrogen receptor-negative cells suggest an estrogen receptor-independent mechanism. The results also suggest that different mechanisms are involved in the regulation of cell growth and c-myc gene expression in different cancer cells by estrogen and tamoxifen.

The group 7 mite allergens react with IgE in 50% of sera from allergic patients.To determine the molecular and antigenic characteristics and heterogeneity of Der f 7 in mite extracts.Monoclonal antibodies (MoAbs) produced from mice immunized with recombinant Der f 7 were examined for crossreactivity to Der p 7 and then used for immunoblotting of 1 and 2-D gel electrophoresis. Deglycosylation was studied with N-glycosidase-F and N-terminal sequencing by Edman degradation. The epitopes of the monoclonal antibodies were compared by cross-inhibitory immunoassays.Immunoblotting of D. farinae extracts with all the anti Der f 7 MoAbs showed major reactivities at 31, 30 and 25 kDa. The strongest immunostaining was at 25 kDa which contrasted with Der p 7 where the 31 and 30 kDa bands were strongest. The relative strength of staining however varied between extracts. The 31 and 30 kDa components were glycosylation products of the 25 kDa form which had the N-terminal sequence predicted from cDNA analysis. Two MoAbs stained an 18 kDa band consistent with a degradation product. The 2-D gels showed that different components with pIs from 5.6-6.4. Both species-specific and Der p 7 crossreactive MoAbs were produced and a two-site ELISA assay for detecting group 7 allergen was developed with MoAbs recognizing different epitopes.Der f 7 has been defined by its natural N-terminal sequence and MoAbs. It apparently exists as different glycosylation and degradation products in mite extracts, the relative abundance of which differs with different preparations. A two-site ELISA to measure the allergen was developed.

The effects of the macromolecular synthesis inhibitors 5,6-dichloro-1-beta-D-ribofuranosyl benzimidazole (DRB), actinomycin D, and cycloheximide on the human gastric cancer TMK-1 cell line were studied. These agents inhibited DNA, RNA, or protein synthesis efficiently and induced cell death rapidly in a wide range of concentrations. After 8 hr of exposure to these agents, the cells exhibited morphological features of apoptosis, including cell shrinkage, nuclear condensation, DNA fragmentation, and formation of apoptotic bodies. Western blot analysis revealed that these inhibitors altered the protein levels of apoptosis-related gene products such as c-Myc, Bcl-X(S), and the mutant p53 (mp53) in TMK-1 cells markedly. The c-myc mRNA and protein levels were decreased initially and were then induced markedly to a new level after 4 hr of exposure to DRB, a RNA polymerase II inhibitor. The Bcl-X(S) levels were increased rapidly after treatment with all of these agents, whereas the levels of Bcl-X(L) and Bax remained largely unchanged. Northern blot analysis indicated that the c-myc overexpression is concomitant to DRB-induced DNA fragmentation and that the increased mp53 protein level was mainly a posttranscriptional event. Our observations suggest that the up-regulation of Bcl-X(S) may serve as an important mechanism for the apoptosis triggered by these inhibitors. This study also provides evidence for the notion that interference with the cellular survival pathway may lead to apoptosis.

The crystal structure of cobalt-substituted azurin from Pseudomonas aeruginosa has been determined to final crystallographic R value of 0.175 at 1.9 A resolution. There are four molecules in the asymmetric unit in the structure, and these four molecules are packed as a dimer of dimers. The dimer packing is very similar to that of the wild-type Pseudomonas aeruginosa azurin dimer. Replacement of the native copper by the cobalt ion has only small effects on the metal binding site presumably because of the existence of an extensive network of hydrogen bonds in its immediate neighborhood. Some differences are obvious, however. In wild-type azurin the copper atom occupies a distorted trigonal bipyramidal site, while cobalt similar to zinc and nickel occupy a distorted tetrahedral site, in which the distance to the Met121,S(delta) atom is increased to 3.3-3.5 A and the distance to the carbonyl oxygen of Gly45 has decreased to 2.1-2.4 A. The X-band EPR spectrum of the high-spin Co(II) in azurin is well resolved (apparent g values gx' = 5.23; gy' = 3.83; gz' = 1.995, and hyperfine splittings Ax' = 31; Ay' = 20-30; Az' = 53 G) and indicates that the ligand field is close to axial.

The crystal structures of the azurin mutant Met121Ala and its azide derivative Met121Ala-azide from Pseudomonas aeruginosa have been determined. The final crystallographic R values are 21.3 and 19.4% for the two structures, respectively. In the Met121Ala mutant, the distance between the copper ion and His117 increases by 0.34 A compared with the wild-type structure. The removal of the methionine in the apical position induces a shortening of the distance from the copper ion to the carbonyl O atom of Gly45 from 2.97 to 2.74 A. In the Met121Ala-azide structure, the azide anion occupies the cavity created by replacing the Met121 side chain with the smaller methyl group of Ala. The azide anion binds with a terminal N atom to the copper ion at a distance of about 2.04 A. In addition, the copper ion has moved out of the trigonal plane by about 0.26 A towards the azide anion. Thus, the copper site in this structure has a distorted tetrahedral arrangement. The spectroscopic characteristics show, in addition, that the copper sites in the two structures are distinctively different. The Met121Ala mutant still maintains the properties of an ordinary type 1 copper site while the Met121Ala-azide derivative has an absorption maximum at about 409 nm and the copper hyperfine coupling has increased to a value intermediate between those of type 2 copper and the wild-type azurin.

The purposes of this study were to investigate the effects of hemodialysis patients' health belief, knowledge on uremia, and social support upon their compliance behavior. The convenience sample of 330 hemodialysis (HD) patients was obtained at HD centers in southern Taiwan. With the use of a questionnaire developed by the researcher, all subjects were interviewed during hemodialysis. Data analyses were processed by a personal computer with SPSS/PC. Pearson correlation, ANOVA, chi-square, multiple regression and factor analysis were selected as the analysis methods for this study. The results indicated: (1) The average overall rate of compliance was 72.1% by patient self report for fluid limit, diet restriction and taking PBM. (2) Two factors of subject's health belief were identified by factor analysis. (3) Those who had more positive motivation for compliance with therapeutic regiments, more knowledge on uremia and stronger social support were positively correlated with compliance behaviors. (4) The best predictive variables of compliance behaviors of HD patients included positive motivation, knowledge on uremia, educational level, current daily urine amount and age; these five variables explained 23% of variance in compliance behaviors. (5) The instruments with a satisfactory validity and reliability developed by the researcher could provide a valuable basis for relevant future research. Implications of these findings for nursing practice are also discussed.

Cervical cancer is a predomiannt gynecologic neoplasia in Taiwan. The associations of carcinoembryonic antigen (CEA), tissue polypeptide antigen (TPA) and squamous cell carcinoma antigen (SCC-Ag) for detection of cervical cancers with type of cancer, clinical stage and lymph node metastasis were studied. Serum samples were collected from 395 patients with cervical cancers (61 CIN3, 334 squamous cell carcinoma, 18 adenocarcinoma), 72 patients with benign gynecologic diseases and 115 healthy controls. The sensitivities of CEA, TPA, and SCC-Ag were 15.2%, 33.6%, and 49.3%. The specificities were 89.3%, 96.2%, and 95.1%, respectively. The positive rate of CEA was found significantly higher among patients with adenocarcinoma than those with squamous cell carcinoma. The positive rate of CEA and SCC-Ag increased with advance of the clinical stages. There was no significant difference in the rate of each tumor marker between patients with lymph node metastasis and those without metastasis. The positive rate obtained by SCC-Ag alone was comparable to those by the combinations of two or three antigens. The result of the present study indicates that SCC-Ag is the most useful tumor marker in the detection of cervical cancers.

The trend towards higher gas turbine inlet temperatures is a natural consequence of the pursuit of higher turbine operating efficiencies. More efficient disk cooling technology is therefore a prime need. The sealing characteristics of a advanced air-cooled turbo-expander disk cavity have been studied using laser sheet flow visualization and static pressure measurements. Experiments were performed on a simplified half-scale model of an actual low pressure turbo-expander first-stage disk cavity. The rotor-stator geometry tested was equipped with a double-toothed rim (DTR) seal at the outer periphery and a labyrinth seal (or shaft seal) at the inner periphery of the cavity. This is one of the first studies that incorporates the effects of shaft sealing flows. Experiments were conducted in the absence of an external flow stream for various labyrinth seal flow rates, and rotational Reynolds numbers up to 1.52 × 106. The results confirm the adequacy of previously estimated design data for this disk cavity configuration. Pressure measurements reveal that a pressure difference criterion based on the differential pressure across the teeth of the rim seal can be used to detect ingress of mainstream flow. The superior performance of the seal geometry studied was confirmed by a comparison against a single-toothed rim seal and a simple axial rim seal.Copyright © 1992 by ASME

The antimutagenic effects of Maillard reaction products (MRPs) prepared by heating three sugars (fructose, glucose and xylose) and four amino acids (arginine, glycine, lysine and tryptophan) at 100 degrees C for 10 hr was evaluated in the Salmonella/microsome assay. The highest extent of browning was found in the MRPs of sugars-lysine and xylose-amino acids. The MRPs of xylose-amino acids showed stronger antioxidative activity and reducing power than did the other combinations. No mutagenicity or toxicity in Salmonella typhimurium TA98 was observed with any of the MRPs in the presence of S-9. Most MRPs, especially those of sugars-tryptophan and xylose-amino acids, strongly inhibited the mutagenicity of 2-amino-3-methylimidazo(4,5-f)quinoline (IQ), 3-amino-1,4-dimethyl-5H-pyridol-(4,3-b)indole (Trp-P-1) and 2-amino-6-methyldipyrido(1,2-a:3',2'-d)imidazole (Glu-P-1) in the presence of S-9. However, the MRPs of fructose-glycine and fructose-arginine increased the mutagenicity of Trp-P-1. The antimutagenic effect of the MRPs was well correlated with their antioxidative activity and reducing power. The mutagenicity of benzo[a]pyrene was moderately inhibited by most MRPs, but was increased by the MRP of glucose-arginine. Aflatoxin B1 mutagenicity was increased greatly by all the MRPs except that of xylose-tryptophan. The findings suggested that MRPs might have a bifunctional property of co-mutagenicity and antimutagenicity in certain cases.

A model describing the time constant of a wide-band microwave differentiator is presented. The time constant combined with the amplitude response dictates the circuit behaviour of a microwave differentiator. By representing the formulations of differentiators in the discrete-time (or Z) domain, the authors implement the differentiators with equal-length transmission lines. The approach method consists of discrete signal processing techniques, transfer functions in the Z domain and optimisation algorithms. Three differentiators with different time constants and frequency bands are built and tested. The experimental results are in good agreement with theoretical values.
